Abstract The cytosine (C)-rich sequences that can fold into tetraplex structures known as i-motif are prevalent in genomic DNA. Recent studies of i-motif–forming sequences have shown increasing evidence of their roles in gene regulation. However, most of these studies have been performed in short single-stranded oligonucleotides, far from the intracellular environment. In cells, i-motif–forming sequences are flanked by DNA duplexes and packed in the genome. Therefore, exploring the conformational dynamics and kinetics of i-motif under such topologically constrained environments is highly relevant in predicting their biological roles. Using single-molecule fluorescence analysis of self-assembled DNA duplexes and nanocircles, we show that the topological environments play a key role on i-motif stability and dynamics. While the human telomere sequence (C3TAA)3C3 assumes i-motif structure at pH 5.5 regardless of topological constraint, it undergoes conformational dynamics among unfolded, partially folded and fully folded states at pH 6.5. The lifetimes of i-motif and the partially folded state at pH 6.5 were determined to be 6 ± 2 and 31 ± 11 s, respectively. Consistent with the partially folded state observed in fluorescence analysis, interrogation of current versus time traces obtained from nanopore analysis at pH 6.5 shows long-lived shallow blockades with a mean lifetime of 25 ± 6 s. Such lifetimes are sufficient for the i-motif and partially folded states to interact with proteins to modulate cellular processes.

Resumen
Objetivo: elaborar e implementar um material educativo que possuiu como finalidade orientar os diabéticos sobre os cuidados na prevenção do COVID-19 em um centro especializado em diabetes do Distrito Federal . Materiais e método:Trata-se de uma construção dialógica na qual foi elaborado um folder com orientações quanto ao covid-19, o motivo da necessidade do controle glicêmico e cuidados de higiene para indivíduos diabéticos diante da pandemia. Resultados e discussão: Foi elaborado, impresso e entregue os folders aos usuários diabéticos em um centro especializado em diabetes no Distrito Federal com orientações quanto ao covid-19 e gestão do autocuidado para prevenção da doença. Outros estudos demonstram a importância e influência da utilização das tecnologias em saúde no manejo do autocuidado e de doenças como a Diabetes Mellitus. Conclusões: A utilização das tecnologias em saúde tem sido uma ferramenta manejada pela equipe de enfermagem na promoção de saúde e do autocuidado de usuários dos serviços de saúde, assim como foi utilizada em um centro especializado de diabetes no Distrito Federal para orientação do auto manejo e prevenção da covid-19. Espera-se que folder seja utilizado nas unidades de saúde, na versão digital ou impressa, para expandir a informação e orientar os indivíduos.

The proton-coupled folate transporter (PCFT) mediates intestinal folate absorption, and loss-of-function mutations in this gene result in the autosomal recessive disorder hereditary folate malabsorption. The current study, focused on a structure-functional analysis of this transporter, identified Gly-189 and Gly-192 (a GxxG motif) located in the fifth transmembrane domain as residues that could not be replaced with alanine without a loss of function. In contrast, function was preserved when Gly-56 and Gly-59 (the other conservative GXXG motif in human PCFT) were replaced with alanine. Similarly, Gly-93 and Gly-97, which constitute the only conserved GXXXG dimerization motif in human PCFT, tolerated alanine substitution. To explore the role of this region in folate binding, the residues around Gly-189 and Gly-192 were analyzed by the substituted cysteine accessibility method. Both I188C and M193C mutants were functional and were inhibited by membrane-impermeable sulfhydryl-reactive reagents; this could be prevented with PCFT substrate, but the protection was sustained at 0°C only for the I188C mutant, consistent with localization of Ile-188 in the PCFT folate binding pocket. The functional role of residues around Gly-189 and Gly-192 is consistent with a molecular structural model in which these two residues along with Ieu-188 are accessible to the PCFT aqueous translocation pathway.

The Dictyostelium Gα5 subunit has been shown to reduce cell viability, inhibit folate chemotaxis and accelerate tip morphogenesis and gene expression during multicellular development. Alteration of the D-motif (mitogen-activated protein kinase docking site) at the amino terminus of the Gα5 subunit or the loss of extracellular signal-regulated kinase (ERK)1 diminished the lethality associated with the overexpression or constitutive activation of the Gα5 subunit. The amino-terminal D-motif of the Gα5 subunit was also found to be necessary for the reduced cell size, small aggregate formation and precocious developmental gene expression associated with Gα5 subunit overexpression. This D-motif also contributed to the aggregation delay in cells expressing a constitutively active Gα5 subunit, but the D-motif was not necessary for the inhibition of folate chemotaxis. These results suggest that the amino-terminal D-motif is required for some but not all phenotypes associated with elevated Gα5 subunit functions during growth and development and that ERK1 can function in Gα5 subunit-mediated signal transduction.

Haesevoets, Reinders Folmer, and Van Hiel (2015) have shown limited consistency of people's behaviour across various mixed–motive games. According to these authors, the modest relationships among these games call into question the general idea that all mixed–motive games render the conflict between selfish interests and concern for others equally salient. Thielmann, Böhm, and Hilbig (2015), however, argued that these findings can be explained in terms of the motivational differences that underlie the games. In this article, we demonstrate that Thielmann et al.'s descriptive model of the different motives underlying selfish and prosocial choices cannot be straightforwardly applied to the empirical data at hand. Analogous to our previous article, we again stress the need for further empirical research investigating the underlying motivational basis of each mixed–motive game. Ausgehend von einigen Beispielen der Moralisierung der Politik, wird zunächst der Begriff des politischen Handelns erläutert und werden wichtigste operative Mittel der Politik vorgestellt. Sodann werden in einer längerfristigen Sicht abendländischer Entwicklungen die Entdeckung der Interessen und die Legitimitätsproblematik politischer Herrschaft behandelt und auf die wichtige Unterscheidung zwischen Wert- und Identitätskonflikten einerseits und Interessen- und Verteilungskonflikten andererseits hingewiesen. Unsere zentrale These lautet, dass der Vorrang von Wert- und Identitätskonflikten vor Interessen- und Verteilungskonflikten mit ausgeprägten und für die Stabilität demokratischer Herrschaftssysteme problematischen Moralisierungs- und Emotionalisierungsneigungen einhergeht. Im Anschluss daran wird zwischen drei verschiedenen Erscheinungsformen der Moralisierung der Politik unterschieden und die Problematik »gesinnungsethischen« politischem Handeln erörtert.

Le principe de la thérapie génique repose sur l'apport du gène, qui est défectueux ou déficitaire, dans les cellules du patient. Dans la première partie, nous présentons une étude bibliographique sur les lipides membranaires d'archaebactéries présentant des propriétés favorables à la vectorisation d'acides nucléiques. Afin d'envisager un ciblage passif ou actif des cellules visées, nous décrivons l'intérêt de la présence de chaînes polyéthylène glycol et de ligand de reconnaissance greffés sur la partie lipidique. Dans la deuxième partie est décrite la synthèse d'une famille de vecteurs atypiques, reposant sur un squelette lipophile pégylé et à motif acide folique analogue aux lipides membranaires d'archaebactéries. La troisième partie présente la préparation, la caractérisation et les études de transfection des complexes lipide / ADN incorporant les vecteurs préparés comme co-lipide
The aim of gene therapy is to bring the defective or overdraw gene in the patient's cells which recover at that time their fully functionnality. In the first part, we present a bibliographic update on archaebacteria membrane lipids exhibiting their particular interest in vectorisation of drugs and DNA. In order to consider a passive or active cells targeting, we discuss the interest of the presence of polyethylene glycol chains and a ligand of recognition grafted at the end of the lipidic structure. In the second part, the synthesis of an atypical vectors family is described consisting on folate bioconjugates derived from archaebacteria membrane lipid analogues. The third part presents the preparation, the characterisation and the efficiency of transfection of the lipid / DNA complexes incorporating such vectors prepared as co-lipids

ZusammenfassungZiel dieser Arbeit war es, Folgen der Digitalisierung für die politische Protestpartizipation am Beispiel der Umweltschutz-Bewegung zu diskutieren und individuelle Voraussetzungen und Motive von Bürger*innen für eine Partizipation herauszuarbeiten. Dabei lag der Fokus insb. auf Ressourcen, Emotionen und Affekten, den konkreten Online- und Offline-Praktiken und den persönlichen Einstellungen der Individuen zu Straßenprotest, Online-Aktivismus und der Kritik des Clicktivism, sowie zum Verständnis von Bürgerschaft. Durch die Zusammensetzung des Samples mit älteren und jüngeren Interview-Partner*innen konnte untersucht werden, wie sich einerseits die konkreten Protestpraktiken einzelner Bürger*innen über die Jahrzehnte verändert haben, die früher nur Offline-Möglichkeiten zur Verfügung hatten und nun sowohl online als auch offline agieren können, und andererseits Digital Natives auf die Möglichkeit reagieren, sich sowohl im Netz als auch auf der Straße bzw. in Organisationen einbringen zu können. Die Erkenntnisse aus der Analyse des Interviewmaterials decken sich mit vielen in der bisherigen Literatur beschriebenen Forschungsergebnissen. Die vorliegende Arbeit kann jedoch auch relevante und notwendige Ergänzungen zum Thema beitragen und schlägt Überarbeitungen früherer Konzepte mit Blick auf die Auswirkungen der Digitalisierung vor. In der bisherigen Forschung nicht ausreichend berücksichtigt wurde z. B. das Thema Frustrationstoleranz.

Der Beitrag rekonstruiert am Beispiel des im Februar 1878 gegründeten Wissenschaftlichen Vereins für neuere Sprachen zu Leipzig – dem achten seiner Art, der sich nur wenig später den Namen Akademischer Verein für neuere Philologie zulegen sollte – zunächst die Motive der Gründung, die Rolle der Hochschullehrer, das Vereinsleben sowie die weitere Entwicklung dieser neuphilologischen Studentenvereine im späten 19. und frühen 20. Jahrhundert. Anschließend wird nach der Bedeutung gefragt, die den Vereinen bei der Konstruktion des idealtypischen ‚Neuphilologen‘ sowie in der Diskussion um die Reform von Studium und Berufspraxis des neusprachlichen Lehramts bis 1914 zugeschrieben werden kann. Die Geschichte der akademisch-neuphilologischen Studentenvereine, die sich über die Zeit des preußisch-deutschen Kaiserreichs erstreckt, kann grundsätzlich in drei Phasen unterschieden werden. Auf die Gründungsphase von 1874 bis in die Mitte der 1880er Jahre, folgte ein Jahrzehnt des relativen Stillstands. Diesem schloss sich eine etwa zwanzig Jahre währende Phase der (Re-)Konsolidierung und Transformation an, die mit dem Beginn des Ersten Weltkrieges im August 1914 ein jähes Ende fand. Abstract In the summer of 1989, one of the glossiest men’s magazines appeared with a picture of Sean Connery on the cover. Dressed in a creamy ivory tuxedo, standing with arms folded against a beige background, visible only to the waist, he embodied elegance, sensuality, and virility. The caption proclaimed him “The Last Real Man in America.” Why should the Last Real Man in America be a British actor? The taunting ambiguity implies that no American can claim true manliness anymore; to see it at all is to see it vanishing. Only a working-class Scotsman who secured his international image as James Bond, an Englishman equally skilled in civility and violence, can temporarily import virility to American shores. Yet the pleasurable visual framing of his face as white on white on beige seems curiously reassuring. Connery represents the last, best hope, the master of everything not quite seen, dark, and below the belt. He can protect a civilized, yet effete Us from a barbaric though enviably violent Them. The July cover of Gentleman’s Quarterly slipped from my mind until late August, when I was standing in a checkout line at a Florida supermarket and noticed the September cover of Celebrity Plus, a decidedly downscale fan magazine. This cover featured Harrison Ford in a woodsy setting, with a yellow blurb announcing, “Rare Interview! The ‘Last’ Real Man.”‘ Faced with such a flagrant intertextual rip-off, I began wondering: in what other country could variations on that come-on sell magazines? To ask the question exposes the strange mixture of bravado, anxiety, and nostalgia in the motif. Male readers in more traditionally patriarchal cultures might well feel insulted. Why did I feel a bit wistful, even teased? The motif has a beleaguered quality to it, as if urban, yuppie, corporate, feminist America had become intrinsically emasculating.

Resumen
La palma de aceite es uno de los cultivos más importantes en Colombia. Tiene un área sembrada de alrededor de 540.000 ha, de las cuales 87.000 ha corresponden a cultivares del híbrido interespecífico OxG. Este cruzamiento genético les confiere características de alta productividad y otras no tan deseadas, que fueron identifica das por múltiples investigaciones. En vivero se han descubierto anomalías asociadas al genotipo, tales como palmas enanas, rizadas, retorcidas o cloróticas, y palmas que presentan retraso en su desarrollo, quimeras, gigantes y erectas. La identificación de estas palmas anormales en vivero permite la selección de plantas de calidad para su posterior siembra en el sitio definitivo. Sin embargo, en el campo se han detectado otras anomalías como el disturbio de amarillamiento, el cual no corresponde a deficiencias nutricionales y está asociado a algunas palmas particulares. Otras anomalías relacionadas con el manejo agronómico son la desaparición de la lámina foliar (DLF) asociada a un desbalance nutricional en la relación Ca/B y al malogro de racimos, que implica un secamiento de racimos antes de llegar a su punto óptimo de cosecha. Esta anomalía tiene relación con procesos inadecua dos de polinización asistida y con otras variables que son motivo de investigación. Actualmente se continúa en la búsqueda de problemáticas que ameriten nuevas investigaciones en cultivares del híbrido interespecífico OxG.

Resumen
During daily studying and working, people have to research massive amounts of information on the internet and download numerous files. Some of these files can be easily categorized to relevant files. However, there are always some files left unorganized due to their difficulty in categorization [1]. Such files pile up in the download folder as time passes, making the folder extremely messy. Many people do not have the motive to clean up the folder as it requires a lot of energy and time. Based on this common problem, my group developed an app that can clean the messy folder up. After applying our program which is based on machine learning, the files will firstly be divided into five general parts – document, video, music, photos and package. Then the files will be further categorized based on the contents they present. For example, photos are divided into animals, families and so on. In order to achieve the content-categorizing function, several powerful apis were introduced in our program, and they helped us to achieve the optimal results.

Resumen
Das Fahren ohne (gültigen) Fahrschein im ÖPNV ist bereits seit den 1960er Jahren ein Problem zahlreicher Verkehrsunternehmen. Auch für die ÖPNV-Nutzenden stellt es ein Problem dar, da Personen, die den ÖPNV ohne (gültiges) Ticket nutzen in Deutschland eine Straftat begehen. In der Wissenschaft wurde das Thema aus unterschiedlichen Perspektiven heraus untersucht (v.a. Rechtswissenschaften, Betriebswirtschaften, Kriminologie sowie einige sozialwissenschaftliche Ansätze), jedoch konzentriert sich die Forschung vorrangig auf sozio-demographische Charakteristika, Marktsegmentierungen und die Folgen des Fahrens ohne (gültigen) Fahrschein für Verkehrsunternehmen und -verbünde. Die Motive und Beweggründe des Fahrens ohne (gültigen) Fahrschein werden in den vorhandenen Studien lediglich objektiv betrachtet. Das Arbeitspapier zeigt die Ergebnisse einer Untersuchung der Motive des Fahrens ohne (gültigen) Fahrschein im Bediengebiet des Rhein-Main-Verkehrsverbundes (RMV), die mithilfe von qualitativen Interviews mit Personen, die bei einer Fahrkartenkontrolle kein (gültiges) Ticket vorzeigen konnten, exploriert wurden.
