Resumen
Human oral squamous cell carcinomas (OSCCs) have high cancer mortality and a 5-year survival rate lower than that of most other carcinomas. New therapeutic strategies are required for the treatment and prevention against OSCCs. An approach to cancer therapy using plant-derived natural compounds has been actively in progress as a trend. Falcarindiol (FALC), or its isolated form Ostericum koreanum Kitagawa (O. koreanum), is present in many food and dietary plants, especially in carrots, and this compound has a variety of beneficial effects. However, biological activity of FALC has not been reported in OSCCs yet. This study aimed to demonstrate the antitumor effects of FALC against OSCCs, YD-10B cells. In this study, FALC was selected as a result of screening for compounds isolated from various natural products in YD-10B cells. FALC suppressed cell growth, and FALC-induced apoptotic cell death was mainly accompanied by the dephosphorylation of PI3K, AKT, mTOR, and p70S6K. The apoptotic cell death was also associated with autophagy as evidenced by the expression of Beclin-1, the conversion of LC3-II, and the formation of autophagosome. FALC-induced autophagy was accompanied by MAPKs including ERK1/2 and p38. Furthermore, FALC caused the antimetastatic effects by inhibiting the migration and invasion of YD-10B cells. Taken together, the findings suggest the potential value of FALC as a novel candidate for therapeutic strategy against OSCCs.

Resumen
Fiber reinforced aerated lightweight concrete (FALC) was developed to reduce concrete's density and to improve its fire resistance, thermal conductivity, and energy absorption. Compression tests were performed to determine basic properties of FALC. The primary independent variables were the types and volume fraction of fibers, and the amount of air in the concrete. Polypropylene and carbon fibers were investigated at 0, 1, 2, 3, and 4% volume ratios. The lightweight aggregate used was made of expanded clay. A self-compaction agent was used to reduce the water-cement ratio and keep good workability. A surfactant was also added to introduce air into the concrete. This study provides basic information regarding the mechanical properties of FALC and compares FALC with fiber reinforced lightweight concrete. The properties investigated include the unit weight, uniaxial compressive strength, modulus of elasticity, and toughness index. Based on the properties, a stress-strain prediction model was proposed. It was demonstrated that the proposed model accurately predicts the stress-strain behavior of FALC.

Resumen
Abstract Innate immune responses are important in combating various microbes during the early phases of infection. NK cells are innate lymphocytes that, unlike T and B lymphocytes, do not express antigen receptors but rapidly exhibit cytotoxic activities against virus-infected cells and produce various cytokines. Here we report a new type of innate lymphocyte present in a novel lymphoid structure associated with adipose tissues in the peritoneal cavity. These cells do not express lineage (Lin) markers but do express c-Kit, Sca-1, IL7R and IL33R. Similar lymphoid clusters were found in both human and mouse mesentery and we term this tissue ‘FALC’ (fat-associated lymphoid cluster). FALC Lin-c-Kit+Sca-1+ cells are distinct from lymphoid progenitors and lymphoid tissue inducer cells. These cells proliferate in response to IL2 and produce large amounts of Th2 cytokines such as IL5, IL6 and IL13. IL5 and IL6 regulate B-cell antibody production and self-renewal of B1 cells. Indeed, FALC Lin-c-Kit+Sca-1+ cells support the self-renewal of B1 cells and enhance IgA production. After helminth infection and in response to IL33, FALC Lin-c-Kit+Sca-1+ cells produce large amounts of IL13, which leads to goblet cell hyperplasia—a critical step for helminth expulsion. In mice devoid of FALC Lin-c-Kit+Sca-1+ cells, such goblet cell hyperplasia was not induced. Thus, FALC Lin-c-Kit+Sca-1+ cells are Th2-type innate lymphocytes, and we propose that these cells be called ‘natural helper cells’.

Resumen
The plant Saussurea costus (Falc) Lipsch has many biological activities and a strong curative and preventive power against a variety of diseases including cancer, diabetes, and hemorrhoids. In the current study, phytochemical screening was carried out as well as an investigation of the antilithiatic and antioxidant activities of aqueous and ethanolic extracts of this plant. The results showed that aqueous and ethanolic extracts were effective in reducing cystine stone mass and that the aqueous extract of Saussurea costus (Falc) Lipsch had the highest percentage of dissolution (6.756 ± 1.024) (p < 0.05). A turbidimetric method and a crystallization test were used to evaluate the antilithiatic activity of an aqueous and ethanolic extract of this plant on calcium oxalate crystallization. The results of these methods revealed that the ethanolic extract of this plant has a significant inhibitory effect on calcium oxalate crystallization, with a percentage inhibition of (91.017 ± 0.299) (p < 0.05) for a concentration of 2 mg mL−1. The DPPH method revealed that the ethanolic extract of Saussurea costus (Falc) Lipsch with a concentration of (IC50 = 0.12325 mg mL−1) had the highest IC50, whereas the FRAP method revealed that the aqueous extract of Saussurea costus (Falc) Lipsch with a concentration of 300 µg mL−1 has the most significant reducing power with (OD = 0.56 ± 0.05). These findings indicate that aqueous and ethanolic extracts of Saussurea costus (Falc) Lipsch had a significant effect on whewellite and weddellite and a greater free radical scavenging effect but had no effect on cystine dissolution.

Resumen
Abstract Fibroblastic reticular cells (FRCs), a subpopulation of stromal cells in lymphoid organs and fat-associated lymphoid clusters (FALCs) in peritoneal fat, play immune-regulatory roles in the host defense. However, the mechanisms underlying the regulation of FRC pathobiology, especially in response to Toll-like receptor (TLR) signaling is unclear. Therefore, we aimed to understand the roles of TLR signaling in FRC pathobiology during sepsis. FALC cells were isolated from mesenteric adipose tissue in wild-type mice before and after cecal ligation and puncture (CLP). The transcriptomic profile of FALC cells were assessed using single cell RNA sequencing. Notably, FRCs were a major population of FALC cells (50 +/− 10.2 % in control, 30 +/− 8.6% in CLP). One population of FRCs were identified in control mice, and two populations of FRCs were identified in CLP mice. According to gene set enrichment analysis, FRCs after CLP are enriched in gene expression associated with inflammatory response and chemotaxis. Interestingly, activation of TLR4 (LPS) and TLR3 (Poly I:C) signaling in mesenteric adipose tissue derived FRCs significantly increased the expression of chemokines known to attract neutrophils (Cxcl2 and Cxcl5), monocytes (Cxcl3), and lymphocytes (Cxcl13 and Ccl20), while activation of TLR9 (ODN1518) significantly decreased these chemokine gene expressions. Importantly, we found similar regulatory effects of TLR3, 4, and 9 on chemokine production in human adipose tissue-derived FRCs. Together, our results indicate that TLRs plays critical roles in regulating chemokine production by FRCs. These data form a knowledge basis upon which to design new therapeutic strategies of FRC-based treatments for sepsis.

Resumen
Abstract Fibroblastic reticular cells (FRC) are highly heterogeneous. Distinct FRC subsets play unique roles in the formation of secondary lymphoid organs and immune responses. We have previously shown that TLR9 signaling plays a critical role in regulating peritoneal immunity via suppressing chemokine production in FRC in the fat associated lymphoid clusters (FALC). However, the subset-specific roles of TLR9 in FRC of FALCs remain unknown. Using single-cell RNA-sequencing, we identified three distinct subsets of FRC (CD55hi, CD9hi and CD55loCD9lo) in mouse mesenteric FALCs at baseline. Based on the gene set enrichment analysis, the Cd55hi subset was enriched in gene expression related to cell differentiation. The CD9hi subset was enriched in gene expression related to immune response. The CD55loCD9lo subset was enriched in the gene expression related to extracellular matrix formation. Furthermore, we found that CD9hi FRC from Tlr9−/− mice increased gene expression associated with inflammation. Using flow cytometry and bulk-RNA seq, we successfully isolated and validated these three subsets in mouse FALCs. Interestingly, activation of TLR9 signaling using ODN1585 significantly decreased CD9 expression in FRC in vivo and in vitro. Furthermore, treatment of ODN1585 suppressed the proliferation of CD9hi FRC, evidenced by decreased expression of Ki67. These results indicate that CD9hi FRC in FALC are immunoregulatory. TLR9 signaling may modulate peritoneal immunity via regulation of the proliferation of CD9hi FRC. Understanding the mechanism of how TLR9 regulates CD9hi FRC may lead to discovering new therapies for inflammatory diseases.

The latency of visually guided saccades executed after a cueing condition was adopted as a more ecological index to explore how spatial attention, usually assessed by manual responses, operates in the visual field. Subjects executed saccades aimed towards one of 4 possible positions, equally distributed around a central fixation cross. Before the onset of the saccadic target, a visual cue was briefly presented at the same or at a different spatial location. The visual cue was non-predictive of the target position. Two experimental sessions were carried out, differing for the onset asynchrony between cue and target. A time-dependent coupling between the task-irrelevant location of the cue and the direction of a following overt shift of attention emerged. Clinical applications of the adopted experimental setting to neurological and psychiatric patients with motor impairments are discussed.

Resumen
Abstract Visceral adiposity in elderly is associated with alterations in adipose tissue immune cells leading to inflammation and metabolic dysfunction. The Nlrp3 inflammasome is a critical regulator of macrophage activation, inflammation, and immunometabolism in visceral adipose tissue during aging; however, the potential contribution of adipose tissue B cells is unexplored. Here, we show that aging expands adipose-resident B cells and fat-associated lymphoid clusters (FALCs) in visceral white adipose tissue of female mice. Adipose tissue B cells exhibit a memory-like B cell profile similar to the phenotype of aged B cells that are increased in spleen of old mice. Mechanistically, the age-induced FALC formation and adipose B cell expansion, but not B cell transcriptional program, is dependent on the Nlrp3 inflammasome. Furthermore, B cell depletion in aged mice improves insulin sensitivity and metabolic capacity of adipose tissue. These data reveal that inhibiting Nlrp3-dependent B cell accumulation can be targeted to reverse metabolic impairment in aging adipose tissue.

Resumen
Context: Protein 53 (p53) is a well-known tumor suppressor protein, while murine double minute 2 (MDM2) acts as a negative regulator of p53, leading to p53 inactivation and cancer development. Aucklandia costus Falc. or Saussurea lappa contains bioactive compounds, particularly terpenoids, known for their anticancer activity against various cancer cells. Targeting the p53-MDM2 protein interaction and inhibiting MDM2 are crucial strategies in cancer therapy. Aims: To analyze the anticancer properties of A. costus terpenes against MDM2 protein. Methods: The compounds costunolide, dehydrocostus lactone, lappadilactone, and cynaropicrin were docked with MDM2 (PDB ID: 4HG7) using AutoDockTools 1.5.6. Additionally, the physicochemical, pharmacokinetic, and toxicity properties were predicted using pkCSM. Results: Lappadilactone exhibited the highest binding energy value, surpassing both the control and the native ligand. Following lappadilactone, cynaropicrin, costunolide, and dehydrocostus lactone displayed decreasing binding energies. When assessing ADMET properties with pkCSM, all compounds exhibited good permeability, suggesting their ability to penetrate intestinal cell membranes, and showed no signs of hepatotoxicity. Conclusions: Lappadilactone emerges as a promising candidate with high intestinal absorption, distinctive distribution characteristics, and a lack of mutagenic or hepatotoxic effects.

Resumen
Aucklandia costus Falc. (Synonym: Saussurea costus (Falc.) Lipsch.) is a perennial herb of the family Asteraceae. The dried rhizome is an essential herb in the traditional systems of medicine in India, China and Tibet. The important pharmacological activities reported for Aucklandia costus are anticancer, hepatoprotective, antiulcer, antimicrobial, antiparasitic, antioxidant, anti-inflammatory and anti-fatigue activities. The objective of this study was the isolation and quantification of four marker compounds in the crude extract and different fractions of A. costus and the evaluation of the anticancer activity of the crude extract and its different fractions. The four marker compounds isolated from A. costus include dehydrocostus lactone, costunolide, syringin and 5-hydroxymethyl-2-furaldehyde. These four compounds were used as standard compounds for quantification. The chromatographic data showed good resolution and excellent linearity (r2 ˃ 0.993). The validation parameters, such as inter- and intraday precision (RSD < 1.96%) and analyte recovery (97.52–110.20%; RSD < 2.00%),revealed the high sensitivity and reliability of the developed HPLC method. The compounds dehydrocostus lactone and costunolide were concentrated in the hexane fraction (222.08 and 65.07 µg/mg, respectively) and chloroform fraction (99.02 and 30.21 µg/mg, respectively), while the n-butanol fraction is a rich source of syringin (37.91 µg/mg) and 5-hydroxymethyl-2-furaldehyde (7.94 µg/mg). Further, the SRB assay was performed for the evaluation of anticancer activity using lung, colon, breast and prostate cancer cell lines. The hexane and chloroform fractions show excellent IC50 values of 3.37 ± 0.14 and 7.527 ± 0.18 µg/mL, respectively, against the prostate cancer cell line (PC-3).

Resumen
Abstract This paper aims to investigate the stress and deformation characteristics of double-row piles in deep foundation pit excavation by selecting representative strata in the Changchun area. The changes in bending moment, displacement, and earth pressure of double-row piles during the excavation are established using the FALC 3D software. The findings indicate that the deformation of the front pile is larger than that of the back pile, and the earth pressure distribution of the fornt pile deviates from the conventional Rankine earth pressure theory. Moreover, there are noticeable variations in earth pressure at the soil layer interface of the rear pile.

Resumen
Monotheca buxifolia is an ethnomedicinally and economically important threatened fruit bearing plant species in Malakand Division Pakistan. The genetic diversity among the 92 various genotypes of Monotheca buxifolia was carried out using sodium dodecyl sulfate poly acrylamide gel electrophoresis (SDS-PAGE) method. A considerable amount of inter districts genetic diversity (66.70%) was observed among the genotypes of M. buxifolia. Protein profiling was conducted on 12% gel electrophoresis. A total of 6 protein bands were observed in M. buxifolia genotypes. SDS-PAGE practice is a convenient scheme for the examination of both genetic diversity and relationship. Particularly, L-4 and L-5 were monomorphic in the inter districts Monotheca buxifolia genotypes and was recognized as species specific. The remaining other loci were polymorphic. In this investigation, the high inter and intra- districts specific diversity was observed demonstrating SDS-PAGE is an authoritative procedure for categorizing the genetically diverse germplasms in M. buxifolia. The findings from this study could be useful in the identification and selection of suitable M. buxifolia genotypes for future conservation programmes. Today, there is still a need to examine the genetic diversity and protect genetic resources, in particular wild species, for possible benefits in plant conservation programmes. To the best of our knowledge, this is the first ever report that addresses genetic variability in M. buxifolia.

Resumen
The present study was conducted in the cold desert region of the Lahaul valley in Himachal Pradesh. Saussurea costus (Falc.) Lipsch. an endangered medicinal herb, was studied in terms of its distribution pattern, cultivation practices and problems in cultivation. Seed germination showed better results in hot chamber in the month of May, June and July. Seeds sown in hot chamber at 30oC showed better germination (99±1.33%) in the month of June in comparison with cold chamber at 20oC. Energy and monetary efficiencies revealed output/input ratio of 7.5 and 0.8, respectively. Introducing of seasonal cash crops like Pea, Potato and Hop, need for a mandatory permit for cultivation and export, longer cultivation cycle (3 years), lower and continuously fluctuating market rates were found to be the main causes of discarding kuth cultivation in the valley. The possibilities for its revival and popularising kuth cultivation have been discussed in the present study.

Resumen
Saussurea costus (Falc) Lipsch is a traditional herb used to treat kidney stone problems because it contains several molecules used to treat this health problem, such as quercitrin. Infectious stones are the most painful of all urinary tract disorders, with ammonium phosphate (struvite) and carbapatite stones being the most common, caused by a bacterial infection with urease activity. These stones are treated with antibiotics, but antibiotic resistance is on the rise. The current study investigated the anti-urolithic activities of S. costus aqueous and ethanolic extracts of against struvite crystals synthesized using microscopic crystallization and turbidimetric methods, respectively. The utilized methods indicated that the ethanolic extract of this plant has a significant inhibitory effect on struvite crystallization, with a percentage inhibition of (87.45 ± 1.107) (p < 0.001) for a concentration of 1 mg mL−1 and a decrease in the number of struvite crystals, reaching values less than 100/mm3. For the number of struvite crystals inhibited by cystone, we found a value of 400/mm3 and with the aqueous extract we found 700/mm3. The antibacterial activity of the plant extracts studied was examined against several urease-producing bacteria, and this activity was evaluated by qualitative and quantitative evaluation methods; the highest minimum inhibitory concentration was seen in the ethanolic extract, with an MIC of 50 mg mL−1 for Staphylococcus aureus followed by an MIC of 200 mg mL−1 for Klebsiella pneumoniae. It showed a minimal bactericidal concentration (MBC) against S. aureus and K. pneumoniae (>50 mg mL−1 and >200 mg mL−1, respectively). Furthermore, to determine the extract’s anti-inflammatory activity, in vivo anti-inflammatory activity was investigated in rats. The results show that at a dose of 400 mg kg−1, the ethanolic extract has a maximum edema inhibition of 66%.

Diseases and infections of the respiratory tract are common global causes of morbidity and mortality. Our study attempts to elucidate a novel remedy for respiratory ailments, in addition to identifying and quantifying the metabolites of Saussurea costus root extract (SCRE) using HPLC. Then, in vitro antiviral and in vivo lung protective effects were elucidated. The in vitro antiviral potential of SCRE was analyzed via plaque assay against the low pathogenic human coronavirus (HCoV-229E) and human influenza virus (H1N1). The value of the half maximal inhibitory concentrations (IC50) of SCRE against HCoV-229E and H1N1 influenza virus were 23.21 ± 1.1 and 47.6 ± 2.3 µg/mL, respectively. SCRE showed a histological improvement, namely a decrease in inducible nitric oxide synthase (iNOS) and caspase-3 immunoexpression in in vivo cyclophosphamide (CP)-induced acute lung injury (ALI). Moreover, there was a considerable decline in microRNA-let-7a gene expression and a significant rise in heme oxygenase-1 (HO-1) gene expression, with a marked decrease in the malondialdehyde (MDA) level. Molecular docking studies revealed that the major constituents of SCRE have a good affinity for caspase-3, HO-1, and iNOS proteins. In conclusion, a traditional plant SCRE could be a promising source of novel therapeutic agents for treating and protecting respiratory tract diseases. More future investigations should be carried out to reveal its efficacy clinically.

Resumen
Plants are rich source of medicinally important compounds such as phenolics compounds and flavanoid used as antioxidant and have chemo-preventive role against the risk of oxidative stress-related diseases. Saussurea costus (Falc.) Lipsch. ( Asteraceae ) have medicinal properties, such as anti-inflammatory, antioxidant, hepatoprotective, antiulcer, anticancer, immunomodulatory and pesticidal activities. Phytochemical investigation was carried out with hexane, chloroform, methanol and ethanol extract. six extracting solvents, methanol, ethanol, aqueous methanol (80% v/v), aqueous ethanol (80% v/v), aqueous methanol (70% v/v) and aqueous ethanol (70% v/v) were applied for their efficacy to extract antioxidants from air-dried, sun-dried and oven-dried roots of Saussurea costus. There was a significant difference (P < 0.05) in the extracting ability of each of the solvents. The aqueous solvents were found superior in their ability to extract the antioxidants and aqueous ethanol was reported more efficient than aqueous methanol. Highest and lowest extractive value and antioxidants compounds were found from respectively Oven dried (40°C) and air dried (ambient, approx 25°C) Saussurea costus roots. There was excellent correlation between extraction yield, antioxidant activity and total phenolic content.

Resumen
The local community of the Suleiman and Hindukush mountain systems in Pakistan has largely depended on the natural resources of the environment since ancient times. The ecosystem of these regions is under huge pressure due to a lack of awareness and the uncontrolled interference of communal, commercial, security, political, and ecological conditions. The present study was designed to illuminate the link between mountain society and the consumption of the benefits from Monotheca phytocoenoses using the ecosystem services concept from the sphere of the socio-ecological system to cultural relations. The use of this approach is very important due to the visible role and dominant status of Monotheca vegetation within the ecological system of the region. M. buxifolia is strongly connected with both local and cultural traditions and is counted as a key species, particularly for high-mountain inhabitants. We report that Monotheca phytocoenoses provide several services including shelter, food, fodder, medicines, and wood, etc., to the indigenous community and is highly valued in the local culture because of the poor economic condition of the society. The concept of this cultural keystone species is crucial for understanding ecosystem services and must be considered for the protection and conservation of these habitats. The results of field and social studies have shown that the stable maintenance of Monotheca phytocoenosis forests ensures the existence of key species as the most important providers of ecosystem services, e.g., provisioning, regulation, maintenance and cultural services, indicating the close relations between society and the protection of mountain areas. According to the results obtained, the mountains community of the studied area believes that tree species like M. buxifolia, F. palmata, O. ferruginea, P. granatum, A. modesta, J. regia, etc., are the key components contributing to the function of both the mountain ecosystem and communities’ well-being. This approach will be extremely useful for ensuring an inclusive management of the socio-ecological system of the Hindukush and Suleiman Mountain ranges of Pakistan.

Resumen
Background: Rice is an essential component of the diets of people all over the world, and it accounts for a sizeable amount of the calories that they consume. The world's food supply suffers huge economic losses due to fungal deterioration on rice seeds. The medicinal plant Saussurea costus contains bioactive chemicals and is well-known for its many therapeutic uses. There are a number of species of Aspergillus that are considered to be diseases in vegetables and fruits. One of these species is Aspergillus niger. The purpose of this current investigation was to investigate the impact of the crude Alkaloid, Flavonoid, and Terpenoid chemicals derived from Saussurea costus' roots on Aspergillus species Purified from Rice seeds obtained from various markets in Hillah, Iraq. Methods: In vitro, antifungal activity against Aspergillus species was achieved using the food poisoning method. Three concentrations 5, 10, and 15mg/ml of each crude compound were prepared and compared to a positive control represented by Carbendazim500g/l and a negative control represented by 10% dimethyl sulfoxide. Results: An objective of the current investigation was to manage the Aspergillus species growth identified in Rice seeds by utilizing secondary compounds derived from the roots of Saussurea costus. The study's data indicated that the extracts represented by active compounds such as Alkaloids, Flavonoids, and Terpenoids obtained from Saussurea costus roots exhibited a significant decrease in the of Aspergillus species growth, particularly at a concentration of 100mg/ml, in contrast to the negative control. This effect was equivalent to that of the positive control, Carbendazim 500g/l.Conclusion: Finally, Saussurea costus roots have shown the most efficacy in controlling Aspergillus species, particularly through the presence of Alkaloid, Flavonoid, and Terpenoid chemicals at a concentration of 100mg/ml.Keywords: Antifungal activity; Saussurea costus; Alkaloids; Flavonoids; Terpenoids

Resumen
This study investigates the quality composition of Aba knife fish (Gymnarchus Niloticus) predominant in South-eastern Nigeria. The fish sample require proper processing techniques to preserve their organoleptic qualities for a substantial period. The objective of the study was to investigate the influence of oven-drying process on the proximate and organoleptic qualities of Gymnaruchus Niloticus. The studied fresh fish fillets were rinsed in clean water, brined and spread in trays before taken for oven drying. An oven drier (FALC STF-F-52) was used for the oven-drying process. Analyses of proximate compositions (moisture content, crude lipid, crude protein, ash content and crude fiber) were carried out on the fresh and oven dried fish samples. The study showed that oven drying decreases the moisture content to a safe level of 38.13 ± 0.01% dry basis(d.b), and increase the crude protein content (18.23±0.01), fat content (4.34±0.01), ash content (3.40±0.01), carbohydrate content (35.96±0.01) of the investigated fish samples. There were no crude fibre contents (0.00) in the fresh fish samples. Therefore, the oven drying process had no noticeable effect on the fibre content. The oven dried fish samples were rated high in terms of taste, aroma, colour, texture and general acceptability by the panel of assessors. The significance of the obtained results as well as recommendations for further studies were offered.
