With the new, state-of-the-art printing devices and equipment, there has been rapid growth in the counterfeiting of banknotes. Traditional security features on banknotes are easy targets for counterfeiters, and they can easily imitate the original banknotes with fake ones. Conventional methods for validating currency require specialized devices for the authentication of banknotes. However, cost and lack of mobility of sophisticated banknote validation devices are big problems for general consumers. Modern digital solutions are attempting to complement the traditional security features through embedding radio frequency identification (RFID) chips in the banknotes, for example, Euro currency. Unfortunately, the requirement of specialized RFID readers for banknote validation impedes their widespread proliferation among consumers. To overcome this problem, a new method of banknote validation using an RFID chip and an NFC-enabled smartphone is presented. The consumer sends a banknote validation request to the Monetary Agency (MA) using her or his smartphone and an Internet connection. TheMAreplies by sending a random challenge to the consumer’s smartphone. The RFID chip in the banknote receives the challenge, via the NFC, and calculates an equivalent response to theMA’s challenge. If any of the messages are incorrect, authentication is denied. By the proposed method, consumers can easily and instantly check the originality of currency notes with theMAusing their smartphones and an Internet connection. The proposed system is less expensive, computationally, than regular methods and preserves the privacy of people who carry banknotes.

To control pricing in passenger transportation, monitor the functioning of urban transport networks, reduce direct contacts during the COVID-19 pandemic, it is advisable to implement a system of NFC validation of fares. NFC (Near-Field Communication) technology provides primary information on public transport and travel fares. The article aims to improve the method of application of the technology of automated NFC-validation of fares in urban passenger transport to account for and control provided transport services. The tasks include research of mechanisms of the automated identification and counting of quantity of the transported passengers to account incomes of the motor transport enterprises; selection of calculation units for automated determination of the cost of provided transport services; development of control methods of drivers work, the operation of vehicles, transportation of privileged categories of citizens, pricing, etc. Theoretical and methodological aspects of NFC-validation technologies for public transport fares for accounting purposes have been studied based on general research methods – institutional and innovative; economic and mathematical methods of analysis and polynomial trend using Excel spreadsheets were used to predict the pace of implementation of NFC-devices; methods of bibliographic and comparative analysis using the information resource «ResearchGate»– to determine promising areas for new technologies in the provision of transport and passenger services. It is proved that the most effective unit of cost calculation of transport services is «passenger-kilometer», which corresponds to the peculiarities of the functioning of urban passenger transport, as it takes into account the number of passengers and distance of travel. The procedure for distribution of income from the sale of electronic tickets and budget subsidies between carriers based on information on the total number (including privileged categories) of transported passengers has been developed. The method of automated cost accounting by the «passenger-kilometer» meter for fuels and lubricants, staff salaries, depreciation of vehicles, and its current repairs, which define the cost of transport services, is improved. Implementing NFC fare validation technology will facilitate automated accounting of costs, revenues, and cash receipts from transport services, the introduction of effective control over the work of drivers, vehicles, concessional transportation, pricing, timeliness, and reliability taxes on urban transport.

The system has the advantages of high security and low cost. In addition, the whole process of logistics could be supervised without special equipment. These lay a solid foundation for “release through” supervision of agricultural products supplied to Hong Kong.

BackgroundAn accurate assessment of nailfold capillaroscopy (NFC) images has great importance in the diagnosis and prognosis of systemic sclerosis (SSc). To overcome some of the inherent problems with NFC image analysis (operator/observer bias, time requirements), there is an interest to automate and standardize NFC image assessment using computer vision algorithms, such as the Vision Transformer (ViT), which are based on recent advances in deep learning.ObjectivesOur aims were (1) to implement and assess the performance and the reliability of ViT in detecting changes on NFC images, and (2) to compare the performance of ViT to that of practicing rheumatologists.MethodsFor this study, we used NFC images of patients with SSc enrolled in our European Scleroderma Trials and Research group (EUSTAR) and Very Early Diagnosis of Systemic Sclerosis (VEDOSS) local registries. Concretely, we included routine NFC images (all NFC images available – digit II to V of both hands-, irrespective of any image artefacts) of patients aged ≥ 18 years with visits between 2012 and 2021, who fulfilled either the 2013 American College of Rheumatology / European League Against Rheumatism classification criteria for SSc (established disease) or the preliminary criteria for VEDOSS (mild/early disease).ViT was trained to identify the following NFC signs of microangiopathy: enlarged capillaries (apex diameter of the capillary > 20 µm, and < 50µm), giant capillaries (> 50 µm), loss of capillaries (< 7 capillaries/mm), and microhaemorrhages. Absence of any of these signs was considered as being a “normal NFC”, while presence of any of these signs was seen as an abnormal NFC image. Its performance was evaluated in a cross-fold (k = 5) validation setting, and the gold standard was the treating physician. Area under the ROC curve (AUC) was used as indicator of the performance. Considered cut-offs for AUC were 90-100% - excellent, 80-89.9% - good, 70-79.9% - fair, 60-69.9% - poor, 50-59.9% - fail. From the first cross-fold, a randomly sampled reliability set was created which was used to compare the NFC assessment performance of four rheumatologists (three invited and trained annotators and the treating physician) with that of of ViT.ResultsWe analysed 17,126 NFC images of 234 EUSTAR patients (14.6% males, 67.8% limited cutaneous SSc, median age 57 years, median disease duration 9 years) and 55 VEDOSS patients (92.7% females, median age 44 years, median time elapsed since first Raynaud’s phenomenon 5 years). ViT had fair to excellent performance in identifying the different NFC changes across all five folds (of 3443 NFC images each), with an area under the ROC curve ranging from 78.59% to 90.4% (Figure 1a).Figure 1.Performance of the ViT: (a) AUCs values for ViT in diagnosing the respective feature on NFC image. Good AUCs were seen in depicting capillary loss (AUC = 87.3%), microhemorrhages (AUC = 85.9%) and the abnormal/normal NFC classification (AUC = 84.7%). The rheumatologists had generally higher performance in assessing NFC images. AbstractNFC (Near Field Communication) devices are used in contactless payment systems, similar to those used in credit cards and electronic ticket smart cards, and they allow a mobile payment to replace or supplement these systems. The trend of increasing the quality of transport services also includes the use of progressive information technologies. The article focuses on the use of NFC technology in regional passenger transport. In the first proposal, the implementation of NFC technology in the form of an electronic wallet in a regional contactless chip card is being considered. In the second proposal we deal with the implementation of NFC technology in regional passenger transport using mobile phones with an active NFC chip. The proposals use Check-In/Check-Out validation, where validators are placed in vehicles. In both proposals it would be possible to create a link to EMV (Europay/MasterCard/VISA) technology, and thus to provide a linking of a contactless smart card, possibly a mobile phone to the passenger’s personal account.

The purpose of the present study was to adapt and validate the Need for Cognition Scale–Short Form (NfC-SF) in the Greek language. A multistep process was followed, including (a) the translation and adaptation of the questionnaire, (b) a reliability analysis of the instrument’s items in combination with an exploratory factor analysis with 177 secondary school students, and (c) a confirmatory factor analysis for defining the underlying structure of the scale, using a sample of 532 secondary school students. The statistical analyses validated a 14-item version of the NfC-SF for measuring the cognitive motivation of secondary school, Greek-speaking students. The present study also extends previous research about the underlying structure of the NfC by suggesting that method effects should be considered in measurement models for improving scale validity.

Abstract To ensure the privacy of users in transport systems, researchers are working on new protocols providing the best security guarantees while respecting functional requirements of transport operators. In this paper1, we design a secure NFC m-ticketing protocol for public transport that preserves users’ anonymity and prevents transport operators from tracing their customers’ trips. To this end, we introduce a new practical set-membership proof that does not require provers nor verifiers (but in a specific scenario for verifiers) to perform pairing computations. It is therefore particularly suitable for our (ticketing) setting where provers hold SIM/UICC cards that do not support such costly computations. We also propose several optimizations of Boneh-Boyen type signature schemes, which are of independent interest, increasing their performance and efficiency during NFC transactions. Our m-ticketing protocol offers greater flexibility compared to previous solutions as it enables the post-payment and the off-line validation of m-tickets. By implementing a prototype using a standard NFC SIM card, we show that it fulfils the stringent functional requirement imposed by transport operators whilst using strong security parameters. In particular, a validation can be completed in 184.25ms when the mobile is switched on, and in 266.52ms when the mobile is switched off or its battery is flat.

Bionanocomposites of polylactide (PLA), plasticized with poly(ethylene glycol) (PEG) (7.5 wt%, 400 and 1500 g/mol) and reinforced with nanofibrillated cellulose (NFC) (1, 3, and 5 wt%) were sequentially compounded, and injection and compression molded. All of the stages caused structural and morphological consequences, more relevant in the plasticized PLA, especially with low molar PEG. Small percentages of NFC (1 and 3 wt%) acted as crystalline nucleating agents and improved thermo-oxidative stability. Given the substantial degradation caused by (re)processing, a downgrading validation strategy was applied, assessing the mechanical and water contact performance during fictional first and second service life applications. After the first processing, PEG increased the ductility and reduced the strength and elastic modulus, while NFC buffered the fall in stiffness and increased rigidity compared to their PLA-PEG counterparts. Once reprocessed, PEG increased the water affinity of the blend, especially for low molar mass PEG. Low percentages of NFC (1 and 3 wt%) modulated water diffusivity and permeability, regardless of the water temperature. Overall, although reprocessing caused significant degradation, the mechanical valorization possibilities of these green bionanocomposites were proven, and are pointed out as sustainable candidates for food packaging or agricultural applications where modulated mechanical or water contact behaviors are required.

Airline ticket reservations can be completed via ticket counters or online. To reduce passenger data redundancy, Single Identity (SI) may be required to input passenger data. In Indonesia, SI is utilised in identity cards using Near Field Communication (NFC), called electronic <em>Kartu Tanda Penduduk</em> (e-KTP). However, the use of e-KTP is not optimal yet because it is still used in the same way as the conventional one. Data input using SI can be done by tapping the e-KTP on a NFC-enabled machine, and the user is required to select the passengers’ names. The process of data input may be simplified, resulting in a higher transaction speed. This research result demonstrated that there was a slight difference of the time required between the two methods for ticket reservations. The time efficiency using SI through ticket counters was 48%, while the online reservation using SI was 45%. Further research may implement the validation methods, such as using PIN or fingerprint.

The discrimination of non-focal class (NFC) and focal class (FC), is vital in localizing the epileptogenic zone (EZ) during neurosurgery. In the conventional diagnosis method, the neurologist has to visually examine the long hour electroencephalogram (EEG) signals, which consumes time and is prone to error. Hence, in this present work, automated diagnosis of FC EEG signals from NFC EEG signals is developed using the Fast Walsh–Hadamard Transform (FWHT) method, entropies, and artificial neural network (ANN). The FWHT analyzes the EEG signals in the frequency domain and decomposes it into the Hadamard coefficients. Five different nonlinear features, namely approximate entropy (ApEn), log-energy entropy (LogEn), fuzzy entropy (FuzzyEn), sample entropy (SampEn), and permutation entropy (PermEn) are extracted from the decomposed Hadamard coefficients. The extracted features detail the nonlinearity in the NFC and the FC EEG signals. The judicious entropy features are supplied to the ANN classifier, with a 10-fold cross-validation method to classify the NFC and FC classes. Two publicly available datasets such as the University of Bonn and Bern-Barcelona dataset are used to evaluate the proposed approach. A maximum sensitivity of 99.70%, the accuracy of 99.50%, and specificity of 99.30% with the 3750 pairs of NFC and FC signal are achieved using the Bern-Barcelona dataset, while the accuracy of 92.80%, the sensitivity of 91%, and specificity of 94.60% is achieved using University of Bonn dataset. Compared to the existing technique, the proposed approach attained a maximum classification performance in both the dataset.

The evolution of modern mobile devices towards novel Radio Frequency (RF) capabilities, such as Near Field Communication, leads to a potential for delivering innovative mobile services, which is still partially unexplored. Mobile proximity payment systems are going to enhance the daily shopping experience, but the access to payment security resources of a mobile device (e.g. the “Secure Element”) by third party applications is still blocked by smartphone and Operating System manufacturers. In this paper, the IDA-Pay system is presented, an innovative and secure NFC micro-payment system based on Peer-to-Peer NFC operating mode for Android mobile phones. It allows to deliver mobile-to-POS micro-payment services, bypassing the need for special hardware. A validation scenario and a system evaluation are also reported to demonstrate the system effectiveness and performance.

Introduction. Further development of urban formations requires optimization of passenger traffic management in the public transport network. Ensuring the efficiency of traffic flow management involves improving accounting with the use of innovative NFC- validation and GPS-navigation systems simultaneously for all participants in passenger traffic. The purpose of the article. The main aim is to provide proposals for improving the cost and revenue accounting of carriers and issuers of electronic tickets for NFC validators in terms of information interaction of all participants in passenger traffic to ensure the effectiveness of automated passenger traffic management in the smart city.Methods. General scientific and empirical methods of cognition of processes and phenomena are used to reveal the purpose of the article.Results. A method of using information from the automated passenger traffic management system using NFC-validation and GPS-navigation technologies on the number of transported passengers to identify costs and revenues of public transport participants has been developed. The procedure for determining the cost of transportation of one passenger on the basis of additive accumulation of costs of carriers and issuers of electronic tickets has been improved. It is proposed to recognize the funds received from the sale of electronic tickets as targeted revenue (targeted funding). The method of accounting for revenues of issuers of electronic tickets and carriers has been improved. Implementation of the proposed method of parallel recognition and accounting of revenues and costs simultaneously for issuers of electronic documents and passenger carriers provides optimization of management in all market participants of passenger transport, which will lead to optimization: transport routes, service quality, reliability of accounting, socio-economic processes making managerial decisions, etc.Perspectives. Further research will be aimed at improving the methodology of control over the functioning of passenger carriers and the transport network of public entities in the implementation of an automated passenger traffic management system.

Security remains a top priority for those users in the hotel, even with the advent of innovative technological advances. This is because many tragic incidents, such as theft and crime, have occurred with unrestricted access. This paper proposes an intelligent door access system that would allow hotel guests to authenticate into their rooms without resorting to traditional closeness access methods. Therefore, research was conducted to solidify the understanding and refine the capabilities of the proposed system. This project aims to promote high-security aspects access system technology, which is Near-Field Communication with the use of application that have the function of simulated smart keys for explicit validation access. A Host-Card Emulator opens the opportunities for efficient financial benefit and the launch of a protective mechanism in the post-pandemic period. The suggested method was statistically and analytically accessed on hotel guests and staff from various hotels in Malaysia. The proposed system is a contactless NFC access control system that employs smartphone Host Card Emulation application technology to reduce the need for appropriate physical access, enhance security, and publicize the use of mobile access systems in the hospitality industry.

The standard protocol of near field communication (NFC) has concentrated primarily on the speed of communication while ignoring security properties. Message between an NFC-enabled smartphone and a point of sale are exchanged over the air (OTA), which is a message considered an authentication request for payment, billing, ticketing, loyalty services, identification or access control. An attacker who has an antenna can intercept or manipulate the exchanged messages to take advantage of these. In order to solve this problem, many researchers have suggested authentication methods for NFC communications. However, these remain inadequate transaction security and fairness. In this paper, we will propose a technique that ensures mutual authentication, security properties, and strong fairness. Mutual authentication is a security property that prevents replay attacks and man-in-the-middle attacks. Both fair exchange and transaction security are also significant issues in electronic transactions with regards to creating trust among the parties participating in the transaction. The suggested protocol deploys a secure offline session key generation technique to increase transaction security and, importantly, make our protocol lightweight while maintaining the fairness property. Our analysis suggests that our protocol is more effective than others regarding transaction security, fairness, and lightweight protocol. The proposed protocol checks robustness and soundness using Burrows, Abadi and Needham (BAN) logic, the Scyther tool, and automated validation of internet security protocols and applications (AVISPA) that provide formal proofs for security protocols. Furthermore, our protocol can resolve disputes in case one party misbehaves.

One of the problems associated with the distribution of psychotropic drugs is that there is a lot of abuse and smuggling because of the lack of supervision. Therefore, it is necessary to make a supporting application, namely the application of monitoring the distribution of psychotropic drugs by transmitting data via the internet. By utilizing NFC technology on smartphones, Delphi programming, PHP, and also the MySQL database, an application is made to function as a system. Making this application is done using the Delphi 7 application as a user application and the Notepad ++ application to build a security web that is connected to the database on localhost php myadmin for data delivery monitoring the distribution of psychotropic drugs. From the test results of this application, the results of the application performance test run well as expected, and encrypted URLs can only be logged if there is a validation process with a database server, supported by an NFC tag system that has a reading distance of 3 cm proves that the application security system is good.

Abstract The distinctive security features of the Spanish electronic national identity card, known as Documento Nacional de Identidad electrónico, allow us to propose the usage of this cryptographic smart card in an authentication framework that can be used during the registration and login phases of internet services where the validation of the user’s age and real identity are key elements, as it is the case for example of the so-called social networks. Using this mechanism with NFC-capable devices, the identity and age of the potential user can be determined, allowing or denying the access to the service based on that information.

In this paper the authors propose a Secure and Optimized Proximity Mobile Payment (SOPMP) Framework using NFC (Near Field Communication) technology, WPKI (Wireless Public Key Infrastructure), UICC (Universal Integrated Circuit Card). The novelty of this proposed mobile payment framework is messages are exchanged in the form of Digital Signature with Message Recovery (DSMR) and merchant sends Invoice in the form of Digital Invoice Certificate (DIC) (which is digitally signed by the merchant). The communication link between mobile phone and merchant POS (Point Of Sale) is NFC. Digital Signature with Message Recovery based on ECDSA eliminates the need of adopting PKI cryptosystems thereby reducing the consumption of resources i.e. it consumes less computational and communication cost. DSMR eliminates the need of certificates validation and removes the hurdle of PKI thereby reducing storage space, communication cost and computational cost. The authors proposed protocol ensures Authentication, Integrity, Confidentiality and Non Repudiation, achieves Identity protection from merchant and Eavesdropper, achieves Transaction privacy from Eavesdropper and Payment Gateway, achieves Payment Secrecy, Order Secrecy, forward secrecy, and prevents Double Spending, Overspending and Money laundering. In addition to these our proposed protocol withstands Replay, Man in the Middle and Impersonation attacks. The security properties of the proposed protocol have been verified using AVISPA and Scyther Tools and presented with results.

This study aims to estimate and validate the accuracy of estimated Total Digestible Nutrient (TDN) content in forages based on the chemical composition of feed. Data on nutrient composition for estimates were obtained from the BR-CORTE database consisting of 86 types of forages and primary data from observations consisting of 19 types of forages. Analysis data is used Pearson correlation, multiple linear regression, determination coefficient (R2), and root mean square error (RMSE) for estimation models, variance test (ANOVA), and T-test for validation. The results showed that forage TDN had a negative correlation with Neutral Detergent Fibre (NDF) (P <0.01) and lignin (P <0.05), but was positively correlated with Non-Fibre Carbohydrate (NFC) (P <0.01), Ether Extract (EE) and Crude Protein (CP) (P<0.01). Thus, the TDN value can be estimated from the chemical composition in the feed. The regression equation that will be used to estimate forage TDN content is TDN = 0.482 NDF + 1.538 EE + 0.699 NFC + 0.718 CP (R2=0.994). Then, this equation is validated and compared to the Hartadi’s and Wardeh’s equations. The results of the T-test showed that the TDN value estimated is non significantly different with TDN Hartadi for cattle (P>0.05). It concluded TDN value of local forage can be estimated based on this estimation models.

An efficient and practical method for identifying mandarin juice over-blended into not from concentrate (NFC) orange juice was established. Juices were extracted from different cultivars of sweet orange and mandarin fruits. After being pasteurized, the volatile organic compounds (VOCs) in the juice samples were extracted using headspace solid-phase microextraction, and qualitatively and quantitatively analyzed using gas chromatography–mass spectrometry detection. Thirty-two VOCs contained in both the sweet orange juice and mandarin juice were used as variables, and the identification model for discriminating between the two varieties of juice was established by principal component analysis. Validation was applied by using common mandarin juices from Ponkan, Satsuma and Nanfengmiju cultivars blended at series of proportions into orange juices from Long-leaf, Olinda, and Hamlin cultivars. The model can visually identify a blending of mandarin juice at the volume fraction of 10% or above.

A new generation of public displays demands high interactive and multiscreen features to enrich people’s experience in new pervasive environments. Traditionally, research on public display interaction has involved mobile devices as the main characters during the use of personal area network technologies such as Bluetooth or NFC. However, the emergent Smart TV model arises as an interesting alternative for the implementation of a new generation of public displays. This is due to its intrinsic connection capabilities with surrounding devices like smartphones or tablets. Nonetheless, the different approaches proposed by the most important vendors are still underdeveloped to support multiscreen and interaction capabilities for modern public displays, because most of them are intended for domestic environments. This research proposes multiscreen interactive middleware for public displays, which was developed from the principles of a loosely coupled interaction model, simplicity, stability, concurrency, low latency, and the usage of open standards and technologies. Moreover, a validation prototype is proposed in one of the most interesting public display scenarios: the advertising.

Bentuk realisasi yang telah dilakukan diantaranya pembuatan National Fullfillment Center (NFC), proses validasi, cycle count, stock moving, stock opname, repacking, validation, replenishment, reporting aging, breakage, perhitungan buffer stock dan empty check. Strategi ini juga sangat penting diterapkan karena sebagian produk yang disimpan dan dijual merupakan produk rentan mengalami kerusakan (perishable) dan memiliki umur simpan yang realtif singkat. Metode penelitian ini menggunakan pendekatan ABC Analysis dan First in First Out (FIFO) sedangkan prosedur penelitian dilakukan dengan observasi secara langsung. Hasil penelitian ini didapatkan tabel analisis ABC dari 12 SKU, Tabel FIFO yang didukung oleh remaining shelf life untuk barang-barang perishable, dan perhitungan buffer stock. Proses perhitungan buffer stock didasarkan pada permintaan (tren) dan ketepatan penyimpanan. Berdasarkan permintaan (demand) produk dan ketersediaan stok, penerapan metode ABC dan FIFO berimplikasi dalam meningkatkan produktivitas seperti keseimbangan lini (Line Balancing). Wireless power transfer (WPT) devices represent one of the most efficient and increasingly used technologies for the transfer of data and power in the near-field range. This work analyzes and describes a new type of device: a ferrite-cored, variable gap, high-frequency power and data transfer system. The classic theoretical models existing in the literature for near-field communication (NFC) and WPT devices have foreseen a lumped-parameters characterization based on the representation of an equivalent circuit model (ECM). The strict interdependence between the different physical domains has clearly increased the difficulty in predicting the behavior of the device, due to the unwanted continuous and chaotic variation of the parameters. The proposed paper aims to provide a general and reliable multi-physics model based on the co-simulation of a Spice®-based ECM analysis and the ESRF Radia®-based 3D finite volume methodology (3DFVM), placing particular emphasis on the intrinsic sensitivity with respect to variables that cannot be directly controlled, such as the variation of the air gap between the coupled coils interfaces. Furthermore, this work outlines a detailed and effective experimental methodology for the estimation of static and dynamic electro-magnetic parameters and the validation of the numerical models in both the time and frequency domain, through the analysis of a real coupled WPT device.

Public transport sustainability is becoming a major driver for public transport development. Public transport ridership represents one of the key performance indicators of sustainability in the sense of balancing the economic, social and environmental aspects of public transport. There are various methods for improving the attractiveness of public transport for passengers by reducing resistances, which discourage potential and existing passengers to use public transport. Transport ticketing is one of the methods. This article presents a methodological framework for evaluating transport ticketing technologies with the use of a transport ticketing convenience model developed by the authors as well as some survey results through the application of the developed framework on traditional smart ticketing and contactless payment card ticketing technologies. First, a methodological framework for modelling ticketing convenience based on end-to-end passenger experience is presented. Second, a ticketing convenience model for barrier-free and double-sided validation baseline ticketing systems is developed. Third, the ticketing system based on contactless bank payment cards is compared with traditional smart ticketing systems in terms of convenience. It is shown that a contactless payment cards ticketing system has greater convenience or a better, more seamless travel experience than traditional smart systems. Finally, some research perspectives on enhancing the ticketing convenience model by using mobile smartphones with NFC, BLE and GPS technology, as well as the inclusion of technologies related to ticketing such as passenger information systems into the model, are contemplated.

Nowadays, applications for the Internet of Things (IoT) have been introduced in different fields of medicine to provide more efficient medical services to the patients. A systematic mapping study was conducted to answer ten research questions with the purposes of identifying and classifying the present medical IoT technological features as well as recognizing the opportunities for future developments. We reviewed how cloud, wearable technologies, wireless communication technologies, messaging protocols, security methods, development boards, microcontrollers, mobile/IoT operating systems, and programming languages have been engaged in medical IoT. Based on specific inclusion/exclusion criteria, 89 papers, published between 2000 and 2018, were screened and selected. It was found that IoT studies, with a publication rise between 2015 and 2018, predominantly dealt with the following IoT features: (a) wearable sensor types of chiefly accelerometer and ECG placed on 16 different body parts, especially the wrist (33%) and the chest (21%) or implanted on the bone; (b) wireless communication technologies of Bluetooth, cellular networks, and Wi-Fi; (c) messaging protocols of mostly MQTT; (d) utilizing cloud for both storing and analyzing data; (e) the security methods of encryption, authentication, watermark, and error control; (f) the microcontrollers belonging to Atmel ATmega and ARM Cortex-M3 families; (g) Android as the commonly used mobile operating system and TinyOS and ContikiOS as the commonly used IoT operating systems; (h) Arduino and Raspberry Pi development boards; and finally (i) MATLAB as the most frequently employed programming language in validation research. The identified gaps/opportunities for future exploration are, namely, employment of fog/edge computing in storage and processing big data, the overlooked efficient features of CoAP messaging protocol, the unnoticed advantages of AVR Xmega and Cortex-M microcontroller families, employment of the programming languages of Python for its significant capabilities in evaluation and validation research, development of the applications being supported by the mobile/IoT operating systems in order to provide connection possibility among all IoT devices in medicine, exploiting wireless communication technologies such as BLE, ZigBee, 6LoWPAN, NFC, and 5G to reduce power consumption and costs, and finally uncovering the security methods, usually used in IoT applications, in order to make other applications more trustworthy.

Cystic fibrosis (CF) predisposes patients to bacterial colonization and infection of the lower airways. Several species belonging to the genus Burkholderia are potential CF-related pathogens, but microbiological identification may be complicated. This situation is not in the least due to the poorly defined taxonomic status of these bacteria, and further validation of the available diagnostic assays is required. A total of 114 geographically diverse bacterial isolates, previously identified in reference laboratories as Burkholderia cepacia (n = 51), B. gladioli(n = 14), Ralstonia pickettii(n = 6), B. multivorans(n = 2), Stenotrophomonas maltophilia(n = 3), and Pseudomonas aeruginosa(n = 11), were collected from environmental, clinical, and reference sources. In addition, 27 clinical isolates putatively identified as Burkholderia spp. were recovered from the sputum of Dutch CF patients. All isolates were used to evaluate the accuracy of two selective growth media, four systems for biochemical identification (API 20NE, Vitek GNI, Vitek NFC, and MicroScan), and three different PCR-based assays. The PCR assays amplify different parts of the ribosomal DNA operon, either alone or in combination with cleavage by various restriction enzymes (PCR-restriction fragment length polymorphism [RFLP] analysis). The best system for the biochemical identification of B. cepacia appeared to be the API 20NE test. None of the biochemical assays successfully grouped theB. gladioli strains. The PCR-RFLP method appeared to be the optimal method for accurate nucleic acid-mediated identification of the different Burkholderia spp. With this method, B. gladioli was also reliably classified in a separate group. For the laboratory diagnosis of B. cepacia, we recommend parallel cultures on blood agar medium and selective agar plates. Further identification of colonies with a Burkholderiaphenotype should be performed with the API 20NE test. For final confirmation of species identities, PCR amplification of the small-subunit rRNA gene followed by RFLP analysis with various enzymes is recommended.

e13623 Background: The high false-negative rate (FNR) of sentinel lymph node biopsy (SLNB) alone results in unnecessary axillary lymph node dissection (ALND) in initially clinically node-positive (cN+) breast cancer after neoadjuvant chemotherapy (NAC). This study aimed to reduce the FNR by establishing a longitudinal MRI radiomics-assisted SLNB model compared with SLNB alone. Methods: We established and tested a machine learning algorithm (RMLM) based on pre-NAC, post-NAC and longitudinal MRI radiomics features to detect axillary complete response (ypN0) to neoadjuvant chemotherapy in cN+ breast cancer. In total, 234 eligible patients who underwent SLNB and ALND were retrospectively recruited in the primary cohort (PC), 723 patients were in external validation cohorts (EVC) 1-3, and 81 patients from two multicentre prospective clinical trials (ClinicalTrials.gov, NCT03154749, NCT04858529) were enrolled in the prospective validation cohort (PVC). The U test and least absolute shrinkage and selection operator with tenfold cross-validation were used to select the most significant features. The specificity and FNR of RMLM-assisted SLNB were compared with those of SLNB alone. Results: In the primary (n=234 patients), external validation (n=723) and prospective validation sets (n=81), RMLM achieved excellent performance in detecting metastatic axillary lymph nodes (AUC=0.938 in the primary cohort, 0.842-0.913 in the external cohorts, 0.865 in the prospective validation cohort). RMLM-assisted SLNB achieved an FNR of 4.96% in PC, 5.83-8.08% in EVC1-3, and 9.09% in the prospective validation cohort. The removal of more than two SLNs combined with RMLM could further reduce the FNR to 2.78% in PC, 3.28-7.27% in EVC1-3, and 6.67% in the prospective validation cohort. Conclusions: RMLM combined with SLNB provides a potential tool to evaluate residual nodal disease after NAC and reduce unnecessary ALND.

Objective: To investigate the value of delta-radiomics after the first cycle of neoadjuvant chemotherapy (NAC) using dynamic contrast-enhanced (DCE) MRI for early prediction of pathological complete response (pCR) in patients with breast cancer. Methods: From September 2018 to May 2021, a total of 140 consecutive patients (training, n = 98: validation, n = 42), newly diagnosed with breast cancer who received NAC before surgery, were prospectively enrolled. All patients underwent DCE-MRI at pre-NAC (pre-) and after the first cycle (1st-) of NAC. Radiomic features were extracted from the postcontrast early, peak, and delay phases. Delta-radiomics features were computed in each contrast phases. Least absolute shrinkage and selection operator (LASSO) and a logistic regression model were used to select features and build models. The model performance was assessed by receiver operating characteristic (ROC) analysis and compared by DeLong test. Results: The delta-radiomics model based on the early phases of DCE-MRI showed a highest AUC (0.917/0.842 for training/validation cohort) compared with that using the peak and delay phases images. The delta-radiomics model outperformed the pre-radiomics model (AUC = 0.759/0.617, p = 0.011/0.047 for training/validation cohort) in early phase. Based on the optimal model, longitudinal fusion radiomic models achieved an AUC of 0.871/0.869 in training/validation cohort. Clinical-radiomics model generated good calibration and discrimination capacity with AUC 0.934 (95%CI: 0.882, 0.986)/0.864 (95%CI: 0.746, 0.982) for training and validation cohort. Delta-radiomics based on early contrast phases of DCE-MRI combined clinicopathology information could predict pCR after one cycle of NAC in patients with breast cancer.

The popularity of Web is increasing with their associated systems. These systems play a significant key role as their compromise has a technical effect, and their need is rising. One of these systems is called Technology wireless (NFC). NFC is becoming an active area nowadays. The complexity of setting the NFC increases as the application, system architecture, and associated parts must satisfy various requirements of ever involving project scenarios. Several organizations are getting the benefit from the NFC. This paper will explore and develop an approach of developing a technique that will permit to exchange by technology wireless (NFC) data between the parties within a Web services environment. This technique has an advantage that facilitates the possibility of adopting such a system to decrease the time-cost and effort for staff and customers. Several means of mobile payments technique have been entitled and adopted this service in many countries, e.g., Europe, USA, Asia, and in the Middle East such as Saudi Arabia. Many users of these countries using a smartphone to pay services via their mobile phones for a wide range of activities. Consequently, this paper involves an agile technique of validating Finite State Machine (FSM) and JFLAP tools.

AbstractNortheast China (NEC) has sustained economic losses in recent years because of extreme precipitation events. Despite many efforts, it remains very difficult to predict these extreme events. In this study, we documented the characteristics of extreme precipitation days (EPD) over NEC and established a seasonal prediction model using a year-to-year increment (DY) approach. The results show that most of the EPD over NEC occurred during midsummer, along with large values concentrated over the Greater and Lesser Khingan Mountains and Changbai Mountain. Two variables—the preceding early spring soil moisture DY over central Asia and the sea surface temperature DY in the tropical Atlantic Ocean—were used to construct the statistical model to predict the EPD DY over NEC. These two factors influenced the EPD by modulating the moisture transport over NEC. Cross-validation tests for the period from 1962 to 2016 and independent hindcasts for the period from 1997 to 2016 indicated that the two variables gave good predictions of the EPD over NEC. The observed and predicted year-to-year increments in EPD were well correlated, with a correlation coefficient of 0.65 for the period from 1962 to 2016 in the cross-validation test. In addition, the EPD DY covaried coherently with the midsummer precipitation amount DY over NEC, and those two predictors also gave good predictions for the midsummer precipitation amount over NEC. The correlation coefficient is 0.68 between the observed and predicted year-to-year increment in the amount of midsummer precipitation from 1962 to 2016 in a cross-validation test.

e12090 Background: Pathological complete response (pCR) of axillary lymph nodes (ALNs) is frequently achieved in patients with clinically node-positive breast cancer after neoadjuvant chemotherapy (NAC), and ALN status is an important prognostic factor for breast cancer patients. Our goal is to develop a new predictive clinical model to assess the axillary lymph node pCR rate after NAC. Methods: A retrospective series of 547 patients who had biopsy-proven positive ALNs at diagnosis and undergoing axillary lymph node dissection from 2007 to 2014 in National Cancer Center/Cancer Hospital of Chinese Academy of Medical Sciences. We analyzed the clinicopathologic features and developed a nomogram to predict the probability of ALN pCR. Univariate assessment was performed using a logistic regression model. A multivariate logistic regression stepwise model was used to generate a nomogram to predict ALN pCR in node positive patients Variables with P < 0.05 on multivariable analysis were included in the nomogram. The adjusted area under the receiver operating characteristic curve (AUC) was calculated to quantify the ability to rank patients by risk. Internal validation was estimated using 50-50 hold out validation method. Nomogram was validated externally with the prospective cohort of 167 patients from 2016 to 2018. Results: In retrospective study, there were 172 (31.4%) patients achieved axillary pCR after NAC. Multivariate analysis indicated that clinical nodal (N) stage, hormone receptor (HR) status and clinical response of primary tumor after NAC were significant independent predictors for axillary pCR ( P< 0.05). The NAC nomogram was based on these three variables. In the internal validation of performance, the AUCs for the training and test sets were 0.719 and 0.753, respectively. The nomogram was validated in an external cohort with an AUC of 0.734. Conclusions: We developed a nomogram to predict the likelihood of axillary pCR in node positive breast cancer patients after NAC. The predictive model performed well in prospective external validation. This practical tool could provide information to surgeons regarding whether to perform additional ALND after NAC.

Abstract Background: Molecular-biomarker-based prediction models, whether based on statistics or artificial intelligence, are becoming increasingly popular in the medical community as an alternative or complement to more traditional prediction tools, such as staging systems. Population-based cohorts are the preferred study design for building and validating such models, but they are generally expensive and frequently unfeasible, particularly, when the outcome of interest is rare, and when the collected data include expensive or difficult-to-obtain molecular biomarkers. As such, studies often develop or validate risk models in cohorts obtained based on data availability, rather than on the representativeness of a target population. Nested case-control (NCC) is an efficient study design for building and validating such models in a rare outcome setting, but it requires a fully enumerated source population, and appropriate methodologies to accommodate the under-sampling of the controls and any matching. Unfortunately, these methodologies are not systematically described in a form easily accessible to clinicians. Methods: We systematically characterized how to correctly develop and evaluate the performance of prediction models in NCC cohorts, by weighing the subjects based on sampling probabilities to account for the sub-cohort sampling. We illustrated the use of weighted metrics for the NCC, with a validation of a model that predicts breast cancer development - BOADICEA version 5 - in the population-based Rotterdam study. We used the C-index, threshold-based metrics, observed-to-expected events ratio (O/E ratio), calibration slope, and decision curve analysis as performance metrics. We compared the metrics obtained in the full cohort with those obtained in NCC cohorts sampled from the Rotterdam study, with and without a matched design. Results: NCC cohorts of 326 women were derived from the full cohort of 4,377 women from the Rotterdam Study. Performance metrics without weight adjustment were biased on the NCC cohorts: the unweighted C-index of the BOADICEA model was 0.61 (0.58-0.63) for the unmatched design. However, with weight adjustment, the C-index in the NCC cohorts (0.65 (0.61-0.69)) corresponds to the C-index in the full cohort (0.65 (0.62-0.69)), despite the NCC cohort being much smaller in size. Similarly, weighted adjustments of O/E ratio, threshold-based metrics, and net benefit for decision curves were unbiased estimates of the corresponding metrics in the full cohort, while the corresponding unweighted metrics were biased. Conclusions: We provided a practical guide for clinicians on how to develop and validate prediction models in NCC cohorts. We showed that this study design is an efficient solution, in case of expensive or difficult-to-obtain biomarkers, and when the outcome is rare; but the performance metrics must be appropriately adjusted to the sampling procedure. Although social validation of the goals, methods, and outcomes of evidence-based practices (EBPs) in autism treatment is a significant factor in their selection and effective use, EBPs are typically identified on the basis of the technical soundness of research without consideration of social validity. The authors investigated EBPs and emerging treatments identified by the National Autism Center (NAC) and National Professional Development Center on Autism Spectrum Disorders (NPDC) to determine which interventions have evidence of social validity, and the types of social validation addressed. A review of 828 articles cited by the NAC and NPDC determined that only 221 articles (26.7%) demonstrated direct evidence of the measurement of social validation. Of seven social validity categories analyzed, only consumer satisfaction, clinically significant behavioral change, and socially important dependent variables were consistently reported. A list of EBPs with varying levels of social and empirical validation is presented, and implications for future research are discussed.

A fiber temperature sensor based on no core fiber-few mode fiber-no core fiber (NCF-FMF-NCF) is proposed. It consists of two segments of NCF and a segment of FMF, with the NCF fused at both ends of the FMF. Meanwhile, the lengths of the NCF and FMF were optimized by simulation simulations and experimental validation. The results show that the sensor has a high sensitivity to the external refractive index (RI) changes, and enables a wide range of ambient temperature measurement. A sensitivity of 0.09445nm/? was obtained in a temperature range of 25-70?. The sensor has the advantages of high stability, good linear fit and simple structure.

PURPOSE: To describe the difficulties of implementing the protocol of experimental necrotizing enterocolitis (NEC) in order to obtain a larger number of newborns affected with the disease and a lower mortality. METHODS: Term Sprague-Dawley newborns rats (22 days) were divided into four groups of 12 fetuses each (n = 48): EC - breastfed newborns; IH - breastfed newborns and subjected to a stress protocol by ischemia and hypothermia; ESB - formula-fed newborns (Esbilac®, PetAg, Hampshire, IL, USA) and NEC - formula-fed newborns and subjected to stress protocol. The parameters set for the study protocol were: milk concentration (0.19 g ml or 0.34 g/ml), diet instilled volume (according to body weight - 200 kcal/day/Kg - or progressive, according to acceptance), weight (gain, loss or maintenance) and duration of the experiment (72 hours or 96 hours). Data of body weight (BW), intestinal weight (IW) and the IW/BW ratio were obtained. Samples of terminal ileum were collected and analyzed by the degree of injury to the intestinal wall. Statistically significance was set to p<0.05. RESULTS: The established protocol with less mortality and increased number of NEC was with Esbilac® at a concentration of 0.19 g/ml of diet instilled volume of 0.1 ml, every 3 hours, for 72 hours. All infants fed with artificial milk lost weight. In the degree score of intestinal injury, the ESB, IH and NEC groups were considered positive for NEC with greater histological injury in the latter. CONCLUSION: The described NEC protocol in rats allowed a greater survival of puppies with a greater number of animals affected by the disease.

The aim of our retrospective study is to develop and externally validate an 18F-FDG PET-derived radiomics model for predicting pathologic complete response (pCR) after neoadjuvant chemotherapy (NAC) in breast cancer patients. A total of 87 breast cancer patients underwent curative surgery after NAC at Soonchunhyang University Seoul Hospital and were randomly assigned to a training cohort and an internal validation cohort. Radiomic features were extracted from pretreatment PET images. A radiomic-score model was generated using the LASSO method. A combination model incorporating significant clinical variables was constructed. These models were externally validated in a separate cohort of 28 patients from Soonchunhyang University Buscheon Hospital. The model performances were assessed using area under the receiver operating characteristic (AUC). Seven radiomic features were selected to calculate the radiomic-score. Among clinical variables, human epidermal growth factor receptor 2 status was an independent predictor of pCR. The radiomic-score model achieved good discriminability, with AUCs of 0.963, 0.731, and 0.729 for the training, internal validation, and external validation cohorts, respectively. The combination model showed improved predictive performance compared to the radiomic-score model alone, with AUCs of 0.993, 0.772, and 0.906 in three cohorts, respectively. The 18F-FDG PET-derived radiomic-based model is useful for predicting pCR after NAC in breast cancer.

Over ten years of Visible Infrared Imaging Radiometer Suite (VIIRS) Environmental Data Records (EDR) Imagery Team activities have included primarily imagery validation, but also product generation and display and user interactions. VIIRS imagery validation starts with pre-launch preparations leading up to producing first-light imagery shortly after the launch of each Joint Polar Satellite System (JPSS) satellite. Imagery quality is scrutinized for typical imagery visualization problems, as well as the overall ability to utilize VIIRS imagery for analysis and forecasting purposes. Then, long-term monitoring of imagery continues through the lifetime of each VIIRS instrument. The VIIRS EDR Imagery Team has undertaken four major ground system code changes. The first of these code changes was needed in 2013 when Near Constant Contrast (NCC) Imagery at night was not routinely being generated from the Day-Night Band (DNB) due to incorrect sensitivity limits. The second applied Terrain Correction to the VIIRS EDR Imagery in 2020. The third, in 2021, was needed to fix an imagery banding anomaly in the NCC, which was masked for years by the natural variability of most NCC Imagery. The fourth was the increase from 6 M-band EDRs to all 16 M-band EDRs in 2021, allowing for the display of true-color and other multi-band imagery products from VIIRS Imagery EDRs. Here, we summarize the efforts of the VIIRS EDR Imagery Team which have resulted in a valuable suite of quality-controlled imagery products for the user community.

Herpesviral nuclear egress is a regulated process of viral capsid nucleocytoplasmic release. Due to the large capsid size, a regular transport via the nuclear pores is unfeasible, so that a multistage-regulated export pathway through the nuclear lamina and both leaflets of the nuclear membrane has evolved. This process involves regulatory proteins, which support the local distortion of the nuclear envelope. For human cytomegalovirus (HCMV), the nuclear egress complex (NEC) is determined by the pUL50–pUL53 core that initiates multicomponent assembly with NEC-associated proteins and capsids. The transmembrane NEC protein pUL50 serves as a multi-interacting determinant that recruits regulatory proteins by direct and indirect contacts. The nucleoplasmic core NEC component pUL53 is strictly associated with pUL50 in a structurally defined hook-into-groove complex and is considered as the potential capsid-binding factor. Recently, we validated the concept of blocking the pUL50–pUL53 interaction by small molecules as well as cell-penetrating peptides or an overexpression of hook-like constructs, which can lead to a pronounced degree of antiviral activity. In this study, we extended this strategy by utilizing covalently binding warhead compounds, originally designed as binders of distinct cysteine residues in target proteins, such as regulatory kinases. Here, we addressed the possibility that warheads may likewise target viral NEC proteins, building on our previous crystallization-based structural analyses that revealed distinct cysteine residues in positions exposed from the hook-into-groove binding surface. To this end, the antiviral and NEC-binding properties of a selection of 21 warhead compounds were investigated. The combined findings are as follows: (i) warhead compounds exhibited a pronounced anti-HCMV potential in cell-culture-based infection models; (ii) computational analysis of NEC primary sequences and 3D structures revealed cysteine residues exposed to the hook-into-groove interaction surface; (iii) several of the active hit compounds exhibited NEC-blocking activity, as shown at the single-cell level by confocal imaging; (iv) the clinically approved warhead drug ibrutinib exerted a strong inhibitory impact on the pUL50–pUL53 core NEC interaction, as demonstrated by the NanoBiT assay system; and (v) the generation of recombinant HCMV ∆UL50-ΣUL53, allowing the assessment of viral replication under conditional expression of the viral core NEC proteins, was used for characterizing viral replication and a mechanistic evaluation of ibrutinib antiviral efficacy. Combined, the results point to a rate-limiting importance of the HCMV core NEC for viral replication and to the option of exploiting this determinant by the targeting of covalently NEC-binding warhead compounds.

We compared the accuracy of prediction of the response to neoadjuvant chemotherapy (NAC) in osteosarcoma patients between machine learning approaches of whole tumor utilizing fluorine−18fluorodeoxyglucose (18F-FDG) uptake heterogeneity features and a convolutional neural network of the intratumor image region. In 105 patients with osteosarcoma, 18F-FDG positron emission tomography/computed tomography (PET/CT) images were acquired before (baseline PET0) and after NAC (PET1). Patients were divided into responders and non-responders about neoadjuvant chemotherapy. Quantitative 18F-FDG heterogeneity features were calculated using LIFEX version 4.0. Receiver operating characteristic (ROC) curve analysis of 18F-FDG uptake heterogeneity features was used to predict the response to NAC. Machine learning algorithms and 2-dimensional convolutional neural network (2D CNN) deep learning networks were estimated for predicting NAC response with the baseline PET0 images of the 105 patients. ML was performed using the entire tumor image. The accuracy of the 2D CNN prediction model was evaluated using total tumor slices, the center 20 slices, the center 10 slices, and center slice. A total number of 80 patients was used for k-fold validation by five groups with 16 patients. The CNN network test accuracy estimation was performed using 25 patients. The areas under the ROC curves (AUCs) for baseline PET maximum standardized uptake value (SUVmax), total lesion glycolysis (TLG), metabolic tumor volume (MTV), and gray level size zone matrix (GLSZM) were 0.532, 0.507, 0.510, and 0.626, respectively. The texture features test accuracy of machine learning by random forest and support vector machine were 0.55 and 0. 54, respectively. The k-fold validation accuracy and validation accuracy were 0.968 ± 0.01 and 0.610 ± 0.04, respectively. The test accuracy of total tumor slices, the center 20 slices, center 10 slices, and center slices were 0.625, 0.616, 0.628, and 0.760, respectively. The prediction model for NAC response with baseline PET0 texture features machine learning estimated a poor outcome, but the 2D CNN network using 18F-FDG baseline PET0 images could predict the treatment response before prior chemotherapy in osteosarcoma. Additionally, using the 2D CNN prediction model using a tumor center slice of 18F-FDG PET images before NAC can help decide whether to perform NAC to treat osteosarcoma patients.

Purpose: While a pathologic complete response (pCR) is regarded as a surrogate endpoint for positive outcomes in breast cancer (BC) patients receiving neoadjuvant chemotherapy (NAC), forecasting the prognosis of non-pCR patients is still an open issue. This study aimed to create and evaluate nomogram models for estimating the likelihood of disease-free survival (DFS) for non-pCR patients. Methods: A retrospective analysis of 607 non-pCR BC patients was conducted (2012–2018). After converting continuous variables to categorical variables, variables entering the model were progressively identified by univariate and multivariate Cox regression analyses, and then pre-NAC and post-NAC nomogram models were developed. Regarding their discrimination, accuracy, and clinical value, the performance of the models was evaluated by internal and external validation. Two risk assessments were performed for each patient based on two models; patients were separated into different risk groups based on the calculated cut-off values for each model, including low-risk (assessed by the pre-NAC model) to low-risk (assessed by the post-NAC model), high-risk to low-risk, low-risk to high-risk, and high-risk to high-risk groups. The DFS of different groups was assessed using the Kaplan–Meier method. Results: Both pre-NAC and post-NAC nomogram models were built with clinical nodal (cN) status and estrogen receptor (ER), Ki67, and p53 status (all p < 0.05), showing good discrimination and calibration in both internal and external validation. We also assessed the performance of the two models in four subtypes, with the triple-negative subtype showing the best prediction. Patients in the high-risk to high-risk subgroup have significantly poorer survival rates (p < 0.0001). Conclusion: Two robust and effective nomograms were developed to personalize the prediction of DFS in non-pCR BC patients treated with NAC.

Background. Immune deficiency is a cardinal feature of CLL and its predecessor MBL. To understand this issue, we studied individuals with MBL since a higher proportion of normal B cells (NBC) are present in this condition as compared to CLL and none of the individuals have been exposed to therapies that might alter immune function. Objectives. Weevaluated the transcriptomes of NBC from people with MBL (NBC-MBL) and compared these to NBC of healthy controls (NBC-HC) and to MBL clonal B cells (Clonal-MBL). Also, we compared the transcriptomic profiles of CD5ˉ NBC-M-MBL, NBC-U-MBL, and NBC-HC. Methods. PBMCs from 13 people with MBL (9 M-MBL, 4 U-MBL) and from 13 HC were FACS-isolated to obtain CD5 + clonal B cells (CD19 +CD20 DimCD5 +Igκ + or Igλ +) for MBL, and up to 4 B-cell fractions of NBC (CD19 +CD20 BrightCD5ˉ or CD5 +/Igκ + or Igλ +) for MBL and HC. Each cell fraction was &gt;99% pure. In all, 76 cell fractions were collected, 34 from MBL and 42 from HC. RNA was sequenced using SMART-Seq v4 Ultra Low Input and HiSeq platform. DESeq2 was used to analyze RNA-seq data. PCA clustered samples based on transcriptomic variance. Differentially expressed genes (DEG) were obtained with adjusted P&lt;0.05 and |FC|≥3. IPA identified relevant biological pathways. DEG protein products were validated by flow cytometry (FC) in 6 MBL (4 M-MBL and 2 U-MBL) and 8 HC. Results. PCAof all B-cell fractions showed that NBC-HC, NBC-MBL, and Clonal-MBL fell into 3 distinct groups, indicating each group has distinct transcriptomes. Focusing on NBC from HC and from MBL based on IGHV-mutation status, 3 groups with a similar gradient were identified: NBC-HC &gt; NBC-M-MBL &gt; NBC-U-MBL ( Fig 1), showing that NBC-HC differ from NBC-MBL and NBC differ within MBL based on IGHV-mutation types. Next, IPA was used to determine up- and down-regulated pathways ( Fig 2). For CD5ˉ NBC-MBL from all MBL people vs. NBC-HC (C1; 1,704 DEG), IPA revealed activation of inhibitory (e.g., FcγRIIB, the most significant pathway, which can inhibit B-cell function) and stimulatory pathways (e.g., [1] S100 signaling, the second most significant pathway, which can trigger NF-κB activation; [2] phospholipase C signaling, the third most significant pathway, involved in cell survival and proliferation; and [3] IL-4, essential for B-cell survival and maturation). To discriminate contributions of NBC from M-MBL and U-MBL, IPA was first performed for CD5ˉ NBC-M-MBL vs. NBC-HC (C2; 1,086 DEG), showing 76% (13/17) of pathways significantly inhibited (e.g., HMGB1, TNFR1, IL17, and IL6). Notably, of the only 4 upregulated pathways, 2 were inhibitory, IL-10 signaling (the most significant pathway) and NFKBIE signaling. These findings suggest downregulation/anergy of NBC in M-MBL. Strikingly, the CD5ˉ NBC-U-MBL vs. NBC-HC comparison (C3; 10,959 DEG) showed that 100% (11/11) of IPA pathways were activated and stimulatory (e.g., S100 signaling), consistent with increased NBC-U-MBL proliferation and migration. FC analyses detected significant ( P&lt;0.05) protein overexpression of CD24, CD27, CD300a, CD32 and IL10RA in CD5ˉ NBC from all MBL vs. NBC-HC; CD27, CD300a and IL10RA overexpression in CD5ˉ NBC-M-MBL vs. NBC-HC; and CD27 and TLR10 overexpression in CD5ˉ NBC-U-MBL vs. NBC-HC, validating gene expression results. Conclusions. PCA showed clear distinctions between NBC-MBL compared to NBC-HC, for both U-MBL and M-MBL. NBC-M-MBL appear to be functionally suppressed presumably due to IL-10 and NFKBIE signaling. In contrast, NBC-U-MBL appear to be stimulated through the S100 pathway. This activation of NBC-U-MBL seems paradoxical since in CLL there is greater immune suppression in U-CLL than M-CLL. Background. Current recommendations for the best views for the left main coronary artery (LMCA) ostium intervention are empirical. Objectives. To determine the optimal projection to visualize the LMCA ostium using only fluoroscopy. Methods. The optimal projection to visualize the LMCA ostium was determined using fluoroscopic images of superimposing the lowest points of the distal ends of two J tipped wires in the noncoronary cusp (NCC) and right coronary cusp (RCC). This was validated independently using 3-dimensional computed tomography (3D-CT) reconstruction. Results. Satisfactory images of the overlapping wires in NCC and RCC could be obtained in 90% (45/50). Between the fluoroscopic and the 3D-CT reconstruction approaches, the mean difference for NCC and RCC overlapping at horizontal axes is -1.8 with a 95% limit of agreement between −3.94 and 0.34 p = 0.10 and at vertical axes −1.6 with a 95% limit of agreement between −3.46 and 0.26 p = 0.09 ; and the mean difference for the optimal projection to visualize the LMCA ostium at horizontal axes is −3.22 with a 95% limit of agreement between -7.26 and 0.81 p = 0.11 and at vertical axes −2.31 with a 95% limit of agreement between −5.83 and 1.21 p = 0.09 . The 3D angulation deviation for the optimal projection to visualize the LMCA ostium was 8.5° ± 4.7° when the LMCA ostium faced the NCC-RCC commissure (n = 32) and 22.3° ± 16.0° p = 0.009 when it did not (n = 13). Conclusions. The optimal projection for LMCA ostial intervention can be determined using fluoroscopic images of superimposing wires in the NCC and RCC when the LMCA ostium faces the NCC-RCC commissure, as was the case in 71% of the patients studied.

Abstract BACKGROUND Alterations in NF2 underlie meningioma tumorigenesis, but tumor suppressor functions of the NF2 gene product, Merlin, are incompletely understood in meningiomas. Here we integrate proteomic proximity-labelling mass spectrometry with CRISPR interference (CRISPRi), RNA sequencing, and biochemical approaches to discover Merlin drives meningioma apoptosis and susceptibility to cytotoxic therapy. METHODS RNA sequencing was performed on triplicate M10G meningioma cells stably expressing CRISPRi machinery and either non-targeting control sgRNAs, sgRNAs suppressing NF2, or sgRNAs suppressing NF2 with Merlin rescue. QPCR in IOMM-Lee and MSC1 meningioma cells expressing non-targeting control shRNAs or shRNAs suppressing NF2 was used for orthogonal validation in vitro. RNA sequencing of euploid meningiomas (n=52) or meningiomas with loss of NF2 as the only copy number variant (n=28) was used for orthogonal validation in vivo. Merlin interactors in meningioma cells were identified using APEX proteomic proximity-labelling mass spectrometry. Mechanistic and functional studies were performed using biochemical, molecular, and cell biology approaches in meningioma cells and CH-157MN meningioma xenografts treated with cytotoxic chemotherapy or ionizing radiation. RESULTS Merlin suppression in meningioma cells and xenografts inhibited pro-apoptotic interferon regulatory factor (IRF) target genes and attenuated meningioma apoptosis. Merlin suppression did not alter IRF stability or subcellular localization in meningioma cells, and proteomic proximity-labelling mass spectrometry revealed a novel interaction between wildtype Merlin and ARHGAP35, a DNA binding factor that inhibits glucocorticoid receptor expression (NR3C1). NR3C1 inhibits IRF activity to prevent apoptosis, and Merlin suppression in meningioma cells induced NR3C1expression, which was inhibited by Merlin rescue. Further, NR3C1 suppression rescued meningioma cell apoptosis in the absence of Merlin, and NR3C1 expression was increased in human meningiomas with loss of NF2 compared to euploid meningiomas. CONCLUSIONS These data shed light on a novel pro-apoptotic tumor suppressor function of Merlin regulating glucocorticoid signalling and susceptibility to cytotoxic therapy in meningiomas.

593 Background: HER2-targeted neoadjuvant chemotherapy (NAC) possesses heterogeneous outcomes and currently lacks clinically-accepted markers of response. A means of predicting which patients will benefit prior to the treatment could reduce toxicity and the delay to effective intervention. Computational analysis of MRI via a deep neural network has shown promise in identifying NAC responders among mixed receptor subtype and treatment regimen cohorts, but faces challenges due to reproducibility across institutions and has not yet been explored in the context of HER2-targeted therapy. Here we present a deep learning approach for predicting response to HER2-targeted NAC from pre-treatment MRI. Methods: 100 HER2+ breast cancer patients who received NAC with docetaxel, carboplatin, trastuzumab, and pertuzumab at Cleveland Clinic (CCF) and had pre-treatment contrast-enhanced MRI’s were included in this analysis. 49 patients achieved pathological complete response (pCR, ypT0/is), while 51 patients retained presence of residual disease following NAC (non-pCR). 85 patients were used to train a convolutional neural network to predict pCR based on pre- and post-contrast MRI images, and the model design was optimized based on performance within a 15 patient internal validation cohort. An external, held-out testing dataset consisting of 28 patients (16 pCR, 12 non-pCR) imaged and treated at University Hospitals (UH) Cleveland Medical Center was used to validate the performance of the model. Performance was assessed by area under the receiver operating characteristic curve (AUC), accuracy, sensitivity, and specificity. A multivariable model incorporating age, hormone receptor status, stage, and tumor size was developed and similarly evaluated. Results: The neural network was able to predict the response to HER2-targeted NAC in the internal validation cohort (AUC = 0.93) as well as in an independent cohort from a separate institution (AUC = 0.85). This model offered superior performance compared to a multivariate clinical model, which achieved AUC = 0.67 and AUC = 0.52, in internal validation and external held-out testing cohorts, respectively. Conclusions: Deep learning analysis of contrast-enhanced MRI could be used to better target anti-HER2 therapy by pre-treatment prediction of response.[Table: see text]

e12625 Background: Neutrophil-to-lymphocyte ratio (NLR) has been found to offer clear predictive utility for the overall survival (OS) and pathologic complete response (pCR) in breast cancer (BC) patients receiving neoadjuvant chemotherapy(NAC). However, previous studies mainly focused on pre-NLR. The aim of this study was to explore the role of pre-/post-NLR towards OS and longitudinal NLR kineticsonpCR for BC patients undergoing NAC. And we also tried to build a nomogram for OS prediction based on these parameters. Methods: In this study, we retrospectively collected 501 female patients with locally advanced BC receiving 4-8 cycles of NAC from 2009 to 2018. Clinicopathological characteristics, NLR at pre-, mid-(every two cycles of NAC) and post-treatment were collected. The primary endpoint was OS. Among the patients, 421 patients with available pre- and post-NLR were included in the survival analysis. These patients were randomly divided into a training cohort (n = 224) and a validation cohort (n = 197). Multivariate survival model was built by including all the significant prognostic factors from the univariate analysis in the training cohort, and a nomogram model was established by “R” version 3.4.3. The performance of the model was further tested in the validation cohort by the concordance index. The second endpoint was pCR. Longitudinal analysis of NLR was performed using a mixed-effects regression model to predict pCR among 176 patients who finished 8 cycles of NAC. Results: The median follow-up time was 43.2 months for 421 patients. In the training cohort, multivariate analysis revealed that ER status, clinical node stage , pCR , pre-NLR, and post-NLR (all Ps<0.05) were independent predictors of OS. Nomogram for OS prediction was established by combining all these significant factors. The C-indexes of the nomogram were 0.764 and 0.693, respectively in the training and validation cohort. In the longitudinal analysis, patients achieved pCR experienced a reduction of NLR every 2 cycles (Coef = -0.032, std error = 0.014, P = 0.024). Conclusions: This study demonstrated the prognostic value of pre-NLR and post-NLR towards BC patients received NAC. Based on that , a novel nomogram was established to predict the 3- and 5- year OS for BC patients. And we also found patients who experienced a decline of NLR during NAC seems to be more likely to achieve pCR from chemotherapy. Routine assessment of NLR may be an easy and affordable tool for defining prognosis.

Necrotising enterocolitis (NEC) is a microbiome-dependent gut disease in preterm infants in early life. Antibiotic treatment is a common intervention for NEC. How NEC lesions, with or without antibiotics, affect plasma metabolome was explored in this study. Formula-fed preterm pigs were used as a model for human NEC and treated with saline, parenteral or oral antibiotics (n = 15–17) for four days after delivery. Gut tissues were collected for evaluation of NEC-like lesions and plasma for metabolomic analysis by proton nuclear magnetic resonance spectroscopy (1H-NMR). Metabolites were annotated, quantified and subjected to statistical modelling to delineate the effects of NEC and antibiotic treatment. Presence of severe NEC lesions, not antibiotic treatment, was the main drive for plasma metabolite changes. Relative to other pigs, pigs with severe NEC lesions had higher levels of alanine, histidine and myo-inositol, and lower levels of 3-hydroxybutyric acid and isobutyric acid. Across NEC lesion states (healthy, mild, severe), antibiotics directly affected only a few metabolites (tryptophan, 3-phenyllactic acid). Together and independently, NEC and antibiotic treatment affected circulating metabolites in preterm pigs. Amino acids and plasma metabolites, partly related to the gut microbiome, may be helpful to monitor progression of NEC lesions after proper validation.

Berita kriminalitas merupakan berita yang selalu menjadi trending topik di setiap media massa, khususnya media massa online. Media massa online terlah menyediakan beberapa fasilitas untuk mempermudah masyarakan dalam mencari sebuah berita berdasarkan topik. Media massa online melabeli suatu berita berdasarkan kategorinya. Namun, media massa online tidak memberikan sub kategori pada berita tersebut. Sebagai contoh jika seorang pengguna membuka kategori kriminal, maka yang ditampilkan adalah semua jenis berita kriminal tanpa memberikan informasi yang spesifik dari jenis kriminalitasnya. Permasalahan tersebut dapat diatasi dengan mengklasifikasikan berita kriminalitas berdasarkan subkategori. Penelitian ini menggunakan metode Naïve Bayes Classifier (NBC) untuk mengklasifikasi berita berdasarkan sub kategorinya. Adapun subkategori terbagi kedalam 5 kategori yaitu korupsi, narkoba, pencurian, pemerkosaan dan pembunuhan. Penelitian ini bertujuan untuk mengetahui kemampuan NBC dalam mengklasifikasi berita dengan melakukan pengujian menggunakan teknik K-Fold Cross Validation dengan nilai K dari 3 sampai 10. Hasil pengujian menyatakan bahwa NBC memiliki kemampuan dalam klasifikasi berita kriminal dengan nilai precision sebesar 98,53 %, nilai recall sebesar 98,44 % dan nilai accuracy sebesar 99,38 %.

Purpose. Previous studies have investigated the transcriptional modulations of aldosterone overproduction of aldosterone-producing adenomas (APAs). We aimed to systematically study the genes and pathways associated with molecular mechanism underlying APA by bioinformatics analysis and experimental validation for the expression profile. Methods. This study was performed based on three gene expression profiles (GSE64957, GSE8514, and GSE60042). Differentially expressed gene (DEG) investigation, function and pathway enrichment analysis, and protein-protein interaction (PPI) network analysis were performed by the bioinformatics analysis. For the validation with quantitative PCR, tissues from 11 patients with nonfunctioning adrenal adenoma (NFA) and 13 with APA were included in our cohort. Results. In this study, the bioinformatics analysis was performed and 182 upregulated and 88 downregulated DEGs were identified. As expected, the upregulated DEGs were primarily involved in calcium ion homeostasis ( p = 2.00X10−4). In the KEGG pathway analysis, calcium signaling pathway ( p = 4.38X10−6) and the aldosterone synthesis and secretion ( p = 8.73X10−6) were enriched. Moreover, quantitative PCR was performed to detect the expression of 7 upregulated genes (PCP4, ATP2A3, CYP11B2, CLCN5, HTR4, VDR, and AQP2) among the intersection of DEGs. The mRNA levels of CYP11B2, HTR4, and AQP2 were significantly increased in APA samples compared to NFA (24.420 folds of NFA, p < 0.001; 3.753 folds of NFA, p = 0.002; and 11.487 folds of NFA, p = 0.018). Conclusion. In summary, the present study showed several candidate genes with high expression from bioinformatics analysis and our cohort. Also, the DEGs were enriched in aldosterone synthesis and secretion and calcium signaling pathway as expected.
