A PCR procedure based on the intergenic region (IR) separating two genes encoding a recently identified mycobacterial two-component system, named SenX3-RegX3, was developed and was shown to be suitable for identifying Mycobacterium bovis BCG. ThesenX3-regX3 IR contains a novel type of repetitive sequence, called mycobacterial interspersed repetitive units (MIRUs). All tested BCG strains exclusively contained 77-bp MIRUs within the senX3-regX3 IR, whereas all non-BCGM. tuberculosis complex strains contained a 53-bp MIRU, in addition to the 77-bp MIRUs. All 148 strains analyzed so far could be divided into eight different groups according to the copy numbers of the 77-bp MIRU and to the presence or absence of the 53-bp MIRU. BCG strains contained either one, two, or three 77-bp MIRUs. The other strains contained one to five 77-bp MIRUs invariably followed by a 53-bp MIRU. The consistent absence of the 53-bp MIRU in BCG strains and its presence in virulent strains allowed us to develop an enzyme-linked immunosorbent assay using specific capture oligonucleotide probes to distinguish between BCG and other M. tuberculosis complex strains.

Using 243 Mycobacterium tuberculosis isolates obtained in 2001 in Osaka City, Japan, the discriminatory power of variable numbers of tandem repeats (VNTRs) of 12 standard mycobacterial interspersed repetitive units (MIRUs) was assessed. The biggest cluster defined by MIRU-VNTRs consisted of 57 (23.5 %) isolates and they belonged to the Beijing family based on spoligotyping. When additional VNTR loci were included in the MIRU-VNTR analysis, the 57 originally clustered strains were further differentiated by the addition of Queen's University Belfast (QUB)-VNTRs, but not exact tandem repeat-VNTR. The allelic diversity of additional VNTR loci such as VNTR 3232 (QUB-3232), VNTR 2163a (QUB-11a), VNTR 2163b (QUB-11b) and VNTR 1982 (QUB-18) was high in the 57 strains. When the 243 M. tuberculosis isolates were analysed using 16-locus VNTR (the 12 standard MIRUs and the 4 QUB loci) and IS6110 RFLP, the respective Hunter–Gaston discriminatory indexes were 0.9966 and 0.9971. The discrimination power of 16-locus VNTR was equal to that of IS6110 RFLP analysis. If appropriate loci are added to the standard MIRU analysis, VNTR genotyping could be a valuable tool for strain typing and epidemiological research of M. tuberculosis in Japan.

The successful use of DNA amplification for the detection of tuberculous mycobacteria crucially depends on the choice of the target sequence, which ideally should be present in all tuberculous mycobacteria and absent from all other bacteria. In the present study we developed a PCR procedure based on the intergenic region (IR) separating two genes encoding a recently identified mycobacterial two-component system named SenX3-RegX3. The senX3-regX3 IR is composed of a novel type of repetitive sequence, called mycobacterial interspersed repetitive units (MIRUs). In a survey of 116Mycobacterium tuberculosis strains characterized by different IS6110 restriction fragment length polymorphisms, 2 Mycobacterium africanum strains, 3 Mycobacterium bovis strains (including 2 BCG strains), and 1Mycobacterium microti strain, a specific PCR fragment was amplified in all cases. This collection included M. tuberculosis strains that lack IS6110 ormtp40, two target sequences that have previously been used for the detection of M. tuberculosis. No PCR fragment was amplified when DNA from other organisms was used, giving a sensitivity of 100% and a specificity of 100% in the confidence limit of this study. The numbers of MIRUs were found to vary among strains, resulting in six different groups of strains on the basis of the size of the amplified PCR fragment. However, the vast majority of the strains (approximately 90%) fell within the same group, containing two 77-bp MIRUs followed by one 53-bp MIRU.

The thelytokous egg parasitoid Ooencyrtus mirus Triapitsyn and Power (Hymenoptera: Encyrtidae) was recovered from brassica plant debris in Pakistan in an effort to find a biological control agent of the invasive bug Bagrada hilaris (Burmeister) (Heteroptera: Pentatomidae) in North America. As the first step in determining the overall host range of this parasitoid, adult females were exposed to the eggs of eight alternate pentatomid host species, two non-pentatomid heteropterans, and two lepidopterans, in choice and no-choice tests. Although O. mirus was more successful on B. hilaris than the other species in terms of the number of the eggs laid, the number of emerged progeny, and the developmental time of the progeny, it was able to reproduce on all of the alternate hosts except for one of the lepidopterans, whose eggs appeared too small for this parasitoid. The results show O. mirus to be a generalist parasitoid species with a preference for B. hilaris. The results also indicate that there is a linear relationship between the mean body length of O. mirus females and the mean host egg weight with an adjusted R 2 of 0.90. The implications of this study on the release of O. mirus for the control of B. hilaris are discussed.

The highly successful pathogen Mycobacterium tuberculosis (Mtb) has evolved strategies to adapt to various stress conditions, thus promoting survival within the infected host. The two-component regulatory system (2CRS) senX3-regX3, which has been implicated in the Mtb response to inorganic phosphate depletion, is believed to behave as an auto-regulatory bicistronic operon. Unlike other 2CRS, Mtb senX3-regX3 features an intergenic region (IR) containing several mycobacterium interspersed repetitive units (MIRU) of unknown function. In this study, we used a lacZ reporter system to study the promoter activity of the 5′ untranslated region of senX3, and that of various numbers of MIRUs in the senX3-regX3 IR, during axenic Mtb growth in nutrient-rich broth, and upon exposure to growth-restricting conditions. Activity of the senX3 promoter was induced during phosphate depletion and nutrient starvation, and IR promoter activity under these conditions was directly proportional to the number of MIRUs present. Quantitative reverse transcriptase (qRT)-PCR analysis of exponentially growing Mtb revealed monocistronic transcription of senX3 and regX3, and, to a lesser degree, bicistronic transcription of the operon. In addition, we observed primarily monocistronic upregulation of regX3 during phosphate depletion of Mtb, which was confirmed by Northern analysis in wild-type Mtb and by RT-PCR in a senX3-disrupted mutant, while upregulation of regX3 in nutrient-starved Mtb was chiefly bicistronic. Our findings of differential regulation of senX3-regX3 highlight the potential regulatory role of MIRUs in the Mtb genome and provide insight into the regulatory mechanisms underlying Mtb adaptation to physiologically relevant conditions.

Indian species of the genus Sakencyrtus Hayat are reviewed and illustrated. A new species, Sakencyrtus bialbifuniculus sp. nov., is described. The two species known from India, S. mirus Hayat and S. albiclavus Hayat, are redescribed. The macropterous forms and males of S. mirus are described for the first time. An identification key to species is provided.

Resumen
The True’s beaked whale (Mesoplodon mirus, True 1913) is a poorly known member of the Ziphiidae family. Its distribution in the northern hemisphere is thought to be restricted to the temperate or warm temperate waters of the North Atlantic, while a few stranding records from the southern hemisphere suggest a wider and antitropical distribution, extending to waters from the Atlantic coast of Brazil to South Africa, Mozambique, Australia and the Tasman Sea coast of New Zealand. This paper (i) reports the first molecular confirmation of the occurrence of the True’s beaked whale at the southern limit of its distribution recorded in the northeast Atlantic: the Azores and Canary Islands (macaronesian ecoregion); (ii) describes a new colouration for this species using evidence from a whale with molecular species confirmation; and (iii) contributes to the sparse worldwide database of live sightings, including the first underwater video recording of this species and close images of a calf. Species identification was confirmed in two cases using mitochondrial DNA control region and cytochromebgene markers: a subadult male True’s beaked whale that stranded in El Hierro, Canary Islands, in November 2012, and a subadult male found floating dead near Faial, the Azores, in July 2004. The whale that stranded in the Canary Islands had a clearly delimited white area on its head, extending posteriorly from the tip of the beak to cover the blowhole dorsally and the gular grooves ventrally. This colouration contrasts with previous descriptions for the species and it may be rare, but it exemplifies the variability of the colouration of True’s beaked whales in the North Atlantic, further confirmed here by live sightings data. The recording of several observations of this species in deep but relatively coastal waters off the Azores and the Canary Islands suggests that these archipelagos may be unique locations to study the behaviour of the enigmatic True’s beaked whale.

We here describe Merothrips meridionalis sp.n. based on specimens originally treated by J.D. Hood as an apterous form of M. mirus. The available data on the distribution of this new species suggest that it is widespread through Brazil, living in sympatry with M. floridensis, M. mirus and M. tympanis. However, the distribution of this new species is possibly further south than other Merothripidae species recorded from South America.

Australodelphis mirus (Delphinidae n. gen., n. sp) is a small extinct Early Pliocene dolphin known from five individuals from shallow-water strata of the Sørsdal Formation, Vestfold Hills, East Antarctica. Australodelphis mirus is the first higher vertebrate named from the Oligocene-Pleistocene interval on land in Antarctica, and is the first cetacean fossil from the polar margin of circum-Antarctic Southern Ocean that postdates the break-up of Gondwana. The dolphin is convergent in skull form with some living beaked whales (Mesoplodon spp.; Family Ziphiidae) in its long, narrow and toothless upper jaw and face, but skull suture patterns, basicranial sinuses, and ear-bones indicate close relationship with living long-beaked dolphins (Delphinidae). Australodelphis mirus perhaps was a suction-feeding squid-eater which occupied quiet near-shore shelf waters influenced by glaciers but probably lacking major sea-ice. Possible ecological equivalents of A. mirus (small ziphiids, long-beaked dolphins) do not occupy Antarctic waters today, perhaps excluded by cold conditions and/or sea-ice cover. Earlier Pliocene cetaceans worldwide reveal significant extinct and sometimes bizarre taxa, and extant families with ranges quite different from today, pointing to climate-related changes in cetacean ecology in the last 2–3 million years.

Das d-dimensionale Vektorpackproblem (d-VPP), welches aus Planungsaufgaben resultieren kann, ist eine Verallgemeinerung des eindimensionalen Zuschnittproblems (1CSP) und deshalb NP-schwer. Die stetige Relaxation, die mittels Spaltengenerierung gelöst werden kann, ergebe den optimalen Zielfunktionswert zC, während der optimale Zielfunktionswert der ganzzahligen Aufgabe zD ist. In der Dissertation werden obere Schranken für das Gap Δ = zD-zC hergeleitet und systematisch Instanzen des 1CSPs mit großem Δ (bis zu 6/5) konstruiert. Die im Teilbarkeitsfall des 1CSPs bekannte Abschätzung Δ < 2 wird zu Δ < 7/5 verschärft. Im d-VPP mit d > 1 gilt die MIRUP-Hypothese Δ < 2 nicht. Dies und die Unbeschränktheit des Wertes einer Variante bei d gegen unendlich werden an speziellen Beispielen gezeigt. Außerdem wird eine Heuristik vorgeschlagen und erprobt.
