Resumen
During a natural outbreak of hepatic fatty cirrhosis (HFC) in western Texas, 500 2-6-year-old Rambouillet ewe sheep were sequentially studied to determine the pattern of lesion development. All sheep developed lesions of HFC. Grossly, changes first began in the subcapsular hepatic parenchyma along the porta hepatis and spread peripherally until, in the final stages of the disease, approximately 80% of the liver was affected. Ascites, hydropericardium, and acquired hepatic vascular shunts were present in sheep with severe HFC. Light microscopic lesions initially appeared as accumulations of fine lipid droplets in the cytoplasm of periacinar hepatocytes but, with time, involved all hepatocytes of the lobule. The fat vacuoles in the periacinar hepatocytes coalesced to form larger vacuoles; and after rupture of adjacent fat-laden hepatocytes, fatty cysts appeared. Fibrosis began in the periacinar zone associated with the ruptured fatty cysts and continued until there was widespread bridging periacinar fibrosis. Islands of regenerating hepatocytes were frequently sequestered within the bands of fibrous tissue. Characteristically, the hepatic and posterior mediastinal lymph nodes, lung, and spleen contain ceroid. No lesions of hepatic encephalopathy were found in any animal. HFC is a progressive, chronic disease of sheep, and the morphology of the hepatic lesions is similar to lipotrope-deficient forms of nutritional cirrhosis. These findings are discussed in relation to similar nutritional deficiencies and toxicoses in sheep.

Abstract. Acute physical exercise stimulates the activity of the hypothalamus-pituitary-adrenal axis in man. In the present study we measured plasma adrenocorticotropin, beta-endorphin, beta-lipotropin and cortisol levels in 27 male trained athletes in basal conditions, 60 min before and immediately after an official competition. The endocrine responses were evaluated in different groups of athletes participating in races (100 m, 1500 m, 10 000 m) or in the disc throw. The athletes competing for the runs showed a statistically significant increase in plasma adrenocorticotropin, beta-endorphin, beta-lipotropin and cortisol levels after the race (P <0.01), whereas the disc throwers showed no significant change in the hypothalamus-pituitary-adrenal axis hormones after the competition. The percent increase in plasma adrenocorticotropin, beta-endorphin, beta-lipotropin and cortisol was higher in the athletes who run 1500 m and 10 000 m than in those participating in the short distance race (100 m). The present results showed that plasma proopiomelanocortin-related peptides and cortisol levels increase in trained athletes following running competition and that this increase is related to the duration of the physical exercise.

In women during perimenopause, changes in metabolic parameters are detected, such as weight gain, which triggers a cascade of pathological reactions leading to the formation of metabolic disorders (lipid, carbohydrate metabolism) and endothelial dysfunction, during the development of which a deficiency of sex hormones plays a certain role in the onset of menopause. There is a connection between hormonal changes and an increase in the amount of abdominal and/or visceral fat, which is accompanied by a weakening of oxidative processes in adipose tissue, a decrease in energy consumption and a predisposition to the development of metabolic syndrome. As a nutritional correction, it is advisable to introduce into the diet lipotropic substances that help catalyze the breakdown of fats and ensure the processes of hydroxylation and methylation of toxic substances in the liver, such as methionine, inositol and choline. Each of the lipotropic substances affects the mobilization of fats, and their combination can provide a synergistic effect and increase the natural metabolic function of the liver.

Searchingof drugs that normalize the function of the digestive glands are important because disruption of digestion underlie the pathogenesis of many diseases such as gastroesophageal reflux disease, hepatitis, gastric ulcer, irritable bowel syndrome, cancer etc. The use of herbal objects containing fibres is a promising direction for solving of the mentioned problem. We were attracted by fruits of Prunus domestica which are rich in fibres (homo- and heteropolsaccharides) and are used in folk medicine as a laxative and hepatoprotective agent. The aim of this experimental study was to investigate the lipotropic properties of «Prunofit» extract. The study of lipotropic properties of the «Prunofit» was carried out in the conditions of subacute toxic liver damage caused by the introduction of ethanol.The content of total lipids (TL), total cholesterol (TCh), triacylglycerols (TGs), unsaturated fatty acids (UFA) and total phospholipids (TPL) were determined in liver and blood serum. Obtained results of the lipotropic properties study of the «Prunofit» extract at a dose of 200 mg/kg against the background of alcoholic liver damage showed a decrease in the intensity of lipolysis, fatty hepatosis, and manifestations of hyperlipidemia. It has occurred due to a decrease in the content of total lipids, cholesterol, triglycerides and free fatty acids in the homogenate rat liver by 27.9%, 7.2%, 27.5% and 38%, respectively, and rat serum by 28%, 42.2%, 8.15%, 47.1%, respectively, compared with the control pathology. Against the background of model pathology, «Prunofit» extract tended to increase the content of total phospholipids in the liver homogenate by 37.34% and in serum by 30.2% compared with the control pathology. According to its ability to inhibit fatty liver infiltration, the «Prunofit» extract was at the level of the reference drug «Methionine» at a dose of 155 mg/kg.

We have measured levels of beta-lipotropin, beta-endorphin, and N-acetyl-beta-endorphin in the plasma, cerebrospinal fluid (CSF), and caudal medullary brain containing the respiratory-related portion of the nucleus tractus solitarius (NTS) of 2.5 +/- 1.0- (SD) and 38.2 +/- 3.7-day-old naive uninstrumented piglets. Time of day, ambient atmosphere, temperature, handling, sound, light, and nutritional status were kept constant. Experimental procedure included decapitation and rapid collection, processing, and freezing of tissues until analysis by radioimmunoassay. Young, compared with older piglets, have higher measured levels of beta-lipotropin in the plasma and CSF and of N-acetyl-beta-endorphin in all three body compartments. Although measured levels of beta-endorphin-like immunoreactivity are also higher in the plasma and CSF of the young group, the calculated level of beta-endorphin is higher only in the CSF. In the NTS, both the measured and calculated active endorphin appear higher in the older group, but this difference is not significant. Excess beta-endorphin in the CSF of neonates may explain the relative immaturity of their respiratory functions at birth.

Abstract. Histamine, which acts as a neurotransmitter, stimulates the release of the pro-opiomelanocortin derived peptides ACTH, β-lipotropin, and β-endorphin. Since stress affects the hypothalamic turn-over of neuronal histamine, we investigated the role of histaminergic neurons in the mediation of the stress-induced release of ACTH and β-endorphin immunoreactivity in male rats. In control animals histamine receptor antagonists had no effect on the release of ACTH or β-endorphin immunoreactivity. Restraint and ether stress increased plasma ACTH 3- and 2-fold, respectively. The responses were almost prevented by intracerebroventricular or intra-arterial infusion of the H2-receptor antagonists cimetidine and ranitidine. Infused intracerebroventricularly the H1-receptor antagonist mepyramine inhibited the ACTH response to restraint by 45% (P< 0.01), but had no effect on the response to ether. Infused intra-arterially the H1-receptor antagonists mepyramine or SKF-93944 had no effect. Restraint and ether stress increased plasma β-endorphin immunoreactivity 6- and 5-fold, respectively. Sephadex G-50 gel chromatography of plasma showed that the β-endorphin immunoreactivity in stressed rats coeluted with β-lipotropin and β-endorphin, whereas the immunoreactivity in control animals co-eluted almost exclusively with β-endorphin. The H2-receptor antagonists cimetidine and ranitidine infused intracerebroventricurlarly inhibited the responses of β-endorphin immunoreactivity to restraint and ether stress by 90 and 70%, respectively, whereas intra-arterial infusion of these antagonists inhibited the responses by only 50 and 60%, respectively. The H1-receptor antagonist mepyramine infused intracerebroventricularly inhibited the β-endorphin immunoreactivity response to restraint and ether stress by 40 and 25%, respectively, whereas intra-arterial infusion of mepyramine or the other H1-receptor antagonist SKF-93944 prevented the response to restraint stress, but had no effect on the response to ether stress. We suggest that hypothalamic histamine is involved in the mediation of the stress-induced release of the proopiomelanocortin-derived peptides ACTH, β-endorphin and β-lipotropin. The effect is mediated preferentially via H2-receptors, but H1-receptors may also play a role.

The article presents the results of studies of medium mass molecules concentration in the blood serum of cows with different physiological conditions and correction of this state with livazen, which has an antioxidant and lipotropic effect on liver cells. It was determined that the use of livazen contributed to a decrease in cholesterol in the group of pregnant cows by 21.0 % and in the group of fresh cows by 15.9 %, with a decrease in the concentration of average molecular peptides by 4.0-11.8 % depending on the length of exposure.

Resumen
Two of the commonest in practice postparturient liver diseases of dairy cows, ketosis and fatty liver, are discussed in this paper. It is not uncommon for both diseases to occur simultaneously, or each to act as a predisposing factor for the other. Clinical findings are nonspecific and certain biochemical parameters can be used as diagnostic tools. Treatment is focused in restoring glucose and energy balance of the affected animals with the administration of glucose, glucogenic substances, corticosteroids, insulin and lipotropic agents. Prevention aims in proper herd management, especially in terms of feeding rations.

Pro-opiomelanocortin (POMC), is a polyprotein expressed in the pituitary and the brain where it is proteolytically processed into peptide hormones and neuropeptides with distinct biological activities. It is the prototype of multipotent prohormones. The prohormone theory was first suggested in 1967 when Chrétien and Li discovered γ-lipotropin and observed that (i) it was part of β-lipotropin (β-LPH), a larger polypeptide characterized 2 years earlier and (ii) its C-terminus was β-melanocyte-stimulating hormone (β-MSH). This discovery led them to propose that the lipotropins might be related biosynthetically to the biologically active β-MSH in a precursor to end product relationship. The theory was widely confirmed in subsequent years. As we celebrate the 50th anniversary of the sequencing of β-LPH, we reflect over the lessons learned from the sequencing of those proteins; we explain their extension to the larger POMC precursor; we examine how the theory of precursor endoproteolysis they inspired became relevant for vast fields in biology; and how it led, after a long and arduous search, to the novel proteolytic enzymes called proprotein convertases. This family of nine enzymes plays multifaceted functions in growth, development, metabolism, endocrine, and brain functions. Their genetics has provided many insights into health and disease. Their therapeutic targeting is foreseeable in the near future. Thus, what started five decades ago as a theory based on POMC fragments, has opened up novel and productive avenues of biological and medical research, including, for our own current interest, a highly intriguing hypocholesterolemic Gln152His PCSK9 mutation in French-Canadian families.

Abstract. In conscious male rats intracerebroventricular infusion of histamine increased the plasma concentrations of ACTH and β-endorphin immunoreactivity 2.5-fold (P < 0.01). Gel filtration of plasma revealed two peaks of β-endorphin immunoreactivity corresponding to β-endorphin and β-lipotropin. The two fractions increased almost equally in histamine-stimulated animals, whereas most of the circulating β-endorphin immunoreactivity in control animals corresponded to β-endorphin. Central infusion of the H1-receptor agonist 2-thiazolylethylamine and of the H2-receptor agonists dimaprit or 4-methylhistamine increased the plasma ACTH and β-endorphin immunoreactivity concentrations 2- and 3-fold, respectively (P < 0.01). Infused intracerebroventricularly, the H2-receptor antagonists cimetidine or ranitidine prevented the histamine-induced increase in plasma ACTH and β-endorphin immunoreactivity (P < 0.01), whereas the H1-receptor antagonist mepyramine inhibited the peptide responses by 70% (P < 0.01). Infused intra-arterially cimetidine or ranitidine inhibited the histamine-induced increase in plasma ACTH by 80% (P < 0.01) and plasma β-endorphin immunoreactivity by 45% (P < 0.05), whereas mepyramine or the other H1-receptor antagonist SKF-93944 inhibited the ACTH response by 50% (P < 0.05), but had no effect on the β-endorphin immunoreactivity. The results indicate that histamine increases the release of the pro-opiomelanocortin derived peptides ACTH, β-lipotropin and β-endorphin from the anterior pituitary lobe, whereas an effect of histamine on the release of β-endorphin from the neurointermediate lobe is possible. The effect of histamine seems primarily mediated by H2-receptors, whereas H 1-receptors appear to play a minor role.

Leptospirosis is a disease caused by Leptospira spp., zoonotic Gram-negative bacteria. This study aimed to determine the handling of a leptospirosis case in a dog at the Pet Vet Clinic, Jakarta. Handling began with anamnesis and continued by examining the patient's physical condition, followed by serological, microscopic, and polymerase chain reaction examinations. An examination revealed that the dog had Leptospirosis. The therapy given was the administration of crystalline procaine and penicillin-G antibiotics. Supportive treatment was given with hepatoprotective, lipotropic, and diuretic supplements. Ringer's lactate was used as fluid therapy. The treatment administered showed positive, progressive results in the patient.

Flunarizine (10 mg/day for 60 days) was given to eight postmenopausal women with common migraine. Plasma LH pulsatility fluctuation, peripheral concentrations of prolactin (PRL), cortisol, β-endorphin (β-EP), β-lipotropin (β-LPH) and Pain Total Index (PTI) were evaluated before and after treatment. PTI was significantly reduced by flunarizine, which did not affect β-LPH, β-EP and cortisol plasma levels. On the contrary, both PRL values and amplitude, and length of LH pulses had increased at the end of treatment. Flunarizine reduced head pain in postmenopausal women. However, the enhancement of both PRL and LH release indicates that this calcium antagonist might interfere with the dopaminergic tonus.

Many important fields of research had a humble origin. In the distant past, A J P Martin’s discovery that amino acids could be separated by paper chromatography and Moore and Stein’s use of columns for quantitative amino acid analysis provided the first steps towards the determination of structure in complex biologically active molecules. They opened the door to reveal the essential relationship that exists between structure and function. In molecular endocrinology, for example, striking advances have been made by chemists with their expertise in the identification of structure working with biologists who contributed valuable knowledge and experience. Advantage was gained from the convergence of different background, and it is notable that the whole is greater than the sum. In the determination of structure, it may be recalled that four of the world’s great pioneers (Archibald Martin, Rodney Porter, Fred Sanger and Vincent du Vigneaud) were acknowledged for their fundamental contributions when individually they were awarded the Nobel Prize. They foresaw that the identification of structure would prove of outstanding importance in the future. Indeed, study of the structures of β-endorphin and enkephalin and the different forms of opiate activity they engender has led to a transformation in our understanding of chemical transmission in the brain.

Vesicular transport within the secretory pathway can be arrested by incubating cells at 15 degrees C or 20 degrees C to block exit from the endoplasmic reticulum or trans-Golgi network, respectively. Using this powerful tool we have compared the intracellular sites of endoproteolytic processing of proopiomelanocortin and two prohormone processing enzymes in AtT-20 mouse pituitary corticotrope tumor cells. For comparison, proopiomelanocortin processing was also evaluated in primary neurointermediate pituitary cultures. AtT-20 cells synthesize and store endogenous proopiomelanocortin and prohormone convertase 1; AtT-20 cells expressing high levels of integral membrane or soluble peptidylglycine alpha-amidating monooxygenase were generated by stable transfection. Cells were incubated with [35S]methionine and chased at 4 degrees C, 15 degrees C, 20 degrees C or 37 degrees C. The endoproteolytic processing of peptidylglycine alpha-amidating mono-oxygenase, prohormone convertase 1, and proopiomelanocortin was compared following immunoprecipitation. Endoproteolytic processing of integral membrane and soluble peptidylglycine alpha-amidating monooxygenase proteins was completely blocked by incubation of cells at 20 degrees C. In contrast, prohormone convertase 1 processing from the 87 kDa precursor to the 81 kDa intermediate proceeded to completion at both 15 degrees C and 20 degrees C, while cleavage to generate the 63 kDa prohormone convertase 1 protein was completely blocked at 20 degrees C. In AtT-20 cells and neurointermediate pituitary cultures, generation of beta-lipotropin from proopiomelanocortin continued at a slow but significant rate at 20 degrees C, while processing of beta-lipotropin to beta-endorphin was blocked. Thus prohormone convertase 1 processing begins in the endoplasmic reticulum and is not completed until after the trans-Golgi network, while peptidylglycine alpha-amidating monooxygenase processing begins after the trans-Golgi network. Selected proopiomelanocortin cleavages begin before entry into immature granules.

Aim is to optimize treatment of non-alcoholic fatty liver disease (NAFLD) combined with type 2 diabetes mellitus. 100 patients with NAFLD in stage of non-alcoholic steatohepatitis (NASH) were enrolled into study. Application of S-adenosylmethionine during the treatment of patients with non-alcoholic fatty liver disease (NAFLD) had high efficacy in the normalization of the functional, biochemical and cytokine profile of the blood. Combination of gliclizide and pioglitazone led to the decrease in the levels of cytolysis and mesenchymal-inflammatory syndromes, stabilization of lipid metabolism. This fact confirms the positive lipotrophic effect of the complex effect of gliclizide and pioglitazone on the functional state of hepatocytes, indicates decrease in insulin resistance and improves cell function. Complex therapy with combination of hypoglycemic and lipotropic drugs (gliclizide, pioglitazone, S-adenosylmethionine) showed good results, confirming the positive complex effect of S-adenosylmethionine, gliclizide and pioglitazone on the functional state of hepatocytes, led to decrease in insulin resistance and production of antiinflammatory effect. Pain syndrome remained significant in 9 patients (30%, 1.8 times less than before treatment); dyspeptic syndrome – in 10 patients (33,3%, decreased by 1,7 times), anorexia – in 10 patients (33,3%, a decrease of 1,9 times), asthenia – in 11 patients (36,7 %, decrease by 1.9 times). Combination of hypoglycemic, lipotropic therapy and basic therapy positively influenced on the state of hepatocytes decreasing activity of ALT, AST, alkaline +phosphatase. USG study showed reduction of the hepatic tissue changes compared to the visualization results before treatment. Particularly important was a significant decrease in the number of patients on the stage 3 of hepatosis having a decrease in the levels of TNF-α, IL-6 and C-reactive protein and increase of adiponectin. According to the results, application of the proposed agents affecting specific pathogenic links of NAFLD is believed to be an effective way to treat NAFLD.
