Rhizobium leguminosarum bv. viciae strains expressing different degrees of tolerance to metal stress were used in this work to study the basic mechanisms underlying heavy metal tolerance. We used various parameters to evaluate this response. The strains' growth responses under different Cd2+ concentrations were determined and we reported variation in Cd2+ tolerance. Total soluble protein content decreased drastically, revealing the toxic effects that intracellular Cd2+ imposes on cellular metabolism, but this decrease in protein content was particularly evident in sensitive and moderately tolerant strains. Tolerant strains presented the highest intracellular and wall-bound Cd2+ concentrations. Cd2+ induced increases in the expression of some specific proteins, which were identical in all tolerant strains. Glutathione levels remained unaltered in the sensitive strain and increased significantly in tolerant and moderately tolerant strains, suggesting the importance of glutathione in coping with metal stress. This work suggests that efflux mechanisms may not be the only system responsible for dealing with heavy metal tolerance. A clear correlation between glutathione levels and Cd2+ tolerance is reported, thus adding a novel aspect in bacteria protection against heavy metal deleterious effects.Key words: glutathione, heavy metal, protein expression, rhizobia, thiol quantification.

Five metal-tolerant endophytic isolates (Bipolaris sp. LF7, Diaporthe miriciae LF9, Trichoderma asperellum LF11, Phomopsis asparagi LF15, Saccharicola bicolor LF22), with known metal-tolerance attributes and biocontrol activities against Ganoderma boninense, were tested for growth-promoting activities independent of (in vitro) and associated with plants (height, weight, root mass and stem circumference) (in vivo). Results revealed that metal-tolerant endophytes did not significantly render benefit to host plants as plant growth was compromised by the presence of metals. Lower production of indole-acetic acid (0.74-21.77 μg mL-1), siderophores (8.82-90.26%), and deaminase activities of 1-aminocyclopropane carboxylic acid (3.00-69.2 μmol mg protein-1 hr-1) were observed.

Present study dealt with identification of some heavy metal tolerant bacteria from contaminated industrial soils of Dhaka Export Processing Zone (DEPZ) at Savar, tannery area at Hazaribagh and uncontaminated agricultural soils of Dhamrai and Kushtia in Bangladesh and determination of their tolerance to chromium (Cr6+) and cadmium (Cd2+). A total of 15 isolates from four soil samples were provisionally identified as different species of Bacillus, Micrococcus and Pseudomonas based on their morphological, physiological, and biochemical characteristics. Among them eight colonies were separated based on high level of heavy metal tolerance and identified at molecular level by PCR technique and 16S rRNA gene sequencing as Micrococcus luteus strain P43 (E4), Bacillus pocheonensis strain TR2-6 (T6), Bacillus megaterium strain H2 (T8), Bacillus amyloliquefaciens strain SCSAAB0007 (D10), Bacillus cereus isolate PGBw4 (D11), Bacillus cereus strain ES-4a1 (K12), Bacillus subtilis strain 1320, (K13), and Bacillus subtilis strain DP14 (K14). The Maximum Tolerable Concentration (MTC) of bacterial strains to Cr6+ and Cd2+ ranged between 250-1250 ?g/ml and 30-150 ?g/ml, respectively in nutrient broth medium. From the metal tolerance investigation Bacillus was found as the most heavy metal tolerant to both Cr6+ and Cd2+ among the three genera. Sugar industrial effluents possess high amounts of toxic pollutants and contaminate the receiving sites. Treatment of contaminated sites by using microorganisms provides an alternate to conventional methods hence demands in the identification of metal tolerant microorganisms has been increasing day by day. Therefore in this study soil samples collected from Tanuku sugar factory residual effluent point (bank of Gosthani river), west Godavari district A.P were analyzed for the bacterial tolerance to Copper (Cu), Zinc (Zn) and Lead (Pb) in their chloride forms. Additionally, the study was carried out to identify the metal tolerant bacteria by morphological, biochemical and 16S rRNA gene sequencing studies. Four potential bacterial isolates were selected to analyze metal tolerance against CuCl2, ZnCl2, and PbCl2. The sequences were compared with those in NCBI and submitted in gene bank with accession numbers MK100333 (Paenibacillus cookie), MK100334 (Bacillus cereus), MK100335 (Aneurini bacillus sp) and MK100387 (Paenibacillus sp.). A Phylogenetic tree was constructed to Paenibacillus sp. the highly efficient bacterial strain among the four isolates using MEGA 7 soft ware. The results of this study showed that P. dentritiformis had multiple metal tolerances (Cu, Zn and Pb) up to 500mg/L after 72 hrs. The identified bacterial strain proved to be the strong heavy metal tolerant bacterial strain. Hence, its usage will be helpful in the treatment of heavy metals specifically Cu, Zn and Pb contaminated soils and further optimization of these cultures is required to improve its metal resistant capacity.

Bryophytes are widely used to monitor air quality. Due to the lack of a cuticle, their cells can be compared to the roots of crop plants. This study aimed to test a hypothetical relation between metal tolerance and cell shape in biomonitoring mosses (Hypnum cupressiforme, Pleurozium schreberi, Pseudoscleropodium purum) and metal sensitive species (Physcomitrium patens, Plagiomnium affine). The tolerance experiments were conducted on leafy gametophytes exposed to solutions of ZnSO4, ZnCl2, and FeSO4 in graded concentrations of 1 M to 10−8 M. Plasmolysis in D-mannitol (0.8 M) was used as a viability measure. The selected species differed significantly in lamina cell shape, cell wall thickness, and metal tolerance. In those tested mosses, the lamina cell shape correlated significantly with the heavy metal tolerance, and we found differences for ZnSO4 and ZnCl2. Biomonitoring species with long and thin cells proved more tolerant than species with isodiametric cells. For the latter, “death zones” at intermediate metal concentrations were found upon exposure to ZnSO4. Species with a greater tolerance towards FeSO4 and ZnSO4 had thicker cell walls than less tolerant species. Hence, cell shape as a protoplast-to-wall ratio, in combination with cell wall thickness, could be a good marker for metal tolerance.

Biodiversitas 18: 1481-1486. The bacterial study is a part of human calling in preserving the earth. Many indigenous bacteria isolated from heavy metal contaminated sites had resistance to heavy metal toxicity and could be used for heavy metal removal. The aims of this study were to isolate heavy metal-tolerant indigenous bacteria from the industrial sewage of Kemisan River in Tangerang, Banten, Indonesia. The potency of bacterial isolates to remove heavy metals was also determined. The heavy-metal tolerance was determined by measuring the minimum inhibitory concentration. The potency of bacterial isolate for removing heavy metals from the medium was determined by an atomic absorption spectrophotometer. The results showed that there were eight heavy metal-resistant bacteria isolated from Kemisan River with minimum inhibitory concentration ranging from 7 mM to 11 mM. Isolate PbSI1 was the highest lead tolerant bacteria, and also tolerant to copper and zinc. The isolate was able to remove 91.25% lead, 73.38% zinc, and 98.57% copper from medium supplemented with the mixture of these heavy metals. The addition of 9 mM of lead in the medium affected the morphological appearance of isolate colonies i.e PbSI1 and PbSI3 to become darker which might occur due to the survival mechanism of bacteria by absorbing the lead inside the cells. The finding of this study indicated that isolate PbSI1 was a promising bacterium, which could be further developed for heavy metal removal.

Abstract Heavy metal contamination is an increasingly pressing global ecological concern adversely affecting plant growth. Phytoremediation is an eco-friendly and low-cost approach to help solve this problem by using plants to remove metals. This study aimed to evaluate the phytoremediation potential of four Plantago species, exposing them to different concentrations (0, 150, 300, 600 and 900 ppm) of zinc, copper, and iron during germination and early seedling growth. These are generally the phases of the plant life cycle most sensitive to stress. The germination percentage (GP), mean germination time (MGT), radicle, hypocotyl and cotyledons length, biomass, water content and tolerance index (TI) were recorded under controlled conditions. The results indicated that metal-induced stress significantly reduced GP, increased MGT, and inhibited seedling growth with increased metal concentration. The relative toxicity of the tested metals could be ranked as Fe > Cu > Zn. Regarding the Plantago species, P. tunetana and P. lanceolata could be considered highly tolerant, P. albicans moderately tolerant, and P. afra low-tolerant to metal toxicity during germination. Therefore, P. tunetana and P. lanceolata present an excellent potential for phytoremediation of metal-contaminated zones.

This work aimed to evaluate density of associative diazotrophic bacteria populations in soil and grass root samples from heavy metal contaminated sites, and to characterize isolates from these populations, both, phenotypically (Zinc, Cadmium and NaCl tolerance in vitro, and protein profiles) and genotypically (16S rDNA sequencing), as compared to type strains of known diazotrophic species. Densities were evaluated by using NFb, Fam and JNFb media, commonly used for enrichment cultures of diazotrophic bacteria. Bacterial densities found in soil and grass root samples from contaminated sites were similar to those reported for agricultural soils. Azospirillum spp. isolates from contaminated sites and type strains from non-contaminated sites varied substantially in their in vitro tolerance to Zn+2 and Cd+2, being Cd+2 more toxic than Zn+2. Among the most tolerant isolates (UFLA 1S, 1R, S181, S34 and S22), some (1R, S34 and S22) were more tolerant to heavy metals than rhizobia from tropical and temperate soils. The majority of the isolates tolerant to heavy metals were also tolerant to salt stress as indicated by their ability to grow in solid medium supplemented with 30 g L-1 NaCl. Five isolates exhibited high dissimilarity in protein profiles, and the 16S rDNA sequence analysis of two of them revealed new sequences for Azospirillum.

Resumen

Mining is widespread and an economically important industry. Unfortunately, acid mine drainage (AMD) can pollute ecosystems with a cocktail of contaminants too complex for accurately forecasting its health consequences. However, through quantification of fundamental toxic events, the effects of complex mixtures can be observed. This project explored two potentially insightful and convenient endpoints. First, oxygen consumption (MO2), a well-established and sensitive indicator of respiratory impairment was utilized. Second, protein carbonyl content (PCC), an experimental ecological biomarker widely lauded in biomedical circles as a highly conserved indicator of health status was assessed for its utility in a metal tolerant aquatic macroinvertebrate, Arctopsyche grandis. A. grandis were exposed to eight environmentally relevant target concentrations (in duplicate) of AMD for eight days at a temperature controlled greenhouse containing artificial flow-through streams. As expected, MO2 was inversely related to treatment concentration (R 2=0.35, p=0.015). Protein carbonyl content, however, diverged from predictions. Protein carbonyl content analysis detected significantly more oxidative protein injury in control treatments than in metal-rich AMD treatments (p<0.001). Moreover, there was not a significant difference in PCC between different AMD concentrations. Protein carbonyl content’s departure from anticipated results likely is the consequence of dynamic interactions between direct and indirect effects at the chemical, biochemical, physiologic and behavioral levels. The results of this project illustrate flaws of utilizing a single biochemical marker to observe effects of a toxic mixture. Rather, a broad suite of biomarkers should be assayed to determine sublethal toxicity. These results also illustrates how multiple stressors can yield unanticipated outcomes.

There is an influx in heavy metals into soils and ground water due to activities such as increased mineral mining, improper watering and the use of heavy metal contaminated fertilizers. These heavy metals are able to increase the ROS species within plants which may result in plant metabolism deterioration and tissue damage. Heavy metals may also directly damage plants by rendering important enzymes non-functional through binding in metal binding sites of enzymes. The heavy metal focused on in this study was vanadium due to South Africa being one of the primary produces of this metal. Two related Brassica napus L cultivars namely Agamax and Garnet which are economically and environmentally important to South Africa were exposed to vanadium. Physiological experiments such as cell death, chlorophyll and biomass determination were conducted to understand how these cultivars were affected by vanadium toxicity. A low cost, sensitive and robust vanadium assay was developed to estimate the amount of vanadium in samples such as water, soils and plant material. The oxidative state as well as the antioxidant profile of the two cultivars were also observed under vanadium stress. A chlorophyll assay which was conducted on the two cultivars xiv exposed to vanadium showed a marked decrease in chlorophyll A in the suspected sensitive cultivar which was Garnet. However, the suspected tolerant cultivar Agamax fared better and the decrease in chlorophyll A was much less. A similar trend was observed for the two cultivars when the cell death assay was conducted. The vanadium assay showed that Garnet had higher concentrations of vanadium within its leaves and lower concentrations in its roots when compared to Agamax. This observation displayed that Agamax had inherent mechanisms which it used to localize vanadium in its roots and which assisted in its tolerance to the vanadium stress. The oxidative state was determined by doing assays for the specific reactive oxygen species namely hydrogen peroxide and superoxide. It was observed that vanadium treated Garnet leaves had higher reactive oxygen species (ROS) production when compared to the Agamax treated leaves. In-gel native PAGE activity gels were conducted to determine the antioxidant profile for the two cultivars which were exposed to vanadium. The antioxidant enzymes which were under investigation were ascorbate peroxide (APX), superoxide dismutase (SOD) and glutathione-dependent peroxidases (GPX-like) as these enzymes are known to be responsible for controlling the ROS produced in the plants. The GPX-like profile consisted of three isoforms. No isoforms were inhibited by vanadium treatments but one isoform had increased activity in both the Garnet and Agamax treated samples. The SOD profile for Garnet consisted of six isoforms xv and Agamax had seven isoforms. One isoform which was visualized in both Agamax as well as Garnet was inhibited by vanadium treatments. Agamax also had two isoforms which were up-regulated however the corresponding isoforms in Garnet showed no change. The Ascorbate peroxidase profile consisted of seven isoforms for both Garnet and Agamax. No isoforms were inhibited by vanadium treatment. Three isoforms were up-regulated in Garnet and Agamax under vanadium treatments. Here, it is illustrated that Garnet lacked certain mechanisms found in Agamax (and thus experienced more cell death, yield and chlorophyll loss) and performed worst under high vanadium concentrations. Although Garnet increased the activity of some of its antioxidant isoforms in response to increasing ROS levels it was not adequate to maintain a normal oxidative homeostasis. This disruption in oxidative homeostasis lead to plant damage. Agamax was observed to produce less ROS than Garnet and was able to control the ROS produced more effectively than Garnet and thus less damage was observed in Agamax.

The development of metal-tolerance and antibiotic resistance in bacteria may be caused by metals polluting a particular environment. During mining and mineral processing activities, large quantities of metals are deposited into the soil. These high concentrations of metals are evolutionary pressures selecting for microorganisms tolerant to these metals. Metaltolerance maybe conferred to these organisms by mobile genetic elements such as plasmids. This study describes the characteristics of plasmids isolated from various bacteria that displayed an ability to withstand high metal concentrations. The isolated plasmids were individually transformed into Escherichia coli JM109. Transformants were then evaluated for metal-tolerant capabilities using a microdilution approach. Plasmids were then isolated from the transformants and the concentration of the plasmid DNA ranged between 11.75 – 118.06 ng/μl. These plasmids were of the same size as the original ones. This demonstrated that successful transformations with plasmid DNA were conducted. In order to determine the compatibility group, plasmids were subjected to PCR amplification using IncQ, IncP-9 and IncW specific primers. Only the IncW provided positive results. To demonstrate that the plasmids were free of genomic DNA, a 16S rDNA PCR test was included. The plasmids that were positive for IncW PCRs were all negative for the rDNA PCRs. Plasmids were stably inherited and at least three, isolated from three different Gram positive species, belonged to the Inc W group of plasmids. These were originally isolated from Paenibacillus ginsingari, Paenibacillus lautus and Bacillus cereus. Minimum inhibition concentrations (MICs) were carried out to determine the ability of transformed E. coli JM109 to tolerate metals at varying concentrations. Results indicated that transformed E. coli JM109 developed ability to grow in the presence of several heavy metals. Some strains were resistant to high concentrations (+10 mM) of Ni2+/Al3+, Pb2+ and Ba2+. The order of metal resistance was Ni/Al=Pb>Ba>Mn>Cr>Cu>Co=Hg. All the x transformants were sensitive to 1 mM of Co2+ and Hg2+. Moreover, protein profiling was used to determine the impact of plasmids on E. coli JM109. Proteins were extracted from both transformed and un-transformed E. coli JM109 using acetone-SDS protocol and subjected to one-dimensional (1D) and two-dimensional (2D) Sodium Dodecyl Sulphate Polyacrylamide Gel Electrophoresis (SDS- PAGE). Transformed E. coli JM109 were grown under the metal stress. One dimension SDS-PAGE illustrated general similarity of the profiles except for two banding positions in the 30 to 35 kDa region where bands were present in the transformants that were grown in the Ni/Al alloy containing media. Twodimensional electrophoresis PAGE analysis showed that some of the proteins were upregulated while others were down-regulated. The largest numbers of proteins were from 15 – 75 kDa. The majority of these proteins had isoelectric points (pI) between 5 and 6. It was concluded that plasmids isolated from various heavy metal-tolerant bacterial species were successfully transformed into E. coli JM109 rendering various new metal-tolerant E. coli JM109 strains. Furthermore, the study showed that metal resistance was due to the presence of the plasmids. Two-dimensional SDS-PAGE resolved more differences in the protein expression profiles. Since the plasmids rendered the E. coli JM109 tolerant to metals tested, it also can be concluded that the change in the protein profiles was due to the effects of the plasmids. Furthermore, plasmids were also re-isolated from the transformants and these plasmids were of the same size as the original ones.. All the plasmids in this study were also stably inherited, a feature associated with IncW plasmids. More detailed genetic characterization of these plasmids is required. Plasmids isolated and characterized in this study may hold biotechnology potential. Such features should be exploited in follow-up experiments.

Resumen
Thermal barrier coatings (TBCs) are applied to hot gas path surfaces to reduce metal temperatures and thermal stresses for improved turbine component durability. Improvements in engine performance are achieved when TBCs enable reductions in cooling air usage and turbine blade tip-shroud clearances. This paper describes the development of thick TBCs with superior strain tolerance. A pattern of grooves or slant steps incorporated into the surface to be coated enables the development of shadow gaps in the ceramic layer during plasma spray deposition. These gaps segment the TBC, permitting ceramic-metal thermal expansion mismatch and thermal strains to be accommodated by free expansion.

Condition being studied: Immunotherapy is the classic treatment for allergic rhinitis. Intralymphatic immunotherapy is a new type of treatment. Streams at Glacier National Park are important because they have some of the cleanest water in the world, they offer awe-inspiring views, and these ecosystems are home to a globally significant community of animals, including imperiled species. This report summarizes trends in chemistry, habitat, and biology for Stream Ecological Integrity (SEI) monitoring at six sentinel stream and river sites in Glacier. From 2007 to 2019 we sampled four stream sentinel sites, three or four times annually. Two additional sites were added at request of Glacier in 2018 to augment information for important Wild and Scenic large rivers. Overall, the ecological integrity of sentinel stream sites in Glacier remained in a reference condition from 2007 to 2019. Our data do not suggest that the largely pristine water quality or biodiversity of these streams has changed in any profound way. Nevertheless, our data may be beginning to reveal some emergent issues. Climate directly affects water temperature, which increased 1 to 1.5 ?C per decade at four of Glacier?s sentinel stream sites between 2007 and 2019. We saw the effect of warmer stream water on stream macroinvertebrates, with increasing warm water tolerant species over the period of our monitoring. Trace metals in streambed sediments increased or were higher than expected, including copper and zinc at Swiftcurrent Creek, cadmium and arsenic at Reynolds Creek, and arsenic at the North Fork of the Flathead River at the border. Glacier?s sentinel stream physical habitat was largely in good condition and stable, except a low cover of invasive, exotic plants at most sites.

Objectives. This systematic review assesses postpartum care for individuals up to 1 year after pregnancy. We addressed two Key Questions (KQs) related to the comparative effectiveness and harms of: (1) alternative strategies for postpartum healthcare delivery and (2) extension of postpartum health insurance coverage. Data sources and review methods. We searched Medline®, Embase®, Cochrane CENTRAL, CINAHL®, and ClinicalTrials.gov from inception to November 16, 2022, to identify comparative studies in the United States and Canada (for KQ 1) and in the United States (for KQ 2). We extracted study data into the Systematic Review Data Repository Plus (SRDR+; https://srdrplus.ahrq.gov). We assessed the risk of bias and evaluated the strength of evidence (SoE) using standard methods. The protocol was registered in PROSPERO (registration number CRD42022309756). Results. We included 50 randomized controlled trials (RCTs) and 14 nonrandomized comparative studies (NRCSs) for KQ 1 and 28 NRCSs for KQ 2. Risk of bias was moderate to high for most RCTs and all NRCSs. KQ 1: Regarding where healthcare is provided, for general postpartum care (6 studies), whether the visit is at home/by telephone or at the clinic may not impact depression or anxiety symptoms (low SoE). For breastfeeding care (8 studies), whether the initial visit is at home or at the pediatric clinic may not impact depression symptoms up to 6 months postpartum, anxiety symptoms up to 2 months, hospital readmission up to 3 months (summary relative risk [RR] 1.38, 95% confidence interval [CI] 0.90 to 2.13; 4 studies), or other unplanned care utilization up to 2 months (low SoE, all outcomes). Regarding how care is provided, for general postpartum care (4 studies), integration of care (i.e., care provided by multiple types of providers) may not impact depression symptoms or substance use up to 1 year (low SoE). Regarding when care is provided, for contraceptive care (9 studies), compared with later contraception, earlier contraception start is probably associated with comparable continued IUD use at 3 and 6 months but greater implant use at 6 months (summary RR 1.36, 95% CI 1.13 to 1.64; 2 RCTs) (moderate SoE). Regarding who provides care, for breastfeeding care (19 studies), compared with no peer support, peer support is probably associated with higher rates of any breastfeeding at 1 month (summary effect size [ES] 1.13, 95% CI 1.03 to 1.24; 4 studies) and 3 to 6 months (summary ES 1.22, 95% CI 1.06 to 1.41; 4 studies) and of exclusive breastfeeding at 1 month (summary ES 1.10, 95% CI 1.02 to 1.19; 6 studies) but probably yields comparable rates of exclusive breastfeeding at 3 months and nonexclusive breastfeeding at 1 and 3 months (all moderate SoE). Compared with no lactation consultant, breastfeeding care by a lactation consultant is probably associated with higher rates of any breastfeeding at 6 months (summary ES 1.43, 95% CI 1.07 to 1.91; 3 studies) but not at 1 month or 3 months (all moderate SoE). Lactation consultant care may not be associated with rates of exclusive breastfeeding at 1 or 3 months (moderate SoE). Regarding coordination/management of care, provision of reminders for testing is probably associated with greater adherence to oral glucose tolerance testing up to 1 year postpartum but not random glucose testing or hemoglobin A1c testing (moderate SoE). Regarding use of information or communication technology (IT; 8 studies), IT use for breastfeeding care is probably associated with comparable rates of any breastfeeding at 3 months and 6 months and of exclusive breastfeeding at 3 months (all moderate SoE). Because of sparse evidence, inconsistent results, and/or the lack of reporting of prioritized outcomes, no conclusions related to interventions targeting healthcare providers are feasible (4 studies). KQ 2: Regarding health insurance (28 studies), more comprehensive health insurance is probably associated with greater attendance at postpartum visits (moderate SoE) and may be associated with fewer preventable readmissions and emergency room visits (low SoE). Conclusion. Most studies included in this systematic review enrolled predominantly healthy postpartum individuals. Researchers should therefore design studies that, either entirely or in part, enroll individuals at high risk of postpartum complications due to chronic conditions, pregnancy-related conditions, or incident or newly diagnosed conditions. New high-quality research is needed, especially for interventions targeting healthcare providers and the impact of more comprehensive or extended health insurance on postpartum health. Patient-reported outcomes, such as quality of life, should also be reported. Researchers should report separate data for various population subgroups, which could help close gaps in health outcomes among the races of postpartum individuals in the United States.

Resumen
The first use of radiographic standards for inspection and acceptance of finished girth welds was adopted by API in 1953. This standard was largely based on the Unfired Pressure Vessel Code� which was adopted by ASME in 1931. At that time and to this day, weld defect acceptance standards are based on workmanship principles. In other words, the so called "Workmanship defect acceptance standards"� reflect the weld quality levels produced by. a trained welder using satisfactory materials, equipment and procedures. For many years exclusive use was made of the API 1104 workmanship defect acceptance levels for all field pipeline and related facilities welds. This, and very similar defect acceptance standards (e.g. BS 4515, CSA 2184, AS 1697, etc.) are used in many countries throughout the world. Though the specified acceptance levels have no scientific basis, the use of workmanship standards has provided an adequate level of quality control for many years. Particularly worthy of attention in this context is the fact that the currently stipulated levels have been developed for welds in lower strength pipe grades. Unfortunately, the developments in acceptance levels have not kept pace with those occurring in-pipeline technology. Furthermore, pipe size, strength and toughness, weld metal properties or pipeline operating conditions are not considered in the present-day defect acceptance standards. Problems of this kind have led companies involved in gas/oil transporting activities to develop their own standard requirements or at least to produce a supplement to the general standards. This report describes the results of an investigation into the engineering significance of girth weld defect acceptance criteria based upon weld quality (or workmanship) considerations. To this end, research efforts were divided into a theoretical and an experimental part. The theoretical study involved a comparison of internationally used weld quality standards and codes for pipeline welding in order to identify the general features common to them and to obtain a clear view of the different acceptability criteria. The experimental part was designed to provide factual information on the failure behaviour of defective girth welds in large pipe diameter pipe lines. The focal point of the experimental examinations was to compare, on the basis of wide plate test specimen behaviour, the performance levels of girth welds containing planar weld defects which were grossly out of tolerance with respect to most present-day weld quality (workmanship standards) specifications.
