Today, robotics has made remarkable advances in a number of industries, including the food and beverage sector. One area of particular interest is the development of an automated bartender capable of efficiently preparing cocktails and drinks. In this context, a state-of-the-art robotic bartender has been designed and implemented following the VDI 2206 methodology, developed by the Association of German Engineers (VDI). This methodology has allowed a more precise definition of the product and accurate estimations in all phases of the mechanical, electrical, and electronic design, and for the mechanical design of the robotic bartender, it prioritizes a solid and safe structure to support the weight of the bottles and components while focusing on an attractive design to improve the user experience. The system features precise sensors to detect and measure ingredients and intelligent control algorithms to ensure accurate dosing. The user interface is intuitive, allowing for uncomplicated cocktail selection and drink customization, and Proteus software was used for the electrical and electronic design, which facilitated extensive simulation and debugging. The system incorporates a PIC16F877A microcontroller to manage and coordinate various functions such as sensors, actuators, and communications. Finally, the pilot implementation has demonstrated the system's effectiveness in preparing a wide variety of cocktails with reduced preparation time. The robotic bartender can produce 10–12 cocktails every 10 minutes, which improves the productivity of the establishments and benefits both bartenders and customers.

Upamanyu Chatterjee (born in 1959) is an Indian Administrative Servant who currently serves as Joint Secretary to Government of India on the Petroleum and Natural Gas Regulatory Board. He is a 1983 batch I.A.S from Maharashtra Cadre. He is a published author and best known for his novel ‘English August’ also adapted into a film of the same title. His literary taste is so amazing and appreciating that he could write many realistic novels with his hard and busy duty of an administrative official. It’s another matter that Upamanyu Chatterjee is a novelist of a disillusioned age. He is by his education and heritage, a misfit in the modern world. He had a profound sense of his age. Being an administrative officer Upamanyu Chatterjee digs deep into the root of the Indian administrative system realistically He is pre-eminently concerned with urban life. Though the places in his novels are the tiny cities and villages, but he portrays the features of big cities realistically. His novels are of streets, houses and people and not of the woods, fields and flowers. We are much impressed by his characters and their gross selfishness and sensuality. The novels of Chatterjee are pervaded by symbolism such as dissmelled steak in passage ways, of stale beer, of cocktails and cigarettes, of dusty paper flowers, of females in shuttered. These things altogether show the inevitable gloom as well as the realistic, if not naturalistic picture of a metro city, Delhi, quite apart and away from the life of Madna, the life of insanity and boredom. In a way, he exposes the reality of his contemporary society and politics as well with his own vision that’s his realism and for this purpose,he employs symbolism to express the bitter things indirectly. My research paper has attempted to focus on his novelistic art of the above mentioned I.A.S.

This article considers Bernard DeVoto’s defense of Frederick Jackson Turner’s influential essay, “The Significance of the Frontier in American History” (1893). DeVoto and other western historians championed Turner’s thesis celebrating westward expansion and Manifest Destiny at the same time that Julia Child was living in France, writing Mastering the Art of French Cooking (1961). While a group of heterosexual males were reviving Turner’s account of the US frontier experience in the mid–twentieth century, Child—along with a cohort of female collaborators and several gay men writing back home in the states—were positing a non-isolationist vision of America and a more cosmopolitan imaginary of its relations with other countries. They did so by writing about seemingly apolitical subjects, such as European cuisine and mixed alcoholic concoctions called “cocktails,” during World War II and the Cold War era.

Slowing human aging with pharmaceuticals is now recognized as a feasible strategy. However, the design of clinical trials is still focused on single drug approaches. The process of aging has multiple pathways, which no current drug has been shown to effectively target. Therefore, it is of interest to study combinations, or cocktails, of drugs. A recently published article reported that a drug cocktail of rapamycin, acarbose and phenylbutyrate slowed aging in middle-aged mice treated for three months. The impact of this report is discussed, with the implications for determining endpoints in humans for testing drug cocktails as well as testing other drug combinations. Geroscience is based on the concept that therapeutic approaches that work for aging will also work for age-related diseases, including neurodegenerative diseases such as Alzheimer’s disease (AD). Single drugs have been ineffective in treating AD, so it seems reasonable to consider using multiple drugs in combination (cocktails) for a more effective treatment approach. However, initial screening of drug cocktails in animal models is costly and time-consuming. The human neuroblastoma cell line SH-SY5Y has neuronal properties and can be stressed with chemicals or transfected with adeno-associated virus (AAV) Aβ and/or pTau vectors for an in vitro model of neurotoxicity. Drug cocktails can then be easily screened for intervention efficacy compared to each individual drug in the cocktail.

AbstractFreshwater salinization is an emerging global problem impacting safe drinking water, ecosystem health and biodiversity, infrastructure corrosion, and food production. Freshwater salinization originates from diverse anthropogenic and geologic sources including road salts, human-accelerated weathering, sewage, urban construction, fertilizer, mine drainage, resource extraction, water softeners, saltwater intrusion, and evaporative concentration of ions due to hydrologic alterations and climate change. The complex interrelationships between salt ions and chemical, biological, and geologic parameters and consequences on the natural, social, and built environment are called Freshwater Salinization Syndrome (FSS). Here, we provide a comprehensive overview of salinization issues (past, present, and future), and we investigate drivers and solutions. We analyze the expanding global magnitude and scope of FSS including its discovery in humid regions, connections to human-accelerated weathering and mobilization of ‘chemical cocktails.’ We also present data illustrating: (1) increasing trends in salt ion concentrations in some of the world’s major freshwaters, including critical drinking water supplies; (2) decreasing trends in nutrient concentrations in rivers due to regulations but increasing trends in salinization, which have been due to lack of adequate management and regulations; (3) regional trends in atmospheric deposition of salt ions and storage of salt ions in soils and groundwater, and (4) applications of specific conductance as a proxy for tracking sources and concentrations of groups of elements in freshwaters. We prioritize FSS research needs related to better understanding: (1) effects of saltwater intrusion on ecosystem processes, (2) potential health risks from groundwater contamination of home wells, (3) potential risks to clean and safe drinking water sources, (4) economic and safety impacts of infrastructure corrosion, (5) alteration of biodiversity and ecosystem functions, and (6) application of high-frequency sensors in state-of-the art monitoring and management. We evaluate management solutions using a watershed approach spanning air, land, and water to explore variations in sources, fate and transport of different salt ions (e.g. monitoring of atmospheric deposition of ions, stormwater management, groundwater remediation, and managing road runoff). We also identify tradeoffs in management approaches such as unanticipated retention and release of chemical cocktails from urban stormwater management best management practices (BMPs) and unintended consequences of alternative deicers on water quality. Overall, we show that FSS has direct and indirect effects on mobilization of diverse chemical cocktails of ions, metals, nutrients, organics, and radionuclides in freshwaters with mounting impacts. Our comprehensive review suggests what could happen if FSS were not managed into the future and evaluates strategies for reducing increasing risks to clean and safe drinking water, human health, costly infrastructure, biodiversity, and critical ecosystem services.

Meanwhile, the term bartender refers to individuals who are able to translate and realize these techniques and concepts practically. Usually a bartender plays a direct role at the bar, serving and interacting with customers, and if necessary, interpreting customer needs regarding the desired drink.

Loss of physical performance, as seen in humans by decreased grip strength and overall physical fitness, is generally accepted to be a consequence of aging. Treatments to delay or reduce these changes or increase resilience to them are generally not available. In this preliminary study, 20-month-old male and female C57BL/6 mice were given either a standard mouse diet or a formulated mouse diet containing rapamycin (14 ppm), acarbose (1000 ppm), and phenylbutyrate (1000 ppm), or a diet containing one half dose of each drug, for 3 months. At the end of the study, performance on a rotarod and grip strength test was compared. In general, mice fed the full dose drug cocktail diet performed better on these assays, with significant improvements in rotarod performance in females fed the full dose cocktail and in grip strength in males fed the full dose cocktail, and females fed the low dose cocktail. These observations provide support for the concept that short term treatment with a cocktail of drugs that targets multiple aging pathways can increase resilience to aging, and suggests that this prototype cocktail could be part of a clinical therapeutic strategy for delaying age-related loss of physical performance in people. Although fatty acids enhance preadipocyte differentiation in the presence of adequate hormone cocktails, little is known regarding their effects in the absence of these hormones. We have now shown that palmitate, a common long-chain saturated fatty acid, induced apoptosis in both mouse 3T3-L1 and rat primary preadipocytes grown in a normal serum-containing medium. Treatment of preadipocytes with palmitate induced multiple endoplasmic reticulum (ER) stress responses, evidenced by increased protein content of CHOP and GRP78 and splicing of XBP-1 mRNA, as well as altered phosphorylation of eIF2α and increased phosphorylation of JNK and Erk1/2. Intriguingly, palmitate induced an early activation of Akt but diminished both Akt activation and its protein mass after prolonged incubation (>6 h). In association with these changes, palmitate reduced expression of β-catenin and its downstream target, c-Myc and cyclin D1, two key prosurvival proteins. Overexpression of constitutively active Akt did not block the apoptotic effect of palmitate. Cotreatment with unsaturated fatty acids (oleate, linoleate) or with LiCl (a glycogen synthase kinase-3β inhibitor) attenuated the palmitate-induced apoptosis. Subsequent analysis suggested that the unsaturated fatty acids probably counteracted palmitate by reducing, not eliminating, ER stress, whereas LiCl probably improved viability by activating the Wnt signaling pathway. Cotreatment of palmitate with a standard adipogenic hormone cocktail also abolished the apoptotic effect and promoted adipocyte differentiation. Collectively, our results suggest that palmitate causes multiple cellular stresses that may lead to apoptosis in preadipocytes in the absence of adipogenic stimuli, highlighting the importance of exogenous hormones in directing cell fate in response to increased fatty acid influx.

I last saw my friend Harun Farocki a few days before the opening of his exhibition at the Hamburger Bahnhof in late January 2014. Astonishingly, this was his first major one-person show in Berlin, a city that he called home and that had shaped his intellectual and artistic sensibility for over half a century. “I should have been born in Berlin,” he muses in his autobiographical “Written Trailers” (2009). Farocki was initially drawn to West Berlin in the early 1960s because the island city had been spared the Wirtschaftswunder (economic miracle) of the 1950s that had reshaped the rest of West Germany. It retained a forlorn rawness, a sense of bohemia, and a countercultural public sphere that attracted hippies, draft dodgers, political outcasts, and artists of all kinds. Farocki was a member of the first Deutsche Film und Fernsehakademie (Berlin Film Academy) class, along with Helke Sander, Holger Meins, and Wolfgang Petersen. He lived in a commune, wrote criticism, and produced relatively obscure agitprop films such as Herstellung eines Molotow-Cocktails (How to Make a Molotov Cocktail) (1968), Anleitung, Polizisten den Helm abzurissen (How to Remove a Police Helmet) (1969), and the better-known Nicht löschbares Feuer (Inextinguishable Fire) (1969). As Berlin changed over the years, however, so, too, did Farocki and his filmmaking practice.

In 1959, the year the Stratford Globe and Mail playwriting competition started receiving submissions from across the country, the Stratford Festival was already being labelled a disaster area by Antony Ferry of the Toronto Star. Arguing that all hopes for a true Canadian theatre had been buried there, he predicted that the theatre would die when It had done Shakespeare’s major works. While this may have enlivened some cocktail conversations, it was fundamentally incorrect for two important reasons: the first being the newly formed Canada Council, which pulled Stratford out of a deficit season in 1959 with a grant of $50,000; the second was the barely perceptible but soon-to-be-realized shift in the theatre’s mandate. For it was in 1959 that the winners of the Globe and Mail contest were announced, and new Canadian work was promised its first audience at Stratford.

Abstract Introduction Recent developments of novel immunotherapeutic drugs have shown promising results for patients with hematologic malignancies, however, an unmet need for accurate and specific biomarkers persists. To address this need, we developed a novel integrative analysis procedure for the automated analysis of multidimensional flow cytometry data obtained from the peripheral blood of patients with chronic lymphocytic leukemia (CLL). State of the art flow cytometry analysis is accomplished by manual sequential segmentation, or gating, of cell populations based on similarities in fluorescence and light scatter characteristics through visualization of the data in one- or two-dimensional plots. This approach has a number of limitations, including the subjective nature of the gating and the inability to fully utilize the high-dimensional data. Recent efforts have produced sophisticated computational methods that overcome many of these limitations; however, these newer computational methods have not been rigorously tested in a clinical context and have focused on the rigorous and automated analysis of samples from individual patients, with substantially less effort towards the analysis of patient populations. The ultimate goal of our analysis is to develop computational approaches that will enable an identification of subsets of patients with distinct immunological markers. Methods We developed a novel analysis framework that facilitates automated identification of both common cell types and patient population subgroups, based on post-processing of individual sample analysis with the FLOCK program. FLOCK identifies clusters of putatively similar cells in an individual sample by multidimensional clustering of the fluorescence marker and light-scattering measurements. We developed a rigorous hierarchical clustering approach to identify common “cell signatures” across multiple patients. The cell signatures were then mapped back onto the individual patient samples and used in a second clustering that identified patient subgroups based on similar abundances of specific cell types. Results We used our analytic framework to analyze multidimensional flow cytometry data (26 cell surface markers in 4 different antibody cocktails) from peripheral blood specimens of a heterogeneous group of 55 CLL patients and 13 healthy controls. Our analysis revealed distinct differences between controls and CLL patients. Analyzing the non-malignant peripheral blood cell types, we were furthermore able to differentiate between distinct clinical subpopulations of patients (e.g. identify treatment-naïve patients from those that had previously undergone chemotherapy). Conclusion/Discussion Using a novel integrative analysis procedure to analyze complex flow cytometry data of the peripheral blood from CLL patients, we are able to identify distinct cell type distributions. We propose that this information is a marker for the overall health/disease status of the corresponding patient, and could ultimately be used for diagnosis, prognosis, and selection of optimal treatment. In the context of multiple novel treatment options for CLL patients, such a tool will be crucial for defining individual patient prognosis, and defining an accurately matched treatment plan. The treatment of acutely disturbed patients is a stressful area of psychiatric practice. Castle et al (1994) found that MRCPsych Part II trainees expressed a general dissatisfaction with training in the management of violence, and in our experience junior trainees are even less confident in the treatment of the disturbed patient. A survey by Cunnane (1994) found that no consensus exists among consultant psychiatrists on which drug treatment is most appropriate for the management of the aggressive, acutely disturbed psychotic patient. There was uncertainty as to optimal management, low expectation of quick results, unwillingness to express an opinion and the use of drug cocktails of dubious rationale.

This review presents an overview of a challenging problem in auditory perception, the cocktail party phenomenon, the delineation of which goes back to a classic paper by Cherry in 1953. In this review, we address the following issues: (1) human auditory scene analysis, which is a general process carried out by the auditory system of a human listener; (2) insight into auditory perception, which is derived from Marr's vision theory; (3) computational auditory scene analysis, which focuses on specific approaches aimed at solving the machine cocktail party problem; (4) active audition, the proposal for which is motivated by analogy with active vision, and (5) discussion of brain theory and independent component analysis, on the one hand, and correlative neural firing, on the other.

Previous studies indicated that the use of a phage cocktail, composed of bacteriophages vB_SenM-2 and vB_Sen-TO17, is effective in killing cells of Salmonella enterica serovars Typhimurium and Enteritidis in vitro and in the Galleria mellonella animal model as efficiently as antibiotics (enrofloxacin or colistin) and induced fewer deleterious changes in immune responses. Here, we investigated the effects of this phage cocktail on the hematological parameters and selected biochemical markers in chickens infected with S. enterica serovar Typhimurium, in comparison to those caused by enrofloxacin or colistin. We found that treatment with antibiotics (especially with enrofloxacin) caused nonbeneficial effects on red blood cell parameters, including hematocrit, MCV, MCH, and MCHC. However, Salmonella-induced changes in the aforementioned parameters were normalized by the use of the phage cocktail. Importantly, hepatotoxicity was suggested to be induced by both antibiotics on the basis of increased alanine transaminase (ALT) and aspartate aminotransferase (AST) activities, in contrast to the phage cocktail, which did not influence these enzymes. We conclude that phage therapy with the cocktail of vB_SenM-2 and vB_Sen-TO17 in Salmonella-infected chickens is not only as effective as antibiotics but also significantly safer for the birds than enrofloxacin and colistin.

Cellulose is the most abundant polysaccharide in lignocellulosic biomass, where it is interlinked with lignin and hemicellulose. Bioethanol can be produced from biomass. Since breaking down biomass is difficult, cellulose-active enzymes secreted by filamentous fungi play an important role in degrading recalcitrant lignocellulosic biomass. We characterized a cellobiohydrolase (AfCel6A) and lytic polysaccharide monooxygenase LPMO (AfAA9_B) from Aspergillus fumigatus after they were expressed in Pichia pastoris and purified. The biochemical parameters suggested that the enzymes were stable; the optimal temperature was ~60 °C. Further characterization revealed high turnover numbers (kcat of 147.9 s−1 and 0.64 s−1, respectively). Surprisingly, when combined, AfCel6A and AfAA9_B did not act synergistically. AfCel6A and AfAA9_B association inhibited AfCel6A activity, an outcome that needs to be further investigated. However, AfCel6A or AfAA9_B addition boosted the enzymatic saccharification activity of a cellulase cocktail and the activity of cellulase Af-EGL7. Enzymatic cocktail supplementation with AfCel6A or AfAA9_B boosted the yield of fermentable sugars from complex substrates, especially sugarcane exploded bagasse, by up to 95%. The synergism between the cellulase cocktail and AfAA9_B was enzyme- and substrate-specific, which suggests a specific enzymatic cocktail for each biomass by up to 95%. Cancer is a complex devastating disease with enormous treatment challenges, including chemo- and radiotherapeutic resistance. Combination therapy demonstrated a promising strategy to target hard-to-treat cancers and sensitize cancer cells to conventional anti-cancer drugs such as doxorubicin. This study aimed to establish molecular profiling and therapeutic efficacy assessment of chloroquine and/or tioconazole (TIC) combination with doxorubicin (DOX) as anew combination model in MCF-7 breast cancer. The drugs are tested against apoptotic/autophagic pathways and related redox status. Molecular docking revealed that chloroquine (CQ) and TIC could be potential PI3K and ATG4B pathway inhibitors. Combination therapy significantly inhibited cancer cell viability, PI3K/AkT/mTOR pathway, and tumor-supporting autophagic flux, however, induced apoptotic pathways and altered nuclear genotoxic feature. Our data revealed that the combination cocktail therapy markedly inhibited tumor proliferation marker (KI-67) and cell growth, along with the accumulation of autophagosomes and elevation of LC3-II and p62 levels indicated autophagic flux blockage and increased apoptosis. Additionally, CQ and/or TIC combination therapy with DOX exerts its activity on the redox balance of cancer cells mediated ROS-dependent apoptosis induction achieved by GPX3 suppression. Besides, Autophagy inhibition causes moderately upregulation in ATGs 5,7 redundant proteins strengthened combinations induced apoptosis, whereas inhibition of PI3K/AKT/mTOR pathway with Beclin-1 upregulation leading to cytodestructive autophagy with overcome drug resistance effectively in curing cancer. Notably, the tumor growth inhibition and various antioxidant effects were observed in vivo. These results suggest CQ and/or TIC combination with DOX could act as effective cocktail therapy targeting autophagy and PI3K/AKT/mTOR pathways in MCF-7 breast cancer cells and hence, sensitizes cancer cells to doxorubicin treatment and combat its toxicity.

ABSTRACT Background Development of successful neutralizing antibodies is dependent upon broad epitope coverage to increase the likelihood of achieving therapeutic function. Recent advances in synthetic biology have allowed us to conduct an epitope binning study on a large panel of antibodies identified to bind to Ebola virus glycoprotein with only published sequences. Methods and Results A rapid, first-pass epitope binning experiment revealed seven distinct epitope families that overlapped with known structural epitopes from the literature. A focused set of antibodies was selected from representative clones per bin to guide a second-pass binning that revealed previously unassigned epitopes, confirmed epitopes known to be associated with neutralizing antibodies, and demonstrated asymmetric blocking of EBOV GP from allosteric effectors reported from literature. Conclusions Critically, this workflow allows us to probe the epitope landscape of EBOV GP without any prior structural knowledge of the antigen or structural benchmark clones. Incorporating epitope binning on hundreds of antibodies during early stage antibody characterization ensures access to a library’s full epitope coverage, aids in the identification of high quality reagents within the library that recapitulate this diversity for use in other studies, and ultimately enables the rational development of therapeutic cocktails that take advantage of multiple mechanisms of action such as cooperative synergistic effects to enhance neutralization function and minimize the risk of mutagenic escape. The use of high-throughput epitope binning during new outbreaks such as the current COVID-19 pandemic is particularly useful in accelerating timelines due to the large amount of information gained in a single experiment.

ABSTRACT“Welcome to Caesars, I am your slave” intoned toga‐clad cocktail waitresses wearing high‐heeled sandals on the opening day of this stately pleasure dome. This single sentence typified the intent of the creator and builder of this sumptuous development, Jay Sarno (1922‐1984). He dreamed of building a casino‐hotel that afforded every “reveler”—a unique gaming experience: posh accommodations, fine dining, star‐studded entertainment, and up‐scale shopping. Sarno chose replicas of the world's most famous art to adorn his creation. This paper analyzes the realization of his dream through the theory of two philosophers fascinated by the reproduction of images. Roland Barthes (1915‐1980) elaborated upon the objective nature of art and what the viewer brings to it. Walter Benjamin (1892‐1940) developed the notion of phantasmagoria to describe the illusions of sound and light shows in Paris, an apt metaphor for this establishment in Paradise. Benjamin's thoughts on lithographs illuminate the characteristics of the art in Caesars: accessibility, flexibility, decorativeness, and anonymity. These attributes transformed this casino‐resort into an icon of popular culture. The authors take a deeper look into the simulacrum, pastiche, and theme of a place that caters to men's wants and desires. The authors also discuss the Forum, an extension of Caesars Entertainment Corporation's holdings. The Forum provides an opportunity for consumers to shop without guilt for the most exclusive products the world has to offer in a Roman themed environment. The authors conclude with an analysis of the Forum as a cathedral of consumption, an entrepreneur's dream where people pay, not for the intrinsic worth of goods and services; but for the status attached to them.

We present a new approach to the cocktail party problem that uses a cortronic artificial neural network architecture (Hecht-Nielsen, 1998) as the front end of a speech processing system. Our approach is novel in three important respects. First, our method assumes and exploits detailed knowledge of the signals we wish to attend to in the cocktail party environment. Second, our goal is to provide preprocessing in advance of a pattern recognition system rather than to separate one or more of the mixed sources explicitly. Third, the neural network model we employ is more biologically feasible than are most other approaches to the cocktail party problem. Although the focus here is on the cocktail party problem, the method presented in this study can be applied to other areas of information processing.

Abstract Breast cancer is frequently characterized by calcifications in mammography. Although breast calcifications were previously thought to be produced passively as the final stage of cell regression, recent studies have shown active production. In another context, vascular smooth muscle cells have been found to produce vascular calcifications by acquiring osteoblast-like characteristics. The same mechanism has been suggested to account for breast calcifications. A report showed that breast calcifications are clearly generated by the mouse breast cancer cell line 4T1 through osteoblast-like characteristics. However, the details underlying such a mechanism of mammary calcification is unknown. Understanding this mechanism will improve diagnostic accuracy for breast cancer. We induced calcifications using osteogenic cocktail on the human breast cancer cell line MDA-MB-231. Our hypothesis was that breast cancer calcifications are generated in a process similar to that of osteoblast differentiation. By a comprehensive analysis of transcription compared with osteoblastic cell line MC3T3-E1 in a microarray, we showed that placental alkaline phosphatase (PLAP) is involved in producing calcifications through PI3K-Akt signaling pathways. Herein, we demonstrated that calcifications occur and that alkaline phosphatase enzyme activity increases in MDA-MB-231 cells cultured using an osteogenic cocktail-containing medium. Microarray transcript analysis showed that the PI3K-Akt signaling pathway was significantly involved, with recruitment of placental alkaline phosphatase. Calcifications and alkaline phosphatase enzyme activity were suppressed by silencing placental alkaline phosphatase using a small interfering RNA. Inhibition of the PI3K-Akt signaling pathway suppressed phospho-c-Jun and placental alkaline phosphatase and resulted in absence of calcifications. These findings reveal that breast cancer cells acquire alkaline phosphatase enzyme activity via placental alkaline phosphatase expression and suggest that breast calcification formation is closely associated with the PI3K-Akt signaling pathway. Our results suggest that PLAP produces ALP enzyme activity required for calcification in MDA-MB-231 cells. PLAP is clinically expressed in some breast cancers. A report showed that dexamethasone induces PLAP enzyme activity and mRNA in the human breast cancer BC-M1 cell line. Therefore, dexamethasone in osteogenic cocktail may be associated with PLAP expression in the MDA-MB-231 cells. On the other hand, c-Jun binds to the promoter region of ALPP in MDA-MB-231, and we showed that c-Jun phosphorylation appears in MDA-MB-231 cells cultured using osteogenic cocktail. We suggest that PLAP transcript would be activated when phospho-c-Jun enters the nucleus and binds to the promoter region of PLAP. Our proposed mechanism in breast calcifications is that osteogenic cocktail induces c-Jun phosphorylation via PI3K-Akt signaling pathway and that PLAP transcription would be activated when phospho-c-Jun enters the nucleus and binds to the promoter region of PLAP. Future research might assess whether increased PLAP levels are the main driver of intracellular calcifications in breast cancer, as well as whether the PI3K-Akt signaling pathway has a similar effect in vivo that we demonstrated to cause calcifications in vitro. MicroRNA (miRNA)-based therapies are an emerging class of targeted therapeutics with many potential applications. Ewing Sarcoma patients could benefit dramatically from personalized miRNA therapy due to inter-patient heterogeneity and a lack of druggable (to this point) targets. However, because of the broad effects miRNAs may have on different cells and tissues, trials of miRNA therapies have struggled due to severe toxicity and unanticipated immune response. In order to overcome this hurdle, a network science-based approach is well-equipped to evaluate and identify miRNA candidates and combinations of candidates for the repression of key oncogenic targets while avoiding repression of essential housekeeping genes. We first characterized 6 Ewing sarcoma cell lines using mRNA sequencing. We then estimated a measure of tumor state, which we term network potential, based on both the mRNA gene expression and the underlying protein-protein interaction network in the tumor. Next, we ranked mRNA targets based on their contribution to network potential. We then identified miRNAs and combinations of miRNAs that preferentially act to repress mRNA targets with the greatest influence on network potential. Our analysis identified TRIM25, APP, ELAV1, RNF4, and HNRNPL as ideal mRNA targets for Ewing sarcoma therapy. Using predicted miRNA-mRNA target mappings, we identified miR-3613-3p, let-7a-3p, miR-300, miR-424-5p, and let-7b-3p as candidate optimal miRNAs for preferential repression of these targets. Ultimately, our work, as exemplified in the case of Ewing sarcoma, describes a novel pipeline by which personalized miRNA cocktails can be designed to maximally perturb gene networks contributing to cancer progression.

And since I had to work, I might as well contribute to the common weal, and do nothing that could harm the social fabric. After several years of experimenting with social work, sales, bookkeeping, cocktail waitressing, organizing Camp Fire Girls, census-taking, and other sundry occupations, it was finally librarianship that demanded the most of my top-notch liberal arts education and my desire to do good in a world so complex that it was often impossible to know what was good from what was not.

The overproduction of eumelanin leads to a panel of unaesthetic hyper-pigmented skin diseases, including melasma and age spots. The treatment of these diseases often requires the use of tyrosinase inhibitors, which act as skin whitening agents by inhibiting the synthesis of eumelanin, with harmful side effects. We report here that laccase from Trametes versicolor in association with a cocktail of natural phenol redox mediators efficiently degraded eumelanin from Sepia officinalis, offering an alternative procedure to traditional whitening agents. Redox mediators showed a synergistic effect with respect to their single-mediator counterpart, highlighting the beneficial role of the cocktail system. The pro-oxidant DHICA sub-units of eumelanin were degraded better than the DHI counterpart, as monitored by the formation of pyrrole-2,3,5-tricarboxylic acid (PTCA) and pyrrole-2,3-dicarboxylic acid (PDCA) degradation products. The most effective laccase-mediated cocktail system was successively applied in a two-component prototype of a topical whitening cream, showing high degradative efficacy against eumelanin.

We study the cocktail party problem and propose a novel attention network called Tune-In, abbreviated for training under negative environments with interference. It firstly learns two separate spaces of speaker-knowledge and speech-stimuli based on a shared feature space, where a new block structure is designed as the building block for all spaces, and then cooperatively solves different tasks. Between the two spaces, information is cast towards each other via a novel cross- and dual-attention mechanism, mimicking the bottom-up and top-down processes of a human's cocktail party effect. It turns out that substantially discriminative and generalizable speaker representations can be learnt in severely interfered conditions via our self-supervised training. The experimental results verify this seeming paradox. The learnt speaker embedding has superior discriminative power than a standard speaker verification method; meanwhile, Tune-In achieves remarkably better speech separation performances in terms of SI-SNRi and SDRi consistently in all test modes, and especially at lower memory and computational consumption, than state-of-the-art benchmark systems.

A Cuban cocktail called mulata inspires an examination of the mulata body. Beyond an analysis of the cocktail as a commercial commodity, the mulata body can be placed within an intercultural space shaped by the processes of colonization, slavery and race relations. By examining the grammars in the mulata cocktail, the discussion moves the subject through other texts and discourses in order to mediate the mulata's embodied genealogy as a form of transculturation. As a hybrid body that inhabits a ‘racialized’ performativity, the mulata's subaltern agency is imagined beyond the exoticism charged to its presence in the Latin American and Caribbean contexts. A closer look at the mulata body helps to trace not only the process of objecthood affected by masculinist power and desire, but also by the way the process of subjecthood is performatively achieved.

The present paper explores the immunomodulating, prophylactic, and therapeutic potential of Ganga water. Ganga water is reported to have 200 isolates of phages which is ten times more than Yamuna and Narmada. This diversity of phages suggests that these may act as a wide-spectrum phage cocktail. Forty-five Volunteers administered Ganga water to themselves and assessed the change in their condition on a 1-10 Likert Scale on ten morbidities. Pre-screening or pre-selection was not done. The results were sorted according to 10 morbidities and changes in the self-assessment were noted. The results indicate that Ganga water provides health benefits for a number of morbidities simultaneously and could work as a wide-spectrum phage cocktail.

Water of Ganga river is reported to have more than 200 isolates of phages. This study has used the naturally available cocktail of phages in the Ganga water as a treatment for chronic Psoriasis. In the conventional Phage Therapy (PT) phages that are active against specific bacteria are first identified; then isolated, multiplied and administered to the patient. We have made a novel innovation of administering the naturally available cocktail of phages in the water of the Ganga river without first identifying the target bacteria and isolating specific phages that may be active against them. In doing so, we enable the large numbers of phages to self-identify the bacteria that are present and act against them. This approach shortcuts the tortuous process of conventional PT. Further, the phages act against a number of bacteria simultaneously and provide good results in psoriasis which has multiple causes.Patients who took Ganga water for only 2 weeks showed benefit but the benefit did not sustain after stoppage of the treatment and the disease relapsed to the pre-treatment levels. The same patients showed sustained benefit after they took Ganga water for four weeks. Conclusion is that Ganga water can be used for therapeutic purposes as long as the treatment is continued for at least four weeks. The study underscores the need to establish more robust protocol for treatment of dermatological and possibly other diseases with the cocktail of phages available in the waters of the Ganga river.

The actin cytoskeleton is regulated by factors that influence polymer assembly, disassembly, and network rearrangement. Drugs that inhibit these events have been used to test the role of actin dynamics in a wide range of cellular processes. Previous methods of arresting actin rearrangements take minutes to act and work well in some contexts, but can lead to significant actin reorganization in cells with rapid actin dynamics, such as neutrophils. In this paper, we report a pharmacological cocktail that not only arrests actin dynamics but also preserves the structure of the existing actin network in neutrophil-like HL-60 cells, human fibrosarcoma HT1080 cells, and mouse NIH 3T3 fibroblast cells. Our cocktail induces an arrest of actin dynamics that initiates within seconds and persists for longer than 10 min, during which time cells maintain their responsivity to external stimuli. With this cocktail, we demonstrate that actin dynamics, and not simply morphological polarity or actin accumulation at the leading edge, are required for the spatial persistence of Rac activation in HL-60 cells. Our drug combination preserves the structure of the existing cytoskeleton while blocking actin assembly, disassembly, and rearrangement, and should prove useful for investigating the role of actin dynamics in a wide range of cellular signaling contexts.

Abstract SARS-CoV-2 gains entry to human cells through its spike (S) protein binding to angiotensin-converting enzyme 2 (ACE2). Therefore, the receptor binding domain (RBD) of the S protein is the primary target for neutralizing antibodies. Selection of broad-neutralizing antibodies against SARS-CoV-2 and SARS-CoV is attractive and might be useful for treating not only COVID-19 but also future SARS-related CoV infections. Broad-neutralizing antibodies, such as 47D11, S309, and VHH-72, have been reported to target a conserved region in the RBD of the S1 subunit. The S2 subunit required for viral membrane fusion might be another target. Due to their small size and high stability, single-domain antibodies might have the ability to be administered by an inhaler making them potentially attractive therapeutics for respiratory infections. A cocktail strategy combining two (or more) antibodies that recognize different parts of the viral surface that interact with human cells might be the most effective.

Salmonella Enteritidis (SE) is one of the main serovar of Salmonella enterica involved in foodborne infections. When intestinal infection by SE requires treatments using antimicrobial, cases can be aggravated if the cause bacteria are resistant to the drugs used. A possible alternative to control these antimicrobial resistant bacteria is phagotherapy, which is characterized by the use of bacteriophages that will lead to the lysis of the target bacteria. The objective was to evaluate the in vitro efficacy of a bacteriophage cocktail for the inactivation of SE in fecal fermentation. Fecal samples were collected from healthy donors and the fecal fermentation preparation was carried out. For the tests, a pool with three different SE isolates was used, and the bacteriophage cocktail was elaborated with the phages UPF_BP1, UPF_BP2 and UPF_BP3. Four different treatments were evaluated: the group called ASF, where the action of the bacteriophage cocktail against a pool of SE in fecal fermentation was tested; the CP1 group (positive control 1), in which only SE was inoculated in fecal fermentation; the CP2 group (positive control 2), in which the bacteriophage cocktail was inoculated in the fecal fermentation; and the CN group (negative control), containing only fecal fermentation. In all treatments the quantification of SE was performed, and in the ASF group, the quantification of phages was also performed. In the CP1 group there was a growth of 3,76 log10 UFC/mL, while in the ASF group there was a decrease in Salmonella showing 0,77 log10 UFC/mL. The results showed that the use of the bacteriophage cocktail against SE in fecal fermentation was able to reduce the amount of Salmonella in the sample. Despite not showing a significant difference (p=0.059), this in vitro sudy demonstrates the ability of phages to act against Salmonella in fecal fermentation, bringing positive evidence that corroborates the continuity of the study for future uses of phage therapy.

To date, about 37 million people are living with the human immunodeficiency virus (HIV) and an estimated 680,000 people have died from acquired immune deficiency syndrome (AIDS) related illnesses globally. While all countries have been impacted by HIV, some have been significantly more impacted than others, particularly countries in sub-Saharan Africa. The purpose of this paper was to identify progress made in HIV prevention globally, particularly in the areas of voluntary counseling and testing (VCT) uptake, access to antiretroviral therapy (ART), and HIV-related stigma. With the development of ART, a cocktail of medications for the treatment of HIV, VCT uptake increased, as it became apparent that the medication would only be prescribed after an HIV diagnosis through testing. Widely considered a critical gateway to HIV prevention and treatment, VCT is being implemented in many countries, and as a result, about 38 million people living with HIV in 2018 had access to ART. Regardless of this success, major challenges still remain. We did an electronic search of 135 articles in English related to global HIV progress and challenges indexed in PubMed, ResearchGate, Google, and other search engines from 1998 to 2021. Sixty articles met the inclusion criteria for this paper. Data on trends in ART coverage were obtained from the Joint United Nations Programme on HIV/AIDS (UNAIDS) website. These data were used to show ART coverage globally in World Health Organization (WHO) regions. It was found that while global successes have been chalked in the areas of VCT uptake and ART coverage, HIV-related stigma has impeded greater success. This paper summarizes and discusses global successes and challenges in HIV prevention efforts in the past four decades with a focus on VCT, ART, and HIV-related stigma.

This is widely, though with much exaggeration, conceived to be the major prop of authoritarian government. It is also, more correctly, seen as hostile to the idea of human rights possessed by persons against governments. In one sense, the decision to incorporate the European Convention on Human Rights into UK domestic law is an authoritative repudiation of the Diceyian heritage.

This essay makes the first sustained study of the Okinawan artist Makishi Tsutomu (1941–2015) who used American Pop Art vocabularies to describe the complex realities of US-occupied Okinawa. Focusing on his 1972 installation Commemorating the Reversion to the Great Empire of Japan, the essay examines the critical ambivalence of Makishi's Political Pop as a translation strategy. Despite his critique of both American and Japanese imperialism, Makishi was aware that Okinawa was inseparably entangled in it, especially in the context of the Vietnam War, which brought violence, but also economic benefits, to Okinawa. Despite his use of the American Pop idiom as a new lingua franca for contemporary art, Makishi's work did not reach either mainland or international audiences as the artist exhibited almost exclusively in Okinawa. By comparing Makishi's artistic strategies with those of a representative Okinawan novelist, Ōshiro Tatsuhiro, especially as articulated in his 1967 novella The Cocktail Party, the essay situates the significance of Makishi's project within the emerging discourse on the global neo-avant-garde.
