Rohsig, Lopez Nicole Sofia. Electronic Thesis or Diss., Bordeaux, 2024. https://theses.hal.science/tel-04828656.

Texto completo
This CIFRE thesis was done in partnership with Decathlon, particularly in the Watersports Center in Hendaye, where they develop gear for aquatic sports. This thesis aims to explore the transition of the company to the circular economy, an industrial system that replaces the linear logic of “extract- produce-use-throw away”. This system proposes closing the material loops to extract their maximum embedded value, resulting in the elimination of waste through the superior design of materials, products, systems, and business models (Ellen MacArthur Foundation, 2013).In a press release from March 2024, Decathlon positioned the development of Circular Business Models at the core of its strategy to achieve a 20 % reduction in absolute CO2 emissions by 2026 and a 42 % reduction in absolute CO2 emissions by 2030 and net zero by 2050. Circular Business Models (CBM) involve cycling, extending, intensifying, and/or dematerialising material and energy loops to reduce resource inputs, waste, and emission leakage (Geissdoerfer et al. , 2017). In this context, the question that guided the beginning of our research emerges: How can we help develop circular business models in the sports industry, given its diverse nature of products? After mapping 154 internal CBM projects, project leaders mentioned challenges such lack of strong governance and a structured circular network to facilitate the progression of circular projects, besides the difficulty of engaging consumers to adopt circular solutions. This motivated us to explore what factors could influence consumer behaviour towards CBMs, whether internal or external. We interviewed 60 aquatic sports practitioners to identify internal factors (e.g., economic, environmental, time and effort, aesthetics) and external factors (e.g. access to circular solutions, prices, product condition). Following this step, we launched a survey with 1735 respondents to measure the consumer interest in different CBMs (repair, second-hand, short-term rental, subscription, end-of-life alternatives) regarding specific product natures (protective equipment, first-layer textile, second-layer textile, budget and premium shoes, accessories, and bulky equipment) for certain sports (swimming, soccer, cycling, hiking, fitness, and running).Building on these results and the literature findings, we created a tool to help sports design teams identify suitable CBM for their products and get recommendations on how to improve circularity, considering product characteristics and customer behaviour. The tool was tested and approved by product managers, especially for beginners, regarding circular economy adoption. Our contribution relies on in the creation of an interactive decision-aid tool that is easy to use, enabling a better understanding of how to apply circular economy considering both technical and behavioural aspects. Future works could explore the adaptation of the tool for other industries, as well as the understanding of the acceptance of circularity by consumers from other cultures outside France

Vivarelli, Agathe.

This thesis, structured as a series of essays, explores the various facets of the digitalization of the sports spectacle experience from the perspectives of both managers and consumers, both within and outside sports venues. The first essay introduces an experience delivery system and a mapping of the sports spectacle consumer journey, highlighting why and how different stakeholders, particularly clubs, integrate technologies into the experience. The second essay examines how club mobile applications transform consumer experiences, emphasizing the tensions between real and virtual social connections, as well as the ambivalent effects of digitalization (value creation versus value destruction). Finally, the third essay expands the analysis by focusing on player-embedded technologies (such as bodycams), which alter media coverage and, consequently, the sports spectacle experience itself. These technologies lead spectators through a transition from a form of embodiment to a sense of disembodiment in relation to the athlete on the field. Together, these three essays offer a broad and comprehensive view of how digitalization is reshaping the sports spectacle experience, revealing both the opportunities and challenges it creates

In this thesis we defend the idea according to which it is possible to think strategies of brand positioning according to a socio-political approach to markets to the heart of which is the social field concept. The main contribution of our research is to attempt to produce an original typology of strategy of positioning of the challenger firms, or at least to have formulated a proposition. This is not only descriptive but presents the advantages to be constructed theoretically: it constructs itself on the basis of the report by the rules dictated by the market enterprise leaders. The second contribution of our research is to offer a framework that permits to think better the position of enterprises leaders identifying its strategic problem. In order to become the market leader and to preserve and to perpetuate this position this requires thinking through the positioning in terms of legitimacy. The Weber classification of the three types of domination legitimate appears to us especially indicated like a possible reading grid. The third contribution of our research is linked with the sociological perspective of the market in which we take place. Rather than to consider the market only like a place where meets the offer and the demand of a certain good, we consider it more of a dynamic field

Resumen
Sport, once an amateur pursuit has evolved in to a lucrative industry. The most recent development in the evolution of Australian sport has been the emergence of industrial regulation. Unlike other Australian labour markets, the sports administrators labour market was entirely market regulated until 1994. Over the last five years the sports administrators labour market has transformed into a centralised award based system. On the surface it appears that there is no obvious explanation for the dramatic re-regulation of this labour market. In order to determine the factors behind the re-regulation, this thesis investigated Australian sports administrators attitudes to unionism, awards and enterprise bargaining, including their preferences to awards and enterprise bargaining agreements. The attitudes of 229 Australian sports administrators were surveyed. The response rate was 67.25%. Findings from the survey indicated a number of points: (1) union membership predicted 8% of their attitude to unionism, (2) non union members were more likely to have positive and accepting attitudes to unions, (3) sports administrators working under Enterprise Bargaining agreements had more positive and accepting attitudes of unions, (4) Enterprise Bargaining was considered to be more beneficial than not in sport, (5) Enterprise Bargaining had more than double the support of Award regulation, and (6)Award regulation had almost equal amounts of rejection and support. Other results indicated that the sports administrators labour market remained market regulated until 1994 because a majority of sports administrators belonged to demographic groups which were less inclined to become union members. Factors behind the reregulation were determined to be: strong support for targeted services within workplaces rather than generic services across an industry, and strong support for increased union interaction when negotiating terms and conditions of employment which effects sports administrators' attitudes to unionism. It was evident that the re-regulation was not caused by a large shift in the attitudes of sports administrators or a result of problems stemming from the market being entirely by market regulation. It is more than likely that the sudden re-regulation of the sports administrators labour market was the sports industry's first step towards industrial maturity.

Resumen
Tourism and Sport
D.B.A.
Esports, or competitive video gaming competitions, have grown in popularity to have millions of global fans, spectators, and participants. Sport organizations, including leagues and teams, are increasingly affiliating, or dovetailing, with esports organizations. Although the meteoric rise of esports presents sport organizations with growth opportunities to connect with these markets, esports remain shrouded in uncertainty, stigma, and stereotypes that impede the convergence of industries. The growing affiliation between these two industries allows researchers to gain insight into the strategic actions of sport organizations despite the institutional constraints that influence their behavior. This dissertation includes three essays that address—with respect to esports—the (1) strategic resources and core competencies of sport organizations; (2) sensemaking of actors within sport organizations; and (3) institutional creation strategies within heavily regulated fields. Essay one consists of an explanatory, qualitative study of professional sport teams diversifying into the esports industry. As sport organizations mature, they will need to seek growth opportunities beyond their core industry and compete with formally tangentially related firms. The nascent esports industry has been embraced by sport organizations (e.g., leagues, teams), many of whom use existing resources and best practices from traditional sport to manage their esports property. This trend provides a salient context to identify how sport organizations are using their resources in a new marketspace and what they can provide to non-sport organizations. Guided by strategic management perspectives of the resource-based view (RBV), knowledge-based view (KBV), and resource orchestration (RO), the purpose of essay one is to examine how sport organizations leverage their core competencies to create a sustainable competitive advantage. The findings support the hypothesis that the existing resources of sport organizations such as physical venues and existing departments (e.g., legal, marketing, sales departments) are used to support sport organizations operations in the esports industry. Moreover, the findings identify that the tacit knowledge embedded in the human capital of sport organizations is a salient resource that helps them generate a competitive advantage against non-sport organizations. While tacit knowledge is a driver of competitive advantage, it is the supporting structures and departments that allows sport organizations to use this knowledge efficiently and effectively. Thus, by operating at the intersection of sport, entertainment, and media, sport organizations are increasingly competing with formally tangentially related firms, and can use their embedded resources and structure when competing with these firms. Essay two focuses on the integration of esports by a first-mover sport organization. Essay two employs an exploratory case study of the Philadelphia 76ers of the National Basketball League (NBA) and their integration of an esports team. The 76ers were the first North American professional sports organization to purchase and integrate an esports team. Novel technologies and practices, such as esports, are surrounded by uncertainty and are generally met with resistance (Huber, 1990) with their integration contingent upon internal and external constituent support. Actors, individuals within the 76ers, had to position and garner support for esports, which are representative of the novel practices and technologies influencing the way sport organizations are managed. In essay two an institutional creation work perspective is linked with sensemaking and related constructs to examine how actors within the 76ers helped create and give meaning—sensemaking—to esports. The findings of essay two support how actors within the 76ers were able to foster a progressive culture to create a shared understanding of esports and use this understanding to influence the sensemaking of others—sensegiving. In particular, the 76ers linked the managerial components of operating a sports team to operating an esports team. Yet the 76ers were deliberate in their approach, limiting cross-promotions between the fanbases of the 76ers and their esports team. By connecting institutional creation work with sensemaking, essay two contributes to how actors can proactively take actions to garner constituent support. Moreover, the findings of essay two support that sensemaking is a critical antecedent of sensegiving, as a shared understanding within an organization is a necessary prerequisite to influence the sensemaking of others (i.e., sensegiving). Essay two provides actors within the field of sport integrating novel practices and activities (e.g., augmented and virtual reality, in-game sports betting, wearable fitness devices, mediated sports consumption) strategies to proactively garner support for their integration. Essay two focuses on the integration of esports by professional sport organizations. Essay three utilizes an exploratory qualitative approach to identify the institutional creation strategies associated with integrating collegiate esports programs within the heavily regulated field of U.S. collegiate athletics. The findings of essay three support how actors need to go beyond creating a shared understanding to integrate novel activities. Actors must also influence the cognitive schema of other actors to facilitate the integration of novel practices and activities. Specifically, they can build on the concept of sportification (Heere, 2018) to both communicate and present esports in a manner consistent with traditional sports, using the concept of sport as a legitimizing agent. Collectively, the three essays support how sport organizations can seize growth opportunities with respect to their institutional environment. Sport organizations must recognize their institutional confines, but also can be strategic in their actions by focusing on their financial performance and sustainability in lieu of their constraints. This research contributes to a deeper understanding regarding how the institutional and strategic concerns of sport organizations influence their efficient and effective management. The research lays a foundation for a stream of future research on the strategic growth and long-term viability of sport organizations both within and beyond the esports industry.

Microbial contaminants of safety concern represent most of time, in canned food, an industrial risk which is well mastered. However, the spoilage flora, due to its high heat resistance, is responsible for major economic losses. Nevertheless, these bacteria remained poorly characterized. Based on the works realized during last 10 years within the EMaiRIT'S unit of microbiology of the CTCPA (expertise unit of the French Technical Center of the Preservation of Food, focused on Management of Industrial Risk liked to Heat Resistant Spores), the main objective of this thesis were: i) to identify and to characterize, with the aim of its later control, the spoilage spore forming bacteria florae ii) to identify the origin of these florae in canning factories and finally iii) to determine ways of control. For that purpose, a current inventory of spore forming bacteria in spoiled canned food was made with the cooperation of 122 canning factories over more than 10 years in France. This characterization of the spoilage species allowed the elaboration of a molecular biology tool (SporeTraQTM) for quick identification of these germs or their detection within a complex population. In parallel, the improvement of the knowledge about the heat resistance of these species, main characteristic of the spores, was led. In addition, the chemical resistance of spores was investigated. When identified, we tried to localize these spores on canning factories lines, with several sampling plans, on various vegetables. At the end, the specific spore forming bacteria related to the industrial canning process was identified, characterized and localized, allowing to improve the microbial risk control either by a more efficient cleaning, and through optimized process schedules. Furthermore, this work was driven within a benefic / risk approach representing the future of the food-processing evolution with improvement of the nutritional quality and the preservation of the sanitary control. This thesis leans on 5 publications of rank A

Resumen
This dissertation is conducted to the study of a current topic in the economic scope, which is the result of the application of techniques and methodologies of analysis typical of Economics to the sports field. Sports Economics is a rather novel discipline, compared to other branches of Economics, whose results have a growing consideration within the academic world, sparking the interest of many researchers. The final motivation of the three chapters presented in this work is to delve into some crucial aspects of sports such as production function, efficiency, competitive balance and heterogeneity of the inputs included in the team, from an economic perspective. In all of them this dissertation tries to contribute to the literature on the sports economics with new variables and experiments, as well as to debate about the most spread theories and assumptions. The first chapter analysis the efficiency and the evolution of the Total Productivity in Spanish First division Basketball teams during their productive process, just considering the performance on the field. The empirical part relies on the use of non-parametric techniques of optimization, measuring efficiency with respect to the best observations of the sample through a data envelopment analysis (DEA). This technique is the more suitable for sports given that it does not consider a specific functional form for the frontier as well as the absence of errors. The results obtained make sense for both, the teams themselves (coaches and managers) and for the regulatory body. Fort he case of the teams as much as they can undertake actions that improve their productivity. Fort he regulatory body the results are determinant to take initiatives, which encourage the evolution of the efficiency of the teams as a whole. The second part addresses the issue of the competitive balance comparing the two conferences in which are divided the NBA teams, and its relationship with demand, which is not clear in the literature. The methodology of analysis is based on Gibrat’s clasical model using the number of relevant competitors as an indicator of competitive balance which avoids some of the problems that the typical indexes of concentration present when it comes to sports competitions. This paper delves into the relationship between competitiveness and spectator’s demand, keeping in mind the Rottenberg’s hypothesis, or which is the same, the willingness of attending the sport event increases with the level of competitiveness. The Granger causality tests are not conclusive due to the level of spectacle reached by the competition independently of the sport game in particular, which makes very common the sold-out in almost every game. Chapter 3 tackles the configuration of the teams in terms of optimal level of diversification providing experimental evidence on the relationship between outcomes achieved and heterogeneity of the inputs. The methodology is based on the diversification index of Gollop and Monahan whose decomposition provides much more information. Every component of the index make sense by themselves and are used in the second part of the paper in different estimations as determinants of the performance of the teams. All in all, the three pieces contained in this dissertation address the relationship between economy and sports, but every paper can be read independently considering the different analysis accomplished in each of them.

Texto completo
Resumen
Spore germination is a common and fundamental event in fungal development and in many instances an essential phase of fungal infection and dissemination. Spore germination is also critical for hyperparasites to function as biocontrol agents as well as in fermentation proceses. Our common objective is to understand the mechanisms which regulated spore germination and identify factors involved in pathogenicity related prepenetration development. Our approach is to exploit the overall similarity among filamentous fungi using both a plant pathogen (Colletotricum trifolii) and a model system that is genetically sophisticated (Neurospora crassa). The simulataneous use of two organisms has the advantage of the available tools in Neurospora to rapidly advance the functional analysis of genes involved in spore germination and development of an economically important fungal phytopathogen. Towards this we have isolated a protein kinase gene from C. trifolii (TB3) that is maximally expressed during the first hour of conidial germination and prior to any visible gene tube formation. Based on sequence similarities with other organisms, this gene is likely to be involved in the proliferative response in the fungus. In addition, TB3 was able to functionally complement a N. crassa mutant (COT-1). Pharmacological studies indicated the importance of calmodulin in both germination and appressorium differentiation. Using an antisense vector from N. crassa, direct inhibition of calmodulin results in prevention of differentiation as well as pathogenicity. Both cAMP dependent protein kinase (PKA) and protein kinase C (PKC) like genes have been cloned from C. trifolii. Biochemical inhibition of PKA prevents germination; biochemical inhibitors of PKC prevents appressorium differentiation. In order to analyze reversible phosphorylation as a regulatory mechanism, some ser.thr dephosphorylative events have also been analyzed. Type 2A and Type 2B (calcineurin) phosphatases have been identified and structurally and functionally analyzed in N. crassa during this project. Both phosphatases are essential for hyphal growth and maintenance of proper hyphal architecture. In addition, a first novel-type (PPT/PP5-like) ser/thr phosphatase has been identified in a filamentous fungus. The highly collaborative project has improved our understanding of a fundamental process in fungi, and has identified targets which can be used to develop new approaches for control of fungal plant pathogens as well as improve the performance of beneficial fungi in the field and in industry. In addition, the feasibility of molecular technology transfer in comparative mycology has been demonstrated.

The apparel industry in Haiti is poised to move beyond the "cut and sew" processes which have characterized its operations to the present time. Chemical usage has largely been limited to spot cleaning and some washing activities, but as washing increases and other fabric treatment processes are integrated into the Haitian industry, chemical usage at these factories will increase and become more varied. The factories must be prepared to identify and control the chemical hazards associated with these newly integrated operations which may pose risks to workers, the community and the environment.

Texto completo
Resumen
Recent environmental concerns have led to calls for reduced hydrocarbon emissions to the atmosphere from a variety of sources. One source of emissions being examined in this regard is natural gas spot sampling methods that vent pipeline gases to the atmosphere. Some sampling techniques and equipment have been developed that do not emit greenhouse gases, but the need exists to test these methods for their ability to collect accurate, representative samples. Another related concern is the accuracy of samples drawn from streams near their hydrocarbon dew point (HDP). While the spot sampling methods recommended by current industry standards perform well on streams far above their HDP, little data are available on their performance near or at the HDP, where poor sampling methods can cause heavy hydrocarbons to condense from the sample and distort the analysis. This project evaluated the ability of four natural gas spot sampling methods, including two zero emissions sampling methods, to capture accurate, representative samples of gas streams at or near their hydrocarbon dew point (HDP). Two of the sampling methods tested were variations on the GPA fill-and-empty method, with additional steps intended to heat the sampling equipment above the HDP or clear condensed hydrocarbon liquids from the sample line. The other two sampling methods, which use the A+ Q2 sample cylinder and a constant-pressure floating-piston sample cylinder, were developed to prevent condensation of heavy hydrocarbons during the sampling process.

Texto completo
Resumen
This report documents the work performed to evaluate the possibility that temperature log anomalies (cold spots or departures from temperature linearity with depth) can be indicators of well leaks. The natural gas cavern storage industry does not have a methodology for accurate gas-filled cavern well Mechanical Integrity Tests (MITs). Analyses of some temperature log anomalies in North American gas cavern well completions revealed that temperature log anomalies can be indicators of well leaks. The challenges in applying this technology to quantify gas-filled cavern MITs are threefold: (1) Does a temperature anomaly (cold spot) always indicate a leak? (2) Can a leak magnitude be correlated to a temperature log anomaly magnitude? and (3) What protocol should be used for executing such an MIT? This report is not intended to completely address all three issues noted above. However, the research presented here is one of many steps needed to evaluate the possibility of temperature logging as an MIT. Research activities discussed in this report involve computational fluid dynamics (CFD) modeling of both a well and cavern. Specifically, the response from a representative cold spot introduced in the cement sheath of a well and the response from an actual leak are addressed.

Resumen
Increased automation for aircraft systems holds the promise to increase safety, precision, and availability for manned and unmanned aircraft. Specifically, established aviation segments, such as general aviation and light sport, could utilize increased automation to make significant progress towards solving safety and piloting difficulties that have plagued them for some time. Further, many emerging market segments, such as urban air mobility and small unmanned (e.g., small parcel delivery with drones) have a strong financial incentive to develop increased automation to relieve the pilot workload, and/or replace in-the-loop pilots for most situations. Before these advances can safely be made, automation technology must be shown to be reliable, available, accurate, and correct within acceptable limits based on the level of risk these functions may create. However since inclusion of these types of systems is largely unprecedented at this level of aviation, what constitutes these required traits (and at what level they must be proven to) requires development as well. Progress in this domain will likely be captured and disseminated in the form of best practices and technical standards created with collaboration from regulatory and industry groups. This work intends to inform those standards producers, along with the system designers, with the goal of facilitating growth in aviation systems toward safe, methodical, and robust inclusion of these new technologies. Produced by members of the manned and unmanned small aircraft community, represented by ASTM task group AC 377, this work strives to suggest and describe certain fundamental principles, or “pillars”, of complex aviation systems development, which are applicable to the design and architectural development of increased automation for aviation systems.

Israel, Alvaro y John Merrill. Production of Seed Stocks for Sustainable Tank Cultivation of the Red Edible Seaweed Porphyra. United States Department of Agriculture, 2006. http://dx.doi.org/10.32747/2006.7696527.bard.

Texto completo
Resumen
Porphyra species (commonly known as ‘nori’ or ‘purple laver’) are edible red seaweeds rich in proteins, vitamins and other highly valued biogenic compounds. For years Porphyra has been cultured using seeded nets extended in the open sea, and its biomass consumed primarily in the Far East. While demands for international markets have increased steadily at an average of 20% per year, supplies are on the verge and not expected to meet future demands. Alternatively, land-based cultivation of seaweed has become attractive in the mariculture industry since (1) important growth parameters can be controlled, (2) is environmentally friendly and (3) perfectly matches with integrated aquaculture leading to sustainable, high quality products. During the last few years a tank cultivation technology for Porphyra has been developed at the Israeli institution. This technology is based on indoor production of asexual spores and their subsequent growth to 1-2 mm seedlings. The seedlings are then transferred to outdoor tanks and ponds when seawater temperatures drop to 20 °C, or below, and days become shorter during winter time. However, the current technology efficiently serves only about 100 m2 of ponds during one growth season. In order to produce seedlings in sufficient amounts, it is critical to address both technical and biological aspects of seedling production, securing optimal up-scale to commercial-size cultivation farms. We hypothesize that massive production of spores is related to thalli origin, thalli age and sporulation triggers, and that seedling survival and their subsequent growth potential is determined by the seawater quality and overall indoor growth conditions imposed. A series of bio-reactors were constructed and tested in which spore release and spore growth were separately studied. The main assessment criteria for optimal viability of the seedlings will be by determining their electron transport rate using PAM fluorometry and by subsequent growth and biomass yields in outdoor ponds. Altogether the project showed (1), controlled sporulation is possible in big outdoor/growth chamber settings provided initial stock material (small frozen seedlings) is at hand, (2), contamination problems can be almost completely avoided if stock material is properly handled (clean as possible and partially dehydrated prior to freezing), (3), spore release can significantly be enhance using high nutrient levels during thawing for P. yezoensis and P. haitanensis, but not for P. rosengurttii, (4), PAM fluorometry is an efficient tool to estimate growth capacity in both seedlings and juvenile thalli. The BARD funding also served to explore other aspects of Porphyra biology and cultivation. For example, the taxonomical status of Porphyra strains used in this study was defined (see appendix), and the potential use of this seaweed in bioremediation was well substantiated. In addition, BARD funding supported a number of opportunities and activities in the Israeli lab, direct or indirectly related to the initial objectives of the project such as: additional molecular work in other seaweeds, description of at least 2 new species for the Israeli Mediterranean, and continuous support for the writing of a book on Global Change and applied aspects of seaweeds. The technology for Porphyra cultivation in land-based ponds is readily available. This study corroborated previous know-how of Porphyra growth in tanks and ponds, and yet offers important improvements regarding seedling production and their handling for successful cultivation. This study supported various other activities opening additional important issues in the biology/cultivation/use of Porphyra and other seaweeds.

Resumen
Electric resistance welded (ERW) pipe has been used in the natural gas transmission industry for many years. The Department of Transportation (DOT) has recently expressed interest in the integrity of the weld seam in pipelines made from ERW pipe that was manufactured prior to 1970. Specifically, the DOT has requested that natural gas transmission and hazardous liquid pipeline operators determine whether or not their pipelines that meet this description require hydrostatic proof testing. The initial concern from the DOT was for seam weld selective corrosion, although reference has since been made to growth of manufacturing discontinuities in the ERW seam. Early ERW pipe was manufactured using either direct current or low-frequency alternating current, processes that were prone to producing incomplete fusion discontinuities. These discontinuities (also referred to as cold welds, penetrators, etc.), if present in a pipeline, can grow under normal service or under upset operating conditions resulting in leaks or ruptures. There exists a need for a method that a pipeline operator could use to demonstrate the integrity of pipelines that were made from this older ERW pipe other than hydrostatic testing, which is expensive and potentially harmful to pipeline integrity. The use of a nondestructive-examination (NDE) technique would enable an operator to sample the integrity of a suspect pipeline during other routine in-service maintenance operations without the need for hydrostatic testing. The detection of incomplete fusion discontinuities is difficult if not impossible with conventional NDE techniques. The intimate mechanical contact between compressed surfaces causes a small amount of reflection and a large amount of transmission of ultrasonic signals. Recently however, several ultrasonic techniques have been introduced to study these types of discontinuities in resistance spot welds and inertia friction welds. These new techniques are based on ultrasonic spectroscopy, that is, the frequency dependence of the various ultrasonic interfacial parameters. These parameters include reflection and transmission coefficients and the frequency dependence of ultrasonic volumetric parameters (e.g., velocity and attenuation).

Resumen
The body condition (BC) score is recognized in the dairy industry as an essential tool for managing the energy reserves of the dairy cow, which is essential for sustaining optimal and efficient production over several lactations. The current use of BC scoring depends on the accuracy of subjective visual estimates, and this limits its kusefulness as a management aid in the dairy industry. A measuring tool that would frequently provide objective data on the cow's body reserves would be a major contribution to efficient dairy herd management. Ultrasonic sensors have the potential to be developed into an efficient BC measuring device, and the experimental use of such sensors for subcutaneous fat thickness (SDFT) estimates, as an indication for BC in beef cattle, supports this assumption. The purposes of this project were: 1. To compare visual BC scoring and ultrasonic fat thickness with on-line automated body weight (BW) measurements as monitors of nutritional adequacy of dairy cows at various stages of lactation. 2. To determine the effects of variation in digestive fill in early and late lactation on the accuracy of body weight measurements in lactating cows. 3. To modify an existing ultrasonic system and develop a specialized, low-cost sensor for repeatable determination of body condition scores by users with minimal training and skill. 4. To develop a standard for the assignment of body condition scores based on ultrasonic measurements of subdermal fat thickness. The procedure to execute these objectives involved: 1. Frequent measurement of BW, milk yield (MY), BC (visually scored) and subdermal fat thickness ultrasonically measured of dairy cows, and data analysis on average and individual basis. 2. Testing and selection of an appropriate special-purpose sensor, finding an optimum body location for working an ultrasonic measurement, prcessing the signals obtained, and correlating the resulting measurements with performance responses in lactating cows. Linking the ultrasonic signals to BC scores, and developing a BC scoring data acquisition system are the first steps towards fulfilling the necessary requirements for incorporating this device into an existing dairy herd management system, in order to provide the industry with a powerful managment tool. From the results obtained we could conclude that: 1. BC does not correlate with BW changes during all stages of lactation, although in general terms it does. These results were confirmed by individual cow BW and BC data obtained during the course of lactation, that were supported by individual objective ultrasonic measurement of SDFT. 2. BW changes reflect energy metabolism reliably ony after peak milk yield; early in lactation, a decrease in BW expresses mobilization of body reserves only qualitatively, and not quantitatively. 3. Gastrointestinal content increases throughout the whole period during which dry matter intake (DMI) increases. The drastic increase very early in lactation prevents the use of BW changes as a basis for quantitative estimatio of energy meatabolism; at this stage of lactation, konly a BC score or any other direct measurements willl provide a quantitative estimate of energy metabolism. 4. Ultrasonic measurements of subdermal fat thickness can be used to quantify changes that correlate with the actual condition of the cow, as assessed by performance and the traditional way of scoring. 5. To find the best site on the cow's body at which to obtain responses to BC and its changes in the course of lactation, additional sites have to be examined. From the present study, it seems that the sites between ribs 12 and 13 have the potential for this purpose. 6. The use of templates made it easier to repeat measurements at a desired site and spot. However, the convenient easy-to-handle way to standardize the measurement, described in this study, koffers scope for improvement. 7. The RF peak values of the A-mode are better indicators of the location of fat layer borders than image analysis, from the point of view of future commercial development. 8. The distances between the RF peaks of the A-mode can be automatically measured by suitable software, for future commercial development. 9. Proper analysis of daily body weight and milk yield data can provide the necessary information on body condition changes during lactation, until a direct BC measurement device is developed. 10. In any case, at least one visual BC assessment has to be done, preferably immediately after calving, for calibration purposes.

Resumen
Objectives: The overall objective as defined in the approved proposal was the development of a whole-cell sensor panel for the detection of endocrine disruption activities of agriculturally relevant chemicals. To achieve this goal several specific objectives were outlined: (a) The development of new genetically engineered wholecell sensor strains; (b) the combination of multiple strains into a single sensor panel to effect multiple response modes; (c) development of a computerized algorithm to analyze the panel responses; (d) laboratory testing and calibration; (e) field testing. In the course of the project, mostly due to the change in the US partner, three modifications were introduced to the original objectives: (a) the scope of the project was expanded to include pharmaceuticals (with a focus on antibiotics) in addition to endocrine disrupting chemicals, (b) the computerized algorithm was not fully developed and (c) the field test was not carried out. Background: Chemical agents, such as pesticides applied at inappropriate levels, may compromise water quality or contaminate soils and hence threaten human populations. In recent years, two classes of compounds have been increasingly implicated as emerging risks in agriculturally-related pollution: endocrine disrupting compounds (EDCs) and pharmaceuticals. The latter group may reach the environment by the use of wastewater effluents, whereas many pesticides have been implicated as EDCs. Both groups pose a threat in proportion to their bioavailability, since that which is biounavailable or can be rendered so is a priori not a threat; bioavailability, in turn, is mediated by complex matrices such as soils. Genetically engineered biosensor bacteria hold great promise for sensing bioavailability because the sensor is a live soil- and water-compatible organism with biological response dynamics, and because its response can be genetically “tailored” to report on general toxicity, on bioavailability, and on the presence of specific classes of toxicants. In the present project we have developed a bacterial-based sensor panel incorporating multiple strains of genetically engineered biosensors for the purpose of detecting different types of biological effects. The overall objective as defined in the approved proposal was the development of a whole-cell sensor panel for the detection of endocrine disruption activities of agriculturally relevant chemicals. Major achievements: (a) construction of innovative bacterial sensor strains for accurate and sensitive detection of agriculturally-relevant pollutants, with a focus on endocrine disrupting compounds (UK and HUJ) and antibiotics (HUJ); (b) optimization of methods for long-term preservation of the reporter bacteria, either by direct deposition on solid surfaces (HUJ) or by the construction of spore-forming Bacillus-based sensors (UK); (c) partial development of a computerized algorithm for the analysis of sensor panel responses. Implications: The sensor panel developed in the course of the project was shown to be applicable for the detection of a broad range of antibiotics and EDCs. Following a suitable development phase, the panel will be ready for testing in an agricultural environment, as an innovative tool for assessing the environmental impacts of EDCs and pharmaceuticals. Furthermore, while the current study relates directly to issues of water quality and soil health, its implications are much broader, with potential uses is risk-based assessment related to the clinical, pharmaceutical, and chemical industries as well as to homeland security.
