Lipid membranes in nature adapt and reconfigure to changes in composition, temperature, humidity, and mechanics. For instance, the oscillating mechanical forces on lung cells and alveoli influence membrane synthesis and structure during breathing. However, despite advances in the understanding of lipid membrane phase behavior and mechanics of tissue, there is a critical knowledge gap regarding the response of lipid membranes to micromechanical forces. Most studies of lipid membrane mechanics use supported lipid bilayer systems missing the structural complexity of pulmonary lipids in alveolar membranes comprising multi-bilayer interconnected stacks. Here, we elucidate the collective response of the major component of pulmonary lipids to strain in the form of multi-bilayer stacks supported on flexible elastomer substrates. We utilize X-ray diffraction, scanning probe microscopy, confocal microscopy, and molecular dynamics simulation to show that lipid multilayered films both in gel and fluid states evolve structurally and mechanically in response to compression at multiple length scales. Specifically, compression leads to increased disorder of lipid alkyl chains comparable to the effect of cholesterol on gel phases as a direct result of the formation of nanoscale undulations in the lipid multilayers, also inducing buckling delamination and enhancing multi-bilayer alignment. We propose this cooperative short- and long-range reconfiguration of lipid multilayered films under compression constitutes a mechanism to accommodate stress and substrate topography. Our work raises fundamental insights regarding the adaptability of complex lipid membranes to mechanical stimuli. This is critical to several technologies requiring mechanically reconfigurable surfaces such as the development of electronic devices interfacing biological materials.

The effect of hormones on the morphology and cell surface polarity of the epithelial cell line MDCK was examined. When MDCK cells were seeded in high densities in media containing FCS a regular monolayer was formed. However, in serum-free medium supplemented with insulin, transferrin, prostaglandin E1, hydrocortisone and triiodothyronine, the development of a multilayer with intercellular lumina was observed. In hormone-depletion studies we identified PGE1 as the inducer of these multilayers. Since dibutyryl cyclic AMP and the phosphodiesterase inhibitor isobutyl methylxanthine could substitute for PGE1, we conclude that an elevated intracellular cAMP level resulted in formation of the multilayer. Further analysis by electron microscopy and immunocytochemistry revealed a polarized organization of the multilayered cells. Junctional complexes, enclosing microvilli-rich membrane domains, were found at the apices of adjacent cells facing the medium and those surrounding the intercellular lumina. Surprisingly, cells participating in the formation of both the free surface and the surface of the intercellular lumen, exhibited two distinct membranes with microvilli, each separated by junctional complexes. Immunolocalization of membrane marker proteins demonstrated that an apical 114 kDa membrane protein was localized to the free cell surfaces, the same membrane domains where extensive microvilli were also observed. The distribution of a basolateral 58 kDa membrane protein was restricted to sites of cell contact. These results provided evidence that nontransformed epithelial MDCK cells form multilayers in response to elevated cAMP levels; however, they retain the potential of developing cell surface polarity.

Self-assembled silk fibroin (SF)-polyethylenimine (PEI) multilayered films were fabricated on ethanol treated electrospun SF nanofibrous substrates via the electrostatic layer-by-layer (LBL) adsorption. The film coated membranes were characterized using scanning electron microscopy (SEM), transmission electron microscope (TEM) and X-ray photoelectron spectrophotometer (XPS). The SEM images showed that the multilayers of SF-PEI were formed on the surface of the ethanol treated SF nanofibres. The characteristics such as the fiber shape and porous structure were well maintained as the number of the coated SF-PEI bilayers was less than five. However, obvious adhesive substances and blocked pores were observed on the surface of the fibers as the number of bilayers of SF-PEI increased to six. Furthermore, the obvious core-shell structures were observed by TEM. The thickness of five SF-PEI bilayers was approximately 80nm. Additionally, the XPS results also revealed that the SF-PEI multilayer composite membranes formed. The adsorption mainly depended on a simple electrostatic interaction between the layers of SF and PEI. These SF-PEI multilayer assembled nanofibrous membranes could be a promising material for use as a sensor, gene delivery agent and scaffolds.

Some metals belonging to groups IV and V show a high permeability to hydrogen and have been studied as possible alternatives to palladium in membranes for hydrogen purification/separation in order to increase their sustainability and decrease their costs. However, to date, very few alloys among those metals have been investigated, and no membrane studies based on 4–5 element alloys with low or zero Pd content and quasi-amorphous structure have been reported so far. In this work, new membranes based on ZrVTi- and ZrVTiPd alloys were tested for the first time for this application. The unprecedented deposition of micrometric-based multilayers was performed via high-power impulse magnetron sputtering onto porous alumina substrates. Dense Pd/ZrxVyTizPdw/Pd multilayers were obtained. The composition of the alloys, morphology and structure, hydrogen permeance, selectivity, and resistance to embrittlement were tested and analyzed depending on the deposition conditions, and the membrane with the enhanced performance was tuned. The environmental impact of these membranes was also investigated to ascertain the sustainability of these alloys relative to more common Pd77Ag23 and V93Pd7 thin-film membranes using a life cycle assessment analysis. The results showed that the partial substitution of Pd can efficiently lead to a decrease in the environmental impacts of the membranes.

The performance of nine different vapor barrier systems comprising polyethylene (PE) membranes were assessed. The vapor barrier systems comprised membranes of virgin PE, 100% new PE, regenerated PE and multilayered virgin and regenerated PE. Membranes were joined either with tape suited to the individual system or an adhesive base on butyl rubber. The vapor barrier systems were evaluated and compared using standard laboratory tests. Chemical analytical techniques and physicomechanical tests were used. Mechanical properties were assessed using laboratory tests recommended by the harmonized standard EN 1385. Chemical analyses followed standard laboratory protocols performed with specialized equipment and visual examination. Chemical and mechanical properties were determined before and after exposure to an aging regime comprising 168 days at 70 °C in total. The chemical stability of the plastic present in each membrane was further evaluated after an additional exposure to an aging regime comprising 50 days followed by another 30 days at 70 °C. Additional aging indicated chemical changes in the membrane material with time. However, it was not possible to distinguish between aging properties for membranes containing virgin PE, 100% new PE, regenerated PE or multilayered virgin and regenerated PE.

The water transport through nanoporous multilayered graphene at 300k is investigated using molecular dynamics (MD) simulation with different water models in this study. We used functionalized and non-functionalized membranes along with five different 3-point rigid water models: SPC (simple point charge), SPC/E (extended simple point charge), TIP3P-FB (transferable intermolecular potential with 3 points—Force Balance), TIP3P-EW (transferable intermolecular potential with 3 points with Ewald summation) and OPC3 (3-point optimal point charge) water models. Based on our simulations with two water reservoirs and a porous multilayered graphene membrane in-between them, it is evident that the water transport varies significantly depending on the water model used, which is in good agreement with previous works. This study contributes to the selection of a water model for molecular dynamics simulations of water transport through multilayered porous graphene.

Thin film composite nanofiltration (NF) membranes are relatively new membranes compared to other types of pressure-driven membranes. However, they attract interest from researchers due to their versatility to be used in various applications. In this work, a new class of NF membrane was successfully fabricated through spin-assisted layer-by-layer assembly by depositing alternate layers of branched polyethylenimine (PEI) and poly(sodium 4-styrenesulfonate) (PSS) on ultrafiltration polysulfone (PSF) membrane. The suitability of the fabricated membranes for removal of divalent ions was investigated. It was found that the membrane consisting of (PEI/PSS)10–0.05 M NaCl showed MgCl2 rejection rate of 93.95% and permeation flux of 0.9 L/m2·h bar during tests performed using a crossflow permeation cell at a crossflow velocity of 0.65 m/s, MgCl2 feed concentration of 6530 ppm, pressure of 10 bar, temperature of 32.5°C, and pH of 6.5. This result suggests that this new fabrication method is suitable for producing polyelectrolyte multilayered (PEM) NF membranes that exhibit comparable membrane performance to commercial ones.

The internal stress and internal friction of electrodeposited multilayered Cu–Ni(100) thin films were investigated as a function of temperature by a sensitive vibrating membrane technique. Each membrane was in a state of biaxial stress (approaching the yield strength) resulting from contributions of the growth stress in the as-deposited film and from the thermal stress as the membrane was heated. The measured internal friction of these stressed multilayered films was enhanced by over an order of magnitude compared to the predicted value of classic damping related to the modulus defect when the effect of the internal stress was taken into account.

Lipid transport between membranes within cells involves vesicle and protein carriers, but as agents of nonvesicular lipid transfer, the role of membrane contact sites has received increasing attention. As zones for lipid metabolism and exchange, various membrane contact sites mediate direct associations between different organelles. In particular, membrane contact sites linking the plasma membrane (PM) and the endoplasmic reticulum (ER) represent important regulators of lipid and ion transfer. In yeast, cortical ER is stapled to the PM through membrane-tethering proteins, which establish a direct connection between the membranes. In this review, we consider passive and facilitated models for lipid transfer at PM–ER contact sites. Besides the tethering proteins, we examine the roles of an additional repertoire of lipid and protein regulators that prime and propagate PM–ER membrane association. We conclude that instead of being simple mediators of membrane association, regulatory components of membrane contact sites have complex and multilayered functions.

The paper presents the investigation results of an opportunity to create a new type membrane with increased characteristics for the pure hydrogen release from hydrogenous gas mixtures. A multilayer coating formed from alternating layers of Nb–Ti and Ti/TiNi on the substrates was used to create hydrogenselective membranes. The substrates of two types were used: plates of porous metal with submicron pore sizes and nickel plates with submicron columnar structure grown on its surface by means of nickel synthesis via a template.

Ovarian granulosa cells are derived embryologically from two keratin-positive epithelia of mesodermal origin, the ovarian rete and the ovarian surface epithelium. In the rat, presumptive granulosa cells still express keratin at birth but as they acquire functions related to oocyte support and steroidogenesis in the maturing ovary they lose this epithelial differentiation marker. Using double-label immunofluorescence microscopy, we examined the distribution of keratin-expressing granulosa cells in rat ovaries on days 1-10 postpartum in relation to (i) laminin and collagen type IV in follicular basement membranes, (ii) the zona pellucida, and (iii) 3β-hydroxysteroid dehydrogenase activity. Keratin was present in most (pre)granulosa cells on days 1-3. As the cells became multilayered in growing follicles, keratin was retained by granulosa cells adjacent to follicular basement membranes but disappeared from cells that were displaced towards follicular centers. From day 7 on, large follicles lacked keratin altogether. Laminin was a consistent component of follicular basement membranes at all ages, while collagen IV varied and diminished in parallel with keratin. 3β-Hydroxysteroid dehydrogenase was demonstrable in stromal interstitial cells from day 7 on. Zona pellucida first appeared in primary follicles adjacent to keratin-positive cells and subsequently became surounded with keratin-negative granulosa cells in growing follicles. The results suggest different roles for laminin and collagen IV in follicular basement membranes and support the hypothesis that keratin expression by granulosa cells depends on paracrine interactions with the ovarian stroma. In early growing follicles, these interactions may be interrupted by physical removal from the vicinity of the basement membranes as the granulosa cells become multilayered. In the more mature follicles, the loss of keratin from all granulosa cells suggests that the required stromal signals cease, perhaps as the perifollicular stroma differentiates into the theca.Key words: ovary, differentiation, keratin, basal membrane, development.

Poly(vinyl-alcohol) hydrogels have already been successfully utilised as drug carrier systems and tissue engineering scaffolds. However, lacking mechanical strength and suturability hinders any prospects for clinical and surgical applications. The objective of this work was to fabricate mechanically robust PVA membranes, which could also withstand surgical manipulation and suturing. Electrospun membranes and control hydrogels were produced with 61 kDa PVA. Using a high-speed rotating cylindrical collector, we achieved fibre alignment (fibre diameter: 300 ± 50 nm). Subsequently, we created multilayered samples with different orientations to achieve multidirectional reinforcement. Finally, utilising glutaraldehyde as a cross-linker, we created insoluble fibrous-hydrogel membranes. Mechanical studies were performed, confirming a fourfold increase in the specific loading capacities (from 0.21 to 0.84 Nm2/g) in the case of the monolayer samples. The multilayered membranes exhibited increased resistance from both horizontal and vertical directions, which varies according to the specific arrangement. Finally, the cross-linked fibrous hydrogel samples not only exhibited specific loading capacities significantly higher than their counterpart bulk hydrogels but successfully withstood suturing. Although cross-linking optimisation and animal experiments are required, these membranes have great prospects as alternatives to current surgical meshes, while the methodology could also be applied in other systems as well.

Purpose: Copolyamide 6,10 (coPA) electrospun mats were covered with multilayered (ML) and single-layered (SL) MXene (Ti3C2Tx) as a membrane for the separation of water/vegetable oil emulsions. Methods: Prepared membranes were characterized by atomic force microscopy (AFM), profilometry, the contact angle measurements of various liquids in air, and the underwater contact angle of vegetable oil. The separation efficiency was evaluated by measuring the UV transmittance of stock solutions compared to the UV transmittance of the filtrate. Results: The MXene coating onto coPA mats led to changes in the permeability, hydrophilicity, and roughness of the membranes and enhanced the separation efficiency of the water/vegetable oil emulsions containing 10, 100, and 1000 ppm of sunflower vegetable oil. It was found that membranes were highly oleophobic (>124°) under water, unlike in air, where the membranes showed high oleophobicity (<5°). The separation efficiency of water/oil emulsions for both types of covered membranes reached over 99%, with a surface coverage of 3.2 mg/cm2 Ti3C2Tx (for ML-Ti3C2Tx) and 2.9 mg/cm2 (for SL-Ti3C2Tx). Conclusions: The separation efficiency was greater than 98% for membranes covered with 2.65 mg/cm2 of ML-Ti3C2Tx, whereas the separation efficiency for membranes containing 1.89 and 0.77 mg/cm2 was less than 90% for all studied emulsion concentrations.

The creation of partial or complete human epidermis represents a critical aspect and the major challenge of skin tissue engineering. This work was aimed at investigating the effect of nano- and micro-structured CHT membranes on human keratinocyte stratification and differentiation. To this end, nanoporous and microporous membranes of chitosan (CHT) were prepared by phase inversion technique tailoring the operational parameters in order to obtain nano- and micro-structured flat membranes with specific surface properties. Microporous structures with different mean pore diameters were created by adding and dissolving, in the polymeric solution, polyethylene glycol (PEG Mw 10,000 Da) as porogen, with a different CHT/PEG ratio. The developed membranes were characterized and assessed for epidermal construction by culturing human keratinocytes on them for up to 21 days. The overall results demonstrated that the membrane surface properties strongly affect the stratification and terminal differentiation of human keratinocytes. In particular, human keratinocytes adhered on nanoporous CHT membranes, developing the structure of the corneum epidermal top layer, characterized by low thickness and low cell proliferation. On the microporous CHT membrane, keratinocytes formed an epidermal basal lamina, with high proliferating cells that stratified and differentiated over time, migrating along the z axis and forming a multilayered epidermis. This strategy represents an attractive tissue engineering approach for the creation of specific human epidermal strata for testing the effects and toxicity of drugs, cosmetics and pollutants.

In the gametophyte of the fern Anemia phyllitidis synthesis of linolenic acid esterified in monogalactosyldiglyceride requires light. By induction-reversion experim ents it could be demonstrated that this light-dependent step is mediated by phytochrome. There is also evidence for phytochrome control of galactolipid and hexadecatrienoic acid synthesis. In continuous blue light the synthesis of linolenic acid is inhibited and linoleic acid accumulates. It is concluded that the blue light photoreceptor affects an inhibition of linoleic acid desaturase. In continuous blue light chloroplasts contain abundant multilayered thylakoids, the grana regions are not as distinct as in white light, and membranes appear less appressed. In continuous red light the membranes are reduced in number and contain less grana-like appressions. It is concluded that both photoreceptors are necessary for a coordinate synthesis and assembly of the individual components of the chloroplast membrane.

Environmental contextPerchlorate from rocket fuel plants or firework manufacturing units can seriously contaminate drinking water. We developed a separation skin on a microfiltration membrane and on sand that can remove perchlorate from water in the presence of competing ions. This method is suitable for a domestic water purification unit selective for perchlorate removal. AbstractThis study reports an interactive separation of perchlorate (ClO4−) by polyethyleneimine (PEI) and poly (styrene sulfonate) (PSS) deposited on a microfiltration membrane and on sand surfaces. The variation of the interaction with respect to deposition and feed variables was assessed. The 9 bilayered ((PEI/PSS) 0.15M NaCl, pH 6)) membranes showed a ClO4− rejection of ~80%. An increase in the feed concentration to 25mgL−1 reduced the rejection to 58%. With a feed pH from 4 to 10, the rejection varied between almost 100% and 16%. The presence of ions reduced the rejection percentage of ClO4− with the interference by the ions in the order of SO42−&gt;HCO3−&gt;NO3−&gt;Cl−. The interference is attributed to the characteristics of the competing ions and the nature of the multilayers. A positive impact of post-treatment (98%) and capping layers on rejection percentage (80% to nearly complete) for synthetic and ClO4− contaminated field water samples is clearly established. The presence of competing ions is also accounted for by a capped membrane system. The selectivity of the competing ions increases with capping layers of 1M NaCl in the order of HCO3−&gt;NO3−&gt;SO42−. In the field water samples up to a SO42− concentration of 3.0mgL−1, the capping layers with 0.4M NaCl result in a near complete rejection of ClO4−, whereas further enhancement requires a capping layer of 1M NaCl. A sand filtration system was developed by incorporating the pre-optimised polyelectrolyte multilayer on sand. Deposition of a single bilayered PEI/PSS multilayer on sand effectively (nearly completely) removes ClO4−.

Elevated levels of certain membrane proteins, including the sterol biosynthetic enzyme HMG-CoA reductase, induce proliferation of the endoplasmic reticulum. When the amounts of these proteins return to basal levels, the proliferated membranes are degraded, but the molecular details of this degradation remain unknown. We have examined the degradation of HMG-CoA reductase-induced membranes in the fission yeast, Schizosaccharomyces pombe. In this yeast, increased levels of the Saccharomyces cerevisiae HMG-CoA reductase isozyme encoded by HMG1 induced several types of membranes, including karmellae, which formed a cap of stacked membranes that partially surrounded the nucleus. When expression of HMG1 was repressed, the karmellae detached from the nucleus and formed concentric, multilayered membrane whorls that were then degraded. During the degradation process, CDCFDA-stained compartments distinct from preexisting vacuoles formed within the interior of the whorls. In addition to these compartments, particles that contained neutral lipids also formed within the whorl. As the thickness of the whorl decreased, the lipid particle became larger. When degradation was complete, only the lipid particle remained. Cycloheximide treatment did not prevent the formation of whorls. Thus, new protein synthesis was not needed for the initial stages of karmellae degradation. On the contrary, cycloheximide promoted the detachment of karmellae to form whorls, suggesting that a short lived protein may be involved in maintaining karmellae integrity. Taken together, these results demonstrate that karmellae membranes differentiated into self-degradative organelles. This process may be a common pathway by which ER membranes are turned over in cells.

Ultrapure water production is vital for sustainable green hydrogen production by electrolysis. The current industrial process to generate ultrapure water involves energy-intensive processes, such as reverse osmosis. This study demonstrates a facile method to produce ultrapure water from simulated seawater using a low capital cost and low-energy-consuming membrane distillation (MD) approach that is driven by the waste heat from photovoltaic (PV) panels. To optimize the PV-MD operation, modeling efforts to design a multilayered MD system were carried out. The results were used to guide the construction of several prototype devices using different materials. The best performing PV-MD device, containing evaporation and condensation regions made from steel sheets and polytetrafluoroethylene (PTFE) membranes, can produce high-purity water with conductivity less than 40 mS and flux higher than 100 g/m2 h, which is suitable for typical electrolyzer use.
