Texto completoThis work considers variable aspects on the implication of fatty acids for daphnids. Fatty acids from seston, algae, and daphnids were analysed by gas chromatography for investigations in respect to nutrition, physiology, and reproduction. Experiments with daphnids were carried out in batch and flow-through cultures. Due to their heterogenic composition, lake seston contained always a vast number of omega-6- and omega-3-fatty acids. Despite great differences in phytoplankton composition of Bautzen Reservoir and Lake Großer Väter, lake specific patterns of sestonic fatty acid composition were not detected. The fatty acid composition of daphnids is strongly influenced by their food. Nevertheless, nutritional DHA was transformed completely to shorter chained omega-3-fatty acids. This transformation is possibly an adaption against fish predation. The growth of juvenile daphnids was limited by food quality in Lake Großer Väter 1998. Neither fatty acids nor another known factor of food quality were responsible for this limitation. According to the results it seems questionable that alpha-linolenic acid and/or EPA are essential for daphnids. Further, it was demonstrated that food concentration and temperature determine clutch size in daphnids. In Lake Großer Väter the seasonal changes of specific fatty acid concentration [µg per individual] of adult daphnids could be explained by changes in the duration of egg development time which is determined by temperature. Large clutches were found nearly exclusively at low temperatures.

Texto completoCutaneous mast cells are considered as key immune effector cells in the pathogenesis of canine atopic dermatitis (CAD; OLIVRY et al. 1997). These cells release immediate-phase and late-phase mediators of inflammation. Dietary fatty acids are incorporated in cellular membranes and seem to influence mediator production and release. A dietary intervention with n6- and n3-fatty acids with a ratio from 5 to 1 alleviated clinical symptoms in 40% of atopic dogs (SCOTT et al. 1997). The purpose of this study was to examine the effects of n6- and n3-fatty acids on the fatty acid composition and the production of prostaglandins in canine mastocytoma cells (C2) as a possible model for CAD. Cells were cultured in a basic medium (DEH) or with additional 14 µM linoleic (C18:2n6, DEH-LA), gammalinolenic (C18:3n6, DEH-GLA), arachidonic (C20:4n6, DEH-AA), a-linolenic (C18:3n3, DEH-LnA), eicosapentaenoic (C20:5n3) or docosahexaenoic acid (C22:6n3, DEH-DHA). Cell growth was examined for 11 days in all media. The cells were harvested after 4 or 8 days, washed twice with phosphated buffered saline and dried under nitrogen for fatty acid analysis. The fatty acid composition was determined by gas chromatography after extraction and transesterification of the phospholipids using di-C17-phosphatidylcholin as internal standard. For measurment of prostaglandin (PG) D2 and E2 the C2 were stimulated with the wasp venom peptide mastoparan. PGD2 was measured by PGD2-methoxim-enzymimmunoassay (EIA) and PGE2 was determined by radioimmunoassay (RIA). Cell growth increased from day 1 to 8 and decreased thereafter in all media conditions. The supplied fatty acid did not influence the cell growth. Added fatty acids increased the concentration of these fatty acids in C2 (LA 4.9-fold, GLA 6.9-fold, AA 6-fold, LnA 9.3-fold, EPA 6.5-fold, DHA 8.4-fold). Futhermore elongated and D6-desaturated products of the corresponding fatty acids were significantly elevated, however D5-desaturated products were not measurable. An increased time dependent incorporation was only detectable for LA after culturing C2 in DEH-LA. The results let us assume that C2 has no activity of the D5-desaturase. If the assumed low activity of these desaturase is one of the mechanisms underlying the pathogenesis of CAD, C2 seems to be an adequate model for CAD. The production of PGE2 after stimulation with mastoparan was significantly reduced when C2 were cultured in DEH-LnA and DEH-DHA and was significantly increased when C2 were cultured in DEH-AA and DEH-EPA. The reason for the different PGE2-production in C2 after the treatment with the n3-fatty acids (LnA, EPA or DHA) being unsettled. The observed results suggest, that C2 could be used to investigate the mechanisms of production and release of prostaglandins or other mediators as a model to improve our understanding of the pathogenesis of canine or human atopic dermatitis

Texto completoThe n-3 polyunsaturated fatty acids (n-3 PUFAs) eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), protect from cardiovascular disease, especially from fetal arrhythmia. Moreover, in vitro studies proved that recombinant cytochrome P450 (CYP) enzymes not only accept the physiologically most important n-6 PUFA arachidonic acid (AA), but also EPA and DHA as alternative substrates, thereby generating regio- and stereospecific biologically active epoxy- and hydroxymetabolites, CYP-eicosanoids. These metabolites serve as second messengers regulating vascular, renal and cardiac function. The precise underlying molecular mechanisms are only partially understood and need further investigation. The first aim of the thesis was to show that the endogenous CYP-eicosanoid profile depends on the availability of the precursor fatty acids. The results of a clinical trial with 20 volunteers, show that n-3 PUFAs serve also in vivo as alternative CYP-dependent substrates and are even preferentially metabolized compared to AA. After EPA/DHA-supplementation 17,18 EEQ was generated as a major metabolite, potentially an important mediator of cardiovascular effects originally attributed to n-3 PUFAs. To test the anti-arrhythmic effect of EPA and 17,18-EEQ, an established cell model with neonatal rat cardiomyocytes (NRKMs) was used. The negative chronotropic effect of EPA was mimicked by 17,18-EEQ, attributed only to the R,S-enantiomer. A structure activity relationship study revealed synthetic analogs, exerting the same biological effect as 17,18-EEQ. Strict structural requirements were found for agonistic function, hinting at a specific interaction with cellular targets, like GPCRs. The search of a molecular target of CYP-eicosanoids led to a putative receptor, which was tested for ligand binding specificity. If the preliminary results on the ligand binding are confirmed in future experiments this receptor might be a novel target for the treatment of cardiac arrhythmia.

Texto completoThis study examines the metabolic incorporation of selected fatty acids into the lipids of porcine spermatozoa and evaluates the physiological state of spermatozoa subsequent to low temperature storage supplementation with selected free fatty acids. The aim was to understand the role of fatty acids in relation to the (cryo-)preservation of spermatozoa and successful reproduction in more detail. All lipids present in porcine spermatozoa were analysed using gas chromatography (GC) and mass spectrometry (MALDI-TOF-MS). The main representatives of the polar lipid classes are glycerophospholipids (in particular GPC and GPE). The main representatives of the neutral lipid classes are diacylglycerols (DAG). Metabolic incorporation of fatty acids into lipids was radiochemically monitored using [14C]-octadecadienoic acid in the supplied spermatozoa-preservation medium. Temperature and incubation time were shown to be particularly important determinants. The added fatty acids were incorporated into both the spermatozoas’ neutral (DAG) and polar lipids (diacyl-GPC). The affected lipids were characterised by means of MALDI- and Q-TOF-MS subsequent to the supplementation of uniformly 13C-labelled octadecadienoic acid. DAG (18:2/18:2), GPC (16:0/18:2) and GPC (18:2/18:2) could be identified and a de-novo biosynthesis of DAG (18:2/18:2) could be proven. The same results were obtained when spermatozoa were supplemented with hexadecenoic, octadecenoic and octadecatrienoic acids. Finally, it was shown that the physiological state of the spermatozoa, especially those supplemented with endogeneously present fatty acids, led to an enhanced vitality and motility in spermatozoa subsequent to low temperature storage. It was also observed that acrosomal damage was reduced and that hexadecenoic acid significantly stabilised all the vitality parameters. In conclusion, supplementing spermatozoa with selected fatty acids is an effective solution for the storage of spermatozoa at 4 to 6°C.
