Texto completoIn recent years, a large number of technologies have been created to help people who have difficulty when reading written texts. The proposed systems integrate speech technologies (reading aloud) or visual aids (setting and/or coloring of fonts or increasing the space between letters and lines). However, it is essential to also propose transformations on the texts’ content in order to have simpler and more frequent substitutes. The purpose of this thesis is to contribute to develop a reading aid system that automatically provides a simplified version of a given text while keeping the same meaning of words.The presented work addresses the problem of semantic ambiguity (quite common in natural language processing) and aims to propose solutions for Word Sense Disambiguation (WSD) by using unsupervised and knowledge-based approaches from lexico-semantic resources. First, we propose a state of the art of the WSD approaches and semantic similarity measures which are crucial for this process. Thereafter, we compare various algorithms of WSD in order to get the best of them. Finally, we present our contributions for creating a lexical resource for French that proposes disambiguated and graduated synonyms according to their level of difficulty to be read and understood. We show that our resource is useful and can be integrated in a lexical simplification of texts module

Texto completoMost cases of Duchenne muscular dystrophy (DMD) are caused by dystrophin gene mutations that disrupt the mRNA reading frame. In some cases, forced exclusion of a single exon can restore the reading frame, given rise to a shorter, but still functional dystrophin protein. Our objective in this work was to produce antisense sequences targeting splice junctions of dystrophin gene to induce removal of disease-associated exons during pre-mRNA processing. To achieve this exon-skipping, we proposed to use the U7 small nuclear RNA as carrier and we first developed AAV vectors harboring chimeric U7snRNA carrying antisense sequences able to promote skipping of exon 23 of the murine dystrophine gene. After intramuscular or intra-arterial injection in mdx mice, we detected efficient skipping of the exon 23 and a long term rescue of dystrophin expression. We next evaluated this strategy in the canine GRMD model and showed the possibility to skip several exons, leading to a very large restoration of dystrophin in injected muscles. These promising results obtained on the mouse and canine models led us to develop the strategy on the human dystrophin gene and especially on the exon 51. We confirmed the skipping of the exon 51 both in vitro in patient myoblasts after transduction with the lentiviral vector and in vivo after intramuscular injection of an AAV-U7ex51 vector in the transgenic hDMD mouse. This study provides evidence on the efficiency of the U7snRNA mediated exon skipping strategy for Duchenne muscular dystrophy, that could concern more than 80% of patients and offers very promising tools for clinical treatment of DMD

Texto completoBien que plusieurs recherches ont souligné l’importance de la métacognition dans l’apprentissage et le développement musical, il existe peu d’études sur son utilisation dans l’enseignement collectif et hétérogène des instruments de musique dans le cadre des programmes de premier cycle en enseignement de la musique, notamment en ce qui concerne les instruments à vent de la famille des cuivres. Ainsi, dans le cadre d’un paradigme interprétatif / constructiviste, nous avons mené une recherche qualitative de nature exploratoire, dont l’objectif était de décrire l’utilisation des stratégies métacognitives dans les processus d’enseignement et d’apprentissage de ces instruments et de la lecture de la notation musicale, durant le cours Instrument complémentaire - Vents 1, d’un programme universitaire de premier cycle en enseignement de la musique, dans une université publique du nord-est du Brésil. Nous avons réalisé une intervention pédagogique de 32 heures par le biais de matériels didactiques et d’approches pédagogiques conçus dans l’intérêt de promouvoir, outre l’apprentissage de ces instruments et la lecture de la notation musicale, le développement métacognitif des étudiants. L’échantillon était composé de huit étudiants (n = 8), quatre femmes et quatre hommes, âgés de 19 à 22 ans. La collecte des données a été effectuée à partir d’un Questionnaire sur les données sociodémographiques et les connaissances musicales antérieures, de neuf Compte rendus de réflexion, d’un Questionnaire de réflexion et d’un Test pratique d’étude et de performance musicale. Pour l’analyse des données, nous avons utilisé la technique de l’analyse de contenu, à partir de la codification fondée sur les écrits de Garcia et Dubé (2012) et de Whitebread et al. (2009). De cette façon, nous avons identifié un total de 856 indices métacognitifs qui nous ont permis de décrire les caractéristiques du processus d’apprentissage, en soulignant les aspects liés aux connaissances métacognitives sur les personnes, les tâches et les stratégies, ainsi que la gestion des processus mentaux liés à la planification, au contrôle et à la régulation. Compte tenu des résultats de notre recherche, nous concluons que la métacognition peut être un outil important dans l’enseignement collectif des instruments de musique, permettant non seulement l’apprentissage et/ou le perfectionnement musical, mais aussi le développement de compétences pédagogiques.
Although several studies have emphasized the importance of metacognition in musical learning and development, only a small number have examined its use in the collective and heterogeneous teaching of musical instruments in undergraduate music education programs, particularly with respect to wind instruments of the brass family. Therefore, within the framework of an interpretive/constructivist paradigm, the goal of this qualitative research study of an exploratory nature was to describe the use of metacognitive strategies in the processes of teaching and learning these instruments and reading music notation, during the course Complementary Instrument - Winds 1, in an undergraduate program in music education at a public university in Northeastern Brazil. A 32-hour pedagogical intervention was carried out employing didactic materials and pedagogical approaches designed in the interest of promoting, in addition to learning these instruments and reading music notation, the metacognitive development of the students. The sample was comprised of eight students (n = 8), four females and four males, aged 19-22 years old. Data collection tools included a Questionnaire on Socio-demographic Data and Prior Musical Knowledge, nine Reflection Reports, a Reflection Questionnaire and a Music Study and Performance Practice Test. A content analysis was conducted using coding strategies based on the work of Garcia and Dubé (2012) and Whitebread et al. (2009). A total of 856 metacognitive indices were identified that allowed the researcher to describe the characteristics of the learning process, highlighting aspects related to metacognitive knowledge about people, tasks and strategies, as well as the management of mental processes related to planning, control and regulation. The results of this research study suggest that metacognition can be an important tool in the contexts of collective teaching of musical instruments, allowing not only for learning and/or musical development, but also for the development of pedagogical skills.
Embora pesquisas diversas tenham evidenciado a importância da metacognição no aprendizado e desenvolvimento musical, são escassos os estudos sobre seu uso no ensino coletivo e heterogêneo de instrumentos musicais no âmbito de cursos de licenciatura em música, principalmente no que se refere aos instrumentos de sopro da família dos metais. Assim, dentro de um paradigma interpretativo / construtivista, realizamos uma pesquisa qualitativa de natureza exploratória, cujo objetivo foi descrever o uso de estratégias metacognitivas nos processos de ensino e aprendizagem destes instrumentos e da leitura de notação musical, no âmbito da disciplina Instrumento Complementar – Sopros 1, de um curso de licenciatura em música de uma universidade pública do nordeste brasileiro. Contexto em que realizamos uma intervenção pedagógica de 32 horas/aula, mediada por materiais didáticos e abordagens pedagógicas concebidas com o interesse de promover, além do aprendizado destes instrumentos e da leitura de notação musical, o desenvolvimento metacognitivo dos estudantes. A amostra foi composta por oito alunos (n = 8), sendo quatro do sexo feminino e quatro do sexo masculino, com idades que variavam entre 19 e 22 anos. A coleta de dados foi feita a partir de um Questionário Sobre Dados Sociodemográficos e Conhecimentos Musicais Anteriores, nove Diários de Reflexão, um Questionário de Reflexão e um Teste Prático de Estudo e Performance Musical. Para análise dos dados utilizamos a técnica de Análise de Conteúdo, partindo de codificação fundamentada pelos artigos de Garcia e Dubé (2012) e Whitebread et al. (2009). Desta forma, identificamos um total de 856 indícios metacognitivos que nos permitiram descrever as características do processo de aprendizagem, ressaltando aspectos referentes aos conhecimentos metacognitivos sobre as pessoas, sobre as tarefas e sobre as estratégias, bem como, referentes à gestão de processos mentais vinculados ao planejamento, controle e regulação. Considerando os resultados de nossa pesquisa, concluímos que a metacognição pode ser uma importante ferramenta no ensino coletivo de instrumentos musicais, proporcionando não somente o aprendizado e/ou aprimoramento musical, mas também o desenvolvimento de competências pedagógicas.

Texto completoIf we take the following definition literally: « Reading is understanding », Reading Comprehension thus, becomes the sole significant measure to take into account when we evaluate the capacity to read a text.Indeed, the students who learn Arabic are unable to master semantic reading at the end of their Lycée (High school) cycle. When we consider the results of their oral exams, we note that they decipher a text without really being able to introduce, explain and analyse it. They do not really understand its meaning. They often spend their time talking about it at a superficial level, without being able to nuance their explanations. In the end, they remain no more than novice readers who can only decipher! These difficulties can also be noted with their written exams, therefore affecting the oral and comprehension of the students who can speak the dialect as well as those who cannot.Yet, The Common European Framework of Reference for Languages (CEFRL) clearly recommends a totally fresh approach whose aim is to rethink the objectives and the teaching methods of foreign languages, and most of all, it sets a common basis for the conceptions of curricula. Indeed, the CEFRL considers that the teaching/learning of a foreign language must be done through media or textbooks that take into account not only the social and cultural background, but also the language itself in its evolution and in its diglossic reality, unlike what is unfortunately still too often the case in Arabic language textbooks, which are randomly devised according to a normative, orthographic and grammatical inspiration. In chapter 6 of the CEFRL, the requirement is on methodology and can be epitomized in two questions: “How can we acquire and learn a new foreign language? What can be done to facilitate the learning and acquisition process?”That is how, from 2005 onwards, how some language textbooks designers started to devise methods, books, toolboxes, which in their introduction parts claim that they are productions that comply with the recommendations of the CEFRL, and labelled them A1, A2. These language textbooks unfortunately keep considering the acquisition of the reading skills as a deciphering or decoding operation. They are presented according to a linguistic and grammatical progression without taking into account the fact that a number of our students not only belong to heterogeneous social and cultural backgrounds, and that many come to the Arabic class already speaking an Arabic dialect, with its multiple regional varieties.As for other course books, honourable efforts can indeed be observed. Some designers take into account the different dialectal varieties by integrating some dialectal references so as to reassure the students who speak a dialect about the existing and possible bridges between the different language levels and registers of Arabic, and to enable the learners who do not speak a dialect to use expressions that will be socially understandable and ready to be used in order to make them aware of the diglossic problems of Arabic.It is true that “Official Instructions” recommend that Arabic teachers should leave an important place to dialectal varieties and to communication, but unfortunately, a lot of course books simply ignore all that wealth, for they are guided by an all normative approach.We are convinced today that French students (of Arabic origin or not) are not taught the Arabic language the same way and with the same objectives, Arabic students from Morocco, Lebanon or Egypt are

Texto completoThe thesis focuses on motifs of the circular code X, an error-correcting code found in protein-coding genes, which have the ability to synchronize the reading frame. We first investigated the evolutionary conservation of X motifs in genes of different species and identified specific selective pressures to maintain them. We also identified a set of universal X motifs in ribosomal RNAs, which are located in important functional regions of the ribosome and suggest that circular codes represented an important step in the emergence of the standard genetic code (SGC). Then, we investigated the functional role of X motifs in modern translation processes and identified a strong correlation between X motif enrichment in genes and translation levels. Finally, we compared the frameshift optimality of the circular code X with the SGC and other maximal circular codes, and identified a new functionality of the code X in minimizing the effects of translation errors after frameshift events

Texto completoMany researchers have highlighted the major roles of top and middle managers in the strategy process (Jarzabkowski, 2005 ; Ikävalko, 2005; Mantere, 2005), yet rare are those who describe these roles precisely and concretely. On the basis of a literature review, this research first emphasizes the need for researchers in strategic management to adopt more dynamic and integrating frames for the future when studying phenomenon such as managerial roles. It appears particularly fruitful to take behavioral and socio-psychological dimensions into account. Thus, from this perspective, relying on the young Strategy-as-Practice research field, combined with the Theory of Social Representations (Moscovici, 1961), this research proposes construction of a theoretical model intended to explain the constitution of managerial roles in strategy process. More largely, this theoretical model is anchored in critical management studies, especially those linked with Foucault’s work. From a discursive perspective, the present research invites consideration that the strategic discourses to which managers are exposed influence managerial roles in strategy process via the social representations these discourses contribute to developing. These roles are simultaneously constructed at the crossroads between strategic discourses and strategic practices. The theoretical model proposed here then leads to an empirical study of managerial roles in strategy process. This study is notably based on the three steps proposed by Clémence et al. (1994) to study social representations. A 79 item questionnaire was constructed and administered to top and middle managers : 1033 complete responses were collected. Some items enable highlighting manager's social representations concerning strategy; others are intended to interpret these social representations in terms of managerial roles. On the basis of both qualitative and quantitative methodologies, four conceptions of managerial role have been emphasized. This typology highlights the influence of strategic discourses, and socio-psychological and socio-demographic variables in the constitution of managerial roles in strategy process

Texto completoThe early novels of John Dos Passos are part of the modernist literary movement. They are characterized by an original organization of the words on the page. The text is no longer a monolithic block, but is divided into sections and its separations are highlighted by a vast paratext. It also contains a variety of collages presented in various styles and in different fonts. These observations are the starting point of this research, which focuses on the materiality of the text in the early novels of John Dos Passos, Manhattan Transfer and the trilogy USA, published between 1925 and 1938. These novels question the gaze of the reader on the text, showing what is placed in front of and around the eye of the observer. The reader becomes aware of what influences his vision. The novel displays its internal structure and various thresholds, allowing the reader to continue or to stop reading, to go backwards and to create connections between the different sections of the text. The novel displays how it has been constructed, highlighting that it is the product of the rearrangement of previous texts. This visible internal structure is also built out of gaps and empty spaces. It reflects the tension between the ambition to be exhaustive, to contain everything and to acknowledge the difficulty to tell a story. The interplay with the blank space on the page, the use of punctuation and phonetic indicators reinforce the written aspect of the text, while at the same time making the words stand off the page and resistant to closure

Texto completoDuring my thesis, my researches have mainly concerned the modular properties of dysferlin, a protein involved in muscular dystrophies, in order to identify the most promising therapeutic approaches.My work has been oriented within two research axes, a mini-proteins approach and an exon-skipping approach, both based on proofs of concept obtained in patients. We first tested an exon-skipping approach for the exon 32 of DYSF, based on the identification of a protein lacking the encoded part of this exon, in an asymptomatic person. We have established the functionality of this truncated protein by allowing its expression in dysferlin-deficient patients' cells. This suggests that the C2D domain (encoded by exons 31 to 34) is not essential for dysferlin since the absence of a part of it don't block its functionality.In the same time we analyzed the characteristics of the others domains of dysferlin, by creating miniproteins containing different combinations of domains. By studying the abilities of this constructs, we have showed that the C-ter part of dysferlin (composed of the last C2 domains and the transmembrane domain) was essential and sufficient for the functionality of dysferlin in muscle. All these results demonstrate that several domains of dysferlin are dispensable, paving the way for studying gene therapy approaches, based on minigenes or exon-skipping for dysferlinopathies

Texto completoThe analysis of Julien Gracq's Un balcon en forêt is based on propositions elaborated in Temps et récit by Paul Ricoeur, who considers fiction as the "time-keeper". The study aims at linking comprehension, interpretation and explanation of the functioning of the text (the "configuration"), at questioning the relation between history (the "phoney war") and fiction, ans at making clear the operation of "refiguration" performed by the reader to build meaning. It first proposes an interpretation of the frame of the fiction in its paratextual organization and its strategic places. The analysis, then, strives to demonstrate how the gracquian text makes the marks of enunciation in fiction waver. The study of temporality does demonstrate that beyond the linearity of fiction a subtle game of anachronies appears, which tends to refuse every kind of referential reading, as the text weaves singular and iterative narrations. As time seems to have stopped, space then occupies an important place : conflictual, it takes on a symbolical and poetical signification, fundamental for the comprehension of the fiction. The study of diegesis in its three elements (pragmatic, cognitive, thymic) shows that in Un balcon en forêt, action is superseded by what is felt and suffered. Thus, a very strict analysis of the textual manifestation allows us to demonstrate how fictional writing is contested by poetical writing, which prefers a paradigmatic structuring. However, even if the gracquian text works in a "closed circumference" it avoids all formalist closure ; on one hand, it appears as a sequence of "imaginative variations on time", on the other hand it leaves initiative to the reader who has become the true producer of an ambiguous and elliptical fiction, and finally, it is "fertilized" by literature and makes sense with reference to an intertext which is always solicited

Texto completoAt a time where welfare at work seems to be a priority for managers and organizations, and where mobile technologies allow permanent access to business information, increasing the porosity of borders between private and professional life; this thesis questions the impact of mobile technologies on employees’ psychological health and more specifically about their well-being (effects of nomadic technologies on the border between private and professional life, interpersonal relations, etc.). To explore this issue, we mobilized the Resources Conservation Theory (Hobfoll, 1989), a motivational theory which is based on the variety of resources available to a person in order to maintain, or reach, a situation of well-being and balance. The sales teams of a large French group, which were fitted in 2012 with Mobile Technologies (MT) constitute our field of investigation. The literature review and the results of an exploratory qualitative analysis, conducted using a semi-structured interview, with 34 commercials and managers, lead us to submit a structural model of the influence of mobile technologies on well-being at work, combining organizational, individual, and technological variables. This model was tested via a survey at a national level. From the 850 questionnaires which were sent, 400 usable responses were studied. First, our empirical results lead us to identify five categories of mobile technologies use, both depending on the nature of the technological support and on the performed task. The five obtained variables, became the input variables, through the modeling stage realized by the structural equations method. The test of research hypotheses emphasizes the role of certain resources such as autonomy, the meaning of work, the usefulness of technologies or organizational social support, especially to cope with the invasion of technology into the private sphere and the pressure related to its use. Overall, our results suggest that, despite the changes brought by the arrival of MT, there is no duality regarding the direct or indirect effect of these technologies on the stress level or on the welfare at work. Few interviewees are adamant on purely negative or purely positive impact of mobile technologies about welfare at work. However, a large majority of commercial and interviewed managers considered that the arrival of mobile technologies has "somewhat" favored well-being at work, despite the increased workload and technological invasion that it induced. In the end, MT seem to constitute constrained resources. To conclude this work, the theoretical and managerial contributions are presented as well as future opportunities of research

The first one, close to the notions of objective, interest, utility, forms the foundations of a classical vision of the manager’s work. However, a second one, based on Aristotle, Thomas Aquinas and Spaemann’s philosophies, arose. This alternative conception differs from objective and interest as it is not elaborated by the subject. It allows a different view on three essential dimensions of the manager’s work: the management by objectives, the body of contradictory requirements that built up on the manager and the dilemma between ethics and efficiency. Finally, a field survey showed that this alternative conception of purpose, linked to virtue-based ethics, has a certain place in the FDJ’s managers’ message, together with consequentialism, which expresses the utilitarian conception of purpose, and deontological ethics, which evacuates any purpose

Among them, viral diseases (re)emerge following environmental, climatic and societal changes: Ebola, Dengue and Zika viruses have recently been the object of special attention. The development of safe and efficient vaccines against emerging viruses is a major challenge for global public health. This thesis work is in line with this issue. Using the yellow fever virus (YFV, genus Flavivirus) as a model, we tried to define new strategies for the design of live-attenuated vaccines for viral infections prevention. After a genomic analysis that allowed to go further into a procedure for virus modification named “re-encoding”, we generated and characterised both in vitro and in vivo mutant strains of YFV. In parallel, a rodent model was set up to test in vivo both the safety and the protective efficiency of the re-encoded viruses

Texto completoThe family meal is a relatively unknown object of study being developed in different scientific fields such as public health, sociology, or anthropology. Regarding eating disorders, the frequency of the number of meals shared with the family is inversely proportional to the emergence of such disorders as well as other psychiatric disorders in adolescence such as violent behavioral disorders, depression and suicidal thoughts. Notwithstanding its frequency, the meal is an essential moment for the development of an individual – also psychologically. Within the family life, it is a daily time of sharing and meeting which brings together the children and those who are close to them. The meal is a moment of privileged exchange, rich in behaviors and interactions: it certainly mirrors the relationship between a family and food, but it accounts even more for the bonds between the family members. The table scene constantly proposes a narrative animating and featuring those who partake of the meal. In this context, several authors have introduced a family meal as part of children’s psychiatric care. Our thesis work will be divided into three main parts. A first theoretical part will report on the role of family meals in the context of psychiatric care for adolescents. A second part will describe the program called "therapeutic family meal" as it is set up in our ward: we will explain how the therapeutic family meal is used by our medical team specialized in the care of teenagers suffering from anorexia nervosa. A third part will develop on a research conducted among caregivers participating in this program. Concerning the involved caregivers, their experience –both individual and collective– has been the subject of our research work. According to a qualitative methodology resulting from the grounded theory and from interviews made with a reasoned sample of participants in this program, we will develop, as results, the different themes characterizing this experience. Our analysis will articulate these themes through several concepts resulting from the symbolic interactions that we will have defined beforehand. Finally, we will address the cultural and methodological limits of our approach. At the end of this last part, we will discuss the concept of paradoxical proximity that emerges from the caring practice in this particular context. We will also discuss the transition at the end of this hospitalization and the setting of a family therapeutic meal involving families and caregivers. This transmission is part of the passage rite embodied by hospitalization for the sick teenager and her family. The issues concerned by the therapeutic family meal, necessary to better frame it, are the ritual approach, a socio-anthropological reflection, the question about the status of the sick teenager (at the hospital and at home), the transition between the hospital and home, and the nature of the caregiver-receiver relationship. Based on two clinical cases, and on our interviews with caregivers, we will discuss the psychopathological contributions of this practice and the new care standpoints that it implies

Texto completoCoding CAG repeat disorders have been associated with nine neurodegenerative disorders. Our group has previously shown that during the translation of mutant ataxine-3 (Atx3), the protein involved in Spinocerebellar Ataxia type 3 (SCA3), a ribosomal frameshift occurs and leads to the reading of a GCA frame rather than a CAG frame. This new reading frame causes the production of polyalanine in the polyglutamine peptide which increases its toxicity. Ribosomal frameshifts are known to occur in all organisms but little is known about this phenomenon in human. To study ribosomal frameshift along the ATXN3 transcript, we generated a transgenic Drosophila model in which we looked at the toxicity of the mRNA. Also, we developed a toeprinting assay to precisely evaluate where the change of reading frame occurs along the mRNA. Our results suggest that the toxicity observed in our Drosophila model results from the production of polyalanine and not from the presence of the mRNA per se. Moreover, the change in reading frame seems to occur at the 48th CAG codon and this pausing of the ribosome also occurs in other polyQ tracts. Because CAG repeat disorders share many characteristics, an alteration of the frameshift could alleviate symptoms of SCA3 patients as well as of many other diseases with coding CAG repeats.

Texto completoThe human immunodeficiency virus type 1 (HIV-1) is responsible for the acquired immune deficiency syndrome (AIDS). HIV-1 develops a resistance towards the inhibitors used to treat infected patients. It is thus important to identify new targets for the development of novel antiretroviral agents. The aim of our work was to better characterize the programmed -1 ribosomal frameshift which generates the precursor of HIV-1 enzymes. The frameshift occurs at a specific sequence of HIV-1 full-length messenger RNA (mRNA), the slippery sequence, and is performed by a minority of the ribosomes translating this mRNA. The frameshift efficiency is controlled by the frameshift stimulatory signal (FSS), an irregular stem-loop located downstream of the slippery sequence. FSS structure is unfolded by every ribosome translating this region and can refold afterwards. We showed that HIV-1 frameshift efficiency is affected by changes in the rate of translation initiation. We transfected Jurkat-T and HEK 293T cells with a bicistronic reporter that contains the frameshift region of HIV-1 between the Renilla luciferase (Rluc) and the firefly luciferase (Fluc) genes. Rluc is produced by all ribosomes translating this reporter whereas only ribosomes that make a –1 frameshift produce Fluc. The translation of the reporter is initiated via a cap-dependant mode, like the majority of cellular mRNAs. We first determined the effect of three inhibitors of translation initiation. We showed that their presence increases the frameshift efficiency. We next determined the impact of the TAR stem loop, which is located at the 5’end of every HIV-1 mRNA. TAR is known to impair the binding of the small subunit of the ribosome (40S) to the mRNA. TAR also modulates the activity of the double-stranded RNA-dependent protein kinase (PKR). When PKR is activated, it phosphorylates the eukaryotic initiation factor 2 (eIF2), inhibiting translation initiation. The inhibition of PKR has the opposite effect. We studied the effect of TAR on PKR by positioning TAR at a distance of the 5’ end where it cannot interfere with the binding of the 40S. Our results showed that a small amount of TAR, which activates PKR, increases the frameshift efficiency whereas a large amount of TAR, which inhibits PKR, decreases it. A model is presented where the variations of translation initiation modulate HIV-1 frameshift efficiency by altering the distance between the elongating ribosomes. This influences the probability that these ribosomes encounter or not a folded FSS. We next observed the effect of the 5’ untranslated region (UTR) of HIV-1 full length mRNA on its frameshift efficiency. This 5’UTR contains several structured parts, including TAR at the 5’end, which can inhibit translation initiation. This mRNA has a cap and an internal ribosome entry site (IRES) and could then use a cap dependent and an IRES-dependent mode of translation initiation. We replaced the 5’UTR of our bicistronic reporter mRNA by the complete 5’UTR of HIV-1 full-length mRNA or a part of it. Our results showed that the presence of the complete 5’UTR inhibits cap-dependent initiation of translation and increases the frameshift efficiency. Those effects are mostly due to the presence of TAR followed by a Poly(A) stem-loop. We also constructed a tricistronic reporter where the ribosomes translating the luciferases have to use an IRES-dependent initiation mode. The rate of this initiation was low and the frameshift efficiency obtained was also low. We proposed a hypothesis accounting for this situation. We also observed that when both initiation modes are available, the cap-dependent mode seems to be highly favored. Finally, we studied the impact of the Tat viral protein on translation initiation and frameshift efficiency. We showed that the presence of Tat increases translation initiation and decreases the frameshift efficiency. Those effects are more important when TAR is present at the 5’end of mRNA. We propose a model explaining the effects of Tat on translation initiation by the inhibition of PKR and by changes in the expression of cellular proteins that are able to unfold TAR. Our results allow us to better understand the mechanisms controlling HIV-1 frameshift, which will help in the development of drugs targeting the HIV-1 frameshift.

Texto completoThis research reveals how textual genres can be exploited in digital work environments to improve the textual practices of managers and secretaries in the context of a municipality and the Canadian federal government. The first objective of this research assesses the suitability of digital work environments to support the textual practices of managers and secretaries in their reading, writing and manipulation of texts. The second objective describes the various roles of textual genre during the managerial and secretarial textual practices. Using email as a focal point, the third objective examines how genre can be exploited to advance the benefits of textual practices in the digital work environments. This qualitative research entails a two-phase methodology. By the study of 17 secretaries and 17 managers, the first phase consists of a thorough examination of the current textual practices in the Canadian federal government and municipal contexts and the difficulties encountered during these practices. This phase also considers the various roles of genre in the digital work environments along with the salient clues sought during email management. This study deployed three data collection techniques: semi-structured interviews, diary journals and cognitive inquiries. The results are examined using several qualitative content analysis techniques. The second phase of this research consists of developing an email processing sequence to further expand our understanding of textual genre and its exploitation in the design of digital work environments. The data for this phase uses a corpus of 1703 messages developed from a sample of two governmental managers. The results provide an encompassing overview of practices relating to the reading, writing and manipulation of texts that are both common and specific to managers and secretaries. With over 40% of events recorded in the diary journal relating to email, the importance of this type of system in digital work environments is clearly emphasized. The difficulties encountered in the digital work environments are also described. The role of genre during textual practices is examined according to a matrix illustrating both the individual and collective dimensions of genre in addition to its three main facets: the form, the content and the purpose. We present next an analytic framework of the prominent cues affecting email management to summarize the process of interpreting messages by the recipient. A typology of the categorization patterns of managers is also developed and used in a statistical experiment aiming to automatically describe and categorize email. Resulting from this experiment, we observe specific linguistic behaviours that characterize each email category. It is also revealed that automatic categorization based on message lexicon is more efficient than non-lexical categorization. At the conclusion of this research, we suggest to enrich the traditional human-computer interaction paradigm with a semiotics of genre in the digital work environments. The study also offers a reflection regarding email membership to a specific genre using the theoretical concepts of hypergenre, genre and sub-genre. The success of the automatic categorization of email according to genre-related facets (the content, the form and the purpose) uncovers valuable insights and perspectives in designing digital work environments with the objective of facilitating the vital performance of textual practices by employees.

Texto completoEighteen severe human diseases have thus far been associated with trinucleotide repeat (TNR) expansions coding for either polyalanine (encoded by a GCN repeat tract) or polyglutamine (encoded by a CAG repeat tract) in specific proteins. Among them, oculopharyngeal muscular dystrophy (OPMD), spinocerebellar ataxia type-3 (SCA3), and Huntington’s disease (HD) are late-onset autosomal-dominant disorders characterised by the presence of intranuclear inclusions (INIs). We have previously identified the OPMD causative mutation as a small expansion (2 to 7) of a GCG repeat tract in the PABPN1 gene. In addition, we and others have reported the occurrence of -1 ribosomal frameshifting events in expanded CAG repeat tracts in the ATXN3 (SCA3) and HTT (HD) genes, which result in the translation of a hybrid CAG/GCA repeat tract and the production of a polyalanine-containing peptide. Data from OPMD suggests that polyalanine-induced toxicity is very sensitive to the dosage and length of the alanine stretch. To validate our ATXN3 -1 frameshifting hypothesis in animal models, we set out to reproduce our findings in Drosophila and mammalian neurons. Our results show that the transgenic expression of expanded CAG repeat tract ATXN3 cDNA led to -1 frameshifting events, and that these events are deleterious. Conversely, the expression of polyglutamine-encoding expanded CAA repeat tract ATXN3 cDNA was neither frameshifted nor toxic. Furthermore, expanded CAG repeat tract ATXN3 mRNA does not contribute to the toxicity observed in our models. These observations indicate that expanded polyglutamine repeats in Drosophila and mammalian neuron models of SCA3 are insufficient for the development of a phenotype. Hence, we propose that -1 ribosomal frameshifting contributes to the toxicity associated with CAG repeat tract expansions in the ATXN3 gene. To further investigate ribosomal frameshifting in expanded CAG repeat tract diseases, we sought to create an antibody capable of detecting the frameshifted product. Here we report the characterization of a polyclonal antibody that selectively recognizes pathological expansions of polyalanine in the protein implicated in OPMD, PABPN1. Furthermore, our antibody also detects the presence of alanine proteins in the intranuclear inclusions (INIs) of SCA3 and HD patient samples.

Texto completoIntroduction: Alternative reading frames (ARFs) are used by multiple viruses in order to generate different proteins from a single nucleotide sequence. Cryptic epitopes, which comprise antigens derived from proteins encoded in ARFs, have recently been the focus of studies pertaining to antiviral immunity and cytotoxic T lymphocytes. Methodology: In order to verify the immunological potential of epitopes encoded in programmed ARFs, three cassettes were constructed to permit the expression of three welldescribed epitopes (GAG77–85 epitope of human immunodeficiency virus type 1; NS31406-1415 epitope of hepatitis C virus; core18-27 epitope of hepatitis B virus) from three overlapping reading frames. The first cassette permits alternative translation initiation, the second cassette includes signals inducing ribosomal frameshifting and the third cassette serves as a control. These elements were introduced into adenoviral vectors. Recombinant adenoviruses were used to immunize C57BL/6 transgenic mice expressing HLA-A*0201 and HLA-DR1. The immune response induced was measured one week following immunization using IFN ELISpot assays. Results: In the context of vaccine cassettes, peptides derived from alternative translation initiation and ribosomal frameshifting can be expressed and detected by the immune system in an animal model. Conclusion: These findings suggest the possibility of designing vaccination strategies in the hope of preventing or curing certain diseases associated with chronic viral infections, such as those caused by human immunodeficiency virus and hepatitis C virus.

Texto completoThis study aims to review the biosciences component of preregistration nursing programmes in higher education institutions across the UK through the experiences and perceptions of lecturers involved in nursing education. Studies suggest that some qualified nurses lack confidence in explaining the bio-scientific rationale for their clinical practice. Biosciences can be difficult to understand and integrate into clinical decision-making and require protected time within preregistration nurse education. In the absence of explicit national guidelines, it is unclear as to the depth and extent biosciences are taught across different institutions and the level achieved at the point of registration. A survey approach was adopted to generate quantitative and qualitative feedback. Data were collected using a semi-structured questionnaire seeking the experiences and views of lecturers involved in teaching biosciences to nursing students across the UK. Data received from 10 institutions were analysed using descriptive statistics and thematic analysis. Lecturers reported that the hours of taught biosciences ranged from 20-113 hours, principally within the first year. This represents between 0.4-2.4% of time within a preregistration nursing programme (4600 hours). Large group lectures predominate, supplemented by smaller group or practical work, and online materials. The biosciences are assessed specifically in half the institutions surveyed and as part of integrated assessments in the rest. In relation to student feedback, all respondents stated that students consistently requested more time and greater priority for biosciences in their programme. This survey suggests that the number of hours spent teaching biosciences is minimal and varies widely between higher education institutions. All respondents expressed concern about the challenges of teaching difficult bio-scientific concepts to large groups in such a limited time and called for greater clarity in national guidelines to ensure that all nurses are adequately educated and assessed in bioscience subjects. Failure to understand the biosciences underpinning care has implications for safe and competent nursing.
