Resumen
The purpose of this study was to identify faculty teaching goals at senior research universities and to determine variables which could be used to predict these goals. The eight disciplines used in this study were: accounting, chemistry, computer science, economics, English, math, mechanical engineering, and psychology. The independent variables were: gender, academic rank, course level, tenure, and four Biglan categories (pure-hard, pure-soft, applied-hard, and applied-soft). Using the Teaching Goals Inventory (Angelo & Cross, 1993), the teaching goals and primary teaching role of faculty at senior research universities across eight academic disciplines were identified (N = 352). Through a principal axis factor analysis, seven factors emerged for the data gathered from the Teaching Goals Inventory, results of which were slightly different from that of previous research. Further analyses indicated differences between the teaching goals and primary teaching role of faculty at senior research universities, community colleges, and four-year colleges. Seven one-way ANOVAs and subsequent post-hoc comparisons were conducted which indicated significant differences (p < .05) among the factor means across the eight academic disciplines. Comparisons of the primary teaching role across the eight academic disciplines also indicated significant differences (p < .05) in the percentage of faculty selecting each of the six roles. Furthermore, this study provides additional evidence to support the theory that the Biglan categories help explain the differences in teaching goals across academic disciplines. Significant differences (p < .05) were detected in the teaching goals and primary teaching role of senior research university faculty across the four Biglan categories. Through regression analyses, three of the four Biglan categories, gender, and level of course entered as predictors of teaching goals. Academic rank and tenure did not enter into any of the regression equations; however, further analyses indicated these variables were intercorrelated with several other independent variables. Implications for these findings are discussed.

Ayant vécu de 1771 à 1841, Édouard Bignon incarne la génération d’hommes qui a traversé, non sans encombre, la Révolution, le Directoire, le Consulat, l’Empire, la Restauration et la monarchie de Juillet. Sa carrière est le reflet des changements politiques français. Précepteur à la fin de l’Ancien Régime, il s’enrôle dans l’armée en 1793 pour fuir les proscriptions de la Terreur. Sous le Directoire, en 1797, il intègre la carrière diplomatique au sein de laquelle il reste dix-neuf années, jusqu’en 1815 : il y connaît une progression continue, passant du rang de second secrétaire de légation en 1797 à celui de ministre plénipotentiaire en Pologne en 1813. Après avoir participé aux Cent-Jours, il se retire de la vie publique. Deux ans après Waterloo, il est élu député et siège dans les rangs de l’opposition libérale jusqu’à la chute de la monarchie bourbonienne. Indépendamment de ses mandats de députés, il se lance dans une carrière de publiciste et d’historien qui lui assure une notoriété grandissante. En effet, dans son testament, Napoléon Ier lui a commandé une histoire diplomatique de la France à laquelle Bignon s’attelle dès 1821. Divisée en quatorze tomes, cette Histoire de France paraît à partir de 1829. L’année suivante, Bignon participe à la chute de Charles X et devient proche des gouvernements de Louis-Philippe : il est nommé pair de France en 1837, couronnement d’une longue carrière au service de la France. Cette étude biographique, qui embrasse la totalité de sa carrière, entend mettre à l’honneur le parcours d’un diplomate napoléonien à l’envergure secondaire, les activités d’un parlementaire sous la Restauration et la monarchie Juillet, et l’originalité d’un historien de l’Empire
Born in 1771 and died in 1841, Édouard Bignon embodies the generation who has live, with some trouble, through the French Revolution, the Directory, the Consulate, the First Empire, the Restoration and the July Monarchy. His career reflects all the French political changes. A private tutor, he enlisted in 1793 while trying to avoid being arrested. During the Directory, in 1797, he joined the Foreign Office and stayed in this departement until 1815 : he was constantly promoted, from the position of 2nd secretary in 1797 to minister plenipotentiary in Poland in 1813. As he joined the ranks of Napoleon during The Hundred Days, he had to retire from public life once Louis XVIII came back. Two years after Waterloo, he became a member of Parliament and sat among the liberals until the Bourbons fall. Separately from his political activities, he published several historical books which rose his popularity. In his will, Napoleon asked him to write the diplomatic history of his reign. Begun in 1821, the Histoire de France was issued from 1829 to 1850 and split into 14 volumes. In 1830, he contributed to the July Monarchy’s birth and became one of its support in Parliament : Louis-Philippe rewarded him and appointed him as peer of France in 1837, as a public recognition for his duties. This study which tackles all Bignon’s activities, aims to emphasize a medium scale diplomatic experience, parliamentarian’s occupations during the Restoration and the July Monarchy and Napoleon historian’s originality

A contact structure is a maximally non-integrable hyperplane field $\xi$ on an odd-dimensional manifold $M$. In $3$-dimensional contact geometry, there is a fundamental dichotomy, where a contact structure is either tight or overtwisted. Making use of Giroux's correspondence between contact structures and open books for $3$-dimensional manifolds, Honda, Kazez, and Mat\'{i}c proved that verifying whether a mapping class is right-veering or not gives a way of detecting tightness of the compatible contact structure. As a counter-part to right-veering mapping classes, right-veering closed braids have been studied by Baldwin and others. Ito and Kawamuro have shown how various results on open books can be translated to results on closed braids; introducing the notion of quasi right-veering closed braids to provide a sufficient condition which guarantees tightness. We use the related concept of $P$-bigon right-veeringness for closed braids to show that given a $3$-dimensional contact manifold $(M, \xi)$ supported by an open book $(S, \phi)$, if $L \subset (M, \xi)$ is a non-$P$-bigon right-veering transverse link in pure braid position with respect to $(S, \phi)$, performing $0$-surgery along $L$ produces an overtwisted contact manifold $(M', \xi')$. Furthermore, if we suppose $L \subset (M, \xi)$ is a pure and non-quasi right-veering braid with respect to $(S, \phi)$, performing $p$-surgery along $L$, for $p \geq 0$, gives rise to an open book $(S', \phi')$ which supports an overtwisted contact manifold $(M', \xi')$.

Actualmente, se dan cerca de 26000 viajes en el distrito de San Juan de Lurigancho y eso sólo considerando a las cuatro asociaciones de mototaxis más grandes del distrito. Se sabe que existen más de quince y que cada una tiene, en promedio, entre 80 y 120 conductores que forman parte de ellas. Por otro lado, todos los conductores usan un Smartphone, mas no lo utilizan para buscar mayores ingresos en el negocio. Entonces, tomando en cuenta que el transporte en tramos cortos en el distrito a través de estos vehículos es tan frecuentes, de hecho, son los más utilizados y existe una gran demanda por ellos llega BIGA, una propuesta de negocio que busca revolucionar el transporte en tramos cortos. Nuestra propuesta de valor se basa en la seguridad y disponibilidad y responde a las necesidades actuales de la población de transportarse en vehículos con mayor confianza, seguridad y no verse expuestos a asaltos o accidentes. Sin embargo, lo que hace realmente novedosa a esta propuesta es su innovación tecnológica. Con el boom de UBER y su éxito rotundo en diferentes países, buscamos aplicar la misma fórmula, sólo que orientada a nuevas necesidades en sectores diferentes. Así, nuestra plataforma permitirá unir a conductores de mototaxis con clientes que busquen transportarse de un punto a otro. También, buscamos combatir la informalidad y dar la oportunidad a nuestros asociados de incrementar sus ingresos y redefinir su imagen ante los pasajeros, lo cual les permitirá crecer económicamente y formar parte de una red que promete ser un éxito rotundo. Resumen

Biglycan is a small, leucine-rich repeat proteoglycan that has been shown to be elevated in the intervertebral discs (IVDs) of older adults (Cs-Szabo, 2002) and can interact with many different signaling molecules and structural proteins. TNF-alpha (TNF) is a general modulator of inflammation and may be a trigger in the chronic inflammatory response that leads to disc degeneration. Since biglycan has been shown to interact with TNFalpha (Tufvesson and Westergren-Thorsson, 2002), it may play a role in modulating its response in the IVD. Western (NFkB) and RT-PCR (MMP-1, -13, NGF and VEGF) assays were performed on both human and bovine cultured cells from IVD tissue. Treatment with biglycan alone was found to be almost indistinguishable from control groups; while biglycan in combination with TNF-alpha was able to significantly reduce transcription and expression response when compared with just TNF-alpha. Biglycan may help modulate the effects of TNF-alpha in the IVD.

Changes in extracellular matrix (ECM) deposition along with increases in airway smooth muscle (ASM) mass are characteristic features of airway remodeling which contribute to asthma pathophysiology. Modifications in ECM include increased collagen deposition and altered proteoglycans (PGs) levels. While biglycan, a small leucine-rich PG (SLRP), deposition is generally increased, changes in decorin, another SLRP, are more variable. The interaction between the cell and the surrounding ECM triggers numerous cellular responses, which modulate functions such as survival, proliferation and differentiation. Since decorin and biglycan are known to influence cell behaviour in various types of cells, we hypothesized that these molecules would affect human airway smooth muscle cell (HASMC) function. We sought to determine the effects of decorin and biglycan on HASMC proliferation, apoptosis and attachment, and to explore the putative mechanisms responsible for these effects. Collagen type I was used as a control.Culture on a decorin matrix caused a decrease in HASMC number, which was attributed to a decrease in proliferation and an increase in apoptosis. Culture on biglycan caused an initial decrease in cell number that was not sustained. Collagen caused a significant increase in cell number. Neither biglycan nor collagen caused changes in proliferation or apoptosis. Subsequent experiments showed that cell anchorage to decorin and biglycan matrices caused abnormal focal adhesion and cytoskeletal organization, as well as, increases in alpha2 integrin protein levels as compared to cells seeded on collagen or plastic. To assess integrin activation, focal adhesion kinase (FAK) protein levels and activation were measured. FAK levels were significantly decreased compared to cells cultured on plastic or collagen I, but activation levels were unchanged.These studies demonstrate that PGs have differential effects on HASMC growth. Whereas decorin reduces ASM proliferation and increases apoptosis, biglycan has no effect. Cell-matrix adhesions were irregular in HASMCs seeded on decorin or biglycan compared to cells on collagen I or plastic. Decreases in decorin in the asthmatic airway wall, as observed in fatal asthma cases, may permit more exuberant ASM hyperplasia, suggesting that decorin's presence could serve a protective role by modulating the increase in ASM mass in asthma.

Des expériences ultérieures ont démontré que l'ancrage des cellules musculaires lisses des voies aériennes sur des matrices de décorine et de biglycane était anormal, ainsi que la formation d'adhésions focales et l'organisation du cytosquelette, en comparaison avec les cellules ensemencées sur du collagène ou du plastique. De plus, l'augmentation d'expression protéique de la sous-unité alpha2 d'intégrine a également été observée. Pour évaluer l'activation des intégrines, le niveau de protéine, ainsi que l'activation de la kinase des adhésions focales (FAK) ont été mesurés. En présence de décorine, l'expression protéique de FAK était significativement diminuée comparativement aux niveaux produits par les cellules ensemencées sur du plastique ou du collagène bien que le niveau d'activation était inchangé. Ces études démontrent que les PGs ont des effets distincts sur la croissance des cellules musculaires lisses des voies aériennes: la décorine a réduit la prolifération et a augmenté l'apoptose des cellules, tandis que le biglycane n'a pas modifié ces paramètres. Les adhésions cellules-matrice des cellules musculaires lisses des voies aériennes ensemencées sur la décorine ou le biglycane étaient irrégulières par rapport aux cellules sur le collagène de type I ou le plastique. La diminution de la quantité de décorine dans la paroi des voies aériennes observée dans les cas d'asthme fatal pourrait entraîner une hyperplasie plus prononcée des cellules musculaires lisses des voies aériennes. Ceci suggère que la présence de la décorine pourrait jouer un rôle protectif dans l'asthme, en modulant l'augmentation de la masse du muscle lisse des voies aériennes.

Cardiovascular disease is now the leading cause of death worldwide. Serum amyloid A (SAA), a positive acute phase reactant, along with C-reactive protein is used clinically as a marker of cardiovascular disease risk. However, recent data has shed light on a possible causal role of SAA in the development of atherosclerosis, the most pervasive form of cardiovascular disease. Several inflammatory diseases such as diabetes and obesity are known to confer increased risk of developing cardiovascular disease. Individuals with these diseases all have modest but persistent elevation of SAA. To determine if SAA caused the development of atherosclerosis, apoe-/-chow fed mice were injected with either an adenoviral vector expressing human SAA1 (ad-hSAA1), a null adenoviral vector (ad-Null) or saline. Human SAA levels rapidly increased, albeit briefly then returned to baseline within 14 days in mice that received ad-hSAA1. After 16 weeks, mice that received ad-hSAA1 had significantly increased atherosclerosis compared to controls on the aortic intimal surface (p<0.0001), aortic sinus (p<0.05) and the brachiocephalic artery (p<0.05). According to the “response to retention” hypothesis; lipoprotein retention by vascular wall proteoglycans is a key initiating event in the development of atherosclerosis. We previously reported that SAA-stimulated vascular smooth muscle cells expressed biglycan with increased glycosaminoglycan chain length and increased binding affinity for low density lipoprotein. To further test the role of biglycan on the development of atherosclerosis we generated biglycan transgenic mice. These mice were crossed to the ldlr-/- mouse on a C57BL/6 background and fed a pro-atherogenic western diet for 12 weeks. There was a significant increase in atherosclerotic lesion area on the aortic intimal surface (p<0.05) and the aortic sinus (p<0.006), as well as a significant correlation between vascular biglycan content and aortic sinus atherosclerotic lesion area (p<0.0001). These data demonstrate that transiently increased SAA resulted in increased atherosclerosis compared to control mice, possibly via increased vascular biglycan content. In support of this we found that biglycan transgenic mice had significantly increased atherosclerosis compared to wildtype controls, likely through increased lipid retention in the vascular wall.

Resumen
The topic of this PhD thesis was to study organelles in plant cells, analysing in particular the mitochondrial morphology. Recent papers have, in fact, reported that regulation of mitochondrial morphology is important not only for maintaining mitochondrial number during cell division, and mitochondrial distribution within each cell (Westermann, 2010; Logan, 2003), but also for executing many biological functions. In animals, a shift in mitochondrial morphology towards the organelle fragmentation is an early event during apoptosis (Suen et al., 2008). In plants, a link between mitochondrial morphology and senescence-associated cell death (i.e. a genetically controlled programme of self destruction) has been reported in Medicago truncatula cultured cells (Zottini et al., 2006). The understanding of molecular mechanisms, used by plants to regulate senescence, might allow biotechnological applications through genetic manipulations of key elements affecting senescence and provide interesting applicative outputs, especially in agronomically relevant species. In light of these findings, I focused on the study of mitochondrial morphology during senescence in grapevine (Vitis spp.) plants, one of the most important crops in Mediterranean area that has also the potentiality of becoming a model organism for fruit trees due to the knowledge of its genome sequence (Jaillon et al., 2007; Velasco et al., 2007). Suspension cell cultures of Vitis spp. were chosen as an initial model system to study mitochondrial morphology during senescence/PCD, because they are an accessible and not complex experimental system, sharing some of fundamental regulatory mechanisms with PCD processes in plant. Then, in order to study changes in mitochondrial morphology during different stages of leaf development and senescence, transgenic grapevine plants, expressing GFP (green fluorescent protein) fluorescent marker targeted to mitochondria, were generated. In the grapevine leaves the senescence/PCD process was characterized by analyzing the chlorophyll degradation and the expression of several VvSAGs genes (i.e. Vitis homologues of Arabidopsis senescence associated genes). Analyses performed in grapevine senescent leaves showed mitochondria prefunding altered in their morphology. Then, we moved to investigate to the molecular aspects of these morphological changes. Indeed, it is known that in eukaryotic cells, the number, size and distribution of mitochondria are regulated by continuous cycles of mitochondrial fusions and fission events (Logan, 2003) but so far in plants, no genes involved in mitochondrial fusion machinery have been identified, while several genes involved in mitochondrial fission have been recently described in Arabidopsis thaliana, a plant model organism. Hence, I moved to Arabidopsis plant and I focused on the study of the subcellular localization, expression and interaction of two proteins (ELM1 and BIGYIN), known to be involved in mitochondrial fission. ELM1 and BIGYIN subcellular localization was carefully analyzed, by using several organelle fluorescent markers and adopting different methods of transient expression in leaf cells, combined with confocal microscope analyses. Yet, I investigated whether in vivo interactions between ELM1 and BIGYIN occurred in plant cells. Then, I analyzed ELM1 and BIGYIN expression pattern in plant to verify if their presence in the same tissues was indeed detected. The analysis was carried out through histochemical GUS assay in transgenic plants, stable expressing the ELM1 and BIGYIN promoter region fused to the -glucuronidase (GUS) reporter. In order to define the physiological role of BIGYIN, a detailed subcellular localization of BIGYIN was performed in the whole plant, stably expressing BIGYIN under control of its own promoter, in order to mimic physiological conditions. These analyses allowed us to establish an association between BIGYIN and chloroplasts/plastids and, in particular, to identify protrusions, marked by YFP::BIGYIN, extruding from chloroplast, mitochondria and peroxisomes. Such kind of tubular protrusions has already been observed and termed matrixules, peroxules stromules respectively, and it has been proposed to play a role in physical inter-organellar interactions, by increasing the transfer efficiency of metabolites/molecules among organelles (Scott et al., 2007). The presence of BIGYIN on these protrusions is indeed an unexpected finding that led us to investigate more in detail this peculiar BIGYIN localization. I focused in detail on the dynamics and behaviour of these YFP::BIGYIN marked tubules, and investigated, in different tissues and different developmental stages, whether a protein trafficking through these organelles indeed occurred. The understanding of physical inter-organellar interactions is connected with the importance of elucidating relationships and cross-talks among organelles, key events to understand the mechanisms of interactions between plant and its environment that can be of particular importance for the coordination of the senescent program.
In questa tesi di dottorato, è stata condotta un’analisi degli organelli in cellule vegetali, studiando, in particolare, i mitocondri e la loro morfologia in diverse condizioni fisiologiche. La regolazione della morfologia mitocondriale è importante sotto molteplici aspetti, infatti controlla il numero dei mitocondri durante la divisione cellulare e la loro distribuzione all’interno di ogni cellula (Wastermann, 2010; Logan, 2003), controlla infine alcune funzioni biologiche. Nelle cellule animali, per esempio, la frammentazione dei mitocondri è un evento precoce che caratterizza il processo di apoptosi (Suen et al., 2008). Un’associazione tra morfologia mitocondriale e senescenza/morte cellulare programmata (PCD) è stata riportata anche in colture cellulare vegetali (Zottini et al., 2006). La comprensione dei meccanismi molecolari, utilizzati dalla pianta per regolare il processo di senescenza/PCD, può essere importante da un punto di vista biotecnologico, in quanto potrebbe permettere una modulazione dei parametri di crescita e di sviluppo della pianta attraverso miglioramenti genetici e manipolazioni di fattori ambientali che regolano il processo di senescenza. I risvolti applicativi di tale approcio sono molto interessanti, soprattutto per le specie di rilevanza agronomica. Alla luce di tali premesse, la morfologia mitocondriale è stata dettagliatamente analizzata durante il processo di senescenza/PCD in vite (Vitis spp). La vite rapprensenta una delle più importanti piante coltivate della zona del Mediterraneo, ed a seguito del sequenziamento del suo genoma (Jaillon et al., 2007; Velasco et al., 2007), è ritenuta un organismo modello negli studi sugli alberi da frutto. Inizialmente l’analisi della morfologia mitocondriale durante il processo di senescenza/morte cellulare programmata è stata condotta in colture cellulari di Vitis spp.. In seguito, lo studio è stato continuato in pianta, in particolare è stata portata avanti un’analisi dettagliata della morfologia mitocondriale durante i differenti stadi di sviluppo e di senescenza della foglia. A tal fine, sono state prodotte ed analizzate piante transgeniche, esprimenti un marcatore fluorescente mitocondriale. Le analisi condotte hanno dimostrato che in foglie senescenti i mitocondri presentano delle morfologie caratteristiche. Questi risultati ci hanno indirizzato ad uno studio degli aspetti molecolari coinvolti nella regolazione della morfologia mitocondriale. In numerose cellule eucariotiche, la morfologia mitocondriale è regolata dal continuo alternarsi di eventi di fusione e fissione mitocondriale (Logan, 2003). Nelle piante, i componenti molecolari coinvolti nel meccanismo di fusione mitocondriale non sono stati ancora individuati, mentre i componenti proteici implicati nella fissione sono stati recentemente descritti in Arabidopsis. A tuttoggi non è stato ancora compreso nè il preciso ruolo svolto da tali proteine, nè le loro precise interazioni fisiche, responsabili del processo di fissione mitocondriale. Durante il mio dottorato, ho quindi analizzato, nel sistema modello Arabidopsis thaliana, la localizzazione subcellulare, l’espressione e l’interazione di due proteine (ELM1 ed BIGYIN), coinvolte nel processo di divisione mitocondriale. La localizzazione subcellulare di ELM1 e BIGYIN è stata determinata in cellule vegetali, utilizzando differenti metodi di espressione transiente combinati con analisi di microscopia confocale. Sono state poi eseguite analisi per verificare l’interazione di queste due proteine in vivo in cellule vegetali. Successivamente è stata condotta in pianta un’analisi del pattern di espressione di BIGYIN ed ELM1, in modo tale da verificarne l’espressione di queste due proteine negli stessi tessuti, prerequisito fondamentale per una loro eventuale interazione. A tal fine, sono state prodotte ed analizzate mediante saggio istochimico-colorimetrico piante transgeniche di Arabidopsis thaliana stabilmente esprimenti il promotore di tali geni fuso al gene che codifica per l’enzima -glucuronidasi (GUS). Per definire il ruolo fisiologico di BIGYIN, una dettagliata localizzazione subcellulare di questa proteina è stata eseguita in piante di Arabidopsis, stabilmente esprimenti il costrutto YFP::BIGYIN sotto il controllo del proprio promotore. Queste analisi hanno messa in evidenza un’associazione tra BIGYIN e cloroplasti/plastidi ed ha portato ad individuare particolari protrusioni, marcate con la proteina di fusione YFP::BIGYIN, che si estondono dai cloroplasti, mitocondri e perossisomi. Protrusioni simili erano già state riportate in letteratura e prendono il nome di ‘stromuli’, ‘matrixuli’, ‘peroxuli’ a seconda che si estendano rispettivamente da cloroplasti, mitocondri o perossisomi. Recentemente, è stato ipotizzato che tali le protrusioni abbiano un ruolo nelle interazioni fisiche tra i differenti organelli, aumentando i contatti fisici tra gli organuli e migliorando l’efficienza di scambio di metaboliti/molecule (Scott et al., 2007). Tuttavia nessun dato è stato riportato a conferma di tale ipotesi. L’inattesa presenza di BIGYIN su tali protrusioni, ci ha permesso di studiare in dettaglio la loro dinamicità e di analizzare la presenza di interazioni fisiche tra gli organelli. La comprensione delle interazioni fisiche tra gli organelli si colloca nel campo di indagine delle relazioni tra tali compartimenti subcellulari che viene ora considerato un campo fondamentale per la conoscenza dei meccanismi di base della biologia cellulare vegetale.

The purpose of this review is to summarize the experiments that have been presented regarding guaranteed Basic Income, performed around the world; to see if a BI might fit into ”the Swedish model”; and what further research would be needed to understand the mechanics of BI. A theory has been used, designed to compare the effects of cash transfer programs irrespective of economic level of the countries. The experiments show many similarities, such as improved school attendance and achievements, improved health, enhanced economic equality, a sense of inclusion and security, and also, where this has been studied, less crime. Where BI has been beneath the level of economic sustenance a growth of working hours including self employment has been noted, whereas in countries with an income high enough to sustain oneself a slight diminished work supply has been reported. The effects shown in the review seem to be well aligned with the visions of the Swedish welfare state, although might not be in alignment with the growing use of “work fare” in Sweden. Further research should focus on more long-lasting experiments and also to find a common ground, on which to perform them to be able to compare results.

Os mecanismos celulares envolvidos na hiperplasia prostática benigna (HPB) não são bem compreendidos e pouco se sabe sobre como os proteoglicanos estão relacionados com esta condição. Neste estudo foi avaliada a expressão de proteoglicanos de superfície celular e do estroma na HPB. As amostras de próstata com HPB foram colhidas de pacientes submetidos à prostatectomia aberta e ressecção transuretral da próstata (RTUP), enquanto as amostras do grupo controle consistiram na zona de transição de próstatas normais de adultos jovens. Foram usados anticorpos primários anti-sindecan-1, anti-biglican e anti-decorin. A imunomarcação foi avaliada determinando-se a área relativa marcada pelo anticorpo, ou usando-se um escore atribuído à intensidade da coloração. Os resultados mostraram que, no grupo controle, a expressão do sindecan-1 foi mínima ou nula. Na HPB, no entanto, a imunomarcação deste proteoglicano foi intensa e localizada principalmente nas células basais do ácino prostático e com menor intensidade na superfície basolateral das células secretoras. Como não houve diferença entre as amostras obtidas através da prostatectomia aberta e da RTUP, esses grupos foram combinados em um único grupo HPB. A área marcada pelo anticorpo anti-sindecan-1 no epitélio das amostras de HPB foi nove vezes maior do que na próstata normal (p<0,001), e não houve correlações significativas entre a marcação do sindecan-1naHPB e o volume da próstata, o PSA ou a idade do paciente. Quanto ao biglican e ao decorin, a marcação foi exclusivamente no estroma, tanto no grupo controle quanto no grupo HPB, e não houve diferença significativa na extensão e intensidade da coloração entre estes dois grupos. Em conclusão, a imunomarcação do sindecan-1 na HPB é intensa e está localizada no epitélio glandular exclusivamente, mas a intensidade não se correlaciona com o tamanho da próstata ou PSA. A expressão do decorin e do biglican, no entanto, não foi alterada na HPB.
The cellular mechanisms involved in benign prostatic hyperplasia (BPH) are not well understood, and little is known about how proteoglycans are affected. Here we investigated the protein expression of stromal and cell surface proteoglycans in BPH. BPH samples were colected from open prostatectomy and transurethral resection of the prostate (TURP), while controls consisted of the transitional zone of normal prostates from young adults. We used primary antibodies against the proteoglycans syndecan-1, biglycan, and decorin. Immunolabeling was evaluated by determining the relative area of staining, or by using a score for staining intensity. The results showed that, in controls, syndecan-1 was mostly negative. In BPH, however, the labeling of this proteoglycan was intense and located mainly in acinar basal cells, but could extended into the secretory cells. As there were no differences in samples from open prostatectomy and TURP, these groups were combined into a BPH group. Syndecan-1 labeling area in the epithelium of BPH samples was nine times greater than that of controls (p<0,001), and there were no significant correlations between syndecan-1 labeling in BPH and prostate volume, PSA, or patients age. Biglycan and decorin labeling were in the stroma exclusively, in control and BPH samples, and there were no significant differences in extent and intensity of the staining between these two groups. In conclusion, syndecan-1 immunolabeling in BPH is prominent and is located in the glandular epithelium exclusively, but intensity does not correlate with prostate size or PSA. Decorin and biglycan labeling, however, were unchanged in BPH.

Arabic text recognition was not researched as thoroughly as other natural languages. The need for automatic Arabic text recognition is clear. In addition to the traditional applications like postal address reading, check verification in banks, and office automation, there is a large interest in searching scanned documents that are available on the internet and for searching handwritten manuscripts. Other possible applications are building digital libraries, recognizing text on digitized maps, recognizing vehicle license plates, using it as first phase in text readers for visually impaired people and understanding filled forms. This research work aims to contribute to the current research in the field of optical character recognition (OCR) of printed Arabic text by developing novel techniques and schemes to advance the performance of the state of the art Arabic OCR systems. Statistical and analytical analysis for Arabic Text was carried out to estimate the probabilities of occurrences of Arabic character for use with Hidden Markov models (HMM) and other techniques. Since there is no publicly available dataset for printed Arabic text for recognition purposes it was decided to create one. In addition, a minimal Arabic script is proposed. The proposed script contains all basic shapes of Arabic letters. The script provides efficient representation for Arabic text in terms of effort and time. Based on the success of using HMM for speech and text recognition, the use of HMM for the automatic recognition of Arabic text was investigated. The HMM technique adapts to noise and font variations and does not require word or character segmentation of Arabic line images. In the feature extraction phase, experiments were conducted with a number of different features to investigate their suitability for HMM. Finally, a novel set of features, which resulted in high recognition rates for different fonts, was selected. The developed techniques do not need word or character segmentation before the classification phase as segmentation is a byproduct of recognition. This seems to be the most advantageous feature of using HMM for Arabic text as segmentation tends to produce errors which are usually propagated to the classification phase. Eight different Arabic fonts were used in the classification phase. The recognition rates were in the range from 98% to 99.9% depending on the used fonts. As far as we know, these are new results in their context. Moreover, the proposed technique could be used for other languages. A proof-of-concept experiment was conducted on English characters with a recognition rate of 98.9% using the same HMM setup. The same techniques where conducted on Bangla characters with a recognition rate above 95%. Moreover, the recognition of printed Arabic text with multi-fonts was also conducted using the same technique. Fonts were categorized into different groups. New high recognition results were achieved. To enhance the recognition rate further, a post-processing module was developed to correct the OCR output through character level post-processing and word level post-processing. The use of this module increased the accuracy of the recognition rate by more than 1%.

This thesis presents the results of a multidisciplinary investigation into the Cenozoic tectono-stratigraphic evolution and mineralization of the Biga Peninsula, northwestern Turkey. This has involved the mapping and analysis of extensional and strike-slip fault systems and of the associated magmatic and volcanic events with a particular focus on the structural controls on the porphyry Cu-Au deposits and epithermal gold mineralization of the region. Detailed field mapping, structural analyses, core-logging of exploration drill holes together with Ar/Ar geochronology, remote sensing analyses and aeromagnetic interpretations were undertaken and integrated to develop new tectono- stratigraphic and magmatic models for the Cenozoic structural evolution and mineralization of the Biga Peninsula. The Biga Peninsula is characterised by the NE-trending Kazdag Massif formed by late Hercinian high grade gneiss and amphibolite metamorphic basement. During the latest Oligocene, re-crystallization and granitoids plutonism (26 - 25 Ma) was followed by metamorphic core complex detachment fault systems. The pre-Cenozoic hanging wall of the Kazdag Massif is composed by Permo- Trassic metasediments (Karakaya Complex) and by Late Cretaceous ophiolites (Cetrni Melange). These are flanked by Oligo-Miocene extensional volcano- sedimentary basins internally deformed by domino-style -NE trending hanging wall extensional faults. Porphyry style Cu-Au mineralization is associated with the granitoid intrusions in the footwall of the detachment fault system. High sulfidation style epithermal Au deposits are found in the hanging wall basins above the core complex detachment fault system. Gold in these major strata- bound silicified lithocaps is controlled by the steep hanging wall extensional faults and associated fracture architectures. The Biga peninsula sits within a system of back-arc extensional basins caused by slab roll-back during the Eocene, Oligocene and Miocene. Regional extension followed the closure of the Vardar-Izmir-Ankara Ocean as the Pontides and Anatolides terranes collided in the Late Cretaceous. This research has identified five stages of back-arc extension with magmatism and volcanic activity associated with magmatic-hydrothermal ore systems. The Middle Eocene and Latest Oligocene magmatic arcs of the Biga Peninsula produced porphyry type and epithermal style mineralization as part of a major NW-SE to E-W arc-shaped magmatic-metallogenic zone running from the Dinarides and the Rhodopes to the Pontides of northwestern Turkey. Since the Early Pliocene, the westward propagation of the North Anatolian Fault generated dextral transtension and new strike-slip fault systems together with increased extension on pre-existing extensional fault systems.

Resumen
This essay traces two trends in current philosophical and theological debates concerning forgiveness. One, advocated by Vladimir Jankélévitch and Jacques Derrida, I label “impossible” forgiveness. The second, advanced by John Milbank and L. Gregory Jones, I label “communal” forgiveness. I explore and critically examine each of these positions in the first two sections of the thesis. In the last section of the thesis I examine a recent conversation amongst religious ethicists against the background of the theoretical conversations described in the first half of the essay. Bringing the theoretical conversation together with the religious ethicists’ conversation, I argue that whether or not we embrace forgiveness depends in large part in what tradition, religious or secular, we place ourselves.

The Biga Peninsula in northwestern Turkey is part of the Western Tethyan Metallogenic Belt, known to be one of the most prospective belts for porphyry and epithermal style copper and gold deposits in the world. However, the limited spatial extent of mineral tenure boundaries around many deposits encumbers the correlation of geological and structural features beyond tenure boundaries and to a district and regional scale. High sulphidation epithermal Ag-Au-Cu and Au-Cu porphyry deposits in the Biga Peninsula are hosted by two temporally discrete magmatic rock formations and are structurally reconfigured by neotectonic faulting. New geological mapping between epithermal and porphyry deposits in the central Biga Peninsula, coupled with petrographic characterization and geochronological constraints on magmatism and mineralization, is used to correlate the Küçükdağ and Kirazlı high sulphidation epithermal Au-Ag-Cu deposits across the TV Tower District and regionally. Identifying prospective magmatic formations and relating structural controls on ore mineralization to regional structures will increase the probability of new discoveries.

Resumen
Most current automatic speaker recognition system extract speaker-depend features by looking at short-term spectral information. This approach ignores long-term information. I explored approach that use the fundamental frequency and energy trajectories for each speaker. This approach models prosody dynamics on single fonemes or syllables. It is known from literature that prosodic systems do not work as well the acoustic one but it improve the system when fusing. I verified this assumption by fusing my results with state of the art acoustic system from BUT. Data from standard evaluation campaigns organized by National Institute of Standarts and Technology are used for all experiments.

En Turquie-nw, trois localités types ont été sélectionnées afin de montrer la diversité des skarns bi-metasomatiques et les minéralisations qui leur sont respectivement associées: à Karakoy, entre la granodiorite du pluton d'Euciler et les horizons calcareux du groupe de Kazdag, nous observons les paragenèses du skarn calcique et des cornéennes qui mettent en évidence les conditions de formation du facies des cornéennes à pyroxène (600-600c, (1,5-2,0 kb). L'important dépôt de wollastonite s'est forme au-dessus de 500c en raison de la forte activité de si et de h#2o ainsi que d'un dégagement rapide de co#2. A Bakirlik Tepe, entre la monzonite du pluton de gurece et les olistostromes calcareux de la formation de Karakaya, nous observons un skarn à pyroxène à minéraux manganésifères et une minéralisation zincifère dans les conditions suivantes: tc<400c, fugacité élevée de l'oxygène, environnement enrichi en h#2o. A Derekoy, entre la monzonite et la dolomie de Kapakli (jurassique supérieur), nous observons un endoskarn à composition de monzonite à olivine et un skarn magnésien à forsterite fassaite, ainsi qu'une importante minéralisation de magnétite

The present work introduces a bibliographical research with critical reflection on the impact of the collaborative processes in the network, specifically in mobile applications, in the smart cities. The study was based on the contextualization of the theme, with a brief description of the concept of smart cities and origin of the term, as well as the survey and definition of concepts related to the processes of collective intelligence and its derivations, and concepts related to big data and networked organizations, based on the work of Clay Shirky, Manuel Castells, Lucia Santaella, James Surowiecki and Henry Jenkins. After the support of the historical survey, this work approaches the case studies of two companies engaged as mobilizing agents, focused on one of the basic principles of improvement of the city, main focus of this work: urban mobility, with the common ground on data update from a collaborative system, collective intelligence and big data. In order to investigate how and to what extent the collective power is applied in smart cities, acting both a collector of urban data and a agent of change, it was necessary, in short, to understand the functioning of the smart city; to detect how the constitution of communities takes place; the influence of ICT on the development of the city; to analyze how people engage, from the participatory culture and the formation of communities in the applications; in conclusion, to understand the convergence between big data and crowdsourcing in the smart city
O presente trabalho apresenta uma pesquisa de caráter bibliográfico, com reflexão crítica referente ao impacto dos processos colaborativos em rede, especificamente em aplicativos móveis, nas cidades inteligentes. O estudo se deu a partir da contextualização do tema, com uma breve descrição do conceito de cidades inteligentes e origem do termo, para assim entendêla na atualidade, bem como o levantamento e definição de conceitos relativos aos processos de inteligência coletiva e suas derivações, assim como os conceitos relativos ao big data e às organizações em rede, baseando-se nos trabalhos de, principalmente, Clay Shirky, Manuel Castells, Lucia Santaella, James Surowiecki e Henry Jenkins. Após o respaldo do levantamento histórico, chega-se nos estudos de casos de duas empresas engajadas como agentes mobilizadores, voltadas para um dos princípios básicos de aprimoramento da cidade, ponto focal deste trabalho: mobilidade urbana, tendo como ponto em comum a atualização de dados a partir de um sistema colaborativo, partindo da inteligência coletiva e big data. Afim de investigar como e em até que ponto o poder do coletivo é aplicado em cidades inteligentes, atuando tanto como coletor de dados urbanos, quanto agente de mudanças, foi necessário, em suma, compreender o funcionamento da cidade inteligente; detectar como se dá a constituição de comunidades; a influência das TIC no desenvolvimento da cidade; analisar como se dá o engajamento das pessoas, a partir da cultura participativa e da formação de comunidades nos aplicativos; em suma, entender a convergência entre big data e crowdsourcing na cidade inteligente

Freqüentment els escultors parlen tot emprant termes com "el llenguatge de la matèria” o "el respecte per la naturalesa dels materials" . Això passa especialment a una part dels escultors nascuts en la primera meitat del segle, encara que també fan servir aquestes paraules altres escultors més joves. Generalment, es parla de matèries i no de materials, en considerar que les matèries tenen un pes específic originat pel que hem anomenat comportaments propis i la seva utilització històrica, mentre que als materials els manca aquesta peculiaritat històrica: són generalment novedosos, molts d'ells d'origen industrial, i s'han incorporat a la pràctica escultòrica recentment. Diversos autors atribueixen a Brancusi l'origen de la proposta que estableix el respecte per les propietats de la matèria com a una característica de l'escultura moderna. Són molts els escultors que han partit d'aquest principi i han continuat elaborant el llenguatge de la matèria com un recurs escultòric, fins a l'actualitat. La proposta d'investigació parteix d'aquesta filosofia en que es considera la matèria com un agent, més que com un pacient; és a dir, com una entitat capaç de manifestar les seves peculiaritats. Pressuposem que, quan l'escultor escull la matèria amb un plantejament similar a l'exposat en el paràgraf anterior, és perquè ja té una actitud d'estima cap a ella, en la mesura que intenta valorar la peculiar idiosincràsia del suport escollit. Per tant, es considera que l'elecció de la matèria ja condiciona, o ve condicionada (indistintament) pel propòsit inicial, per la disposició vers l'escultura, pel món referencial, pel mètode de treball, pels procediments, i pels resultats. En la nostra recerca, i amb el fi de concretar més, hem delimitat la hipòtesi de treball de la manera següent: La utilització de la fusta en la pràctica escultòrica, des de la perspectiva del comportament, pressuposa l'assumpció del pes cultural i tradicional que ha acumulat aquesta matèria al llarg de la seva història; hi destaquen tres aspectes fonamentals: la percepció plàstica de la natura i de l'arbre; la participació dels components simbòlics, tradicionals o primitius implícits en les societats primàries i arcaiques; i el contacte directe amb la matèria, amb la conseqüent aplicació dels procediments artesanals. En la proposta inicial d'aquesta tesi tenim en compte el matis temporal regressiu que comporta aquesta pràctica, en la mesura que s'hi recullen aspectes derivats d'una dinàmica obsoleta, generalment relacionada amb cultures de tecnologia rudimentària i que tenen un contacte directe amb la natura.

O decorim e o biglicam são membros da família dos pequenos proteoglicanos ricos em leucina e possuem importantes funções no controle da proliferação, migração e invasão do citotrofoblasto extraviloso (TEV). O objetivo deste trabalho foi de caracterizar a expressão diferencial e a imunolocalização de decorim e biglicam em placentas humanas normais a termo (PNT), na placenta acreta (PA), na mola invasora (MI) e no coriocarcinoma (CO). Na PNT, as células deciduais apresentaram positividade para o decorim, enquanto o TEV foi negativo. O decorim foi fracamente expresso na matriz endometrial, mas negativo no fibrinoide do tipo matriz, enquanto foi positivo para biglicam. Na PA e na MI, o TEV mostrou positividade para decorim e biglicam. No CO, somente o citotrofoblasto foi positivo para ambos proteoglicanos. Portanto, o decorim e o biglicam são expressos diferencialmente em placentas normais e patológicas, sugerindo que os padrões de expressão desses proteoglicanos nas patologias estudadas indicam um papel na modulação da migração e da invasão do trofoblasto.
Decorin and biglycan are family members of the small leucine-rich proteoglycans family, and they have many functions as controlling proliferation, migration and invasion of extravillous trophoblast cells (EVT). The aim of this study was to characterize decorin and biglycan differential expression and immunolocalization in human normal term placenta (NTP), in placenta accreta (PA), in invasive mole (IM), and in choriocarcinoma (CH) samples. In PNT, deciduas cells were positive to decorin whereas EVT was negative. Decorin was faintly stained at endometrial matrix, but negative at matrix-type fibrinoid, although it was positive for biglycan. In PA and IM, the EVT was positive for decorin and biglycan. In CH, only cytotrophoblast cells were positive for both proteoglycans. Therefore, decorin and biglycan are differentially expressed in normal placenta and in placenta pathologies, suggesting that the expression patterns of the proteoglycans in studied pathologies indicate a role in modulating trophoblast migration and invasion.

Le handicap ou les décès en rapport avec l'asphyxie périnatale dans les pays développés correspondent à 0,2 à 1,3/1000 naissances vivantes. L’évaluation clinique et l’IRM conventionnelle permettent de distinguer les formes modérées des formes graves d’Encéphalopathies Hypoxiques Ischémiques (EHI). Le pronostic des EHI modérées reste difficile à préciser. L’imagerie des tenseurs de diffusion (DTI) mesure la diffusion de l'eau dans les tissus cérébraux et les paramètres quantitatifs tels que la Fraction d’Anisotropie et le Coefficient Apparent de Diffusion de l’eau (ADC) reflètent les propriétés microstructurales tissulaires. Nous avons mené une étude clinique et une étude expérimentale. L'objectif de l'étude clinique était d'évaluer les paramètres d'IRM, de SRM et de DTI chez les nouveau-nés asphyxiques à terme. Nous avons montré que l'IRM (y compris la diffusion conventionnelle) et les ratios du Lactate en SRM sont des outils intéressants pour l’évaluation neurologique précoce de ces nouveau-nés. Les paramètres de DTI (FA et ADC) dans des zones spécifiques cérébrales (capsules internes, pédoncules cérébraux, putamen/pallidum) sont fortement corrélés avec l'évolution neurologique précoce (J8) et à 6 mois, reflétant de possibles différences dans les mécanismes physiopathologiques impliqués. Des études sont nécessaires pour évaluer à long terme la valeur pronostique de ces paramètres. L'objectif de l'étude expérimentale était d'évaluer les effets à court terme de l'érythropoïétine (Epo) sur les lésions cérébrales en imagerie de diffusion et l’expression de l’aquaporine-4 en immunohistochimie dans un modèle HI de rat nouveau-né. L'effet protecteur potentiel de l'Epo humaine recombinante sur les ratons a été étudié. L'Epo a un effet neuroprotecteur précocement dans notre modèle (réduction de la surface de l’accident vasculaire cérébral AVC, augmentation de la valeur d’ADC dans l’AVC chez les rats traités), éventuellement grâce à une « up-régulation » de l’expression de l'aquaporine-4. Un mécanisme possible de la diminution de l’œdème cérébral pourrait être une meilleure élimination de l’'excès d'eau des tissus cérébraux vers la circulation sanguine via l'aquaporine-4
Severe neurological impairment or death in relation to perinatal asphyxia in developed countries concern 0.2 to 1.3/1000 live births. Clinical evaluation and conventional Magnetic Resonance Imaging (MRI) parameters are efficient to distinguish severe to minor Hypoxic-Ischemic Encephalopathy (HIE), but it remains discussed regarding long term outcome for babies with moderate HIE. Diffusion Tensor Imaging (DTI) measures the diffusion of water in cerebral tissue and quantitative parameters such as Fractional Anisotropy (FA) and Apparent Diffusion Coefficient (ADC) reflect the microstructure properties in local areas. We conducted in the same time a clinical and an experimental study. The aim of the clinical study was to evaluate MRI, MR Spectroscopy (MRS) and DTI parameters in asphyxiated full-term neonates. We showed that MRI (including conventional diffusion) and MRS represent interesting tools to evaluate the early neurological outcome in HIE neonates. DTI parameters in specific brain areas had strong correlations with early and at 6 months neurological outcome in babies with HIE. Studies are needed to evaluate the long-term prognostic value of DTI parameters in neonates with moderate injury. The aim of the experimental study was to evaluate the short-term effects of erythropoietin (Epo) on brain lesions (MR-Diffusion Imaging) and aquaporin-4 (immunohistochemistry) expression in a HI neonatal rat model. The potential protective effect of human recombinant Epo on a HI pup rat model was studied. We conclude that Epo has a short-term protective effect in HI pup rat model (reduction of stroke area; increase of ADC value in stroke area in treated rats), possibly through an early up-regulation of aquaporin-4 expression. One possible mechanism could be a better clearance of water excess from brain tissues to blood by aquaporin-4

Resumen
The dissertation analyses the reliefs and paintings on thirty-one different tombs in Western Anatolia erected between 600 and 450 BC, in order to illuminate the ways in which non-Greek elites were identified on their memorials. The tombs from three areas are treated: Phrygia, Hellespontine Phrygia and Lydia, where the primary language groups were Phrygian, Mysian and Lydian. There is little literary evidence for these regions, and what there is tends to focus on political developments. Descriptions of people and society are few, and tend to represent them from an outside perspective, grouping them according to cultural characteristics which differentiate them from Greeks. It is clear, however, that the regions were important, prosperous places, controlled by illustrious grandees and land marked with a relatively high proportion of monumental tombs. Of these monumental tombs, there is a relatively high number decorated with striking and articulate images. There is much to be gained from examining the images on these tombs, as ‘indigenous’ sources for how elite Western Anatolians described themselves. Previous approaches to the tombs and their images have tended to look at them individually or in smaller groups, and to concentrate on the transmission and reception of Persian and Greek culture in the Achaemenid provinces. This dissertation contributes a broader comparative study of the decorated tombs, focussing on the kinds of statuses the images represent and the cultural forms these took. By comparing the various methods of self-representation, it clarifies patterns of identities in Western Anatolia and their relationship to historical circumstances. The dissertation is divided into five chapters. An introduction outlines the scope and sample, the historical background, previous studies of the monuments, the definition of ‘identity’ and the methods of analysis adopted here. Three case study chapters present the regions and the decorated monuments within them. A concluding chapter synthesises three aspects: social identities (roles and spheres of life represented); geographic and chronological patterns; and cultural affiliations and orientations. The dissertation concludes that a tension between Persian identities and local traditions is evident in some of the tomb images, which relates to the political upheavals in Western Anatolia and the Aegean at the time of the Persian Wars.
