Addressing an area of high importance to the South African industry, the research activity soon grew into a research unit, commercial centre / centre of excellence, technology transfer unit and innovation support centre. Above all, the research started to impact on product development practices to deliver improved products. The paper considers the development of the available technology platforms at the CUT'S CRPM to become a technology power-house on the African continent, and how it impacted on Institutional Research Development in South Africa.

Texto completoViruses are highly efficient vectors that have been used for vaccination and gene therapy applications. However, their complexity renders downstream process particularly challenging since devices and strategies especially designed for virus purification are still lacking or need further optimization. After an introduction to the challenges of virus purification and the current strategies being employed, this dissertation presents the study of three different stages of the downstream process: clarification, ultrafiltration and chromatography. A novel clarification procedure based on diatomaceous earth was evaluated. Small-scale batch incubations led to the identification of Divergan RS – a synthetic non-charged material – as the most promising candidate for integration in a scalable filtration set-up. Ultrafiltration was addressed with the evaluation of cassette and hollow fiber modules. The results obtained show that cassette module with cut-offs in the 500 kDa range and highly hydrophilic materials enable complete recovery of infective Adenovirus while reducing process time in half when compared with the best hollow fibers. Despite the encouraging results with Adenovirus, the experiments using Retrovirus resulted in low yields and possible optimization strategies were identified. Membrane technology was also evaluated as an alternative to the packed-bed chromatography columns. By using a scale-down 96-well device, the impact of ligand density, membrane structure and feed conductivity were evaluated for the purification of Adenovirus by ion exchange chromatography. The hydrogel-grafted membrane with ligand density of 2.4 μmol cm-2 operated in bind/elute mode shown the best compromise between yield and purity. Overall, this thesis contributed to the advancement of virus purification field by exploiting innovative technologies.

Texto completoThe present work investigates the technology development of state-of-the-art SiGe and SiGeC Heterojunction Bipolar Transistors (HBT) by means of technology computer aided design (TCAD). The objective of this work is to obtain an advanced HBT very close to the real device not only in its process fabrication steps, but also in its physical behavior, geometric architecture, and electrical results. This investigation may lead to achieve the best electrical performances for the devices studied, in particular a maximum operating frequency of 500 GHz. The results of this work should help to obtain more physical and realistic simulations, a better understanding of charge transport, and to facilitate the development and optimization of SiGe and SiGeC HBT devices.The TCAD simulation kits for SiGe/SiGeC HBTs developed during our work have been carried out in the framework of the STMicroelectronics bipolar technology evolution. In order to achieve accurate simulations we have used, developed, calibrated and implemented adequate process models, physical models and extraction methodologies. To our knowledge, this work is the first approach developed for SiGe/SiGeC HBTs which takes into account the impact of the strain, and of the germanium and carbon content in the base, for both: process and electrical simulations.In this work we will work with the successive evolutions of B3T, B4T and B5T technologies. For each new device fMAX improves of 100 GHz, thus the technology B3T matches to 300 GHz, B4T and B5T to 400 and 500 GHz, respectively.Chapter one introduces the SiGe SiGeC heterojunction bipolar technologies and their operating principles. This chapter deals also with the high frequency AC transistor operation, the extraction methods for fMAX and the carrier transport in extremely scaled HBTs.Chapter two analyzes the physical models adapted to SiGeC strained alloys used in this work and the electrical simulation of HBT devices. This is also an important work of synthesis leading to the selection, implementation and development of dedicated models for SiGeC HBT simulation.Chapter three describes the B3T TCAD simulation platform developed to obtain an advanced HBT very close to the real device. In this chapter the process fabrication of the B3T technology is described together with the methodology developed to simulate advanced HBT SiGeC devices by means of realistic TCAD simulations.Chapter four describes the HBT architectures developed during this work. We will propose low-cost structures with less demanding performance requirements and highly performing structures but with a higher cost of production. The B4T architecture which has been manufactured in clean-room is deeply studied in this chapter. The impact of the main fabrication steps is analyzed in order to find the keys process parameters to increase fMAX without degrading other important electrical characteristics. At the end of this chapter the results obtained is used to elaborate a TCAD simulation platform taking into account the best trade-off of the different key process parameters to obtain a SiGeC HBT working at 500 GHz of fMAX

Consequently, the parts shall not be measured only when they are complete. For that reason, some specifications are verified at several intermediate stages of manufacturing. These specifications are called manufacturing specifications.Manufacturing transfer is the activity of determining the manufacturing specifications. With this PhD, Safran Aircraft Engines wants a manufacturing transfer method to be proposed. This method shall be automated in a software. The software inputs shall come from the digital environment of the company.In order to address this issue, PITOL (Production and Intermediate TOLerancing) and DMPITOL (Deviation Model for PITOL) are proposed. A manufacturing transfer method has been developed. It enables one to efficiently realize specification synthesis, tolerance analysis and tolerance optimization. Then the method is automated in a software, so that it is useable in industry.In order to ease the use of the method by Safran Aircraft Engines, some rules are proposed which make possible the calculation of manufacturing transfer for two specific processes. Moreover solutions are proposed to make the method suitable with the industrialization process

Texto completoIn some circumstances, a person may be deprived of natural abilities, such as walking and speaking, perhaps due to limb amputation, spinal cord injuries (SCI), or degenerative diseases. Assistive technology devices allows motor-impaired people to overcome their limitations promoting greater independence. Particularly suitable in the case of people with severe motor impairment, electrical biosignals have been successfully utilised to operate alternative communication devices. For over half a century, information extracted from the electromyographic signal for the purpose of operating a given device has not considered the information provided by the basic unit of the muscle: the motor unit. The objective of this study is to investigate how accessing information at motor unit level would improve the operator's performance during a given task. The hypothesis is that the proposed methodology would allow generating more precise control commands, when compared to traditional approaches relying on global information obtained by conventional electromyographic signal acquisition and processing. A system to detect motor unit action potentials from the electromyographic signal was devised, including the electrode design, and the performance evaluated by measuring the time taken to perform several cursor control tasks. The specifications of the cursor control task were extracted from a different study, which used the traditional electromyographic signal-processing approach. Comparing the results from both studies proved that the novel approach provides better control than the traditional one, being 27\% faster in the most difficult task

Texto completoAt the level of the middle school, the technology is a general discipline which addresses all the pupils. So, more than ever, in 21th century, the knowledge of the world of objects is a part of the culture of the citizen. Nevertheless, the technology in middle school is a discipline still under construction, the objects of knowledge are partially defined and the research in didactics of the technology still has numerous questions to investigate. This thesis discusses the design activity of students in middle school (pupils aged 11-15) in France and particularly to designing object. At first, our purpose is to answer, in an empirical way, the following question: what are the difficulties met by the pupils in situation of design? In the exploratory way, we analyze pupils' activity resolving collectively a problem of design. We intend secondly to analyze more particularly the difficulties bound to the production of representations of the possible solutions through the role which play the computing tools of computer-aided design. It is a way to think about the integration of these tools in the teaching of the technology in middle school. Would the use of graphic intermediaries modify and in which sense the performances of the pupils of 5th in the phase of research for solutions? The recourse to the theories of the action is relevant to observe the joint activity of the teacher and the pupil, especially in a domain or we expect from the research that she brings one so much one few answers. Through the results which she proposes, this thesis contributes to the community of the didacticiens of the sciences and the technology

Texto completoThis thesis deals with a methodology of Reverse Engineering (RE) of mechanical assemblies from heterogeneous data in a routine context. This activity consists, from the existing data of a part or an assembly, in rebuilding their digital mock-up. The data used in entrance of our process of RE can be drawings, photos, points clouds or another existing version of the digital mock-up. The proposed approach, called Heterogeneous Data Integration for Reverse Engineering (HDI-RE), is divided into three steps : the segmentation, the signature and the comparison of the initial data with a knowledge database. The signatures of the studied object are compared with the signatures of the same type existing in the database in order to extract components ordered by similarity (distance with the object). The parameterized digital mock-up which is the most similar to the object is then extracted and its parameters identified from the initial data. Data set processing, called "heterogeneous" data, requires a solution which is able to manage on one hand, the heterogeneousness of the data and the information which they contain and on the other hand, the incompleteness of some data which are in link with the noise (into points cloud) or with the technology used to digitize the assembly (ex: scanner or photography)

Texto completoThis research aims at studying online interactions in a social network that is specific to teaching and learning French as a Foreign Language (FFL). It questions the way the virtual environment incorporates with the training device so to provide new contexts of interactions and language learning.The data used for this research belong to an academic and language learning context which enabled an ecological analysis of the uses.Furthermore a discourse analysis from various participants was undertaken to highlight their impressions of the device, the noticed affordances and their appropriation with the online environment. This study contributes at elaborating useful tools for the analysis of participation and a better understanding of social interactions in the case of a mediated communication in a pedagogical context.The analyses are based on (1) participative activities, their conversational and enunciative format, (2) the various participation modes and relationnal social commitment that flourishes in a socio-pedagogical network and (3) the spatial and attentionnal features that are specific to a virtual learning environment.The study shows that there are few uses of the socio-pedagogical network, and the various ones that have been noticed did not benefit from the network’s wide access, conversational and social design.As a certain number of affordances have been noted by the participants, few of them were used in the case of our study. Considering those limited uses, the analysis tries to spot the restraints and obstacles to these changing practices by taking into account the participant’s discourse and suggested activities. From this observation, the research suggests didactical guidelines that include this social and relational challenge - and to consider this socio-pedagogical network as an environment which mediates social activities and digital litteracy

Texto completoThe prosthetic socket, an essential interface element between the patient's stump and prosthetic device, is most often the place where the degree of prosthetic success is defined. It is the most critical part of the prosthesis, customized to fit with the unique residual limb of the amputee. Without a proper socket shape and fit, the prosthesis becomes uncomfortable, or even unusable, and causes pain and skin issues. The state-of-the-art prosthetic production is still missing universal numerical standards to design a socket. The current practice is expensive and relies on the manual refinements of the orthopedic technician, and the fit quality strictly correlates with his skills as well as the subjective feedback of the patient. The thesis aims to conduct a deep analysis of an optimal design of the prosthetic socket by studying and developing an alternative computer-aided design process. This process is fully based on the virtual model of the patient’s residual limb and relies on the calculation of the socket-stump interaction. A fast calculation is favorable in this case, that’s why we propose to use the Mass-Spring System (MSS) instead of the widely used FE method to model the soft tissues of the residual limb. A new configuration of the MSS model is proposed to respect the non-compressibility property of the soft tissues by adding non-linear “Corrective Springs”. The numeric model is to be generated from the scanned model of the stump. For this purpose, we propose a fusion scheme of four RGB-Depth sensors for a rapid and low-cost scan with error reduction techniques. Finally, the virtual residual limb is used in the socket designing phase. A parametric design method is proposed and investigated. The design problem is transformed into a constraint-satisfaction-problem whose constraints are derived from the inverse calculation of the stump-socket interaction. The inverse approach has been chosen to eliminate the need for expensive contact formulation. This fact leads to rapid calculations, and consequently, allows to provide real-time numerical feedback during the designing process. The validation was done by comparing the results of our system with the output of FE simulations. The system has been implemented with a user-friendly graphical interface and virtually tested and numerically validated. This system reduces the limitations of the current practices. However, a lot of works is still ahead to refine and develop the system and validate it with clinical experiments

Texto completoBipolar complementary metal-oxide-semiconductor (BiCMOS) processescan be considered as the most general solution for RF products, as theycombine the mature manufacturing tools of CMOS with the speed and drivecapabilities of silicon-germanium (SiGe) heterojunction bipolar transistors(HBTs). HBTs in turn are major contenders for partially filling the terahertzgap, which describes the range in which the frequencies generated bytransistors and lasers do not overlap (approximately 0.3THz to 30 THz). Toevaluate the capabilities of such future devices, a reliable prediction methodologyis desirable. Using a heterogeneous set of simulation tools and approachesallows to achieve this goal successively and is beneficial for troubleshooting.Various scientific fields are combined, such as technology computer-aided design(TCAD), compact modeling and parameter extraction.To create a foundation for the simulation environment and to ensure reproducibility,the used material models of the hydrodynamic and drift-diffusionapproaches are introduced in the beginning of this thesis. The physical modelsare mainly based on literature data of Monte Carlo (MC) or deterministicsimulations of the Boltzmann transport equation (BTE). However, the TCADdeck must be calibrated on measurement data too for a reliable performanceprediction of HBTs. The corresponding calibration approach is based onmeasurements of an advanced SiGe HBT technology for which a technology specific parameter set of the HICUM/L2 compact model is extracted for thehigh-speed, medium-voltage and high-voltage transistor versions. With thehelp of the results, one-dimensional transistor characteristics are generatedthat serve as reference for the doping profile and model calibration. By performingelaborate comparisons between measurement-based reference dataand simulations, the thesis advances the state-of-the-art of TCAD-based predictionsand proofs the feasibility of the approach.Finally, the performance of a future technology in 28nm is predicted byapplying the heterogeneous methodology. On the basis of the TCAD results,bottlenecks of the technology are identified

Zudem sind HBTs bedeutendeWettbewerber für die teilweise Überbrückung der Terahertz-Lücke, derFrequenzbereich zwischen Transistoren (< 0.3 THz) und Lasern (> 30 THz).Um die Leistungsfähigkeit solcher zukünftigen Bauelemente zu bewerten, isteine zuverlässige Methodologie zur Vorhersage notwendig. Die Verwendungeiner heterogenen Zusammenstellung von Simulationstools und Lösungsansätzenerlaubt es dieses Ziel schrittweise zu erreichen und erleichtert die Fehler-_ndung. Verschiedene wissenschaftliche Bereiche werden kombiniert, wie zumBeispiel der rechnergestützte Entwurf für Technologie (TCAD), die Kompaktmodellierungund Parameterextraktion.Die verwendeten Modelle des hydrodynamischen Simulationsansatzes werdenzu Beginn der Arbeit vorgestellt, um die Simulationseinstellung zu erläuternund somit die Nachvollziehbarkeit für den Leser zu verbessern. Die physikalischenModelle basieren hauptsächlich auf Literaturdaten von Monte Carlo(MC) oder deterministischen Simulationen der Boltzmann-Transportgleichung(BTE). Für eine zuverlässige Vorhersage der Eigenschaften von HBTs muss dieTCAD Kon_guration jedoch zusätzlich auf der Grundlage von Messdaten kalibriertwerden. Der zugehörige Ansatz zur Kalibrierung beruht auf Messungeneiner fortschrittlichen SiGe HBT Technologie, für welche ein technologiespezifischer HICUM/L2 Parametersatz für die high-speed, medium-voltage undhigh-voltage Transistoren extrahiert wird. Mit diesen Ergebnissen werden eindimensionaleTransistorcharakteristiken generiert, die als Referenzdaten fürdie Kalibrierung von Dotierungspro_len und physikalischer Modelle genutztwerden. Der ausführliche Vergleich dieser Referenz- und Messdaten mit Simulationengeht über den Stand der Technik TCAD-basierender Vorhersagenhinaus und weist die Machbarkeit des heterogenen Ansatzes nach.Schlieÿlich wird die Leistungsfähigkeit einer zukünftigen Technologie in28nm unter Anwendung der heterogenen Methodik vorhergesagt. Anhand derTCAD Ergebnisse wird auf Engpässe der Technologie hingewiesen

The rapid advances in wireless communication and portable computer technologies have made feasible a new style of computing that may lead to a profound change of our life. Mobile computing is bringing us exciting new applications and services that promise to provide information access any where, any time. In this thesis, we study, from an educational point of view, how this new technology and media can be effectively used in computer aided learning. We argue, both theoretically and practically, that mobile computing is an invaluable tool for learners. An ongoing project of a Taiwan Butterflies Learning System is used as an example. The need for a National Learning Infrastructure (NLI) that integrates mobile and traditional communication links, as well as a National Learning Network that provides rich educational resources for everyone is also suggested as a long term goal of our education policy.

A multimedia learning environment combines many formats of materials, such as text, images, graphs and videos. Currently, most teaching representation models with single-monitor system raises a difficulty to present various formats of learning materials simultaneously. Students are benefited rarely by using those single-monitor representation systems in the classroom. Knowledge can be acquired effectively by learning in the models with multiple representations at the same time and by building referential connections among corresponding elements in different representations. This thesis designs a teaching aided system which can be easily used in multi-display environments and can conspicuously improve learning effects by Cognitive Theory of Multimedia Learning. This system not only offers some popular authoring tools such as Microsoft PowerPoint, Microsoft Word, Microsoft Excel, and Adobe Acrobat Reader, but also has universal controls on several multimedia objects like HTML pages, images, films, real-time videos from digital camcorder, and so on. It also realizes some synchronization mechanisms to properly organize events for Dual-Channel (dual-display) learning strategy. Teaching activities through this system can be recorded automatically as a learning object that can be replayed or reused by learners or teachers.
