GreA is a well-characterized transcriptional factor that acts primarily by rescuing stalled RNA polymerase complexes, but has also been shown to be the major transcriptional fidelity and proofreading factor, while it inhibits DNA break repair. Regulation of greA gene expression itself is still not well understood. So far, it has been shown that its expression is driven by two overlapping promoters and that greA leader encodes a small RNA (GraL) that is acting in trans on nudE mRNA. It has been also shown that GreA autoinhibits its own expression in vivo. Here, we decided to investigate the inner workings of this autoregulatory loop. Transcriptional fusions with lacZ reporter carrying different modifications (made both to the greA promoter and leader regions) were made to pinpoint the sequences responsible for this autoregulation, while GraL levels were also monitored. Our data indicate that GreA mediated regulation of its own gene expression is dependent on GraL acting in cis (a rare example of dual-action sRNA), rather than on the promoter region. However, a yet unidentified, additional factor seems to participate in this regulation as well. Overall, the GreA/GraL regulatory loop seems to have unique but hard to classify properties.

Resumen
ABSTRACT Transcription elongation factor GreA induces nucleolytic activity of bacterial RNA polymerase (RNAP). In vitro, transcript cleavage by GreA contributes to transcription efficiency by (i) suppressing pauses and arrests, (ii) stimulating RNAP promoter escape, and (iii) enhancing transcription fidelity. However, it is unclear which of these functions is (are) most relevant in vivo. By comparing global gene expression profiles of Escherichia coli strains lacking Gre factors and strains expressing either the wild type (wt) or a functionally inactive GreA mutant, we identified genes that are potential targets of GreA action. Data analysis revealed that in the presence of chromosomally expressed GreA, 19 genes are upregulated; an additional 105 genes are activated upon overexpression of the wt but not the mutant GreA. Primer extension reactions with selected transcription units confirmed the gene array data. The most prominent stimulatory effect (threefold to about sixfold) of GreA was observed for genes of ribosomal protein operons and the tna operon, suggesting that transcript cleavage by GreA contributes to optimal expression levels of these genes in vivo. In vitro transcription assays indicated that the stimulatory effect of GreA upon the transcription of these genes is mostly due to increased RNAP recycling due to facilitated promoter escape. We propose that transcript cleavage during early stages of initiation is thus the main in vivo function of GreA. Surprisingly, the presence of the wt GreA also led to the decreased transcription of many genes. The mechanism of this effect is unknown and may be indirect.

Small RNA are very important post-transcriptional regulators in both, bacteria and eukaryotes. One of such sRNA is GraL, encoded in the greA leader region and conserved among enteric bacteria. Here, we conducted a bioinformatics search for GraL’s targets in trans and validated our findings in vivo by constructing fusions of probable targets with lacZ and measuring their activity when GraL was overexpressed. Only one target's activity (nudE) decreased under those conditions and was thus selected for further analysis. In the absence of GraL and greA, the nudE::lacZ fusion's β-galactosidase activity was increased. However, a similar effect was also visible in the strain deleted only for greA. Furthermore, overproduction of GreA alone increased the nudE::lacZ fusion’s activity as well. This suggests existence of complex regulatory loop-like interactions between GreA, GraL and nudE mRNA. To further dissect this relationship, we performed in vitro EMSA experiments employing GraL and nudE mRNA. However, stable GraL-nudE complexes were not detected, even though the detectable amount of unbound GraL decreased as increasing amounts of nudE mRNA were added. Interestingly, GraL is being bound by Hfq, but nudE easily displaces it. We also conducted a search for genes that are synthetic lethal when deleted along with GraL. This revealed 40 genes that are rendered essential by GraL deletion, however, they are involved in many different cellular processes and no clear correlation was found. The obtained data suggest that GraL's mechanism of action is non-canonical, unique and requires further research.

The coliphage HK022 protein Nun transcription elongation arrest factor inhibits RNA polymerase translocation. In vivo, Nun acts specifically to block transcription of the coliphage λ chromosome. Using in vitro assays, we demonstrate that Nun cross-links RNA in an RNA:DNA hybrid within a ternary elongation complex (TEC). Both the 5′ and the 3′ ends of the RNA cross-link Nun, implying that Nun contacts RNA polymerase both at the upstream edge of the RNA:DNA hybrid and in the vicinity of the catalytic center. This finding suggests that Nun may inhibit translocation by more than one mechanism. Transcription elongation factor GreA efficiently blocked Nun cross-linking to the 3′ end of the transcript, whereas the highly homologous GreB factor did not. Surprisingly, both factors strongly suppressed Nun cross-linking to the 5′ end of the RNA, suggesting that GreA and GreB can enter the RNA exit channel as well as the secondary channel, where they are known to bind. These findings extend the known action mechanism for these ubiquitous cellular factors.

My thesis analyses the phenomenon of Figwit, a non-speaking elf extra who appeared for only three-seconds in the first instalment of Peter Jackson's 2001 Lord of the Rings movie trilogy. Figwit was initially generated as an online parody by female fans of the movie and as a foil to the 'swooning, drooling girly' fandom that was being directed towards the movie's star actors. However, Figwit evolved into a bona fide, albeit minor, celebrity both on and offline as he attracted attention from worldwide media, a small speaking role in the final movie and genuine adulatory fandom as manifested in the production of Figwit merchandise. In my thesis I argue that Figwit's creation and consequential community formation reflects a dynamic online-offline dialogic in which pre-existing offline and habitus-generated social practices and distinctions, ideal reflexive individuality and celebrity/fandom were dynamically reproduced within online technological frameworks. I also argue that online activity and interactivity is generated by users to strategically express and engage intensified reflexive individuality, affirming sociability and hyper-social distinctions. In this regard I have also argued that these various potentials and imaginaries were significantly enabled by digital architectures and genres of online communication and interactivity. In particular, I discuss the internet's capacity for searchability, traceability, and rhetorical framing processes that facilitate continuous re-editing authorship possibilities, which are not necessarily replicable in face-to-face interactions. Finally, I argue that reflexive online interactivity and identity expressions may transform into online and offline consequences that may be constructive, divergent or even contradictory. The arguments put forward in this thesis are based on a multi-sited ethnography, which utilises a variety of methodologies including participant-observation, subject interviews, communications and media archiving and analysis, and it draws from a variety of sources both online and offline.

This list contains 27 rivers that flow into the Great Lakes, most of which are or have been areas of industrial and chemical pollution. Through installations involving photography, ceramics, and video, I hope to reveal the issues facing the Great Lakes and explore how people can help.

This thesis concerns Aristobulus of Cassandrea, a historian of Alexander the Great, who followed the Macedonian king during his expedition in Asia and wrote a book about these events. This work is lost. At the beginning of this study there is the corpus of Aristobulus’ fragments edited by Felix Jacoby in his Die Fragmente der griechischen Historiker (Leiden 1927). The few biographical information about the historian has been collected and discussed in the first part of our work, where we analyze the 6 testimonia about Aristobulus’ life. Then, we discuss the 62 fragments from the work concerning Alexander the Great, and the 2 spurious, which are not Aristobulus’. Of each fragment we present: an Italian translation, an analysis of major textual problems and a commentary, in which the Aristobulus’ text is compared with the most important ancient sources on Alexander. In the final chapter, we develop an analysis of all the fragments, in order to reconstruct the characteristics of the work of the historian.
Il lavoro di tesi si propone di analizzare le testimonianze e i frammenti di Aristobulo di Cassandrea, storico di Alessandro Magno, che accompagnò il sovrano macedone nella spedizione in Asia e che scrisse un’opera su questi avvenimenti, andata perduta. Punto di partenza per questo studio è il corpus dei frammenti di Aristobulo edito da Felix Jacoby nella grande raccolta Die Fragmente der griechischen Historiker (Leiden 1927). Nella prima parte del lavoro sono state raccolte e discusse, sulla base delle testimonianze, le poche notizie biografiche sullo storico. Si è poi proceduto all’analisi dei 62 frammenti tratti dall’opera su Alessandro Magno e dei 2 spuri, che sono risultati non pertinenti allo storico in esame. Di ogni frammento sono stati affrontati i principali problemi testuali, ove presenti, ed è stata fornita una traduzione in italiano e un commento, nel quale il testo di Aristobulo è stato messo a confronto con quello delle principali fonti antiche su Alessandro. Nelle conclusioni, si presenta un’analisi complessiva dei frammenti di Aristobulo, al fine di ricostruire le caratteristiche peculiari dell’opera dello storico.

This paper presents my master project “Grey Flowers” which is an incarnation of a poetic metaphor to a ceramic flower sculpture. The project is an installation of crafted flowers as a strong symbol of a short lived moment which has now stopped for ages. This sculptural work is based on the notion of decadence of the 19th century and presents a new way which it can been interpreted. Ceramics, clay and firing processes are used as a methods of unpredictability and a “happy accident”. The main aim of the project is to create a feeling of abandonment by showing decaying and dying beauty. The sculptural work investigates the different aspects of the flower and its representation trying to show different approaches tothinking and working while telling about the same idea.

The poems are tied together through similar images or ideas in order to mimic the way the mind works as it jumps from thought to thought. GREY SLATE also mixes different types of poems: from haiku to sonnet to paradelle, and from lyric to narrative to prose poem. GREY SLATE hopes to inspire readers to take a break from searching for truths and indulge in the beautiful mystery that is life with no need for answers.

Background: Corporate Responsibility is not a new concept however in recent times more companies are starting to use different kinds of responsibility disclosures. These disclosures are voluntary and corporations may use them in the way they find suits them best. There is a lot of research in this field and there is no single explanation for what Corporate Responsibility is and how corporations should relate to it. Purpose: The purpose of the study is to examine the relationship between the controlling owners of corporations and CSR disclosure. Method: We use a content analysis and a survey study to examine the relationship between corporate controlling owners and the CSR disclosure. Conclusions: Our results show that controlling owners have little effect on the different dimensions of CSR disclosure. We have however found some differences between the different owner categories where some categories are found to use CSR disclosure to a greater extent.

Resumen
Abstract. As part of the quality of products, corrective actions are necessary. In this context, organizations should use different methods to skillfully analyze product incompatibilities and their causes. The aim of a study is to propose the method for determining the degree of impact of causes on the occurrence of incompatibilities. This method is a new combination of Grey Relational Analysis (GRA) with known methods: brainstorm, causes and effects diagram, and important technique. The method was tested for an example of a cast alloy product.

Resumen
All structures in Switzerland - that is, all buildings, roads, infrastructure constructions and so on - consume over their entire life cycle around 50 % of Switzerland's final energy requirement. They are also responsible for around 30 % of emissions of the greenhouse gas CO2. In recent decades, the energy requirements and CO2 emissions resulting from the use of such structures have fallen sharply. However, the grey energy contained within the structures as well as the CO2 emissions associated with the construction, renovation and demolition of buildings, remain high. There is great potential for improvement here. The joint project “Low energy concrete” provides an important basis for transforming the construction industry into a sustainable sector. It primarily focuses on the building material concrete, which is responsible for an especially high amount of grey energy and significant CO2 emissions. The results of this joint project are summarised and interpreted in this synthesis on “Sustainable Concrete Structures”. The chief objectives of the joint project were as follows: CO2 emissions and grey energy are reduced by drastically decreasing the amount of clinker in the cement. Grey energy is reduced by replacing reinforcing and prestressing steel in concrete structures with wood and plastic. The service life of the structures is extended by professional monitoring and adequate renovation measures; this reduces the average annual grey energy and CO2 emissions. The research work shows that the CO2 emissions caused by concrete and concrete structures can be reduced by a factor of 4, while the bound grey energy can be decreased by a factor of 3.
