Coronilla scorpioides (L.) W.D.J. Koch is known for producing several compounds with pharmaceutical interest, such as the hydroxycoumarins umbelliferone, scopoletin and daphnoretin, the dihydrofuranocoumarin marmesin, and the furocoumarin psoralen. In vitro callus cultures of C. scorpioides were established from hypocotyl, leaf, stem internode and root explants in order to evaluate the possibility of in vitro production of these active secondary metabolites. Calli were obtained with high frequency from all the explant types both in B5 and MS medium. However, after the third subculture, B5 medium, giving the best results, was selected for subsequent transfers. Homogeneous calli were kept either in darkness or in light. Chemical analyses showed that scopoletin and the intermediate products of the biogenetic pathway of psoralen, umbelliferone and marmesin, were always present in the calli and excreted into the media, while daphnoretin was never detected. Light seems to be a prerequisite for psoralen biosynthesis. Root-derived calli produced a significantly higher amount of psoralen (137.5 μg g−1 DW). Principal component analysis showed that umbelliferone, marmesin and psoralen contents are related to variables associated with different explant types.

Water stress can adversely affect seed germination and plant growth. Seed osmopriming is a pre-sowing treatment in which seeds are soaked in osmotic solutions to undergo the first stage of germination prior to radicle protrusion. Seed osmopriming enhances germination performance under stressful environmental conditions, making it an effective method to improve plant resistance and yield. This study analyzed the effect of seed osmopriming with polyethylene glycol (PEG) on seed germination and physiological parameters of Coronilla varia L. Priming treatments using 10% to 30% PEG enhanced germination percentage, germination vigor, germination index, vitality index, and seedling mass and reduced the time to reach 50% germination (T50). The PEG concentration that led to better results was 10%. The content of soluble proteins (SP), proline (Pro), soluble sugars (SS), and malondialdehyde (MDA) in Coronilla varia L. seedlings increased with the severity of water stress. In addition, under water stress, electrolyte leakage rose, and peroxidase (POD) and superoxide dismutase (SOD) activities intensified, while catalase (CAT) activity increased at mild-to-moderate water stress but declined with more severe deficiency. The 10% PEG priming significantly improved germination percentage, germination vigor, germination index, vitality index, and time to 50% germination (T50) under water stress. Across the water stress gradient here tested (8 to 12% PEG), seed priming enhanced SP content, Pro content, and SOD activity in Coronilla varia L. seedlings compared to the unprimed treatments. Under 10% PEG-induced water stress, primed seedlings displayed a significantly lower MDA content and electrolyte leakage than their unprimed counterparts and exhibited significantly higher CAT and POD activities. However, under 12% PEG-induced water stress, differences in electrolyte leakage, CAT activity, and POD activity between primed and unprimed treatments were not significant. These findings suggest that PEG priming enhances the osmotic regulation and antioxidant capacity of Coronilla varia seedlings, facilitating seed germination and seedling growth and alleviating drought stress damage, albeit with reduced efficacy under severe water deficiency.

A costa leste do Uruguay, de Montevideo até La Coronilla, caracteriza-se, no seu lado platense (a W de Punta del Este), por praias arenosas restritas, com escarpas ativas, promontórios formados por rochas do embasamento e pequenas ilhas; e, no seu lado atlântico, pela presença de lagunas e praias mais extensas e homogêneas, com maior exposição à ação do vento e de ondas de tempestade. A caracterização da cobertura vegetal e da morfodinâmica e sedimentologia do sistema praia - dunas frontais, incluindo rumo inferido de deriva litorânea longitudinal, permitiu estabelecer subdivisões desta costa em segmentos com características semelhantes. Os dois segmentos diferenciados pela associação de espécies vegetais foram o de Montevideo a Cuchilla Alta, caracterizado pelo predomínio de Panicum racemosum, e o de Jaurreguiberry até La Coronilla, com alta concentração de Spartina ciliata. Esta subdivisão foi correlacionada a fatores abióticos, como salinidade e intensidade do vento. Correlações estatísticas aplicadas a resultados de análise de variações granulométricas e mineralógicas dos sedimentos mostraram-se insuficientes para deduzir o rumo de deriva litorânea longitudinal. Critérios mais efetivos na determinação desse rumo foram os geomorfológicos, como deflexão das desembocaduras de arroios e lagunas e, principalmente, e assimetria dos arcos de praia com forma em espiral logarítmica. Com base na variação de sentido de transporte litorâneo residual assim deduzido, a costa também foi subdividida em dois segmentos. O primeiro, entre Montevideo e as proximidades da laguna Garzón, apresenta transporte preferencial para SW. No segundo, entre laguna Garzón e a região de La Coronilla, a deriva toma rumo NE. Os principais fatores controladores desta inversão de deriva seriam a mudança de orientação da linha de costa e a refração de ondulações exercida por bancos sedimentares submersos, situados no prolongamento do estuário do rio de la Plata.
The east uruguayan coast, between Montevideo and La Coronilla, is characterized, in the side corresponding to the La Plata River, by active scarps, promontories formed by basement rocks, and small islands. The Atlantic side, on the other hand, has lagoons and extense and homogeneous beaches, under more intense winds and storm waves action. The characterization of the vegetal cover and the morphodynamic and sedimentology of the beach-foredune system, including the infered net longshore drift direction, allowed to divide this coast into sectors. In the foredunes, we observed two distinct sectors as for the association of plant species. The first one is located between Montevideo and Cuchilla Alta and is characterized by the predominance of Panicum racemosum, whereas the second one is located between Jaurreguiberry and La Coronilla and presents a high concentration of Spartina ciliata. The distinction between these two sectors could be correlated to abiotic factors, as salinity and wind strenght. Linear correlation applied to spatial variation of mineralogical and granulometric data has turned out to be insufficient to infere the net longshore drift direction, despite of its success in some regions. The most effective criteria in order to determine this direction were the geomorphological characteristics, mainly the deflection of rivers and lagoons mouths and the assimetry of logarithmic spiral beach lines. By observing the net longshore drift, we identified two distinct sectors. The first one, between Montevideo and the region nearby Laguna Garzón, presents a preferential transport toward SW, whereas the second one, between Laguna Garzón and La Coronila, has longshore drift towards NE. The main factors controlling the longshore drift inversion would be the shifts of the coastline direction and the swell waves refraction exerted by submerse sedimentary bars in the eastern extension of La Plata Estuary.

Coroners have always been associated with investigating death, but this ancient office has undergone considerable reform since its creation in the twelfth century. The role of the coroner of the twenty-first century involves investigating death, conducting autopsies, furnishing reports, issuing death certificates and if necessary, testifying in court hearings. However, the original function of the office of coroner was linked to raising revenue for the King in the form of large fines or “amercements”. By the fourteenth and fifteenth centuries the prestige and fiscal functions of the medieval coroner had diminished as a result of the increased power of justices. The gradual move away from justices’ control during the eighteenth century elevated the status of the coroner, whose role evolved into one that formed a more effective system for reporting and investigating cause of death. This in turn further legitimised the role of the office of coroner in the English legal system. It was this “modernised” institution that formed the foundation of coronial law and practice in Australia.

The investigation and classification of deaths in England and Wales relies upon the application by medical practitioners of diverse reporting standards set locally by coroners and thereafter upon the effectively unconstrained decision process of those same coroners. This research, using extensive analysis of Ministry of Justice (MOJ) and Office for National Statistics (ONS) data, presents comprehensive analysis of local variation in coronial outcomes across the three decision-making stages of whether to report the death, to advance to inquest, and the choice of inquest conclusion. Substantial local variation was found in reporting rates to the coroner over time (12-87%) and in rates of advancing reports of death to an inquest (6-29%). The profiles of inquest verdicts varied widely between coroner areas with some verdicts more prone to varied levels of use. Individual coroner areas were consistent over time in their rates of reporting, advancing to inquest and use of verdicts. The gender of the deceased was a major factor with all coroner areas reporting proportionately fewer female deaths than male, and female deaths being overall half as likely as that of a male to proceed to an inquest. Once at inquest, a woman’s death was more likely to yield a verdict of natural causes than that of a man. Coroners seemed prima facie to be ‘gendered’ in their approach to verdicts; that is, they were consistently more likely to favour a particular verdict when dealing with a death, according to the gender of the deceased. Decision Board Analysis (DBA), comprising three typical coroner case scenarios, was utilised to analyse the decision-making style of incumbent coroners. The DBA allowed for an examination of the way in which available case information was managed prior to coming to a conclusion, and free text respondent comments were captured. Coroners were found to vary widely in their choices of outcomes yet they managed the available information in a similar way, reading concordant amounts of information and agreeing on the relative salience of the available information. Coroners sometimes robustly defended chosen outcomes against alternatives with dichotomous positions being taken. Variation in coronial outcome could not be explained by any variation in decision-making style. Further analysis of coroners’ local practices and their determinants seems necessary.

O carrapato Boophilus microplus, um ectoparasita hematófago, está presente em áreas tropicais e subtropicais no mundo todo. Atualmente, seu controle é baseado no uso de pesticidas. Para auxiliar no desenvolvimento de novas vacinas para este parasita, é importante compreender melhor sua fisiologia. A coronina, uma proteína de citoesqueleto, pertence à Família das Proteínas G. Está localizada no córtex celular e apresenta várias funções, desde locomoção celular, fagocitose, macropinocitose a ligação de microtúbulos, orientação da polaridade de embriões, citocinese, polimerização dos filamentos de actina e regulação da organização do citoesqueleto. No presente trabalho, o cDNA de uma proteína semelhante a coronina foi isolado de uma biblioteca de cDNA de glândulas salivares de partenóginas por triagem imunológica. A análise de seqüência mostrou que o clone SG5 possui identidade entre 15 e 17% com seqüências de proteínas semelhantes a coronina de outras espécies, como as de humanos e de camundongo, e possui um tamanho de 1.938 pb, com uma suposta ORF de 960 nucleotídeos. Na análise da expressão do gene, verificouse que o mRNA deste gene está presente em ovos e larvas de 15 dias e glândulas salivares, intestino e ovário de teleógina e partenógina; em corpo gorduroso sua presença não foi detectada. Um estudo mais detalhado do papel da proteína semelhante a coronina nas glândulas salivares de B. microplus pode auxiliar em uma melhor compreensão da fisiologia deste parasita.

Deaths in custody often attract considerable public interest. While the State’s duties in relation to deaths in custody are clear, the key obstacles faced in trying to prevent these deaths relate to the nature of Western Australia’s police investigation model and the coronial system. Despite the number of deaths in custody remaining relatively stable in recent years, there is a lot more that can be done to prevent deaths occurring. The initial police investigation into any death in custody must be independent, efficient and effective. This allows the police to obtain evidence relating to the death and provide it to the coroner, and if an offence has been committed, to the prosecutor. The Western Australian model of police investigation is not optimal because it relies on police to investigate all deaths in custody, even when the deceased was in police custody or died as a result of police operations. This poses a conflict of interest and must be addressed. The coroner has the unique ability to make recommendations to prevent deaths occurring. There are a number of issues associated with Western Australia’s coronial system which mean that the coroner’s ability to prevent deaths in custody is not being fulfilled. These issues include that there is no system in place to monitor whether coronial recommendations are implemented, and that the coroner relies on the initial police investigation, which is conducted using a flawed model. This paper will discuss several systemic issues associated with police investigations and the coronial system. These issues seriously inhibit the potential to prevent deaths in custody because systemic and non-systemic issues are potentially not being identified or addressed. To resolve these issues, the Western Australian police investigation model and coronial system must be reformed.

The present work describes the study of the secondary metabolites of two species that belong to the Rhamnaceae family: Condalia buxifolia and Scutia buxifolia, and the study of the ratio structure/activity of seven cyclopeptidcs alkaloids and one neutral cyclopeptidc: scutianine-B (24), scutianine-C (14), scutianine-D (15), scutianine-E (16), scutianine-F (18), scutianine-M (37), condaline-A (22) and scutianene-D (38). In the bark of the bole of Condalia buxifolia three metabolites were isolated: lupeol (34), taraxerol (35) and β-sitosterol (33). In the bark of the roots was isolated the cyclopeptidc alkaloid condaline-A (22). In the bark of the roots of Scutia buxifolia the metabolites ursolic acid (36) and β-sitosterol (33) were isolated, whereas the cyclopeptidcs alkaloids scutianine-B (24), scutianine-C (14) and scutianine-D (15) were only identified in this part of the plant by comparison with authentic samples. Additionally, the antimicrobial activity of the portions and the isolated metabolites of Condalia buxifolia and Scutia buxifolia were analyzed by Bioautography test, utilizing different microorganisms: two gram positive bacteria (Staphylococcus aureus (Sa) and Staphylococcus epidermidis (Se)), three gram negative bacteria (Klebsiella pneumonia (Kp), Salmonella setúbal (Ss) and Escherichia coli (Ec)) and two fungus (Saccharomyces cerevisiae (Sc) and Candida albicans (Ca)). In this analysis it was observed that the stereochemistry of the amino acids that compose the alkaloids structure and the presence of the basic group of these structures are important to this sort of activity.
O presente trabalho descreve o estudo dos metabólitos secundários de duas espécies pertencentes à família Rhamnaceae: Condalia buxifolia e Scutia buxifolia, e ainda um estudo da relação estrutura/atividade de sete alcalóides ciclopeptídicos e um ciclopeptídeo neutro: scutianina-B (24), scutianina-C (14), scutianina-D (15), scutianina-E (16), scutianina-F (18), scutianina-M (37), condalina-A (22) e scutianeno-D (38). Da casca do caule de Condalia buxifolia foram isolados três metabólitos: lupeol (34), taraxerol (35) e β-sitosterol (33). Da casca da raiz foi isolado o alcalóide ciclopeptídico condalina-A (22). Da casca da raiz de Scutia buxifolia foram isolados os metabólitos ácido ursólico (36) e β-sitosterol (33), enquanto que os ciclopeptídicos scutianina-B (24), scutianina-C (14) e scutianina-D (15) foram apenas identificados nesta parte da planta através de comparação com amostras autênticas. Adicionalmente, foram analisadas atividades antimicrobianas das frações e dos metabólitos isolados de Condalia buxifolia e Scutia buxifolia, através da técnica de bioautografia utilizando-se diferentes microrganismos: duas bactérias Gram positivas (Staphylococcus aureus (Sa) e Staphylococcus epidermidis (Se)), três bactérias Gram negativas (Klebsiella pneumonia (Kp), Salmonella setubal (Ss) e Escherichia coli (Ec)) e dois fungos (Saccharomyces cerevisiae (Sc) e Candida albicans (Ca)). Observou-se, nesta análise, que tanto a estereoquímica dos aminoácidos que constituem a estrutura dos alcalóides, bem como a presença do grupo básico destas estruturas, são importantes para este tipo de atividade.

The thesis is focused on the Nueva Coronica y Buen Gobierno, by Felipe Guaman Poma de Ayala, written in the Peruvian highlands between the end of the sixteenth and the beginning of the seventeenth centuries. Although specifically designed to superficially appear innocuously and mildly complaining, the author disguises his transcendental subversive intentions by limiting the scope of the referential subject to the members of his cultural group.
While the introduction explains in detail the objective of the thesis and reviews some literature on the subject, the first chapter provides a broad and summary overview of indigenous Andean cosmology, its constitutive elements and its main characteristics. In the second chapter these elements of cosmological philosophy are presented as the only available tools at the disposal of the indigenous population in order to understand that catastrophic event referred to as the Conquest and the inevitable conclusion to which it arrived. The third chapter explores the use of those cosmological tools and the understanding of the disaster in the subversive endeavour of cultural survival in the context of colonization. Finally, the fourth chapter examines the plurality of levels of subversive messages conveyed by the author and his hidden message of rebellious hope. Within the same frame of cosmologically oriented thought patterns and messianistic expectations, Guaman Poma alters one apparently insignificant historical detail, thereby making possible the cosmic regeneration and historical rebirth of the last Inca emperor and his entire people. A section of conclusions and a bibliography follow the fourth and last chapter.

In the late sixteenth-century a Peruvian Indian and Inca nobleman named Guaman Poma de Ayala wrote a one-thousand page history of the world, his Nueva Cronica y Buen Gobierno, recounting the development of Andean and European humanity from the beginning of time up to the period in which the author lived. My analysis focuses on the mode of communication used by Guaman Poma, his use of Renaissance Iberian discursive and visual codes, to articulate his radical views of Spanish rule in Peru. His views, I argue, although articulated in a foreign language and media, express a fundamentally Andean understanding of the world. The conquest and the Spanish people are woven into the Andean mythological order. Resumen
El caso presentado se enmarca en el proyecto de tesis doctoral en curso, más amplio y abarcativo, titulado Discontinuados: El Proyecto territorial del balneario Uruguayo. En el mismo se exploran las transformaciones territoriales aceleradas de la Región Costera Sur del Uruguay y se indaga acerca de los procesos de urbanización en clave multiescalar, donde la conformación de los balnearios juega un rol destacado. En esta oportunidad, y a modo de episodio, se hace foco en el balneario oceánico de La Coronilla, por considerarse un caso crítico de deterioro de su patrimonio ambiental y cultural debido a la desembocadura en un sector inmediatamente adyacente al balneario del Canal Andreoni, una mega obra de ingeniería culminada en 1981, que consiste en el dragado de una suerte de rio artificial para optimizar la producción de arroz y la cría de ganado en las zonas rurales aledañas. The following case study is framed in the ongoing research, broader and comprehensive, titled Discontinued: The territorial Project of the ruguayan seaside resort. It is explored in it the accelerated territorial changes in the South Coastal Region of Uruguay and inquires about the urbanization process in a multiesalar approach, where the conformation of the seaside resorts plays an outsanding role. In this opportunity the focus is on the oceanic seaside resort named La Coronilla, as it is considered a critical case of spoilage of its environmental and cultural heritage. It exists a before and after for La Coronilla. It was a prestigious resort in the Coasts of Rocha, recognized not only for its sandy beaches but also for the lodging infrastructure mounted there, with at least six huge hotels almost built in the shore. Until is decided by 1981 to enlarge significantly the Canal Andreoni, which is was the beginning of the end.
