Resumen
Genetic diversity in modern Arctic communities provides a baseline from which to assess population history. This is augmented by documenting patterns of genetic variation in prehistoric populations using ancient DNA methods, and inferring dietary resource information and adaptive strategies derived from stable isotope analyses. This chapter uses this multidisciplinary approach to examine population history and colonization events in the Aleutians of South Alaska, and the origin and population history of Paleoeskimo and Neoeskimo populations of the North American Arctic. The power to identify past demographic events relies on knowledge of both genetic and isotopic signatures of demographic events, and on acquisition of securely dated and well provenienced samples for analysis.

Molecular exercise physiology is the study of exercise physiology using molecular biology methods. The development of differentiated cell types is regulated by transcription factors like the muscle-making MyoD that specifies cell type, while others regulate the development of muscle, tendons, and bones. Maternal nutrition and exercise commonly affect embryonic development through epigenetic mechanisms. Adaptation to exercise involves sensor proteins detecting exercise-related signals, the processing of signals by signalling proteins and networks, and the regulation of the actual adaptations by effector proteins. Many sport- and exercise-related traits depend on both common and rare DNA sequence variations, including the muscle mass-increasing myostatin (GDF8) loss-of-function and the haematocrit-increasing EPOR gain-of-function mutations. Additionally, common DNA sequence variations contribute to the inherited variability of development, body height, strength, and endurance. Finally, in addition to ethical concerns, current genetic performance tests only explain a fraction of the variation of sport and exercise-related traits.

The life sciences are now a significant contributor to the ever expanding digital universe of data, and stand poised to lead in both the generation of big data and the realization of dramatic benefit from it. We can now score variations in DNA across whole genomes; RNA levels and alternative isoforms, metabolite levels, protein levels, and protein state information across the transcriptome, metabolome and proteome; methylation status across the methylome; and construct extensive protein–protein and protein–DNA interaction maps, all in a comprehensive fashion and at the scale of populations of individuals. This chapter describes a number of analytical approaches aimed at inferring causal relationships among variables in very large-scale datasets by leveraging DNA variation as a systematic perturbation source. The causal inference procedures are also demonstrated to enhance the ability to reconstruct truly predictive, probabilistic causal gene networks that reflect the biological processes underlying complex phenotypes like disease.

Resumen
Recent developments in molecular genetics have given us much information about DNA markers and a detailed look at evolutionary causes of genetic variation in humans. Patterns of genetic variation within populations and genetic differences among populations is best understood in light of our species’ African origin 200,000 years ago and the subsequent dispersion of human populations throughout the world 70,000 to 100,000 years ago. Although an evolutionary focus on genetic variation using methods from the field of population genetics provides us with considerable insight regarding our history, older ideas based on traditional definitions of biological race have no explanatory value.

‘Genes as markers’ shows that most genetic variation does not affect gene function or activity, but it is still of enormous interest. Notably it enables the identification of individual people, useful in forensics in the form of DNA fingerprinting, the establishment of paternity, and other information about family relationships. It also provides some evidence about the migration of human populations in historic and prehistoric times. Genetic variation has also enabled biologists to examine the thorny issue of human racial differences and establish the degree to which there is any biological basis for perceived race.
