Texto completoNowadays, climate change, anthropogenic releases and the increase in the world's population are contributing to an increase in the number of bacteria of concern, drug releases and toxins into the environment. Ochratoxin A, estradiol, and some bacteria are among the contaminants polluting nature and threatening the health of living beings. In order to detect these potentially harmful elements, we worked on the development of an original oligonucleotide labeling. This marking is based on the use of electroactive metal complexes such as redox probes.These complexes are based on the macrocycle ligands DOTA and NOTA, usually mainly used in medical imaging, functionalized with iron (III). The study of their electrochemical properties, carried out by cyclic voltammetry, has shown that they have many advantages competing with the most common redox compounds. In particular, we sought to apply this oligonucleotide labeling to the construction of biosensors, with the first test of a genosensor for the detection of DNA from Vibrio bacteria. Sensor design and target detection were followed by impedance spectroscopy. However, impedance analysis did not achieve the expected results, and in order to extend the scope of our study, another method was tested. Therefore, we sought to couple metal complexes directly to aptamers via a reaction between a thiol function and a maleimide. In the next step, the biosensors were built by immobilizing the modified aptamers on electrodes. In parallel, aptamer-target interactions were quantified by thermophoresis or MST analyses to confirm certain results and validate the binding characteristics of aptamers
