In this paper, authors analyzed the semi-rigid road base asphalt pavement of towns and studied the optimum design methods of it, combining characteristics of town roads, considering natural conditions, vehicle axle,materialsupply,construction techniques, maintenance, transportation and so on, referring to design experiences of asphalt pavement in Tian Jin area, through indoor and outdoor experiments, applying math methods. In addition,optimum pavement structure function and calculating methods is given out, asphalt pavement structure adapt to Tian Jin area is recommended and CAD program is developed.

This paper adopts the unique perspective of Confucian aesthetic principles to investigate their profound impact on the interrelationship between “Ren” (benevolence) and spatial design in classical Chinese architecture. Drawing upon core Confucian concepts such as “Ren”, “Tian Ren He Yi” (the unity of man and nature), and “Jin Shan Jin Mei” (the pursuit of perfection), the study meticulously analyzes specific instances of classical architectural layouts and spatial designs, providing a detailed exposition of the manifestation of “Ren”. Subsequently, it elucidates how Confucian aesthetic principles influence classical architecture by inheriting and developing the spirit of benevolence and adeptly applying an aesthetic consciousness of the unity of space and time. Simultaneously, the paper distinctly presents the profound insights of Confucian aesthetic principles on contemporary spatial design and societal development. This influence extends beyond the formal and structural aspects of architecture, permeating deeply into the cultural connotations embedded in architecture. It injects distinctive and valuable design wisdom into contemporary society, contributing not only to the comprehension of the cultural value of ancient architecture but also furnishing a novel framework for contemporary spatial design. In doing so, it offers concrete and far-reaching guidance for the construction of a harmonious society.

We report the continuous argon ions irradiation of itinerant Fe3GeTe2, a two-dimensional ferromagnetic metal, with the modification to its transport properties measured in situ. Our results show that defects generated by argon ions irradiation can significantly weaken the magnetization (M) and coercive field (Hc) of Fe3GeTe2, demonstrating the tunable magnetism of this material. Specifically, at base temperature, we observed a reduction of M and Hc by up to 40% and 62.4%, respectively. After separating the contribution from different mechanisms based on the Tian-Ye-Jin (TYJ) scaling relation, it’s the skew scattering that dominates the contribution to anomalous Hall effect in argon ions irradiated Fe3GeTe2. These findings highlight the potential of in situ transport modification as an effective method for tailoring the magnetic properties of two-dimensional magnetic materials, and provides new insights into the mechanisms underlying the tunable magnetism in Fe3GeTe2.

The study aims to identify the influence of V. Mayakovsky’s satire on Chinese drama. The paper examines the works of Chinese authors created under the influence of V. Mayakovsky’s satirical play “The Bedbug”. In Tian Han’s play “The Ballad of the Ming Tombs Reservoir” and in Jin Shan’s eponymous film, the idea of moving into the future determines the plot and the compositional structure. Meng Jinghui’s drama “The Bedbug” (the 2000, 2013 and 2017 productions) repeats the main plot of V. Mayakovsky’s play and retains many lines from the original work. This is the first staging of Mayakovsky’s play in China. The scientific novelty of the study lies in the fact that Mayakovsky’s influence on Chinese drama is for the first time being considered from the cultural and historical perspective and reflects the dynamics of perception of Mayakovsky’s satire in China starting with a romantic-utopian view, associated with the concept of the “Great Leap Forward” (Tian Han), to a postmodern view (Meng Jinghui). The study of the Chinese-language material on the reception of Mayakovsky’s satire, which remains unexplored in Russia, will make it possible to expand the understanding of the attitude to the creative work of the Russian futurist poet and playwright abroad not only among literary scholars, but also the reading public. The study found that the satirical comedy by the Russian poet of the turn of the 19th-20th centuries in the deconstructed versions reflects the problems of the formation of Chinese society in the 20th-21st centuries.

To accurately describe the deformation characteristics of clay under long-term cyclic load, based on fractional calculus theory, elastoplastic theory and the basic element model, a variable-order fractional dynamic model designed to predict accumulative strain of clay was exhibited. Firstly, the cyclic load was separated into static and alternating load in accordance with cyclic load characteristics, and the strain of clay under static and alternating load was analyzed. Then, on the basis of the variable-order Abel dashpot model, rheological theory and elastoplastic theory, the expressions of the rheological constitutive model and strain response were both obtained. Finally, in combination with the undrained dynamic triaxial testing of Zhan Jiang clay and Tian Jin soft clay, a series of analyses was carried out on the effectiveness and parameter sensitivity of the model when subjected to long-term cyclic loading. By comparing the dynamic constitutive model with pre-existing models, the superiority of the dynamic constitutive model is revealed. The results show that the dynamic constitutive model can characterize properly the deformation characteristics of clay under the action of long-term cyclic loading, especially in its accelerating stage. The parameter sensitivity of the model exhibits a growing trend with the increment of loading duration.

Oracle-bone scholars have long uniformly read the charges (i.e., the portion that mentions the event being divined) of Shang oracle-bone inscriptions as questions. Since the 1970's, many foreign oracle-bone scholars have proposed a new view that the charges are never questions, or at least are generally not questions. Whether or not the charges are actually questions is a very important question bearing not only on oracle bone studies, but also on research on ancient divination and on ancient Chinese grammar. Based on an examination of a great number of Shang oracle-bone inscriptions, the author of this essay believes that charges that can now be determined to be questions are mainly those choice-type charges in early inscriptions that contain the final particles yi and zhi, and true-or-false type charges that contain yi. Charges of the type “V bu V” (such as “rain not rain” [yu bu yu]) and “V bu,” regarded by many as choice-type questions, are actually comprised of the charge and a verification or use notation, the “bu V” being a verification and the “bu” being either a verification or a use notation (similar to the notation bu yong, “do not use”). Charges that can now be determined not to be questions are mainly in those inscriptions with complex charges, such as “this *spring the king ought not ally with Wang Cheng to attack Xia Wei, (for if he does) he will not receive divine assistance” (jin ?chun wang wu bi Wang Cheng fa Xia? Wei, fu qi shou you you, and “On ren we ought not hunt, (for if we do) it will rain” (ren wu tian, qi yu), the syntax of which shows that they cannot be questions.

Abstract Genomic discoveries of oncogenic driver alterations in lung cancer have led to development of effective target therapeutics; however, patients who initially respond to the therapy inevitably experience regrowth of the disease. Recent studies identified a reversible stage of cancer cells entering a quiescent state to ensure survival, called drug-tolerant persister (DTP) cells. The DTP stage is viewed as a major source of non-genetic drug resistance, while its epigenetic dynamic and regulatory mechanisms remain largely unknown. To determine the epigenetic activity reminiscent of a quiescent state during DTP development, we evaluated repressive epigenetic modifications during DTP progression and found that DTP cells had a markedly increased H3K27me3 level at the beginning of DTP stage. Treatment of EZH2 inhibitor together with target therapy synergistically inhibited cell growth, indicating the essential role of compressed chromatin structure for DTP survival. To further investigate the epigenomic heterogeneity of DTP cells, we performed single cell ATAC-seq on DTP cells. Unsupervised hierarchical clustering analysis revealed that the drug-induced DTP cells had two distinct populations compared with the untreated group with only one major population, suggesting an increased epigenomic heterogeneity. By analyzing the differentially accessible regions, motifs for TEADs and GATAs were specifically enriched in DTP cells, whose family members are well-established regulators of the lung development. This raised a possibility that the evolution of drug resistance shares similarities with organ regeneration, both require terminally differentiated cells regaining cell plasticity and serving as a progenitor cell to direct re-differentiation. Thus, we focused on alveolar lineage genes as a model to test our hypothesis that DTP cells hijack conserved developmental pathways for lung regeneration to ensure their survival and subsequent expansion. Our results showed that specific alveolar marker genes were transiently upregulated and exhibited translocation from cytoplasm to nuclei at early phase of DTP. Moreover, deletion of the alveolar lineage gene decreased cell re-growth under drug treatment. Our finding provides a new perspective of anti-relapse therapeutic strategies, that targeting lung lineage pathways to manipulate the activity of critical lineage factors during lung regeneration, for patients with minimal residual disease and relapse after target therapy. Nature is an excellent source of inspiration for the widespread application of science and technology. Researchers learn from biological principles and gradually develop from the earliest direct use of biological materials to the design and development of new biomimetic materials inspired by nature. Therefore, the innovation of bioinspired nanomaterials will enable tremendous advances in biology and biochemistry with biomimetic capabilities, such as biosensing, cell imaging, and drug delivery. In this presentation, I will discuss my group’s recent work on two bioinspired nanomaterials: nano-peptoids and single-atom nanozymes. The first type of bioinspired materials is a sequence-defined peptide-like materials that can mimic the structure and function of peptides and proteins. Compared with peptides and proteins, peptoids have a high degree of thermal and chemical stability, and are resistant to proteolytic degradation. In addition, due to the lack of intramolecular and intermolecular backbone hydrogen bond donors, it is possible to precisely control the peptoid and peptoid-substrate interactions, leading to bio-inspired synthesis of nanomaterials with a layered structure. As a peptide mimic, the peptoids has biocompatibility, non-toxicity and sequence-specific heteropolymers. Nano-peptoids can be synthesized with polar and hydrophobic monomers and have the following advantages: They can be easily processed into desired shapes (e.g. dendritic, sheets, tubes and vesicles), and the size range is 2-100 nm to enhance cell targeting and uptake. The application of nano-peptoids in cell imaging and cancer theranostics will be discussed. Another type of bioinspired nanomaterials with enzyme-like properties is single-atom nanoenzymes. As a biocatalyst, natural enzymes have the ability to accelerate various reaction rates with extremely high activity and selectivity under mild conditions. However, due to the temperature and pH-related denaturation of proteins and the possibility of contamination by bacteria or other reagents, the shelf life of natural enzymes is limited or uncertain, and their purification and production costs are high. Therefore, natural enzymes must narrow the range of conditions under which they can be used, limiting their further applications in biosensing and biomedicine. R Recent research has focused on the rational design of nanomaterials with inherent enzyme-like catalytic properties (for example, oxidase, peroxidase, catalase, superoxide dismutase, and so on) and exploration of their applications in various fields. Although much progress has been made, the development of nanozymes is still hampered by several challenges. Catalytic activity and substrate selectivity are the two most important issues that need to be resolved to allow the technology to reach full maturity. Therefore, rational design and tuning of nanomaterials is highly desirable for the development of novel enzyme-mimic nanomaterials with precise catalytic sites. We have developed various Fe-N-C based single atomic site catalysts which have similar structure with active site of peroxidase. These atomically-dispersed Fe-N-C materials have enzyme-like properties, therefore they are classified as single-atom nanozymes. Abstract Pancreatic ductal adenocarcinoma (PDAC) is the third leading cause of cancer death in the United States, with a 10% 5-year survival rate. Immunosuppressive myeloid cells, including tumor-associated neutrophils (TANs), contribute to tumor development and treatment resistance. Lysine (K)-specific demethylase 6A (KDM6A) is one of the most frequently mutated epigenetic genes and a tumor suppressor in PDAC. However, the molecular mechanisms by which KDM6A contributes to PDAC development and whether KDM6A impacts the tumor immune microenvironment are unknown. This study established a genetically engineered pancreas-specific Kdm6a-knockout Ptf1aCre;LSL-KrasG12D/+;LSL-p53R172H/+ (KPC) PDAC mouse model to investigate the influence of KDM6A loss on PDAC development and tumor immune microenvironment. We found that KDM6A loss accelerated PDAC progression and increased metastases. Pancreata of mice with Kdm6a deficiency developed aggressive undifferentiated PDACs. Additionally, KDM6A loss led to increased infiltration of TANs and neutrophil extracellular traps (NETs) formation. Mechanistically, we used Bru-seq technology to investigate the impact of KDM6A on nascent RNA transcription. We demonstrated that many chemotactic cytokines related to neutrophil recruitment, specifically CXCL1, were upregulated in KDM6A-knockout PDAC cells. We further confirmed increased CXCL1 mRNA and protein levels in KDM6A-deficient human and mouse PDAC cells. In addition, immunohistochemical staining also confirmed the upregulated CXCL1 expression in both human and murine PDAC cells with KDM6A loss. TANs were found to express CXCR2, the receptor of CXCL1, by immunofluorescent analysis. To further confirm that cancer cells with KDM6A loss can attract neutrophils in vitro, we performed chemotaxis assays using both human neutrophils derived from PLB-985 cells and mouse primary neutrophils isolated from bone marrow and conditioned media from KDM6A- knockout or knockdown PDAC cells. We found that PDAC cells with KDM6A loss promoted neutrophil recruitment in vitro compared to KDM6A-retained PDAC cells. Furthermore, the CXCL1 neutralizing antibody reversed the chemotactic property of KDM6A-deficient PDAC cells, confirming that CXCL1 is the primary chemokine mediating the neutrophil recruitment. In summary, these findings shed light on the mechanism by which KDM6A loss promotes PDAC development, regulates tumor immune microenvironment, and suggests that the CXCL1-CXCR2 axis may be a candidate target for the treatment of PDAC, especially those with KDM6A mutations. Abstract CD70 has emerged as an attractive tumor target with its over-expression in multiple solid and hematological cancers, yet tightly controlled and limited expression in normal cells. In the last decade, multiple anti-CD70 antibody drug conjugates (ADCs) have been developed, but most, if not all, of these ADC programs have been discontinued. Using an expanded genetic code to create Engineered Precision Biologics (EPBs), Ambrx has developed ARX305, a CD70-targeted next-generation antibody drug conjugate (ADC) to potentially overcome the challenges associated with earlier ADCs and treat patients with cancers over-expressing CD70. ARX305 is comprised of a proprietary, high-affinity, humanized anti-CD70 antibody stably and site-specifically conjugated to AS269, a potent microtubule inhibitor. After binding to CD70 expressed on target tumor cells, ARX305 internalizes and releases the cytotoxic payload pAF-AS269 which inhibits cellular proliferation. In vitro testing across a panel of solid and liquid tumor cell lines showed ARX305 elicited highly potent and specific cell killing across multiple tumor types with medium to high CD70 expression levels. In multiple in vivo CD70-expressing xenograft or disseminated models, ARX305 induced significant tumor growth inhibition or regression whereas unconjugated antibody exhibited poor activity. ARX305 utilizes stable oxime conjugation chemistry, a non-cleavable PEG linker, and a membrane-impermeable payload to minimize premature payload release in circulation and associated off-target toxicity. These properties of ARX305 result in high serum stability, with rodent pharmacokinetic studies confirming a similar exposure profile of ARX305 compared to unconjugated antibody and a long serum half-life. Repeat dose toxicokinetic studies in cynomolgus monkeys demonstrated ARX305 was tolerated at exposures well above therapeutic exposures in mouse pharmacology studies, indicating a wide therapeutic index. In summary, the highly potent anti-tumor activity in multiple tumor types and wide therapeutic index of ARX305 support clinical evaluation of this next generation anti-CD70 ADC. ARX305 is currently in a Phase 1 dose escalation study in China, and the IND in the United States is open. Background and Aim: A brand equity might be its monetary value, its position in a company's balance sheet, or customers' loyalty and confidence in the goods offered by the business. This study aims to explore the level of perception of customer-based brand equity of the Tian Shang Ren Jian brand in China's tea industry and to study the guideline for Customer based Brand Equity development of the Tian Shang Ren Jian brand in China's tea industry Materials and Methods: This research is a quantitative research method. The total sample size for the research will be 385 customers of the Tian Shang Ren Jian brand. The questionnaire was used as a tool to collect data. Statistical values such as percentage, frequency, mean, and standard deviation were used for data analysis. Results: The results suggest that each of the variables demonstrated elevated values, with an average score of 4.11 and a deviation from the mean of 0.64. The levels of brand awareness, image, perceived quality, and loyalty were significantly higher. Tian Shang Ren Jian's customer-based brand equity ideas can help China's tea sector build successful partnerships by emphasizing individualization and self-conceptual links. Tian Shang Ren Jian needs to improve its understanding of the brand, sales promotion, and digital media technologies to uphold consumer confidence and enhance brand/product communication. Conclusion: Tian Shang Ren Jian's customer-based brand equity ideas can help China's tea sector build successful partnerships, with an average score of high level from the mean. Managers and marketers of Tian Shang Ren Jian brand should review their situation analysis and rethink their brand identity to improve sustainability.

Abstract Background: Pyrotinib is an irreversible pan-ErbB receptor tyrosine kinase inhibitor targeting human epidermal growth factor receptor 1 (HER1), HER2, and HER4. Exploratory studies of neoadjuvant pyrotinib combined with trastuzumab-based standard chemotherapy for HER2-positive (HER2+) early or locally advanced breast cancer (BC) showed promising pathological complete response (pCR) rates of 51.6%-71%, with a manageable toxicity profile. We are to further investigate this combination strategy versus standard therapy in a randomized trial. Trial design: Pyramid is a randomized, multicenter, open-label, two-stage adaptive enrichment phase 2 trial to evaluate the efficacy and safety of pyrotinib plus trastuzumab and nab-Paclitaxel versus pertuzumab plus trastuzumab and nab-Paclitaxel as neoadjuvant treatment in patients with HER2+ early or locally advanced BC (NCT04900311). Eligible patients will be randomized 1:1 to receive four 21-day cycles of pyrotinib (400 mg, po, qd) or pertuzumab (840 mg loading dose, 420 mg maintenance doses, i.v., day 1) with trastuzumab (8 mg/kg loading dose, 6 mg/kg maintenance doses, i.v., day 1) and nab-Paclitaxel (260 mg/m2, day 1 or 125 mg/m2, i.v., day 1/8/15). After completion of neoadjuvant treatment, patients will undergo surgery, followed by standard EC regime for 4 cycles. Subsequent adjuvant therapy will be decided by investigators. Randomization was stratified by hormone receptor (HR) status (ER and/or PR positive vs. negative) and primary tumor size (2<T≤5 cm vs. T>5 cm). There will be two stage of this trial, and population may be enriched to HR-positive based on the results of interim analysis at Stage I. Eligibility criteria: Key inclusion criteria include female, treatment-naïve, histologically confirmed HER2+ invasive BC (cT2-4/cN0-3/cM0), the diameter of primary tumor > 2 cm, known HR status (ER and PR). Specific aims: Primary endpoint is total pathologic complete response (tpCR) rate per central review assessment. Secondary endpoints include invasive disease-free survival, event-free survival, objective response rate, breast-conserving surgery rate, safety. Exploratory endpoint is to investigate the possible predictive biomarkers of efficacy. Statistical methods: A sample size of 220 patients per arm is calculated to provide a power of 80% with a significance level of 0.1 (one-sided) to detect an estimated increase in the tpCR rate from 39% in the pertuzumab group to 49% in the pyrotinib group. Considering a dropout rate of 10%, 490 patients (245 per arm) will be required to be enrolled. A two-stage adaptive enrichment design will be used. The trial will enroll 186 patients at Stage I and an interim analysis will be conducted by an independent data monitoring committee (IDMC) when the pathologic response has been evaluated in at least 132 patients (including at least 66 HR-positive patients) at Stage I. Then, one of five options might be adapted at Stage II depending on the results, including: i. Continue study as planned in whole population; ii. Re-estimate sample size in whole population; iii. Enrich to HR-positive population with pre-defined sample size; iv. Enrich to HR-positive population and re-estimate sample size; v. Early termination. The Clopper-Pearson method will be used to calculate a 95% confidence interval (CI) for the tpCR rate. The CMH chi-square test will be used to compare the tpCR rate between groups and 95% CIs for the difference will be estimated using the Miettinen-Nurminen method. Background: Bronchiectasis is common in patients with Chronic Obstructive Pulmonary Disease (COPD). COPD with bronchiectasis has been considered a phenotype with worse lung function and more severe exacerbations. There is scarce literature on the characteristics and optimal management of such patients.Methods:Patients with COPD reporting within the one-year study period were subjected to High Resolution Computed Tomography (HRCT) scan of the thorax. Sputum was sent for Gram-stain and culture/sensitivity for patients found to have bronchiectasis. Bronchiectasis Severity Index (BSI) was calculated using the online BSI calculator. Association between presence of bronchiectasis and gender, lung function and frequency of exacerbations was statistically analysed.Results: Total 62 patients with COPD were enrolled. Bronchiectasis was present in 11 (17.7%) patients. The most common bacterial isolate from sputum of patients with bronchiectasis was Haemophilus influenza (54.54%). The prevalence of bronchiectasis was more in females (19.45% compared to 15.4% in males), but this association was not found to be statistically significant(p=0.748). Forced Expiratory volume in 1st second (FEV1) was found to be significantly lower in patients with bronchiectasis (p<0.05). There was increased frequency of exacerbations among patients with bronchiectasis. This association was however not found to be statistically significant (p=0.765), 1 (9.1%) patient had low BSI score (0-4), 3 (27.3%) patients had intermediate BSI score (5-8) and 7 (63.3%) patients had high BSI score (≥9).Conclusions:The presence of bronchiectasis in COPD is a phenotype associated with a poor clinical course. The characteristics of this co-existence are largely unknown. More studies are required to properly characterize and manage patients with this coexistence. 1. Interesting Phenomenon in an Islamic place called “Pondok Pesantren” which is located in Sananrejo village Turen district region of Malang attracts many attentions to a large of citizens. It is issued that the Pondok Pesantren which is well known with the term Pondok Pesantren Bihār Bahr ‘Asal Fadlāil al-Rahmah is a Mosque which is built by spirit where many people called them as “Jin”. In this case, this research is eager to investigate the motivation which become the background of establishing the Pondok Pesantren, how the proses of constructing this huge building with eleven floors and more than one hectare area of building; how the belief and characters which is taught in this Pondok Pesantren; and how the typology as well as ideology of Santri in this Pondok Pesantren. This research applies descriptive qualitative method by which the data collection is carried out using observation and interview. This also brings out phenomenology approach to reveal the ideology of the santris of Bihār Bahr ‘Asal Fadlāil al-Rahmah. Background:TNF has been shown to contribute to osteoclastogenesis independently and in conjunction with M-CSF and RANKL. We have previously demonstrated that TNF enhances the kinetics of RANKL-induced human osteoclastogenesis and that its effects are mitigated more effectively by the anti-TNF biologic adalimumab (ADA) as compared to etanercept (ETN).Objectives:To determine the mechanism responsible for the difference in effectiveness between the two biologics, a label-free quantitative proteomics study was conducted on TNF-activated human osteoclasts (OC) upon biologic treatment.Methods:Human bone marrow-derived OC precursors (OCP, 3 donors) were exposed for 5 days to M-CSF, M-CSF+RANKL (RANKL) alone or in combination with 100 ng/mL TNF +/- 5 ug/mL ADA, ETN or human IgG1 (IGG) as pre-formed complexes. OC differentiation was confirmed by measuring TRAP5b activity. Shotgun proteomics was performed on peptides generated from detergent based cell extracts subjected to methanol/chloroform precipitation and trypsin/Lys-C digestion. Data acquisition was performed with Orbitrap Q Exactive™ HF-X mass spectrometer. MaxQuant was utilized to quantify proteins based on MS1 peak intensities. Data matrix was normalized, imputed and subjected to differential expression analysis by limma. DAVID pathway analysis (DPA) was used to identify pathways impacted by various treatment conditions based on proteins exhibiting significant (p<0.05) 1.2-fold change in expression as compared to RANKL. Protein levels within culture supernatant or cell lysate were verified by ELISA or MSD.Results:Principle component analysis (PCA) of the proteomic profiling data for the 3 donors indicated that exposure of OCP to TNF induced a distinct profile from that of M-CSF and RANKL. The addition of ADA:TNF complexes restored the profile to that of RANKL, whereas those exposed to ETN:TNF complexes exhibited an intermediate profile matching differences observed in TRAP 5b activity levels. DPA identified 3 pathways most associated with osteoclastogenesis: receptor-mediated endocytosis (e.g. CD163, IGFBP2), oxidation-reduction process (e.g. FABP4), and cell adhesion (e.g. ICAM-1, TGFBI) that were significantly impacted by TNF with ADA:TNF being more effective than ETN:TNF in restoring most pathway-associated proteins to RANKL levels. Based on ELISA, two pro-osteoclastogenic factors IGFBP2 and ICAM-1 were increased 2-fold in OCP culture supernatants in response to TNF with only ADA:TNF complexes reducing these levels of both to RANKL. TNF-induced reduction of intracellular levels of CD163, an M2 macrophage polarization marker, and TGFBI were not only restored to RANKL levels by ADA:TNF but brought to a level closer to M-CSF alone, unlike ETN:TNF. One anti-osteoclastogenic factor FABP4 was found to be increased intracellularly above that of RANKL only following exposure to ADA:TNF complexes.Conclusion:Shotgun proteomic profiling of human OCP differentiatedin vitrowith TNF revealed at least 3 novel pathways by which TNF exerts its pro-osteoclastogenic effects. Only with the addition of ADA:TNF, not with ETN:TNF, were most pathway-associated proteins significantly restored to RANKL levels including pro-osteoclastogenic factors ICAM-1 and IGFBP2. Moreover, ADA:TNF complexes enhanced the intracellular levels of CD163 and the anti-osteoclastogenic factor FABP4, suggesting these complexes are exerting an effect on OCP beyond simple TNF neutralization. In pursuit of this spirit, the Union established in 2000 the IUPAC Prize for Young Chemists and has been honoring since then outstanding young research chemists at the beginning of their careers by making annual awards.<br /><br />The prizes are given for the most outstanding Ph.D. theses in the area of the chemical sciences, as described in 1000-word essays. Abstract Objective: Benign breast disease (BBD), especially benign proliferative breast disease (BPBD), is related to increased breast cancer risk. However, few studies have examined whether conventional breast cancer risk factors influence risk of breast cancer among women with BBD. The aim of this study was to evaluate the associations of lifestyle factors with risk of breast cancer among women biopsied for BBD within a multi-center, hospital-based, case-control study in China, in order to provide scientific basis of health guidance for BBD patients and lay the foundation for primary prevention of breast cancer. Methods: A multi-center, hospital-based, case-control study was conducted. Patients with BPBD (n=608) and patients with non-proliferative breast disease (NPBD) (n=366) were collected from 23 hospitals in 11 provinces during April 2012 to April 2013. A face-to-face survey, baseline data and fasting blood was collected with all study subjects. Serum adiponectin levels were assayed using ELISA. After 10 years, the cumulative incidence rate of breast cancer in the two groups was counted through follow-up. Logistic regression analysis was used to obtain the association between specific factors and risk of breast cancer. Results: After 10 years’ follow-up, 388 BPBD and 240 NPBD cases were included in the final analysis (Table 1), of which 16 (4.12%) and 3 (1.25%) developed breast cancer, respectively. The cumulative incidence of breast cancer between the two groups was significant difference (P=0.041). Compared with women in the NPBD group, BPBD group were more likely to be central obesity (with higher waist-to-hip ratio (WHR)) (OR 24.98, 95% CI 1.845-336.203, P=0.015) and less likely to have physical activity (OR 0.626, 95% CI 0.416-0.943, P=0.025) and less often to eat carrots (OR 0.616, 95% CI 0.398-0.953, P=0.030) (Table 2). Subgroup analyze indicated that, physical activity, eat carrots and ham sausage, body weight, BMI, waist circumference and WHR were statistical differences in premenopausal BPBD patients, while only physical activity (OR 0.423, 95% CI 0.269-0.665 P &lt; 0.001) was the independent risk factors. Meanwhile, among the factors of Tea consumption, Glycemia, Body weight, BMI, Waist circumference, WHR and HMW/total adiponectin ratio in postmenopausal group, only HMW/total adiponectin ratio (OR 0.041, 95% CI 0.002-0.820 P=0.037) was statistically significant factor. These stratified multivariate logistic regression analysis results are shown in Table 3. Conclusion: In patients with BBD, physical activity may be the protect factor for breast cancer carcinogenesis in premenopausal women while lower HMW/total adiponectin ratio is a risk factor for postmenopausal women, which can provide direction for primary prevention of breast cancer. We proposed a new idea for testing the thermal decay of light source under the effect of heat where the lux meter is used rather than using the integrating sphere and the thermal camera is used rather than thermocouple. The thermal decay behavior of the light is detected by using the lux meter. The thermal camera can be used to provide the temperature value as well as the temperature distribution of the measured surface which is more convenient than by using the thermocouple. Objectives: This research assesses the impacts of three distinctive crime control activities organized and directed by the neighborhood committees with the assistance of local police in contemporary urban China—Tiao-jie, Bang-jiao, and neighborhood watches. Tiao-jie deals with disputes and minor criminal cases. Bang-jiao provides guidance to residents who have committed minor offenses or have been released from correctional institutions to facilitate reintegration. Neighborhood watches engage local residents in crime prevention under the direction of neighborhood committees. Method: Using survey data recently collected in Tianjin, we examine the effects of indicators of the implementation of these neighborhood-based crime control strategies on residents’ reports of household property victimizations that occurred within the neighborhoods with multilevel logistic regressions. Results: Net of a range of individual-level and neighborhood-level control variables, the indicators of the level of activity of Tiao-jie, Bang-jiao, and neighborhood watches exhibit negative effects on reported household property victimization. Conclusions: Our findings provide suggestive evidence that the traditional strategies of neighborhood-level crime control continue to be relevant in the China of today and that the role of collective efficacy appears to differ from that observed in Western cities.

Abstract Oncogene BRAF is located on chromosome 7q34, consisting of 766 amino acids and 22 exons, encoding a serine/threonine protein kinase belonging to the RAF family and plays a critical role in the MAPK signaling pathway. The mutation of BRAF was documented in nearly 8% of all human cancers including melanoma (60%), thyroid (60%), and lung adenocarcinoma (10%). The most common mutation of BRAF is V600E (Class I), which was found in more than 70% in these cancers. Despite the clinical success of approved small molecule inhibitors of BRAF V600E (vemurafenib, dabrafenib and encorafenib), this remains an area of unmet medical need because of primary or acquired drug resistance. The construction of BRAF drug-resistant cell lines plays a pivotal role in cancer research. It serves as a crucial tool for discovery of the mechanisms of drug resistance. In this study, we have developed a systematic approach for constructing drug-resistant cell lines, inducing resistance through the gradual escalation of drug concentrations. The validation of drug resistance involves morphological recordings of parent and drug-resistant cell lines, drug resistance index in vitro through IC50 measurements and in vivo through the inhibition of tumor growth (TGI) which compare to those in the parental cell line. We successfully established Vemurafenib and Dabrafenib resistant A375 cell lines (A375 Vemurafenib R, A375 Dabrafenib R) with 21 and 31.5 folds of resistance index, respectively. The in vivo drug resistance efficacy was also tested by implanting Vemurafenib resistant cells subcutaneously into NOD-SCID mice and showed that A375 Vemurafenib R exhibited robust tolerance to 50 mg/kg Vemurafenib treatment (P.O.21 days, 50mpk, TGI=17.4%), whereas the parental cells were nearly completely inhibited (P.O.21 days, 50mpk, TGI=92.3%). In addition, single-cell sequencing of A375 Vemurafenib R revealed significant gene expression differences in the MAPK and EGFR signaling pathways, aiding in the identification of new targets, and signaling pathways associated with drug resistance, thereby providing potential targets for new drug development. Kabupaten Buton Utara merupakan salah satu daerah yang potensial hutan mangrovenya baik dari segi hasil hutan kayunya (HHK) maupun hasil hutan bukan kayu (HHBK) dan sangat dipengaruhi oleh kondisi ekologisnya, yang meliputi substrat, suhu, pH dan salinitas air. Lokasi penelitian ini di Desa Dampala Jaya dan Bumi Lapero, Kecamatan Kulisusu Barat, Kabupaten Buton Utara. Penelitian ini bertujuan untuk mengetahui :1) untuk mengetahui jenis vegetasi mangrove di lokasi pnelitian; 2) untuk mengetahui struktur vegetasi penyusun hutan mangrove di lokasi penelitian;3) untuk mengetahui Indeks Nilai Penting mangrove di lokasi penelitian; 4) untuk mengetahui Indeks keanekaragaman mangrove. Metode penelitian yang digunakan adalah menggunakan kombinasi desain jalur dan metode garis berpetak (Onrizal, 2008). Hasil penelitian ini menunjukkan bahwa di lokasi penelitian terdapat 5 (lima) jenis vegetasi mangrove, yaitu Bruguiera gymnorrhyza, Rhizophora stylosa, Xylocarpus granatum, Heriteria littoralis, Bruguiera parviflora. Indeks Nilai Penting di Desa Dampala Jaya yang tertinggi adalah spesies Bruguiera gymnorrhyza baik pada tingat semai (100,05), tiang atau pancang (93,93) dan pohon (95,75); sedangkan Indeks Nilai Penting terendah di tingkat semai Heriteria littoralis (12,71), tingkat tiang atau pancang dan tingkat pohon adalah spesies Bruguiera parviflora,masing-masing dengan nilai 17,71 dan 17,93. Indeks Nilai Penting tertinggi di Desa Bumi Lapero tingkat semai yaitu adalah Bruguiera gymnorrhyza yaitu 82,28, tingkat pancang atau tiang Rhizophora stylosa yaitu 114,07, tingkat pohon Bruguiera gymnorrhyza yaitu 106,04. Indeks Nilai Penting terendah adalah spesies Bruguiera parviflora baik pada tingkat semai (8.00), tingkat tiang atau pancang (12,66) dan tingkat pohon (22,78). Indeks keanekaragaman (H’) di Desa Dampala Jaya tertinggi pada tingkat semai yaitu spesies Bruguiera parviflora (0,21), sedangkan tingkat tiang atau pancang terdapat pada spesies Bruguiera gymnorrhyza dan Rhizophora stylosa masing-masing 0,37 begitu juga pada tingkat pohon terdapat pada spesies Bruguiera gymnorrhyza dan Rhizophora stylosa dengan nilai masing-masing 0,37. Indeks Nilai Penting vegetasi mangrove menggambarkan pentingnya peranan satu jenis spesies mangrove dalam ekosistem hutan mangrove. Jika Indeks Nilai Penting vegetasi mangrove tinggi nilainya, maka spesies mangrove itu sangat mempengaruhi kestabilan ekosistem hutan mangrove. Nilai Indeks Keanekaragaman di Desa Dampala yang tertinggi adalah Rhizophora stylosa pada tingkat semai yaitu 0,36, tingkat pancang atau tiang Bruguiera gymnorrhyza dan Rhizophora stylosa yaitu 0,37, tingkat pohon Bruguiera gymnorrhyza dan Rhizophora stylosa yaitu 0,37. Nilai indeks keanekaragaman di Desa Bumi Lapero yang tertinggi pada tingkat semai adalah Rhizophora stylosa yaitu 0,36, pada tingkat pancang atau tiang Bruguiera gymnorrhyza yaitu 0,37, pada tingkat pohon Rhizophora stylosa yaitu 0,37. Berdasarkan kriteria keanekaragaman Barbour et al. (1987) dalam Syamsuddin, N., (2019); H’= 1-2 (rendah), H’=2-3 (sedang), H’= 3-4 (tinggi), H’= >4 (sangat tinggi), sehingga Nilai Indeks Keanekaragaman di Desa Dampala Jaya dan Desa Bumi Lapero tergolong rendah.

One obstacle that can limit the growth and production of rice is low water availability. Therefore, a comprehensive study of the response of rice plants to drought is very important. The study was conducted in the greenhouse of the Faculty of Animal Husbandry and Agriculture, Diponegoro University, from April to August 2016. The study used a 3 x 3 factorial design with 3 replications. The first factor is three types of rice (Sidenuk, Way Apo Buru, Pepe) and the second factor is water stress treatment (KA <field capacity = enough water, field capacity (KL), water saturation (JA). The observed parameters are protein content, leaf levels rolling (number of leaves rolled), relative water content (RWC), water use efficiency (WUE), N, P, K uptake, rice protein content decreases with increasing water supply. The relative water content of rice plants decreases with increasing stress water that occurs in the limited water supply for the three types of rice Sidenuk rice has the lowest N seed content (2.1%) compared to Way Apo (3.0%) and Pepe (3.1%). The mineral content of P in both biomass and seeds is not really influenced by water stress, as well as K content of seeds. Kelembapan, jamur, dan kumbang merupakan faktor yang dapat menghambat keberhasilan produksi tanaman pisang mas Kirana. Guna mempermudah petani dan pemilik perkebunan yang ingin memulai pembudidayaan tanaman pisang mas Kirana, dibutuhkan suatu sistem yang dapat bertugas membantu mendiagnosa penyakit yang terdapat pada tanaman pisang mas Kirana tersebut, serta memberikan solusi agar dapat menghindari berbagai penyakit yang dapat menghambat pertumbuhan serta produksi dari tanaman pisang mas Kirana. Basis pengetahuan yang digunakan dalam sistem pakar ini yaitu pendekatan algoritma fuzzy logic Tsukamoto. Metode ini dapat bekerja dengan baik karena menggunakan aplikasi nilai monoton. Masukannya berupa gejala-gejala yang dialami kemudian akan diproses dengan kaidah (IF–THEN). Keluaran hasil dari tiap-tiap aturan diberikan secara tegas. Sistem pakar dirancang untuk berinteraksi langsung dengan pemakai dengan format dialog tanya jawab. Metode pengembangan sistem menggunakan waterfall dan pemodelan sistem menggunakan UML. Sedangkan alur penelitian digambarkan dengan diagram alir dan bahasa pemrograman menggunakan PHP.

Taman Wisata Alam (TWA) Jering Menduyung merupakan salah satu hutan konservasi Bangka Belitung yang terletak di Kabupaten Bangka Barat. Pada kawasan ini mempunyai hutan mangrove primer yang mendominasi seluas 1209,7 Ha dari seluruh tutupan lahan. Selain itu, juga terdapat ekosistem hutan rawa 405,519 Ha dan hutan belukar rawa 478,709 Ha. Hutan rawa air tawar pada beberapa tahun terakhir keadaan vegetasi mengalami banyak gangguan seperti adanya aktivitas pertanian. Penelitian ini dilakukan untuk menganalisis tingkat keanekaragaman vegetasi hutan rawa air tawar di TWA Jering Menduyung. Metode analisis vegetasi menggunakan metode kombinasi antara metode jalur dan metode garis berpetak, sedangkan penempatan jalur menggunakan metode purposive sampling. Hasil penelitian ini menunjukkan bahwa hutan rawa air tawar memiliki 25 jenis tumbuhan yang termasuk kedalam 19 famili. Famili Rubiaceae merupakan jenis yang paling banyak, diantaranya yaitu jenis Ixora paludosa (Blume) Kurz, Psychotria sp., Oxyceros longiflorus (Lam.) T.Yamaz, Psychotria sp.. Melaleuca leucadendron L merupakan spesies yang mendominasi pada tingkat pertumbuhan pancang (115,64%), tiang (300%) dan pohon (300%). Sementara tingkat semai, spesies yang mendominasi adalah Cyperus sp. (58,62%). Indeks keanekaragaman Shannon-Weiner yang ditemukan tertinggi berkisar anatara 1,591-2,197 yang ditemukan pada tingkat semai dan pancang. Nilai indeks keanekaragaman Shannon-Weiner pada tingkat tiang dan pohon <1. Distribusi spesies berdasarkan indeks kemerataan Evennes untuk semua tingkat pertumbuhan (semai, pancang, tiang dan pohon) di hutan rawa air tawar menunjukkan distribusi spesies yang tidak sama (0-0,792). Stategi yang tepat dalam pengelolaan hutan rawa air tawar TWA Jerieng Menduyung ini melalui pendekatan partisipatif pada tingkat lanskap untuk perencanaan pengelolaan ekosistem hutan yang efektif dan berkelajutan.

The nanostructures and mechanical properties of nanocomposite nc-TiN/a-(TiB2, BN) and nanostructured multilayers nc-TiN/a-TiBN were investigated using X-ray diffraction (XRD), high-resolution transmission electron microscopy (HRTEM), microindentation, and stress measurements. It was found that the monolayer Ti-B-N thin films consisted of nanometer-sized crystalline (nc-) Ti(N, B) embedded into amorphous (a-) (TiB2, BN) matrix. When B content was below ∼16 at.%, two different-sized nanocrystallites with mean grain sizes of ∼3 and 9 nm respectively were embedded in a-TiB2. With increasing B incorporation (>∼27 at.%), more uniform nanograins was embedded a-(TiB2, BN). Incorporation of B not only decreased the size of nanocrystallites, but also gave rise to twinning deformation in nanocrystallites. A maximum hardness of ∼44 GPa was achieved at B content of 19 at.%. It was also found that the nanostructure and mechanical behaviors of nc-TiN/a-TiBN multilayers was dependent on the modulation length (bilayer thickness Λ). Decrease of Λ made the preferred orientation of nc-TiN gradually transform from (200) to (111). A maximum hardness of ∼30 GPa was achieved at Λ = 1.8 nm. Deflection from this Λ value decreased hardness. By contrast, the residual compressive stress value monotonically increased with decrease of Λ. The enhancement of the hardness was due to the coherent stresses and the structural barriers to dislocation motion in the interface.

In the last decade, considerable research effort was directed to the deposition of multilayer films with layer thicknesses in the nanometer range (superlattice coatings), in order to increase the performance of various cutting tools and machine parts. The goal of the present work was to investigate the main microstructural, mechanical and wear resistance characteristics of a superlattice coating, consisting of alternate multilayer ZrN/TiAlN films, with various bilayer periods (5÷20 nm). The coatings were deposited by the cathodic arc method on Si, plain carbon steel and high speed steel substrates to be used as wear resistance surfaces. The multilayer structures were prepared by using shutters placed in front of each cathode (Zr and Ti + Al). The characteristics of multilayer structures (elemental and phase composition, texture, Vickers microhardness, thickness, adhesion, and wear resistance) were determined by using various techniques (AES, XPS, XRD, microhardness measurements, scratch, and tribological tests). A comparison with the properties of ZrN and TiAlN single-layer coatings was carried out.

Fiber PVA yang memuat partikel nano TiO2 anatase telah berhasil dibuat dengan metode elektrospining. Fiber tersebut dibuat dari larutan yang merupakan campuran serbuk nano TiO2 anatase dalam 10% larutan PVA dengan rasio TiO2/PVA terbesar mencapai 60%. Proses elektrospining dilakukan pada tegangan 15 kV dengan jarak ujung anoda ke kolektor (TCD) sejauh 15 cm. Hasil analisis SEM diameter fiber dari tiap-tiap variasi rasio massa menunjukan perubahan rasio TiO2/PVA berpengaruh terhadap diameter fiber. Perlakuan pemanasan pada suhu 300pada fiber tidak mengubah fase kristalin TiO2 dan juga tidak memperlihatkan pengaruh yang signifikan terhadap diameter fiber. Namun demikian perlakuan panas tersebut dapat mereduksi ukuran partikel TiO2. Fiber yang dihasilkan mempunyai rentang diameter dari 100 -700 nm dan partikel TiO2 yang termuat pada fiber PVA memiliki rentang ukuran partikel 50 -180 nm. Hasil analisis serapan sinar UV menunjukan terjadinya serapan sinar UV pada daerah UVB, dengan demikian fiber hasil elektrospining potensial untuk digunakan sebagai fotokatalis.Kata kunci : Fiber, Polivinil Alkohol, Titanium Oksida, Elektrospining

Penelitian pada Kawasan Hutan Dengan Tujuan Khusus (KHDTK) (Hutan Pendidikan) ± 225,7 ha di Desa Senaru, Kabupaten Lombok Utara bertujuan untuk mengetahui potensi vegetasi dan cadangan karbon tersimpan. Tutupan alami KHDTK Senaru berubah akibat pemanfaatan kayu 1993, penanaman sengon dan mahoni 1996, penanaman gaharu 1998-2001, dan saat ini pemanfaatan dengan sistem agroforestri. Populasi dalam penelitian ini adalah areal KHDTK Senaru yang dikelola intensif oleh masyarakat (± 120 ha), yang ditentukan secara sengaja. Sampel dengan intensitas 1% dan terbagi dalam 30 plot ukuran 20x20 m ditentukan secara random sampling. Analisis data meliputi analisis vegetasi, cadangan karbon atas permukaan tanah dan karbon tanah. Hasil penelitian menunjukkan bahwa terdapat 32 spesies. Pada tingkat semai, pancang, tiang dan pohon ditemukan masing-masing 10, 8, 17 dan 20 spesies. Dua spesies dengan Index Nilai Penting (INP) tertinggi untuk semai dan pancang adalah kopi dan kakao, dengan INP pada tingkat semai sebesar 120,3 dan 34,2 dan pada tingkat pancang sebesar 146,1 dan 92,5. Dadap dan sengon memiliki INP tertinggi pada tingkat tiang dan pohon, dengan INP pada tingkat tiang sebesar 77,9 dan 48,7, dan pada tingkat pohon sebesar 87,1 dan 79,9. Cadangan karbon rata-rata 126,41 ton/ha, yang terdiri atas karbon tingkat pancang (3,36 ton C/ha), tiang (9,32 ton C/ha), pohon (70,61 ton C/ha), tumbuhan bawah tegakan (0,13 ton C/ha), seresah (0,29 ton C/ha) dan tanah (42,7 ton C/ha). Hasil penelitian dapat menjadi masukan dalam pengembangan model pengelolaan KHDTK Senaru dan bahan evaluasi di masa mendatang, dan secara umum dapat menambah informasi sumberdaya hutan yang sudah ada saat ini.Katakunci: hutan, vegetasi, cadangan karbon, Senaru Lombok A Study on the Vegetation and Carbon Stocks of the Special Purpose Forest (KHDTK) of Senaru Village, Northern LombokAbstractStudy was conducted in Forest for Special Purpose (Education Forest) ±225.7 ha in Senaru Village, North Lombok. The study was aimed to determine the potential of vegetation and carbon stocks. Land cover of study area was changed due to the timber management in 1993, planting mahogany and sengon in 1996, planting Aquilaria spin 1998-2001, and currently the implementation of agroforestry. Population for this study was the area of education forest intensively utilized by farmer (±120 ha). Sampling was determined by means of random with the intensity of 1% and distributed into 30 plots of 20x20m. Data analysis included analysis of vegetation, above ground carbon and soil carbon stock. The result shows that 32 species were found. There were 10, 8, 17 and 20 species of vegetation for seedlings, saplings, poles and trees, respectively. The first two highest Important Value Index (IVI) for seedling and saplings were Coffea sp and Theobroma sp, where the IVI for seedling was 120.3 and 34.2, while for saplings were 146.1 and 92.5. Erytrhina sp and Pharaseriantes sp were the two highest IVI for poles and trees, where the IVI for poles was 77.9 and 48.7, while for trees was 87.1 and 79.9, respectively. Carbon stock of study area was 126.41 ton C/ha, which was differentiated into carbon stock for saplings (3.36 ton C/ha), pole (9.32 ton C/ha), trees (70.61 ton C/ha), understory (0.13 ton C/ha), litter (0.29 ton C/ha) and soil (42.7 ton C/ha). The results of this study could be an input in develoving a model of Senaru educational forest as wells as future evaluation. Besides, it could enrich the existing information about forest resources.

Penelitian ini bertujuan untuk mengetahui portofolio efisien yang memberikan return tertinggi dengan risiko tertentu atau return tertentu dengan risiko terendah dan besarnya proporsi dana dari masing-masing saham dalam pembentukan portofolio optimal. Dari sekumpulan kandidat portofolio efisien ini terdapat portofolio optimal. Saham-saham yang diteliti adalah saham-saham yang termasuk dalam Jakarta Islamic Index periode Desember 2016-Mei 2017 dan Juni 2015-Mei 2017. Jumlah saham tiap-tiap periode penelitian ada 30 saham. Dari 30 saham masing-masing periode penelitian terdapat saham pembentuk portofolio optimal periode pertama: UNTR (26%), ASII (20%), TLKM (18%), ADHI (14%), INTP (9%), INDF (8%), UNVR (4%), LPPF (1%), SSMS (1%) dan periode kedua: SMGR (63,96%), LPPF (35,56%), SILO (0,48%), LSIP (0,01%). Apabila dibentuk portofolio optimal sesuai dengan proporsi perhitungan, maka expected return yang dihasilkan masing-masing periode penelitian, yaitu: periode pertama sebesar 0,16% per enam bulan lebih besar dibanding dengan expected return market (IHSG) sebesar 0,09% per enam bulan dan periode kedua sebesar 353,73% per dua tahun lebih besar dibanding dengan expected return market (IHSG) sebesar 0,02% per dua tahun.

Neurotransmitters function in one of two modes, promoting either inhibition or excitation. However, the metabotropic γ-aminobutyric acid receptor (GABABR) system can switch between these modes. In the presence of a small excitatory stimulus, the GABABR mediates a shunting inhibition that suppresses excitation. However, in the presence of a strong excitatory stimulus, the GABABR potentiates the response. This bipartite action is accomplished by linking the GABABR to two electrogenic mechanisms; one activates an outward current and another reduces an outward current. As a consequence, the GABABR serves as a discriminator that reduces the influence of weak signals while augmenting responses to strong signals. In retinal ganglion cells, this mechanism acts to promote the communication of phasic information.

Abstract Background: Glycosylation profoundly influences the interactions between cancer cells and microenvironmental stromal cells during the peritoneal disseminated metastasis of ovarian carcinoma (OC), which is the major cause of cancer-related death. Although the characteristic cancer glycoconjugates are widely used as biomarkers for cancer diagnosis, our knowledge about cancer glycome remains quite fragmented due to the technique limitations in analyzing glycan chains with tremendous structural and functional heterogeneity. Method: Given the dysregulated cancer glycome is defined by the altered glycosylation machinery, here we performed a systematic loss-of-function screen on 498 genes involved in glycosylation for key regulators of OC dissemination. We identified neuraminidase 4 (NEU4), an enzyme capable of hydrolyzing terminal sialic acid from glycoconjugates, as a vital peritoneal dissemination-promoting modifier of OC glycome. What’s more, we also identified membrane proteins whose sialylation was able to be modified by NEU4 in OC cells by pull down and mass spectrum to investigate the functional significance of NEU4-mediated desialylation. Results: In human patients with high-grade serous OC (HGSOC), increased NEU4 was detected in the disseminated OC cells when compared with that in the primary tumor cells, which significantly correlated with the worse survival. Among three alternative splice-generated isoforms of human NEU4, we revealed that only the plasma membrane-localized NEU4 isoform 2 (NEU4-iso2) and intracellular isoform 3 promoted the peritoneal dissemination of OC by enhancing the cell motility and epithelial-mesenchymal transition. We also identified NEU4-iso2-regulated cell surface glycoproteome and found that NEU4-iso2 desialylated the epithelial growth factor receptor (EGFR), in particular at N196 residue, for the hyperactivation of EGFR and its downstream tumor-promoting signaling cascades. Conclusion: Our results provide new insights into how the OC glycome is dysregulated by NEU4 during OC progression and reveals a functionally important glycosite on EGFR for its abnormal activation in cancer. Aims: Fokienia hodginsii is a threatened conifer tree species, known as the dominant nursery-grown species capable of colonizing the challenging woodland environments in southern China due to its strong root penetrating ability. The ecological phenotype of Fokienia hodginsii is not well documented during its breeding process, which limits the potential planting area and its ecological function. This study aims to understand how Fokienia hodginsii associates with microbes to conduct its key ecological function and provide a theoretical basis for further improving the forest nursery management of Fokienia hodginsii. Methods: This study explored the ecological traits of 11 main Fokienia hodginsii provenances in a homogeneous garden experiment by analyzing their nutrient utilization strategies and associated microbial features in the rhizosphere soil and roots. Results: The study found that the paramount difference in the rhizosphere soil among provenances is in Ca and Fe content. Some microbial communities, namely Crenarchaeota, Verrucomicrobiota, and Desulfobacterota, were positively correlated with the amounts of the soil nutrient elements, whereas Abditibacteriota and Dependentiae were negatively correlated. The abundance of N- and Fe-related bacteria in the Fu Jian Chang Ting (FJCT) provenance was significantly higher than that in other provenances, while the C-, P-, K-, and Mg-related fungal communities, respectively, had higher abundances in the FJCT, Fu Jian Long Yan (FJLY), Fu Jian Gu Tian (FJGT), and Fu Jian Xian You (FJXY) provenances than the others. The impacts of the Gui Zhou Li Ping (GZLP), Hu Nan Dao Xian (HNDX), Jiang Xi Shang Yao (JXSY), and Guang Dong Shi Xing (GDSX) provenances on the rhizosphere soil are similar, but the differences in nutrient utilization arise from the plant itself. Conversely, the root nutrient contents of the FJCT, Fu Jian You Xi (FJYX), Fu Jian An Xi (FJAX), FJLY, Fu Jian De Hua (FJDH), FJGT, and FJXY provenances are highly correlated with soil nutrient features. Conclusions: For the native provenances, their economic traits are better than the exotic provenances. The native provenances are more sensitive to local soil conditions, so they should benefit more from human interventions, rendering them more suitable for artificial cultivation. The growth of the exotic provenances is less affected by the soil environment, making them better suited for the ecological transformation of forest stands and soil improvement.

Perkembangan wilayah pesisir Teluk Kendari cukup pesat dengan berbagai macam aktivitas baik berupa jasa kelautan seperti pelabuhan untuk pelayaran dan perikanan maupun kegiatan-kegiatan di sekitar pantai seperti pemukiman, industri, usaha dan pertambakan. Adanya logam berat di perairan, berbahaya baik secara langsung terhadap kehidupan kerang darah (Anadara. granosa) dan kerang bakau (Polymesoda bengalensis), maupun efeknya secara tidak langsung terhadap kesehatan manusia. Penelitian ini bertujuan untuk menganalisis kandungan logam berat Pb dan Zn pada air, sedimen, dan jaringan kerang dan mengkaji faktor bioakumulasi logam berat Pb dan Zn pada dua jenis kerang. Sampel air, sedimen dan kerang diambil pada kedua lokasi pengambilan kerang, tiap lokasi 3 titik pengambilan dengan 3 kali ulangan analisis. Hasil Penelitian menunjukkan bahwa logam Pb dan Zn di air pada tiap lokasi telah melampau baku mutu, kadar tertinggi masing-masing 0,018 mg/L dan 0,793 mg/L, sedangkan pada sedimen tertinggi masing-masing 0,823 mg/Kg dan 6,919 mg/Kg, dan pada jaringan kerang menunjukan hasil bahwa kerang ukuran besar mengandung logam Pb dan Zn lebih tinggi, masing-masing 1,750 dan 9,863 mg/Kg. (sedimen dan kerang belum melampau baku mutu). Nilai BCFo-w logam Pb tertinggi 119,20 pada kerang bakau dan Zn tertinggi 35,99 pada kerang darah. Berdasarkan kategori nilai IFK (BCF) untuk logam Pb dan Zn termasuk dalam kategori rendah hingga sedang.
