Texto completoPositron Emission Tomography (PET) is a nuclear imaging technique widely used in oncology and neuroscience to observe biological activity in the body. Detection of two gamma quanta with the energy 511 keV emitted by positron annihilation in tissues allows one to reconstruct the tracer activity distribution in the body of the patient. Additional measurement of the difference in time detection between the two photons lets us to improve significantly the quality of the reconstructed image (time-of-flight method).In this manuscript, we present the development of two innovative detectors, fast and efficient, used to detect Cherenkov light produced by electrons from the photo-ionization conversions of 511 keV gamma quanta. The first one, intended for use in a brain PET scanner of a high spatial resolution, uses TriMethylBismuth for the detection medium. The second one, planned to be used to construct a whole-body PET scanner, enforces a PbF₂ crystal as Cherenkov radiator. In both configurations a micro-channel photo-multiplier (MCP-PMT) is used to detect Cherenkov photons. We commissioned an electronic detection chain with a time resolution limited to 5 ps (RMS). Using precise MCP-PMT calibration, we were able to develop simultaneously detectors with high efficiency, up to 25 %, and time resolution as good as 200 ps (FWHM).We highlight the limitations of detectors time resolution and suggest several developments in order to improve performances of Cherenkov light detectors

Texto completo[CA] El creixement de les plantes es pot veure afectat per l'estrès abiòtic, sequera, salinitat o altes temperatures. La transducció de senyals d'estrès abiòtic és fonamental per a generar una resposta fisiològica adequada, que implica la participació de diferents hormones vegetals, sent l'àcid abscísic (ABA) el regulador hormonal crític en la regulació de la resposta de la planta a situacions d'estrès per dèficit hídric. La ruta de senyalització d'ABA i els components principals de la ruta estan ben caracteritzats molecularment i bioquímica. Els receptors "Pyrabactin Resistance 1"(PYR)/"PYR1-LIKE"(PYL)/"Regulatory Component of ABA Receptor" (RCAR) exerceixen un paper important en la regulació quantitativa en resposta a l'estrès tant en llavors com en planta. Encara que la funció bioquímica dels receptors PYR/PYL/RCARs d'ABA, està ben caracteritzada en els últims anys, es coneix poc sobre altres aspectes amb rellevància biològica, com les seues modificacions postraduccionals o la regulació de la seua vida mitjana. Un dels avanços recents en aquest camp ha sigut el descobriment d'una nova família d'E3 ligases anomenades RSL1/RFAs ("RING-finger-ABA-related") que consta d'almenys 10 membres, que són reguladors clau de l'estabilitat dels receptors PYR/PYL/RCAR d'ABA en teixits d'arrels i fulles, regulant la seua degradació en diferents ubicacions cel·lulars. Un estudi més detallat d'aquesta família gènica va revelar que RSL1/RFAs es caracteritzen estructuralment per la presència de tres dominis RING putatius en tàndem, denominats "RING1-IN BETWEEN RING-RING2" (RBR), i en conseqüència pertanyen a la família d'E3 ligases de tipus RBR. Cinc membres de la família RSL1/RFA, RSL1 i RFA6-RFA9, contenen un domini TM en l'extrem C-terminal, la qual cosa suggereix que RFA6-RFA9 també es localitzen en la membrana plasmàtica. No obstant això, altres membres d'aquesta família d'E3 ligases com RFA1-RFA5 manquen del domini TM C-terminal i la seua caracterització funcional, així com la seua ubicació cel·lular, encara no ha sigut investigada. Vam mostrar que l'E3 ligasa RFA1 es localitza tant en el nucli com en el citosol, mentre que RFA4 mostra una localització específica en el nucli promovent la degradació nuclear dels receptors ABA. Per tant, els membres de la família RSL1/RFA interactuen amb els receptors ABA en la membrana plasmàtica, el citosol i el nucli, dirigint-los a la seua degradació a través de la vía endosomal/vacuolar (en el cas de RSL1) o el proteosoma 26S (per a RFA1 i RFA4). Proporcionem informació sobre la funció fisiològica d'aquestes E3 ligases de tipus RBR. Realitzant tant mutagènesis com a assajos bioquímics per a identificar la cisteïna 361 (Cys361) en RFA4 com la Cys del lloc actiu, que és una característica distintiva de les E3 ligases de tipus RBR. Hem demostrat mitjançant una anàlisi d'immuno-transferència del mutant amb pèrdua de funció de rfa1rfa4 que els nivells endògens dels receptors d'ABA PYR1 i PYL4 augmenten en comparació amb les plantes de tipus silvestre. D'altra banda, hem identificat un enzim E2, "Ubiquitin Conjugating Enzyme 26" (UBC26), com l'enzim nuclear canònic E2 que interactua amb l'E3 ligasa RFA4 i forma complexos UBC26-RFA4-Receptor, formant agregats nuclears. També generem al·lels ubc26 amb pèrdua de funció que mostraven una major sensibilitat a ABA i acumulació de receptors ABA en comparació amb el tipus silvestre. En definitiva, hem revelat un sofisticat sistema d'ubiquitinació de receptors ABA en diferents ubicacions subcel·lulars dut a terme a través de la família d'E3 ligases RSL1/RFA de tipus RBR. Hem iniciat proves bioquímiques per a identificar la S-acilació en el domini TM de RSL1. Hem generat RSL1C334S, RSL1 C5S i RSL1C6S mitjançant mutagènesis, així com RSL1ΔTM que presenta una delecció del domini TM. [EN] Plant growth is affected by abiotic stress, drought, salinity or high temperature. Signal transduction of abiotic stress is crucial to generate an appropriated physiological response, which involves the participation of different plant hormones, being abscisic acid (ABA) the critical hormonal regulator in regulating the plant's response to situations of stress due to water deficit. The ABA signaling pathway and the major components of the pathway are well characterized molecularly and biochemically. Pyrabactin Resistance 1 (PYR)/PYR1-LIKE (PYL)/Regulatory Component of ABA Receptor (RCAR) ABA receptors play an important role in quantitative regulation of ABA signaling both in seeds and vegetative tissues. Although the biochemical function of the PYR/PYL/RCAR ABA receptors has been well established in recent years, little is known about other aspects with biological relevance, such as their post-translational modifications or the regulation of their half-life. One of the recent advances in this field has been the discovery of a new family of E3 ligases called RSL1/RFAs (RING-finger-ABA-related) that consists of at least 10 members, which are key regulators of the stability of PYR/PYL/RCARs in root and leaf tissues, and regulate the degradation of ABA receptors at different cellular locations. Further inspection of the gene family revealed that RSL1/RFAs are structurally characterized by the presence of three putative RING domains in tandem, named as RING1-IN BETWEEN RING (IBR)-RING2, and accordingly they belong to the RBR-type E3 ligase family. Five members of the RSL1/RFA family, that is, RSL1 and RFA6-RFA9, contain a TM domain at the C-terminal end of the proteins, which suggests that RFA6-RFA9 are also localized in plasma membrane. However, other members of this family of E3 ligases such as RFA1-RFA5 lack the C-terminal TM domain and their functional characterization, as well as their cellular location, has not been investigated yet. In this study we show that the E3 ligase RFA1 is localized both in the nucleus and in the cytosol, while RFA4 shows a specific localization in the nucleus promoting the nuclear degradation of ABA receptors. Therefore, we members of the RSL1/RFA family interact with ABA receptors at the plasma membrane, cytosol and nucleus, targeting them for degradation via the endosomal/vacuolar pathway (in the case of RSL1) or the 26S- proteasome (for RFA1 and RFA4). We provide information on the physiological function of these RBR-type E3 ligases, which are hardly explored in plants. Additionally, we performed mutagenesis and biochemical assays to identify Cys361 in RFA4 as the active site cysteine, which is a distinctive feature of RBR-type E3 ligases. We have shown by immunoblot analysis of the rfa1rfa4 loss-of-function mutant that endogenous levels of ABA receptors PYR1 and PYL4 are increased compared to wild-type plants. On the other hand, we have identified an E2 enzyme, Ubiquitin Conjugating Enzyme 26 (UBC26), as the canonical nuclear enzyme E2 that interacts with the E3 ligase RFA4 and forms UBC26-RFA4-Receptor complexes, forming nuclear aggregates. We also generated loss-of function ubc26 alleles that exhibited higher sensitivity to ABA and accumulation of ABA receptors compared to wild type. We have revealed a sophisticated ubiquitination system of ABA receptors in different subcellular locations carried out through the RBR-type RSL1/RFA family of E3 ligases. We have proceeded with the biochemical and genetic study of the different members of the family. We have started biochemical tests to identify the S-acylation in the TM domain of RSL1. To this end, we have generated RSL1C334S, RSL1 C5S and RSL1C6S by mutagenesis as well as RSL1ΔTM, a deletion of the TM domain. Initial studies have shown that Cys residues close to the TM domain are S-acylated. Finally, we have also generated new combined mutants: rsl1rfa1, rsl1rfa5, rfa1rfa5 and rsl1rfa1rfa5.

Resumo: Pesquisas no campo da tecnologia têm buscado avaliar técnicas e/ou processos econômicos e eficientes para o tratamento de efluentes que contenham metal pesado. Atualmente a bioadsorção vem se apresentando como uma boa alternativa às tecnologias existentes. Este trabalho teve como objetivo a investigação da macrófita Azolla pinnata como bioadsorvente de cinco íons metálicos: Cd2+, Cu2+, Ni2+, Pb2+ e Zn2+ em soluções sintéticas. A caracterização do biomaterial foi realizada utilizando-se técnicas como Microscopia Eletrônica de Varredura (MEV) para análise da superfície do material; análise de Espectroscopia de Energia Dispersiva de Raios-X (EDX), que mostrou que qualitativamente a diminuição de íons de competição (Ca, Na e K) após o processo de adsorção, evidenciando que a troca iônica pode ser um dos mecanismos do processo; e análise de Infravermelho por Transformada de Fourier (FTIR), em que se pôde constatar que alguns grupos funcionais como carboxilas e grupos sulfonados são fundamentais no processo. A cinética de adsorção foi ajustada pelos modelos de pseudoprimeira e pseudossegunda ordem, sendo que o melhor ajuste variou de acordo com o íon estudado. O modelo de difusão intrapartícula mostrou que muitos mecanismos podem ser limitantes na taxa com a qual ocorre o processo. Os dados de equilíbrio foi ajustado pelos modelos de isoterma de Langmuir e de Freundlich em duas temperaturas, 25 e 45 ºC. O processo foi favorecido pelo acréscimo de temperatura para todos os íons. O modelo de Langmuir se ajustou melhor aos dados experimentais de todos os íons e o valor de capacidade máxima de adsorção (qmax) a 25 ºC foi de 0,359; 0,448; 0,254; 0,472 e 0,357 mmol/g para Cd2+, Cu2+, Ni2+, Pb2+ e Zn2+, respectivamente. Os eluentes HCl, EDTA e CaCl2 foram testados no processo de dessorção em batelada e o cloreto de cálcio se mostrou eficiente além de não causar danos ao material nem diminuir a capacidade de adsorção da macrófita. Ensaios dinâmicos em coluna de leito fixo foram realiados para os íons Cu2+ e Pb2+ com soluções mono e bicomponentes. Nos ensaios em sistema binários, pode-se constatar uma grande preferência pela adsorção de íons chumbo e, portanto, uma grande queda na adsorção de cobre. Foram testados ciclos de adsorção/dessorção em coluna utilizando o CaCl2 como eluente e, em três ciclos, a macrófita manteve suas propriedades adsortivas. Com estes resultados, pode-se constatar que a realização deste trabalho contribuiu para a avaliação deste material como bioadsorvente alternativo e que, especificamente, macrófita A. pinnata mostrou-se atrativa para utilização nos processos de tratamento e/ou estabilização de metais pesados presentes em baixas concentrações em efluentes industriais
Resumo: No processo Kraft para produção de papel e celulose, os cavacos de madeira são cozidos em um digestor com uma solução aquosa denominada licor branco, constituída de hidróxido de sódio (NaOH) e sulfeto de sódio (Na2S). Durante o cozimento uma pasta fibrosa é produzida. Esta pasta é filtrada e segue para o branqueamento. O produto filtrado chama-se licor negro, devido à mudança da sua cor nesta etapa. O licor negro possui aproximadamente 15 % de sólidos em massa. Ele é bombeado para a unidade de recuperação, na qual ele será concentrado em evaporadores de múltiplosefeitos até 75 % de sólidos em massa. Em seguida, ele é bombeado para uma caldeira onde é queimado, para recuperar os sais inorgânicos e, principalmente, fornecer calor para geração de calor. Durante a evaporação do licor negro, quando a porcentagem em sólidos ultrapassa 50 %, ocorre a precipitação de sais inorgânicos. Estes sais se depositam nas superfícies de troca térmica dos evaporadores diminuindo sua eficiência e causando prejuízos à indústria. Os principais sais que causam este problema são o carbonato de sódio (Na2CO3) e o sulfato de sódio (Na2SO4). O objetivo do presente trabalho foi testar e validar um método experimental para a determinação da quantidade de Na2CO3 presente em amostras industriais de licor negro. Além disso, também foram analisadas algumas propriedades físico-químicas do licor negro, como massa específica, teor de sólidos e viscosidade. Com os resultados obtidos, tentou-se obter uma correlação entre o teor de carbonato de sódio e as demais propriedades do licor que foram determinadas. Os resultados das análises das características físico-químicas demonstraram coerência e boa reprodutibilidade, porém, os resultados das análises de teor de carbonato de sódio não foram satisfatórios e mostraram que a técnica deve ser aperfeiçoada
Abstract: Research in the technology field has been looking for evaluating efficient and economic techniques and/or process for effluents that contains heavy metal. The biodsorption process has been seemed as a good alternative to the existing technology. This work had the aim to investigate the use of Azolla pinnata macrophyte as biosorbent of five metallic ions: Cd2+, Cu2+, Ni2+, Pb2+ and Zn2+ in synthetic solutions. The biomaterial characterization was carried out by techniques as scanning electron microscopy (SEM) for the analyze of material superficies; the energy dispersive X-ray spectroscopy (EDX), which showed, qualitatively, a decreasing in the competitions ions (Ca, Na e K) after the sorption process, indicating that the ion exchange can be one o the process mechanism; and Fourier Transform Infrared Spectroscopy (FTIR), in which it was possible implied functional groups as carboxyl and sulfonate play a important role in the adsorption. The kinetics was adjusted by primer and second order models and the model which fit better depends on each ion. The intra-particle diffusion model showed that several mechanisms may be limiting to the process rate. The equilibrium data were adjusted by Langmuir and Freundlich models in two temperatures: 25 and 45 ºC. The process was favored by the increasing in the temperature for all five ions. The Langmuir model fit better the experimental data and the maximum adsorption capacity (qmax) were 0,359; 0,448; 0,254; 0,472 and 0,357 mmol/g for Cd2+, Cu2+, Ni2+, Pb2+ and Zn2+, respectively. The HCl, EDTA and CaCl2 eluents were tested in the desorption process and the calcium chloride was efficient and did not cause any damage in the biomaterial or decreasing the biosorption capacity. Dynamics runs were carried out in fixed-bed in mono and bi-components solutions. In the binary were found a great preference for lead instead of copper and as a result, a significant decrease in the removal of copper. Adsorption/desorption cycles were tested in fixed-bed system using CaCl2 as eluent. After three cycles no damage, weight lost or significant decrease in the biosorption capacity was noticed. These results indicating that the A.pinnata macrophyte is an attractive potential new biosorbent to be used in heavy metal removal especially in low concentrations effluents
Abstract: In the Kraft process for pulp and paper production, wood chips are cooked in a digester with an aqueous solution named white liquor, consisted by sodium hydroxide (NaOH) and sodium sulfide (Na2S). During this cooking process, wood chips are transformed generating a fibrous paste and a liquid solution. This paste is filtered and separated from the liquid solution, named black liquor due to its color. The paste goes to the bleaching stage to produce cellulose. Black liquor contains inorganic and organic (mainly lignin) compounds from wood. Initially this black liquor contains approximately 15 mass/% of solids and is sent to the recovery unit to have this concentration raised to approximately 75 mass/% to be burned in a recovery boiler in order to generate power and recover inorganic reactants, recycling them to the process. The concentration process of black liquor occurs in a multiple effect evaporators unit. During this process, when the concentration of solids exceeds 50%, the precipitation of inorganic salts (mainly Na2CO3 and Na2SO4) begins. These salts are deposited in the heat transfer surfaces of the evaporator tubes, reducing their efficiency, leading to a reduction in the production capacity. The scope of this work was testing and validate an experimental method to estimate the content of Na2CO3 present in industrial black liquor samples. Moreover, some black liquor's physical-chemical properties as density, solids content and viscosity were also analyzed. An attempt to develop a predictive model to evaluate sodium carbonate content from the other properties was made. The results obtained for physical-chemical properties have shown a good consistency. However, the results of sodium carbonate analyses were not satisfactory, showing that the technique must be improved

Texto completoThis research was born for our love to pets and we want to give them a better life quality and the need that pet owners to have immediate and quality care for our furry friends when an emergency happens. From that need is born SAFE PET, that is a pet insurance, which offers a standard package that includes many benefits and coverages for affiliated pets. The main source of income for SAFE PET will be the sale of our policies, our initial investment will be S/ 124,890 which will be financed by 60% the 4 partners of the company and 40% with a loan to a financial institution. Due to the research we conducted through surveys of pet owners, most of them told us that they wanted to have insurance for their pet, and then thanks to our landing page we could observe that of 100% of visits to our landing, 54.6% left their data Because you are interested in our insurance, that percentage is much higher than we expected and that encourages us much more to be one of the first in the country to dedicate ourselves exclusively to comprehensive pet insurance. Finally, after the financial study carried out on our company, we have considered in the sensitivity analysis COK (89.71%) and a weighted average cost of capital of WACC (74.53%), to finish after all our financial plan we can conclude that SAFE PET, it is an economically viable company.

Likewise, the current context has been evaluated and the respective study that supports the viability of the project has been prepared. Nowadays people care more about the safety of their pets, there is a petfriendly trend that has led society to increase consumption of feed, veterinary expenses and canine spas. Considering that there is currently no business similar to that proposed, the student authors of the present work have considered that the creation of pet insurance in Peru would be highly profitable. To support the idea presented, all aspects concerning the creation of the legal entity, strategic objectives, internal and external policies, financial analysis and the profitability of the project have been taken into consideration. After analyzing the aforementioned aspects, and considering all aspects that may influence its development, it is concluded that the business is viable.

El servicio está orientado a hombres y mujeres mayores de 25 años, que estén interesados en el mantenimiento de los jardines de sus residencias, por lo que se estima un tamaño de mercado de USD$15,114,390.00 dólares al año. La elección de la provincia de Panamá Oeste responde a 3 motivos principales: por un lado, en los últimos años la provincia ha presentado un crecimiento demográfico significativo, siendo el punto de mayor producción residencial del país. Fenómeno que se ha dado debido a que el 70% de los proyectos residenciales son de interés preferencial, ya que la Ciudad de Panamá no es capaz de cubrir la gran demanda de viviendas que exige la población, por lo que recurren a áreas aledañas a la ciudad donde residir. Otro motivo es que Panamá Oeste es considerada la “Ciudad Dormitorio”, ya que la mayoría de los ciudadanos que residen en esta zona, trabajan en la ciudad en jornadas extendidas, por lo que no disponen de tiempo para realizar actividades como la jardinería. El último y uno de los más relevantes motivos es que en La Chorrera no existe una empresa que brinde el servicio de jardinería, más bien empresas de jardinera en la ciudad deben viajar hasta la provincia para prestar sus servicios. Su equipo gestor está representado por Milagros Macias, Lic. En Ingeniera Marítima, con conocimiento y experiencia en servicio al cliente y manejo de personal. Además cuenta con habilidades en el diseño de jardines utilizando los módulos de AutoCAD. El análisis financiero realizado muestra que el proyecto es factible y que requiere de una inversión inicial de USD$45,000 dólares. El VAN que arroja es de USD$106,227.36 dólares, la TIR de 76% garantiza la capacidad adquisitiva si la tasa de descuento subiera y el análisis de sensibilidad muestra que existe un 69% de probabilidades de éxito en su ejecución, convirtiéndolo en un proyecto que espera una buena rentabilidad.

Texto completoThis thesis aims at developing hybrid nanoparticles based on silica by reverse microemulsion. The silica nanoparticles are the basic matrix containing and protecting organic molecules and/or metallic nanoparticles. The combined incorporation of different entities within the silica opens wide prospects for the introduction of new properties related to the hybrid structure. To develop such objects, we used reverse micelles based on water, Triton X-100, hexanol and cyclohexan as reaction medium. The influence of operating conditions on the control of the size of reverse micelles was first studied. These micelles were then set to be used as nanoreactors for the synthesis of silica nanoparticles by sol-gel using suitable alkoxysilanes precursors. We monitored how it was possible to control the size of silica nanoparticles based on the water to surfactant ratio. It was thus possible to prepare in a reproducible way nanoparticles with sizes varying from 30 nm to 200 nm. We then investigated the possibility to encapsulate, in this nanoscaled matrix, fluorophores and nanoparticles of gold and silver in a controlled manner. To ensure a good colloidal stability in solution, these hybrid nanoparticles were, on the one hand, modified by adding a functional silane and, on the other hand, by click chemistry. We have thus shown that it is possible to perform, in a same micellar media, all of manufacturing process of the hybrid nanoparticle, from the silica matrix to its functionalization passing by the incorporation of functional entities. This method of sequential synthesis allowed us to bypass the purification and redispersion steps that can be problematic in the conventional methods. All this work has been extended to the design of an antibacterial agent based of silver/silica nanoparticles, capable of preventing bacterial growth through the gradual release of silver ions. Tests conducted in solution on the impregnated cotton and polyethylene terephtalate indeed show an interesting antibacterial character of these systems

Texto completo[ES] RESUMEN La señalización por la hormona vegetal ácido abscísico (ABA) desempeña un papel crítico en la regulación del crecimiento de la raíz y en la arquitectura del sistema radical. La promoción de crecimiento de la raíz en condiciones de estrés hídrico mediada por ABA es clave para la supervivencia de las plantas bajo condiciones limitantes de agua. En este trabajo, hemos explorado el papel de los receptores PYR/PYL/RCAR (PYRABACTIN RESISTANCE1 (PYR1)/PYR1 LIKE (PYL)/ REGULATORY COMPONENTS OF ABA RECEPTORS) de Arabidopsis (Arabidopsis thaliana) en la ruta de señalización de ABA en raíz. Así, hemos descubierto que el receptor de ABA PYL8 juega un papel no redundante en la regulación de la percepción de ABA en raíz. Inesperadamente, dada la naturaleza multigénica y la redundancia funcional parcial observada en la familia PYR/PYL/RCAR, el mutante pyl8 fue el único mutante sencillo de pérdida de función de los receptores PYR/PYL/RCAR que mostraba una sensibilidad reducida a la inhibición del crecimiento mediada por ABA en raíz. Este efecto se debe a la falta de inhibición mediada por PYL8 de varias fosfatasas del grupo A tipo 2C (PP2Cs), ya que PYL8 es capaz de interactuar in vivo con al menos cinco PP2Cs, denominadas HYPERSENSITIVE TO ABA1 (HAB1), HAB2, ABAINSENSITIVE1 (ABI1), ABI2, and PP2CA/ABA-HYPERSENSITIVE GERMINATION3 según lo han revelado la purificación por afinidad en tándem (TAP por sus siglas en inglés) y estudios proteómicos de espectrometría de masas. La transducción de la señal del ABA localizada en la membrana plasmática celular juega un papel crucial en los pasos iniciales de la señalización de la fitohormona, pero los mecanismos moleculares que unen los componentes básicos de la señalización y la membrana plasmática no están claros. Estudiando las interacciones de los receptores del ABA PYR/PYL/RCAR con la membrana plasmática hemos encontrado que éstos pueden interaccionar transitoriamente con ella de forma dependiente de calcio gracias a una familia de proteínas con dominios C2 relacionadas con la ruta de señalización de ABA (denominadas C2-domain ABA-related (CAR) proteins). Específicamente, se encontró que PYL4 interacciona de manera independiente de ABA con CAR1 tanto en la membrana plasmática como en el núcleo de las células vegetales. La proteína CAR1 pertenece a una familia multigénica constituida por 10 miembros en Arabidopsis thaliana, desde CAR1 hasta CAR10, y que solo se encuentra en plantas. Los ensayos de complementación bi-molecular de fluorescencia y de co-immunoprecipitación confirmaron la interacción en células vegetales tanto de PYL4-CAR1 como de otras parejas de PYR/PYL-CAR. La cristalización de la proteína CAR4 reveló que, además de un dominio C2 clásico de unión a lípidos dependiente de calcio, las proteínas de la familia CAR presentan un dominio específico que probablemente es responsable de la interacción con los receptores PYR/PYL/RCAR y de su posterior reclutamiento a las vesículas de fosfolípidos. Esta interacción es relevante para la función de los receptores PYR/PYL/RCAR en la señalización del ABA, ya que diferentes mutantes triples car de pérdida de función, que tienen afectados los genes CAR1, CAR4, CAR5, y CAR9, demostraron una reducción de la sensibilidad al ABA en ensayos de establecimiento de plántula y crecimiento de la raíz. En resumen, hemos identificado nueva familia de proteínas que son capaces mediar las interacciones transitorias dependientes de Ca2+ con vesículas de fosfolípidos, lo que a su vez afecta localización de PYR/PYL/RCAR y regula positivamente la señalización de ABA.

La promoció del creixement de l'arrel en condicions d'estrés hídric, regulada per ABA és clau per la supervivència de les plantes sota condicions limitants d'aigua. Amb aquest treball, hem investigat el paper dels receptors PYR/PYL/RCAR (PYRABACTIN RESISTANCE1 (PYR1)/PYR1 LIKE (PYL)/ REGULATORY COMPONENTS OF ABA RECEPTORS) d'Arabidopsis (Arabidopsis thaliana) en el camí de senyalització d'ABA en arrel. Així, hem descobert que el receptor d'ABA PYL8 exerceix un paper no redundant en la regulació de la percepció d'ABA en arrel. Inesperadament, donada la naturalesa multigènica i la redundància funcional parcial que s'observa en la família PYR/PYL/RCAR, el mutant pyl8 va ser l'únic mutant senzill de pèrdua de funció dels receptors PYR/PYL/RCAR que mostrava una sensibilitat reduïda a la inhibició del creixement mitjançada per l'ABA en l'arrel. Doncs aquest efecte es deu a la falta d'inhibició regulada per PYL8 de diverses fosfatases del grup A tipus 2C (PP2Cs), ja que PYL8 té la capacitat d'interactuar in vivo almenys amb cinc PP2Cs, anomenades HYPERSENSITIVE TO ABA1 (HAB1), HAB2, ABAINSENSITIVE1 (ABI1), ABI2, and PP2CA/ABAHYPERSENSITIVE GERMINATION3 segons ho han revelat per una banda la purificació per afinitat en tàndem (TAP són les seues sigles en anglés) i per altra banda, estudis proteòmics d'espectrometria de masses. Pel que fa a la transducció del senyal del l'ABA, la qual es localitza en la membrana plasmàtica cel¿lular, juga un paper molt important en els primers instants de la senyalització de la fitohormona, no obstant això els mecanismes moleculars que uneixen els components bàsics d'aquesta senyalització amb la membrana plasmàtica, no es troben del tot clars. Per tant, s'han estudiat les interaccions que tenen els receptors del ABA PYR/PYL/RCAR amb la membrana plasmàtica, i hem trobat que aquests tenen la capacitat d'interaccionar transitòriament amb la membrana de forma dependent al calci, gràcies a una família de proteïnes amb domini C2, les quals es troben relacionades amb la ruta de senyalització d'ABA(anomenades C2domain ABArelated (CAR) proteins).Específicament, es va trobar que PYL4 interacciona d'una manera independent al ABA amb CAR1, tant en la membrana plasmàtica, com en el nucli de les cèl¿lules vegetals. La proteïna CAR1 pertany a la família multigènica constituïda per 10 components en Arabidopsis thaliana, des de CAR1 fins CAR10, que tan sols es troba en plantes. Els assajos de complementació bimolecular de fluorescència i de co-immunoprecipitació, van confirmar la interacció en cèl¿lules vegetals, tant de PYL4CAR1 com d'altres parelles de PYR/PYL-CAR. La cristal¿lització de la proteïna CAR4 va revelar que, a més d'un domini C2 clàssic de unió a lípids dependent del calci, les proteïnes de la família CAR presenten un domini PYR/PYL/RCAR, i del seu posterior reclutament a les vesícules fosfolipídiques. Doncs, aquesta interacció és rellevant en la funció dels receptors PYR/PYL/RCAR, ja que participa en la senyalització del l'ABA. Aquesta interacció es clau per a la funció dels receptors, ja que diferents mutants triples car de pèrdua de funció, els quals posseïxen afectats els gens CAR1, CAR4, CAR5 i CAR9, van mostrar una reducció de la sensibilitat a l'ABA en assajos d'establiment de plàntula i creixement de l'arrel. En conclusió, hem identificat una nova família de proteïnes amb la capacitat d'organitzar les interaccions transitòries dependents del calci amb vesícules de fosfolípids, fet que al seu torn afecta la localització de PYR/PYL/RCAR i regula positivament la senyalització d'ABA.

It should be noted that we have carried out a thorough investigation, in order to gather valuable information, and in this way, we will achieve efficient and effective service to customers in our assigned segment, and that over time we will be able to extend. In addition, it seeks to satisfy the basic needs of the owners with respect to their pets, providing a differentiated service of high quality and easy access. Our platform aims to connect veterinarians with potential clients who seek to satisfy the needs of their pets, generating an increase in sales to veterinary companies and a portfolio of options for clients so that they can choose the best service that they consider according to their needs. For this reason, the research process will take into account the interviews and metrics to obtain accurate feedback for the project.

Nuestro proyecto pertenece a la industria de servicios, y consiste en una tienda especializada en la venta de jugos de frutas y vegetales. Los jugos naturales a base frutas y verduras, mejor conocidos como jugos verdes , está entrando en un período de rápida evolución, debido al cambio producido en los patrones de consumo en segmentos de la población panameña, cada vez más interesada en el consumo de alimentos naturales para mejorar sus hábitos alimenticios y salud. Además, este mismo segmento de personas optan por productos más prácticos, al no disponer de tiempo suficiente para su preparación. En Panamá existen algunas tiendas dedicadas a ofrecer jugos naturales de frutas y verduras, de la cual destaca GREEN BAR, la cual es percibida como la más especializada hasta el momento, adicional se encuentra ECO-FRUIT el cual adicional vende comida orgánica ligera, y FRESCHO, quien hace entregas a domicilio. La propuesta de valor radica en brindar una experiencia de consumo basada principalmente en servicio personalizado y la oferta de realizar las mezclas de frutas y vegetales al gusto del consumidor. El mercado objetivo son individuos comprendidos entre los 25 y 45 años, que tienen en común hábitos alimenticios y rutina de vida. Hemos identificado los siguientes segmentos: los pragmáticos y los racionales. Debido a que Barra Verde PTY es una marca totalmente nueva, el objetivo de este plan de mercadeo es: Generar conocimiento de marca y lograr que los consumidores asocien a Barra Verde PTY como la mejor experiencia de servicio en la categoría. Por lo tanto el posicionamiento a futuro, deseado es que Barra Verde PTY, se perciba como la mejor experiencia de compra en el mercado de Panamá en su categoría. La estrategia para conseguir los objetivos ya mencionados estará fundamentada en un Plan Comunicacional, teniendo como iniciativas y tácticas, conjuntas de mercadeo digital, promocional y relacional. La tienda está localizada en la ciudad de Panamá, en la exclusiva zona de Costa del Este, caracterizada por la alta presencia de corporaciones nacionales y multinacionales. Texto completoThe objective of this research work is to develop the business proposal, through a virtual platform, for the care of domestic pets, aimed at the owners of these pets and veterinary centers that in recent years have had a growth of 76% at the National Level and 91% only in Metropolitan Lima. A growing market of pet owners (users) has been identified that is looking every day for specialized veterinary centers that provide a high quality service, online connectivity, with updated information and at the same time at the minimum necessary costs. Likewise, veterinary centers (clients) that seek to connect them with their target audience through social networks and all computer media that increase their client portfolio and that allow them to develop the advertising of their services at low costs. In the first part of the work, the strategic planning has been developed in order to collect valuable information that has allowed to corroborate assumptions and determine the points most valued by users and customers with respect to the value proposition and thus be able to use them in the phase later, that is, in our marketing plan. Indeed, in said plan, the target audience was clearly identified and the means by which the proposal will be publicized and disseminated, as well as all the components of the marketing mix, were established. In the subsequent sections, the operations plan, the organizational and human resources structure are detailed, and finally a financial analysis is carried out, which has confirmed the feasibility of the project.

This research project aims to demonstrate the sustainability and profitability of Service Pet 360, with different market studies that will be carried out on our target audience, while developing different marketing strategies with the purpose of achieving a better positioning in the market and offer a variety of services and products. Neste trabalho foram estudados os efeitos da adição de agente compatibilizante, poli[(estireno)-co-(anidirido maleico)] [SMA] e dos ciclos de reciclagem à blenda PET/PS. Inicialmente foi avaliada a miscibilidade da blenda de poli(tereftalatode etileno) [PET] com poliestireno [PS]. Sendo esta blenda imiscível em toda a faixa de composição, foi estudada a influência da adição de SMA nas propriedades térmicas e na morfologia da blenda PET/PS/SMA. O parâmetro de interação de Flory-Huggings (c12), cujo valor está relacionado com as interações específicas entre os componentes, foi calculado a partir das temperaturas de fusão de equilíbrio do componente semicristalino puro e na blenda, como igual a +0,024. O valor positivo sugere interações desfavoráveis entre os componentes, em concordância com os outros métodos. A viabilidade de reutilização do PET em mistura com outro polímero, foi analisada através de sucessivos processos de extusão, de maneira que as propriedades microscópicas dos componentes puros e das blendas foram estudadas. A pequena redução da massa molar média do PET, não deve prejudicar o reaproveitamento do PET pós-consumo. Para as blendas PET/PS/SMA, o aumento da adesão interfacial, de certo modo, minimiza o efeito causado pela redução de massa molar durante o processo de reciclagem.

Texto completoNowadays previous warning of impending natural and environmental disasters, accurate climate prediction, and detailed understanding of the status of global water resources are everyday issues that are extremely important to the global community. The National Meteorological and Hydrological Services throughout the world are responsible for providing this information, which is necessary for the protection of the environment, economic development (transportation, energy, agriculture, etc.) and the safety of life and property. In this context, one of the services widely used worldwide is the International Data Collection System which is composed of geostationary and low-orbit (nongeostationary) satellite networks. INPE develops and operates the Brazilian Data Collection System (BDCS), which is basically composed by the Data Collection Satellite (DCS) 1 and 2, the China-Brazilian Earth Resources Satellite (CBERS) 4, 1100+ Data Collection Platforms (DCPs) distributed in the Brazilian territory, two Receiving Ground Stations (RGS) at Cuiabá-MT, and Alcântara-MA, and one MISSION Center (MC) at the INPE Northeast Regional Center (INPE/NRC) at Natal, RN The BDCS has been operating since 1993 using an analog technology that has become obsolete and must be replaced by digital systems with on-board data decoding and storage. In this scenario, carrier frequency and phase synchronization and symbol time estimation have a fundamental importance for the correct reception of the signal arriving at the receiver. Therefore, it was proposed the development of a solution for these functions using digital signal processing using MatLab codes. For the implementation of frequency synchronization, the chosen solution uses xii a second-order Digital PLL, taking into account requirements such as the Doppler Effect. For the symbol time estimator we used the feedforward algorithm, which uses a simple implementation structure, a simplification of the one proposed in [16]. Also a simple solution of a linear interpolator was used to determine the delay time. The result presented by the frequency / phase synchronizer was satisfactory for operation in part of the range. The symbol time estimator also presented satisfactory results for Eb / N0 in the 0 to 11 dB range, with a loss of less than 0.5 dB over the theoretical value.

The main objective of this work was to study the chemical and physical properties, as the bond distance and energy, of confined atoms via Hartree-Fock method. We studied the H- 'H POT. +' e HE- 'H POT. +' molecules introducing the screening effects via modification of the nuclear repulsion energy between atoms allowing thus to understanding the effects in the bond length and energy. The relevance of this work is of prime importance in material science due to many technological applications as molecules confined to surfaces, in liquid helium, neutral plasma and catalysis. All this applications has a common background of knowledge: the influence of the material physical and chemical properties with the distance and energy between the atoms in the chemical composition of the material.

Texto completoApesar da facilidade de cultivo, a produção de hortaliças é inviabilizada economicamente pela perecibilidade que causa perdas provocadas principalmente pelas condições inadequadas de conservação. A vida útil das hortaliças é beneficiada pela conservação em temperatura em torno de 10oC. Em climas quentes e secos, como no semi-árido nordestino, esta condição é possível mediante equipamentos de refrigeração que demandam custos e energia elétrica, condição esta incompatível com as das populações de baixa renda. O sistema Pot in Pot desenvolvido na Nigéria, consiste na superposição de dois potes de cerâmica de diferentes tamanhos dispostos um dentro do outro e de uma camada de areia molhada inserida entre eles que proporciona o resfriamento do pote interno pela evaporação da água, conseguindo-se aumentar o tempo de vida útil de vegetais. Com o objetivo de avaliar o efeito deste sistema de conservação sobre hortaliças produzidas no Nordeste brasileiro foi desenvolvida esta pesquisa comparando o sistema Pot in Pot (27±0,40C e 83±0,6%UR) com o ambiente de uma câmara climatizada para a condição semelhante a do semi-árido nordestino (37±0,7ºC e 40±0,5%UR) e com outro sistema convencional de conservação: refrigeração em geladeira doméstica (7±0,30C e 53±1%UR), utilizando-se amostras sem embalagem e amostras embaladas em polietileno de alta densidade. Dois tipos de hortaliças foram utilizados: pimentão e tomate, adquiridos no comércio atacadista. Os experimentos foram realizados em 3 repetições e em cada utilizando-se 72 unidades de tomate e 80 de pimentão. Todas as amostras foram selecionadas, limpas, identificadas, pesadas e fotografadas. Em seguida foi retirada uma unidade de cada amostra para ensaios laboratoriais de pH, sólidos solúveis totais (SST), acidez total titulável (ATT) e textura, correspondendo ao tempo zero do experimento. As demais amostras foram divididas em 2 porções iguais onde uma delas foi submetida a sanitizacão. Em seguida, todas as amostras sanitizadas ou não, foram armazenadas nos 3 diferentes ambientes. A cada 7 dias, as amostras foram analisadas quanto às características visuais (cor, brilho, frescor, turgência e enrugamento), perdas de peso, pH, acidez total titulável, sólidos solúveis totais e textura até apresentarem sinais de alteração que as tornaram impróprias para o consumo. Nas análises físico-químicas utilisou-se métodos preconizados por AOAC. Os resultados demonstraram diferenças significativas (p<0,05) entre os ambientes e os períodos de armazenamento. Concluiu-se que: o uso do sistema Pot in Pot proporcionou aumento da vida útil das hortaliças estudadas e foi o mais eficiente para a conservação do tomate; a refrigeração com embalagem foi o melhor ambiente para o pimentão; as características visuais e o aparecimento de fungos foram limitantes na vida útil de pimentão e a turgescência para a conservação de tomate; a sanitização com cloro não contribuiu para a conservação das hortaliças; o aparecimento de fungos em todos os sistemas utilizados aponta para a necessidade da associação de fungicida específico e que a eficiência do sistema Pot in Pot é dependente das características intrínsecas das hortaliças

Texto completoIn this research, the validation of the business idea for the creation of the Pet & Vet Club application is developed. It is a platform of integrated services for pets: teleconsultations, training and online products store. This business is aimed at pet owners, both men and women between 18 and 50 years old, who reside in Metropolitan Lima and belong to the A and B socioeconomic levels. The growth trend of pet-related services was established as a frame of reference to determine the potential of the business model. After the analysis of primary and secondary sources, it was observed that both the market for teleconsultations and the online shopping for pets present an important development that can be taken advantage of by this application, since there are no competitors with the exact offer. The value proposition demonstrated its viability in the Lima market in order to meet the needs of new consumers. Finally, the financial plan of the project was carried out in order to evaluate the feasibility of the project. The initial investment, operating costs, administrative and sales expenses were identified, as well as an estimate of sales based on the concierge and secondary sources. Finally, it was concluded that the project is viable given a positive NPV, an IRR greater than the discount rate and the DPP of 2.72 years.
