Resumen
Cannabinoid (CB) receptors have been found in most vertebrates that have been studied. The location of various CB receptors in the body and brain are known, but their physiological functions are not fully understood. The effects CBs have on the cardiovascular system have been of growing interest in recent years. Increasing reports from emergency departments and law enforcement agencies detail acute cardiovascular and psychological effects from synthetic CB intoxication, such as JWH-018. This major health concern is substantiated by governmental agencies like the CDC and NIDA. This pilot study investigates the acute toxic effects of the synthetic CB, JWH-018, on the cardiovascular and neuroendocrine systems in Ictalurus punctatus (channel catfish). Research in organisms besides the traditional mammal models can provide new insights into CB function and physiology. Ictalurus punctatus lend multiple benefits as a model organism that permits researchers to investigate in vivo effects of both cardiovascular and neuroendocrine systems without much influence from traditional sampling methods, and further more provide ample size and tissue to perform specific cardiovascular experiments. Multiple methods were used to assess cardiovascular function and sympathetic nervous system activation. Two different doses, low (500 µg/kg) and high 1,500 µg/kg, of JWH-018 were evaluated in the study. Delivery of JWH-018, via dorsal aorta cannulation, was administered to channel catfish in order to measure cardiovascular functions and sample blood. Plasma levels of the hypothalamus-pituitary-adrenal/interrenal (HPA/I) biomarkers; ACTH, cortisol, epinephrine, and norepinephrine, were measured using ELISAs. Myocardial and neural tissue was collected after the exposures for rt-PCR analysis on β2 adrenergic and glucocorticoid receptor density change. Acute exposure of JWH-018 in undisturbed channel catfish yielded several findings: (1) High dose of JWH-018 was responsible for cardio depressor effects in catfish with a tendency to produce tachycardia, (2) rt-PCR results showed a 2.7 fold increase of glucocorticoid receptor mRNA density in catfish cardiomyocytes when exposed to each dose of JWH-018, (3) Catfish plasma ACTH levels were increased with high doses of JWH-018, while plasma cortisol was increased by low doses. Channel catfish is an excellent animal model to examine the effects of synthetic cannabinoids and cardiovascular function. Acute exposures to high levels of JWH-018 appear to produce cardiovascular dysfunction providing evidence that substantiates emergency department reports, in addition yields novel information about the interaction of CBs exposure and the increase of glucocorticoid receptors levels on cardiomyocytes. The channel catfish is a new animal model that can aid in further investigations of CB exposure and multiple physiological functions for health and toxicology studies. With relatively easy adjustments from this pilot study, the effects on CBs can be monitored on Ictalurus punctatus with confident results concerning human health.

The primary objective of this work was to determine the tissue-specific bioconcentration factors (BCFs) of the selected pharmaceuticals: norethindrone (NET), ibuprofen (IBU), verapamil (VER), clozapine (CLZ) and fenofibrate (FFB) in two freshwater fishes: fathead minnow and channel catfish. BCF tests on fathead followed the standard OECD 42-day test while a 14-day abridged test design was used in catfish exposures. Additional objectives included a) comparing the measured BCFs to the US EPA's BCFWIN model predicted values, b) comparing the BCF results from the standard and reduced tests, and c) prediction of chronic risk of the pharmaceuticals in fish using the human therapeutic plasma concentrations. Each test included uptake and depuration phases to measure tissue-specific kinetic BCFs. The results indicated that all the pharmaceuticals, except IBU, have the potential for accumulation in fish. Estimated BCFs for NET, VER and FFB may not be significant in view of the current regulatory trigger level (BCF ≥ 2000); however, CLZ's BCF in the liver had approached the criterion level. Significant differences were noticed in the tissue-specific uptake levels of the pharmaceuticals with the following general trend: (liver/kidney) > (gill/brain) > (heart/muscle) > plasma. IBU uptake was highest in the plasma. When compared to the measured BCFs, predicted values for NET, IBU, VER and FFB were slightly overestimated but did not differ largely. However, the measured BCF of CLZ in the liver was approximately two-orders of magnitude higher than the predicted level. The tissue-BCFs for the two species were not widely different indicating the potential usefulness of the reduced BCF test. Comparison of fish and human plasma levels indicated that NET, CLZ and VER have the potential to cause chronic effects in fish.

Channel catfish have pectoral spines that lock to defend against gape-limited predators such as largemouth bass. Previous work indicated that spines increase survival of channel catfish exposed to bass but did not determine whether bass avoid catfish if less dangerous species are available. We presented bass with channel catfish and two alternatives, bluegill and goldfish, and compared order of ingestion, ingestion time, percent of successful strikes, and time spent chasing each prey species. We also presented the three species in a jar study that standardized position in the water column as well as in a net-pen study. The order of ingestion was suggestive of a preference for goldfish, then bluegill and finally channel catfish. Handling time was greater for channel catfish, less for bluegill, and the least for goldfish. Fewer catfish were eaten when other prey were available. Bass appear to avoid channel catfish if alternative prey is available.

The use of fatty acid (FA) analysis has become an important tool in recent years to investigate a broad spectrum of questions in fisheries and aquatic ecology. One of these applications has been the use of FA profiles as biomarkers for indicating diet and recent feeding history of fishes and other aquatic consumers. Differences in forage FA profiles and spatial differences in food web structure within aquatic systems may lead to corresponding spatial patterns of FA profiles in consumers, thereby facilitating the potential applicability of FA biomarkers as indicators of habitat use and origin for mobile consumers such as fishes. However, little information is available regarding differences in FA profiles among habitat types in river-floodplain ecosystems and the time-scale over which fish FA profiles change when fish move between habitats that differ in the FA profiles of prey resources. The objectives of this study were to test whether the tissue FA profiles of channel catfish (Ictalurus punctatus) differed among three reaches of the lower Kaskaskia River and its floodplain lakes, to determine the time-course of tissue FA profile turnover when fish are relocated to a new habitat type, and to compare FA profiles among muscle, liver, and adipose fin tissues in channel catfish. Fatty acid profiles of channel catfish were significantly different among sites, especially between upper and lower river sites, and between river channel and oxbow lake sites, suggesting differing energy sources among habitats and river reaches in the Kaskaskia system. More specifically, there was a significant increase in the essential FAs, 18:2n-6 and 18:3n-3, in channel catfish with increasing distance downstream, which could reflect an increase in river-floodplain connectivity at downstream sites. Channel catfish transplanted from the Kaskaskia River to ponds at Southern Illinois University Carbondale showed a significant linear decrease in n-3 and medium-chain polyunsaturated FAs (MC-PUFA), and a significant linear increase in monounsaturates (MUFA) with longer residence time in a lentic environment. Liver tissue exhibited the fastest turnover rate of the three tissue types (< 2 weeks), with muscle and adipose fin tissue both displaying similar, longer turnover times (approximately 10 weeks). Results of this study support the use of FA profiles as indicators of energy sources for fishes in large river-floodplain ecosystems, including the potential for indentifying habitat-specific (river channel vs. floodplain lake) or river reach-specific energy sources. Additionally, results of this study provide a timeline for FA turnover in channel catfish tissues when fish move among habitats with distinct FA signatures, which is crucial for detecting temporal shifts in use of habitat-specific energy sources by channel catfish. Results also suggest that adipose fin tissue samples may be a less invasive alternative to muscle tissue for analysis of FA profiles in channel catfish. Improved understanding of which habitats or river reaches provide trophic support for riverine fishes through use of FA profile analysis has potentially important implications for habitat conservation and rehabilitation in river-floodplain ecosystem.

This study describes the population characteristics of channel catfish (Ictalurus punctatus) and flathead catfish (Pylodictis olivaris) in the lower 200 miles of the Wabash River in 1998. Densities of channel and flathead catfish did not differ over the study reach for either species. Electrofishing captured twice as many channel as flathead catfish, and hoop nets captured twice as many flatheads as channel catfish. Growth rates of channel catfish were fast as compared to documented growth in other rivers in the United States, with ages ranging from 1-8. Flathead catfish growth was average when compared to that reported in other United States rivers with ages ranging from 0-7. Length-frequency analysis for both species revealed the populations were comprised of small individuals with mortality higher than expected in older fish. This study will aid in the management of the Wabash River, as historic data are limited.

Acoustically, teleost swimbladders have been considered resonant underwater bubbles. Contrary results indicating that bladders are tuned less sharply than such a bubble have been explained by damping of surrounding fish tissue. Recent findings in toadfish and weakfish, however, suggest that the bladder is a highly damped structure and that the frequency of the fish sounds is deternlined as a forced response to sonic muscle movement rather than by resonance of the bladder. In this study I examined acoustics and material properties of swimbladders in Tilapia (an auditory generalist) and the channel catfish (an auditory specialist). The swimbladder was struck with a piezoelectric impact hammer. Amplitude and timing characteristics of bladder sound and displacement were compared for strikes of different amplitudes. Most of the first cycle of sound occurs during swimbladder compression, indicating that the bladder rapidly contracts and expands as force increases during the strike. Harder hits are shorter in duration generate a similar displacement duration with an increasing number of shorter cycles and a 12-15 dB increase in sound amplitude. The frequency spectrum is broad, and the dominant frequency is driven by the strike and not the natural frequency of the bladder. The displacement waveform varies between species catfish exhibit a greater structural stiffness and lower amplitude movement and higher sound amplitude for an equivalent hammer strike. Material properties (peak load, stress, strain, Young's modulus), fiber direction (catfish only), and structural stiffiless of bladders exhibit various patterns suggesting that the bladder walls are not uniform structures. Additionally thickness varies regionally. Notably Young's modulus in both species is similar despite large difference in stress and strain. The modulus of catfish bladder increased 1600-fold when dried. Finally the bladder of both species had a high water content averaging about 70%. These data suggest that viscous damping caused by water in the bladder wall is a major factor responsible for acoustic properties of the teleost swimbladder.

Channel Catfish (Ictalurus punctatus, Rafinesque, 1818) leukocyte immune-type receptors (IpLITRs) are a family of proteins sharing structural and phylogenetic relationships with mammalian immune receptors. Based on their predicted signalling potential and ligand-binding properties, IpLITRs may be important in the control of immune cell effector responses in fish. The main objectives of this thesis were to determine how stimulatory IpLITRs activate cells and to develop assays for the screening of IpLITR ligands. Using cellular transfections, coimmunoprecipitation, and flow cytometry, I determined that stimulatory IpLITRs associate with specific adaptor molecules, which is required for their surface expression and signalling ability. These adaptors assemble with IpLITRs via their charged transmembrane regions and contain cytoplasmic tails encoding tyrosines that may initiate kinase pathways leading to immune cell activation. This study represents the first step towards elucidating how IpLITRs turn on immune cells. Combined with the development of assays to identify IpLITR ligands, my work sets the stage for further investigations into the functional characterization of these receptors.
