ABSTRACT SecB, a protein export-specific chaperone, enhances the export of a subset of proteins across cytoplasmic membranes of Escherichia coli. Previous studies showed that the synthesis of SecB is repressed by the presence of glucose in the medium. The derepression of SecB requires the products of both the cya andcrp genes, indicating that secB expression is under the control of catabolic repression. In this study, twosecB-specific promoters were identified. In addition, 5′ transcription initiation sites from these two promoters were determined by means of secB-lacZ fusions and primer extension. The distal P1 promoter appeared to be independent of carbon sources, whereas the proximal P2 promoter was shown to be subject to control by the cyclic AMP (cAMP) receptor protein (CRP)-cAMP complexes. Gel-mobility shift studies showed that this regulation results from direct interaction between the secB P2 promoter region and the CRP-cAMP complex. Moreover, the CRP binding site on thesecB gene was determined by DNase I footprinting and further substantiated by mutational analysis. The identifiedsecB CRP binding region is centered at the −61.5 region of the secB gene and differed from the putative binding sites predicted by computer analysis.

Yersinia pestis is one of the most dangerous pathogens. The cyclic AMP receptor protein (CRP) is required for the full virulence of Y. pestis, and it acts as a transcriptional regulator to control a large regulon, which includes several virulence-associated genes. The regulatory action of CRP is triggered only by binding to the small molecule cofactor cyclic AMP (cAMP). cAMP is synthesized from adenosine triphosphate by the adenylyl cyclase encoded by cyaA. In the present work, the regulation of crp and cyaA by CRP was investigated by primer extension, LacZ fusion, electrophoretic mobility shift assay, and DNase I footprinting. No transcriptional regulatory association between CRP and its own gene could be detected under the growth conditions tested. In contrast, CRP bound to a DNA site overlapping the core promoter −10 region of cyaA to repress the cyaA transcription. The determination of cellular cAMP levels further verified that CRP negatively controlled cAMP production. Repression of cAMP production by CRP through acting on the cAMP synthesase gene cyaA would represent a mechanism of negative automodulation of cellular CRP function.

ABSTRACT We have shown that the Escherichia coliphosphate-starvation-inducible psiE gene is regulated by both phosphate and the carbon source by using both lacZ and chloramphenicol acetyltransferase gene (cat) fusions. Yet, under all conditions tested, a single transcriptional start site lying 7 bp downstream of a predicted −10 region was revealed by primer extension analysis. DNase I footprinting showed that the PhoB transcriptional-activator protein protects two predictedpho boxes lying upstream of and near the −35 promoter region. Similar analysis showed that the cyclic AMP (cAMP)-cAMP receptor protein (cAMP-CRP) complex binds a region that overlaps with the downstream pho box. These results, together with measurements of the in vivo psiE promoter activity under various conditions, show that expression of the psiE gene is under direct positive and negative control by PhoB and cAMP-CRP, respectively.

Defisiensi kalsitriol dapat meningkatkan faktor risiko terjadinya penyakit kardiovaskular. Hal ini dikaitkan dengan penurunan fosforilasi troponin I (TnI) di Ser23/24. Fosforilasi TnI di Ser23/24 berperan dalam regulasi Mg-ATPase miofilamen dan sensitivitas miofilamen terhadap Ca2+ yang selanjutnya mempengaruhi respon frekuensi dan kekuatan ejeksi ventrikel. Ser23/24 TnI ini diketahui merupakan substrat spesifik PKA. Penelitian ini bertujuan untuk mempelajari apakah efek kalsitriol terhadap peningkatan fosforilasi TnI mempunyai jalur mekanisme aksi yang juga melalui cAMP/PKA. Penelitian ini merupakan eksperimental murni menggunakan rancangan pre-post test with control group dengan sampel kultur primer kardiomiosit Sprague-Dawley jantan dewasa. Kardiomiosit diinkubasi kalsitriol selama 5 menit kemudian kadar cAMP diukur menggunakan metode ELISA. Hasil penelitian tidak menunjukkan adanya korelasi antara peningkatan TnI terfosforilasi dengan kadar cAMP (p>0,05). Penelitian ini masih belum dapat membuktikan adanya jalur aktivasi cAMP/PKA oleh kalsitriol dalam mekanisme peningkatan TnI terfosforilasi pada kardiomiosit.

Penerapan Whatsapp Business telah memberikan kontribusi positif terhadap Higland Camp Curug Panjang dengan calon pengunjung, dengan adanya layanan pelanggan bisa membuat pengelola lebih leluasa untuk berkomunikasi dengan calon pengunjung untuk memberikan informasi – informasi yang di butuhkan oleh calon pengunjung. Layanan pelanggan yang dilakukan melalui fitur chat atau telpon dengan calon pengunjung digunakan untuk menumbuhkan rasa nyaman dengan calon pengunjung sehingga calon pengunjung ini tertarik untuk berkunjung ke Highland Camp Curug Panjang. Penelitian ini bertujuan untuk mengetahui penggunaan Whatsapp Business dalam layanan pelanggan untuk meningkatkan minat pengunjung. Pendekatan penelitian ini kualitatif fenomenologi dengan metode deskriptif kualitatif. Teknik pengumpulan data menggunakan data primer diperoleh dengan observasi pengamatan layanan pelanggan Whatsapp Business dan wawancara sedangkan data sekunder dari dokumentasi. Hasil penelitian menunjukan bahawa penggunaan Whatsapp Business melalui layanan pelanggan memberikan dampak baik untuk meningkatkan minat pengunjung di Highland Camp Curug Panjang.

Expression of the surfactant protein A (SP-A) gene is lung specific, developmentally induced, and regulated by adenosine 3',5'-cyclic monophosphate (cAMP) and glucocorticoids. Humans have two highly similar genes encoding SP-A (SP-A1 and SP-A2). In the companion paper [S.M. McCormick, V. Boggaram, and C.R. Mendelson Am. J. Physiol. 266 (Lung Cell. Mol. Physiol. 10): L354-L366, 1994] we report that SP-A1 and SP-A2 RNA transcripts are alternatively spliced at their 5' ends, resulting in nine different primer-extended transcripts. In the present study, primer extension was used to assess the relative levels of expression of the SP-A1 and SP-A2 genes in human adult lung tissue and in fetal lung tissues maintained in organ culture in the absence or presence of dibutyryl (DB)cAMP (1 mM) and dexamethasone (Dex, 10(-4) M). Primer extension and Northern analysis were used to assess the effects of these agents on the levels of expression of these genes. In human adult lung tissue, 65% of the SP-A mRNA transcripts were derived from the SP-A2 gene, whereas only 35% were from SP-A1. On the other hand, in lung tissue from a 28-wk gestation neonate, only SP-A1 mRNA transcripts were detected, and, in midgestation fetal lung cultured in control medium, 65% of the SP-A mRNA was found to be SP-A1 and 35% was SP-A2.(ABSTRACT TRUNCATED AT 250 WORDS)

Para ello, se buscan las analogías morfológicas de las estructuras con las generadas por procesos productivos conocidos y se cruzan los restos documentados con las posibilidades económicas e históricas de contexto, llegando a poder articular dos hipótesis-marco en este punto de la investigación.

Han pasado ya casi seis décadas desde que en 1964 Susan Sontag publicara «Notes on Camp», reivindicando la atención hacia una sensibilidad estética que históricamente había sido reprimida y perseguida. Aunque ha sido referenciado en los estudios culturales académicos como un concepto esquivo, aquello que Sontag denominó «camp» se extendió como un marco interpretativo en una gran parte de las formas de expresión artística y cultural, revelándose como una estética compleja cuya naturaleza política desafiaría el status quo de la normalización y normatividad de las sociedades occidentales. Caracterizada por romper los equilibrios entre el arte elevado y el abrazo a la cultura popular, la compleja y descarada estética camp ha sido de inspiración para múltiples producciones culturales de las últimas décadas, haciendo alarde de modos distintos de habitar los espacios, de entender el cuerpo y la interacción social desde la subalternidad, la ironía, el artificio, la teatralidad y la exageración, asuntos asociados a las identidades homosexuales en un primer momento. Sin embargo, el montaje de la exposición en el año 2019 con el nombre Camp: Notes on Fashion en el Metropolitan Museum of Arts de Nueva York plantea interesantes lecturas sobre la evolución del concepto, las nuevas relaciones de la vestimenta con la identidad de género y la apertura de las prácticas museísticas a las ideologías que promueven la representación disidente, realizando distintos despliegues a través de merchandising y planteando interesantes debates sobre los formatos de performatividad de los fashion studies.

Certain mouse strains (e.g., DBA/2) contain two renin genes (termed Ren-1 and Ren-2) and express higher renin levels in nonkidney tissues than strains with a single renin gene. The 5'-flanking regions of the Ren-1 and Ren-2 genes contain several TATA boxes preceding putative transcriptional start sites. These initiators are termed P1a, P1, P2 (from 5' to 3'), and their function (with the exception of P2) is largely unknown. In this study, we mapped the renin transcriptional start sites in renal and extrarenal tissues [adrenal, brain, testis, heart, and submandibular gland (SMG)] and examined the effect of adenosine 3',5'-cyclic monophosphate (cAMP) on tissue specific promoter usage. Our results showed that, in the unstimulated state, P2 (the predicted initiator) is active in all DBA/2 mouse tissues. Additional transcriptional start sites were detected in the adrenal and testis (originated by P1a and P2) and the SMG (originated by P1a, P1, and P2). The administration of 8-bromoadenosine 3',5'-cyclic monophosphate led to selective stimulation of P1a in the adrenal but did not affect the selective usage of initiation sites in other organs. A locus-specific ddNTP primer extension assay was used to verify which renin gene is induced by cAMP. Results indicated that both Ren-1 and Ren-2 responded to cAMP treatment in identical fashion. Taken together, these data indicate that more than one form of renin transcript is present in several mouse tissues. There is tissue specificity in promoter usage in the unstimulated state and in response to cAMP.

The museum is a permanent, non-profit institution in the service of society and its development, open to the public. It collects, preserves, researches, communicates and exhibits material testimonies of man and his environment, for the purpose of study, education and entertainment. In recent times, museums are primarily communication centres that retain and develop all other functions of a traditional museum. The main form of communication is an exhibition, although any transmission of information is considered as communication. However, the museum is not the only institution where objects of this purpose can be found. Namely, there are many alternative types of collecting and storing things from the past, although not enough attention has been paid to them or real significance attributed. A true collector is a special type of collector, their purpose being to put together a collection of related items as complete, unique and as representative as possible. A collector is a person who is passionate about collecting specific items for his own pleasure. There are a large number of people like this, because in fact it is very difficult to think of any object that nobody would collect. Some of the typical examples vary from the collectors of works of art and precious vases, to the collectors of the most ordinary, trivial, useless, discarded items that are searched for in attics and basements, sometimes even in the landfills... In search of at least one such unusual collector, I came across a small heritage museum created based on geographical affiliation: the objects collected in this collection are all representative of the past of Banat.

A genomic clone for the rat liver gap junction protein (connexin-32) was isolated and characterized by restriction enzyme mapping and sequence analysis. While the complete coding sequence is contained within one uninterrupted block, the 5′-untranslated region of the transcript contains a 6.1 kb intron. Both S1 nuclease protection and primer extension assays indicate multiple transcription start sites. Sequences homologous to cAMP response elements are found near the transcription start sites and within the 3′-end of the intron.

L’especialització d’Elias en el camp de la decoració amb esmalt, sobre vidre i sobre ceràmica, va ser coneguda i celebrada tant durant els anys abans de la Guerra Civil com durant la postguerra. Aquest article en recupera la història i el llegat.

In the human fetal lung, surfactant protein A (SP-A) is encoded by two highly similar genes, SP-A1 and SP-A2, which are developmentally and hormonally regulated. Using primer extension analysis, we evaluated the levels of SP-A1 and SP-A2 mRNA transcripts in human fetal lung explants and in a human adult lung adenocarcinoma cell line (H441 cells) cultured in the absence or presence of either dibutyryladenosine 3′,5′-cyclic monophosphate (DBcAMP, 1 mM), dexamethasone (10−7 M), or insulin (2.5 μg/ml). In the human fetal lung explants, the content of SP-A1 mRNA was approximately four times that of SP-A2 mRNA. DBcAMP increased SP-A1 mRNA levels by 100% and SP-A2 mRNA levels by 500%, thus reducing the ratio of SP-A1 mRNA to SP-A2 mRNA to ∼1:1. Dexamethasone inhibited all of the SP-A1 and SP-A2 mRNA transcripts to the same extent, by ∼70%, whereas insulin inhibited all SP-A mRNA transcripts by ∼60%. The ratio of SP-A1 to SP-A2 mRNA in dexamethasone- or insulin-treated explants was the same as the ratio observed in controls. In the H441 cells, SP-A1 mRNA levels were ∼1.5 times that of SP-A2 mRNA levels. DBcAMP increased both SP-A1 and SP-A2 mRNA levels by 100%. Dexamethasone inhibited SP-A1 mRNA levels in the cell line by 60%, whereas SP-A2 mRNA levels were not significantly affected. Insulin inhibited SP-A1 mRNA levels in the cell line by 40% without affecting SP-A2 mRNA levels. These findings suggest that the two human SP-A genes are regulated differently in the two model systems.

The paper illuminates the enlightenment work of Zaharija Orfelin from the viewpoints of broader cultural and historical and narrower linguistic and didactic guidelines. In the social context of 18th-century Serbia, which cannot be considered one-sided, amidst the fusion of cultural values of the East and West, Orfelin conceptualized the key of national values in education. The Primer is more than the first book - it is a latent proclamation of the coming of the Enlightenment ideas. The aim of this work is point to the fact that in the cultural history of Serbia he was the precursor of modern Serbian language teaching long before it came to life in our modern teaching under the influence of foreign methodologists.

The research aimed at analyzing spike in blockers of Iraqi volleyball league clubs as well as finding a correlation relation between spiking and tactical performance of blockers. The researcher used the descriptive method on (96) Iraqi volleyball primer league club players of the season (2020 – 2021). The clubs who came first five (40) were selected and given a questioner for evaluation spiking in volleyball. Click and Scout program was used to come up with the conclusion that state a significant correlation relation between spiking and tactical performing of blocking in primer league volleyball players.

En el tombant de segle francès, l’anomenat debat sobre nacionalisme i literatura comprenia algunes veus reticents a la importació de literatura estrangera. En el text editat, Gide s’oposa a aquestes prevencions. «Dones i terres catalanes medievals» és un estudi de llarga durada, a l’espai de la Catalunya Vella i la Nova. Les dones, majoritàriament, són dones amb poder: primer les feminae, després les dominae. Un poder que es palesa en l’assumpció de les polítiques masculines, però que, alhora, traspua autoritat, refl ectida en les polítiques femenines. L’objectiu principal, doncs, ha estat refl exionar sobre el rol de les dones al camp català als segles X-XIII.La metodologia emprada és la pròpia de la recerca històrica feminista, a més dels mètodes empírics clàssics: recull de dades arxivístiques, sistematització, tractament, comparació, exposició de resultats. Pel que fa a l’estat de la qüestió bibliogràfi ca, no hi ha referències per a l’època i l’òptica triada; per tant, aquesta anàlisi s’ha construït prenent com a base fonts primàries d’arxiu.

The current study comprising molecular identification of the mutagenic effects of gamma rays and sodium azide on Catharanthus roseus using scot markers took place at the Department of Biology, University of Kufa, Iraq. The seeds of two cultivars, i.e., ‘victory carmine’ and a local variety (pink variety) of Catharanthus roseus L (carmine var.) received irradiation with two doses of gamma rays (50 Gray and 75 Gray). Afterward, these got soaked in sodium azide with four concentrations (0.2%, 0.4%, 0.6%, and 0.8%) to induce the genetic variation in the cultivar. The non-treated seeds served as control treatments for comparison with treated ones. Accomplishing irradiation was at an average of 18 Gy/h using Cobalt-60. The effects of gamma rays and sodium azide treatments’ evaluation were through 10 start codon targeted (SCoT) DNA markers. The higher number of distinctive fingerprints was three produced by primers SCoT-26, while primer SCoT-60 provided a distinct fingerprint for only one treatment. However, primer SCoT-40 failed to recognize any treatment with a distinctive fingerprint. The highest molecular size (2488 bp) came from primer SCoT-54, whereas the lower molecular size (143 bp) resulted in primer SCoT-12. Primer SCoT-33 provided the highest value for chief and unique bands, reaching 33 and 18, respectively. The highest number of amplified bands was 77 bands in primer SCoT-54. Primer SCoT-60 produced the highest value for monomorphic bands, primer SCoT-12 gave the highest value for polymorphic bands, polymorphism, and discriminatory (15, 68.18, and 20.83, respectively), and primer SCoT-9 produced the highest value for efficiency.

Background: Primary resistance is the resistance that occurs in patients who have never received treatment OAT or had ever received treatment OAT is less than 1 month. The incidence of primary MDR in Central Java in 2006 2.07%. Extrapulmonary TB about 15-20% of all cases of TB, and TB lymphadenitis is the highest form (35% of all extrapulmonary TB). Patients with decreased immune systems (SLE) can increase the incidence of TB. Research in Spain get 6x higher TB incidence in patients with SLE. Case: We present the case of 19 year old woman SLE who received treatment for 11 months whose came with shortness of breath and chronic cough since 2 month prior to admission. In physical examination we found right and left submandibula lymphonodi enlargement as solid, slightly mobile nodule with diameter 3 cm. Chest X ray showed lung inflammation which suspicion of specific process and minimal left pleural effusion, and concluded as pulmonary TB. FNAB confirmed lymphadenitis TB with granulomatous inflammation. One of AFB result is positive and Gene Xpert is MTB positive with rifampicin resistant that make this patient categorized as primary MDR TB with lymphadenitis TB. This patient received Pirazinamid 1500 mg, Ethambutol 800 mg, Kanamicin 750 mg, Levofloxacin 750 mg, Ethionamide 500 mg, Cicloserin 500 mg, and B6 100 mg. Conclusion: MDR TB in general occur in patients with a history of OAT previous TB (MDR TB secondary). Primary MDR TB with lymphadenitis tb is a rare case, but can occur on the condition that decreases the immune system, one of SLE. This involves multiple immune disorders caused by the use of long-term immunosuppressive therapy.

Este artículo orbita en torno a la representación audiovisual de uno de los capítulos más oscuros dela historia de la humanidad: el Holocausto. Partiendo de la proposición wittgensteniana de que “ética y estética son una”, hemos llevado a cabo un análisis hermenéutico y semiótico de la película húngara El hijo de Saúl (Saul fia, László Nemes, 2015), tratando aspectos entre los que destacan el montaje, la imagen, el foco, el audio y el color. El objetivode este estudio es reivindicar la importancia de este filme como objeto cultural que reúne una serie de decisiones estéticas que la sitúan éticamente en el mundo y en el panorama cinematográfico, frente a otros que trivializan, estetizan o monetizan el exterminio judío a manos de los nazis. El hijo de Saúl respeta lo extraordinario del acontecimiento, e interroga al cine como medio para encontrar una vía de representación que no degenere en esa banalización del dolor. Así, llegamos a tres conclusiones. En primer lugar, que en ese constante preguntarse por las posibilidades del cine y los límites estéticos de la representación, se encuentra el comportamiento ético por parte de los cineastas. En segundo, que el espectador deberá aceptar el reto de enfrentarse a un cine alejado del modo de representación institucional (MRI) y formar parte activa del intercambio de información. Por último, que el cine como vehículo de cambio social debe servir al fortalecimiento de la democracia y contribuir a garantizar la no repetición del desastre.

<p>Me ocuparé aquí de abordar una tensión profunda y fundamental presente en la Teoría de las Formas de Platón, a saber, la tensión existente entre comprender las Formas como arquetipos por un lado y como esencias por otro. Un arquetipo es un ejemplo preeminente de su clase que incorpora los estándares más altos en sus virtudes particulares. Una esencia, en cambio, es el denominador común más alto de una clase de cosas, sus condiciones necesarias y suficientes si se quiere. Platón parece inclinarse más por una u otra concepción en diferentes momentos de su obra, pero no parece advertir la diferencia entre ambas. Una manera útil de captar la distinción, al menos parcialmente, es pensando en lo que significa la palabra ‘ideal’. La palabra tiene (al menos) dos significados: por un lado, puede significar “concebido o considerado como perfecto o excelente en forma suprema en su clase”. Por otro lado, su segundo significado carece de esta fuerza normativa y significa, en forma evaluativamente neutra, “perteneciente al mundo de la mente -en lugar de a la realidad concreta”. (Piénsese en la diferencia entre “un matrimonio ideal” y la ficción de la química teorética, “un gas ideal”). A veces las Formas de Platón parecen ser entendidas como ‘ideales’ en el primer sentido normativo; en otras ocasiones, parecen más neutrales, con ‘ideal’ usado en el segundo sentido. ¿Es la ‘cama ideal’ la mejor posible de las camas? ¿O es la ‘cama ideal’ simplemente el conjunto de propiedades que las camas tienen en común, la idea de cama? ¿Es el arquetipo de ‘cama’ o la esencia de ‘cama’? Cualquiera de las opciones que tomemos resulta problemática.</p>

Abstract: Moderate thrombocytosis can accompany several diseases (bleeding, inflammation, iron deficiency, or autoimmune diseases), but hematologic examination is strongly recommended in a patient with persistent platelet count above 450 G/L unless reactive origin can be confirmed. The 47-year-old woman’s medical history included hypertonia, asthma bronchiale, and endometriosis. In March 2015, she underwent laboratory examination due to weight loss and lack of appetite. Her results showed elevated thrombocyte count (617 G/L), but no iron deficiency. She presented in our clinic on 07. 04. 2015 with acute pain below her left hypochondrial region, but simple imaging examinations showed no difference to explain it. Abdominal CT revealed a 4.5 cm thrombus which protruded into the left renal artery, blocking it. We started APTI- (activated partial thromboplastin time) monitored continuous intravenous treatment with unfractionated heparin. The JAK2V617F mutation analysis came back positive. Subsequent bone marrow examination revealed prefibrotic/early stage myelofibrosis, prompting treatment with hydroxyurea. The applied treatments led to the disappearance of the patient’s symptoms accompanied by the gradual normalisation of the thrombocyte count. Moderate thrombocytosis is often secondary, but if it persists and is accompanied by mainly thromboembolic events, the risk of diseases of the haematopoietic system, primarily Philadelphia chromosome negative chronic myeloproliferative disease should also be considered. Clinically, essential thrombocythaemia and the prefibrotic/early stage of myelofibrosis can be very similar. Differential diagnosis is only possible through the histological examination of the bone marrow, which becomes indispensible due to the difference in prognosis and treatment options. El objetivo de esta investigación fue evaluar la inclusión de compost de cama profunda (CP) de cascarilla de arroz y porquinaza en la alimentación de vaquillas cruzadas de doble propósito. Para ello se utilizaron 18 vaquillas alimentadas con CP procesada en 12 meses obtenidos de la Unidad de Docencia, Investigación y Vinculación Hato Porcino de la Escuela Superior Politécnica Agropecuaria de Manabí “Manuel Félix López”. Se aplicó un DCA con arreglo bifactorial [primer factor: edad (menor a 25 meses de edad y mayor a 25 meses de edad y preñadas de diferente tiempo de gestación); segundo factor: niveles de compost de cama profunda (0%, 10% y 20%)], se consideró como covariable al peso inicial. La composición nutricional del compost de cama fue determinada mediante un análisis proximal con 5,71% de proteína cruda. No se encontraron diferencias significativas (P>0,05) en cada uno de los factores y en la interacción de estos, el grupo de animales menores a 25 meses de edad tuvo un mejor peso al final promedio de la investigación con 308,48 Kg (±2,24), a la vez que los animales que fueron alimentados con una dieta que incluyó 20% de CP tuvieron un peso medio de 310,18 Kg (±2,10) al final del experimento. Se concluye que el contenido nutricional encontrado en el compost de cama profunda no desfavoreció la ganancia de peso y conversión alimenticia, por lo que las dietas suministradas 20% de porquinaza podrán ser utilizadas para mejorar el rendimiento productivo en vaquillas mestizas.

Male and female Chinese soft-shelled turtles (Pelodiscus sinensis) have sex-dimorphic growth patterns, and males have higher commercial value because of their larger size and thicker calipash. Thus, developing sex-specific markers is beneficial to studies on all-male breeding in P. sinensis. Here, we developed an accurate and efficient workflow for the screening of sex-specific sequences with ZW or XY sex determination systems. Based on this workflow, female and male P. sinensis reference genomes of 2.23 Gb and 2.26 Gb were obtained using de novo assembly. After aligning and filtering, 4.01 Mb female-specific sequences were finally identified. Subsequently, the seven developed sex-specific primer pairs were 100% accurate in preliminary, population, and embryonic validation. The presence and absence of bands for the primers of P44, P45, P66, P67, P68, and P69, as well as two and one bands for the PB1 primer, indicate that the embryos are genetically female and male, respectively. NR and functional annotations identified several sex-determining candidate genes and related pathways, including Ran, Eif4et, and Crkl genes, and the insulin signaling pathway and the cAMP signaling pathway, respectively. Collectively, our results reveal that a ZW-type sex-determination system is present in P. sinensis and provide novel insights for the screening of sex-specific markers, sex-control breeding, and the studies of the sex determination mechanism of P. sinensis.

There are approximately 400 binding sites per cell, with a binding constant of about 100 pmol/L. Before the proliferative response, the affinity constant of the membrane particulate fraction to 35S-labeled guanosine triphosphate-gamma-S (35S-GTP gamma S) and the intracellular cyclic adenylate monophosphate (cAMP) level increased in the presence of G-CSF to about 2.5-fold and about fivefold higher, respectively, than the levels seen in the absence of G-CSF. The increases were time-dependent, with a peak occurring 15 minutes after the addition of G-CSF at 37 degrees C. These findings suggest that, following the binding of the G-CSF to its surface receptors, the activation of the guanosine triphosphate (GTP)-binding protein/adenylate cyclase system may be involved in the proliferation of immature myeloid cells.

Teknik pengumpulan data yang dilakukan dengan cara membaca/mencari beberapa informasi atau data dari berbagai sumber baik berupa buku ataupun artikel-artikel yang terkait dengan objek penelitian. Hasil penelitian dari beberapa peneliti terkait penelitian ini menjadi data primer. jenis penelitian ini adalah penelitian pustaka yang menganalisis hasil-hasil penelitian yang dilaksanakan oleh peneliti-peneliti sebelumnya, yang terkait dengan judul penelitian ini. Perapan model pembelajaran Quantum Teaching, aktivitas belajar menjadi lebih aktif dan cenderung positif dalam mengikuti proses pembelajaran yang diberikan oleh guru. Dengan kondisi tersebut maka tingkat pemahaman akan meningkat dan pada gilirannya dapat meningkatkan motivasi belajar.

La clinoterapia o “reposo en la cama” fue introducida como tratamiento para dolencias mentales en 1852, con el propósito de inhibir la “conciencia de movimiento” y la excitación periférica y, así, posibilitar un “reposo psíquico”. En un primer momento la clinoterapia, junto con otras medidas como el open door y el non restraint, significó una alternativa a las celdas de reclusión, las camisas de fuerza y otros métodos restrictivos. Sin embargo, a la postre se vio reducida al simple aislamiento en una habitación, muchas veces en condiciones muy poco apropiadas.

Des de l’any 2006, i amb diferents equips, hem estat treballant sobre parelles mixtes o matrimonis binacionals en sengles projectes del Plan Nacional de I+D+I:1 primer amb matrimonis entre homes espanyols i dones estrangeres, els majoritaris entre les parelles mixtes espanyoles, i després incorporant també les unions de dones espanyoles amb homes estrangers. Per bé que ambdues recerques han estat de caire qualitatiu, sempre hem pres en consideració amb finalitats analítiques i de plantejament el conjunt de l’Estat. Teníem interès, doncs, a dur a terme una recerca més modesta quant al territori a abastar que ens permetés així mateix focalitzar l’atenció en qüestions més específiques i localitzades. El projecte concedit per l’Institut Català de les Dones, que hem dut a terme en el període 2010-2011, ens ha donat l’oportunitat de realitzar-la.

ABSTRAKTujuan dalam penelitian ini adalah menganalisis preferensi konsumen yakni masyarakat kota Jambi terhadap kopi lokal dan menganalisis hubungan usia terhadap preferensi masyarakat kota Jambi dalam mengkonsumsi kopi lokal. Adapun hipotesis dalam penelitian ini adalah terdapat hubungan antara usia konsumen kopi dengan preferensi dalam mengkonsumsi kopi lokal Jambi.Penelitian ini dilakukan di Kota Jambi dengan mengumpulkan data primer dan sekunder. Penelitian ini dilaksanakan pada bulan Februari sampai Juni 2020. Data yang digunakan adalah data cross section. Data konsumen dikumpulkan dari 3 kedai kopi yang terdapat di Kota Jambi yakni Hello Sapa, Camp Coffee, dan Kopi Juaro. Penarikan sample dilakukan dengan metode simpel random sampling. Penghitungan sampel dengan rumus Slovin. Hasil penelitian menunjukkan bahwa Mayoritas konsumen kopi memilih kopi lokal dibandingkan kopi non lokal dan Hubungan usia konsumen dengan preferensi konsumsi kopi adalah tidak signifikan. Sample calculation using the Slovin formula. The results showed that the majority of coffee consumers prefer local coffee than non-local coffee and the relationship between consumer age and coffee consumption preference is not significant. Trabajar con Redes Inalámbricas de Sensores (RIS) en zonas urbanas es una tarea complicada de realizar, en donde diversos factores se deben tomar en cuenta, debido a la naturaleza propia del entorno. Es aquí en donde herramientas analíticas como los simuladores juegan un papel importante para el éxito de las redes. El objetivo de este trabajo es contar con un entorno virtual 3D para el simulado de RIS que sea confiable, escalable, modular y robusto que permita dar certidumbre durante el diseño de redes. En este trabajo se presenta el desarrollo de un entorno 3D de la ciudad de Torreón, haciendo uso del simulador de eventos discretos OMNeT++, con herramientas de INET framework. La cama de simulación 3D comprende una sección de 1 km2 de objetos físicos como edificaciones y vegetación, el mapa de la ciudad de Torreón y datos de elevación de la SRTM. Se presentan dos casos comparativos entre simulaciones y datos de RIS desplegadas en la ciudad, obteniendo la eficiencia de la red y el RSSI, a diferentes distancias entre nodos. El primer caso de estudio corresponde al estándar IEEE 802.15.4 en distancias de 25, 50 y 100m, y el segundo caso al modo TI Legacy Long Range a 90, 135 y 200m. Los resultados muestran una coincidencia del 92% y el 95% en promedio respectivamente en paquetes recibidos entre datos simulados y los recolectados con las RIS. En cuanto al RSSI se muestra un comportamiento acorde a lo esperado, debilitándose a mayores distancias. La cama de simulación 3D es una herramienta confiable para simular redes con diferentes niveles de complejidad y escala, facilita tener referencias visuales y permite entender el comportamiento de las RIS que se busquen diseñar, sirviendo de apoyo en los esfuerzos para el monitoreo de calidad de aire que existen en la región.

A l’Estat espanyol, el desenvolupament eòlic ha seguit un model centralitzat i concentrat en territoris rurals i empobrits, cosa que ha perpetuat una dinàmica extractiva: mentre que els beneficis i l’electricitat són transferits a altres llocs, les zones productores són assimilades a un erm, és a dir, desvalorades econòmicament i moralment. Aquest article, basat en un treball de camp etnogràfic de llarga durada a la Catalunya meridional, té dos objectius principals. El primer, mostrar que aquest moviment dialèctic —valoració del vent i desvaloració del territori i dels seus habitants— és funcional per als interessos dels promotors eòlics, ja que permet justificar la seva activitat i abaratir-ne els costos. El segon, examinar les resistències, especialment les de caràcter quotidià que el desenvolupament eòlic ha generat. En aquest sentit, paro especial atenció a la demanda de dignitat que ha articulat aquestes resistències a la regió, argumentant que cal entendre-la com a part d’una trajectòria de lluita orientada a reproduir les condicions objectives i subjectives que fan possible el sosteniment i la valoració de les formes de vida locals.

In the course of experiments designed to assess the potential role of alternative open reading frames (ORF) present in the 5′-terminal untranslated region (5′-UTR) of poliovirus type 1 (Mahoney strain) genomic RNA, we came across a double mutation that completely abrogated the infectivity of full-length cDNA clones. The infectivity was rescued in trans by cotransfecting COS-1 cells with short RNA transcripts of the wild-type 5′-UTR of poliovirus type 2 Lansing, provided a free 3′-OH was available. Direct sequencing of the viral RNA revealed that the infectious viruses recovered were recombinants Lansing/Mahoney, with variable points of ‘crossing-over’. A novel mechanism of RNA–RNA recombination, which we propose to call ‘primer alignment-and-extension’, is described that would explain the high rate of recombination of RNA viruses observed in natural conditions.

Para este doble homenaje a la revista y a su conceptor, se me ocurre en primer lugar una imagen: la viñeta diseñada por Kim bajo las instrucciones del guionista Antonio Altarriba que representa al joven estudiante Antonio en su improvisado rincón-cama-despacho de la carbonera donde su familia vivió durante años. Estando alojados en un sótano en el que se almacenaba el carbón, su madre le inventó una ventana con cortinas. Me lo figuro estudiando e imaginando mundos que, por diferentes causas explicitadas en El Ala Rota, le fueron totalmente prohibidos a sus padres.

Abstract We have cloned the human perforin (P1) gene and sequenced 6.2-kb genomic DNA, containing 1.4-kb 5'-flanking region, the 5' untranslated region, the complete coding region and the beginning of the 3' untranslated region. The P1 gene including at least 95-bp 3' untranslated region is organized in only three exons: the first exon (97 bp) contains all but four nucleotides of the 5' untranslated region and was determined by primer extension and S1 nuclease mapping. This exon is separated by 1.7 kb from the second exon containing the remaining (4 bp) 5' untranslated region, the leader peptide and the N-terminal region of P1 up to--but not including--the C9 homologous region. The third exon is separated by a 1.2-kb intron and contains the remainder of the molecule, including at least 90 bp of the 3' untranslated region. This simple gene organization differs from that of the more complicated C9 gene. Because of the unusual intron in the 5' untranslated sequence the transcription initiation (cap) site is located almost 1.8 kb upstream of the ATG start signal. The more immediate 5' flanking sequence contains a CCAAT and GC box but lacks other known promoter elements. Instead, we find three different sequence repeats. One of them, a hexanucleotide sequence with the consensus GCCCTG of unknown significance occurs 19 times within a stretch of 240 bp. Further upstream we localized sequences homologous to the following enhancer and promoter elements: c-fos proto-oncogene, IFN-gamma and phorbol ester response elements, five cAMP response elements, and three motifs corresponding to general inducer elements. In addition, a sequence conserved in the 5'-flanking region of several T cell genes was identified. The 5' flanking regions of P1. CCP1 (granzyme B) and CCP2 (granzyme C) (kindly provided by Dr. Bleackley) contain as only significant homology cAMP response elements. These findings are consistent with a tight control and regulation of P1, which appears to be distinct from that of granzymes.

The programmable sequence specificity of CRISPR has found uses in gene editing and diagnostics. This manuscript describes an additional application of CRISPR through a family of novel DNA enrichment technologies. CAMP (CRISPR Associated Multiplexed PCR) and cCAMP (chimeric CRISPR Associated Multiplexed PCR) utilize the sequence specificity of the Cas9/sgRNA complex to target loci for the ligation of a universal adapter that is used for subsequent amplification. cTRACE (chimeric Targeting Rare Alleles with CRISPR-based Enrichment) also applies this method to use Cas9/sgRNA to target loci for the addition of universal adapters, however it has an additional selection for specific mutations through the use of an allele-specific primer. These three methods can produce multiplex PCR that significantly reduces the optimization required for every target. The methods are also not specific to any downstream analytical platform. We additionally will present a mutation specific enrichment technology that is non-amplification based and leaves the DNA in its native state: TRACE (Targeting Rare Alleles with CRISPR-based Enrichment). TRACE utilizes the Cas9/sgRNA complex to sterically protect the ends of targeted sequences from exonuclease activity which digests both the normal variant as well as any off-target sequences.

Objective: To explore the effect of plasma homocysteine (Hcy) levels on methylation and expression of thrombomodulin (TM) gene in patients with retinal vein occlusion (RVO).Methods: 11 cases of patients who were diagnosed as RVO in Department of Ophthalmology in Baogang Hospital from January 2019 to December 2020 were included in this research. 11 cases of healthy people who came to our physical examination center for physical examination during the same period were included into the control group. Fasting cubital venous blood was collected in the morning, plasma Hcy levels were measured by means of enzyme cycle assay, TM gene methylation was detected by methylation-specificity polymerase chain reaction (MSP), and the correlation between characteristic changes in plasma Hcy and TM gene methylation was analyzed in the research subjects.Results: Plasma Hcy expression levels were higher in the RVO group than those in the control group (p < .01). In the control group, the TM primer set region was basically unmethylated, while in the RVO group, the TM primer set region was partially methylated.Conclusions: Plasma Hcy affects the occurrence and development of RVO through methylation of TM encoding gene.

Abstract: Primary angle closure glaucoma (PACG) is the leading cause of blindness in Asia. In PACG, degenerative changes in the trabecular meshwork resulting from iridiotrabecular contact (ITC) cause high intraocular pressure resulting in glaucomatous optic atrophy. We reported a 58-year-old woman came with the chief complaint of pain in the right eye for three months, accompanied by red and watery eye, and glare when exposed to light. Patient also complained of headaches and sometimes nausea. There was a history of hypertension since one year ago, but not taking any drugs. In ophthalmological examination of the right eye, visual acuity was 6/20, intraocular pressure 39 mm Hg, conjunctival injection, corneal edema, Van Herick 1 shallow anterior chamber, iris atrophy, round shaped and centrally located pupil with a diameter of 5 mm and a NO3NC3 lens. In the posterior segment of the right eye, non-uniform fundal reflexes were found, papillae were round, vital, well-defined with a cup and disc ratio of 0.7-0.8. The patient was diagnosed with primary angle-closure glaucoma in the right eye and a trabeculectomy was performed on the right eye. After trabeculectomy, the intraocular pressure of the right eye was 17.7 mm Hg. In conclusion, trabeculectomy has a high success rate in terms of controlling intraocular pressure in primary angle closure glaucoma. Pada PACG, perubahan degeneratif dalam trabecular meshwork yang dihasilkan dari kontak iridiotrabecular (ITC) menyebabkan tekanan intraokular tinggi yang menghasilkan glaucomatous optic atrophy. Kami melaporkan kasus eorang perempuan berusia 58 tahun datang dengan keluhan nyeri mata kanan sejak tiga bulan yang lalu, disertai dengan mata merah, berair dan silau saat terkena cahaya. Pasien juga mengeluh adanya nyeri kepala dan kadang disertai mual. Riwayat hipertensi sejak satu tahun sebelumnya, tidak mengonsumsi obat-obatan. Pada pemeriksaan oftalmologi mata kanan didapatkan visus 6/20, tekanan intraokuler 39 mmHg, konjungtiva injeksi, kornea edema, bilik mata depan dangkal Van Herick 1, iris atrofi, pupil berbentuk bulat letak sentral dengan diameter 5 mm serta lensa NO3NC3. Pada segmen posterior mata kanan ditemukan refleks fundus non uniform, papil bulat, vital, berbatas tegas dengan cup and disc ratio 0,7-0,8. Pasien didiagnosis dengan glaukoma sudut tertutup primer mata kanan dan dilakukan tindakan trabekulektomi pada mata kanan. Setelah tindakan trabekulektomi, tekanan intraokuler mata kanan menjadi 17,7 mmHg. Simpulan kasus ini ialah trabekulektomi terbukti memiliki tingkat keberhasilan yang tinggi dalam hal mengontrol tekanan intraokuler pada glaukoma penutupan sudut primer. Aboriginal law has developed to require Aboriginal peoples to be “consulted and accommodated” if their rights may be impacted by a government decision, including a government permit or approval of a project of a private proponent. For example, hunting rights often exist throughout a First Nation’s treaty or traditional territory (i.e. far beyond the limits of reserves), and the duty to consult and accommodate can be triggered by mining and other resource development. Contrary to the common understanding of some of those unfamiliar with this area of law, this duty applies not only to activities undertaken under federal authorization, but also to those under provincial authorization. The Crown’s “duty to consult and accommodate” Aboriginal peoples has become a central theme in the discussion of natural resource development in Canada. In response to various decisions of Canadian courts, the Government of Ontario significantly overhauled its Mining Act in 2009 to provide for some consultation with Aboriginal communities. Those changes came in to effect in the spring of 2013. This paper will describe the constitutional duty to consult as it has been described and elaborated on by courts in Canada and some of the implications it has for resource extraction in Ontario. It will then undertake a case study discussing Ontario’s attempt to respond to its duty to consult by amending the Mining Act regime. Finally, the paper will consider the flaws in the Mining Act and the reasons that exploration companies and surveyors working for them should be prudent and pro-active when undertaking intrusive activities in the traditional territories of Aboriginal peoples.

The genomic structure and transcriptional regulation of the rat apelin receptor (APJR) were analysed by rapid amplification of 5′ cDNA ends (5′-RACE), transient expression assays and DNA–protein interaction. Analysis of the 5′-flanking region of a rat genomic clone shows no TATA box, but a putative CAAT box and several putative binding sites for transcription factors are present. Two transcriptional start sites were identified by 5′-RACE, RNase protection and primer extension analyses. Promoter activity was exhibited in the APJR- expressing SH-SY5Y cell line as well as in COS-7 and Chinese hamster ovary (CHO-K1) cells. Consecutive 5′-deletion analysis revealed the highest promoter activity in a region between bp −966 and −165. DNaseI footprint analysis revealed seven protected regions and electrophoretic mobility shift, super-shift and competition assays identified individual DNA–protein complexes capable of binding Sp1, estrogen receptor (ER)α, glucocorticoid receptor and CCAAT enhancer binding protein (C/EBP)γ transcription factors. Site-directed mutagenesis identified an individual Sp1 motif that plays a major role in activation of the APJR promoter and also demonstrated constitutive transcriptional regulation of the promoter by estrogen and glucocorticoid receptors. Promoter regulation by the cAMP-dependent signal cascade was also shown.

A change in the management of pusaka tinggi (high heritage assets) in Nagari Kubang Putiah, Minangkabau, prompted this research. This article examines the position of pusaka tinggi in Nagari Kubang Putiah, as well as whether high inheritance management in Nagari Kubang Putiah is governed by Islamic law. The primary data came from interviews, while the secondary data came from literature studies on high heirlooms in Kubang Putiah. The findings show that, first, the position of high heirloom in Nagari Kubang Putiah is as a customary inheritance; second, the management of high heritage assets in Kubang Putiah has changed, not following the applicable provisions; and third, the management of high inheritance assets is following Islamic law, such as hibah (grants). Penelitian ini dilatarbelakangi oleh adanya suatu perubahan pengelolaan harta pusaka tinggi di Nagari Kubang Putiah, Minangkabau. Artikel ini menganalisis bagaimana kedudukan harta pusaka tinggi di Nagari Kubang Putiah, apakah pengelolaan harta pusaka tinggi di Nagari Kubang Putiah itu sesuai dengan Hukum Islam. Artikel ini merupakan hasil penelitian lapangan. Sumber data primer diperoleh dari wawancara, sedang data sekunder dari kajian literatur yang terkait dengan harta pusaka tinggi di Kubang Putiah. Hasil penelitian menunjukkan bahwa, pertama, kedudukan harta pusaka tinggi di Nagari Kubang Putiah adalah sebagai harta waris adat; kedua, pengelolaan harta pusaka tinggi di Kubang Putiah mengalami perubahan; ketiga, dari perspektif hukum Islam, pengelolaan harta pusaka tinggi dapat disamakan dengan harta hibah.
