Copolyester of -caprolactone and -valerolactone with controlled molar mass and composition are synthesized by ring opening polymerization with stannous octanoate and octanol. Crystallinity of copolyester are measured. The evolution of water uptake, hydrophobicity properties and hydrolytic degradation of the binders surfaces have been investigated during the immersion in controlled medium. Copolymers are incorporated in an antifouling paint formulation and then immersed in natural conditions. Efficiency was validated during eight months of time immersion in the Kernevel harbour

Texto completoBiofouling induces important economic and ecological problems. This phenomenon includes microorganisms (bacteria, microalgae…) giving the microfouling which allows the macrofouling development with algae, invertebrates and sponges. These organisms colonize every immersed surface as boat hull. The colonization induces reduced speed of ships and fuel overconsumption. In the past, the utilization of AF coatings with heavy metals or pesticides caused environmental problems. The purpose of the study is to find an alternative to AF compounds (copper) respecting the marine medium. The marine environment is an inspiration; a biomimetic approach could be an interesting strategy to inhibit biofouling. In a first part, a screening of one hundred compounds is realized against marine and terrestrial bacteria. Eight molecules from two families (batatasins and hemibastadins) are studied to understand the way of action. Among selected compounds, Dibromohemibastadin-1 (DBHB) from hemibastadin family shows promising AF activities. This molecule is able to inhibit the biofilm formation with an IC50 of 6,44 µg/mL against the bacterium Pseudomonas aeruginosa PAO1 and 12,8 µg/mL for the marine bacterium Paracoccus sp. 4M6. To identify the way of action of DBHB, the impact on the bacterial communication named quorum sensing is investigated. The molecule shows an anti-quorum sensing property. In a second part, another group participating at microfouling is studied: microalgae. In order to evaluate the impact of hemibastadin family in particular DBHB, microalgae adhesion and biofilm formation are characterized. DBHB induces inhibition only on the adhesion and the biofilm formation of the diatom Cylindrotheca closterium. The last part presents the formulation of coatings containing six compounds from the batatasin and hemibastadin families. These coatings have been immersed in a harbor to evaluate the impact on natural microfouling. Furthermore, a new method for the evaluation of AF coatings is developed in controlled conditions, in a photobioreactor. This method allows the evaluation of coatings on the formation of a mixed biofilm (bacteria and microalgae). This method has been established to avoid environmental constraints by immersion in natural condition. This study allows the characterization of the AF property of DBHB. This compound provides promising research path to limit biofouling. Moreover, the development of a test allowing adhesion and microalgae biofilm formation in dynamic condition improves the characterization of compounds activities

Texto completoThe development of biofilms causes serious problems in the marine and medical fields. Their high tolerance to commonly used chemical agents’ disinfectants, antibiotics, biocides) makes their eradication difficult. Moreover, the use of biocide molecules is widely controversial, considering their catastrophic environmental impact. Research has therefore focused on systems that, by their composition, limit biocontamination. Among them are amphiphilic systems that can be composed of a hydrophobic polydimethylsiloxane (PDMS) matrix and an amphiphilic PDMS-PEG copolymer. Despite their efficiency, these systems are questioned because of the petrochemical origin of PDMS. The objective of this thesis project is to substitute PDMS with a biopolymer, poly(hydroxyalkanoate) (PHA). A system was formulated with PHA as a hydrophobic matrix and a PHA-PEG copolymer as an amphiphilic additive. Two types of PHA were used in this study, PHBHV (short chain length) and PHAmcl (medium chain length). The formulated coatings were characterized physically, chemically and mechanically. Then their anti-adhesive, anti-biofilm and fouling-release capacities were evaluated on different microorganisms. Two opportunistic pathogenic bacteria, Staphylococcus aureus and Pseudomonas aeruginosa, a marine bacterium, Bacillus 4J6 and two benthic diatoms, Phaeodactylum tricornutum and Navicula perminuta. Texto completoMarine biofouling represents the undesirable accumulation of biological organisms on the surfaces of structures submerged in seawater. Unfortunately, this natural phenomenon has serious economic, environmental and material consequences. Since the ban of some biocides in antifouling paints (TBT in January 2008) because of their toxicity on the nontargeted marine species and their accumulation in the marine environment, research has focused on the development of new efficient, durable and environmentally friendly antifouling coatings without releasing toxic species. Thus, the work of this thesis deal with the functionalization of glassy carbon surface by carbohydrates linked to an electrostimulable conjugated system via a triazole link in order to develop surfaces with antifouling activity. Indeed, this kind of coating was designed to intervene in the first steps of biofouling. First, the carbohydrate, which is very hydrophilic, should fight against the formation of the conditioning film by surronding itself with an aqueous barrier resistant to proteins. On the other hand, the continuous modification of charge state by applying an electric current to the electroactive conjugated system is expected to disrupt the bacterial colonization delaying the installation of marine biofilm. Our study is therefore based on the synthesis and the immobilization of electrostimulable carbohydrates on a glassy carbon surface by aromatic amine oxidation in organic and aqueous media. A microbiological test was carried out on one of the carbohydrate coatings in the presence of the TC8 bacterial strain in the wells of a microplate containing electrochemical cells connected to a potentiostat. Electrical stimulation of this coating allowed to improve its antibacterial properties
