Using a spatially-resolving x-ray spectrometer system, toroidal impurity rotation in Alcator C-Mod plasmas was measured. The propagation of the rotational velocity from the edge to the core of the plasma column was measured during the L- to H-mode transition. Momentum transport was measured in both Ohmic and ICRF-heated discharges, which produced EDA and ELM-free H-modes. The momentum transport was modeled by a simplified diffusion model, in which momentum diffusivity was substantially higher than neoclassically predicted values.

Empirical energy confinement scalings play a crucial role in the design of tokamak fusion reactors, measuring how quickly energy is transported by turbulence from the fusion-producing core to conduction loss at the edge. Unfortunately, experiments often exhibit discontinuous changes in scaling behavior as the plasma parameters are varied, termed confinement transitions. Navigating these transitions requires an understanding of the physical origin and limits of confinement scalings, and is crucial for retiring the physics risk of extrapolating empirical results to future reactors. This thesis explores the connection between two universally observed transitions in tokamak transport: the Linear to Saturated Ohmic Confinement (LOC/SOC) transition and the concomitant intrinsic rotation reversal. Analysis and modeling of rotation reversal hysteresis experiments show that a single turbulent bifurcation underlies both transitions on Alcator C-Mod.
Plasmas on either side of the reversal exhibit different toroidal rotation profiles and therefore different turbulence characteristics despite profiles of density and temperature which are indistinguishable within measurement uncertainty. Elements of this bifurcation are also shown to persist for auxiliary heated L-modes. Within a reduced quasilinear transport model, the deactivation of subdominant (in linear growth rate and contribution to heat transport) ion temperature gradient (ITG) and trapped electron mode (TEM) instabilities is identified as the only possible change in turbulence across the reversal which is consistent with the measured profiles and inferred heat and particle fluxes. Experimental constraints on a possible change from strong to weak turbulence, outside the description of the quasilinear model, are also discussed.
These results indicate an explanation for the LOC/SOC transition that provides a mechanism for the hysteresis through the dynamics of subdominant modes and changes in their relative populations, and does not involve a change in the most linearly unstable ion-scale drift-wave instability. This work highlights the importance of considering the dynamics of the entire mode spectrum, and not just the dominant modes, in making predictions about transport and confinement regimes.

We perform extensive analysis on the physics of L-mode tokamak fusion reactors to identify (1) a favorable parameter space for a large scale steady-state reactor and (2) an operating point for a minimum scale steady-state reactor. The identification of the large scale parameter space is part of the 2008 MIT Nuclear Systems Design Project, which also includes sustainability and economic optimizations to identify a plausible operating point for a large scale (a 14 m major radius) hydrogen production reactor dubbed HYPERION. Due to the potentially prohibitive capital cost (a $50 billion) and exorbitant thermal power (a 35 GWth) of HYPERION, we identify a conservative estimate for the minimum scale of a similar steady-state L-mode reactor of approximately 7.5 meters, half the size of HYPERION and only 20% larger than ITER. This minimum scale reactor would require an on-coil magnetic field of a 16 T and a blanket power density of ~ 5 MW/m 2 . It would produce 7 GWth of power with a power gain of 30, and it would operate far from all stability and confinement limits. To confirm the viability of this operating point, we perform various 1-D calculations. The crucial advantage of a steady-state (or fully non-inductive) reactor is that it is not limited by flux swing and can operate continuously, recharging its solenoid during operation. The crucial advantages of L-mode are that it avoids instabilities associated with edge localized modes (ELMs) and that it allows volumetric heating in the mantle due to the absence of a pedestal. Steady-state L-mode tokamak reactors could be the future of controlled fusion research and even play an important role in meeting the world's clean energy needs.

The production of hepcidin is modulated according to the iron demands of the body : in the presence of iron, the synthesis of hepcidin is increased to avoid the excess of the metal which is toxic, and, on the contrary, hepcidin is decreased to allow an effective mobilization of the metal. I participated in the characterization of the phenotype of mouse KO HEPC1 which present a multivisceral iron overload with a particularly important hepatic and pancreatic accumulation of iron. In this murin model, I tried to establish the link between the iron metabolism and the glucose metabolism. I studied the mechanisms of physiopathological regulation of the hepcidin by the iron in a system of primary culture of hepatocytes. To finish this subject, the works considering directly the stakes of a pluriprofessional health team, will be presented. The second part of this thesis is devoted to the practical research. Observations were led during the synthesis meetings in four parisian wards : -unity of pediatric hemodialisis, unity of bone marrow transplant, ward of general pediatric and ward of infantile cardiology. What was said between all the present professionnals, is analysed with a quantitative and qualitative approach. The author concludes to the sporadic aspect of the work in a pluridisciplinary team in a pediatric ward in the sample hereby studied. (431 ref. )

The most prevalent criticism of fission-fusion hybrids is simply that they are too exotic - that they would exacerbate the challenges of both fission and fusion. This is not really true. Intriguingly, hybrids could actually be more viable than stand-alone fusion reactors while mitigating many challenges of fission. This work develops a conceptual design for a fission-fusion hybrid reactor in steady-state L-mode tokamak configuration with a subcritical natural or depleted uranium pebble bed blanket. A liquid lithiumlead alloy breeds enough tritium to replenish that consumed by the D-T fusion reaction. Subcritical operation could obviate the most challenging fuel cycle aspects of fission. The fission blanket augments the fusion power such that the fusion core itself need not have a high power gain, thus allowing for fully non-inductive (steady-state) low confinement mode (L-mode) operation at relatively small physical dimensions. A neutron transport Monte Carlo code models the natural uranium fission blanket. Maximizing the fission power while breeding sufficient tritium allows for the selection of an optimal set of blanket parameters, which yields a maximum prudent fission power gain of 7.7. A 0-D tokamak model suffices to analyze approximate tokamak operating conditions. If the definition of a "reactor" is a device with a total power gain of 40, then this fission blanket would allow the fusion component of a hybrid reactor with the same dimensions as ITER to operate in steady-state L-mode very comfortably with a fusion power gain of 6.7 and a thermal fusion power of 2.1 GW. Taking this further can determine the approximate minimum scale for a steady-state L-mode tokamak hybrid reactor, which is a major radius of 5.2 in and an aspect ratio of 2.8. This minimum scale device operates barely within the steady-state L-mode realm with a thermal fusion power of 1.7 GW. This hybrid, with its very fast neutron spectrum, could be superior to pure fission reactors in terms of breeding fissile fuel and transmuting deleterious fission products. It could operate either as a breeder, producing fuel for pure fission reactors from natural or depleted uranium, or as a deep burner, fissioning heavy metal and transmuting waste with a cycle time of decades. Despite a plethora of potential functions, its primary mission is deemed to be that of a deep burner producing baseload commercial power with a once-through fuel cycle. Although hybrids are often purported a priori to pose an elevated proliferation risk, this reactor breeds plutonium that could actually be more proliferation-resistant than that bred by fast reactors. Furthermore, a novel method (the "variable fixed source method") can maintain constant total hybrid power output as burnup proceeds by varying the neutron source strength. As for engineering feasibility, basic thermal hydraulic analysis demonstrates that pressurized helium could cool the pebble bed fission blanket with a flow rate below 10 m/s. The Brayton cycle thermal efficiency is 41%. This device is dubbed the Steady-State L-Mode Non-Enriched Uranium Tokamak Hybrid (SLEUTH). The purpose of this work is not any sort of elaborate design, but rather the exploration of an idea coupled with corroborating numerical analysis. At this point in the hybrid debate, viable conceptual designs are persuasive while intricate build-ready designs are superfluous. This work conceives such a conceptual design, demonstrates its viability, and will perhaps, incidentally, spur a profusion of pro-fusion sentiment!

Texto completoRecognizing the current and future requirements of extending Telemetry, TT&C, Earth Resources operations into the C-Band (4400-5200 MHZ) arena, Orbit Communication Systems and Orbit Communication Ltd. are currently designing and testing a Tri-Band auto-tracking / receive / transmit feed technology combining dual polarization with multiple frequency band coverage (L/S/C) into a single feed that can be fitted onto new or existing antenna systems. This technology reduces footprint and minimizes life cycle cost, both of which are important considerations for both our commercial and military customers. This technology has been applied to a number of systems, including flyaway, fixed (or teleport) and shipboard, and is readily applicable to ground and mobile applications. The multiband feed design consists of coaxial, concentric waveguide cavities operating in TE11 and TE21 modes (Patent Pending). The large outermost cavities of the feed operate in the L/S frequency band while the innermost cavities operate at C-Band. The antenna is fed with orthogonally polarized inputs/outputs enabling polarization diversity in all bands. The coaxial cavity feed is ideal for this application because the feed produces high-efficiency, near-optimum illumination patterns and coincident phase centers in all three bands simultaneously. Because it is a waveguide design, the input power levels used in Telemetry and Satellite systems do not present breakdown problems. This type of feed has proved to be an ideal multiband illuminator for parabolic reflectors, and therefore could be used in many military and commercial applications that require multiband operation.

What is described herein is the final configuration as delivered, antenna test range results, on-site results, and lessons-learned during the development and implementation phases. However, through procedures of control, the context of implementation has totally changed. CNC are increasingly complex. The NC programming has to be optimised and controlled. This evolution necessitates the development of convivial and interactive Human/machine interfaces that induced constraints in training and in utilisation. Through the elaboration of a teaching software destined for acquire the ability to programme CNC machine tool with ISO language, we approached the pedagogical dimension of mechanic. Furthermore, a verification of the precision of CNC programs is realised. The operational context (Characteristic of CNC, choice of tools) has been processed so as to put the operator in the context. A convivial interface, seconded by different types of help, authorises the use of this module by beginners or by experts. Syntactic, lexical and grammatical analysis of typed information's is undertaken in real manner time in order to address to the user relevant messages adapted at its level. The analysis module uses an inference method and algorithmic one, they are developed in C++ language. Developed with collaboration of industrial partners, the software realised shows the feasibility of strongly interactive products integrating real constraints of the industrial world

Texto completoThe main purpose of this thesis concerns the problem of spatial prediction using some nonparametric conditional models where the covariate variable is a functional one. More precisely, we treat the nonparametric estimation of the conditional mode and that of the conditional quantiles as spatial prediction tools alternative to the classical spatial regression of real response variable given a functional variable.Concerning the first model, that is the conditional mode, it is estimated by maximizing the spatial version of the kernel estimate of the conditional density. Under a general mixing condition and the concentration properties of the probability measure of the functional variable, we establish the almost complete convergence (with rate), the Lp consistency (with rate) and the asymptotic normality of the considered estimator. The usefulness of this estimation is illustrated by an application on real meteorological data.The model of the conditional quantiles is considered in the second part of this thesis and is treated as the inverse function of the conditional cumulative distribution function which is estimated by a double kernel estimator. Under the same general conditions as in the first model, we give the convergence rate in the Lp- norm and we show the asymptotic normality of the constructed estimator. These asymptotic results are closely related to the concentration properties on small balls of the probability measure of the underlying explanatory variable and the regularity of the conditional cumulative distribution function.Our study generalizes to spatial case some existing results in functional times series case. Finally, we highlight what our models brings compared to classical regression, discussing the use of our results as preliminary works to construct predictive regions

Texto completoDue to its phylogenetic position at the base of the phylogenetic tree of Vertebrates, the dogfish is an emergent model to study fundamental biological process such as spermatogenesis and to identify bioactive peptides. During this work, we have shown that GFRα1and POU2/POU5F1 are conserved markers of undifferentiated spermatogonia from Chondrychtyes to Mammals whereas PLZF and c-KIT presented a less specific expression pattern and that HMGB2, HMGB3 and MCM6 may be new markers. Then, the development of a culture system of dogfish spermatogonia allowed us to show that GDNF function on self-renewal maintenance was also conserved. However, sequences study showed that the ARTN may replace the GDNF to induce self-renewal through GFRα1 in the dogfish. In parallel, the study of the action mode of three peptides isolated from dogfish testis and of two from the epigonal tissue showed antineoplastic effects by affecting various cellular processes. Thus, even if two peptides were further characterized, the three others seem to directly affect the cytoskeleton, which was followed by various cellular perturbations depending on the cell line and the peptide, such as cell-cycle slowing down, autophagy inhibition, necrosis or membrane destabilization. Complementary studies must be done to confirm the first target and identify which cytoskeleton element is involved. Those results suggest finally that the dogfish is a suitable model to study spermatogenesis evolution as well as to search bioactive peptides

Resumo: Introdução: A candidíase oral é uma infecção fúngica oportunista da cavidade oral, cujas taxas de prevalência e incidência vêm aumentando significativamente em todo o mundo. Assim, novas estratégias orientadas para gerir esta doença têm sido propostas, dentre as quais está o uso de óleos essenciais (OE) com propriedades antifúngicas. Evidências indicam que o OE de Coriandrum sativum L. (coentro) é um forte agente antifúngico contra Candida e, portanto, investigações devem dar continuidade ao conhecimento gerado. Objetivo: Este estudo buscou avaliar a atividade antifúngica e modo de ação do OE de C. sativum sobre Candida spp., e determinar os alvos moleculares afetados na expressão global do genoma humano. Material e Métodos: C. sativum foi obtido a partir do Banco de Germoplasmas do Centro Pluridisciplinar de Pesquisas Químicas, Biológicas e Agrícolas (Universidade Estadual de Campinas, SP, Brasil) cujo OE e fração ativa tiveram o perfil fitoquímico determinado por cromatografia gasosa acoplada a espectrometria de massa. Posteriormente, foram realizados testes com cinco cepas de referência de Candida: Determinação da Concentração Inibitória e Fungicida Mínima (CIM/CFM); modo de ação antifúngica (ensaio do sorbitol e ergosterol); Microscopia Eletrônica de Varredura (MEV) de biofilmes de Candida e testes de inibição de aderência em biofilme. Utilizou-se nistatina, anfotericina B ou caspofungina como controles positivos, além de controles negativos. Também, foi testado o efeito do OE sobre a atividade proteolítica de C. albicans. Por fim, um ensaio de farmacogenômica identificou quais alvos moleculares no genoma humano foram afetados pelo OE e fração ativa de C. sativum. Os testes foram realizados em triplicata de experimentos independentes e os dados foram tratados estatisticamente (ANOVA, pós-teste de Tukey, ?=0,05). Os dados da análise farmacogenômica foram processados nas plataformas GeneGo MetaCore® e David Bioinformatics Resources. Resultados: O perfil fitoquímico EO indicou monoterpenos (37,9%) e sesquiterpenos (62,1%) como compostos principais. Os valores de CIM/CFM para o OE variaram de 15,6 a 62,5 µg/mL. Quanto ao modo de ação, o OE de C. sativum parece se ligar ao ergosterol da membrana celular fúngica, aumentando a permeabilidade iônica e causando morte celular; entretanto, o OE não atua sobre vias de biossíntese da parede celular. Estes achados confirmam as alterações na integridade da morfologia do biofilme verificadas nas análises por MEV. Além disso, o OE apresentou atividade antiaderente em biofilme em baixas concentrações (15,6-62,5 µg/mL) contra as cepas testadas, bem como atividade contra proteases produzidas por C. albicans, sendo estatisticamente significante na CIM (p<0,05). Finalmente, o OE e sua fração ativa apresentaram baixa citotoxicidade em células humanas com CI30 de 359,8 e 366,7 µg/mL, respectivamente. As principais vias afetadas estão relacionadas com quimiocinas e MAP-quinase (apoptose, proliferação) bem como proteínas de adesão. Conclusões: O OE das folhas de C. sativum tem forte atividade antifúngica e antiaderente sobre Candida spp. e atividade anti-proteolítica sobre C. albicans, e atua aumentando a permeabilidade iônica da membrana celular, provavelmente devido ao efeito sinérgico de mono e sesquiterpenos. Análise farmacogenômica indicou baixa citotoxicidade do OE e sua fração ativa com alvos moleculares específicos afetados no genoma humano, o que incentiva o desenvolvimento de novas pesquisas pré-clínicas toxicológicas e clínicas nesta área
Abstract: Introduction: Oral candidiasis is a common opportunistic fungal infection of the oral cavity with increasingly significant worldwide prevalence and incidence rates. Accordingly, novel specific-targeted strategies to manage this ailment have been proposed, among which is the use of essential oils (EO) with antifungal properties. Coriandrum sativum L. (coriander) EO has proven antifungal activity against Candida species and thus deserves further investigation. Objective: This study aimed to evaluate the antifungal activity and mode of action of the EO from Coriandrum sativum L. leaves on Candida spp., and to determine the molecular targets affected in the whole-genome expression in human cells. Material and Methods: C. sativum was obtained from the Germoplasm Bank of the Research Center for Chemistry, Biology and Agriculture (University of Campinas, SP, Brazil) whose EO and active fraction had their phytochemical profile determined by Gas Chromatography coupled to Mass Spectrometry. Then, we carried out the following tests with five reference strains of Candida spp. (CBS): Minimum Inhibitory and Fungicidal Concentration (MIC/MFC); antifungal mode of action (sorbitol and ergosterol assays); Scanning Electron Microscopy (SEM) analysis of Candida biofilm and tests of inhibition of biofilm adherence. We used nystatin, amphotericin B or caspofungin as positive controls, in addition to negative controls. Also, we tested the effect of C. sativum EO on the proteolytic activity of C. albicans. Next, a pharmacogenomic assay identified which molecular targets in the human genome were affected by C. sativum EO and its active fraction. Tests were performed in triplicate of independent experiments and data were statistically treated (ANOVA, Tukey¿s post-test, ?=0.05). Pharmacogenomic data were processed on GeneGo MetaCore® and DAVID Bioinformatics Resources. Results: The EO phytochemical profile indicated monoterpenes (37.9 %) and sesquiterpenes (62.1 %) as major compounds. The MIC/MFC values for the EO ranged from 15.6 to 62.5 µg/ml. With regard to the mode of action, C. sativum EO may bind to membrane ergosterol, increasing ionic permeability and causing membrane damage to cell death, but it does not act on cell wall biosynthesis-related pathways. This mode of action confirms the changes in the integrity of the biofilm morphology as verified in the analyses by SEM. The EO showed anti-adherent activity at low concentrations (31.2 ¿ 62.5 µg/ml) against all strains tested, as well as activity against proteases produced by C. albicans, with statistical significance at MIC (P < 0.05). Finally, the EO and its active fraction had low cytotoxicity on human cells with IC30 of 359.8 and 366.7 µg/ml, respectively, affecting the pathways of chemokines and MAP-kinase (apoptosis, proliferation), as well as adhesion proteins. Conclusions: The EO from C. sativum leaves has strong antifungal and anti-adherent activity against Candida spp., as well as anti-proteolytic activity against C. albicans, and acts by increasing cell membrane ionic permeability, probably due to the synergistic effect of mono- and sesquiterpenes. Pharmacogenomic analyses revealed low cytotoxicity with specific targets affected in the human genome, which encourages further pre-clinical toxicological screening and clinical research in this field

Texto completoThe adenosyl-L-homocysteine hydrolase (AdoHcy hydrolase) is regarded today as a good pharmacological target fot the design of new broad spectrum antiviral agents. Throughout this study, we sought to exploit the catalytic mechanism of the enzyme and the structural data recently published to cause its inhibition. The family of esters homologous of adenosine was revealed to be a family of the type I inhibitors assigning cofactor NAD+. The thionucleosides reached their target : Cysteine 195. Studies of inactivated protein in ESI-MS then studies of the peptides obtained after digestion by EndoLys C of inhibited enzyme enable us to confirm the mechanism of action of these compounds. The evaluation of the acyclonucleosides is always in hand but the preliminary results remain encouraging and open the way towards new structures

Texto completoResults showed that the ash content, crude proteins, available sugars, total oxalate, starch and phosphorus content increased significantly with the maturity stage. On the other hand the amylose content decreased as function of the maturity. The water adsorption capacity, the water solubility index, the temperature and the enthalpy of gelatinization of the flour and starch increased significantly with the maturity stage. Irrespective of the degree of maturity the starch granules diffractogramm were exhibited an "A" type cristallinity. The browning indicators increased significantly (p<0.05) with the maturity stage of the tuber. The color coordinate L* of the flours and that of its dough increased while a* decreased with the maturity stage. These two coordinates lend themselves better with electric drying than solar drying. The sensory characteristics of the taro Sosso dough were positively affected by the maturity stage and electric drying. The overall acceptability of taro Sosso dough was correlated with its color. These results suggest that 10 months of the taro Sosso maturity and the electric drying (40°C) are the best conditions to obtain the acceptable pastes. These optimum conditions might be used to produce the taro flour at industrial scale

Texto completoThis thesis deals with the study of asymptotic properties of e kernel (Parzen-Rosenblatt) density estimate under associated and censored model. In this setting, we first recall with details the existing results, studied in both i.i.d. and strong mixing condition (α-mixing) cases. Under mild standard conditions, it is established that the strong uniform almost sure convergence rate, is optimal. In the part dedicated to the results of this thesis, two main and original stated results are presented : the first result concerns the strong uniform consistency rate of the studied estimator under association hypothesis. The main tool having permitted to achieve the optimal speed, is the adaptation of the Theorem due to Doukhan and Neumann (2007), in studying the term of fluctuations (random part) of the gap between the considered estimator and the studied parameter (density). As an application, the almost sure convergence of the kernel mode estimator is established. The stated results have been accepted for publication in Communications in Statistics-Theory & Methods ; The second result establishes the asymptotic normality of the estimator studied under the same model and then, constitute an extension to the censored case, the result stated by Roussas (2000). Texto completoSpatial representation of modeling area in travel demand models has changed little over the course of last several decades. In this regard, the state-of-the-art still widely relies on the same centroid-connector system that has been used in classic models. In this approach continuum bidimensional space is lumped on centroids. It is an aggregate approach which ignores the physical variability linked to the scatteredness of disaggregate residence- and activity-places over the local space. Consequently the modeling performance in explaining route and mode choice behavior degrades at local scales: In route choice, disaggregate location influences the propensity between a distant interchange to a highway, or a nearby road. In mode choice, feeder service to public transportations influences the auto vs. transit modal share. We propose a disaggregate approach for spatial representation. Based on a zoning system, a stochastic disaggregate representation is used to characterize the space within a traffic analysis zone. For each zone, anchor-points are defined as the network nodes that are used for accessing to the network from within the local space. An itinerary between a pair of zones is then considered as a chain of legs composed of two terminal legs, corresponding to the intrazonal route sections, and one main leg between two anchor points. The route choice problem is transformed to a joint choice of a pair of anchor points. The vector of random terminal travel times is Multivariate Normal resulting in a Multinomial Probit model of choice of a pair of anchor points. To extend to the multimodal context, a transit composite mode is defined as a chain of access, main, and egress modal legs, and transit platforms are considered as anchor points connecting the feeder legs to the main line-haul leg. A Multinomial Logit mode choice model is estimated based on the 2001 Paris Household Travel Survey for the auto mode and the composite transit modes. It is joined with the two Multinomial Probit models corresponding to the choice of anchor points. The result is a joint model of mode and station choice with a disaggregate representation of the local space
