Texto completoPositron Emission Tomography (PET) is a nuclear imaging technique widely used in oncology and neuroscience to observe biological activity in the body. Detection of two gamma quanta with the energy 511 keV emitted by positron annihilation in tissues allows one to reconstruct the tracer activity distribution in the body of the patient. Additional measurement of the difference in time detection between the two photons lets us to improve significantly the quality of the reconstructed image (time-of-flight method).In this manuscript, we present the development of two innovative detectors, fast and efficient, used to detect Cherenkov light produced by electrons from the photo-ionization conversions of 511 keV gamma quanta. The first one, intended for use in a brain PET scanner of a high spatial resolution, uses TriMethylBismuth for the detection medium. The second one, planned to be used to construct a whole-body PET scanner, enforces a PbF₂ crystal as Cherenkov radiator. In both configurations a micro-channel photo-multiplier (MCP-PMT) is used to detect Cherenkov photons. We commissioned an electronic detection chain with a time resolution limited to 5 ps (RMS). Using precise MCP-PMT calibration, we were able to develop simultaneously detectors with high efficiency, up to 25 %, and time resolution as good as 200 ps (FWHM).We highlight the limitations of detectors time resolution and suggest several developments in order to improve performances of Cherenkov light detectors

Texto completo[CA] El creixement de les plantes es pot veure afectat per l'estrès abiòtic, sequera, salinitat o altes temperatures. La transducció de senyals d'estrès abiòtic és fonamental per a generar una resposta fisiològica adequada, que implica la participació de diferents hormones vegetals, sent l'àcid abscísic (ABA) el regulador hormonal crític en la regulació de la resposta de la planta a situacions d'estrès per dèficit hídric. La ruta de senyalització d'ABA i els components principals de la ruta estan ben caracteritzats molecularment i bioquímica. Els receptors "Pyrabactin Resistance 1"(PYR)/"PYR1-LIKE"(PYL)/"Regulatory Component of ABA Receptor" (RCAR) exerceixen un paper important en la regulació quantitativa en resposta a l'estrès tant en llavors com en planta. Encara que la funció bioquímica dels receptors PYR/PYL/RCARs d'ABA, està ben caracteritzada en els últims anys, es coneix poc sobre altres aspectes amb rellevància biològica, com les seues modificacions postraduccionals o la regulació de la seua vida mitjana. Un dels avanços recents en aquest camp ha sigut el descobriment d'una nova família d'E3 ligases anomenades RSL1/RFAs ("RING-finger-ABA-related") que consta d'almenys 10 membres, que són reguladors clau de l'estabilitat dels receptors PYR/PYL/RCAR d'ABA en teixits d'arrels i fulles, regulant la seua degradació en diferents ubicacions cel·lulars. Un estudi més detallat d'aquesta família gènica va revelar que RSL1/RFAs es caracteritzen estructuralment per la presència de tres dominis RING putatius en tàndem, denominats "RING1-IN BETWEEN RING-RING2" (RBR), i en conseqüència pertanyen a la família d'E3 ligases de tipus RBR. Cinc membres de la família RSL1/RFA, RSL1 i RFA6-RFA9, contenen un domini TM en l'extrem C-terminal, la qual cosa suggereix que RFA6-RFA9 també es localitzen en la membrana plasmàtica. No obstant això, altres membres d'aquesta família d'E3 ligases com RFA1-RFA5 manquen del domini TM C-terminal i la seua caracterització funcional, així com la seua ubicació cel·lular, encara no ha sigut investigada. Vam mostrar que l'E3 ligasa RFA1 es localitza tant en el nucli com en el citosol, mentre que RFA4 mostra una localització específica en el nucli promovent la degradació nuclear dels receptors ABA. Per tant, els membres de la família RSL1/RFA interactuen amb els receptors ABA en la membrana plasmàtica, el citosol i el nucli, dirigint-los a la seua degradació a través de la vía endosomal/vacuolar (en el cas de RSL1) o el proteosoma 26S (per a RFA1 i RFA4). Proporcionem informació sobre la funció fisiològica d'aquestes E3 ligases de tipus RBR. Realitzant tant mutagènesis com a assajos bioquímics per a identificar la cisteïna 361 (Cys361) en RFA4 com la Cys del lloc actiu, que és una característica distintiva de les E3 ligases de tipus RBR. Hem demostrat mitjançant una anàlisi d'immuno-transferència del mutant amb pèrdua de funció de rfa1rfa4 que els nivells endògens dels receptors d'ABA PYR1 i PYL4 augmenten en comparació amb les plantes de tipus silvestre. D'altra banda, hem identificat un enzim E2, "Ubiquitin Conjugating Enzyme 26" (UBC26), com l'enzim nuclear canònic E2 que interactua amb l'E3 ligasa RFA4 i forma complexos UBC26-RFA4-Receptor, formant agregats nuclears. També generem al·lels ubc26 amb pèrdua de funció que mostraven una major sensibilitat a ABA i acumulació de receptors ABA en comparació amb el tipus silvestre. En definitiva, hem revelat un sofisticat sistema d'ubiquitinació de receptors ABA en diferents ubicacions subcel·lulars dut a terme a través de la família d'E3 ligases RSL1/RFA de tipus RBR. Hem iniciat proves bioquímiques per a identificar la S-acilació en el domini TM de RSL1. Hem generat RSL1C334S, RSL1 C5S i RSL1C6S mitjançant mutagènesis, així com RSL1ΔTM que presenta una delecció del domini TM. [EN] Plant growth is affected by abiotic stress, drought, salinity or high temperature. Signal transduction of abiotic stress is crucial to generate an appropriated physiological response, which involves the participation of different plant hormones, being abscisic acid (ABA) the critical hormonal regulator in regulating the plant's response to situations of stress due to water deficit. The ABA signaling pathway and the major components of the pathway are well characterized molecularly and biochemically. Pyrabactin Resistance 1 (PYR)/PYR1-LIKE (PYL)/Regulatory Component of ABA Receptor (RCAR) ABA receptors play an important role in quantitative regulation of ABA signaling both in seeds and vegetative tissues. Although the biochemical function of the PYR/PYL/RCAR ABA receptors has been well established in recent years, little is known about other aspects with biological relevance, such as their post-translational modifications or the regulation of their half-life. One of the recent advances in this field has been the discovery of a new family of E3 ligases called RSL1/RFAs (RING-finger-ABA-related) that consists of at least 10 members, which are key regulators of the stability of PYR/PYL/RCARs in root and leaf tissues, and regulate the degradation of ABA receptors at different cellular locations. Further inspection of the gene family revealed that RSL1/RFAs are structurally characterized by the presence of three putative RING domains in tandem, named as RING1-IN BETWEEN RING (IBR)-RING2, and accordingly they belong to the RBR-type E3 ligase family. Five members of the RSL1/RFA family, that is, RSL1 and RFA6-RFA9, contain a TM domain at the C-terminal end of the proteins, which suggests that RFA6-RFA9 are also localized in plasma membrane. However, other members of this family of E3 ligases such as RFA1-RFA5 lack the C-terminal TM domain and their functional characterization, as well as their cellular location, has not been investigated yet. In this study we show that the E3 ligase RFA1 is localized both in the nucleus and in the cytosol, while RFA4 shows a specific localization in the nucleus promoting the nuclear degradation of ABA receptors. Therefore, we members of the RSL1/RFA family interact with ABA receptors at the plasma membrane, cytosol and nucleus, targeting them for degradation via the endosomal/vacuolar pathway (in the case of RSL1) or the 26S- proteasome (for RFA1 and RFA4). We provide information on the physiological function of these RBR-type E3 ligases, which are hardly explored in plants. Additionally, we performed mutagenesis and biochemical assays to identify Cys361 in RFA4 as the active site cysteine, which is a distinctive feature of RBR-type E3 ligases. We have shown by immunoblot analysis of the rfa1rfa4 loss-of-function mutant that endogenous levels of ABA receptors PYR1 and PYL4 are increased compared to wild-type plants. On the other hand, we have identified an E2 enzyme, Ubiquitin Conjugating Enzyme 26 (UBC26), as the canonical nuclear enzyme E2 that interacts with the E3 ligase RFA4 and forms UBC26-RFA4-Receptor complexes, forming nuclear aggregates. We also generated loss-of function ubc26 alleles that exhibited higher sensitivity to ABA and accumulation of ABA receptors compared to wild type. We have revealed a sophisticated ubiquitination system of ABA receptors in different subcellular locations carried out through the RBR-type RSL1/RFA family of E3 ligases. We have proceeded with the biochemical and genetic study of the different members of the family. We have started biochemical tests to identify the S-acylation in the TM domain of RSL1. To this end, we have generated RSL1C334S, RSL1 C5S and RSL1C6S by mutagenesis as well as RSL1ΔTM, a deletion of the TM domain. Initial studies have shown that Cys residues close to the TM domain are S-acylated. Finally, we have also generated new combined mutants: rsl1rfa1, rsl1rfa5, rfa1rfa5 and rsl1rfa1rfa5.

Resumo: Pesquisas no campo da tecnologia têm buscado avaliar técnicas e/ou processos econômicos e eficientes para o tratamento de efluentes que contenham metal pesado. Atualmente a bioadsorção vem se apresentando como uma boa alternativa às tecnologias existentes. Este trabalho teve como objetivo a investigação da macrófita Azolla pinnata como bioadsorvente de cinco íons metálicos: Cd2+, Cu2+, Ni2+, Pb2+ e Zn2+ em soluções sintéticas. A caracterização do biomaterial foi realizada utilizando-se técnicas como Microscopia Eletrônica de Varredura (MEV) para análise da superfície do material; análise de Espectroscopia de Energia Dispersiva de Raios-X (EDX), que mostrou que qualitativamente a diminuição de íons de competição (Ca, Na e K) após o processo de adsorção, evidenciando que a troca iônica pode ser um dos mecanismos do processo; e análise de Infravermelho por Transformada de Fourier (FTIR), em que se pôde constatar que alguns grupos funcionais como carboxilas e grupos sulfonados são fundamentais no processo. A cinética de adsorção foi ajustada pelos modelos de pseudoprimeira e pseudossegunda ordem, sendo que o melhor ajuste variou de acordo com o íon estudado. O modelo de difusão intrapartícula mostrou que muitos mecanismos podem ser limitantes na taxa com a qual ocorre o processo. Os dados de equilíbrio foi ajustado pelos modelos de isoterma de Langmuir e de Freundlich em duas temperaturas, 25 e 45 ºC. O processo foi favorecido pelo acréscimo de temperatura para todos os íons. O modelo de Langmuir se ajustou melhor aos dados experimentais de todos os íons e o valor de capacidade máxima de adsorção (qmax) a 25 ºC foi de 0,359; 0,448; 0,254; 0,472 e 0,357 mmol/g para Cd2+, Cu2+, Ni2+, Pb2+ e Zn2+, respectivamente. Os eluentes HCl, EDTA e CaCl2 foram testados no processo de dessorção em batelada e o cloreto de cálcio se mostrou eficiente além de não causar danos ao material nem diminuir a capacidade de adsorção da macrófita. Ensaios dinâmicos em coluna de leito fixo foram realiados para os íons Cu2+ e Pb2+ com soluções mono e bicomponentes. Nos ensaios em sistema binários, pode-se constatar uma grande preferência pela adsorção de íons chumbo e, portanto, uma grande queda na adsorção de cobre. Foram testados ciclos de adsorção/dessorção em coluna utilizando o CaCl2 como eluente e, em três ciclos, a macrófita manteve suas propriedades adsortivas. Com estes resultados, pode-se constatar que a realização deste trabalho contribuiu para a avaliação deste material como bioadsorvente alternativo e que, especificamente, macrófita A. pinnata mostrou-se atrativa para utilização nos processos de tratamento e/ou estabilização de metais pesados presentes em baixas concentrações em efluentes industriais
Resumo: No processo Kraft para produção de papel e celulose, os cavacos de madeira são cozidos em um digestor com uma solução aquosa denominada licor branco, constituída de hidróxido de sódio (NaOH) e sulfeto de sódio (Na2S). Durante o cozimento uma pasta fibrosa é produzida. Esta pasta é filtrada e segue para o branqueamento. O produto filtrado chama-se licor negro, devido à mudança da sua cor nesta etapa. O licor negro possui aproximadamente 15 % de sólidos em massa. Ele é bombeado para a unidade de recuperação, na qual ele será concentrado em evaporadores de múltiplosefeitos até 75 % de sólidos em massa. Em seguida, ele é bombeado para uma caldeira onde é queimado, para recuperar os sais inorgânicos e, principalmente, fornecer calor para geração de calor. Durante a evaporação do licor negro, quando a porcentagem em sólidos ultrapassa 50 %, ocorre a precipitação de sais inorgânicos. Estes sais se depositam nas superfícies de troca térmica dos evaporadores diminuindo sua eficiência e causando prejuízos à indústria. Os principais sais que causam este problema são o carbonato de sódio (Na2CO3) e o sulfato de sódio (Na2SO4). O objetivo do presente trabalho foi testar e validar um método experimental para a determinação da quantidade de Na2CO3 presente em amostras industriais de licor negro. Além disso, também foram analisadas algumas propriedades físico-químicas do licor negro, como massa específica, teor de sólidos e viscosidade. Com os resultados obtidos, tentou-se obter uma correlação entre o teor de carbonato de sódio e as demais propriedades do licor que foram determinadas. Os resultados das análises das características físico-químicas demonstraram coerência e boa reprodutibilidade, porém, os resultados das análises de teor de carbonato de sódio não foram satisfatórios e mostraram que a técnica deve ser aperfeiçoada
Abstract: Research in the technology field has been looking for evaluating efficient and economic techniques and/or process for effluents that contains heavy metal. The biodsorption process has been seemed as a good alternative to the existing technology. This work had the aim to investigate the use of Azolla pinnata macrophyte as biosorbent of five metallic ions: Cd2+, Cu2+, Ni2+, Pb2+ and Zn2+ in synthetic solutions. The biomaterial characterization was carried out by techniques as scanning electron microscopy (SEM) for the analyze of material superficies; the energy dispersive X-ray spectroscopy (EDX), which showed, qualitatively, a decreasing in the competitions ions (Ca, Na e K) after the sorption process, indicating that the ion exchange can be one o the process mechanism; and Fourier Transform Infrared Spectroscopy (FTIR), in which it was possible implied functional groups as carboxyl and sulfonate play a important role in the adsorption. The kinetics was adjusted by primer and second order models and the model which fit better depends on each ion. The intra-particle diffusion model showed that several mechanisms may be limiting to the process rate. The equilibrium data were adjusted by Langmuir and Freundlich models in two temperatures: 25 and 45 ºC. The process was favored by the increasing in the temperature for all five ions. The Langmuir model fit better the experimental data and the maximum adsorption capacity (qmax) were 0,359; 0,448; 0,254; 0,472 and 0,357 mmol/g for Cd2+, Cu2+, Ni2+, Pb2+ and Zn2+, respectively. The HCl, EDTA and CaCl2 eluents were tested in the desorption process and the calcium chloride was efficient and did not cause any damage in the biomaterial or decreasing the biosorption capacity. Dynamics runs were carried out in fixed-bed in mono and bi-components solutions. In the binary were found a great preference for lead instead of copper and as a result, a significant decrease in the removal of copper. Adsorption/desorption cycles were tested in fixed-bed system using CaCl2 as eluent. After three cycles no damage, weight lost or significant decrease in the biosorption capacity was noticed. These results indicating that the A.pinnata macrophyte is an attractive potential new biosorbent to be used in heavy metal removal especially in low concentrations effluents
Abstract: In the Kraft process for pulp and paper production, wood chips are cooked in a digester with an aqueous solution named white liquor, consisted by sodium hydroxide (NaOH) and sodium sulfide (Na2S). During this cooking process, wood chips are transformed generating a fibrous paste and a liquid solution. This paste is filtered and separated from the liquid solution, named black liquor due to its color. The paste goes to the bleaching stage to produce cellulose. Black liquor contains inorganic and organic (mainly lignin) compounds from wood. Initially this black liquor contains approximately 15 mass/% of solids and is sent to the recovery unit to have this concentration raised to approximately 75 mass/% to be burned in a recovery boiler in order to generate power and recover inorganic reactants, recycling them to the process. The concentration process of black liquor occurs in a multiple effect evaporators unit. During this process, when the concentration of solids exceeds 50%, the precipitation of inorganic salts (mainly Na2CO3 and Na2SO4) begins. These salts are deposited in the heat transfer surfaces of the evaporator tubes, reducing their efficiency, leading to a reduction in the production capacity. The scope of this work was testing and validate an experimental method to estimate the content of Na2CO3 present in industrial black liquor samples. Moreover, some black liquor's physical-chemical properties as density, solids content and viscosity were also analyzed. An attempt to develop a predictive model to evaluate sodium carbonate content from the other properties was made. The results obtained for physical-chemical properties have shown a good consistency. However, the results of sodium carbonate analyses were not satisfactory, showing that the technique must be improved

Texto completoThis research was born for our love to pets and we want to give them a better life quality and the need that pet owners to have immediate and quality care for our furry friends when an emergency happens. From that need is born SAFE PET, that is a pet insurance, which offers a standard package that includes many benefits and coverages for affiliated pets. The main source of income for SAFE PET will be the sale of our policies, our initial investment will be S/ 124,890 which will be financed by 60% the 4 partners of the company and 40% with a loan to a financial institution. Due to the research we conducted through surveys of pet owners, most of them told us that they wanted to have insurance for their pet, and then thanks to our landing page we could observe that of 100% of visits to our landing, 54.6% left their data Because you are interested in our insurance, that percentage is much higher than we expected and that encourages us much more to be one of the first in the country to dedicate ourselves exclusively to comprehensive pet insurance. Finally, after the financial study carried out on our company, we have considered in the sensitivity analysis COK (89.71%) and a weighted average cost of capital of WACC (74.53%), to finish after all our financial plan we can conclude that SAFE PET, it is an economically viable company.

Texto completoThis work proposes the start-up of an innovative business, for this, the external and internal factors that could have an impact on its development have been considered. Likewise, the current context has been evaluated and the respective study that supports the viability of the project has been prepared. Nowadays people care more about the safety of their pets, there is a petfriendly trend that has led society to increase consumption of feed, veterinary expenses and canine spas. Considering that there is currently no business similar to that proposed, the student authors of the present work have considered that the creation of pet insurance in Peru would be highly profitable. To support the idea presented, all aspects concerning the creation of the legal entity, strategic objectives, internal and external policies, financial analysis and the profitability of the project have been taken into consideration. After analyzing the aforementioned aspects, and considering all aspects that may influence its development, it is concluded that the business is viable.
