Medina, Molano Natalia Stefania. "Contribution of radioactivity to catalytic performance in heterogeneous media". Electronic Thesis or Diss., Université de Lille (2022-....), 2023. http://www.theses.fr/2023ULILR025.
This thesis aimed to identify applications of noble metals recovered from spent nuclear fuels such as 107Pd. To study the effect of the radiation three different applications were selected: heterogeneous catalysis, with the hydrogenation of cinnamaldehyde (CNA) as model reaction; photocatalysis, with the decolourisation of methyl orange (MO) as model reaction; and the regeneration of catalysts, deactivated with the synthesis of hydroxymethyl furfural. Protocols were developed for the synthesis of the catalysts and the performance testing in restricted nuclear environment, namely in glove box and for the implementation of the reactions in glove box and extractor column as appropriate.Regarding the results it was found that β-radiation did not show any effect on the hydrogenation of CNA independent of the applied reaction conditions. On the other hand, the decolourisation of MO was successfully activated by the β-radiation of the catalyst. A decrease in absorbance (at the isosbestic point) was observed and was correlated to the amount of the catalyst employed, evidencing the effect of the irradiation doses. Finally, the regeneration of the Pd-based catalyst showed promising results after γ-irradiation, notably with a partial recovery of the initial catalytic performance after -irradiation. Meanwhile, the Ru-based catalyst, although generally not performing well, showed increased activity compared to the fresh catalyst upon -irradiation

Study of the relationship between the clinical radiosensitivity of a patient and his lymphocytes irradiated in vitro in order to establish a predictive test of individual radiosensitivity. Clinical radiosensitivity was quantified by the intensity of radiotherapy side effects and cellular sensitivity was measured by the kinetics of appearance/disappearance of radiation-induced DNA double-strand breaks (marked by gamma-H2AX foci). To make the protocol viable for clinical application, sample viability and analysis speed were improved. Finally, we focused on low-dose response in another context: radio-adaptive response. This phenomenon is characterized by a weaker cellular response to a high dose exposure when it is preceded by a low-dose exposure. This type of response was observed for the two-way translocation rate in CD4-positive lymphocytes but not for gamma-H2AX signaling

Using cell lines representative of hemopoietic differentiation, we showed that immature hemopoietic CD34+ cells are less sensitive to ionizing radiation than mature CD34- cells. Ln immature cells, ionizing radiation activate apoptotic cell death related to neutra! sphingomyelinase stimulation responsible for nuclear sphingomyelin hydrolysis and nuclear ceramide generation. Ln CD34+ cells, ionizing radiation do not produce neither ceramide nor apoptosis, and activate delayed reproductive cell death (mitotic cell death). These results suggest that nuclear neutra! sphingomyelinase plays a pivotai role in the cellular response of ionizing radiation. Ln a second part, we tried to determine the mecanisms responsible for negative regulation of nuclear sphingomyelinase in CD34+ cells. We first considered nuclear proteolytic events. Thus, we provided evidences for the presence of the serine-protease, Granzyme B, in the nucleus of CD34+ cells. We showed that ionizing radiation up-regulates Granzyme B expression both in the nucleus and the cytoplasm of these cells. This overexpression is not responsible for the lack of apoptotic response of CD34+ cells to ionizing radiation. Lnterestingly, we observed that Granzyme B overexpression confers a potent cytotoxic ability to these cells towards target cell lines of myeloid and lymphoid origin. Our results suggest that when irradiated, CD34+ cells acquire cytotoxic potential toward cells of the medullar environment. Conversely, in cytotoxic T lymphocytes, ionizing radiation triggers down-regulation of Granzyme B expression correlated with the loss of cytotoxic function. These results suggest immunosuppressor effects of ionizing radiation, related to their capacity to modulate Granzyme B expression

TMI-Orion, a company specialising in systems for acquiring physical parameters (pressure, temperature, humidity, etc.) for harsh environments, wishes to diversify, particularly in the civilian nuclear markets or the new space. These two markets are subject to strong radiative stress. The company, which is at the origin of the thesis, wishes to develop its skills with regard to radiative stress in order to be able to propose applications in these sectors. This thesis was therefore conducted with the main objective of providing as much information as possible on radiative stress to TMI-Orion. We have therefore identified four areas of work to address this issue. In the context of this thesis, we began by describing the radiative environments encountered in the space and civilian nuclear fields. Then the mechanisms of radiation-matter interaction were defined. Finally, the effects of radiation on electronics have been given. The second area of work consists of a study of the degradation mechanisms of elementary electronic components (MOS and bipolar transistors) as a function of the temperature to which they are subjected during irradiation. The effect of low (up to 100 K) and high (up to 400 K) irradiation temperatures on the degradation of elementary components was studied. During this study we were also able to compare the degradation obtained between a cobalt-60 irradiation and an X-ray irradiation. The third area of work consists of a simulation study on shielding against radiative stress. As part of this work, we have developed two calculation codes on GEANT4. The first one allows to study the dose deposited by a cobalt-60 source for different shielding conditions. The second one allows to model the X-ray generator of the IES laboratory and gives its energy spectrum. The effect of different filters on the spectrum is also studied. The last axis of this thesis work consists of a complete reliability study on autonomous data loggers developed by TMI-Orion. One particular recorder, the PicoVACQ, is used as an example. The reliability of this recorder was calculated by an analytical method, studied using the feedback available within the company, measured against thermal stress and measured against radiative stress. Finally, the search for a microcontroller, a central component of a radiation-tolerant data acquisition system, was carried out. One microcontroller in particular appears to have good resistance to dose effect and will probably be used by the company in its future projects

En anglais : Nanocomposites fabricated with ion-based techniques have a number of attractive characteristics. However, the main and most crucial diculty in obtaining commercial NPs-based devices is the inability to produce a suitable narrow size and spatial NP distributions. The objective of this thesis is twofold: i) to go further in the description of the behavior of the ion-driven NPs and ii) to overcome the limitations related to the ion-beam techniques providing a guideline methodology to rationalize the synthesis of NPs when ion-beams are used. Thus, a model system is fabricated. It consists of chemically synthesized metallic nanoparticles sandwiched between two silica layers. We show how the ion irradiation and the temperature can be used to tune the size distribution of the embedded NPs. Moreover, we show that when an initially large NPs size distribution is considered, the study of the growth kinetic of the NPs under irradiation can be problematic. Our model system is than used to investigate in detail the behavior of the NPs under irradiation. We show that the evolution of the precipitate phase under irradiation is successfully described by an Ostwald ripening mechanism in an open system limited by the diusion. Moreover, the concentration threshold for nucleation as well as the surface tension and the gold diusivity in silica under irradiation is estimated. Finally, direct and inverse Ostwald ripening processes under irradiation are systematically investigated and the existing theoretical models experimentally checked

Various studies were carried out on the ageing of epoxy resins under irradiations. In all cases, pure polymers were studied. The aim of our work managed by the CEA and the CNRS consists on studying the part of fillers and particularly the part of silica on ageing process under electron beam irradiations. Because of their wide use in industrial applications and especially in nuclear environment, the DGEBA-TETA resins (DiglycidylEther of Bisphenol A - TriEthyleneTetraAmine) were chosen. Those epoxy resins are difficult to analyse because of their insolubility. Some pure and nanometric silica filled chemical models which chemical structure very close to the one the DGEBA/TETA resin were synthesized and analysed with classical methods in organic chemistry. A major phenomenon of rupture of the C-O and C-N chemical bonds with creation of phenolic extremities, methylketon extremities, of primary and tertiary amines and notably enamine functions were revealed by the analyses. The quantitative 1H and 13C NMR analyses revealed the screen effect due to the silica and the reactions between the chemical species created by the irradiations and the silica surface. Thermic and thermodynamic analyses of the different epoxy resins in function of the irradiation dose and of the kind of silica showed the decrease of the glass transition temperature, of the relaxation temperature and of the crosslink density confirming the major phenomenon of bond ruptures during irradiations. With silica, the decrease of the crosslink density is slowed. This phenomenon can be explained with interactions between the nanometric silica surface and the epoxy resin offsetting the effect of the chain rupture on the resin mechanical properties. The 13C solid state NMR analyses confirmed the choice of the chemical models and permitted to detect the chemical species created by the irradiations. The analyse of the polarization transfers with 13C CP-MAS NMR spectroscopy revealed the rigidification of the nanometric silica filled resin and the reactions between the resin and the silica surface during irradiations. This study permits to assert that the silica fillers in the epoxy resins protect the network from the irradiations

This work falls under a research program which aims to synthesize new organic and organometallic compounds having antioxidant activity and/or an interesting radioprotective or radiosensitive activity without toxicity. That is why we carried out the synthesis of new derivatives of pulvinic acid, salicylic acid, naphtyl-methyl-imidazoline, benzothiazole, thiadiazole and naphtalimides. These compounds were characterized by NMR (1H, 13C, 31P), by mass spectrometry, elementary analyzes and for some of them by X-rays diffraction. The activity of the majority of them was evaluated by in vitro and in vivo tests in collaboration with the “CEA” (Saclay), the university of Strasbourg (ULP) and the “Centre de Recherche du Service de Santé des Armées” of La Tronche (CRSSA). The experimental results show a structure/activity relationship between pulvinic acid and antioxidant properties. The modifications of biological properties due to organometallic ligands have also been developed. Moreover, new classes of antioxidants, radioprotective and radiosensitive agents have been defined

In a radiological examination, low-energy X-radiation is used (<100 keV). For other radiological procedures, the energy used is several MeV. ICRP in publication 103 has currently considered that photons irrespective of their energy have the same radiation weighting factor. Nevertheless, there are topological differences at the nanoscale of X-ray energy deposition as a function of its energy spectrum, meaning that the different interactions with living matter could vary in biological efficacy. To study these differences, we characterized our irradiation conditions in terms of initial photon energies, but especially in terms of energy spectra of secondary electrons at the cell nucleus level, using Monte Carlo simulations. We evaluated signaling of DNA damage by monitoring a large number of γH2A.X foci after exposure of G0/G1-phase synchronized human primary endothelial cells at a dose from 0.25 to 5 Gy at 40 kV, 220 kV and 4 MV X-rays. Number and spatial distribution of γH2A.X foci were explored. In parallel, we investigated cell behavior through cell death and ability of a mother cell to produce two daughter cells. We also studied the missegregation rate after cell division. We report a higher number of DNA double-strand breaks signaled by γH2A.X for 40 kVp and/or 220 kVp compared to 4 MVp for the highest tested doses of 2 and 5 Gy. We observed no difference between the biological endpoint studies with 40 kVp and 220 kVp X-ray spectra. This lack of difference could be explained by the relative similarity of the calculated energy spectra of secondary electrons at the cell monolayer. The energy spectrum of secondary electrons seems to be more closely related to the level of DNA damage measured by γH2A.X than the initial spectrum of photon energy or voltage settings. Our results indicate that as the energy spectrum of secondary electrons increases, the DNA damage signaled by γH2A.X decreases and this effect is observable beyond 220 kVp

Modulation of ceramide's metabolism in the response of leukaemic cells to ionising radiation. Apoptosis induced by antitumoral drug or radiotherapy leads to an increase in the intracellular concentration of the apoptotic second messenger ceramide (Cer) increases. Cer production occurs either through the stimulation of its de novo synthesis via Ceramide synthase, or through sphingomyelinase-induced sphingomyelin degradation. .
