Three near-infrared emitters (2TPA-QBT, 2MeTPA-BT and TPA-QBT-MeTPA) were rationally designed and synthesized. Density functional theory (DFT) and time-dependent density functional theory (TDDFT) calculations showed that the introduction of mono- or di-methyl groups between the donors and acceptor could result in the spatial configuration changing greatly for 2MeTPA-QBT and TPA-QBT-MeTPA compared to their parent compound 2TPA-QBT. The emission of TPA-QBT-MeTPA had a more obvious hybridized local and charge transfer feature (HLCT) based on the influence of the steric hindrance of the methyl substituent. Attributed to their different spatial configurations and luminescence mechanisms, different emission wavelengths with photoluminescent quantum yields of 26%, 38% and 34% in toluene, as well as 24%, 27% and 31% in 4,4′-bis(N-carbazolyl)-1,1′-biphenyl (CBP) doped film, were observed for 2TPA-QBT, 2MeTPA-QBT and TPA-QBT-MeTPA, respectively. The constructed organic light-emitting devices (OLEDs) displayed electroluminescence with emission peaks at 728, 693 and 710 nm, with maximum external quantum efficiencies of 1.58%, 1.33% and 3.02% for the 2TPA-QBT, 2MeTPA-QBT and TPA-QBT-MeTPA-doped OLEDs, respectively. This work illustrated the effect of spatial configuration changes on the luminescence properties of donor-acceptor-type organic emitters.

We investigated differences in physical activity (PA) levels between black and white South African 9-yr-old children and their association with bone mineral content (BMC) and density (BMD) by using dual-energy X-ray absorptiometry. PA was analyzed in terms of a metabolic (METPA; weighted metabolic score of intensity, frequency, and duration) and a mechanical (MECHPA; sum of all ground reaction forces multiplied by duration) component. There were significant ethnic differences in patterns of activity. White children expended a significantly greater energy score (METPA of 21.7 ± 2.9) than black children (METPA of 9.5 ± 0.5) ( P < 0.001). When children were divided into quartiles according to the amount and intensity of sport played, the most active white children (using METPA scores) had significantly higher whole body BMD and higher hip and spine BMC and BMD than less active children. White children in the highest MECHPA quartile also showed significantly higher whole body, hip, and spine BMC and BMD than those children in the lowest quartile. No association between exercise and bone mass of black children was found. In this population, PA has an osteogenic association with white children, but not black children, which may be explained by the lower levels of PA in the black children. Despite this, black children had significantly greater bone mass at the hip and spine (girls only) ( P < 0.001) even after adjustment for body size. The role of exercise in increasing bone mass may become increasingly critical as a protective mechanism against osteoporosis in both ethnic groups, especially because the genetic benefit exhibited by black children to higher bone mass may be weakened with time, as environmental influences become stronger.

Resumen
Tender coconut water tastes sweet and is enjoyed by consumers, but its commercial development is restricted by an extremely short shelf life, which cannot be explained by existing research. UPLC-MS/MS-based metabolomics methods were used to identify and statistically analyze metabolites in coconut water under refrigerated storage. A multivariate statistical analysis method was used to analyze the UPLC-MS/MS datasets from 35 tender coconut water samples stored for 0–6 weeks. In addition, we identified other differentially expressed metabolites by selecting p-values and fold changes. Hierarchical cluster analysis and association analysis were performed with the differentially expressed metabolites. Metabolic pathways were analyzed using the KEGG database and the MetPA module of MetaboAnalyst. A total of 72 differentially expressed metabolites were identified in all groups. The OPLS-DA score chart showed that all samples were well grouped. Thirty-one metabolic pathways were enriched in the week 0–1 samples. The results showed that after a tender coconut is peeled, the maximum storage time at 4 °C is 1 week. Analysis of metabolic pathways related to coconut water storage using the KEGG and MetPA databases revealed that amino acid metabolism is one of the main causes of coconut water quality deterioration.

Cancers develop in complex microenvironments whose importance was emerging during last years. In fact, cancer microenvironment influences tumor progression and leads to the raising of chemotherapics resistance. Thus, a shift of the focus from cancer cells to cancer cells in their environment is crucial for studying the molecular and metabolic features of tumors in physiological contexts. Within the microenvironment, cancer associated fibroblasts (CAF) are attracting the attention of scientific community since, up to date, it is clear that they are the main component involved in the organization of tumor tissue and that they interact with cancer cells and affect their behavior. Importantly, stromal cell types within the microenvironment are genetically stable, contrary to the tumor counterpart that frequently shows genetic instability and seriously altered physiological mechanisms. Accordingly, they represent a valuable therapeutic target. For these reasons, in this work we investigated the interplay between prostate cancer cells and tumor microenvironment, focusing on CAFs' role and on alterations of cancer cells occurring due their presence. In the first part of the study we collected expression profile data from cultures and co-cultures as well as metabolic data in order to characterize prostate cancer cells (PC3), CAFs and normal fibroblasts and alterations occurring in these cells n co-culture conditions. We found deregulated genes, strictly related to activation of processes which ease conditions favoring tumor progression in PC3 cells and in CAFs. More, we found deregulated genes in CAFs which usually are described as deregulated in transformed cells from patients and studied in PC3s, highlighting the importance of considering the stromal component as playing a major role in driving tumor progression. Through analyses of metabolic data we shed light on metabolic needs of PC3s and CAFs and on the metabolic behavior adopted by CAFs to enhance PC3s' growth. In the second part of the study, the retrieved data were used to build a metabolic model which in turn was used to validate an algorithm we developed, Metpath, designed toto improvestudies of unknown pathways' alterations. The models we built, a single cell model for PC3 and S9 and one model representing interacting PC3 and CAF cells, showed to be able to recapitulate the main metabolic reactions occurring in the corresponding conditions and represent a valid starting ground on which refined models for further studies could be developed. Moreover, MetPathrevealed altered metabolic pathways in PC3 and in CAF, which deserve further studies to deepen the knowledge about the way they can confer advantages to tumor. Taken together, all the results obtained in this work demonstrate the importance of considering and studying the tumors not only as a set of cancer cells but as part ofa complex microenvironment which influences tumor's progression. Importantly, this workhighlights the importance of the modern approaches to cancer study. In fact, simulation studies on --omics integrated models, like the ones elaborated in this work, can really push forward research in the multidisciplinary field of cancer research.

Current understanding of potential toxicity of engineered nanomaterials to aquatic microorganisms is limited for risk assessment and management. Here we evaluate if the MetPLATE™ test can be used as an effective and rapid screening tool to test for potential aquatic toxicity of various metal-based nanoparticles (NPs). The MetPLATE bioassay is a heavy metal sensitive test based on β-galactosidase activity in Escherichia coli. Five different types of metal-based NPs were screened for toxicity: (1) citrate coated nAg (Citrate-nanosilver), (2) polyvinylpyrrolidone coated nAg (PVP-nAg), (3) uncoated nZnO, (4) uncoated nTiO2 and (5) 1-Octadecylamine coated CdSe Quantum Dots (CdSe QDs); and compared with their corresponding ionic salt toxicity. Citrate-nAg was further fractionated into clean Citrate-nAg, unclean Citrate-nAg and permeate using a tangential flow filtration (TFF) system to eliminate residual ions and impurities from the stock Citrate-nAg suspension and also to differentiate between ionic- versus nano-specific toxicity. Our results showed that nAg, nZnO and CdSe QDs were less toxic than their corresponding ionic salts tested, while nano- or ionic form of TiO2 was not toxic as high as 2.5 g L− 1 to the MetPLATE™ bacteria. Although coating-dependent toxicity was noticeable between two types of Ag NPs evaluated, particle size and surface charge were not adequate to explain the observed toxicity; hence, the toxicity appeared to be material-specific. Overall, the toxicity followed the trend: CdCl2 > AgNO3 > PVP-nAg > unclean Citrate-nAg > clean Citrate-nAg > ZnSO4 > nZnO > CdSe QDs > nTiO2/TiO2. These results indicate that an evaluation of β-galactosidase inhibition in MetPLATE™ E. coli can be an important consideration for rapid screening of metal-based NP toxicity, and should facilitate ecological risk assessment of these emerging contaminants.

Silver nanoparticles (AgNPs) synthesized using numerous types of coating materials may exhibit different toxicity effects. The study evaluated coating material dependent toxicity by selecting 3 types of AgNP synthesis methods with different coating materials (citrate, polyvinyl pyrrolidone, and branched polyethyleneimine, coated AgNPs as citrate-AgNPs, PVP-AgNPs, and BPEI-AgNPs respectively). Two acute aquatic toxicity tests were performed; 48hr D. magna and MetPLATE E. coli toxicity tests. Significantly different toxicity effects were observed in D. magna test exhibiting lethal median concentrations (LC50) for citrate-AgNPs, PVP-AgNPs, and BPEI AgNPs respectively as, 2.7, 11.2, and 0.57μg/L. Median inhibitory concentrations (EC50) for MetPLATE tests were 1.27, 1.73, and 0.31mg/L respectively with significant different toxicity effects. Silver ion fractions were detected in the range of 2.4-19.2% in tested NP suspensions. Study suggests the toxicity effects are due to the cumulative action of ionic and nanoparticle fractions in the suspensions.
