Texto completoTo enter poems through the concept of voice is to engage in a poetics of listening intently to bodies: the bodies which write, speak aloud, perform and translate, as well as the bodies of the reader and the spectator. When we undertake to deconstruct the metaphor of the voice and observe its forms of passage, we discover the decisive links that exist between language as a physical force and poetics as ethics and politics. The concept of the passage of voice gives rise to critical levers which involve other deconstructions, thereby changing the way we think about many dualisms. These latter usually prevent us from perceiving the relational activity of language, foremost among which is the opposition between oral and written language. To consider voices not as signatures – as stylistics or literary history often do – but as processes is to listen to the rhythms of subjectivations whose energy unfolds from the enunciative life of language, with their transformative or even emancipatory value. This implies discoveries about our conception of places and temporality, not as thematic or ontological categories, but as anthropological ones.This thesis explores three main forms of voice passages and sets out their issues and effects: movements of diffraction, spatialisation and presentation. To do so, it questions theoretical texts which focus on investigating the conceptual issues of voice, rhythm and subject (Meschonnic, Martin, Rabaté, Dessons, Bernadet), based on a main corpus (in the form of texts, performances and videos) made up of the protean works of the contemporary French poets Stéphane Bouquet, Christophe Manon and Frank Smith

Texto completoOral administration still remains the route of choice for the majority of pharmaceutical dosage forms due to higher patient comfort and reduced cost of treatment. One of the major drawbacks of using the oral route to deliver drugs is that these molecules need to cross biological membranes in order to pass to the bloodstream and reach their physiological target. A large number of drugs with high clinical potential have not yet been employed because of their limited bloodstream access. Therefore, there is a real need to develop new methodologies addressing the problem of low permeable drugs used in oral administration. In this work we studied retention using an external magnetic field as a mean to maintain magnetic carriers near the absorption site of the drug. It was proven that an effective retention can cause over-concentration of the drug near the intestinal membrane thus increasing its bioavailability. For this purpose, we designed magnetic carriers, first, at the lab scale, and then using industrial processes. Magnetic retention efficiency was subsequently studied using ex vivo and in vivo experiments. Pharmacokinetics studies coupled with imaging techniques showed that magnetic retention provided a threefold increase in drug bioavailability. Finally, it was shown that retention of magnetic carriers near the absorption window of the drug can lead to overconcentration of the drug and increased transport through intestinal membrane resulting in a higher absorbed fraction reaching the bloodstream

Texto completoMany nanocarriers have been developed to improve the delivery of molecules into the skin. In this PhD thesis, we are interested in lipid-based Janus nanoparticles (JNP), an innovative galenic form characterized by the combination of two compartments of opposite chemical polarity, an aqueous compartment associated to a lipid compartment. The main aim was the characterization of JNP. ATR-FTIR spectroscopy allowed to identify an infrared descriptor to follow the physical stability of JNP in open air and over time. The same descriptor allowed to follow their behavior on the surface of the skin, and to note a significant penetration from 3 hours of application. AFM-IR has been shown to be a promising technique for studying the nanostructure of the human skin. In addition, it has shown that after 24 hours of application, JNP were accumulated in the first layers of the SC with a gradient in the deeper layers of the SC. However, it was not possible to conclude if they have penetrated in the intact or degraded form. JNP seem to have an influence on the cutaneous penetration of the hyaluronic acid, they allowed a significant increase of its penetration flux. The characterization of the lipophilic phase of JNP by different techniques (LC-MS, DLS, Cryo-TEM, X-ray diffraction...) allowed to better understand their instability at high temperatures (32°C - 43°C)

Texto completoAs a physiologic nitric oxide (NO) donor, S-nitrosoglutathione (GSNO) has potential therapeutic application for the treatment of cardiovascular disease (CVD). With a longer in vivo half-life than NO, GSNO is still sensitive to many factors leading to poor applicability. This study aimed at the development of nanocomposite particles (NCP) based on synthetic polymeric nanoparticles encapsulating GSNO (GSNO-NP) embedded in a polysaccharidic matrix for oral delivery of GSNO. Although GSNO-NP, with a high encapsulation efficiency, showed an in vitro burst release, they succeeded in the preservation of GSNO stability and bioavailability for smooth muscle cells as they delayed in vitro protein S-nitrosation (NO biomarker) until 18 h. Therefore, to reach the sustained release, GSNO-NP were embedded in a matrix of alginate (a), chitosan (c) or a blend (acNCP). GSNO-acNCP with high encapsulation efficiency (76%) and an in vitro release until 24 h, promoted the highest permeation rate of GSNO through an intestinal barrier model (Caco-2). With this daily oral treatment compatibility Wistar Rat pretreatment by gavage with GSNO-acNCP 17 h before aorta removal decreased the maximal contractile effect induced by phenylephrine (PHE) on isolated aortic rings. Furthermore, the N-acetylcysteine (a thiol displacing NO stores from tissues) produced the relaxation of PHE precontracted aortic rings, proving NO storage in the vessel wall. By increasing the residence time in the gastrointestinal tract thus promoting GSNO crossing through the intestinal barrier, GSNO-acNCP induced a long lasting effect (17 h after administration) through NO storage in vessels

Texto completoIn E. M. Forster’s novels the representation of the body reveals an ambiguous view of the world and a disillusioned concept of literature. At first reading there appears to be no trace of the human body in Forster’s works, which focus on silence and the invisible, and prefer to suggest rather than describe. In A Room with a View, Howards End and Maurice, the event is commented upon without really being narrated, and the reader has to read between the lines. Here the eye and the voice point up a process of secret and retention in the text. The three novels then open out to write the senses and display an idealist body, that can be seen and touched in its immediacy, and is both the object and origin of a voice linking vision and visibility in a necessary, almost phenomenological, complementarity. The Forsterian novel would thus favour representation in space over chronological narration, showing some form of aesthetic modernity which is actually confirmed in the three film adaptations by James Ivory through their insistence on the eye and the voice, and the way they call into question narration on screen. Although the films first seem to silence the Forsterian voice, they shed new light on the novels. Forster’s last choices in A Passage to India invalidate these dynamics as the writer eventually renounces the body and writing itself, both equally unsuited to show or tell the world

Texto completoThe pulmonary route is of increasing interest for the systemic administration of therapeutic proteins and peptides, still largely administered parenterally. A microdevice was designed to facilitate and accelerate the in vitro screening studies of various active biomolecules and to select the most suitable formulations for penetration through the lung epithelium, in order to select the best candidates for an administration via the lungs. Organized in two distinct configurations, this microdevice allows as a first step the culture of tight polarized bronchial epithelial barriers (Calu-3 cells) in 7 days in 1 mm² microwells, without the need for medium renewal or the use of an external apparatus. A simple manufacturing technique was developed and glass culture platforms containing 12 parallel microwells can be obtained in a standardized manner. The ability of molecules to cross the pulmonary barrier is then performed in the second configuration of the microdevice, which is dedicated to the permeability measurement of the tight epithelial Calu-3 barriers cultured in microwells. Among the different candidates studied (nanoparticules and biomolecules), the pulmonary barrier permeability regarding PLGA nanoparticules coated with chitosan and regarding insulin has been successfully demonstrated. Finally, capillary electrophoresis with laser induced-fluorescence (CE-LIF), a technique compatible with the low volumes handled in this microdevice, has been exploited for insulin detection and quantification after its transport across the miniaturized pulmonary barriers. To this end, insulin was either FITC-labeled or complexed with a fluorescent antibody or aptamer. Currently, only the derivatization method can be used for a quantification purpose, but the use of an aptamer to indirectly quantitate insulin has shown encouraging results

Texto completoFrom our clinical experience with prison inmates, our research work proposes a theoretical paradigm of the concept of acting-out in relation with the superego instance. The purpose is thus to bring out the metapsychological stakes and issues of the superego concept as they appear in the clinical approach of the acting-out, and more specifically in relation with the criminal act, while tracing back the conditions of its theoretical elaboration. Even if the foundations that Freud laid is essential to our reflection, the kleinian concepts of primitive superego and of paranoid-schizoid position will be far more precious to us in this endeavour to reflect on the reappearance of certain modes of functioning which are under the supremacy of a particularly cruel and destructive superego. This aspect, which Lacan has reformulated with his view of an imperative superego, will provide the basis for our discourse. While the superego instance is first identified through its function of moral consciousness, as a legislating instance, there also exists a more paradoxical double polarity whereby the reverse side of the Law is expressed through a ferocious and cruel injunction to have enjoyment (“jouissance”). Within this perspective, the criminal acting-out appears like the irrepressible response to the injunction of a strong Superego pushing to commit harsh and virulent actions. This thesis notably insists on the criminal act seen as a last resort. A resort which aims at releasing oneself from the position where one has become the object of the Other's “jouissance”, while one tries, at the same time, to be a subject

Texto completoThe invocative drive goes through the work of Jacques Lacan, although he never really developed this theme: its object is the voice, at the very moment when it is lacking: our hypothesis is that the subject passes to the act when the voice is silenced, there is no cogito. Intricate to the scopic drive, the invocative drive has the silence as a signature. The passage to the act proceeds from a "I do not think", from language and not from affects that the subject would suffer. This thesis considers the dimension of real of the father in the passage to the act, in a psychotic decompensation case, in his relation to the invocative drive. Laws have been established to differentiate the actions of the designated as crazy, psychotic subjects from those of the poor, weak and criminals, which specifies the framework of mental expertise that is not foreign to our research. The texts of the laws of 1838 and 1990 were modified not so much to ensure a better protection of the patients as for security reasons. Thus, the various modes of hospitalization were revised: free hospitalization, hospitalization at the request of a third party, duty of hospitalization ... until the recent concept of duty of care. Our thesis begins by revisiting these historical aspects to situate the context of passage to the act which will then be analyzed in the light of Lacan's teaching. We envision the voice as a tentative to substitute for the foreclosure of the Name of the Father, the voice supplies and completes the silence of the Other. To do this, we develop several clinical illustrations. In the case of Juan, we show how the therapeutic work allowed him not to go into silence and look for his name by addressing to us. The work of speech prevents the passage to the act. Duras' novel, Summer Rain, allows us to hear the invocative drive as the echo of the voice of the Other and its bodily resonance. With Seth, the voice of the father and the devil are confused, a pact of alliance between the terrible interdicting fathers and the female figures. With Mrs. X, we are getting closer to the aspects of Lacan's formula: in psychosis, the gaze sounds. We can then take inconsideration what happened to Chris who cannot stand the voice. The question of the reverse side of self-punishment is discussed with the patient Medea. We then rely on Gabriel Garcia Marquez with a chronic of a deadly attempt announced. Finally, we try to understand the outbreak of psychosis in Telfusa, whom the voices according to her constantly accompany her, even when she speaks. The treatment will be like this: no to Juana the Crazy but yes to freedom. This thesis attempts to highlight the proper value of the word introduced by psychoanalysis, by illuminating the clinical practice of the passage to the act in the field of psychosis
La pulsion invocante atraviesa la obra de Jacques Lacan, aunque él nunca desarrolló realmente este tema: su objeto es la voz, en el momento mismo en que ella falta: nuestra hipótesis es que el sujeto pasa al acto cuando la voz se calla, no hay cogito. Intrincada a la pulsión escópica, la pulsión invocante tiene como firma, el silencio. El pasaje al acto procede de un "yo no ienso", del lenguaje, y no de un afecto del cual sufriría el sujeto. Esta tesis considera la dimensión real del padre en el pasaje al acto, en caso de descompensación psicótica, en su relación con la pulsión invocante. Se han establecido leyes para diferenciar los actos de los sujetos designados como locos, psicóticos de los actos de los pobres, débiles y criminales lo que especifica el marco de peritaje psicológico que no es ajeno a nuestra investigación. Los textos de las leyes de 1838 y 1990 se modificaron no tanto para garantizar una mejor protección de los pacientes como por razones de seguridad. Por lo tanto, se revisaron los diversos modos de hospitalización: hospitalización libre, hospitalización por pedido de un tercero (HDT), hospitalización de oficio (HO) ... hasta el concepto reciente de obligación de tratamiento. Nuestra tesis comienza revisando estos aspectos históricos para ubicar el contexto de los pasajes del acto, que luego será analizado a la luz de la enseñanza de Lacan. Vislumbramos la voz como un intento de sustitución a la forclución del Nombre del Padre, la voz que viene a suplir y completar el silencio del Otro. Para hacer esto, desarrollamos varias ilustraciones clínicas. En el caso de Juan, mostramos cómo el trabajo terapéutico le permitió no quedarse en el silencio y buscar su nombre dirigiéndose a nosotros. El trabajo de la palabra impide el paso al acto. La novela de Duras, lluvia de verano, nos permite entender la pulsión invocante como el eco de la voz del Otro y su resonancia corporal. Con Seth, la voz del padre y el diablo se confunden, un pacto de alianza entre los padres terribles que prohíben y las figuras femeninas. Con la Sra. X, nos estamos acercando a los aspectos de la fórmula de Lacan: en la psicosis, la voz sonoriza la mirada. Podemos enseguida tomar en consideración lo que le sucede a Chris que no soporta la voz. La cuestión del reverso del autocastigo la abordamos con la paciente Medea. Nos apoyamos en Gabriel García Márquez con crónica de un intento de muerte anunciado. Finalmente, intentamos de comprender el desencadenamiento de la psicosis en Telfusa, a quien las voces, según ella, la acompañan constantemente, incluso cuando habla. El tratamiento se presenta asi: no a Juana la loca, y si a la libertad. Esta tesis trata de poner en evidencia el valor propio de la palabra introducida por el psicoanálisis, esclareciendo la clínica del acto en el campo de las psicosis

Texto completoThis thesis is an analysis of the trilogy of ʼAḥlām Mustaġānimī, one of the first Algerian authors in Arabic with formal analysis and the works of Genette.The introduction discusses the methodology and the issue. How are the novels woven into a trilogy and why have they been so popular with the Arabic-speaking readership? Which leads to another: how Algerian history is included in the trilogy. The first part shows that these novels are a reflection of Mustaġānimī's life and his commitment, through the dedications and the Arabic language of which generations have been deprived. The second part uses the tools of the formal approach to determine the mechanism of suspense by disassembling the narrative text into the constituent elements of the main story, the love story, and its interruptions. It shows ʼAḥlām Mustaġānimī's great sensitivity to the place of women and to Algerian politics before and after independence. The last part, with intertextualities and linguistic styles, analyzes the text, and the poetry of its writing. This mastery of the Arabic language by Mustaġānimī is one of the causes of his success in the Arabic-speaking readership. This thesis shows the interest of these methods of analysis, explaining and objectifying the feelings of the reader. Finally, Mustaġānimī finds himself in the implicit protagonist of the trilogy, which is therefore an “intrusion” into the life of the writer

Ethnolinguistic fieldwork was carried out among the Yanesha’ people in May-August 2008 to learn about their language and the possible factors leading to its endangerment. This thesis examines the unique linguistic features of Yanesha and its place within the Arawak language family. It also discusses a puberty ritual that plays an important role in preserving musical and linguistic heritage among Yanesha’ women: the ponapnora female initiation ritual.
