Resumen
With his thought-provoking and socially relevant music, American composer Jake Runestad has quickly become one of the most performed choral composers of the 21st century. Although music and social justice have been tied together for centuries, there is a new movement bringing social justice to American choral music in a noticeably increased manor, and Jake Runestad is a leading composer in this movement. In this paper, I provide a detailed analysis into the social justice themes employed by Runestad, interviews with him and several well-respected American choral directors programming and commissioning his music, as well as compositional devices employed within his compositions. The purpose of this study is to show Jake Runestad's place as an American choral composer by offering a historical overview of the social justice themes in American music and Western choral music separately. I will then narrow the scope to Jake Runestad, who since 2013 has been using his choral music to bring awareness to human inequalities within the United States today.

Resumen
My paintings are a mediated autobiography focusing on fragmented, cropped or collaged elements derived from photographic sources. Banal scenes based on my experience growing up in rural Washington state are charged with multivalent interpretations via close ups, veiling and intentional obfuscation. Through these means, altered color and expressionistic paint handling, they subtly point outside of their beginnings as the photographic and personal and into a dialogue with historical and social sites such as punk music, abstract expressionism and existential literature.

This dissertation explores the development of commercial entertainments and film exhibition in the urban South around the turn of the last century through the growth and decline of Jake Wells Enterprises. A former professional baseball player, Wells invested in a wide variety of public amusements, with the core of his early business centered on establishing and organizing a string of vaudeville, popularly priced, and legitimate theaters throughout the largest cities in the region, a network he later transitioned to showing exclusively motion pictures. A thorough analysis of period newspapers, trade journals, and some business records covering Wells’ career provides much-needed evidence for film and cultural historians wishing to understand the genesis and evolution of public amusements in the region, and its negotiation of traditional social and cultural institutions. In the 1890s, Wells played and managed several professional baseball teams in the South. The sport educated players and spectators alike to both the values and creed of New South progress, and to rising tensions confronting the intersection of modern and traditional forms of culture. Using his experiences and contacts gained in baseball, Wells helped foster a culture of entrepreneurship and innovation required for the progress of media industries in the region, establishing social networks of knowledge and improving distribution flows of entertainment. The dissertation explores how race and the genteel emerged as regional characteristics most influential to the success of this conversion in many urban areas. Protestants and evangelical culture served as the bulkhead supporting opposition to new amusements. Wells’ expansion plans and violations of Sabbath day laws evoked a “spatial” battle between commercialism and religion where political, social, and cultural power drawn from place and identity were challenged and reconfigured. Another chapter explores the exhibition and reception of early Civil War films in the region. Wells and other exhibitors were influential in their production and circulation nationwide, and positioned cinema as an alternative shrine to commemorate the Lost Cause in many communities. The last chapter shows how Wells failed to meet local demands and consumer desires in competition with the rise of national chain theaters and Hollywood’s vertical integration.

Each of the artists engages, in various ways, with issues relating to biotechnology and the body. The convergence of technology and the body arouses both utopian desires and dystopian nightmares; many of these desires and fears are prefigured in ancient myth and legend. The artists and artworks are therefore discussed with reference to reoccurring tropes from both ancient myth and science fiction.

American composer Jake Heggie wrote Camille Claudel: Into the Fire in 2012 for the Alexander String Quartet and mezzo-soprano Joyce DiDonato. As Heggie is known for his operas Dead Man Walking, Moby-Dick, and most recently, Great Scott, it is not surprising that his seven-song cycle about this passionate female French sculptor is of operatic dramaturgical and musical quality. Due to the complexities and bias that surround Claudel’s life story, and the relative novelty of Heggie’s music within the art song genre, there is a lack of literature regarding the presentation of an authentic performance of this cycle. This project report concentrates on providing the singer with an interpretive framework based on how Heggie’s musical influences and tendencies, along with Gene Scheer’s historical, first person narrative, reflect Claudel’s life, work, and fiery personality. Through a deeper understanding of Heggie’s music in correspondence with Scheer’s poetry, a singer can effectively embody the unparalleled passion, artistry, and voice of Camille Claudel.

The human mitochondrial genome is a 16,569 base pair double-stranded deoxyribonucleic acid (DNA) molecule located in the mitochondrion. The mitochondria1 genome sequence is conserved, maternally inherited and undergoes no recombination, making its evaluation ideal for evolutionary studies. Certain alterations in the genome are unique to specific human populations and haplogroups. The genetic background, or haplogroup of an individual, may act in concert with disease associated mutations. The ethnicity of an individual is often utilised as an indicator of haplogroup. During this investigation the full mitochondrial sequence of 10 individuals belonging to three ethnic Southern African populations, namely three Xhosa, three Zulu and four Tswana individuals, was generated. The complete nucleotide sequences were compared to one another in order to determine the genetic relationship between individuals. Sequences were also evaluated against the 2001 revised Cambridge reference sequence (RCRS) to detect novel alterations as well as alterations present in all individuals analysed. A total of 222 alterations (207 previously reported, 15 unreported) were detected relative to the RCRS. Five length alterations and 207 nucleotide substitutions were detected. Ninety-eight alterations were detected only once, and 115 were detected in at least two individuals. Haplogroup analysis clustered individuals into haplogroups LO, L2 and L3. No clear correlation between haplogroup assignment and ethnic origin could be observed. The distribution of shared alterations between individuals was in agreement with the haplogroup clustering. Ethnicity can therefore not be utilised as an indicator of haplogroup in the context of the current study. To investigate the association between disease presentation and haplogroup, individuals will have to be randomly sampled and then haplogrouped. Reasons to substantiate the lack of association between ethnicity and haplogroups include the occurrence of gene flow between populations, inaccurate ethnic and incomplete haplogroup classification and populations not being sufficiently divergent.

American composer Jake Heggie's For a Look or a Touch exists as the only classical music work of remembrance based on the topic of remembering homosexual victims of the Holocaust. The composer, after being approached by Mina Miller, wrote this 2007 composition as part of the Music of Remembrance concert series. The music of this work varies in style from a blazing swing dance to a haunting vocalise on "oo." Gene Scheer created the text, delivered in the work by the characters Manfred Lewin and Gad Beck, as a compilation of many influential stories from the documentary film Paragraph 175. For a Look or a Touch chronicles the horrendous treatment of homosexuals before, during, and after World War II. Chapters include an introduction to homosexual persecution during Hitler's reign, a look at current works of remembrance and how For a Look or a Touch came to fruition, an explanation of the texts created by Gene Scheer, a discussion on Jake Heggie's musical setting of this text, and avenues for possible future research. Appendices include text of the German anti-sodomy laws as written in Paragraph 175, the mission statement for the organization Music of Remembrance, transcripts from personal interviews with both Jake Heggie and Gene Scheer, correspondence with Heggie, and the source delineation of Gene Scheer's text.

The aim of this project was to explore the role of the tyrosine kinases JAK1 and JAK3 in cytokine signalling, focusing on interleukin-2 signalling in CD8+ effector T lymphocytes. Initial experiments compared the effects of the pan JAK1/JAK3 inhibitor tofacitinib, the selective JAK1 inhibitor GSK186, and the selective JAK3 inhibitor GSK192 on IL-2 control of effector CD8+ cytotoxic T cells (CTL). On the basis of these preliminary data, a detailed analysis of the effect of tofacitinib on effector CD8+ T lymphocytes was performed. Phosphorylation events regulated by tofacitinib were identified using mass spectrometry analysis of SILAC (stable isotope labelling with amino acids in cell culture) labelled CTL. Tofacitinib regulated a selective number of phosphorylation sites, with less than 1.2% of the CTL phosphoproteome significantly regulated by tofacitinib treatment following 4hrs tofacitinib treatment. Proteins with downregulated phosphorylation sites were enriched in functions related to the Jak-STAT signalling, regulation of gene expression, and MAPK signalling cascades. Proteins with upregulated phosphorylations were also enriched in functions related to regulation of gene transcription. The proteome of tofacitinib treated CTL was defined by label free mass spectrometry. Approximately 4.5% of the CTL proteome was significantly regulated following 24 hours tofacitinib treatment, suggesting tofacitinib regulates the expression of a selective subset of proteins. Tofacitinib treatment resulted in the downregulation of proteins involved in ribosome biosynthesis, steroid biosynthesis, regulation of transcription and the cell cycle; and the upregulation of proteins with hydrolase activity, and with roles in the lysosome and extracellular exosomes. The phosphoproteomic and proteomic data demonstrates that JAK kinase dependent IL-2 signalling regulates essential processes in CTL by controlling a selective number of phosphorylation events and proteins. Validation of proteins identified as regulated following tofacitinib treatment identified new targets of IL-2 signalling in CTL, including the transcription factor NFIL3. NFIL3 was shown to be upregulated in CD8+ T lymphocytes following stimulation with IL-2 and regulated perforin and CD62L expression, suggesting a role in the regulation of CTL effector function.

We propose a straightforward algorithm to estimate large Bayesian time-varying parameter vector autoregressions with mixture innovation components for each coefficient in the system. The computational burden becomes manageable by approximating the mixture indicators driving the time-variation in the coefficients with a latent threshold process that depends on the absolute size of the shocks. Two applications illustrate the merits of our approach. First, we forecast the US term structure of interest rates and demonstrate forecast gains relative to benchmark models. Second, we apply our approach to US macroeconomic data and find significant evidence for time-varying effects of a monetary policy tightening.

In this paper we put forward a Bayesian Model Averaging method aimed at performing inference under model uncertainty in the presence of potential spatial autocorrelation. The method uses spatial filtering in order to account for uncertainty in spatial linkages. Our procedure is applied to a dataset of income per capita growth and 50 potential determinants for 255 NUTS-2 European regions. We show that ignoring uncertainty in the type of spatial weight matrix can have an important effect on the estimates of the parameters attached to the model covariates. After integrating out the uncertainty implied by the choice of regressors and spatial links, human capital investments and transitional dynamics related to income convergence appear as the most robust determinants of growth at the regional level in Europe. Our results imply that a quantitatively important part of the income convergence process in Europe is influenced by spatially correlated growth spillovers.

This paper develops a Bayesian variant of global vector autoregressive (B-GVAR) models to forecast an international set of macroeconomic and financial variables. We propose a set of hierarchical priors and compare the predictive performance of B-GVAR models in terms of point and density forecasts for one-quarter-ahead and four-quarter-ahead forecast horizons. We find that forecasts can be improved by employing a global framework and hierarchical priors which induce country-specific degrees of shrinkage on the coefficients of the GVAR model. Forecasts from various B-GVAR specifications tend to outperform forecasts from a naive univariate model, a global model without shrinkage on the parameters and country-specific vector autoregressions

The Mevalonate Pathway: A Potential Therapeutic Target for JAK2-driven Myeloproliferative Neoplasms Lori Nicole Griner Abstract Myeloproliferative neoplasms (MPNs) are diseases of hematopoietic stem cell origin and are characterized by uncontrolled growth of cells of the myeloid compartment. The Philadelphia chromosome negative classical MPNs, including polycythemia vera, essential thrombocythemia, and myelofibrosis, are diseases of dysregulated JAK2 signaling. In fact, the majority of MPN patients have activating mutations in JAK2 (e.g JAK2-V617F), a tyrosine kinase that contributes to the growth and survival of myeloid cells. While MPNs were first described over sixty years ago, a significant need remains to develop therapeutic strategies for them. Inhibitors of JAK2 are currently being developed, and one inhibitor, ruxolitinib, was recently approved for certain MPN patients. Ruxolitinib has made profound impacts on improving splenomegaly and constitutional symptoms in MPN patients, but it and other JAK2 inhibitors have not significantly reduced the JAK2 mutant allele burden, and thus such inhibitors have not induced remission in these patients. The current consensus in the MPN field supports JAK inhibition for the treatment of patients, but a further understanding of MPNs and JAK2 signaling, as well as improved JAK2 inhibitors, may be necessary for treating MPN patients. The work described in this dissertation has uncovered novel requirements for JAK2-V617F-driven signaling and transformation. We demonstrate that JAK2-V617F co-localizes with lipid rafts, cholesterol-rich microdomains within the plasma membrane that function to serve as platforms for signaling complex formation. Signaling complex formation is a necessary component for dysregulated signaling induced by JAK2-V617F. We provide evidence that cholesterol altering-lipid raft disrupting agents attenuate JAK2-V617F-driven signaling. We also show that cholesterol-lowering statins are effective at downregulating JAK2 signaling and inducing apoptosis in JAK2-V617F-driven cell lines. Importantly, we show that statins, inhibitors of the mevalonate pathway, inhibit the growth of primary MPN cells, while the same statin doses have no effect on healthy controls. Impressively, we demonstrate that statins cooperate with multiple JAK inhibitors, including ruxolitinib, to inhibit cell growth and induce apoptosis of JAK2-V617F-driven cells. This report establishes statin-mediated inhibition of the mevalonate pathway as a potential approach to improve MPN therapeutics. We propose future studies with statins and JAK2 inhibitors in the treatment of MPNs.

In opera, the opera chorus actively shapes the dramatic structure through interactions with the soloists, commentary after events, and momentum provided toward scene endings. Since the chorus traditionally represents the voice of the people, it also provides a natural access point by which audiences may connect to the unfolding drama. To realize its dramatic potential, an opera chorus must have a resonant, vibrant sound that is more “soloistic” than other genres of choral music. Indeed, there are quantifiable acoustic differences between classical solo and choral singing. The characterizations of the chorus must also be convincing. Yet there is only minimal research, to date, describing a systematic approach to rehearsing the opera chorus and applying those rehearsal techniques to specific musical examples. In this disquisition, I summarize existing research regarding choral rehearsal strategies and the role of the chorus master. I then synthesize and apply this research in the form of a chorus master’s analysis of choral excerpts from three contemporary operas recently produced by The Minnesota Opera: Norwegian composer Poul Ruders’ (b. 1949) The Handmaid’s Tale (1998), Jake Heggie’s (b. 1961) Dead Man Walking (2000), and American composer Kevin Puts’ (b. 1972) Silent Night (2011). I argue that a chorus master’s rehearsal strategies for these works must invite efficient, classical vocalism and a dramatic, textually informed interpretation of the elements of melody, harmony, form, rhythm, texture, and timbre. In opera, the opera chorus actively shapes the dramatic structure through interactions with the soloists, commentary after events, and momentum provided toward scene endings. Since the chorus traditionally represents the voice of the people, it also provides a natural access point by which audiences may connect to the unfolding drama. To realize its dramatic potential, an opera chorus must have a resonant, vibrant sound that is more ?soloistic? than other genres of choral music. Indeed, there are quantifiable acoustic differences between classical solo and choral singing. The characterizations of the chorus must also be convincing. Yet there is only minimal research, to date, describing a systematic approach to rehearsing the opera chorus and applying those rehearsal techniques to specific musical examples. In this disquisition, I summarize existing research regarding choral rehearsal strategies and the role of the chorus master. I then synthesize and apply this research in the form of a chorus master?s analysis of choral excerpts from three contemporary operas recently produced by The Minnesota Opera: Norwegian composer Poul Ruders? (b. 1949) The Handmaid?s Tale (1998), Jake Heggie?s (b. 1961) Dead Man Walking (2000), and American composer Kevin Puts? (b. 1972) Silent Night (2011). I argue that a chorus master?s rehearsal strategies for these works must invite efficient, classical vocalism and a dramatic, textually informed interpretation of the elements of melody, harmony, form, rhythm, texture, and timbre. Les adénomes hépatocellulaires (AHC) sont des tumeurs bénignes rares qui se développent le plus souvent chez la femme jeune suite à la prise de contraceptifs oraux. Les complications principales sont l’hémorragie et plus rarement, la transformation maligne en carcinome hépatocellulaire (CHC). Des travaux récents ont permis d’identifier 3 groupes moléculaires principales d’AHC qui se définissent par (1) l’inactivation du facteur de transcription HNF1A (H-HCA), (2) l'activation de la voie Wnt/ß-caténine (bHCA) ou (3) la présence d’infiltrats inflammatoires (IHCA).Afin d’identifier les voies de tumorigenèse associées au développement d’AHC inflammatoires (IHCA), une analyse transcriptomique comparant des IHCA à des foies non tumoraux a été réalisée au laboratoire, ce qui a permis d’identifier dans ce groupe tumoral une activation de la voie IL-6/JAK/STAT3. Nous avons recherché de nouvelles altérations géniques et nous avons caractérisé le mécanisme d'activation de la voie IL-6/JAK/STAT dans les IHCA. Les conséquences fonctionnelles sur la voie STAT3 des différents mutants ont été analysées par une modélisation de leur expression dans des lignées hépatocellulaires. Par ailleurs, nous avons réalisé des études génomiques intégrées (analyse CGH-SNP, méthylome et séquençage exome) sur une large série de 250 AHC avec pour objectif d’affiner la classification moléculaire des AHC, d’identifier de nouveaux gènes altérés dans ces tumeurs et d’élucider les mécanismes de transformation maligne des AHC en CHC.Dans le groupe des IHCA, ces analyses nous ont permis d’identifier de nombreux oncogènes activés par mutation somatique ; de plus, trois de ces gènes n’avaient jamais été décrits comme étant mutés dans des tumeurs humaines. Nous avons identifié des mutations activatrices du récepteur à l’IL-6, gp130 dans 60% des IHCA. Nous avons aussi retrouvé des mutations de FRK, une src-like kinase, dans 10% des IHCA, du facteur de transcription STAT3 dans 5% des IHCA, du gène GNAS dans 5% des cas, et de la tyrosine kinase JAK1 dans 1% des cas. Toutes les mutations identifiées étaient somatiques, monoalléliques et mutuellement exclusives. Nous avons pu montrer, dans des systèmes de lignées cellulaires hépatocellulaires, que l'expression des formes mutées de ces gènes est capable d’activer la voie IL-6/STAT3 en absence du ligand IL-6, contrairement aux protéines sauvages. Nous avons identifié des inhibiteurs pharmacologiques qui permettent d’inhiber de façon spécifique ces mutants et qui pourraient être utilisés en clinique pour le traitement des IHCA.Grâce à une technique de CGH-SNP, nous avons identifié des événements récurrents de pertes et gains de chromosomes associés aux groupes moléculaires d’AHC. De façon similaire, l’étude de la méthylation dans les AHC a permis de mettre en évidence un pattern spécifique à chaque sous groupe. Nous avons montré que l’instabilité chromosomique augmente progressivement dans les lésions borderline et dans les CHC développés sur AHC comparés aux AHC classiques. Le séquençage exome de 5 transformations malignes de AHC en CHC a identifié un nombre plus important de mutations dans les AHC qui ont transformé comparé aux AHC classiques ; ce nombre est significativement augmenté dans la partie CHC des tumeurs. La comparaison de la partie bénigne et maligne des tumeurs a mis en évidence l'activation de ß-caténine comme un évènement précoce dans le processus de transformation et a révélé la présence de mutations somatiques fréquentes dans le promoteur de la télomèrase (TERT), identifiées principalement dans la partie maligne des tumeurs.En conclusion, cette étude a permis d’identifier des mécanismes distincts conduisant à l'activation de STAT3 dans les IHCA, renforçant le rôle de la voie JAK-STAT3 dans la tumorigenèse bénigne hépatocellulaire ainsi que le lien entre Src kinases et inflammation. Ces travaux ont permis d’affiner la classification moléculaire des AHC avec des corrélations étroites
Hepatocellular adenomas (HCA) are rare benign tumors that develop most often in young women after taking oral contraception. The main complications are hemorrhage and rarely, malignant transformation to hepatocellular carcinoma (HCC). Recent work in the laboratory identified three main HCA molecular groups that are defined by (1) inactivation of the transcription factor HNF1A (H-HCA), (2) activation of the Wnt/ß-catenin pathway (bHCA) or (3) the presence of inflammatory infiltrates (IHCA).To identify tumorigenesis pathways associated with the development of inflammatory HCA (IHCA), a transcriptome analysis comparing IHCA to non-tumor liver was performed in the laboratory, leading to the identification of an activation of the IL-6/JAK/STAT3 pathway in these tumors. We sought new gene alterations and we characterized the activation mechanism of the IL-6/JAK/STAT pathway in IHCA. The functional consequences of the different mutants on the STAT3 pathway were analyzed by modeling their expression in hepatocellular cell lines. In addition, we performed integrated genomic studies (CGH-SNP analysis, methylome and exome sequencing) on a wide range of 250 HCA with the aim to refine the molecular classification of HCA, to identify new genes altered in these tumors and to elucidate the mechanisms of malignant transformation of HCA to HCC.In the group of the IHCA, we identified many oncogenes activated by somatic mutation; in addition, three of these genes were never been described as mutated in human tumors. We identified activating mutations in the IL-6 receptor gp130 in 60% of IHCA. We also found mutations in FRK, a src-like kinase, in 10% of IHCA, of the transcription factor STAT3 in 5% of IHCA, of the GNAS gene in 5% of cases, and of the tyrosine kinase JAK1 in 1% of the cases. All identified mutations were somatic and monoallelic and were mutually exclusive. We have shown in hepatocellular cell lines that the expression of mutated forms of these genes is able to activate the IL-6/STAT3 pathway in the absence of the IL-6 ligand, in contrast to wild-type proteins. We have identified pharmacological inhibitors that specifically inhibit the mutants and that could be used for the clinical treatment of IHCA.Using a CGH-SNP technique, we identified recurrent chromosomes gains and losses associated with the HCA molecular groups. Similarly, the study of methylation in HCA highlighted a specific pattern in each subgroup. We showed that chromosomal instability increases gradually in borderline lesions and in HCC developed on HCA compared to classical HCA. Exome sequencing of 5 malignant transformation of HCA to HCC identified a large number of mutations in the transformed HCA compared to classical HCA; and this number is significantly increased in HCC tumors counterpart. Comparison of benign and malignant tumors highlighted the activation of ß-catenin as an early event in the transformation process and revealed frequent somatic mutations in the promoter of the telomerase gene (TERT), identified mainly in the malignant part of tumors.In conclusion, this study has led to the identification of distinct mechanisms leading to the activation of STAT3 in IHCA, strengthening the role of the JAK-STAT3 pathway in benign hepatocellular tumorigenesis and the relationship between Src kinases and inflammation. This work helped to refine the molecular classification of HCA with tight correlations between genotype and phenotype, and led to advances in the identification of major genetic determinants involved in the process of malignant transformation

In my thesis I follow historical development of property rights in Europe. I focus in particular on relationship to land and records of property conditions in law. The important moments are relation between ownership and state and landed property as a policy instrument. The aim is to reveal, when and how the ownership as a legal institution came into being and how the status of an individual reflected his or her ability to acquire property. I pay attention to the most significant law-making factors and to the way how the ownership relations affect social order and economic performance at the particular time. The theoretical part firstly defines relevant terms of law and then deals with the institution of property in the history of philosophy and economic thoughts. The analytical part compares development of property rights in the Czech lands with that of state forms on the territory of nowadays Germany. The comparison sets this development in a wider frame of legal, political and economic history and observes the interaction of the both lands in the area of property conditions in the past.

This thesis covers primarily the detection of operating emission of an old Škoda Felicia car and the emission produced during the manufacturing of a new Toyota Prius, as the selected representative of an ecological car. The thesis also touches on hybrid technology in itself, the question of the greenness of hybrid cars, sphere of emission, but also related problems so called "scrapping" and its influence on environment and also current situation in automobile industry. The target of this study is calculation of operation time of an old Škoda Felicia during which the CO2 emission will reach the same amount of emission produced during manufacturing of a Toyota Prius. The objective is to point out incompetent and therefore not always serious comparison of cars greenness when only the operating CO2 is being compared. Other energy consumption and therefore also the greenhouse gases production is related with the car manufacturing, raw material extraction, petroleum processing and other economic sectors affected by production. Emission laboratory measurement executed by TÜV SÜD Czech discovered that the old Škoda produces 260m/km CO2 during its real running time. For the emission calculation was used an American model EIO-LCA created by Green Design Institute at Carnegie Mellon University that records ecological influence of car production, in stated amount, on all economy sectors. The outputs are quoted in CO2 equivalent. According to this model during manufacturing of 1 hybrid car 13.5 t of CO2e (equivalent CO2). is produced. Recalculation found out that the old Škoda Felicia could run for more than 5 years in order to produce as much emission as the manufacturing of new Toyota Prius third generation.

Jane Austen wrote her novels over two hundred years ago. Today many people, especially women, are still affected by them and her characters. She has become famous through her romantic novels where she writes about young women during the late 18th century who spend their days drinking tea and socializing in order to find a man, marry him and live happily ever after. Even though Austen writes romance and her novels remind the reader of fairy tales, she also focuses on presenting important passages and events that occur in these young women’s lives. Many of the novels Austen has written have features of a so-called Bildungsroman; a novel about education which refers to a character’s growth and self-development. The structure of a Bildungsroman often includes the main character, the protagonist, going on a long journey or quest in search of the meaning of life. In this essay I will analyse the heroine’s education in Austen’s two novels Northanger Abbey and Mansfield Park and how Austen educates the reader with these novels. The purpose of this essay is to show that the heroines in Northanger Abbey and Mansfield Park under a long period of time receive moral education through different people and events during their lives. However, it is not only the characters that are educated, my opinion is that the reader is educated as well. Both the reader and the heroines are taught that happiness can only be achieved by good education and high moral standards. I will use some of the features of a Bildungsroman, such as journey, self-development, obstacles and maturity and by examining these features in the novels, I will support my thesis.

Com o crescimento das redes de computadores mundiais e o aumento do uso de aplicações distribuídas, surge um aumento significativo no tráfego de dados no enlace, demonstrando, assim, a necessidade de primoramento - ou substituição das atuais técnicas de transmissão de dados - do modelo tradicional cliente-servidor. A tecnologia de agentes móveis tem sido alvo de diversas pesquisas na área, por permitir que apenas um código mova--se entre os nós da rede e retorne com os resultados, diminuindo, assim, a carga na rede. Para que um agente móvel possa cumprir sua missão, é necessário que se atenda a um deadline. Entretanto, em um sistema distribuído, há a possibilidade de concorrência por um mesmo recurso. Tratar de forma adequada tal concorrência é de suma importância, especialmente em um ambiente em tempo real. Em face disso, um modelo de execução em que agentes móveis disputam um mesmo recurso em um mesmo host é proposto, o RT-JADE: uma extensão de middleware que possibilita agentes móveis concorrentes cumprirem suas missões, usando métodos de escalonamento em tempo real sobre a plataforma JADE. A eficácia do modelo proposto foi demonstrada através de simulações e comparações entre diversas políticas de escalonamento, com distintas cargas de trabalho (quantidade de agentes concorrentes).
With the growth of worldwide computer networks and increased use of distributed applications, there is a significant increase in data traffic in the link, thus demonstrating the need for improvement # or replacement of existing techniques for data transmission # of the traditional client-server one. The mobile agent technology has been the subject of several researches in area, because it only allows a code to move between network nodes and return with the results, reducing the network#s load. For a mobile agent to accomplish its mission, it is necessary for it to meet a deadline. However, in a distributed system there is the possibility of competition for the same resource. Treating such competition adequately is very important, especially in a real-time environment. In this view, an execution model in which mobile agents compete for the same resource in the same host is proposed # RT-JADE: a middleware extension that allows concurrent mobile agents to achieve their missions, using real-time scheduling methods on the JADE platform. We demonstrate the effectiveness of the proposed model through simulation and comparison of different scheduling policies under different workloads (number of competing agents).

The production of Nitric Oxide (NO) under physiological conditions has beneficial roles in acting as a key signaling component of many biological processes as well as having an anti-microbial effect. However its effects following excess production by the inducible NO pathway is potentially detrimental in the pathogenesis of chronic inflammation including sepsis and several other inflammatory diseases. Understanding the mechanisms that regulate the expression of the inducible nitric oxide synthase (iNOS) responsible for producing the excessive amounts of NO in disease states is therefore critical. In this regards, experiments were carried out to identify the signaling pathways that may mediate this process, focusing specifically on the JAK/STAT cascade. The reason for selecting the latter is because our research group, amongst others, has carried out extensive work investigating other signaling pathways, including the mitogen activated kinases (MAPK). Moreover, studies have also been carried out in an attempt to identify the critical role of JAK/STAT signaling for iNOS induction. These studies however failed to conclusively demonstrate whether, as with the MAPKs, the JAK/STATs may also play an essential role. Furthermore there is indeed controversy in the literature with researchers unable to agree whether expression of iNOS does require JAK/STAT activation. Thus, the aim of the project described in this thesis was to establish unequivocally whether activation of the JAK/STATs preceeds induction of iNOS. The studies were extended to L-arginine transport as well because the latter is widely reported to be induced in parallel with iNOS and substrate supply to iNOS may be critical for sustained NO production. Changes in transporter activity as well as their expression profiles were assessed. All experiments were carried out in either rat aortic smooth muscle cells (RASMCs) or in the J774 macrophage cell line. These cell types were selected because RASMCs are one of the prime targets for induced NO production in vascular inflammation and the macrophages are involved in host defence, acting in part through NO production. To establish the role of JAK/STATs, pharmacological and molecular approaches were used. Pharmacologically, two inhibitors were used and these were AG490 and JAK inhibitor I. The former is reported to be a selective JAK2 inhibitor and the other blocks all known JAK proteins. The potential of the GTPases to regulate the induction of iNOS was also examined using selective inhibitor known to regulate these proteins. In addition to these drugs, siRNA targeting JAK2 was also exploited and western blotting was extensively used to detect expression of various proteins including iNOS, native and phosphorylated JAK2 and TYK2. Changes in iNOS activity was monitored by determining nitrite production using the Griess assay and L-arginine transport was monitored using tritiated arginine (L-[3H]arginine). RASMCs were treated with a combination of LPS (100 µg/ml) and IFN- (100 U/ml) and the macrophages with LPS (1 µg/ml) to induce iNOS and transporter activity. Consistent with previous reports, the above treatment of both cell types resulted in the expression of iNOS, production of NO and enhanced transport of L-arginine. These effects were not affected by AG490 but blocked by JAK inhibitor I. Furthermore, although both cell types expressed the key JAKs (JAK2 and TYK2), neither of these proteins were phosphorylated under conditions of induced NO production. Moreover, siRNA experiments showed that JAK2 expression could be abolished without any significant change in NO production, confirming that at least JAK2 may not be required for this process. Whether TYK2 is involved still remains to be resolved as the phosphor-protein could not be detected. However the conclusive siRNA knockdown studies could not be carried out due to time and cost constraints. Apart from iNOS and NO production, changes in induced L-arginine transport were also not significantly affected under the experimental conditions described above suggesting that like with iNOS, induction of L-arginine transport is independent of at least JAK2. Interestingly however, STAT-1 was phosphorylated and this was blocked by JAK inhibitor I but not AG490. Thus, STAT-1 activation may be essential but its activation may be independent of the JAKs. One possible alternate upstream activator of STAT-1 may be the GTPases. Indeed these proteins have been indicated to phosphorylate STAT-1 independent of the JAKs. However, in this project, inhibition of the GTPase pathway enhanced NO production and L-arginine transport suggesting that the GTPases downregulate these processes. In conclusion, the studies carried out in this thesis have shown that induction of iNOS, NO production and L-arginine transport in both RASMCs and J774 macrophages are independent of JAK2 but require STAT-1 activation which may be phosphorylated independently of the JAKs. The role of other JAKs such as TYK2 although unlikely, will need to be resolved using a more specific approach such as siRNA.

Resumen
Drama character as a dream, acter in person as it?s life. This work is a study of a diference between an actor in person and a drama character. As to my absolvent role of Segismundo in ?Life is a dream? by Calderón de la Barca the objective is to determine the diference in perception of the actor. On one hand as a person (at rehearsals, looking for attributes for his character) and on the other hand as the character (during performances, perceived like that by the audience). This study is completed with specific examples from the above mentioned play, it?s rehearsals at the DISK theatre and my personal interpretation of the Segismundo character. The main source for my approach was ?Člověk v situaci? by Jan Císař.
