Abstract Abstract 1468 Background: Increased YB-1 expression correlates with poor prognosis, drug resistance and metastasis in several different cancers including B cell lymphoma. Phosphorylation and nuclear localization of YB-1 in response to growth factors leads to increased survival through expression of proteins such as survivin and multidrug resistance protein 1. Until now, its role in leukemia has not been investigated. We hypothesized that YB-1 expression is aberrantly regulated in pediatric pre-B acute lymphoblastic leukemia (pre-B ALL), and that YB-1 may be activated downstream of IL-7. This cytokine facilitates the differentiation and survival of pre-B cells and has been implicated in the drug resistance of pre-B ALL. Methods: YB-1 and IL-7Ra protein expression was investigated by flow cytometry in normal pre-B cells (CD19+CD10+CD20−), and mature B cells (CD19+CD10−CD20+) as well as diagnostic and relapsed pre-B ALL (CD19+CD10+/−). Cell surface and cytoplasmic expression was quantified by mean fluorescent intensity (MFI). Bone marrow from healthy donors was used as a source of normal pre-B cells, while mature B cells were derived from PBMCs; leukemic cells at presentation and relapse were obtained following local IRB approval and informed consent. Activation of YB-1 downstream of IL-7 stimulation (25 ng/ml) was examined in pre-B ALL cell lines or NSG (NOD scid gamma) mice-expanded pre-B ALL by Western blotting using anti-phosphoYB-1(S012). Pre-B ALL cell lines used in these experiments were 697, 380, RCH and RS-4;11. Signaling pathways were investigated by pre-treatment of cells with pharmacological inhibitors followed by Western analyses. For the transient overexpression of YB-1, pEGFP or a pEGFP/YB-1 fusion protein was electroporated into freshly isolated mature B cells (which have a low basal expression of YB-1) and YB-1 and IL-7Ra expression was assessed by flow cytometry after 24 h. Results: While intracellular YB-1 expression was significantly higher in leukemia samples at presentation compared to normal pre-B cells, the highest YB-1 levels were found in relapsed pre-B ALL (see figure, part A). All examined pre-B ALL cell lines had levels comparable to those of the relapse samples. Similarly, surface IL-7Ra (CD127) levels (MFI medians; upper-lower range) were increased in onset (221; 150–286), and relapsed (1840; 651–2030) ALL compared to normal pre-B cells (528; 333–2673). (normal pre-B vs. leukemia at presentation, p<0.001, Mann-Whitney). Overexpression of YB-1-GFP in normal mature B cells resulted in increased expression of IL-7Ra (see figure, part B), suggesting an link between the YB-1 and IL-7 signaling pathways. Activated YB-1 is phosphorylated on S102 and relocated to the nucleus. Addition of IL-7 to pre-B ALL cell lines led to phosphorylation of YB-1 within 30 min. Similar results were shown for patient-derived, NSG mice-expanded pre-B ALL samples. Intracellular immunostaining using Imagestream technology (Amnis) showed that IL-7 treatment of pre-B ALL cell lines increased nuclear YB-1 levels 4-fold. As PI3K and MEK1 are involved in signaling downstream of IL-7, we investigated their role in YB-1 signaling in both pre-B ALL cell lines and NSG-mouse expanded pre-B ALL using pharmacological inhibitors. Western analyses showed that inhibition of PI3K using LY294002 did not prevent IL-7-mediated phosphorylation of YB-1 but the MEK1 inhibitor U0126 did, indicating the involvement of MAPK (see figure, part C). Conclusion: We show that YB-1, which is highly expressed in pediatric pre-B ALL compared to normal pre-B cells, is expressed at even higher levels after relapse. We demonstrate a link between the YB-1 and IL-7 signaling pathways which could offer a novel target for the treatment of refractory leukemia. AbstractMelanoma is the most lethal dermal tumor, and a high recurrence rate and skin defects are two main serious problems. An antimelanoma material,which effectively inhibits tumor recurrence and possesses excellent biocompatibility, is urgently needed to treat melanoma. In this study, we developed a novel antitumor Yb3+ [Yb(NO3)3]containing chitosan hydrogel (Yb-CS hydrogel) by dissolving Yb(NO3)3 and chitosan in acetic acid solution and forming composite hydrogels by a freeze-drying process after adding NaOH to the mixed solution. In vitro studies demonstrated that the Yb3+ produces effect of inducing cell death in Yb-CS hydrogel. Moreover, we found that the Yb-CS hydrogel inhibited a focal adhesion kinase (FAK)-dependent signaling pathway and induced B-16 cell anoikis. However, the Yb-CS hydrogel was less effective on L929 normal mouse dermal cells. In vivo studies showed that the Yb-CS hydrogel inhibited the recurrence of melanoma in a mouse bare xenograft tumor model. We concluded that the Yb-CS hydrogel could potentially be used in the antimelanoma field, especially in the inhibition of melanoma recurrence.

Bulky monomeric lanthanoid(III) aryl oxides Ln(Odpp)3(Odpp = 2,6- diphenylphenoxide; Ln = Nd, Sm, Er, Yb or Lu) have been prepared by reaction between the lanthanoid metal, Hg(C6F5)2, and HOdpp in tetrahydrofuran (thf), and Yb(Odpp)3 has also been obtained from an analogous reaction of (PhCC)2Hg. Cleavage of Ln(η5-C5H5)3 (Ln = Nd or Yb) with HOdpp also yields Ln(Opdd)3. The aryl oxides have been isolated as [Ln(Odpp)3(thf)2] (Ln = Nd, Sm or Yb) or as the homoleptic complexes Ln(Odpp)3 (Ln = Nd, Er, Yb or Lu). The X-ray crystal structure of [Yb(Odpp)3(thf)2].thf [crystals monoclinic, space group P21/n, a 11.009(4), b 32.134 (7), c 15.498(6)Ǻ, β 96.72(3)°, Z 4; 2785 data refined to R 0.039] shows the complex to have distorted square pyramidal stereochemistry with apical and transoid basal Odpp ligands, (Yb-Odpp) 2.078(9)Ǻ, and transoid basal thf ligands, {Yb-O(thf)} 2.305(8)Ǻ. Similarly, Yb(Odpp)3 [crystals triclinic, space group Pī, a 15.934(2), b 13.563(1), c 10.968(2)Ǻ, α 116.71(1), β 92.90(2), γ 103.38(1)°, Z 2; 6236 data refined to R 0.031] was shown to have a trigonal pyramidal arrangement of oxygen donor atoms [{Yb-O} 2.065(4) Ǻ;{O-Yb-O} 117.5(1)°], a novel intramolecular chelate Yb…π-arene interaction, {Yb...C} 2.978(6)Ǻ, and an agostic Yb…CH interaction.

Reaction of Yb ( Otbp )2( thf )3( tbp = 2,4,6 But3C6H2; thf = tetrahydrofuran ) with TlOtbp in tetrahydrofuran at room temperature yields thallium metal and Yb( Otbp )3( thf ) (1), which was also obtained by redox transmetallation between TlOtbp and ytterbium metal in refluxing tetrahydrofuran . Some [ Yb ( Otbp )2( �-OH) (thf)]2 (2) was also isolated from the first synthesis and is derived from hydrolysis of Yb ( Otbp )3( thf ) by traces of water. The X-ray crystal structure of (1) [triclinic, P1, a 21.78(1), b 13.89(2), c 10.461(4)� , α; 109.03(5), β 99.03(3), γ; 98.34(5)�, Z 2, No = 4354 'observed' data refined to R 0.0711 shows novel distorted tetrahedral four coordination for ytterbium [0-Yb-0 angles 90.6(5)-121.8(4)�, which is bonded more strongly to the aryl oxide [Yb-0 1.97(1), 2.02(1), 2.09(1) � ] than to thf [Yb-0 2.35(1) �. The X-ray structure of (2) [triclinic, P1, a 15.61(1), b 13.84(2), c 11.085(4) �, α 99.25(9),, β 94.46(4), γ 96.42(9), Z 1, No = 4718 'observed' data refined to R 0.050] indicates a centrosymmetric hydroxo -bridged dimer with a YbO2Yb core. Ytterbium has irregular five coordination with two bridging hydroxide oxygens [Yb-0 2.217(6), 2.199(6) A; 0-Yb-0' 70.2(2)�, Yb-O-Yb, 109.8(3)�, non-bonding Yb…Yb 3.614(2) A], two phenoxide oxygens [2.047(7), 2.073(6) �], and a more distant thf oxygen [2.363(8) � ].

Fusarium crown rot caused by Fusarium pseudograminearum is one of the most devastating diseases of wheat worldwide causing major yield and economic losses. In this study, strain YB-15 was isolated from soil of wheat rhizosphere and classified as Bacillus subtilis by average nucleotide identity analysis. It significantly reduced Fusarium crown rot with a control efficacy of 81.50% and significantly improved the growth of wheat seedlings by increasing root and shoot fresh weight by 11.4% and 4.2%, respectively. Reduced Fusarium crown rot may have been due to direct antagonism by the production of β-1, 3-glucanase, amylase, protease and cellulase, or by the ability of B. subtilis YB-15 to induce defense-related enzyme activities of wheat seedlings, both alone and in seedlings infected with F. pseudograminearum. Improved plant growth may be related to the ability of B. subtilis YB-15 to secrete indole acetic acid and siderophores, as well as to solubilize phosphorus. In addition, the genome of strain YB-15 was determined, resulting in a complete assembled circular genome of 4,233,040 bp with GC content of 43.52% consisting of 4207 protein-encoding genes. Sequencing the B. subtilis YB-15 genome further revealed genes for encoding carbohydrate-active enzymes, biosynthesis of various secondary metabolites, nutrient acquisition, phytohormone production, chemotaxis and motility, which could explain the potential of strain YB-15 to be plant growth-promoting bacteria and biological control agent. B. subtilis YB-15 appears to be a promising biocontrol agent against Fusarium crown rot as well as for wheat growth promotion.

The Y box is a conserved sequence in the promoter of major histocompatibility complex (MHC) class II genes, which contains a CCAAT sequence (CCAAT box). Previously, we partially purified the DNA-binding protein that recognizes the Y box of the I-Aβ gene and showed that it consisted of two components (factors A and B) both of which were necessary for optimal DNA binding. The genes for the heteromeric protein NF-Y (NF-YA and NF-YB), which binds to the I-Eα Y box have been cloned. We subsequently isolated the genes for NF-YA and NF-YB using oligonucleotides designed from the published sequences. NF-YA and NF-YB were tested for binding to the I-Aβ and I-Eα Y boxes. While neither NF-YA or NF-YB alone bound to the Y box, when the components were mixed the complex bound to the I-Aβ Y box with high affinity. Moreover, NF-YA and NF-YB could be complemented for binding to DNA by factor B or factor A, respectively. These results suggest that the active binding protein is NF-YA in factor A extracts and NF-YB in factor B extracts. Finally, antibodies against NF-YA and NF-YB were shown to induce a supershift when nuclear extracts were added to the double-stranded oligodeoxynucleotide covering the Y box of the I-Aβ gene. Antisense expression constructs of both NF-YA and NF-YB were made and their effect on expression from the I-Aβ promoter was tested. Either antisense construction, when transfected into cells, lowered the expression of a reporter gene linked to the I-Aβ promoter. This study provides direct evidence of the identification of NF-YA and NF-YB as the previously described factors A and B. Moreover, these results strongly implicate NF-Y in the expression of the MHC class II gene I-Aβ.

3D geological model for each reservoir unit comprising the Yamama Formation revealed to that the formation is composed of alternating reservoirs and barriers. In Subba and Luhais fields the formation began with barrier YB-1 and four more barriers (YB-2, YB-3, YB-4, YB-5), separated five reservoirs (YR-A, YR-B, YR-C, YR-D, YR-E) ranging in thickness from 70 to 80 m for each of them deposited by five sedimentary cycles. In the Ratawi field the formation was divided into three reservoir units (YR-A, YR-B, and YR-C) separated by two barrier units (YB-2 and YB-3), the first cycle is missing in Ratawi field. The study involves 1 well in Luhais field (Lu-12), 3 wells in Subba field (Su-7, Su-8, and Su-9), and 5 wells in Ratawi field (Rt-3, Rt-4, Rt-5, Rt-6 and Rt-7), the Luhais, Subba, and Ratawi fields located in the Mesopotamia zone (Zubair subzone). The reservoir units (YR-C and YR-D) in Subba oil field, and YR-B in Ratawi oil field represent the major reservoir units that characterized by the best Petrophysical properties (the highest porosity, the lowest water saturation, and the best Net Pay Thickness), Luhais oil field has poor to moderate Petrophysical properties and low oil bearing in YR-A, YR-B and YR-C units, and produce heavy oil and salt water from YR-D and YR-E as indicated by low resistivity log reading, and according to the Drill Steam Test (DST) with the description of cutting in final geological reports.

With the facile displacement being utilized of thf from Yb(pin)2(thf)4 (pin = 2-phenylindol-1-yl, thf = tetrahydrofuran) in toluene solution, the complexes Yb(pin)2(dme)2 (dme = 1,2- dimethoxyethane), Yb(pin)2 (tmen)(tmen = N,N,N′,N′-tetramethylethane-1,2-diamine) and Yb(pin)2(diglyme)(thf) (diglyme = bis(2-methoxyethyl) ether) have been prepared from the respective ligands and Yb(pin)2(thf)4. Yb(pin)2 (diglyme) (thf) [monoclinic, space group P 21 /c, a 15.35(1), b 16.179(5), c 14.45(2) Ǻ, β 107.51(8)°, Z 4, R 0.044 for 2956 (I > 3σ(I)) 'observed' reflections] has a monomeric six-coordinate structure with transoid nitrogen donor atoms, N-Yb-N 143.6(4)° and an irregular coordination polyhedron described as either a distorted trigonal prism or a monocapped square pyramid. Attempted crystallization of Yb(pin)2 (thf) by partial desolvation of Yb(pin)2(thf)4 in hot toluene, containing a trace of dme, gave a mixture of red Yb(pin)2(thf) and orange [Yb(pin)2(dme)]2. The latter was independently synthesized by partial desolvation of Yb(pin)2(dme)2 in toluene. An X-ray crystal structure showed [Yb(pin)2(dme)]2 [monoclinic, space group P 21/c, a 11 .614(2), b 15.945(7), c 15.327(4) Ǻ, β 110.19(2)°, Z 2 dimers, R 0.070 for 2314 (I ≥ 3σ(I)) 'observed' reflections] to be a dimer with two bridging pin ligands, coordinated through nitrogen only. There is an approximately square pyramidal five-coordinate ytterbium environment with an apical dme oxygen, and with two bridging nitrogens, a terminal nitrogen, and a dme oxygen in the basal plane.

This paper is focussed on the development of microstructure during crystallisation heat treatment of B-phase parent glasses with composition (e/o) 35R:45Si:20Al:83O:17N, where R = Er, Yb, Y or a mixture of Y and Yb. Extensive high resolution analytical transmission electron microscopy has shown that the lenticular B-phase crystals take up a substantial range of composition. The element R is always clearly anti-correlated with the Si, and a larger R3+ cation radius moves the composition range to lower R contents. It is suggested that a locally increased density in the bi-dimensional network of randomly oriented (Si,Al)(O,N)4 tetrahedra is associated with an increased density of vacancies in the R3+ cation lattice.

Lanthanoid (II) complexes of bulky aryl oxides, Ln (OC6H2But2-2,6-X-4)2 ( Ln = Yb or Eu ; X = H, Me or But) have been prepared by one or more of the following methods: ( i ) reaction of Ln (C6F5)2 with the appropriate phenol, (ii) reaction of the lanthanoid metal, Hg(C6F5)2 and the phenol, and (iii) reaction of the lanthanoid metal with the corresponding thallium(I) phenolate, in tetrahydrofuran ( thf ), and they have been isolated as Ln (OC6H2But2-2,6-X-4)2(thf)3 complexes. Crystallization of Yb (OC6H2But2-2,6-X-4)2( thf )3 (X = H or Me) from toluene yields Yb (OC6H2But2-2,6-X-4)2( thf )2. The X-ray crystal structure of [ Yb (OC6H2But3-2,4,6)2( thf )3]. thf [monoclinic, a 10.20(1), b 32.97(2), c 16.018(4) Ǻ, β 93.52(8)°, V 5375 Ǻ3, Z 4, 2734 'observed' data refined to R 0.070] shows distorted square pyramidal five coordination for ytterbium with an apical thf oxygen [ Yb -O 2.44(1)Ǻ], and transoid phenolate oxygens [ Yb -O 2.21(1), 2.22(1)Ǻ, O- Yb -O 149.0(6)°] and trans thf oxygens [ Yb -O 2.47(2), 2.48(1)Ǻ, O-Yb -O 177.3(5)°] in the square plane. Four methyl groups ( Yb…C 3.89-4.15 Ǻ) across the square plane away from the apical oxygen may block coordination of an additional ligand. X-Ray structures of Yb (OC6H2But2-2,6-Me-4)2(thf)n showing four coordination (n = 2) and five coordination (n = 3), given in a preliminary communication,1 are further discussed.

We evaluated the magnetic alignment and superconducting magnetic anisotropies of high-Tc superconductor Yb 1 Ba 2 Cu 3 O 7-δ by both X-ray diffraction and magnetization measurements. The sample was grain aligned under a high magnetic field of 6 T. X-ray diffraction pattern revealed normal alignment in Yb -123 by showing the c axis aligned perpendicular to the magnetic field. At low temperatures, magnetic hysteresis ΔM with magnetic field perpendicular to a–b plane is larger than magnetic hysteresis parallel to a–b plane. A crossover was observed at higher temperature in magnetic hystereses, i.e., ΔM⊥a - b < ΔM‖a - b at certain applied field. This crossover can be explained due to difference in vortex structure along both field directions.

YB-1 belongs to the evolutionarily conserved cold-shock domain protein family of RNA binding proteins. YB-1 is a well-known transcriptional and translational regulator, involved in cell cycle progression, DNA damage repair, RNA splicing, and stress responses. Cell stress occurs in many forms, e.g., radiation, hyperthermia, lipopolysaccharide (LPS) produced by bacteria, and interferons released in response to viral infection. Binding of the latter factors to their receptors induces kinase activation, which results in the phosphorylation of YB-1. These pathways also activate the nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB), a well-known transcription factor. NF-κB is upregulated following cellular stress and orchestrates inflammatory responses, cell proliferation, and differentiation. Inflammation and cancer are known to share common mechanisms, such as the recruitment of infiltrating macrophages and development of an inflammatory microenvironment. Several recent papers elaborate the role of YB-1 in activating NF-κB and signaling cell survival. Depleting YB-1 may tip the balance from survival to enhanced apoptosis. Therefore, strategies that target YB-1 might be a viable therapeutic option to treat inflammatory diseases and improve tumor therapy.

Hepatocellular carcinoma is one of the most common cancer types worldwide. In cases of advanced-stage disease, sorafenib is considered the treatment of choice. However, resistance to sorafenib remains a major obstacle for effective clinical application. Based on integrated phosphoproteomic and The Cancer Genome Atlas (TCGA) data, we identified a transcription factor, Y-box binding protein-1 (YB-1), with elevated phosphorylation of Ser102 in sorafenib-resistant HuH-7R cells. Phosphoinositide-3-kinase (PI3K) and protein kinase B (AKT) were activated by sorafenib, which, in turn, increased the phosphorylation level of YB-1. In functional analyses, knockdown of YB-1 led to decreased cell migration and invasion in vitro. At the molecular level, inhibition of YB-1 induced suppression of zinc-finger protein SNAI1 (Snail), twist-related protein 1 (Twist1), zinc-finger E-box-binding homeobox 1 (Zeb1), matrix metalloproteinase-2 (MMP-2) and vimentin levels, implying a role of YB-1 in the epithelial-mesenchymal transition (EMT) process in HuH-7R cells. Additionally, YB-1 contributes to morphological alterations resulting from F-actin rearrangement through Cdc42 activation. Mutation analyses revealed that phosphorylation at S102 affects the migratory and invasive potential of HuH-7R cells. Our collective findings suggest that sorafenib promotes YB-1 phosphorylation through effect from the EGFR/PI3K/AKT pathway, leading to significant enhancement of hepatocellular carcinoma (HCC) cell metastasis. Elucidation of the specific mechanisms of action of YB-1 may aid in the development of effective strategies to suppress metastasis and overcome resistance.

Redox trans- metallation between ytterbium metal and Tl (C5H4PPh2) in l,2-dimethoxyethane ( dme ) gives thallium metal and [ Yb (C5H4PPh2)2(dme )]. The ytterbium-transition metal hetero-bimetallics [ Yb ( thf ) (C2H4PPh2)2Z]. nPhMe [Z = Ni(CO)2, Mo(CO)4 or PtMe2, n = 2/3 or 1] have been prepared by reaction of [ Yb ( thf )(C5H4PPh2)2] with Ni(CO)2(PPh3)2, Mo(CO)4( nbd ) ( nbd = norbornadiene ) or PtMe2(cod) (cod = cycloocta-1,5-diene) in toluene. The latter are also bonded through phosphorus to the Ni(CO)2 group, resulting in tetrahedral stereochemistry for nickel. Infrared spectroscopy suggests the presence of ytterbium- isocarbonyl bonding in solid [ Yb ( thf )(C2H4PPh2)2Z] [Z = Ni(CO)2 or Mo(CO)4].

Abstract Medulloblastoma relapse occurs in 30–40% of patients and is typically fatal. The emergence of therapy resistant sub-clones likely plays a major role in a large proportion of recurrent medulloblastoma. Y-box binding protein 1 (YB-1) is a multifunctional transcription/translation factor and known onco-protein. Overexpression has been described in numerous cancers, where elevated expression and nuclear accumulation correlates with disease progression, metastasis and drug resistance. Genomic analysis of a large medulloblastoma cohort revealed YB-1 up-regulation across all subgroups of medulloblastoma, where elevated expression correlated with poor survival. Immunohistochemical staining of patient tissue microarrays displayed significant YB-1 expression, with a high proportion (83%) of patients exhibiting nuclear accumulation. High YB-1 expression was also observed at both protein and RNA level across medulloblastoma cell lines, with expression highest in Group 3 and 4. Hence, we hypothesised that YB-1 plays a role in medulloblastoma chemoresistance and progression. Treatment of Group 3 (HDMB-03 and D283MED) and SHH (DAOY) cell lines with vincristine and cisplatin and analysis of cellular localisation by nuclear/cytoplasmic fractionation and immunofluorescence demonstrated that YB-1 undergoes nuclear translocation in response to these standard medulloblastoma chemotherapy agents. Chromatin immunoprecipitation (ChIP) analysis of untreated Group 3 cell lines (D283MED and HDMB-03) demonstrated considerable YB-1 interaction with an inverted CCAAT box in the ATP-binding cassette subfamily B member 1 (ABCB1) promoter. RT-PCR analysis of ABCB1 following vincristine and cisplatin treatment revealed differences in transcript expression, indicative of different YB-1 promoter interactions dependent on chemotherapeutic treatment. Our results highlight YB-1 as a novel candidate chemoresistance driver in medulloblastoma.

Cell fate decisions regulating survival and death are essential for maintaining tissue homeostasis; dysregulation thereof can lead to tumor development. In some cases, survival and death are triggered by the same receptor, e.g., tumor necrosis factor (TNF)-receptor 1 (TNFR1). We identified a prominent role for the cold shock Y-box binding protein-1 (YB-1) in the TNF-induced activation and nuclear translocation of nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) p65. In the absence of YB-1, the expression of TNF receptor-associated factor 2 (TRAF2), a central component of the TNF receptor signaling complex required for NF-κB activation, is significantly reduced. Therefore, we hypothesized that the loss of YB-1 results in a destabilization of TRAF2. Consistent with this hypothesis, we observed that YB-1-deficient cells were more prone to TNF-induced apoptotic cell death. We observed enhanced effector caspase-3 activation and could successfully rescue the cells using the pan-caspase inhibitor zVAD-fmk, but not necrostatin-1. Taken together, our results indicate that YB-1 plays a central role in promoting cell survival through NF-κB activation and identifies a novel mechanism by which enhanced YB-1 expression may contribute to tumor development.

Colletotrichum capsici is one of the most important pathogens on chili peppers. Unreasonable application of chemical fungicides will lead to threats to human and animal health, environmental damage, and increased fungicide resistance to pathogens. As an alternative strategy, biological control has been paid more and more attention by academics. In this study, the Yb-1 strain was isolated from healthy cucumber leaves, which could significantly restrict the mycelium growth of C. capsici and Botrytis cinerea, with inhibition rates of 93.49% and 74.03%, respectively. Strain Yb-1 was identified as Bacillus velezensis by morphological features and 16S rRNA gene, gyrA gene, and gyrB gene sequence analysis. When pepper seeds were treated with different concentrations of bacterial suspension, we found that a medium-concentration treatment (104 CFU/mL and 105 CFU/mL) could promote seed germination and growth, while high-concentration treatments (108 CFU/mL) limited seed germination and growth. In addition, the root-irrigation method, acupuncture-leaf method, and leaf-cutting method were used to evaluate the safety of strain Yb-1 on peppers. The results indicated that Yb-1 did not affect the normal growth of peppers. The results of further field trials showed that the control effect of strain Yb-1 on pepper anthracnose was 59.45%. Thus, the strain B. velezensis Yb-1 has great biocontrol potential for pepper anthracnose and promotes plant growth.

Tax1, a potent activator of human T-cell lymphotropic virus type 1 (HTLV-1) transcription, has been shown to modulate expression of many cellular genes. Tax1 does not bind DNA directly but regulates transcription through protein-protein interactions with sequence-specific transcription factors. Using the yeast two-hybrid system to screen for proteins which interact with Tax1, we isolated the B subunit of the CCAAT binding protein NF-Y from a HeLa cDNA library. The interaction of Tax1 with NF-YB was specific in that NF-YB did not interact with a variety of other transcription factors, including human immunodeficiency virus Tat, human papillomavirus E6, and Bicoid, or with the M7 (amino acids 29CP-AS) Tax1 mutant. However, NF-YB did interact with the C-terminal Tax1 mutants M22 (130TL-AS) and M47 (319LL-RS). We also show that in vitro-translated NF-YB specifically bound to a glutathione S-transferase-Tax1 fusion protein. Further, Tax1 coimmunoprecipitated with NF-Y from nuclear extracts of HTLV-1-transformed cells, providing evidence for in vivo interaction of Tax1 and NF-YB. We further demonstrate that Tax1 specifically activated the NF-Y-responsive DQbeta promoter, as well as a minimal promoter which contains only the Y-box element. In addition, mutation of the Y-box element alone abrogated Tax1-mediated activation. Taken together, these data indicate that Tax1 interacts with NF-Y through the B subunit and that this interaction results in activation of the major histocompatibility complex class II promoter. Through activation of this and other NF-Y driven promoters, the Tax1-NF-Y interaction may play a critical role in causing cellular transformation and HTLV-1 pathogenesis.

Herein we highlight for the first time the ability to tune the stoichiometry of metal boride nanocrystals through nanoparticle synthesis in thermally stable inorganic molten salts. Two metal–boron systems are chosen as case studies: boron-poor nickel borides and boron-rich yttrium borides. We show that NiB, Ni4B3, Ni2B, Ni3B, and YB6 particles can be obtained as crystalline phases with good selectivity. Anisotropic crystallization is observed in two cases: the first boron-rich YB4 nanorods are reported, while boron-poor NiB nanoparticles show a peculiar crystal habit, as they are obtained as spheres with uniaxial defects related to the crystal structure. Crystallization mechanisms are proposed to account for the appearance of these two kinds of anisotropy at the nanoscale.

The incommensurate palmierite-like structure of β-K5Yb(MoO4)4, potassium ytterbium tetramolydate, has been refined in the (3 + 1)-dimensional monoclinic superspace group X2/m(0ρ0)00, with X = [0 0 0 0; ½ ½ 0 0; 0 0 ½ ½; ½ ½ ½ ½] and the unit-cell parameters a = 10.4054 (16), b = 6.1157 (12), c = 19.7751 (18) Å, β = 136.625 (10)°; q = 0.6354 (30)b*. The occupations of the K and Yb atomic positions are described by crenel functions. The structure model reveals a balanced interaction between the atoms of the first and second coordination spheres. It is shown that the third coordination sphere should not be neglected in studies of modulated structures. The ordering of the K and Yb atoms appears to be the driving force for the modulation of all the other atoms.

The yrast state of the Ytterbium, Yb isotopes for the neutron range of 82 ≤ N≤ 180 and lead, Pb isotopes for the neutron range of 98 ≤ N≤132 for the even-even nuclei have been studied using the energies of the first excited state in these nuclei. The nuclear deformation parameters, and the reduced quadruple transition probability B (E2)↑ with other intrinsic parameters associated with the nuclei shape were obtained using a MATLAB code. The results revealed that the Pb nucleus with Z = 82 - which is one of the magic numbers have a more ‘spherically’ nuclei shape at the ground state with small degree of deformation as compared to the nuclei shapes in the Yb isotopes. Our study supports the global predictions of the prolate deformation in Yb isotopes around the neutron range of 90 ≤ N ≤ 112.

Resumen
We report on the crystal growth of rare-earth pyrosilicates, R 2Si 2O 7 for R = Yb and Er using the optical floating zone method. The grown crystals comprise members from the family of pyrosilicates where the rare-earth atoms form a distorted honeycomb lattice. C-Yb 2Si 2O 7 is a quantum dimer magnet with field-induced long range magnetic order, while D-Er 2Si 2O 7 is an Ising-type antiferromagnet. Both growths resulted in multi-crystal boules, with cracks forming between the different crystal orientations. The Yb 2Si 2O 7 crystals form the C-type rare-earth pyrosilicate structure with space group C 2 / m and are colorless and transparent or milky white, whereas the Er-variant is D-type, P 2 1 / b , and has a pink hue originating from Er 3 +. The crystal structures of the grown single crystals were confirmed through a Rietveld analysis of the powder X-ray diffraction patterns from pulverized crystals. The specific heat of both C-Yb 2Si 2O 7 and D-Er 2Si 2O 7 measured down to 50 mK indicated a phase transition at T N ≈ 1.8 K for D-Er 2Si 2O 7 and a broad Schottky-type feature with a sharp anomaly at 250 mK in an applied magnetic field of 0.8T along the c-axis in the case of C-Yb 2Si 2O 7 .

Mitochondria play key roles in essential cellular functions, such as energy production, metabolic pathways and aging. Growth factor-mediated expression of the mitochondrial OXPHOS (oxidative phosphorylation) complex proteins has been proposed to play a fundamental role in metabolic homoeostasis. Although protein translation is affected by general RNA-binding proteins, very little is known about the mechanism involved in mitochondrial OXPHOS protein translation. In the present study, serum stimulation induced nuclear-encoded OXPHOS protein expression, such as NDUFA9 [NADH dehydrogenase (ubiquinone) 1α subcomplex, 9, 39 kDa], NDUFB8 [NADH dehydrogenase (ubiquinone) 1β subcomplex, 8, 19 kDa], SDHB [succinate dehydrogenase complex, subunit B, iron sulfur (Ip)] and UQCRFS1 (ubiquinol-cytochrome c reductase, Rieske iron–sulfur polypeptide 1), and mitochondrial ATP production, in a translation-dependent manner. We also observed that the major ribonucleoprotein YB-1 (Y-box-binding protein-1) preferentially bound to these OXPHOS mRNAs and regulated the recruitment of mRNAs from inactive mRNPs (messenger ribonucleoprotein particles) to active polysomes. YB-1 depletion led to up-regulation of mitochondrial function through induction of OXPHOS protein translation from inactive mRNP release. In contrast, YB-1 overexpression suppressed the translation of these OXPHOS mRNAs through reduced polysome formation, suggesting that YB-1 regulated the translation of mitochondrial OXPHOS mRNAs through mRNA binding. Taken together, our findings suggest that YB-1 is a critical factor for translation that may control OXPHOS activity.

In the title compound, [Yb(C24H40BN6)2]·C7H8, the Yb atom is coordinated by two tris(3-tert-butyl-5-methylpyrazol-1-yl)hydridoborate [TptBu,Me] ligands. One ligand binds in the κ3mode, throuh three N atoms of the pyrazolyl rings, the other ligand coordinates through two N atoms of the pyrazolyl rings and the H atom attached to the central Bviaan agostic-type interaction through the B—H group of the second TptBu,Meligand, giving an overall distorted octahedral geometry. One of thetert-butyl groups is disordered over two sites, with occupancies of 0.65 and 0.35.

Abstract The binary compound Yb5Bi3 was synthesized by reaction of the elements in a sealed Ta container. Its crystal structure was determined from single-crystal X-ray diffraction data: β -Yb5Sb3-type, space group Pnma, Pearson code oP32, a = 12.6375(6), b = 9.7243(4), c = 8.4117(5) Å, V = 1033.72(9) Å3, Z = 4, Rgt(F) = 0.028, wRref(F2) = 0.069, T = 290 K. Band structure calculations and analysis of the chemical bonding suggest mainly ionic interactions in the crystal structure and a possible presence of ytterbium in two valence states Yb2+ and Yb3+. The magnetization measurements showed that at low temperatures Yb5Bi3 contains ytterbium exclusively in the 4f14 configuration without fluctuations to the Yb 4f13 configuration up to 400 K. From the Yb-LIII X-ray absorption spectroscopy data the effective valence of ytterbium was found to be 2.11 (89% of Yb in 4f14 configuration).

Nuclear Factor-Y (NF-Y) transcription factors play vital roles in plant abiotic stress response. Here, the NF-Y family in Brassica napus, which is hyper-sensitive to nitrogen (N) deprivation, was comprehensively identified and systematically characterized. A total of 108 NF-Y family members were identified in B. napus and categorized into three subfamilies (38 NF-YA, 46 NF-YB and 24 NF-YC; part of the Arabidopsis NF-YC homologous genes had been lost during B. napus evolution). In addition, the expansion of the NF-Y family in B. napus was driven by whole-genome duplication and segmental duplication. Differed expression patterns of BnaNF-Ys were observed in response to multiple nutrient starvations. Thirty-four genes were regulated only in one nutrient deficient condition. Moreover, more BnaNF-YA genes were differentially expressed under nutrient limited environments compared to the BnaNF-YB and BnaNF-YC subfamilies. Sixteen hub genes responded diversely to N deprivation in five rapeseed tissues. In summary, our results laid a theoretical foundation for the follow-up functional study of the key NF-Y genes in B. napus in regulating nutrient homeostasis, especially N.

Single crystals of quaternary compounds in the solid solution (NaxRE1-x)AlB14 (RE=Li, Mg, rare earths (Tb, Dy, Ho, Er, Tm, Yb, Lu)) were grown by the Na-RE-Al-B system using a molten Al-self flux under an Ar gas. The single crystals of LiAlB14, NaAlB14, MgAlB14 and (NaxMg1-x)AlB14 (0.21≤x≤0) were grown; the quaternary boride (NaxLi1-x)AlB14 and (NaxRE1-x)AlB14 (Tb, Dy, Ho, Er, Tm, Yb, Lu) phases were not obtained at the single crystal growth experiment in the Na-Li-Al-B and Na-RE-Al-B systems. The as-growth LiAlB14, NaAlB14, MgAlB14, (Na0.08Mg0.92)AlB14 and (Na0.21Mg0.79)AlB14 single crystals were subjected to chemical analyses and measurements of unit-cell parameter. The homogeneity range for the solid solutions of (NaxMg1-x)AlB14 (0.21≤x≤0) crystals was studied by XRD and chemical analyses. Measurements of microhardness in air and magnetic susceptibility of crystalline samples of as-grown (NaxMg1-x)AlB14 (x=1, 0.21≤x≤0) have been measured in the temperature range of 300 K to 2 K.
