Texto completoIn the first part of this thesis a numerical method based on a recently proposed up-scaled Poisson-Nernst-Planck type of equation (PNP) and its microscale counterpart is employed to estimate the tortuosity and thus the effective and apparent diffusion coefficients in thin charged membranes. Beside this, a new mathematical approach for estimation of tortuosity is applied and validated. This mathematical approach is also derived while upscaling of micro-scale Poisson-Nernst-Planck system of equations using the volume averaging method. A variety of different pore 2D and 3D micro-geometries together with different electrochemical conditions are studied here. To validate the new approaches, the relation between porosity and tortuosity has been obtained using a multi-scale approach and compared with published results. These include comparison with the results from a recently developed numerical method that is based on macro and micro-scale PNP equations.
Results confirm that the tortuosity value is the same for porous media with electrically uncharged and charged particles but only when using a consistent set of PNP equations. The effects of charged particles are captured by the ratio of average concentration to effective intrinsic concentration in the macroscopic PNP equations. Using this ratio allows to consistently take into account electro-chemical interactions of ions and charges on particles and so excludes any ambiguity generally encountered in phenomenological equations.
Steady-state diffusion studies dominate this thesis; however, understanding of transient ion transport in porous media is also important. The last section of this thesis briefly introduces transient diffusion through bentonite. To do so, the micro Nernst-Planck equation with electro-neutrality condition (NPE) is solved for a porous medium which consists of compacted bentonite. This system has been studied before in another research using an experimental approach and the results are available for both transient and steady-state phases. Three different conditions are assumed for NPE governing equations and then the numerical results from these three conditions are compared to the experimental values and analytical phenomenological solution. The tortuosity is treated as a fitting parameter and the effective diffusion coefficient can be calculated based on these tortuosity values. The results show that including a sorption term in the NPE equations can render similar results as the experimental values in transient and steady state phases. Also, as a fitting parameter, the tortuosity values were found varying with background concentration. This highlights the need to monitor multiple diffusing ion fluxes and membrane potential to fully characterize electro-diffusive transport from fundamental principles (which have been investigated in first part of this thesis) rather than phenomenological equations for predictive studies.

Texto completoOur knowledge of the diversity of marine microbial communities has long been restricted to the precious but incomplete information generated by the culture-based methods. In this study, molecular techniques (PCR, cloning, sequencing, hybridization, metagenomics library construction and genetic markers [16S rRNA, genes coding for enzymes specific to the reverse tricarboxylic acid cycle (acIB, oorA)] were used to circumvent the limits inherent to cultivation methods, and to obtain a more realistic view of the specific and functional diversity of the deep-sea hydrothermal vent microbial communities. This research allowed (i) to confirm the ecological significance of free-living epsilon-Proteobacteria at deep-sea hydrothermal vents, especially during in-situ colonization experiments (this study provides the first example of the prevalence and ecological significance of free-living Arcobacter-like at deep-sea hydrothermal vents, which are supposed to be sulfo-oxidizing bacteria involved in filamentous sulphur formation, (ii) to design and validate a 16S rRNA oligonucleotide probe targeting most of the epsilon-Proteobacteria found in hydrothermal systems, (iii) to obtain the physiology of yet uncultured groups of archea from deep-sea hydrothermal vents using metagenomics. By the combined use of variety of molecular approaches this work enlarges our view of the diversity of microbial communities in deep-sea hydrothermal vents

Texto completoNumerical investigations of an optimized thin airfoil with a passive and an active flow control device (riblets and rotary cylinder) have been performed. The objectives of the thesis were to investigate the tip vortices reduction using riblets and decrease in flow separation, using a rotary cylinder for improved lift-to-drag ratio. The investigations has application potentials in improving performances of Micro Air Vehicles (MAVs). The airfoil has a chord length of 19.66 cm and a span of 25 cm. with the free stream mean velocity was set at 20 m/s. The Reynolds number was calculated as 3 × 10 4. Investigations with base model of the airfoil have shown flow separation at approximately 85% chord length at an angle of attack of 17 degrees. For investigation using passive flow control device, i.e. riblets, investigations were performed for different radial sizes but at a fixed location. It was found that with 1 mm radial size riblet, the tip vortices were reduced by approximately 95%, as compared to the baseline model. Although negligible lift-to-drag improvement was seen, a faster dissipation rate in turbulent kinetic energy was observed. Furthermore, investigations were carried out using the active flow control device. The rotary cylinder with a 0.51 cm in diameter was placed slightly downstream of the location of flow separation, i.e. at x/c = 0.848. Investigations were performed at different cylinder's rotations, corresponding to different tangential velocities of being higher than, equal to and less than the free stream mean velocity. Results have shown approximately 10% improvement in lift to drag ratio when the tangential velocity is near the free stream mean velocity. Further investigation may include usage of the riblets and the rotary cylinder combined, to increase the stability as well as the lift-to-drag ratio of the MAVs.

Texto completoAlumina thin films were deposited on silicon substrate by chemical vapor deposition using an organometallic precursor (trimethylaluminium) introduced in the afterglow of oxygen microwave plasma. Columnar and oxygen over-stoichiometric films presented hydrogen content between 4 and 25 at. % which stabilized gamma alumina at the nanometric scale. An amorphous interphase (5 nm-thick) was detected at the substrate/coating interface. Parametrical study pointed out that lower pressure, higher substrate-holder RF bias and elevated substrate temperature improved the film quality. Correlations between the gas phase composition in radiative species and films properties were complex. Statistical tools were used in order to optimize the process. The influence of the most significant parameters (pressure, RF bias, temperature, substrate/injector distance) on the film properties was studied by mean of a response surface design of experiments. Finally, principal component analyses were conducted to identify quality markers in the gaseous phase

Texto completoIn the context of changes of general practice (uneven distribution of young general practitioners (GPs) across the country, strong feminisation), this thesis focuses on the determinants of the outpatient care supply of general practitioners. Using different micro-econometric analyses, firstly we want to understand the impact of feminization on the incomes of general practitioners and specifically on their private practice behaviours in terms of volume of care provided but also composition of the activity (consultations, home visits). Secondly, we study the determinants of the practice location choice within the region and we identify the levers that could improve the distribution of GPs in the area. Our results confirm that female GPs provide fewer services than male GPs and they also have a different composition of private practice activity. However, we show that male and female GPs adopt a similar practice location choice. Factors characterizing the place of installation as the characteristics associated with the supply of care, the demand for care, or equipment influence the practice location choice

Texto completoWith the population ageing, the expected increase in the long term care demand questions the role our societies want to entrust to family in the care provision for disabled elderly people. We use a micro-econometric framework to study individual and family caregiving behaviours. From a public policy perspective, three mains findings emerge from the analysis proposed. First, the identification of family interactions in individual caregiving decisions highlights the necessity to reconsider the idea of an inexorable decline in family support. Second, the decrease in labour supply induced by the care provision beyond a certain level points out the limits of a public policy aimed at both extending the work lives of seniors and encouraging informal care for disabled elderly people. Finally, the use au publicly funded formal care is associated with a rather modest decline in family support

Texto completoWe explored mechanisms involved in tolerance to organic xenobiotics belonging to PAHs (phenanthrene), in the context of allopolyploid speciation (hybrid genome duplication). We developed a comparative approach, using a recent allopolyploidization model including the hexaploid parental species S. alterniflora and S. maritima, and the allopolyploid S. anglica, which resulted from genome doubling of the F1 hybrid S. x townsendii. Integrative approach based on physiological and molecular analyses highlights that hybridization and genome doubling enhance tolerance to xenobiotics in Spartina. The paternal parent S. maritima exhibits higher sensitivity compared to the maternal parent S. alterniflora. Various transcriptomic analyses were performed, to identify de novo stress responsive transcripts, and to annotate small RNAs (miRNAs, their target genes, and siRNAs) involved in gene expression and transposable element regulations. Differential expression analyses in response to stress allowed us to develop a putative miRNA regulatory network (miRNA/target genes) in response to PAH, functionally validated in Arabidopsis as heterologous system. An exploratory profiling of Spartina rhizosphere microbiome exposed to phenanthrene was also performed to characterize environmental degradation abilities, in the perspective of optimizing green remediation strategies

Texto completoInfra Red femtosecond laser ablation coupled to Mass Spectrometry is a powerful tool for in-situ analysis of geological samples. Nevertheless, implications linked to this type of ablation are still not well defined. Transmission Electron Microscopy (TEM) coupled to Focused Ion Beam milling (FIB) allows direct characterization of ablation damage and particles. Thus, IR-femtosecond laser ablation can be considered as stoechiometric and mainly photo-mechanical. Particles are generated by coalescence and agglomeration of condensates in the laser induced plasma. Chemical segregation of elements noticed on measurements is then confined to these processes, themselves dependant on the target composition. The potential of analytical improvement is substantial, in terms of ablation settings and conception of laser systems dedicated to LA-ICP-MS

Texto completoThe detection and identification of pathogenic microorganisms is a real public health issue for the food industry and the clinics or national interest as illustrated in the biodefense field. Thus, it is important to have identification and detection methods that are fast, sensitive and robust. This PhD thesis aims at contributing to the development of a rapid approach to identify microorganisms without any a priori by tandem mass spectrometry. This innovative approach, called phylopeptidomics, is based on the combination of peptidomics, i.e. large scale analysis of peptides derived from the enzymatic digestion of a biological sample, and the phylogeny of cellular organisms. After extraction of the proteins from the sample of interest, peptides are generated and analyzed by tandem mass spectrometry. The deconvolution of MS/MS signals using the "μOrg.ID" software developed in the laboratory enables the identification and quantification of organisms present in the sample according to the organisms indexed in generalist databases. The study of the proteome of Bacillus atrophaeus, a simulant agent of anthrax, in sporulated and vegetative form, has provided an illustration of a new method of identification of protein biomarkers, which allows determining the ratio between both forms. The limit of detection of phylopeptidomics in pure samples and equimolar mixtures was established with model bacteria of medical and environmental interests. The limit of detection of B. atrophaeus spores in the presence of 14 interfering matrices (food, environmental and others) has highlighted the advantages and limitations of the approach. Finally, a standardized artificial mixture of 24 organisms was developed in order to evaluate bioinformatics tools in metaproteomics

Texto completoArtificial reefs are submerged structures deliberately placed on the seabed to mimic some characteristics of a natural habitat, they have been used for thousands of years by fishermen to improve their fishing practices and recently they are also used for the protection or even the restoration of certain marine habitats. Despite these various objectives, there is still a lack of scientific fundaments for determining the quality of these submerged structures in order to assess their effectiveness. The present works, focused on the study of their intrinsic characteristics, namely their materials and their structures. A combined monitoring of the biofilm and the macrofouling on different substrates, supplemented by an analysis of their heavy metal content allowed to highlight the quality of certain substrates used for the construction of artificial reefs. Simple monitoring of the biofilm has proven to be an efficient indicator for highlighting these differences to validate the use of a particular substrate.In addition, the development of a method to assess the complexity and heterogeneity of artificial reefs has enabled us to give a relevant classification of the various existing structures. This standardized method will allow to assess in situ the influence of structural parameters of artificial reefs on recruitment, abundance, distribution and/or diversity. A first study at the microscopic scale revealed the influence of substrate structure on both the abundance of biofilm and the photosynthetic activity of these communities.A more precise scientific framework is now available to guide the construction of artificial reefs on the choice of materials and the design of artificial reef structures in order to optimize the effectiveness of artificial reef immersion project

Texto completoTumors are composed not only of malignant cells but also contain a vast variety of non-malignant cells, notably immune cells forming the tumor microenvironment (TME). The composition of the TME was shown to be associated with clinical outcome for cancer patients, in terms of survival and therapeutic responses. With the relatively recent development of immunotherapies targeting specific elements of the TME, tumor immunology has risen a strong interest and holds a strong therapeutic potential. Several methodologies have been developed to study the composition of the TME with an increased precision. Notably, some methods such as MCP-counter enable the use of the tumor bulk transcriptome to quantify cell populations composing the TME. The methodological aspect of this PhD project consisted in setting up an enhanced version of MCP-counter that can be readily applied to RNA-Seq data, as well as propose an adaptation of the method for mouse models. Using MCP-counter, the TME of large series of tumors can be easily analyzed. The application part of this PhD work consisted of applying MCP-counter to establish an immune-based classification of soft-tissue sarcoma, a rare, aggressive and heterogeneous cancer type. The immune classification notably allowed to identify immune low and high groups, and a group characterized by a strong vasculature. Interestingly, the classification was notably found to be predictive of the patients' response to immunotherapies. It also highlighted an important role of tertiary lymphoid structures (TLS). TLS are lymph-node-like structures composed of T and B cells that form within the tumor or in close proximity. They are a site of formation and maturation of antitumoral immune responses. TLS are raising a growing interest in many malignancies. In most cancer types, a strong infiltration by T cells, in particular CD8+ T cells, is associated with a favorable clinical outcome. However, clear-cell renal cell carcinoma and prostate cancer are exceptions to this general rule. Indeed, in these urological cancers, an increased infiltration by T cells is associated with a decreased patient survival and with earlier relapse and disease progression. In a third part of this thesis, these exceptions are investigated with more details by scrutinizing the TME, and questioning the implication of the complement system. Overall, this thesis presents how the combination of several analysis methods, in silico, in situ and in vivo, can help achieve an extremely precise description of the TME. Knowing accurately what cell populations and what their functional orientation can help guide patients care and improve clinical outcome. Usually, painting fragments are prepared as polished sections and analyses are carried out on the cross-section surface. This sample preparation is easy to handle, however ends into critical constraints regarding feasibility and efficiency of micro-analyses. We propose to explore a different strategy: preparation and analysis of thin sections. These preparation procedures were first optimized and validated with µFTIR, µXRF, µXRD and µXANES. Besides, new methodological capabilities based on full-field/µXANES were assessed. Paintings were ideal candidates for estimating pros and cons of this new strategy for CH materials in general

Texto completoThis work investigated the performance of a dissolved air flotation (DAF) pilot plant, used to treat drinking water containing 05 Tu and 50 Cu. The particles (micro-bubbles and flocs) and hydraulic characteristics were taken into account. The focus was given in the reaction zone as hypothesized by Reali\'s mathematic model (REALI, 1991). An image analyses was used to assess particles distribution sizes. The hydraulics of the DAF tank was assessed by using a pulse stimulus-response test combined with the three-dimensional flow analyses carried-out with equipment that applied ultra sound (microADV). Therefore, two methods were developed; one for the acquisition, treatment and also to obtain the micro-bubbles and flocs sizes distribution, without the need of extracting samples and another, involved the development of a software (VelDigital3D) in order to treat the microADV data. Initially, the application of the microADV in the DAF process was evaluated. After, the data collection was made and the result was treated by using the VelDigital3D software. Sequentially, the effects of some agents for micro-bubbles coalescence after the releasing point were investigated: i) mixture conditions in terms of hydraulic loading rate (HLR) and detention time (DT); ii) recirculation rate (p); iii) pH variation and, iv) coagulant dosage (\'AL POT.+3\'). Finally, the mathematic model behavior proposed by Reali (1991) was investigated by varying some parameters in the design, such as: detention time in the reaction zone (DTr.z) and hydraulic loading rate in the clarification zone HLRc.z (dowflow); and by varying some operational parameters, such as: particles sizes distribution (micro-bubbles and flocs) and recirculation rate (p). The main conclusions were: i) the microADV probe can be used to obtain the velocity flow profile in water containing micro-bubbles; ii) the flow within the reaction zone showed a well defined pattern of recirculation throughout the height of the unit, confirming the obtained results by using the stimulus-response tests; iii) the variation of micro-bubbles medium size was low (from 20 to 30 \'mü\'m) but some points regarding this aspect were discussed and, iv) the behavior of the mathematical model proposed by Reali (1991) showed a significant adjustment to the experimental data proving that it can be applied to analyzed design parameters.

Texto completoThis thesis is devoted to the study of resonance frequencies of bidimensional optical cavities. More specifically, we are interested in whispering-gallery modes (modes localized along the cavity boundary with a large number of oscillations). The first part deals with the numerical computation of resonances by the finite element method using perfectly matched layers, and with a sensibility analysis in the three following situations: an unidimensional problem, a reduction of the rotationally invariant bidimensional case, and the general case. The second part focuses on the construction of asymptotic expansions of whispering-gallery modes as the number of oscillations along of boundary goes to infinity. We start by considering the case of a rotationally invariant problem for which the number of oscillations can be interpreted as a semiclassical parameter by means of an angular Fourier transform. Next, for the general case, the construction uses a phase-amplitude ansatz of WKB type which leads to a generalized Schrödinger operator. Finally, the numerically computed resonances obtained in the first part are compared to the asymptotic expansions made explicit by the use of a computer algebra software

Texto completoThe acidophilic Acidithiobacillus bacteria play an important role in industrial biomining operations for metal recovery. In this thesis, the genome sequence of a psychrotolerant Acidithiobacillus ferrivorans CF27 were first refined. The comparative genome analysis between CF27 and the closely related Acidithiobacillus genomes revealed: (i) a syntenic conservation of two tRNA array units which are only present in At. ferrivorans CF27 and At. ferrooxidans ATCC 23270 genomes and mainly contribute to the tRNA gene redundancy in both organisms. Moreover, our large-scale genome survey of the tRNA array units in prokaryotic organisms showed that tRNA arrays appear in few phyla; (ii) a high proportion of species-specific genes in CF27 and SS3 strains indicated the high variability of gene content in Acidithiobacillus genomes and therefore the unique nature of each group of species. Given that mRNA expression of some CF27 specific genes were confirmed in Fe(II)-grown cells and sulfur attached cells in CF27, these results highlighted the functional importance of specific genes for CF27 lifestyle; and (iii) the mosaic taxonomic composition of members of the Acidithiobacillia class, and thus confirmed that this group belongs to a particular taxonomic class, distinct to other proteobacterial groups. Taken together, our results provide insights into At. ferrivorans lifestyle as well as the chimeric genome nature of the Acidithiobacillus organisms. Texto completoThe Micro-expressions (MEs) are very important nonverbal communication clues. However, due to their local and short nature, spotting them is challenging. In this thesis, we address this problem by using a dedicated local and temporal pattern (LTP) of facial movement. This pattern has a specific shape (S-pattern) when ME are displayed. Thus, by using a classical classification algorithm (SVM), MEs are distinguished from other facial movements. We also propose a global final fusion analysis on the whole face to improve the distinction between ME (local) and head (global) movements. However, the learning of S-patterns is limited by the small number of ME databases and the low volume of ME samples. Hammerstein models (HMs) are known to be a good approximation of muscle movements. By approximating each S-pattern with a HM, we can both filter outliers and generate new similar S-patterns. By this way, we perform a data augmentation for S-pattern training dataset and improve the ability to differentiate MEs from other facial movements. In the first ME spotting challenge of MEGC2019, we took part in the building of the new result evaluation method. In addition, we applied our method to spotting ME in long videos and provided the baseline result for the challenge. The spotting results, performed on CASME I and CASME II, SAMM and CAS(ME)2, show that our proposed LTP outperforms the most popular spotting method in terms of F1-score. Adding the fusion process and data augmentation improve even more the spotting performance

Texto completoAtmospheric pressure dielectric barrier discharge are more and more used in industry, in particular for ozone generation, surface treatment, material deposit, treatment of pollutants, production of ultraviolet radiation in excimer lamps, etc… At such high pressures, these discharges are generally filamentary, but under certain particular conditions they can be homogeneous. However, the reasons of the transition from one type to another are still badly known. Contrarily to the filament discharges, the homogeneous ones have been widely studied. So, for a better understanding of the behavior of filament discharges in rare gases, a new experimental device was designed in order to carry out the electrical and spectroscopic temporal analysis of a monofilament. The present study deals with pure argon and xenon. The temporal evolutions of both the current flowing through and the voltage drop across the micro-discharge, were calculated by using a macroscopic dynamic electrical model. The injected electric power as well as the ignition voltage was then deduced for various conditions. The establishment of the discharge was recorded: different phases namely, the initial electronic avalanche, followed by the positive streamer and then the formation of the cathode layer were clearly illustrated by short exposure-time snapshots in the visible range. .

We gave an example where the region of damping is at the borderline of the geometric control condition, yet the damped waves exhibit a uniform exponential decay of energy, generalizing an example of Lebeau.In [2] we studied the controllability of the gravity-capillary water wave equation. Under the geometric control condition, we proved in arbitrary spatial dimension the exact controllability for spatially periodic small data. This generalizes a result of Alazard, Baldi and Han-Kwan for the 2D gravity-capillary water wave equation.In [3] we studied the propagation of singularities for the gravity-capillary water wave equation. We defined the quasi-homogeneous wavefront set, generalizing the wavefront set of H¨ ormander and the homogeneous wavefront set of Nakamura, and proved propagation results for quasi-homogeneous wavefront sets by the gravity-capillary water wave equation. As corollaries, we obtained local and microlocal smoothing effects for gravity-capillary water waves with sufficient spatial decay

Texto completoIn the context of household energy provision in the UK, thermodynamic and related carbon analyses of three micro-generation technologies have been carried out. These studies contribute to the research of the SUPERGEN ‘Highly Distributed Power Systems’ Consortium, which has been addressing a broad range of issues regarding micro-generation. The technologies analysed here are a grid-tied micro-wind turbine (rotor diameter 1.7m, rated power 600 W at 12 m/s), a grid-tied solar photovoltaic array (15 m2, 2.1 kWp mono-crystalline silicon), and a solar hot-water system (2.8 m2 flat-plate collector, direct-feed system). Annual energy outputs were estimated and contextualised against the demands of representative UK households. The overall energy-resource and carbon savings provided by the micro-generators were assessed on the basis that they (partially) displace the established supply systems. Savings were then compared with the energy-resource and carbon ‘debts’ of the micro-generators to determine their net performance.
The displaced energy or carbon payback periods of the micro-generators were estimated to be well within their estimated lifetimes: a maximum 2.5 years for the SHW system, 3.1 years for the micro-wind turbine installed in an ‘open’ environment, and 7.4 years for the solar PV system. After payback, net energy-resource and carbon savings accrue. This thesis thus demonstrates that, given appropriate UK installations, all three micro-generators can reduce carbon emissions and enhance energy security by reducing use of, and dependence upon, fossil fuels.
