Texto completoThe Achaemenid Empire, which dominated much of the Near East between 559 and 330 BCE, relied on the economic exploitation of the territories it controlled, organized through a sophisticated administrative network. The satrapy of Transeuphratia, located in the western part of the empire, played a important role within this system. In Idumea, a region in the southern Levant, a corpus of nearly 2,250 ostraca written in Imperial Aramaic was discovered. Although many lack precise archaeological context, having emerged on the antiquities market in the 1970s, comparisons with ostraca from sites such as Maresha, Arad, and Beersheba indicate that they belong to the same period and administrative system. Since the 1990s, these ostraca have been predominantly studied from a philological perspective. However, this research offers an innovative approach by utilizing a relational database to conduct in-depth quantitative and comparative analyses. These documents, primarily economic in nature, provide insight into the local administration, and the aim is to situate them within their historical context to reveal the mechanisms of local governance within the satrapy. The comparative methodology, which juxtaposes the Idumean corpus with similar discoveries from other regions of the empire, helps establish parallels between the administrative practices across different regions. One key corpus for comparison is the Persepolis Fortification Archives, discovered in the 1930s, which transformed the understanding of Achaemenid administration by providing raw economic sources. This model, in which each satrapy maintained its own archives, raises the question of whether a similar system existed in Transeuphratene, and particularly in Idumea. The ostraca under study appear to represent traces of this local administrative network on a smaller scale. These documents reveal a series of economic transactions and the management of local resources. Although the inscriptions are sometimes incomplete or elliptical, they allow for an understanding of the key features of local administrative practices. Using Henkelman's concept of the "institutional landscape," this study proposes a twofold analysis: a microscopic perspective that focuses on local details, and a macroscopic view that situates these details within the broader framework of the empire. One of the primary objectives of this study is to identify the main themes present in the Idumean ostraca and to pinpoint the key actors in the administrative system, such as administrators, scribes, and tax collectors. This approach enables a better understanding of how the central Persian authority interacted with its peripheral provinces, adapting its practices to local specificities. By analyzing the Idumean ostraca and comparing them with other contemporary sources, this study seeks to reconstruct an overview of the regional administrative system. The Idumean ostraca corpus provides a unique opportunity to explore the management of the empire's peripheral territories. Through the study of recurring administrative terms and documented transactions, it is possible to gain a deeper understanding of the imperial administrative structure and the administrative practices in this region

Texto completoMucopolysaccharidosis type I (MPS I) is an autosomal recessive disease caused by mutations in the α-L-iduronidase (IDUA) gene, resulting in lack of the lysosomal enzyme IDUA and consequent accumulation of glycosaminoglycans (GAGs). There are more than 200 disease-causing mutations reported and the mutational profile varies considerably from country to country. In this study, we conducted molecular analysis of 92 DNA samples, representing the largest cohort of Brazilian MPS I patients examined. Additionally, we analysed allelic frequencies from other countries to better understand the composition of our population and the allele distribution worldwide. Forty-two mutations were identified in our sample and 14 of them are newly described: c.78delC, c.159-1_-23del, p.Gln125*, p.Trp175*, c.590-6ins4G, c.763delC, c.973-1G>A, p.Asp349Glu, p.Asn350Lys, p.Lys384Asn, c.1403-12_-4delCAGGCCCCG and c.1522_1525delGAGG. In silico analysis of novel mutations suggests they are possibly pathogenic, supporting previous biochemical and clinical diagnosis. Among recurrent mutations, p.Trp402* and p.Pro533Arg are the most frequent in Brazil, with 38% and 17% of alleles, respectively. The first is also the major allele in Australian patients and in some European and American countries, while the second is found in the Mediterranean region, mainly North Africa. The second most frequent allele worldwide and highly prevalent in Scandinavia, p.Gln70*, was relatively rare in our sample, with only 3% of alleles. These results reveal the great allelic heterogeneity of IDUA not only in Brazilian patients, which differs from all countries analysed, but also between geographically close populations. The knowledge of the genetic background of MPS I may improve diagnosis and management of patients and families with the disease.

Texto completoMucopolysaccharidosis type I (MPS I) is a lysosomal storage disease that leads to neurodegeneration and neurological deficits, among other pathological and clinical consequences. The absence of behavioral parameters from the MPS I mouse model motivated the accomplishment of this work. Our first objective was to analyze behavioral parameters that might be used to essay the effect of different therapies on the CNS. During exploration of an open field, adult MPS I (Idua-/-) mice showed normal locomotion and anxiety, but reduced number of rearings. Idua-/- mice performed normally in a novel object recognition memory task and showed normal short-term retention of inhibitory avoidance training. By contrast, long-term retention of inhibitory avoidance was impaired. The deficit in inhibitory avoidance memory could not be attributed to reduced footshock reactivity. The results indicate that Idua-/- mice present deficits in long-term memory for aversive training and reduced exploratory behavior. The second aim was to evaluate oxidative biomarkers on this mice model. We evaluated antioxidant enzymatic activity, protein damage and lipid peroxidation on forebrain, cerebellum, heart, lung, diaphragm, liver, kidney and spleen. From eight analyzed organs, seven had alteration on at least one parameter. The results indicate that Idua-/- mice can be under oxidative stress.

A theoretical model for evaluating the compaction grouting induced soil strength improvement has been developed based on the initial void ratio, the compression index, and the material parameter (Eui/(1+ν)Sui) of the soil before grouting. The soil strength improved around the compaction grout bulb increases with the decreasing distance, the increasing void ratio, the decreasing compressinon index, as well as the decreasing material parameter. This model has been satisfactorily verified with the laboratory test sata obtained in this study and from other independent investigation.

The primary goal of the study was to investigate the molecular mechanisms involved in the drug resistance of non-small cell lung cancer cell (NSCLC) to cisplatin, by focusing on the inhibition of parthanatos by iduna and the consequences. H460 cells are known to have higher resistance to cisplatin-induced cell death, an effective therapeutics commonly used to treat NSCLC. Parthanatos is a caspase- independent programmed cell death (PCD), relies on the over-activation of PARP-1 (poly(ADP) ribose polymerase-1), synthesis and nuclear efflux of PAR (poly (ADP) ribose) and migration of AIF (apoptosis-inducing factor) from mitochondria to the nucleus to cause large scale (≥ 50 kbp) of DNA fragmentation. Iduna, on the other hand, has recently been found can inhibit parthanatos to protect neurons from ischemic injury. Its role in cancer cells was unclear until now. The experimental strategies of the study were to know could parthanatos be inhibited by iduna in H460 cells that consequently increase cells'' resistance to cisplatin-induced cell death. The potential of the water extract of Graptopetalum paraguayense (WGP) and one of its major components, gallic acid (GA), in inhibiting of iduna, thereby, to increase parthanatos and break the cisplatin resistance of H460 cells were also investigated for the first time in cancer cells. Techniques applied in the study included, MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5 -diphenyltetrazolium bromide) assay, PARP-1 activity assay, Western blotting, DNA fragmentation assay, co-immunoprecipitation assay, RAPsylation assay, ubiquitination assay, mitochondrial permeability assay, shRNA knockdown assay, and chemical blocking assays, using inhibitors such as AG14361, N-phenylmaleimide, MG132. The results showed that compared to A549 and CH27 cell lines, H460 cells revealed the highest resistance to cisplatin-induced cell death that was likely due to the parthanatos inhibition of H460 cells. Cisplatin-induced PARP-1 activation and PARsylation of cytosolic proteins were not inhibited in H460 cells whereas cisplatin-induced mitochondrial permeability, nuclear accumulation of AIF and the large scale of DNA fragmentation were less severe in H460 cells compared with two other cell lines. Chemical inhibition of PARP-1 and AIF evidently blocked the cisplatin-induced parthanatos of A549 and CH27 but not H460 cells further demonstrated that cisplatin-induced parthanatos was inhibited in H460 cells. Cisplatin-inhibited iduna expression was also seen in A549 and CH27 but not H460 cells, indicating iduna may play a part in cisplatin resistance of H460 cells. The results further showed that iduna can prevent the cisplatin-induced mitochondrial permeability, nuclear accumulation of AIF, DNA fragmentation and cell death of H460 cells that explained the increase of cisplatin resistance of H460 cells. Both iduna and AIF were PARsylated in cisplatin-treated H460 cells to promote not only the binding between iduna and AIF but also the ubiquitination and proteasomal degradation of AIF that eventually reduced the amount of nuclear AIF and the occurrence of parthanatos. The study also demonstrated that WGP and GA can inhibit the protein expression and functions of iduna to restore (elevate) the cisplatin-induced parthanatos of H460 cells. In overall, the study has brought the first hand information showing that degree of parthanatos played a critical role in determining the drug (cisplatin) resistance of NSCLC (H460) cells. For the first time, iduna-mediated inhibition of parthanatos has been linked to the drug (cisplatin) resistance of cancer cells (H460), suggesting that iduna may be a potential target in NSCLC therapy. WGP and GA were newly demonstrated as iduna inhibitors and able to break the cisplatin resistance of H460 cells, indicating both drugs were therapeutic useful in NSCLC therapy.
