Jokes are specific discourses that contain cultural, historical and region-specific references. In order to understand the salience of these discourses, the common ground of interlocutors is significantly important. The current study explores how non-natives English speakers comprehend English jokes. In addition, it aims to know the difficulties and the lexical issues encountered by Pakhtoon and Sindhi participants in interpreting English jokes. The researchers used a mixed-method approach for this study. The population of this study includes Pakhtoon and Sindhi undergraduate students.A sample of 20 students was selected and two types of tools were used to check the English jokes comprehension skills of participants. The socio-cognitive approach is used as a theoretical framework and the results of quantitative data were presented with the help of pie charts. Results of this study show that ethnic, religious, racial, and jokes having historic references are difficult to understand. Individuals' social knowledge of English culture is important for comprehending English jokes. Knowledge of semantic expressions, ample knowledge and wisdom of culture-specific words and idioms are important for identifying the salience aspects of English jokes.

The present study evaluates marker assisted forward breeding (MAFB)-derived disease resistant introgression lines (ILs) which do not have the targeted resistance genes for bacterial blight (xa5 + xa13 + Xa21) and blast (Pi2 + Pi9 + Pi54). The ILs were derived in the background of two elite rice cultivars, Krishna Hamsa [Recurrent Parent 1 (RP1)] and WGL 14 (RP2), involving multi-parent inter-crossing. Molecular characterization with gene specific markers for seven reported resistance genes each for bacterial blight (Xa33, Xa38, xa23, Xa4, xa8, Xa27 and Xa41) and blast (Pi1, Pi20, Pi38, Pib, Pitp, Pizt and Pi40) revealed the presence of xa8 and Xa38, in addition to the targeted xa5, xa13 and Xa21 for bacterial blight resistance and Pi1, Pi38, Pi40, Pi20, Pib and Pipt, in addition to the targeted Pi9 and Pi54, for blast resistance in various combinations. A maximum of nine resistance genes xa5 + Xa21 + Pi54 + xa8 + Pipt + Pi38 + Pi1 + Pi20 + Pib was observed in RP1-IL 19030 followed by eight genes xa5 + xa13 + Xa21 + xa8 + Pi9 + Pipt + Pi1 + Pi20 in two RP2-ILs, 19344 and 19347. ANOVA revealed the presence of significant variability for all the yield traits except “days to 50% flowering” (DFF). Box plots depicted the seasonal differences in the phenotypic expression of the yield traits. There was significant positive association of grain yield with days to flowering, tiller number and panicle number. Thousand grain weight is also significantly and positively correlated with grain yield. On the contrary, grain yield showed a significantly negative association with plant height. Multi-parent selective inter-crossing in the present study not only led to the development of high yielding disease resistant ILs but also enhanced recovery of the recurrent parent via selection for essential morphological features. More than 90.0% genetic similarity in the ILs based on SNP-based background selection demonstrated the success of multi-parent selective intercrossing in the development of disease resistant NILs.

This paper describes the effect of input uncertainties on a set of integral parameters (kinf, nuclide compositions) associated with the validation of CASMO-5 against PIE data. The nuclear data under consideration are the cross-sections, fission spectrum and neutron multiplicities and fission yields. Various sources of covariance information are considered, novel ones (ENDFB-VIII.0, JEFF-3.3) as well as more widely distributed ones (JENDL-4.0, ENDF/B-VII.1, Scale 6.1 and Scale 6.2). All possible nuclide reaction pairs (cross sections, fission spectrum and averaged number of neutron per fission) have been perturbed, e.g. all isotopes available in both the respective covariance libraries and the CASMO-5 library. The evolution of the uncertainty estimates with exposure is complemented with sensitivity analysis to determine the main contributors to the uncertainty. The Pearson coefficient defined between the model output and a given input is used in this work to assess the part of the variance in the model output coming from the considered input uncertainty. It is a very promising measure of sensitivity as it is computationally cheap even though it assumes linearity of the output with respect to input perturbations. The evolution of the uncertainty with exposure, both in terms of trends and magnitude are however very different. Sensitivity analysis allows determining why the trends and magnitudes are different.

Resumen
Oligo(phenylimines) hereafter named PI6, PI8 and PI9 were synthesized by condensation reaction between the terephtalaldehyde and diamines of different length size (diaminohexane, diaminooctane and diaminononane) via mechanochemistry. The reactions were carried out without solvent or catalyst and completed after only 90 minutes. The products were characterized by 1H-NMR, UV-Vis, FT-IR spectroscopy, GPC, DSC and Small angle X-ray diffraction. Thin films were prepared by spin coating from concentrated solutions on glass and quartz substrates and mechanically rubbed with a polyacrylonitrile tissue. The oriented films were studied by both Laser Scanning Confocal Microscopy (LSCM) and UV-Vis spectroscopy. With this last technique, the higher optical dichroism was observed for PI6. The oriented films of this oligo(phenylimine) were thus used as patterns to induce molecular orientation of a fluorescent phenyleneethynylene polymer in order to obtain polarized light emission.

Resumen
BackgroundPancreatic ductal adenocarcinoma (PDAC) is an aggressive cancer without effective therapies and with poor prognosis, causing 7% of all cancer-related fatalities in the USA. Considering the lack of effective therapies for this aggressive cancer, there is an urgent need to define newer and more effective therapeutic strategies. Polyinosine–polycytidylic acid (pIC) is a synthetic double-stranded RNA (dsRNA) which directly activates dendritic cells and natural killer cells inhibiting tumor growth. When pIC is delivered into the cytoplasm using polyethyleneimine (PEI), pIC-PEI, programmed-cell death is induced in PDAC. Transfection of [pIC]PEIinto PDAC cells inhibits growth, promotes toxic autophagy and also induces apoptosis in vitro and in vivo in animal models.MethodsThe KPC transgenic mouse model that recapitulates PDAC development in patients was used to interrogate the role of an intact immune system in vivo in PDAC in response to [pIC]PEI. Antitumor efficacy and survival were monitored endpoints. Comprehensive analysis of the tumor microenvironment (TME) and immune cells, cytokines and chemokines in the spleen, and macrophage polarization were analyzed.ResultsCytosolic delivery of [pIC]PEIinduces apoptosis and provokes strong antitumor immunityin vivoin immune competent mice with PDAC. The mechanism underlying the immune stimulatory properties of [pIC]PEIinvolves Stat1 activation resulting in CCL2 and MMP13 stimulation thereby provoking macrophage polarization. [pIC]PEIinduces apoptosis via the AKT-XIAP pathway, as well as macrophage differentiation and T-cell activation via the IFNγ-Stat1-CCL2 signaling pathways in PDAC. In transgenic tumor mouse models, [pIC]PEIpromotes robust and profound antitumor activity implying that stimulating the immune system contributes to biological activity. The [pIC]PEIanti-PDAC effects are enhanced when used in combination with a standard of care (SOC) treatment, that is, gemcitabine.ConclusionsIn summary, [pIC]PEItreatment is non-toxic toward normal pancreatic cells while displaying strong cytotoxic and potent immune activating activities in PDAC, making it an attractive therapeutic when used alone or in conjunction with SOC therapeutic agents, potentially providing a safe and effective treatment protocol with translational potential for the effective therapy of PDAC.

Resumen
ABSTRACT The in vitro development of resistance to the new nonfluorinated quinolones (NFQs; PGE 9262932, PGE 4175997, and PGE 9509924) was investigated in Staphylococcus aureus. At concentrations two times the MIC, step 1 mutants were isolated more frequently with ciprofloxacin and trovafloxacin (9.1 × 10−8 and 5.7 × 10−9, respectively) than with the NFQs, gatifloxacin, or clinafloxacin (<5.7 × 10−10). Step 2 and step 3 mutants were selected via exposure of a step 1 mutant (selected with trovafloxacin) to four times the MICs of trovafloxacin and PGE 9262932. The step 1 mutant contained the known Ser80-Phe mutation in GrlA, and the step 2 and step 3 mutants contained the known Ser80-Phe and Ser84-Leu mutations in GrlA and GyrA, respectively. Compared to ciprofloxacin, the NFQs were 8-fold more potent against the parent and 16- to 128-fold more potent against the step 3 mutants. Mutants with high-level NFQ resistance (MIC, 32 μg/ml) were isolated by the spiral plater-based serial passage technique. DNA sequence analysis of three such mutants revealed the following mutations: (i) Ser84-Leu in GyrA and Glu84-Lys and His103-Tyr in GrlA; (ii) Ser-84Leu in GyrA, Ser52-Arg in GrlA, and Glu472-Val in GrlB; and (iii) Ser84-Leu in GyrA, Glu477-Val in GyrB, and Glu84-Lys and His103-Tyr in GrlA. Addition of the efflux pump inhibitor reserpine (10 μg/ml) resulted in 4- to 16-fold increases in the potencies of the NFQs against these mutants, whereas it resulted in 2-fold increases in the potencies of the NFQs against the parent.

We previously reported that CaCl2 hydrothermal-treated (Ca-HT) titanium (Ti) implants induced a tight sealing at the interface between the implant and peri-implant epithelium (PIE) after implantation. However, it is not clear how long this improved epithelium sealing can be maintained. We subsequently investigated whether the positive effect of Ca-HT to promote sealing between the PIE and implant was sustained longer term. Maxillary molars were extracted from rats and replaced with either Ca-HT implants (Ca-HT group), distilled water-HT implants (DW-HT group) or non-treated implants (control group). After 16 weeks, the majority of implants in the Ca-HT group remained at the maxillary with no apical extension of the PIE. Conversely, half the number of control implants was lost following down-growth of the PIE. The effect of Ca-HT on migration and proliferation of rat oral epithelial cells (OECs) was also investigated. In OECs cultured on Ca-HT Ti plates, protein expression in relation to cell migration decreased, and proliferation was higher than other groups. Surface analysis indicated HT enhanced the formation of surface TiO2 layer without altering surface topography. Consequently, Ca-HT of Ti reduced PIE down-growth via tight epithelial attachment to the surface, which may enhance implant capability for a longer time post-implantation.

Resumen
Cell polarity is characterized by the asymmetric distribution of factors at the cell cortex and in the cytoplasm. Although mechanisms that establish cortical asymmetries have been characterized, less is known about how persistent cytoplasmic asymmetries are generated. During the asymmetric division of the Caenorhabditis elegans zygote, the PAR proteins orchestrate the segregation of the cytoplasmic RNA-binding proteins MEX-5/6 to the anterior cytoplasm and PIE-1, POS-1, and MEX-1 to the posterior cytoplasm. In this study, we find that MEX-5/6 control the segregation of GFP::PIE-1, GFP::POS-1, and GFP::MEX-1 by locally increasing their mobility in the anterior cytoplasm. Remarkably, PIE-1, POS-1, and MEX-1 form gradients with distinct strengths, which correlates with differences in their responsiveness to MEX-5/6. We show that MEX-5/6 act downstream of the polarity regulators PAR-1 and PAR-3 and in a concentration-dependent manner to increase the mobility of GFP::PIE-1. These findings suggest that the MEX-5/6 concentration gradients are directly coupled to the establishment of posterior-rich PIE-1, POS-1, and MEX-1 concentration gradients via the formation of anterior-fast, posterior-slow mobility gradients.

Resumen
This paper argues that the Caland system rests on a Pre-PIE verb-like adjective class, which formed root aorists. The Caland system as we know it came to be when PIE shifted to having a noun-like adjective class, and the Caland roots had to be adapted to the new system via derivation (while the old root aorists were gradually lost). Evidence for root aorists to Caland roots in Vedic is reviewed, and a typologically informed scenario for the shift is proposed. Finally, the paper argues that this scenario clarifies the origin of the *-eh1- stative in Indo-European (following Jasanoff (2002–2003)’s account), which would have arisen as PIE shifted from verb-like adjectives to nominal adjectives, and came to have a switch adjective system based on aspect.

Resumen
We studied contractile effects in isolated electrically driven (1 Hz) atrial preparations from patients undergoing cardiac bypass surgery. ATP concentration dependently (10, 30, and 100 μM) and rapidly decreased force of contraction (negative inotropic effect, NIE) and thereafter more slowly increased force of contraction. The maximum positive inotropic effect (PIE) at 100 μM ATP amounted to 152% of the predrug value ( n = 9) and was stable and could be washed out fast and completely. The PIE did not affect time parameters of contraction (time to peak tension and time of relaxation). Moreover, a similar NIE and PIE were noted with adenosine 5′- O-(2-thiotriphosphate) (100 μM). In contrast 2-methyl-thio-ATP did not exert a NIE but only a PIE. In a second set of experiments, preparations were first incubated for 30 min with purinoreceptor antagonists and, in their continuous presence, 100 μM ATP was applied. However, the PIE and NIE of ATP could neither be blocked with suramin (100 and 500 μM), pyridoxalphosphate-6-azophenyl-2′,4′-disulfonic acid (50 μM), nor reactive blue 2 (30, 100, and 500 μM), which are known blockers for subtypes of P2 receptors, or 1,3-dipropyl-cyclopentvl-xanthine (1 and 10 μM), a subtype (A1 adenosine) P1 receptor blocker. Likewise, the inhibitor of phospholipase C (PLC) activity (U-73122) and the inhibitor of adenylate cyclase activity (SQ-022563) (10 μM each) failed to affect the NIE and the PIE of ATP. We tentatively suggest that the PIE of ATP might be mediated via P2X4-like receptors. In summary, we describe a novel biphasic effect of ATP on force contraction in the isolated human atrium. It is conceivable that ATP plays a physiological role in the human heart, for instance, after cardiac injury to sustain contractility.

We consider the problem of off-policy evaluation (OPE) in reinforcement learning (RL), where the goal is to estimate the performance of an evaluation policy, pie, using a fixed dataset, D, collected by one or more policies that may be different from pie. Current OPE algorithms may produce poor OPE estimates under policy distribution shift i.e., when the probability of a particular state-action pair occurring under pie is very different from the probability of that same pair occurring in D. In this work, we propose to improve the accuracy of OPE estimators by projecting the high-dimensional state-space into a low-dimensional state-space using concepts from the state abstraction literature. Specifically, we consider marginalized importance sampling (MIS) OPE algorithms which compute state-action distribution correction ratios to produce their OPE estimate. In the original ground state-space, these ratios may have high variance which may lead to high variance OPE. However, we prove that in the lower-dimensional abstract state-space the ratios can have lower variance resulting in lower variance OPE. We then highlight the challenges that arise when estimating the abstract ratios from data, identify sufficient conditions to overcome these issues, and present a minimax optimization problem whose solution yields these abstract ratios. Finally, our empirical evaluation on difficult, high-dimensional state-space OPE tasks shows that the abstract ratios can make MIS OPE estimators achieve lower mean-squared error and more robust to hyperparameter tuning than the ground ratios.

Resumen
The aim of this study was to describe the generation of a PCV2 (porcine circovirus type 2) infectious clone (pIC-PCV2) and its infectivity under in vitro and in vivo conditions. The constructed pIC-PCV2 contained the whole PCV2 genome from a German isolate together with a partial duplication of 467 bp. PK-15 cells were transfected with pIC-PCV2 and an indirect immune fluorescence assay (IFA) was performed 7 days post-transfection. The PCV2 Cap gene was expressed in approximately 20 % of the cultured cells, and only the recombination product, and not pIC-PCV2, was subsequently detected by PCR and Southern blot. This result indicated that infection by pIC-PCV2 delivered genomic PCV2 DNA specifically into susceptible cells and led to the expression of a functional virus genome. Eighteen 30- to 40-day-old conventional pigs were distributed into three groups. Group 1 pigs (n=6) were inoculated intranasally (i.n.) with a Spanish isolate of PCV2 propagated in cell culture; pigs from group 2 (n=6) were inoculated with pIC-PCV2 intramuscularly (i.m.), and the last group of pigs (n=6) was inoculated with pIC-PCV2 intraperitoneally (i.p.). All pigs remained clinically healthy during the whole experimental period (35 days). Pigs that received pIC-PCV2 i.p. and i.m., as well as those PCV2 i.n. inoculated, became infected based on an in situ hybridization (ISH), PCR, TaqMan PCR and serological results. The results of this study confirm that cloned PCV2 genomic DNA is infectious both in vitro and in vivo, and is able to cause PMWS-like lesions in i.p. and i.m. experimentally inoculated pigs.

Resumen
A systematic study of the fragmentation pattern of N-diisopropyloxyphosphoryl (DIPP) dipeptide methyl esters in an electrospray ionization (ESI) tandem mass spectrometry (MS/MS) was presented. A combination of accurate mass measurement and tandem mass spectrometry had been used to characterize the major fragment ions observed in the ESI mass spectrum. It was found that the alkali metal ions acted as a fixed charge site and expelled the DIPP group after transferring a proton to the amide nitrogen. For all the N-phosphoryl dipeptide methyl esters, under the activation of a metal ion, the rearrangement product ion at m/z 163 was observed and confirmed to be the sodium adduct of phosphoric acid mono-isopropyl esters (PAIE), via a specific five-membered penta-coordinated phosphorus intermediate. However, no rearrangement ion was observed when a β-amino acid was at the N-terminal. This could be used to develop a novel method for differentiating isomeric compounds when either α- or β-amino acid are at the N-terminus of peptides. From the [M + Na]+ ESI-MS/MS spectra of N-phosphoryl dipeptide methyl esters (DIPP – Xaa1 – Xaa2 – OMe), the peaks corresponding to the [M + Na – Xaa1 – C3H6]+ were observed and explained. The [M + Na]+ ESI-MS/MS spectra of N-phosphoryl dipeptide methyl esters with Phe located in the C-terminal, such as DIPP–Val–Phe–OMe, DIPP–Leu–Phe–OMe, DIPP–Ile–Phe–OMe, DIPP–Ala–Phe–OMe and DIPP–Phe–Phe–OMe, had characteristic fragmentation. Two unusual gas-phase intramolecular rearrangement mechanisms were first proposed for this fragmentation. These rearrangements were not observed in dipeptide methyl ester analogs which did not contain the DIPP at the N-terminal, suggesting that this moiety was critical for the rearrangement.

Abstract: This paper describes the design, construction and working of an IoT based smart mirror. Every morning our day begins by watching ourselves at least once in mirror before leaving our homes. We interact with it psychologically to find out how we look and how our attire is. Smart Mirror or Magic Mirror is one of the applications of Raspberry Pie. A computer screen embedded in mirror looks very futuristic. The Raspberry Pie stays at back scenes and controls the data displayed on mirror. While looking at mirror you can look at various notifications from social sites as well news, weather forecast and more things. Such mirrors can be programmed to work as AI and control home appliances by voice input or touch screen. The Raspberry Pie is connected to monitor via HDMI as well as it also has inbuilt Wi-Fi and Bluetooth interfaces so we can just swipe music and videos to mirror.

Resumen
Coreopsis is a flowering plant belonging to the Asteraceae family. It is an ornamental plant native to the Americas, Asia and Oceania and its flower is used as a raw material for tea and food manufacture in China. In this study, new cultivars of C. rosea (“golden ring”) were developed via radiation-induced mutation of the original cultivar, “pumpkin pie”. The chemical composition and antioxidant activities of flowers belonging to three different Coreopsis cultivars were evaluated: “golden ring”, “pumpkin pie” and “snow chrysanthemum” (coreopsis tea; C. tinctoria). The volatile compounds were characterized via gas chromatography-mass spectrometry (GC-MS) and 50–59 oils representing 95.3–96.8% of the total volatile compounds in these flower materials were identified. ”Golden ring” contained a high amount of fatty acids (38.13%), while “pumpkin pie” and “snow chrysanthemum” teas were rich in aliphatic amides (43.01%) and esters (67.22%), respectively. The antioxidant activities of the volatile oils of these cultivars were evaluated using 1,1-diphenyl-2-picrylhydraxyl (DPPH) and 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging assays. The volatile extract of “golden ring” showed higher antioxidant activities compared with the extracts of the other cultivars. Therefore, “golden ring” can be used for further development as a raw material for tea manufacture or as a dietary supplement.

Resumen
Phosphodiesterase type-5 (PDE-5) inhibitors are novel and important options for the treatment of pulmonary arterial hypertension (PAH). Therefore, we aimed to examine effects of vardenafil, a PDE-5 inhibitor, on the pulmonary arteries isolated from rats with monocrotaline- (MCT-) induced pulmonary hypertension. MCT (60 mg/kg) or its vehicle was administered by a single intraperitoneal injection to 6-week-old male Sprague Dawley rats. Rats were sacrificed 21 days after MCT injection, and the main pulmonary arteries were isolated and then mounted in 20 mL organ baths. Concentration-response curves for vardenafil (10−10–10−5 M) were constructed in phenylephrine- (Phe-) precontracted rings. PAH caused marked rightward shift in the curves to vardenafil whereas maximal responses were not affected. Inhibition of NO synthase (L-NAME, 10−4 M) or guanylyl cyclase (ODQ, 10−5 M) caused similar attenuation in responses evoked by vardenafil. Moreover, contraction responses induced by CaCl2(3×10−5–3×10−2 M) were significantly reduced in concentration-dependent manner by vardenafil. In conclusion, vardenafil induced pulmonary vasodilatation via inhibition of extracellular calcium entry in addition to NO-cGMP pathway activation. These results provide evidence that impaired arterial relaxation in PAH can be prevented by vardenafil. Thus, vardenafil represents a valuable therapeutic approach in PAH besides other PDE-5 inhibitors.

Resumen
The generalization of neural networks is a central challenge in machine learning, especially concerning the performance under distributions that differ from training ones. Current methods, mainly based on the data-driven paradigm such as data augmentation, adversarial training, and noise injection, may encounter limited generalization due to model non-smoothness. In this paper, we propose to investigate generalization from a Partial Differential Equation (PDE) perspective, aiming to enhance it directly through the underlying function of neural networks, rather than focusing on adjusting input data. Specifically, we first establish the connection between neural network generalization and the smoothness of the solution to a specific PDE, namely transport equation. Building upon this, we propose a general framework that introduces adaptive distributional diffusion into transport equation to enhance the smoothness of its solution, thereby improving generalization. In the context of neural networks, we put this theoretical framework into practice as PDE+ (PDE with Adaptive Distributional Diffusion) which diffuses each sample into a distribution covering semantically similar inputs. This enables better coverage of potentially unobserved distributions in training, thus improving generalization beyond merely data-driven methods. The effectiveness of PDE+ is validated through extensive experimental settings, demonstrating its superior performance compared to state-of-the-art methods. Resumen
Expanding the usage of renewable energy in islanded microgrids leads to a reduction in its total inertia. Low inertia microgrids have difficulties in voltage and frequency control. That affected saving its stability and preventing a blackout. To improve low inertia islanded microgrids' dynamic response and save their stability, this paper presented relative rate observer self-tuning fuzzy PID (RROSTF-PID) based on virtual inertia control (VIC) for an islanded microgrid with a high renewable energy sources (RESs) contribution. RROSTF-PID based on VIC's success in showing remarkable improvement in the microgrid's dynamic response and enhancement of its stability. Moreover, it handles different contingency conditions successfully by giving the desired frequency support. Ant colony optimization (ACO) technique is used to find the optimal values of the RROSTF-PID based on VIC parameters. Furthermore, using MATLAB TM/Simulink, RROSTF-PID based on VIC response is compared to Optimal Fuzzy PID (OF-PID) based VIC, Fuzzy PID (F-PID) based VIC, PID-based VIC, conventional VIC responses, and the microgrid without VIC response under different operation conditions.

Resumen
Rice blast, caused by Magnaporthe oryzae, threatens rice production in most of the rice-growing areas in China, especially in regions that have grown Oryza sativa subsp. japonica in recent years. The use of resistance genes is the most effective and economical approach for blast control. In our study, a set of six near-isogenic lines (NIL) were developed by introgression of six resistance alleles of the Piz locus (Pi2, Pigm, Pi40, Pi9, Piz, and Pizt) into a blast-susceptible, high-yielding, high-quality japonica ‘07GY31’ via marker-assisted backcross breeding. Artificial inoculation using 144 M. oryzae isolates collected from the lower region of the Yangtze River, China, revealed that most of the NIL, including NIL-Pi2, NIL-Pigm, NIL-Pi40, NIL-Pi9, and NIL-Pizt, exhibited broad-spectrum resistance against rice blast at the seedling stage, with resistance frequencies (RF) of 93.06 to 98.61%. NIL-Piz was an exception, with an RF of 21.53%, which was slightly higher than the recurrent parent 07GY31. NIL-Pi40 and NIL-Pigm had broad-spectrum resistance (RF of 93.33 and 71.67%, respectively) at the heading stage following inoculation of 60 isolates of M. oryzae. Field trials with artificial inoculation at the seedling and heading stage showed that NIL-Pigm and NIL-Pi40 were highly resistant in four locations under high disease pressure. NIL-Pizt showed effective resistance in three locations from Zhejiang and Jiangsu Provinces. This study shows that O. sativa subsp. japonica alleles of the Piz locus confer resistance to M. oryzae, and provides an effective method to enhance seedling and panicle blast resistance in rice plants in the lower region of the Yangtze River, China.

Resumen
Abstract Cross-linking of IgE or a bacterial product (f-Met-Leu-Phe; FMLP) induces the release of leukotriene C4 (LTC4) and histamine in human basophils. However, the signaling mechanisms in human basophils are only partially understood. It has been demonstrated that extracellular signal-regulated kinases (ERK1/2) specifically regulate the pathway for LTC4 generation, but not for histamine release and interleukin-4 production. More recent studies have suggested that tyrosine kinase (syk)-mediated phosphorylation of shc is responsible for the ras-ERK cascade via the formation of shc-Grb2-Sos2 following stimulation with anti-IgE antibody, but not FMLP, in human basophils. However, while characterizing the role of phosphatidylinositol (PI)-3 kinase in signaling pathways leading to basophil mediator release, it was noted that this pathway might also regulate p21ras activation. Anti-IgE antibody, but not FMLP, resulted in phosphorylation of p85 (regulatory subunit of PI3 kinase), suggesting activation of PI3 kinase. Inhibition of PI3 kinase by selective inhibitor (LY294002) abolished anti-IgE antibody- but not FMLP-induced phosphorylation of MEK1 (MAPK kinase/ERK kinase) and ERKs while inhibiting LTC4 generation as well as histamine release. IgE-mediated activation of ras (upstream of MEK-ERK) was also inhibited. But, further upstream, phosphorylation of syk and of shc and inducible association between shc and Grb2 were not affected. Furthermore, the IgE-mediated cytosolic calcium response ([Ca++]i) was also diminished. These results suggest that functional responses may be dependent on the activity of PI3 kinase, which regulates at least 2 important signaling pathways: by regulating activation of ras for the MEK-ERK pathway and the increase in [Ca++]i.
