Texto completoThe main objective of this study was to examine the relationship between CETP and LDL particle heterogeneity in heterozygous familial hypercholesterolemia. The results of this study suggest that the LDL peak particle diameter was smaller in familial hypercholesterolemia than in controls. Furthermore, the results suggest that several factors are implicated in the LDL particle heterogeneity and that some of them are associated with familial hypercholesterolemia, such as the plasma CETP concentration. This study suggest that increased plasma CETP concentration could lead to significant LDL particle remodeling in familial hypercholesterolemia and could contribute to the pathogenesis of atherosclerosis in these patients.

Texto completoPlasma lipoproteins constantly exchange lipids and apolipoproteins. Besides the action of transfer proteins, the ability of lipoproteins to receive or donate lipids also depends on several other factors. The structure, lipid and protein composition of the lipoproteins may influence the fluidity of the particle and thereby this ability of lipoprotein. In the plasma, the lipoprotein class that is the most affected by lipid transfers is HDL. The current study was designed to establish a practical method to quantify the ability of this lipoprotein to simultaneously receive its main constituent lipid classes, namely phospholipid, free cholesterol, cholesteryl ester and triglycerides. The method is based on the lipids exchange between an artificial nanoemulsion (LDE) that resembles the LDL, used as a donor of radioactive lipids, and plasma lipoproteins. After precipitation of the LDE and the others plasma lipoproteins, the accept capacity of the HDL\' is quantified by the measure of radioactivity in the lipoprotein. In the present study, the validation of this method was carried through. Moreover, possible interferences in the method had been analyzed. In the same study, the transfer of lipids of LDE for the particle of HDL from control subjets was analyzed. The rise of temperature (4 to 37°C), time of incubation (5min to 2h) and of HDL-Cholesterol (33 to 244 mg/dL) resulted in gradual increase in the transfer of the four lipids from LDE to HDL. On the other hand, the increase of pH (6.5 to 8.5) and albumin concentration (3.50 to 7.00 g/dL) had not modified the values of transfers. The plasma sample showed to be unchanged for this assay for period of 12 months (p>0.05), whereas the LDE was unchanged for 15-days (p>0.2). The intra-assay results showed imprecision (C.V) of 0.83, 0.56, 1.49 and 0.51 % for phospholipid, free cholesterol, cholesteryl ester and triglycerides transfer, respectively. The inter-assay showed imprecision for results of phospholipid, free cholesterol, cholesteryl ester and triglycerides transfer of 0.78, 0.59, 1.32 and 0.58%, respectively. The average of phospholipid, free cholesterol, cholesteryl ester and triglycerides transfers from LDE to HDL in the 53 volunteers was of 25.5±2.6, 9.9±1.6; 4.8±1 .3 and 6.9±1 .1 %, respectively. Cholesterol ester, triglyceride and free cholesterol transfers positively correlated among each other. A positive correlation was also found between phospholipid and triglyceride transfers and free cholesterol transfer and HDL cholesterol concentration. The method of lipids transfer to HDL showed to be practical and reproducible, being able, inside of the ideal conditions established in this work, to determine the aceptora capacity of lipids of the HDL.

Texto completoIn the framework of CEA R&D program to develop an industrial prototype of Sodiumcooled Fast Reactor, the present thesis aimed to propose an innovative compact heat exchanger technology. In order to increase the global compactness the basic idea of this work is to design a channel were the fluid flow is as much three-dimensional as possible. In particular the channel can be thought as the result of the superposition of two undulated channels in phase opposition. To numerically provide a physically-consistent model, a new non-linear eddy viscosity named Anisotropic Shear Stress Transport (ASST) model has been developed and implemented into the available solver ANSYS FLUENT. To validate the numerical model, two experimental sections have been used to acquire an extensive aerodynamic database, whereas, to validate the thermal modeling approach, the VHEGAS facility has been built. Once having validated the ASST model, correlations for friction factor and Nusselt number for various geometries could be obtained. Finally, it has been shown that the innovative channel is the most compact one among the most important existing industrial compact heat exchanger technologies

Texto completoThe blood-brain barrier (BBB) protects the central nervous system (CNS) from plasma fluctuations of endogenous, but also exogenous molecules, including therapeutic molecules. The BBB’s restrictive properties are compensated by the presence of different mechanisms that provide transport of nutrients across the BBB, including transcytosis of endogenous ligands mediated by receptors. Our objective is to improve drug delivery across the BBB and we developed “vectors” that target different recpetors. During our thesis we developed and optimized cellular tools and approaches, in particular syngeneic in vitro models of the BBB and blood-spinal cord barrier (BSCB) from both rat and mouse, based on the co-culture of brain (BMECs) or spinal cord (SCMECs) microvascular endothelial cells (MECs) and astrocytes. Among the receptors we studied, we show that the LDL receptor (LDLR) is expressed at the apical plasma membrane of BMECs and confirmed that it is involved in transcytosis of LDL through the vesicular compartment, while avoiding the lysosomal compartment, further establishing its interest as a target receptor. We show that our vectors conjugated to an organic molecule or to a protein cargo are endocytosed by BMECs in a LDLR-dependent manner, avoid the lysosomal compartment and cross the BMEC monolayers. Finally, we developed BBB and BSCB in vitro models in inflammatory conditions, considering that MECs inflammation is associated with many CNS lesions and pathologies. These models will be useful to better understand the inflammatory processes of CNS endothelial cells and to evaluate vectorization strategies preferentially targeting CNS structures in pathological condition

Texto completoThis experimental and numerical study in the field of fluid mechanics deals with jets-in cross flow configurations that are relevant for the cooling of aero engine combustion chambers. Indeed, in presence of instabilities it is important to determine to which extent the film cooling is able to do its job of preserving the combustion chamber walls from the thermal load. The test facility MAVERIC has been upgraded in order to acoustically force the crossflow in which the jets are discharging. The strong sensitivity of the overall flow unsteady properties to the presence of the acoustic forcing has been clearly evidenced. The agreement between the experimental results and large-eddy simulations proved to be quite encouraging for a stationary acoustic wave whereas the case of a propagating acoustic wave investigated only numerically reveals also quite a significant change of the flow topology. In this context, the effect of the acoustic forcing on the wall thermal behavior has been analyzed thanks to the use of the fluid-solid coupled AVBP-AVTP solver

Texto completoThis thesis deals with the study of random methods for learning large-scale data. Firstly, we propose an unsupervised approach consisting in the estimation of the principal components, when the sample size and the observation dimension tend towards infinity. This approach is based on random matrices and uses consistent estimators of eigenvalues and eigenvectors of the covariance matrix. Then, in the case of supervised learning, we propose an approach which consists in reducing the dimension by an approximation of the original data matrix and then realizing LDA in the reduced space. Dimension reduction is based on low–rank approximation matrices by the use of random matrices. A fast approximation algorithm of the SVD and a modified version as fast approximation by spectral gap are developed. Experiments are done with real images and text data. Compared to other methods, the proposed approaches provide an error rate that is often optimal, with a small computation time. Finally, our contribution in transfer learning consists in the use of the subspace alignment and the low-rank approximation of matrices by random projections. The proposed method is applied to data derived from benchmark database; it has the advantage of being efficient and adapted to large-scale data

Texto completoThe objective of this thesis is to evaluate the interest of 3D molded interconnect devices technologies 3D-MID for wireless power transfer (WPT) through electromagnetic induction. WPT systems mostly uses planar coils that allows transfer between receiver and emitter at low and mid-range distance, at the condition that they are well aligned. We studied a specific case with a 3D receiver enclosed in a half meter box with 4 emitting inductors on 4 sides. Three questions were examined: the magnetic resonance of 3D-MID inductors at 6.78 MHz, their dimensions and their 3D shape. The nearly spherical shape 3D-MID receiver (Ø 8 cm) was 3D printed, activated with laser direct structuring (LDS), autocatalytic metallization and electroplating. It has 3 solenoid receiving inductors, each with quality factor above 129 ±10, placed orthogonally on the equators. The experimental results show: (1) the receiver is able to receive 4.33 W at 15.8 % efficiency in the middle of the box and (2) that we can change position and orientation of the receiver in the box, the placement of the inductors allows, in a widely meaning, to mean the received power. In conclusion 3D-MID allows to integrate, relatively easily, inductors for WPT, in the casing of electronics devices, in the same way as for 3D-MID electromagnetic antennas in smartphones. These inductors can 3D-shape the casing, which will allow the design of omnidirectional receivers

Texto completoPyrolysis is the primary step in all thermochemical transformations of solids and therefore the detailed characterization of the pyrolysis products and the understanding of the involved mechanisms are mandatory. In this context, this PhD thesis aims to study the slow pyrolysis of PET using different analytical devices. In the first part, slow pyrolysis experiments (5 °C/min) were carried out in a tubular reactor for four final temperatures ranging from 410 °C to 480 °C. Unlike other studies from the literature, light products as well as heavy products were identified. The non-condensable gas was analyzed online using Gas micro-Chromatography (μ-GC), which allows acquiring the gas temporal profiles. The carbonyl compounds were trapped, derivatized and analyzed by High Performance - Liquid Chromatography (HPLC). The waxy products were characterized by Fourier Transform Infrared Spectroscopy (FTIR) and by Ion Cyclotron Resonance Mass Spectrometry (FT-ICR MS) using an Electrospray Ionization (ESI). Some waxy molecules were quantified by Gas Chromatography / Mass Spectrometry and Flame Ionization Detection (GC/MS-FID). The characterization of the solid residue was performed by FT-ICR MS coupled with Laser Desorption ionization (LDI). In the second part, the formation kinetics of volatile compounds during PET pyrolysis was studied using a Thermogravimetric Analyzer coupled with mild ionization methods (SPI: Single Photon Ionization; REMPI: Resonance Enhanced Multiple Photon Ionization; APCI: Atmospheric Pressure Chemical Ionization). The combination of these different techniques allowed a detailed characterization of the products and consequently the identification of reaction pathways of PET degradation, which are mostly molecular pathways

Texto completoAlthough the prebiotic challenge is important, the study of aerosols in the atmosphere of Titan, Saturn's largest moon, remains a challenge for the scientific community. Indeed, the data collected by the Voyager and Cassini-Huygens probes made it possible to retrieve the first information but not to unlock the secrets of this particle fog. The purpose of this thesis is to study the chemical composition of these aerosols through their reproduction on Earth. The first approach presented is molecular characterization using high resolution mass spectrometry. In this section, the comparison between the soluble and insoluble fraction of these samples and their ageing will be presented. Then, the investigation of the growth of these aerosols is presented through the use of tandem mass spectrometry. The last part of this thesis presents the study of the three-dimensional structure. For this purpose, ion mobility coupled with mass spectrometry has been used and will be reported in two parts, one in TWIMS-Synapt and the other in TIMS-FTICR. Finally, a study in Atomic Force Microscopy will be presented and will make it possible to show real structures of molecules present in aerosols thanks to the magnification provided by this type of microscope
