Texto completoAtom interferometry is applied to absolute measurement of gravity acceleration g, to provide an accurate value for the realization of the LNE watt balance. The atomic source is obtained from a cloud of cold 87 Rubidium atoms. Two vertical counter-propagating are used to generate stimulated Raman transitions, that separate the wave-packets and make them interfere. During the transitions, the phase difference between the beams is printed on the phase of the free-falling atoms. Then the atomic phase shift between the two vertical paths becomes sensitive to the atom acceleration and allows obtaining an accurate value of g. A part of this manuscript is dedicated to the study of noise sources which deteriorate the measurement sensitivity. In particular, we detail the vibrations contribution, which we are able to reduce by a factor of 3 to 10, depending on the configurations, thanks to the measurement of a seismometer and to its analog processing. The best reported sensitivity, in optimal environment, is 1,4 10−8~g/Hz1/2. The study of the measurement accuracy also represents an important part of this work. Although the vacuum chamber was only temporary, we started to list the systematic shifts. According to two comparisons with well-known absolute gravimeters based on optical interferometry, our measurement shows a residual bias of 16 10−9~g

Texto completoSensitivity of plastics explosives is one of the most important problems with energetic materials. In this work, models of plastic-bonded explosives were created with the aim of correlating their energetics to their sensitivity. Three representative RDX-based formulations were simulated where HTPB-DOA, Estane and EVA were used as the polymeric phase for the simulations. A new molecular model was develop using single chain system with a compression–minimization iterative procedure. Molecular mechanics simulations were carried using conditions similar to those found experimentally in hot-spots. Resulting models were analysed in terms of potential energy and conformational and compared to sensitivity values (Drop Weight Impact Test and GAP test) reported in the literature. Then the effect of the chain groups in HTPB was analysis to correlate with sensitivity. Preliminary models show a good correlation between sensitivity of explosives and molecular modelling data.

Nous avons isole les cas ou la sensibilite pouvait etre evaluee en ligne, ou l'hypothese de panne unique etait indispensable pour assurer une sensibilite independante des entrees/sorties, ou une annulation systematique de certaines composantes du residu amenait a une nouvelle analyse de sensibilite sur un residu reduit ces differents resultats constituent un schema d'analyse systematique de sensibilite des residus polynomiaux

Texto completoObjective : To evaluate control methods for measured or unmeasured confusion bias, in observational situation of psychotic or schizophrenic patients. Methods : Propensity score method (for measured confusion bias) and analyses of sensibility (for unmeasured confusion bias) were applied in the field of psychiatric epidemiology, specifically in schizophrenia. In first, the question of residual bias was underlined by the results of a transversal study. The exposition at a contextual parameter (prison) was studied in link with psychotic disorders (DSM IV), with a classic control method.Second, to lead to an unbiased estimation of treatment effect, we compared a classic control method with a method based on propensity score. These approach were applied to a cohort of French schizophrenic patients where we studied the event (relapse) by the treatment exposition (polypharmacy or not).Third, we developed a synthesis on modelisation of uncertainty and non-measured confusion bias. Theories and methods were described, and then applied on results of previous studies. Results : The transversal study, with non-demonstrated results still then, allow us to reach the question of control quality in the case of exposition to a parameter in observational situation. The cohort study permit to compare a classic control method and propensity score (PS). We highlighted different results according to some control method. Stratification method on PS seemed to be the best method to predict relapse according to treatment exposition. Non-measured bias control methods were then described. And a combination of probabilistic methods was applied to the previous studies. Conclusion : In the case of observational studies, the objective was to study, to describe and to apply modelisation methods to take in account differences at baseline, potentially source of confusion bias. This research is at the crossroads of methodology, biostatistics and epidemiology

Texto completoNumerous studies in various alphabetic languages have shown that letter knowledge is one of the best predictors of reading achievement. This research, registered in the theoretical context of emergent literacy, is reported in order to examine the early letter knowledge development and its contribution to reading and spelling skills. A first longitudinal study describes letter name and letter-sound knowledge evolution, assessed across three testing times. The participants were 57 French children, followed from beginning of kindergarten through the beginning of first grade. Uppercase form, high graphonemic frequency of the letters, their graphic proximity, phonological structure of the name of the consonants (VC) as well as the early literacy interest but not the level of print exposure influence the development of these knowledge. A series of regression analyses highlighted the predictive value of letter-name knowledge on the development of letter-sound knowledge, on subsequent learning of reading and writing at the end of first grade and its contribution to word recognition at beginning of Grade 3. A second cross-sectional study carried out with 74 young children (mean age: 4 years, 8 months) described the influence of explicit and implicit letter knowledge in recognition and spelling of written pseudo-words. Letter-name knowledge seems to support letter-sound relationships, emergence of phonemic sensitivity and plays an essential part in the initial development of spelling skills and identification of written words. The conclusions resulting from these two studies suggest that early letter-name knowledge is a critical preschool acquisition and could constitute prop of emergent literacy and fundamental building block of subsequent literacy development

Texto completoThe finite element method allows for the determination of the states of the stress in a mechanical structure under solicitation. Manufacturing processes may be modeled. Nowadays, the reduction of computational costs allows to tackle inverse problems for the identification of some material or geometrical parameters of the structure that are optimal to a given problem. The inverse problem makes use of a mechanical model and an objective function measuring the discrepancies between the observed response and responses resulting from different sets of parameters. This thesis describes a gradient method and the automatic differentiation technique for the solution of two inverse problems in mechanics. The first one is concerned with the identification of a strain-hardening law from uniaxial tension test performed on two different steels and from the academical code \fl. The second one deals with the identification of a viscoelastic law for a sandwich material from experimental data measured on a simply supported sandwich beam and a complex eigenvalue solver developed at LPMM. These two models are differentiable. For each problem, the optimal parameter set is identified as a set for which the gradient of the objective function is zero. We use the automatic differentiation technique to generate codes for the computations of these gradients in a simplified manner. Results we obtained demonstrate the interest of such an approach

Texto completoLegionella pneumophila (Lp) is an accidental human pathogen which can infect alveolar macrophages and pneumocytes. During infection, Legionella have to deal with to various types of antibacterial agents, such as antimicrobial peptides (AMPs) produced by the host, and antibiotics with intracellular activity administered to patients. The mechanism of action of human AMPs against Legionella, and the resistance level to antibiotics of the bacterium are still poorly described. Our work aimed to contribute to a better understanding of the anti-Legionella activity of these molecules. The first part of this study consisted in the evaluation of the susceptibility of clinical Lp sg1 isolates to 8 antibiotics, to determine the epidemiological cut-off values of these different molecules. We demonstrated that all clinical isolates are susceptible to the tested antibiotics. The results revealed the presence of a subpopulation displaying a reduced susceptibility to macrolides. The analysis of the genomes allowed us to correlate this reduced susceptibility to le presence of the LpeAB macrolides efflux pump, found specifically in the sequence types ST1, ST701 and ST1335.The second part of this study was dedicated to the characterization of the antibacterial activity of the human AMPs LL-37 and HBD-3, and to the identification of their mechanism(s) of action against Legionella. All of the experiments show that LL-37 and HBD-3 induce a loss of cultivability by different mode of action. The results suggest that LL-37 is able to permeabilize the membrane of the L. pneumophila cells. Our findings also show that both peptides inhibit the intracellular replication of L. pneumophila, in part through collaboration with the host cell

Texto completoAs part of the piercing operations, billets undergo a reheating process from room temperature to a temperature of about 1250°C. During this heating, the thermal gradient in the billet leads to a deterioration of the axial health and is responsible of the initiation ofdefects on the formed tube.A methodology for the analysis of the mechanisms at the origin of these defects has been established. It is based on first, the characterization of the axial health with hot rheological tests to establish some indicators of ductility and second, on metallurgical analyses to identify the internal defects.Then finite element simulations have been performed to study the thermo-mechanical loadings induced by heating. Critical solicitation zones of the product during the processes have been identified.Finally a characterization of the observed defects into the billets has been led coupling cracks growth tests and X-FeM numerical simulations. The implementation of the experimental stress failure criterion, in the simulation of the heating cycle allows to obtain good values of thermal gradient boundary curves and leads to optimal heating curves.The analysis shows that the conditions at the beginning of the reheating process have a strong influence on the axial health of the billets. The results of these studies lead to friendly industrial solutions for improving productivity

Texto completoThe Internet of Things (IoT) is a new approach in which the Internet connection extends into real-world objects to make them communicate with each other. The communication for IoT can be established using cellular networks or the licence-free frequencies in the Industrial, Scientific and Medical (ISM) bands. There are many communication standards for IoT applications. There are also many modulation techniques. Nevertheless, there is no perfect solution for all the needs and requirements in all domains. The choice of the right communication standard and modulation technique is unique for each use-case and depends on each specific application.In this research, instead of developing a new communication standard or a new modulation technique, we improved the receiver sensitivity while using the existing modulation technique and communication standards. Improving the receiver sensitivity increases the link budget. A better link budget, under certain propagation conditions, increases the communication distance. Different radio communication standards and conventional techniques based on Low Power Wide Area Networks (LPWAN) are presented in the first chapter.The theoretical basis of the time-synchronous averaging (TSA) method as a powerful signal processing method to improve the sensitivity of the RF receiver and the simulation methodologies are presented in chapter two. We also study the impact of phase noise on the MSK modulator in this chapter.The synchronisation problem, as well as the different solutions to improve it, are presented in chapter three. Synchronisation in phase and frequency are studied separately, and an innovative method has been developed to combine the synchronisation process with averaging.The performance of the synchronised averaging method with and without new synchronisation techniques was measured, and the results are presented in chapter four. The processing gain from the theoretical calculation is compared with the measurement results too

Texto completoType 2 diabetes is a major health concern characterized by the association of metabolic syndrome with increased hepatic glucose production (HGP). Increased HGP is usually considered as a consequence of insulin resistance. We wondered whether, on the contrary, suppression of HGP could modulate peripheral insulin sensitivity. Mice deficient for liver G6pc, coding for the Glucose-6-phosphatase catalytic subunit, key enzyme of HGP (L-G6pc-/- mice), exhibited improved glucose tolerance and insulin sensitivity compared to wild type mice. Phosphoinositide 3-kinase/AKT pathway was greater induced by insulin in subcutaneous adipose tissue of L-G6pc-/- compared to wild type mice, whereas insulin similarly induced this pathway in epididymal adipose tissue of L-G6pc-/- and wild type mice. Analysis of adipose tissue metabolism revealed white to brown fat conversion specifically in subcutaneous adipose tissue of L-G6pc-/- mice. This change was characterized by the appearance of multilocular adipocytes expressing Ucp1 and Prdm16, correlated with increased expression of genes specific of oxidative pathway. L-G6pc-/- mice also displayed an increase levels in circulating FGF-21 that could be responsible for modifications of their subcutaneous adipose tissue. This data are the first demonstration that absence of HGP could modify peripheral insulin sensitivity and induce changes in adipose tissue metabolism

Texto completoThe present thesis explores the role of visual and proprioceptive modalities in graphic production. We use a distinction made by Meulenbrock and Thomassen (1991) between two independent spatial reference systems. The anatomical system is required to produce obliques, which are strong candidates from a motoric viewpoint, while the geometrical system results in bias for horizontal and vertical orientations, which are candidates from a perceptual viewpoint, with strong dependence on visual information. Systematic variations of visual feedback have been performed according to two experimental paradigms: visual occlusion and visuo-proprioceptive discordance by means of a mirror. We examine the importance of visual information in the planning, programming and execution of graphic movement patterns as a function of the item structure. Results confirm the role of visual guidance when copying items composed of horizontals and verticals. With regard to items composed of oblique segments, the programmation of movement parameters in terms of extrinsic coordinates or of intrinsic coordinates seems to depend on the motor complexity of movements. Results also show that the mirror-drawing task does not confirm our expectation that horizontals-verticals could induce proprioceptive adaptation, while obliques could induce visual adaptation. Two distinct behaviors appear whatever the item structure in such a task. Some subjects seem to favor a retroactive control of movements on the basis of visual information, while others rely more on a proactive control of movements, the motor system being determinant

Texto completoThe Arctic is warming twice faster than the rest of the world, leading to major changes for marine ecosystems. For example, sea-ice extent is decreasing, and air mass distribution is changing, thus modifying wind and precipitation regimes. In parallel, the Arctic is subject to increasing anthropogenic pollution carried by atmospheric and oceanic circulation, and accentuated by the development of local human activities. In this context, there is an urgent need to understand the ecological impacts of these environmental modifications on the species of this region. Little auks (Alle alle) are the most abundant seabird in the Arctic, and key players of coastal food webs. Although recent works suggested a high resiliency of little auks to global changes, a comprehensive study was needed to understand in detail the impacts of these changes. Through this work, we used a multidisciplinary approach (diet ecology, ecotoxicology, bioenergetics, foraging ecology) on the long term to characterize little auk sensitivity to environmental changes during the breeding season (summer) and in winter. We therefore studied a little auk population in East Greenland. Our results show that little auks are indeed impacted by ongoing changes. During the breeding season, their prey change and their diving effort increase when there is no sea-ice, even if they remain faithful to their feeding grounds on the continental slope. This tends to decrease their body condition and that of their chick, but does not impact their survival. In winter, little auks optimize their migration and their distribution according to the distribution of their prey and their energetic landscape. Therefore, our predictive models indicate that a warming of the North Atlantic could be benefic for little auk populations, by reducing their energetic needs. Finally, we found that little auks ingest microplastics, by confusing them with their prey. This additional source of pollution could increase with sea-ice loss that release microplastics stocked in the ice. This work underlines the importance of long term monitoring programs for the study of global change impacts

Texto completoThis thesis, dedicated to the history and development of the ecological thought, explores the collective mindsets that contribute to the open mindedness of one society to the new ideas of ecology, according to the traditions of thought, cultural conceptions of nature and religious and political cultures at work in a given country. Two case studies attracted our attention : Germany and the United States. Through strong fantasies on nature and a prolific intellectual environmental reflection these two countries appear as privileged matrixes to analyse the collective, ethical and ideological values attached to the protection of nature and the currents of thought at work in the collective environmental mind – from elaborated discursive reasoning to public opinion. This contextualising approach of the emerging ecological cultures allows a broader reflection on the specific affinities between Protestantism and environmentalism through the confrontation of historical considerations and empirical analysis of social representations of nature and the environment in the European public

Texto completoDespite significant clinical benefit from the consideration of molecular context, targeted therapies are still challenging. First part of this work focused on tyrosine kinase inhibitors targeting EGFR in non small cell lung cancers. Thus, improvement of biomarkers detection methods was completed by in vitro characterization of an unreported mechanism of acquired resistance. Briefly, pulmonary cells were exposed to a mutagen agent and a selection pressure was applied with EGFR inhibitors allowing the detection of TBK1 signature. Finally, synergic effect of that co-inhibition was highlighted. Now essentials in gynaecological cancers management, PARP inhibitors represent the second part of that work. Those targeted therapies are based on synthetic lethality. Consequently, BRCA1/2 pathogenic mutations are required for their administration, illustrating the issue of variants of uncertain significance. Toward their functional characterization necessity, a transcriptional analysis of splicing variant was first conducted on mRNA extracted from FFPE samples. Then, to evaluate functional signification of all types of variants, genomic edition was developed. Editing efficiencies of the unknown variant and a silent control one were compared in a haploid model where those genes are essentials. Functional signification of BRCA1/2 variants, and thereby mutations from all essential tumor suppressor genes in our model, can be evaluated in three weeks which is compatible with clinical management

Texto completoThis thesis is part of a multidisciplinary context on the border between the domains of the ambient intelligence, the human-machine interaction and the conversational agents. We study the problem of contextual proactive assistance in the course of the ambient computing. In this regard, our challenge is to provide to the user who wish have a virtual assistant, a proactive services which can communicate according to the user’s context. However proactive ambient intelligence, or contextual proactive assistance, did not follow an important development as reactive ambient intelligence where the user must explicitly express their needs. This is why the objective of this CIFRE thesis is to study and evaluate the concept of contextual proactive assistance through the implementation of an online conversational agent named X-CAMPUS. This acronym stands for eXtensible Proactive Multichannel Conversational Agent for Ubiquitous Services. This system aims to assist the user in their daily tasks thanks to its ability to perceive the state of the environment and interact effectively according to user’s needs (weather, TV programs, etc.). We conducted an experimental study to investigate the interest of X-CAMPUS and evaluate the satisfaction that it provides to users. We describe quantitative and qualitative results of this study obtained through a set of scenarios using different parameters of interaction (multimodal, multi-channels, behavior reactive/proactive agent, type of sensors, context mono/multi-user…)

Texto completoPodosomes are particular sub-cellular F-actin rich structures, composed of a dense actin-core surrounded at it base by numerous proteins that establish a close contact with the extracellular matrix on the ventral face of the cell. Unlike "classical" adhesive structures, podosomes are very dynamic and are able to locally degrade the extracellular matrix. These structures are specifically found in very motile cells, which mainly belong to the myeloid cell lineage, including macrophages, which are key cells of the innate immunity. Despite numerous studies that aimed to decipher podosome architecture and signaling pathways that regulates their formation, several questions remains about the exact role of these structures. Indeed, if the dynamic behavior of podosome seems to participate in the probing activity of the cells for analyzing their surrounding environment, studies of podosome mechanosensory processes is still sporadic. In this context, the purpose of my PhD was an exploratory research in order to decipher mechanosensory and biophysical properties of podosomes in human macrophages in vitro. Thus several complementary approaches were combined such as: (i) Atomic Force Microscopy, (i) micro-contact printing of matrix proteins, (iii) fabrication of matrices with various stiffness, and (iv) the use of a thin compliant suspended substrate. Finally my work enabled to explore podosomes at a nanometer-scale level in living cells and shaded light on several aspects as: (i) the height of podosomes is constant independently of the physicochemical properties of the substrate, (ii) theses structures are able to exert a normal, oscillating and periodic strain on the substrate related to acto-myosin complexes contraction and actin-polymerization activity, (iv) these structures have periodic stiffness oscillations closely related to the periodic strain they exert on the substrate, which are modulated by the stiffness of the substrate
