AbstractField studies conducted in 2018–2019 in the northern Steppe of Ukraine with sunflower hybrids of different maturity groups (LG 50300, LG 5580, LG 5478, LG 5638, LG 5662) showed that the crop density of early-crop hybrid LG 50300 from 55,000 plants/hectare to 70,000 plants/hectare led to a decrease in productivity by 0.11 t ha−1 and a decrease in oil content by 0.9%. The density of middle-early hybrid LG 5580 resulted in a decrease in sowing productivity of 0.21 t ha−1, while oil content remained nearly the same. Another middle-early hybrid LG 5478 showed slight variations in productivity and oil content with an increase of crop density. The study of the mid-season hybrid LG 5038 showed a decrease in sowing productivity by 0.2 t ha−1 with the density up to 70,000 plants/hectare. Mid-season hybrid LG 5662 with density of 70,000 plants/hectare showed productivity increase by 0.14 t ha−1.

This study aims to determine: (1) The effect of brand equity on purchasing decisions, (2) The effect of perceived quality on purchasing decisions, (3) The effect of brand association on purchasing decisions, and (4) The effect of brand loyalty on purchasing decisions. This type of research is a survey research using a questionnaire. The population in this study were all consumers of LG brand LED TVs in the Special Region of Yogyakarta. The samples used are consumers who have made purchases and consumers who use LG branded LED TVs. The sampling method used accidental sampling with a number of respondents 100 people. Data were collected using a questionnaire that has been tested for validity and reliability. The data analysis technique used is multiple linear regression. The results of this study indicate that: based on the results of the t test (1) Brand Equity does not have a significant effect on the decision to buy LED TV products with the LG brand in Yogyakarta with a significance value of 0.427> 0.05. (2) Perception of quality does not have a significant effect on purchasing decisions for LG brand LED TV products in the Special Region of Yogyakarta with a significance value of 0.222> 0.05. (3) Brand Association has a significant effect on purchasing decisions for LG brand LED TV products in Yogyakarta with a significance value of 0.037 <0.05. (4) Brand Loyalty has a significant effect on purchasing decisions for LG LED TV products in the Special Region of Yogyakarta with a significance value of 0.000> 0.05. Meanwhile, based on the F test, it shows that brand association and brand loyalty simultaneously influence the decision to buy LED TV products with the LG brand in Yogyakarta, with a calculated F value of 56,968> 0.05 with a significance value of 0.000 <0.05.

Abstract In wild-type strains of Neurospora crassa, the rDNA is located at a single site in the genome called the nucleolus organizer region (NOR), which forms a terminal segment on linkage group (LG) V. In the quasiterminal translocation strain T(I;V)AR190, most of the right arm of LG I moved to the distal tip of the NOR, and one or a few rDNA repeat units are moved to the truncated right arm of LG I. I report here that, in partial diploid strains derived from T(I;V)AR190, large terminal deletions result from chromosome breakage in the NOR. In most of these partial diploids, chromosome breakage is apparently frequent and the breakpoints occur in many parts of the NOR. The rDNA ends resulting from chromosome breakage are "healed" by the addition of new telomeres. Significantly, the presence of ectopic rDNA creates a new site of chromosome breakage in the genome of partial diploids. These results raise the possibility that, under certain conditions, rDNA is a region of fragility in eukaryotic chromosomes.

Tryptic hydrolysis of whey protein isolate under specific incubation conditions including a relatively high enzyme:substrate (E:S) ratio of 1:10 is known to preferentially hydrolyse β-lactoglobulin (β-LG), while retaining the other major whey protein fraction, i.e., α-lactalbumin (α-LA) mainly intact. An objective of the present work was to explore the effects of reducing E:S (1:10, 1:30, 1:50, 1:100) on the selective hydrolysis of β-LG by trypsin at pH 8.5 and 25 °C in a 5% (w/v) WPI solution during incubation periods ranging from 1 to 7 h. In addition, the use of a pilot-scale continuous high-temperature, short-time (HTST) heat exchanger with an extended holding time (EHT) of 5 min as a means of inactivating trypsin to terminate hydrolysis was compared with laboratory-based acidification to <pH 3 by the addition of HCl, and batch sample heating in a water bath at 85 °C. An E:S of 1:10 resulted in 100% and 30% of β-LG and α-LA hydrolysis, respectively, after 3 h, while an E:S reduction to 1:30 and 1:50 led >90% β-LG hydrolysis after respective incubation periods of 4 and 6 h, with <5% hydrolysis of α-LA in the case of 1:50. Continuous HTST-EHT treatment was shown to be an effective inactivation process allowing for the maintenance of substrate selectivity. However, HTST-EHT heating resulted in protein aggregation, which negatively impacts the downstream recovery of intact α-LA. An optimum E:S was determined to be 1:50, with an incubation time ranging from 3 h to 7 h leading to 90% β-LG hydrolysis and minimal degradation of α-LA. Alternative batch heating by means of a water bath to inactivate trypsin caused considerable digestion of α-LA, while acidification to <pH 3.0 restricted subsequent functional applications of the protein.

It is well known that fruit processing industries lead to the generation of large amounts of by-products every year, such as seeds and peels, which are frequently considered as waste. However, it has already been proved that these by-products can consist in a rich source of bioactive compounds with valuable nutritional and biological effects (1). In this perspective, there is a growing interest in the discovery of natural antioxidants which can be used as an alternative to synthetic substances by pharmaceutical and food industries (2). Jabuticaba (Myrciaria cauliflora) is a typical fruit from Brazil that belongs to Myrtaceae family. Its seeds are usually discarded as waste by industries and studies have already proven that these by-products are rich in bioactive compounds with promising biological effects (3,4). In this study, it was evaluated the antioxidant activity of hydroalcoholic (EtOH), methanolic (MEtOH), aqueous (AQ) and propanone (AQAc) extracts from jabuticaba seeds. This effect was assessed by their ability to scavenge 2,2-diphenyl-1-picrylhydrazyl (DPPH•), 2,2’-azino-bis (ethylbenzothiazoline-6-sulfonic acid) (ABTS•+), hypochlorous acid (HOCl), and superoxide anion (O2 •−) radicals. The redox properties of jabuticaba seed extracts were studied by using differential pulse voltammetry (DPV). Their protective effect to prevent in vitro oxidative cellular damages induced by 2,2-azobis(2-aminopropanone) dihydrochloride (AAPH) in human erythrocytes was also investigated. The obtained results revealed that all extracts were able to scavenge free radicals used in this work, due to their lower IC50 (inhibitory concentration) values. A similarity between E pa values suggested that the extracts were constituted by analogous electroactive species and exhibited similar antioxidant effect in this assay. However, it was observed that the AQAc extract presented the lowest E pa value (115.1 ± 0.95 mV) indicating a higher antioxidant activity when compared to the other extracts (p < 0.05). Furthermore, it was showed a good positive correlation between the results obtained by using DPV and the synthetic and biological radicals (DPPH•: r = 0.8215; ABTS•+: r = 0.9123; HOCl: r = 0.8193, and O2 •-: r = 0.9916). The extracts protected human erythrocytes against oxidative cellular damage caused by AAPH, which was confirmed by using field-emission gun scanning electron microscopy (FESEM) analysis. Taken together, the results reported in this work evidence that jabuticaba seeds are a source of antioxidant compounds that present a remarkable potential to prevent damage related to oxidative stress in biological systems. In this way, our work suggests the use of this fruit byproduct in further biochemical studies aiming its use as a new nutraceutical and food additive. Recognition of conventional foreign antigen by T cells is determined by the expression of multiple variable regions of both alpha and beta chains of the T cell receptor (TCR) alpha/beta heterodimer. In contrast, there exists a class of antigens that appears to interact with the TCR alpha/beta heterodimer through the variable region on the beta chain (V beta), independent of other TCR components, a property that has led to their designation as superantigens. The goal of the present study was to analyze V alpha use in V beta 6+ T cells responsive to the superantigen, Mlaa. Results indicate that while deletion of T cells expressing V beta 6 in Mlsa-expressing mice is essentially complete and therefore appears to occur regardless of V alpha usage, in vitro Mlsa stimulation of T cells from Mlsa-negative mice results in significant skewing of V alpha use among responding V beta 6+ T cells. This indicates that V alpha expression influences recognition of the superantigen, Mlsa by mature peripheral T cells.

Goniopora columna is a stony coral valued for its reef-building potential and its unique appearance. Thus, identifying the optimal culture conditions for G. columna would enable efficient cultivation and prevent the illegal exploitation of marine resources. Light sources are crucial for the growth of corals because zooxanthellae provide them with basic nutrients through photosynthesis. Different corals and zooxanthellae have different photoacclimation characteristics; therefore, selecting a suitable light wavelength remains the key inhibitor of coral maintenance in marine aquariums. Accordingly, this study investigated the effects of different light wavelengths on G. columna. It was illuminated for 6 or 12 h a day under white light, yellow light, red light (LR), green light (LG), blue light (LB), or purple light (LP) for 8 weeks. During the experiment, R(R; i.e., a formula feed that combines sodium alginate, protein and probiotics) of 5% (w/v) of G. columna tissue and skeletal dry weight was fed every day. Coral polyps were counted, zooxanthellae density, chlorophyll a concentration, specific growth rates, and survival rates were calculated; polyp stretching and contractile behaviors were observed; and body composition and digestive enzyme activity were analyzed. LB or LP (but not LG or LR) illumination for at least 6 h per day significantly promoted the growth, survival, protein content, and protease activity of the G. columna specimens. Furthermore, coral polyp extension reached 100% after 30 min of LP and LB light irradiation. Although no significant differences in the zooxanthellae density or chlorophyll a concentration were noted under various light wavelengths, significant reductions were detected in the absence of light. To achieve energy-efficient coral aquaculture with regard to G. columna cultivation, 6 h of LB or LP illumination per day can improve the growth.

The energy efficiency index (EEI) is an important factor used as an indicator either for building energy consumption or electronic device performance; it allows one to select effective devices that save energy. This work studies the performance of different types of LED lamps used in indoor lighting, the lamps currently available in the Egyptian market have been tested according to their photometric and electric parameters, namely: luminous flux, power factor, and EEI. Three different brands E, T, and V have been chosen with the nominal powers 9, 12, and 15 W. The results showed that both 9- and 15-Watt lamps have the same EEI values as 0.14, 0.13, and 0.12 for T, V, and E lamps, respectively, whereas 12-Watt lamps have EEI values of 0.16, 0.13, and 0.13 for T, V, and E lamps, respectively. The experimental testing of these lamps revealed that all the lamps have the same EEI class (A+) regardless of the nominal power. The results also revealed a relationship between the power factor and EEI: as the power factor increases, EEI increases, too. The expanded uncertainty in luminous flux has been calculated.

To test whether antibodies directed to TCR affect T cell tumor growth in vivo, mice were inoculated intravenously with C6VL tumor cells expressing V beta 6 TCR and then treated intraperitoneally with mAb specific for V beta 6 TCR. Administration of anti-V beta 6 antibody prolonged survival of mice bearing V beta 6-expressing tumor cells and it led to the induction of host immunity to the tumor cells in surviving animals. This treatment eliminated not only tumor cells bearing V beta 6 TCR but also normal host T cells expressing V beta 6 T cells receptors. However, the lack of V beta 6-expressing T cells in such treated mice did not result in generalized immune disfunction. These data demonstrate the utility of anti-TCR V segment antibody in the treatment of T cell tumors. Most importantly, since the number of V genes for the T cell antigen receptor is limited, both in man and in mouse, it should be possible to establish a panel of mAbs directed to each V gene product and use such antibodies in the treatment of T cell neoplasms.

Ferguson’s “separate but equal” doctrine; use of the Atlanta Constitution as the propaganda machine of New South Democrats; and his national advocacy for the disfranchisement of Black southerners. It also documents the New South and Grady critique of Black press leaders such as T. Thomas Fortune and Ida B. Wells-Barnett.

Innovation may be divided into two types: incremental and discontinuous (Tidd et al. 2009). The first of these refers to ‘doing what is already done but better.’ The latter may come through the emergence of a new technology, a completely new market or a new business model and refers to decision making under uncertainty. The focus of this chapter is the implementation of discontinuous innovation (DI) in supply chain relationships. The authors review two research-led projects: the development and implementation of the SCRIA (Supply Chain Relationship in Aerospace) Relationship Evaluation Tool (RET) and the development of the concept of Value-Transparency (V-T). Both projects originated in the authors’ academic research in the early 1990s and were aimed at subsequent implementation in the context of supply chain relationships. One was successfully implemented; the other was not. The authors introduce future research exploring some supply relationship context-specific barriers and enablers in the implementation of radical concepts or DI through case based research method.

Various difficulties of classical physics, including inadequate description of atoms and molecules, led to new ways of visualizing physical realities, ways which are embodied in the methods of quantum mechanics. Quantum mechanics is based on the description of particle motion by a wave function, satisfying the Schrodinger equation, which in its “time-independent” form is: ((−h2/8mπ2)⛛2+V)Ψ=E Ψ or, for short: HΨ = EΨ In this equation, H, the Hamiltonian operator, is defined by H = − ((h2/8mπ2)⛛2+V, where h is Planck’s constant (6.6 10−34 Joules), m is the particle’s mass, ⛛2 is the sum of the partial second derivative with x,y, and z, and V is the potential energy of the system. As such, the Hamiltonian operator is the sum of the kinetic energy operator and the potential energy operator. (Recall that an operator is a mathematical expression which manipulates the function that follows it in a certain way. For example, the operator d/dx placed before a function differentiates that function with respect to x.) E represents the total energy of the system and is a number, not an operator. It contains all the information within the limits of the Heisenberg uncertainty principle, which states that the exact position and velocity of a microscopic particle cannot be determined simultaneously. Therefore, the information provided by Ψ(nit) is in terms of probability: Ψ2(x,t) is the probability of finding the particle between x and x + dx, at time t. The Schrödinger equation applied to atoms will thus describe the motion of each electron in the electrostatic field created by the positive nucleus and by the other electrons. When the equation is applied to molecules, due to the much larger mass of nuclei, their relative motion is considered negligible as compared to that of the electrons (Born-Oppenheimer approximation). Accordingly, the electronic distribution in a molecule depends on the position of the nuclei and not on their motion. The kinetic energy operator for the nuclei is considered to be zero. For a many-electron molecule, the Hamiltonian operator can thus be written as the sum of the electrons’ kinetic energy term, which in turn is the sum of individual electrons’ kinetic energies and the electronic and nuclear potential energy terms.

Abstract Laser Additive Manufacturing using Powder Bed Fusion (LAM-PBF) is one of the revolutionary technologies playing a key role in fourth industrial revolution for redefining manufacturing from mass production to mass customization. To upkeep the pace, Raja Ramanna Centre for Advanced Technology (RRCAT) has indigenously developed an LAM-PBF system and it is being used for process and component development for various engineering applications. This paper reports a parametric investigation to evaluate the influence of process parameters on the sample properties and to develop the process window for fabricating complex shaped engineering components. In the present work, an experimental investigation is carried out to investigate the effect of Laser Energy density (LED) on the porosity, microstructure and mechanical properties of SS 316L bulk structures fabricated by LAM-PBF system. LED is a combined parameter simultaneously considering the effect of Laser Power (P), Scan Speed (v), hatch spacing (h) and layer thickness (t). The effect of three LED values such as 83.33 J/mm3, 253.97 J/mm3 and 476.19 J/mm3 is investigated in the present work by building cuboidal samples at a layer thickness of 75 microns. Porosity is estimated using area fraction method in optical microscopy and it is found that the minimum porosity of 0.14 % and pore area of 1.28 mm2 are observed at 253.97 J/mm3. Maximum porosity of 18.85 % is observed at 83 J/mm3 due to incomplete fusion defects. However, porosity observed at 475 J/mm3 is 6.56 % with average pore size of 17.8 mm2. Microstructural studies show primarily columnar growth in all the samples with fine dendrites. The dendrite size is observed to be 3.2 μm, 2.4 μm and 1.46 μm at 83.33 J/mm3, 253.97 J/mm3 and 476.19 J/mm3 respectively. Micro-hardness and single cycle automatic ball indentation studies are found to be in agreement with dendritic size, with lower hardness at larger dendrite size. X-Ray Diffraction (XRD) studies indicate similar peaks at all the conditions, with slight peak shift observed with increase in LED primarily due to higher amount of residual stress. Thus, it can be inferred that LED of 253.97 J/mm3 is suitable for fabricating engineering components due to combination of lower porosity and fine dendritic structure.

During last decade, customers’ requirements of Line Pipes and accessories became more and more stringent. This process is led by the exploitation of fields with more severe conditions of pressure, environment and temperature. For this reason, heavy wall products (both straight pipes and bends) need to be developed. A development program was carried out in order to satisfy more stringent requirements and higher wall thickness. The metallurgical approach to steel design aimed to improve the combination of strength and toughness and increase the control of hardness after quenching and tempering, preserving adequate weldability. Industrial trials were performed to manufacture 60 mm thick hot induction bends in grade X65. Characterization was carried out after various post bending heat treatments (quenching, tempering and post welding). The new low-C steel showed promising results. The full characterization of off-line Q&T bends at various locations (tangent length, transition zone, and bend body) confirmed the achievement of X65 grade, with hardness HV10 < 240, Charpy V-notch transition temperature < −60°C, Crack Tip Opening Displacement > 0.7 mm at −45 °C, and very good HIC and SSC resistance.

Backgroud: "Population migration" is a term that has many meanings. Population migration can manifest itself in such forms as nomadism, pilgrimage, wanderings, urbanization, ruralization, etc. Population migrations have a long history, but are relatively little studied. In Western Europe and North America, population migration became the object of sociological research only from the middle of the 19th century. Interest in the study of population migration has become relevant due to the needs of studying the adaptation of immigrants in host countries and studying the consequences of mass emigration of the working population from donor countries. Purpose: To carry out a systematization and comparative analysis of the main approaches to the study of migration in sociology and other socio-humanitarian disciplines. Methods: The work uses a number of general scientific and special sociological methods: logicalsemantic - for analyzing and deepening the conceptual apparatus of the concept of external migration; comparative analysis of the results of statistical and specifically sociological studies of migration. Results: A significant increase in the scale and intensity of international population migration at the end of the 20th and the beginning of the 21st century led to the interest of Ukrainian sociologists in the issue of migration. The theoretical and methodological approaches of Western researchers echo the approaches of post-Soviet scientists, in particular, in the recognition of the interdisciplinary nature of population migration studies. Thus, there are six sociological approaches to the study of migration. At the same time, V. Iontsev noted that to the sociological approach "it would be possible to add the classification of migration flows according to vertical and horizontal characteristics and the theory of "rational expectations". Conclusion: Within the scope of the comprehensive study, a broad classification of approaches to the study of migration was presented. V. Iontsev's classification included 17 scientific approaches to the study of population migration, which, in turn, united 45 scientific directions, were classified as: the concept of "attraction - repulsion" by E. In this study, we have conducted a combined numerical and experimental study on the Poisson’s ratio of SAC lead free solders. The Poisson’s ratio (PR) is one of the basic mechanical properties used in many material constitutive models. Although often not measured, it is important property in many finite element method (FEM) calculations. The value of the Poisson’s ratio of SAC lead free solders is relatively unexplored compared to other material properties, and for FEA simulations it is typically assumed to be v = 0.3. In the current work, we have shown the effects of the chosen value of the solder joint Poisson’s ratio on the finite element results for BGA components subjected to thermal cycling. In the finite element models, the reliability predictions were based on the Morrow-Darveaux energy-based fatigue model. Several sizes (5, 10, 15 mm) of PBGA components with SAC305 solder joints with 0.4 and 0.8 mm spacing were modeled. The packages were subjected to a time dependent cyclic temperature distribution from −40 to 125 °C. The package assemblies were assumed to be in a stress-free state at 25 °C (room temperature), with no residual stresses induced in the manufacturing process. The simulation results have demonstrated that for specified range of Poisson’s ratio values of 0.15 < v < 0.40, the solder Plastic Work varied over 20% and the Predicted Reliability Varied over 50%. To determine the actual Poisson’s ratio experimentally, uniaxial tensile stress-strain tests were carried out on SAC305 (96.5Sn3.0Ag0.5Cu) specimens using a micro tension/torsion testing machine with two strain rates (0.0001, and 0.00001 (sec−1)), four testing temperatures (T = 25, 50, 75, 100 °C), and several durations of prior aging at T = 125 °C. Deformations and strains in axial and transverse directions were measured using strain gages with automatic data acquisition from LabVIEW software. The recorded transverse strain vs. axial strain data were then fit with a linear regression analysis to determine the Poisson’s ratio values. A test matrix of experiments was developed to study the effects of temperature, strain rate, alloy composition, and solidification cooling profile on the value of solder Poisson’s ratio. The Poisson’s ratio was found to increase with increasing temperature, and decrease with increasing strain rate. Using a slower solidification cooling profile led to an increase in the solder Poisson’s ratio value. Finally, the microstructural coarsening that occurs during isothermal aging lead to an increase in the Poisson’s ratio.

Nanochips® cartridges (Fig 1) are disposable panels with 400 micro arrays (Fig 2) that can be independently used as test sites for various assays like genetic marker. Each site (Fig 3) is a permeation layer coated electrode with 80 μ diameter and 120 μ pitch. Permeation or “perm” layer is a thin film of a proprietary hydrogel material over the proprietary chip design. Perm layers were deposited on the electrodes using spin coating process with a proprietary solution. Performance of the cartridge depended on the quality and thickness of the permeation layer over these 400 electrodes. In this process one millimeter deep tear shaped well was constructed from the ceramic base to which 20×20 array of silicon chip were attached. These wells filled with 180 μl proprietary solution and spun at fixed 1200 RPM for 20 seconds. Process was repeated three times at room temperature in a clean room. Post process treatment included 30 minutes in dry incubator, wash in Milli Q water, and finally, dry at room temperature. Quality of the 1,500 nm thick permeation layer was so demanding that more than half of the cartridges were rejected due to poor quality of the perm layer. Major causes of rejection were bubbles, high SD (Standard Deviation), and thickness (too thick or too thin) of the layer. To understand the problem focus was given to both solution making and spin coating process. Basic hypothesis was that the film thickness was based on viscosity of solution and subsequent evaporation process. Figs 4–6 showed the details of spin coating process for developing a heuristic model. Since viscosity depended strongly on the temperature and time during a chemical reaction, a viscosity profile was developed for the solution during the reaction. From the viscosity curve (Fig. 7) it was established that 75 minute at 50° C was not enough to complete the reaction as was initially thought. The time-Viscosity curve reached the plateau after 150 minutes! So, it was necessary to continue the reaction for 75 more minutes to complete the reaction. This explained a major reason (bubble forming) in the present process. Viscosity of the solution depended on a number of other factors like dispensed volume, temperature of the spinneret, time to dispense and/or in the spinneret, etc. A systematic study on these variables led to an empirical equation of the following form: Hf=Hf0+A*(τe/Vd)*ηf*(Tc−T0)n Where Hf = Predicted Average Thickness in nm. Hf0 is minimum average thickness in nm. T, V, and τ are various temperature, dispensed volume, and time parameters. A and n are curve fitting constants for experimental set-ups. The best of part of this modeling was its ability not only to predict the thickness of the film, rather its ability to control the thickness of the film in real time for a given solution. Above equation allowed appropriate dispensing volume and time to be kept in the well of the chip before spinning for a given solution with a specific viscosity. A tabular form was given to the operator who matched the information to get a specific film thickness. This model helped dropped the rejection rate to less than 10% from more than 50%. Operators were able to control the thickness of the film within 1500 nm +/− 300 nm, as demanded by the Spec. The model further allowed to target, rather control, the film thickness. We were also able to make films as thin as 800 nm or as thick as 1,500 nm with +/− 200 nm variation from the Table developed from the empirical equation. Using the assumption that viscosity plays the most important role in our spin coating process and constructing a corresponding evaporative model, we were able to identify a major shortcoming of an existing process to develop submicron thick permeation layer. Existing process was resulting in more than 50% rejection of an expensive critical component. An empirical model of spin coating process was developed to predict the film thickness within hundreds of nanometers. This dramatically improved the yield to more than 90% from less than 50%. The model allowed to correct the process in real time and allowed targeting the film thickness anywhere around one micron with few hundred nanometer accuracy.

The general objective of this study was to develop rationally designed mimetic antagonists (and agonists) of the PK/PBAN Np class with enhanced bio-stability and bioavailability as prototypes for effective and environmentally friendly pest insect management agents. The PK/PBAN family is a multifunctional group of Nps that mediates key functions in insects (sex pheromone biosynthesis, cuticular melanization, myotropic activity, diapause and pupal development) and is, therefore, of high scientific and applied interest. The objectives of the current study were: (i) to identify an antagonist biophores (ii) to develop an arsenal of amphiphilic topically active PK/PBAN antagonists with an array of different time-release profiles based on the previously developed prototype analog; (iii) to develop rationally designed non-peptide SMLs based on the antagonist biophore determined in (i) and evaluate them in cloned receptor microplate binding assays and by pheromonotropic, melanotropic and pupariation in vivo assays. (iv) to clone PK/PBAN receptors (PK/PBAN-Rs) for further understanding of receptor-ligand interactions; (v) to develop microplate binding assays for screening the above SMLs. In the course of the granting period A series of amphiphilic PK/PBAN analogs based on a linear lead antagonist from the previous BARD grant was synthesized that incorporated a diverse array of hydrophobic groups (HR-Suc-A[dF]PRLa). Others were synthesized via the attachment of polyethylene glycol (PEG) polymers. A hydrophobic, biostablePK/PBAN/DH analog DH-2Abf-K prevented the onset of the protective state of diapause in H. zea pupae [EC50=7 pmol/larva] following injection into the preceding larval stage. It effectively induces the crop pest to commit a form of ‘ecological suicide’. Evaluation of a set of amphiphilic PK analogs with a diverse array of hydrophobic groups of the formula HR-Suc-FTPRLa led to the identification of analog T-63 (HR=Decyl) that increased the extent of diapause termination by a factor of 70% when applied topically to newly emerged pupae. Another biostablePK analog PK-Oic-1 featured anti-feedant and aphicidal properties that matched the potency of some commercial aphicides. Native PK showed no significant activity. The aphicidal effects were blocked by a new PEGylated PK antagonist analog PK-dF-PEG4, suggesting that the activity is mediated by a PK/PBAN receptor and therefore indicative of a novel and selective mode-of-action. Using a novel transPro mimetic motif (dihydroimidazole; ‘Jones’) developed in previous BARD-sponsored work, the first antagonist for the diapause hormone (DH), DH-Jo, was developed and shown to block over 50% of H. zea pupal diapause termination activity of native DH. This novel antagonist development strategy may be applicable to other invertebrate and vertebrate hormones that feature a transPro in the active core. The research identifies a critical component of the antagonist biophore for this PK/PBAN receptor subtype, i.e. a trans-oriented Pro. Additional work led to the molecular cloning and functional characterization of the DH receptor from H. zea, allowing for the discovery of three other DH antagonist analogs: Drosophila ETH, a β-AA analog, and a dF analog. The receptor experiments identified an agonist (DH-2Abf-dA) with a maximal response greater than native DH. ‘Deconvolution’ of a rationally-designed nonpeptide heterocyclic combinatorial library with a cyclic bis-guanidino (BG) scaffold led to discovery of several members that elicited activity in a pupariation acceleration assay, and one that also showed activity in an H. zea diapause termination assay, eliciting a maximal response of 90%. Molecular cloning and functional characterization of a CAP2b antidiuretic receptor from the kissing bug (R. prolixus) as well as the first CAP2b and PK receptors from a tick was also achieved. Notably, the PK/PBAN-like receptor from the cattle fever tick is unique among known PK/PBAN and CAP2b receptors in that it can interact with both ligand types, providing further evidence for an evolutionary relationship between these two NP families. In the course of the granting period we also managed to clone the PK/PBAN-R of H. peltigera, to express it and the S. littoralis-R Sf-9 cells and to evaluate their interaction with a variety of PK/PBAN ligands. In addition, three functional microplate assays in a HTS format have been developed: a cell-membrane competitive ligand binding assay; a Ca flux assay and a whole cell cAMP ELISA. The Ca flux assay has been used for receptor characterization due to its extremely high sensitivity. Computer homology studies were carried out to predict both receptor’s SAR and based on this analysis 8 mutants have been generated. The bioavailability of small linear antagonistic peptides has been evaluated and was found to be highly effective as sex pheromone biosynthesis inhibitors. The activity of 11 new amphiphilic analogs has also been evaluated. Unfortunately, due to a problem with the Heliothis moth colony we were unable to select those with pheromonotropic antagonistic activity and further check their bioavailability. Six peptides exhibited some melanotropic antagonistic activity but due to the low inhibitory effect the peptides were not further tested for bioavailability in S. littoralis larvae. Despite the fact that no new antagonistic peptides were discovered in the course of this granting period the results contribute to a better understanding of the interaction of the PK/PBAN family of Nps with their receptors, provided several HT assays for screening of libraries of various origin for presence of PK/PBAN-Ragonists and antagonists and provided important practical information for the further design of new, peptide-based insecticide prototypes aimed at the disruption of key neuroendocrine physiological functions in pest insects.
