RésuméDe nombreux points de comparaison sont possibles entre la philosophie de Nishida Kitarō (1870-1945) et la philosophie de la religion de Karl Rahner (1904-1984), notamment concernant la temporalité, thème à propos duquel ces deux auteurs réfléchirent radicalement. En réexaminant la notion rahnérienne d’avenir absolu à la lumière de la notion nishidienne de présent absolu, il est possible d’apporter une solution au problème suivant : rejoint-on Dieu (Rahner) ou l’absolu (Nishida) dans un avenir situé au-delà de la temporalité ou bien dans l’instant présent, compris comme le lieu même du jaillissement de la temporalité ? Dans la mesure où elle s’appuie sur une philosophie du néant absolu, la pensée de Nishida permet de dégager la philosophie de la religion de Rahner des cloisons de l’onto-théo-téléologie qui l’enserrent et, la plaçant ainsi sur un horizon plus vaste, la rend apte à exprimer, d’une manière qu’on espère nouvelle, les nombreuses potentialités qu’elle recèle encore. En retour, une lecture de la temporalité nishidienne à travers le regard de Rahner permet de l’approfondir et de faire ressortir des éléments inédits.

Resumen
Current clinical studies confirm that the consumption of oats for people suffering from celiac disease is safe. Some studies have confirmed different levels of immunoreactive gluten epitopes of oats in different cultivars, while others explain these differences due to contamination with gluten-rich species or as random cross-reactivity ELISA of homologous oat epitopes with anti-wheat gliadin antibodies. The aim of our two-year study was therefore to map cross-reactive oat epitopes in a set of 132 oat cultivars using a G12-based ELISA kit. The results were focused on the varietal and annual level of cross-reactivity (interference) of avenin epitopes with the G12 antibody on the identification of potential cultivars with significantly different interferences and assessing the degree of risk of possible false-contamination with external gluten. Although repeated evaluations confirmed high year-to-year variability (RSD ≥ 30%) in approximately 2/3 of the cultivars, the content of interfering avenin epitopes with G12 did not exceed the considered safe limit (20 mg·kg−1) for celiacs. At the same time, not only annual but, above all, significant cultivar dependences in the interference of avenins to the G12 antibody were demonstrated. Genetic dependence was further confirmed in connection with the proven avenin polymorphism as well as immunoblotting with the identification of interfering peptides with the G12 antibody in the 25 and 30 kDa regions. It was the occurrence of two bands around 30 kDa that predominantly occurred in oat cultivars with a relatively higher content of cross-reactive avenins (12–16 mg·kg−1). Due to the fact that the contents of interfering avenins ranged in several cultivars even over 16 mg·kg−1, the choice of a suitable oat cultivar may be crucial for gluten-free food producers, as it reduces the risk of a possible false-response of the commercial ELISA kits when checking the real-gluten contamination.

Cette thèse analyse un mouvement d'opposition, qui, à travers une action antagoniste et conflictuelle, produit son agir et son « sens » de l'action. L'enjeu principal consiste à observer le phénomène de Bure à partir de l'agir et des visions du monde des acteurs, notamment à partir de membres de « la commune de Bure » qui s'opposent à la construction de Cigéo, le projet français d'enfouissement des déchets nucléaires. L'étude, qui s'efforce d'analyser les représentations et les pratiques qui alimentent tant objectivement que subjectivement l'action des individus, s'appuie sur une enquête qualitative fondée sur l'observation participante et sur des entretiens compréhensifs. A cet égard, il s'agit de comprendre l'agir des acteurs à travers leur activité réflexive et subjective, mais aussi leur activité « concrète » observée, en se focalisant sur le sens qu'ils donnent à leurs actions et les éléments qui les poussent à la mobilisation. A Bure, la peur et la menace de la construction d'un centre d'enfouissement ont poussé les individus à se rassembler autour d'une lutte commune, mais ce qui distingue la résistance à Bure d'autres mouvements similaires est la construction progressive d'un savoir partagé, qui s'est étendu peu à peu à des questions et des sujets qui ne concernent pas seulement les déchets nucléaires. A cet égard, tous les acteurs ont en commun un sentiment de malaise, de frustration et de manque de reconnaissance qui les trouble et qui les pousse à l'action. L'expérience et l'expérimentation, la lutte pour la reconnaissance, la volonté de puissance et le besoin de manifester les sentiments forgent le vécu des membres de la commune de Bure et leur permettent d'expérimenter de nouvelles formes des modes-de-vie alternatives. Des espaces d'expérience se façonnent, dans lesquels, ensuite, des alternatives « concrètes » et « imaginées » à la vie et à la société émergent. C'est dans ce cadre que, les représentations et les pratiques transforment les espaces d'expérience en contre-espaces publics. Les structures mentales persistantes, les sentiments profonds et les interactions significatives deviennent le moteur de la mobilisation et la pulsion vers un processus de subjectivation qui concerne les sujets agissants. De plus, il y a une volonté qui se déploie, une volonté de dépassement du malaise et la détermination à créer un futur alternatif. Cette thèse s'efforce d'analyser le potentiel d'agir du cas exemplaire du mouvement de la commune de Bure, sa capacité à travers l'action de créer et d'organiser un espace public délimité qui sera capable de regrouper des visions du monde exprimant une résistance à l'ordre établi et une nouvelle possibilité de changement. This dissertation analyzes an opposition movement that produces its action and its "sense" of action through antagonism and conflict. Particularly it aims to observe the "Bure phenomenon" from the actions and worldviews of the actors, starting from the members of "la commune de Bure" who oppose the construction of Cigéo, the Highly Radioactive Waste Underground Storing Project (Bure, France). The present study, which seeks to analyze representations and practices that feed both objectively and subjectively into the actions of individuals, conducted a qualitative survey based on participant observations and interviews. The challenge is to understand not only the actions of the actors through their reflexive and subjective activity, but also their observed "concrete" activity, focusing on the meaning they give to their actions and the elements that push them to mobilization. The fear to the landfill center in Bure and the perceived threat that it represents have driven individuals to rally around a common struggle. What distinguishes this resistance from other similar opposition movements is the progressive development of a shared knowledge, which has gradually extended to issues that do not only concern nuclear waste. All the actors feel a common unease, frustration and lack of recognition that trouble and encourage them to action. Their shared experience, struggle for recognition, will to power and need to manifest feelings forge the life of the members of "la commune de Bure": spaces of experience are shaped, in which "concrete" and "imagined" alternatives to the dominant forms of life and society emerge. It is in this way that representations and practices transform spaces of experience into counter-publics. Persistent mental structures, deep feelings and meaningful interactions become the motor of mobilization and the impulse towards a process of subjectivation that concerns the acting subjects. In addition, there is a will that unfolds, a will to overcome uneasiness and the determination to create an alternative future. This dissertation aims to analyze the "potentiel d'agir" of an exemplary case by means the Bure's movement, its capacity to create and organize a delimited public space through action able to bring together worldviews of resistance to the established order and new possibilities for change. The experience of conflict and the quest for recognition can make people reflect on the unease that affects society and the possible future alternatives

This chapter explores methodological challenges in understanding the relationships between processes, products, and perspectives within digital multimodal composition. Using examples from a research project concerning multilingual and multimodal composing in an eighth-grade classroom in the United States, the authors describe specific challenges – and possible avenues forward – in relation to screen-capture software, student retrospective design interviews, and multimodal timescapes. The chapter concludes with implications from these challenges for research and instruction for emergent bilingual students.

AbstractEcolabels and green taxes aim to achieve more sustainable market outcomes by affecting suppliers’ production and sales behaviour, consumers’ purchasing behaviour, or both. In this chapter, we present the economic rationale for how these approaches may impact suppliers and consumers in various settings and review recent published empirical research on the topic. We focus specifically on examples where ecolabels and green taxes have been used to protect oceans and fisheries by reducing plastic waste and reducing purchases of less sustainable seafood. We conclude by discussing other possible policy instruments and highlight important avenues for future work in pursuing more sustainable market outcomes.

AbstractBy unraveling the dimensions and dynamics of the ‘cycle of protractedness’, the concluding chapter contributes to the development and reexamination of key concepts in the field of International Relations, providing a renewed analysis of the role of the Israeli-Palestinian protracted peace process in the conflict and its transformation. It concludes that, even though dehumanization dominates in ongoing conflicts, it is possible to identify activities undertaken by local and international actors in the societal level that have developed alongside—as a consequence—and within the very framework—as a desired outcome—of the protracted peace process. In terms of policy design and policy making, the findings of this book provide some clues for the intensification of new—underexplored—avenues for conflict transformation that favor reconciliation rather than reinforcing dehumanization. Although focused on a single case study, being that of Israel and Palestine, the findings of this research can be applied to other cases of protracted social conflicts by way of lessons learned.

AbstractThis chapter summarizes the key insights derived from a three-dimensional perspective on EU–Turkey relations that combines the analytical lenses of (1) theories and concepts, (2) institutions, and (3) policies. It furthermore reflects upon the different periodizations of the EU–Turkey relationship. On this basis, we offer a systematic survey of the conditions under which cooperative trends in EU–Turkey relations could be (re)invigorated. The analysis shows that despite the high potential for reciprocity inside and outside the accession framework, the relationship currently rests on unfavorable conditions for cooperation on both sides. Important enablers of cooperative behavior—trust, communication, reputation, fairness, enforcement, and common identity—cannot properly operate in the current setup. The chapter puts forward possible means to allow for these enablers to facilitate cooperative behavior in EU–Turkey relations in the future. It concludes by discussing the future trajectory of the relationship and pinpointing avenues for a future research agenda for EU–Turkey studies.

AbstractThe research on software startups has increased lately, focusing on describing how these companies’ unique context influences development practices. The next step for research is the creation of specific practices for these companies grounded in scientific results. An obstacle in this path is which dependent variable these novel practices should improve. A natural answer is these companies’ success. This position paper reviews the literature on new ventures and startups’ success to show that telling if a startup is successful or not is a complex issue. As a solution to this problem, this paper proposes a conceptual framework, suggesting that novel practices should improve success determinants or reduce inhibitors rather than focusing on the startups’ success. Three examples illustrate the framework’s use: hypotheses engineering, microservices, and BizDev. The identification of contributors and inhibitors for success of software startups could enrich the framework and indicate possible avenues for the creation of development practices specific tailored for these companies.

The present study addresses the implicit monolingual bias in our understanding of polar question-answer sequences by examining them in bilingual conversation. Drawing upon a corpus of naturally-occurring conversational data amongst native Spanish-English bilinguals in the southwestern United States, I target and explore the ‘fit’ between polar questions and answers in this community of practice. In this report, I focus specifically on the dimension of language (non-)concordance between particle answers and the questions they address. Evidence is offered that speakers produce response particles that are concordant with the language of the question as the pragmatically unmarked answer format, whereas response particles that are non-concordant with the language of the question (i.e., ‘code-switched’) are marked, produced agentively and for cause, routinely indexing an emergent stance that is at variance with the terms established by the question. Some possible avenues for future comparative work on the expression of agency in question-answer sequences are explored in the Conclusion.

In the presented study a combination of document analysis, semi-structured interviewing and observation was used to study current assessment practices in the context of three different funding calls to which inter- or trans-disciplinary research proposals were submitted. Findings from the three calls were compared to best practices presented in the existing literature and used to make recommendations for possible improvements, reflect on possible barriers for effective reforms, and suggest further research avenues for a larger potential follow-up project.

High harmonics generated with a femtosecond laser provides virtually all of the properties that one would wish to apply to photoemission investigations of electronic states in materials. Harmonic light is monochromatic and polarized, as determined by the input laser. It is tunable over a wide range of photon energies. For even modest input intensities (0.5 mJ per pulse in our system) we have been able to generate harmonics with useful fluxes to 80 eV. The generated harmonic light is co-linear with the input laser so that reflective optics such as normal or grazing incidence gratings of reasonable size can be used to collect and select a particular harmonic of interest and then refocus this light onto the material under study. With efficient electron detection schemes useful photoelectron count rates can be obtained. With these properties alone, harmonics makes possible photoelectron spectroscopy experiments in the laboratory setting that were previously the sole domain of large scale synchrotron labs. But an additional, and singularly important, aspect of harmonic light is its femtosecond, and possibly even attosecond, time structure which opens avenues for investigations of the temporal evolution of excited electronic systems.

Al igual que ocurre en otras ciudades, el Centro de Montevideo ha visto desplazada su vitalidad fundacional. La investigación se propuso develar el paisaje de nuestra principal avenida, frente a esta situación de deterioro. Para ello buscó identificar, relevar y analizar las construcciones de las manzanas frentistas a la avenida 18 de Julio entre Plaza Independencia y Ejido. Culmina señalando las oportunidades de inversión en estructuras existentes poniendo a disposición un catálogo de posibles intervenciones para desencadenar un proceso de revitalización del Centro de Montevideo. En la identificación de edificaciones se utilizaron seis categorías analíticas: fósiles, construcciones subutilizadas, baldíos, esqueletos, edificios públicos degradados y galerías, y se definieron tres estrategias: colmatación, recuperación de fósiles y recuperación de galerías. Se identificaron 114 padrones pasibles de la aplicación de alguna de las tres estrategias. Cuando dos o más de estos padrones son linderos se considera que se está frente a una zona de oportunidad, que habilita a transformaciones de mayor impacto. As is the case in other cities, the Downtown of Montevideo has been displaced it foundational vitality. This research aimed to reveal the landscape of our main Avenue, in relation to the situation found, of continuing deterioration. This sought to identify, survey and analyze the constructions belonging to the 18 de Julio Avenue between Plaza Independencia and Ejido street. The work finishes with investment opportunities in existing structures by providing a catalog of possible interventions to trigger a process of revitalization of downtown Montevideo. In identifying buildings used six analytical categories: fossil, underutilized buildings, vacant, skeletons, degraded public buildings and galleries, and defined three strategies: completion, fossil recovery and recovery of galleries. We identified 114 sites where it is possible the implementation of any of the three strategies. When two or more of these sites are boundaries being considered an area of opportunity, which enables high impact changes.

Engineers spend a significant amount of time searching for the right product information during the design and manufacturing process. A large amount as well as various types of product information have been generated and are available within the engineering databases. But representing and indexing the products effectively and retrieving them efficiently, remains a challenge. The purpose of this survey is to document the current state of research and development. We then identify avenues for exploration and provide a comparison of the advantages, disadvantages, and limitations among the various techniques. This survey concludes by suggesting possible future research directions.

This paper presents SCATR: a novel, high-efficiency electricity generation system with carbon separation. The system uses oxygen from an air separation unit to burn methane in supercritical water, expanding the resulting products stream through turbines to generate power. The water is condensed, preheated, and recycled through the plant, leaving a stream of CO2 compressed to pipeline pressures. The details of the model are discussed, and an exergy analysis of the system is performed, providing insight into the operation of the plant as well as possible avenues for improvements. The efficiency of the system, including air separation and CO2 separation, is found to be 42.3% relative to the HHV of methane (or 47.0% relative to its LHV).

Goal recognition is the task of recognizing the objective of agents based on online observations of their behavior. Goal recognition design (GRD), the focus of this survey, facilitates goal recognition by the analysis and redesign of goal recognition models. In a nutshell, given a model of a domain and a set of possible goals, a solution to a GRD problem determines: (1) to what extent do actions performed by an agent reveal the agent’s objective? and (2) what is the best way to modify the model so that the objective of an agent can be detected as early as possible? GRD answers these questions by offering a solution for assessing and minimizing the maximal progress of any agent before recognition is guaranteed. This approach is relevant to any domain in which efficient goal recognition is essential and in which the model can be redesigned. Applications include intrusion detection, assisted cognition, computer games, and human-robot collaboration. This survey presents the solutions developed for evaluation and optimization in the GRD context, a discussion on the use of GRD in a variety of real-world applications, and suggestions of possible future avenues of GRD research.

Bayesian nonparametric space partition (BNSP) models provide a variety of strategies for partitioning a D-dimensional space into a set of blocks, such that the data within the same block share certain kinds of homogeneity. BNSP models are applicable to many areas, including regression/classification trees, random feature construction, and relational modelling. This survey provides the first comprehensive review of this subject. We explore the current progress of BNSP research through three perspectives: (1) Partition strategies, where we review the various techniques for generating partitions and discuss their theoretical foundation, `self-consistency'; (2) Applications, where we detail the current mainstream usages of BNSP models and identify some potential future applications; and (3) Challenges, where we discuss current unsolved problems and possible avenues for future research.

Se evaluará un proceso de re-composición de la forma urbana del Centro Histórico Tradicional de la ciudad de Bogotá, usando el espacio público como elemento y estructura de sutura urbana, sutura de la forma y sutura social, de los microterritorio urbanos. Los nodos de articulación de la forma urbana serán los elementos constitutivos del espacio público, asociados a los espacios arquitectónicos, urbanos y naturales que por su uso y/o afectación, puedan ser vinculados como elementos constitutivos de la categoría del espacio público efectivo. El eje de la Avenida de los Comuneros se establece como una oportunidad urbanística, debido a que la intervención para la ampliación de la sección vial, produjo un detrimento urbano-arquitectónico en ambos lados de la vía, que está sujeto a la destrucción de las morfologías regulares por la segmentación de las manzanas, resultando un perfil “configurado” por culatas, sin relación edificio-calle, contribuyendo a la perdida de la vitalidad y el carácter del territorio específico. A process of re-composition the urban form of Historic Traditional Center of Bogotá will be evaluated using the public space as an element and structure of urban suture, form suture and socially suture, of the urban micro territories. The form articulating nodes will be determinates as the components of the public space, associated with possible architectural spaces, urban and natural sites that because of its use and/or commitment could be linked as constituent elements of the effective public space category. The axis of the “Avenida de los Comuneros” is set as an urban planning opportunity, because the intervention to expand the avenue section was an urban-architectural detriment on both sides of the road, which are subject of destruction of the regular morphologies by block segmentation, ending up in a profile “set” by stocks, with no building-street relationship, contributing to the vitality`s loss and identity of the specific territory.

The Eddy Dissipation Concept (EDC), proposed by Magnussen (1985), advances the concept that the reactants are homogeneously mixed within the fine eddy structures of turbulence and that the fine structures may therefore be regarded as perfectly stirred reactors (PSRs). To understand more fully the extent to which such a sub-grid scale stirred reactor concept could be applied within the context of a computational fluid dynamics (CFD) calculation to model local or global extinction phenomena: (1) various kinetic mechanisms are investigated with respect to CPU penalty and predictive accuracy in comparisons with stirred reactor lean blowout (LBO) data and (2) a simplified time-scale comparison, extracted from the EDC model and applied locally in a fast-chemistry CFD computation is evaluated with respect to its capabilities to predict attached and lifted flames. Comparisons of kinetic mechanisms with PSR lean blowout data indicate severe discrepancies in the predictions with the data and with each other. Possible explanations are delineated and discussed. Comparisons of the attached and lifted flame predictions with experimental data are presented for some benchscale burner cases. The model is only moderately successful in predicting lifted flames and fails completely in the attached flame case. Possible explanations and research avenues are reviewed and discussed.

The Earth system and human health are inextricably linked. In this period of widespread and rapid global change, understanding the challenges and opportunities at the intersection of health, geoscience, and the Earth system (broadly termed “geohealth”) has taken on a new level of importance. Scientific discovery and innovation can play a key role in helping humanity understand and respond to environmental, technological, and societal drivers of global changes so as to enable security, prosperity, and positive health outcomes for all. This AMS Policy Program study synthesizes input from the AMS community on the various connections, gaps, and opportunities that currently exist at the geohealth interface. Through these community discussions this study identifies: 1) a set of critical throughlines for effective convergence in geohealth research, 2) overarching challenges that currently impede progress, and 3) potential solution areas where significant progress might be made quickly. The geohealth professionals contributing to this report covered a wide range of geohealth topics. In addition to identifying overarching challenges that impact most of all areas of the field, the contributors suggested more narrowly focused areas where rapid progress might be possible. The report includes these as examples that might foster productive new avenues for research funding, while recognizing that the rapid nature of this study meant that it could not be exhaustive in its coverage of the field and that a different set of contributors might have provided a much different set of examples.

The city of Kampala in Uganda provides an illustrative example of how institutional and administrative reform, without widespread policy change, can generate substantial increases in municipal revenues. Through the implementation of more efficient digitalised systems, attracting higher capacity staff, and a focus on the ‘citizen as a client’, the city has managed to increase own-source revenues three-fold from UGX 30 billion (US$8.2 million) in 2010/11 to UGX 90 billion (US$25 million) in 2018/191, as well as crowd in more central government and donor funds. These reforms were made possible by strong leadership, a political window to act, and strong support from development partners. What was striking was the administration’s reflection that its most significant success was not in doing something new, but rather in doing its job as it is meant to be done. Furthermore, the reforms contributed to achieving an investment-grade credit rating in 2015, creating the potential for increased funding opportunities for large-scale investments in the future. This, coupled with recent regulatory change to remove the previously restrictive 10 per cent cap on borrowing, provides promising new avenues for attracting investments. However, a number of stakeholders noted concerns that, despite overcoming regulatory hurdles, the city is still not equipped with the capacity to develop bankable plans and projects. For development partners, the example of Kampala highlights the need for collaboration to build capacity, both in reforming systems and in designing implementable strategies and bankable projects worthy of external finance. However, concerns around the way development finance skews prioritisation of projects, as well as the difficulties in providing for ongoing maintenance which are often not accounted for in development partner investments, were also highlighted.

Latin America and the Caribbean is the most violent region in the world, with an annual homicide rate of more than 20 per 100,000 population and with an increasing trend. Yet most evidence of crime concentration, geo-temporal patterns, and event dependence comes from cities in high-income countries. Understanding crime patterns in the region and how they compare to those in high-income countries is of first-order importance to formulate crime reduction policies. This paper is the first to analyze crime patterns of cities in five Latin American countries. Using micro-geographic units of analysis, the paper finds, first, that crime in Latin America is highly concentrated in a small proportion of blocks: 50 percent of crimes are concentrated in 3 to 7.5 percent of street segments, and 25 percent of crimes are concentrated in 0.5 to 2.9 percent of street segments. This validates Weisburd's "law of crime concentration at place" (Weisburd, 2105). These figures are fairly constant over time but sensitive to major police reforms. The second finding is that hot spots of crime are not always persistent. Crime is constantly prevalent in certain areas, but in other areas hot spots either appear or disappear, suggesting a possible rational adaptation from criminals to police actions that cause crime displacement in the medium run to other areas. Finally, the paper finds a significant pattern of repeated crime victimization in location and time for property crimes. There are striking similarities with the developed world in crime concentration, although crime levels are much higher and usually increasing. There are also some differences in terms of the persistence of hot spots that pose interesting policy implications and avenues for future research.

The overall objective of the current proposal is to develop procedures to improve the pregnancy rate achieved following transfer of fresh or cryopreserved embryos produced in the laboratory into heat-stress recipients. The overall hypothesis is that pregnancy rate in heat-stressed lactating cows can be improved by use of embryo transfer and that additional gains in pregnancy rate can be achieved through development of procedures to cryopreserve embryos, select embryos most likely to establish and maintain pregnancy after transfer, and to enhance embryo competence for post-transfer survival through manipulation of culture conditions. The original specific objectives were to 1) optimize procedures for cryopreservation (Israel/US), 2) develop procedures for identifying embryos with the greatest potential for development and survival using the remote monitoring system called EmbryoGuard (Israel), 3) perform field trials to test the efficacy of cryopreservation and the EmbryoGuard selection system for improving pregnancy rates in heat-stressed, lactating cows (US/Israel), 4) test whether selection of fresh or frozen-thawed blastocysts based on measurement of group II caspase activity is an effective means of increasing survival after cryopreservation and post-transfer pregnancy rate (US), and 5) identify genes in blastocysts induced by insulin-like growth factor-1 (IGF-1) (US). In addition to these objectives, additional work was carried out to determine additional cellular determinants of embryonic resistance to heat shock. There were several major achievements. Results of one experiment indicated that survival of embryos to freezing could be improved by treating embryos with cytochalasin B to disrupt the cytoskeleton. An additional improvement in the efficacy of embryo transfer for achieving pregnancy in heat-stressed cows follows from the finding that IGF-1 can improve post-transfer survival of in vitro produced embryos in the summer but not winter. Expression of several genes in the blastocyst was regulated by IGF-1 including IGF binding protein-3, desmocollin II, Na/K ATPase, Bax, heat shock protein 70 and IGF-1 receptor. These genes are likely candidates 1) for developing assays for selection of embryos for transfer and 2) as marker genes for improving culture conditions for embryo production. The fact that IGF-1 improved survival of embryos in heat-stressed recipients only is consistent with the hypothesis that IGF-1 confers cellular thermotolerance to bovine embryos. Other experiments confirmed this action of IGF-1. One action of IGF-1, the ability to block heat-shock induced apoptosis, was shown to be mediated through activation of the phosphatidylinositol 3-kinase pathway. Other cellular determinants of resistance of embryos to elevated temperature were identified including redox status of the embryo and the ceramide signaling pathway. Developmental changes in embryonic apoptosis responses in response to heat shock were described and found to include alterations in the capacity of the embryo to undergo caspase-9 and caspase-3 activation as well as events downstream from caspase-3 activation. With the exception of IGF-1, other possible treatments to improve pregnancy rate to embryo transfer were not effective including selection of embryos for caspase activity, treatment of recipients with GnRH.and bilateral transfer of twin embryos. In conclusion, accomplishments achieved during the grant period have resulted in methods for improving post-transfer survival of in vitro produced embryos transferred into heat-stressed cows and have lead to additional avenues for research to increase embryo resistance to elevated temperature and improve survival to cryopreservation. In addition, embryo transfer of vitrified IVF embryos increased significantly the pregnancy rate in repeated breeder cows.

This project was based on the hypothesis that nematodes interacting with plants as either parasites or beneficial saprophytes are attracted to their host by natural products. This concept was supported by numerous observations that parasitic nematodes are attracted to root exudates. Our overall goal was to identify nematode sensory compounds from root exudates and to use that information for reducing nematicide applications. We applied skills of the investigators to achieve three specific objectives: 1) Identify nematode behavioral cues (e.g., attractants or repellents) in root exudates; 2) Identify new natural nematicidal compounds; and 3) Combine a natural attractant and a nematicide into a nematode trap. Because saprophytic nematodes benefit plants by mineralizing organic matter, we sought compounds attractive primarily to parasitic nematodes. The project was constructed on several complementary foundations. First, data from Dr. Spiegel’s lab showed that under aseptic conditions Ditylenchus dipsaci, a parasite on onion, is attracted to certain fractions of onion root exudates. Second, PI Phillips had a sizeable collection of natural plant products he had identified from previous work on Rhizobium-legume interactions, which could be tested “off the shelf”. Third, Dr. Ferris had access to aseptic and natural populations of various saprophytic and parasitic nematodes. The project focused on five nematode species: D.dipsaci, Heterodera avenae, and Tylenchulussemipenetransat ARO, and Meloidogyne javanicand Caenorhabditis elegans at UCD. Ten pure plant compounds, mostly flavonoids, were tested on the various nematode species using six different assay systems. Results obtained with assorted test systems and by various scientists in the same test systems were essentially irreproducible. Many convincing, Many convincing, i.e. statistically significant, results in one system or with one investigator could not be repeated with other assays or different people. A recent report from others found that these compounds, plus another 30, were inactive as attractants in three additional parasitic nematode species (Wuyts et al. Nematology 8:89- 101, 2006). Assays designed to test the hypothesis that several compounds together are required to attract nematodes have thus far failed to find a reproducibly active combination. In contrast to results using pure plant compounds, complex unfractionated exudates from aseptic onion root reproducibly attracted D. dipsaci in both the ARO and UCD labs. Onion root exudate collection, separation into HPLC fractions, assays using D. dipsaci and MS-MS experiments proceeded collaboratively between ARO and UCD without any definitive identification of an active compound. The final active fraction contained two major molecules and traces of several other compounds. In the end, analytical studies were limited by the amount of onion root exudate and the complexity of the purification process. These tests showed that aseptic plant roots release attractant molecules, but whether nematodes influence that release, as insects trigger release of attractants from plants, is unknown. Related experiments showed that the saprophyte C. elegans stimulates its prey, Pseudomonas bacteria, to increase production of 2, 4-diacetylphloroglucinol (DAPG) a compound that promotes amino acid exudation by plant roots. It is thus possible that saprophytic nematodes are attracted primarily to their bacterial or fungal prey and secondarily to effects of those microorganisms on root exudation. These observations offer promising avenues for understanding root-zone interactions, but no direct routes to controlling nematodes in agriculture were evident. Extracts from two plant sources, Chrysanthemum coronarium and Sequoia sempervirens, showed nematicidal activity at ARO and UCD, respectively. Attempts to purify an active compound from S. sempervirens failed, but preliminary results from C. coronarium are judged to form a potential basis for further work at ARO. These results highlight the problems of studying complex movement patterns in sentient organisms like nematodes and the issues associated with natural product isolation from complex mixtures. Those two difficulties combined with complications now associated with obtaining US visas, slowed and ultimately limited progress on this project. As a result, US investigators expended only 65% of the $207,400 originally planned for this project. The Israeli side of the project advanced more directly toward its scientific goals and lists its expenditures in the customary financial report.

Background : Banana being a monocot and having distinct peel and pulp tissues is unique among the fleshy fruits and hence can provide a more comprehensive understanding of fruit ripening. Our previous research which translated ripening discoveries from tomato, led to the identification of six banana fruit-associated MADS-box genes, and we confirmed the positive role of MaMADS1/2 in banana ripening. The overall goal was to further elucidate the banana ripening signaling pathway as mediated by MADS-boxtranscriptional regulators. Specific objectives were: 1) characterize transcriptional profiles and quality of MaMADS1/2 repressed fruit; 2) reveal the role of additional MaMADSgenes in ripening; 3) develop a model of fruit MaMADS-box mode of action; and 4) isolate new components of the banana ripening signaling pathway. Major conclusion: The functions of the banana MaMADS1-5 have been examined by complimenting the rinor the TAGL1-suppressed lines of tomato. Only MaMADS5 exhibited partial complementation of TAGL1-suppressed and rinlines, suggesting that while similar genes play corresponding roles in ripening, evolutionary divergence makes heterologous complementation studies challenging. Nevertheless, the partial complementation of tomato TAGL1-surpessed and rinlines with MaMADS5 suggests this gene is likely an important ripening regulator in banana, worthy of further study. RNA-seqtranscriptome analysis during ripening was performed on WT and MaMADS2-suppressed lines revealing additional candidate genes contributing to ripening control mechanisms. In summary, we discovered 39 MaMADS-box genes in addition to homologues of CNR, NOR and HB-1 expressed in banana fruits, and which were shown in tomato to play necessary roles in ripening. For most of these genes the expression in peel and pulp was similar. However, a number of key genes were differentially expressed between these tissues indicating that the regulatory components which are active in peel and pulp include both common and tissue-specific regulatory systems, a distinction as compared to the more uniform tomato fruit pericarp. Because plant hormones are well documented to affect fruit ripening, the expressions of genes within the auxin, gibberellin, abscisic acid, jasmonic acid, salicylic and ethylene signal transduction and synthesis pathways were targeted in our transcriptome analysis. Genes’ expression associated with these pathways generally declined during normal ripening in both peel and pulp, excluding cytokinin and ethylene, and this decline was delayed in MaMADS2-suppressed banana lines. Hence, we suggest that normal MaMADS2 activity promotes the observed downward expression within these non-ethylene pathways (especially in the pulp), thus enabling ripening progression. In contrast, the expressions of ACSand ACOof the ethylene biosynthesis pathway increase in peel and pulp during ripening and are delayed/inhibited in the transgenic bananas, explaining the reduced ethylene production of MaMADS2-suppressed lines. Inferred by the different genes’ expression in peel and pulp of the gibberellins, salicylic acid and cytokinins pathways, it is suggested that hormonal regulation in these tissues is diverse. These results provide important insights into possible avenues of ripening control in the diverse fruit tissues of banana which was not previously revealed in other ripening systems. As such, our transcriptome analysis of WT and ripening delayed banana mutants provides a starting point for further characterization of ripening. In this study we also developed novel evidence that the cytoskeleton may have a positive role in ripening as components of this pathway were down-regulated by MaMADS2 suppression. The mode of cytoskeleton involvement in fruit ripening remains unclear but presents a novel new frontier in ripening investigations. In summary, this project yielded functional understanding of the role and mode of action of MaMADS2 during ripening, pointing to both induction of ethylene and suppression of non-ethylene hormonal singling pathways. Furthermore, our data suggest important roles for cytoskeleton components and MaMADS5 in the overall banana ripening control network. Implications: The project revealed new molecular components/genes involved in banana ripening and refines our understanding of ripening responses in the peel and pulp tissues of this important species. This information is novel as compared to that derived from the more uniform carpel tissues of other highly studied ripening systems including tomato and grape. The work provides specific target genes for potential modification through genetic engineering or for exploration of useful genetic diversity in traditional breeding. The results from the project might point toward improved methods or new treatments to improve banana fruit storage and quality.
