Institué par ordonnance royale en 1839 le consistoire luthéro-réformé d’Alger comprenait, outre les pasteurs, douze délégués laïcs, choisis parmi les notables protestants de la ville. Longtemps les réformés dominèrent cette assemblée et ce n’est qu’après les réformes de 1859 et de 1867 que les luthériens furent assez nombreux pour compter, puis qu’ils accédèrent à la parité. Dix figures particulièrement remarquables sont évoquées ici à partir des archives de l’Église de la Confession d’Augsbourg et de celles de l’Église protestante d’Algérie. Elles sont assez représentatives de la composition de cet organe ecclésial atypique à l’époque, puisque appartenant officiellement à la fois à l’Église de la Confession d’Augsbourg et à l’Église réformée.

Cet article vise à révéler les grandes composantes de la rhétorique discursive lors du règlement de litiges fonciers en pays atchan (peuple ébrié situé au sud-est de la Côte d’Ivoire). Le jeu des échanges verbaux autour des lois coutumières sont le reflet d’une vision du monde qui met le bien-être communautaire au centre de toute préoccupation. Nous avons utilisé comme technique de recherche qualitative l’observation d’un fait socioculturel. Aussi, avons -nous assisté à une assemblée de notables, pour régler un litige foncier dans le village d’Akoue-djεmɛ. Le déroulé de cette procédure judiciaire coutumière a été transcrit, puis analysé selon l’organisation de la rhétorique discursive. Ce travail a permis de découvrir les trois grandes parties de la séance du règlement dudit conflit. La rhétorique du discours social atchan est faite de jeux intertextuels ponctués par des formules ritualisées et proverbiales. This article aims at showing the major components of the discursive rhetoric during land dispute resolution among the Atchan community (The Ebrie people located in the South west of Cote d’Ivoire). The way of going about the oral communication talking about customary laws truly reflects a world vision that sets the community well-being as the prior preoccupation. The qualitative methodology is the one used. We took part in a gathering of dignitaries solving a land dispute in the Akoue-djεmɛ village. The event of this customary court has been transcribed and analyzed according to the discursive rhetoric organization. This study enabled us to discover the three major components of the sitting. The rhetoric of the Atchan social speech is made up with inter-textual games with rituals and proverbs.

Abstract Motivation RNA sequencing (RNA-Seq) is a widely used approach to obtain transcript sequences in non-model organisms, notably for performing comparative analyses. However, current bioinformatic pipelines do not take full advantage of pre-existing reference data in related species for improving RNA-Seq assembly, annotation and gene family reconstruction. Results We built an automated pipeline named CAARS to combine novel data from RNA-Seq experiments with existing multi-species gene family alignments. RNA-Seq reads are assembled into transcripts by both de novo and assisted assemblies. Then, CAARS incorporates transcripts into gene families, builds gene alignments and trees and uses phylogenetic information to classify the genes as orthologs and paralogs of existing genes. We used CAARS to assemble and annotate RNA-Seq data in rodents and fishes using distantly related genomes as reference, a difficult case for this kind of analysis. We showed CAARS assemblies are more complete and accurate than those assembled by a standard pipeline consisting of de novo assembly coupled with annotation by sequence similarity on a guide species. In addition to annotated transcripts, CAARS provides gene family alignments and trees, annotated with orthology relationships, directly usable for downstream comparative analyses. In this paper we demonstrate the power of a model of tile self-assembly based on active glues which can dynamically change state. We formulate the Signal-passing Tile Assembly Model (STAM), based on the model of Padilla et al. [24] to be asynchronous, allowing any action of turning a glue on or off, attaching a new tile, or breaking apart an assembly to happen in any order. Within this highly generalized model we provide three new solutions to tile self-assembly problems that have been addressed within the abstract Tile Assembly Model and its variants, showing that signal passing tiles allow for substantial improvement across multiple complexity metrics. Our first result utilizes a recursive assembly process to achieve tile-type efficient assembly of linear structures, using provably fewer tile types than what is possible in standard tile assembly models. Our second system of signal-passing tiles simulates any Turing machine with high fuel efficiency by using only a constant number of tiles per computation step. Our third system assembles the discrete Sierpinski triangle, demonstrating that this pattern can be strictly self-assembled within the STAM. This result is of particular interest in that it is known that this pattern cannot self-assemble within a number of well studied tile self-assembly models. Notably, all of our constructions are at temperature 1, further demonstrating that signal-passing confers the power to bypass many restrictions found in standard tile assembly models.

The paper is devoted to the position of the French aristocracy regarding the projects of the Controller General of Finance Calonne, presented for discussion at the Assembly of notables in 1787. The budget deficit by the end of the 80s of 18th century reached over 80 million livres a year, the country was on the verge of an economic crisis. The Controller General of Finance Calonne proposed to the king to convene in 1787 an Assembly of notables, prominent representatives of the French nation, to approve a plan of government reforms. Although the Assembly included notables from the three estates of the kingdom, the aristocracy, which was widely represented in it, played an important role. The titled French nobility at the end of the Old Order still retained influence in the state, thanks to their economic position, social status and positions at court, in the army and in the state apparatus. The Assembly of notables had no legislative force, but Calonne convinced the king that the reform plan approved by the assembly would break the resistance of parliaments and gain the approval of the whole society. The presented reforms affected the interests of the privileged estates, but the government expected that the notables would accept the proposals and vote for the reforms, which according to Calonne contributed to the huge budget deficit, educational ideas about equality, physiocrats projects announced earlier and the chosen composition of notables, many of which were occupied by liberal and pro-government position. The Notables put forward their ideas on taxation and the creation of provincial assemblies and expressed the idea of convening the States General as a body competent to adopt such significant reforms for French society.

(1) Background: Dementia and mild cognitive impairment (MCI) are still underdiagnosed in the general population. Impaired odor identification has been identified as an early marker of MCI and dementia. We aim to investigate whether short tasks, in which simple forms must be assembled from single building blocks based on a template or while considering specific re-strictions, could increase the diagnostic quality of established cognitive screening tests in detecting MCI or dementia. (2) Methods: A brief assembly test, where participants had to assemble simple animal shapes from Lego® Duplo® building blocks, the Frontal Assessment Battery, and the Mini-Mental State Exam (MMSE) were administered to a consecutive series of 197 patients (89 with mild dementia, 62 with mild cognitive impairment, and 46 without cognitive impairment) referred for neuropsychological testing. (3) Results: Both participants with dementia and with MCI performed badly in the assembly tasks. The assembly tasks and the Frontal Assessment Battery were substantially correlated. Complementing MMSE scores with the assembly tasks improved the diagnostic accuracy of individuals with dementia and MCI. (4) Conclusions: People with suspected dementia or MCI may already benefit from simple assembly tasks. Although these tests require little additional time, they can notably increase sensitivity for dementia or MCI.

Molecular self–assembly is the automatic formation of functional assemblies of different structural components through weak, reversible, non–covalent interactions on the basis of molecular recognition. Amphiphilic molecules have a natural advantage in self–assembly at the gas/liquid interface. In this work, two amphiphilic molecules with a special molecular structure, indocyanine green (ICG) and a derivative of indocyanine green (CCS), were combined with two dye molecules (tetraphenylporphyrin tetrasulfonic acid hydrate (TPPS) and nickel (II) phthalocyanine–tetrasulfonic acid tetrasodium salt (TsNiPc) for self–assembly through the Langmuir–Blodgett (LB) technique. The nanostructure and assembly behavior in ordered self–assembled films are effectively regulated by inducing dye molecules to form different types of aggregates (H– and J–aggregates). In addition, we prepared composite films containing the same functional components using the conventional drop–casting technique and performed a series of comparative experiments with LB films. The degree of hydrophilicity was found to be related to roughness, with LB composite films being flatter and denser, with the lowest roughness and the best hydrophobicity compared to drop–casting films. Notably, the LB films showed better optoelectronic properties under the same conditions, providing new clues for the application of optoelectronic functional ultrathin film devices.

In this study, Russian olive trees exhibiting witches’-broom symptoms were collected from urban green areas in Tabriz, in the northwest of Iran. PCR analysis confirmed that phytoplasma caused the disease and, according to the resulting Sanger sequencing electropherogram, a mixed infection with two or more phytoplasma species within the Russian olive trees was revealed. Next-generation sequencing analyses, using the Illumina method, were performed on total DNA from the infected Russian olive plants to recognize the microbial genomic content and assemble the whole genome of the causative pathogen(s). The use of MetaphlAn2 and Kraken2 to analyze species composition revealed the very diverse and unique compositions of different Prokaryotic and Eukaryotic species within the infected plants. Several bacteria and fungi were discovered inside the samples, among which Mycoplasmatota was significantly dominating. Interestingly, the results also revealed a high level of endosymbiotic bacteria and Archaea (Methanobacteria) genome contents within the samples. Bowtie2, metaSPAdes, and CD-HIT pipelines were used to perform the initial genome assembly, and the whole genome of the notable phytoplasma species was assembled and submitted to Genbank.

Recent advances in sequencing technologies and platforms have enabled to generate metagenomics sequences using different sequencing platforms. In this study, we analyzed and compared shotgun metagenomic sequences generated by HiSeq3000 and BGISEQ-500 platforms from 12 sediment samples collected across the Norwegian coast. Metagenomics DNA sequences were normalized to an equal number of bases for both platforms and further evaluated by using different taxonomic classifiers, reference databases, and assemblers. Normalized BGISEQ-500 sequences retained more reads and base counts after preprocessing, while a slightly higher fraction of HiSeq3000 sequences were taxonomically classified. Kaiju classified a higher percentage of reads relative to Kraken2 for both platforms, and comparison of reference database for taxonomic classification showed that MAR database outperformed RefSeq. Assembly using MEGAHIT produced longer assemblies and higher total contigs count in majority of HiSeq3000 samples than using metaSPAdes, but the assembly statistics notably improved with unprocessed or normalized reads. Our results indicate that both platforms perform comparably in terms of the percentage of taxonomically classified reads and assembled contig statistics for metagenomics samples. This study provides valuable insights for researchers in selecting an appropriate sequencing platform and bioinformatics pipeline for their metagenomics studies.

AbstractDynamic gene expression seen during whole-body regeneration is likely controlled by genomic regulatory elements that dictate the spatiotemporal activity of the regeneration transcriptome. Identifying and characterizing these non-coding regulatory sequences are key to understanding how genes are connected into networks to deploy the process of whole-body regeneration. Here, we describe the application of the Assay for Transposase Accessible Chromatin (ATAC-seq) in the acoel Hofstenia miamia to identify regions of open chromatin that represent putative regulatory elements. Notably, when paired with gene knockdown techniques such as RNAi, ATAC-seq can be implemented in a functional genomics approach to validate putative regulatory elements. ATAC-seq requires no species-specific reagents, is amenable to small input cell numbers, and can be completed in a single day, making it an ideal assay to identify dynamic chromatin at high resolution during whole-body regeneration in virtually any species with a quality genome assembly.

AbstractDespite massive flood controlling investments, perennial flooding continues to be a major challenge in the Greater Accra Metropolitan Assembly in Ghana. Previous studies have mostly considered the vulnerability of Accra to flooding induced by urbanization and climate change. This chapter examined the impacts of and adaptation strategies to flooding in two flood-prone residential areas in Accra. A survey was conducted among 320 household heads to ascertain local impacts of floods and community adaptation strategies. To obtain a broader picture of government interventions and challenges, key stakeholders such as personnel from ministries, departments, and agencies who are involved in city planning, and private urban planning consultants were interviewed. The study found that a notable driver of floods in Accra is blocked waterways, and flawed and ad hoc engineering works. About three-quarters of the households interviewed have suffered flood-related losses over the past decade such as housing damage, income, and even a death of a relative. Key flood control interventions included dredging prior to start of rains and sporadic demolition of unauthorized buildings on or near waterways to allow free flow of water. However, these interventions only seem to be ephemeral due to the rapid rate of littering and re-siltation of the waterways after few rain events. The study highlights the need for more pragmatic and robust engineering solutions to build resilience of Accra to floods.

The chapter follows the progressive politicization of Colson and his neighbors during the so-called pre-Revolutionary period (1787–89), taking note of the extent to which he and they were taken totally by surprise by the events of that period. It examines, notably, Colson’s reaction to the Assembly of Notables of 1787; to the long struggle between Louis XVI and the Parlement of Paris; to the convocation of the Estates General for May 1789; and to the electoral assemblies and the statements of grievances (cahiers de doléances) drawn up in those assemblies. It also emphasizes Colson’s descriptions of the “hurricane” of July 1788 that destroyed a large proportion of the crops in northern France, and of the terrible winter of 1788–89 and its effects on the population of Paris.

Abstract Keeping Montlinot as inspector of the depot de mendicite at Soissons against his better judgment was not the last concession Calonne would be obliged to make to enlightened opinion. Historians date the “prerevolution” in France from Calonne’s decision to convoke an Assembly of Notables in February 1787. He calculated that by this extraordinary measure he might circumvent the obstruction that the parle ment would certainly raise against his plan for a general land tax-a tax to be collected without exemptions for the privileged and with the aid of consultative assemblies in each province. The strategy backfired. The Assembly of Notables called for a wholesale redress of grievances. Calonne turned over the reins of finance to his most outspoken critic in the Assembly, the archbishop of Toulouse, Lomenie de Brienne.

Nuclear power is a proven, safe, plentiful and clean source of power generation, and AP1000 is the only Generation III+ reactor to receive Design Certification from the U.S. Nuclear Regulatory Commission (NRC). One of the major features of this advanced reactor is the utilization of the passive safety systems and a steel Containment Vessel (CV). As the boundary isolated to keep the radioactive waste to release to outside and also the medium to transfer the heat, CV is a mainly part of the Passive Containment Cooling (PCS) System, which is the key to move the resident heat from the core to the air. The CV is made up with Sub-Assemblies, such as Bottom Head (CVBH), 1st Ring, 2nd Ring, 3rd Ring, and Top Head (CVTH), which are assembled with steel plates. So to build the Containment Vessel on site, we have to assemble each individual part with steel plates fabricated in shop first, transport the Sub-Assembly to Nuclear Island (NI) and lift them in place. Right now, the first AP1000 in the world is under construction at Sanmen, China. Based on the statement above, the construction of CVBH of the 1st AP1000 is a notable achievement and implementation without experience proven, the constructability analysis about work sequence, detailed logic, activity duration, production rate and improvement of them during the process is worthy to study.

Gas foil bearings (FB) satisfy many of the requirements noted for novel oil-free turbomachinery. However, FB design remains largely empirical, in spite of successful commercial applications. The mechanical structural characteristics of foil bearings, namely stiffness and damping, have been largely ignored in the archival literature. Four commercial bump-type foil bearings were acquired to measure their load capacity under conditions of no shaft rotation. The test bearings contain a single Teflon coated foil supported on 25 bumps. The nominal radial clearance is 0.036 mm for a 38 mm journal. A simple test set up was assembled to measure the FB deflections resulting from static loads. The tests were conducted with three shafts of increasing diameter to induce a degree of preload into the FB structure. Static measurements show nonlinear FB deflections, varying with the orientation of the load relative to the foil spot weld. Loading and unloading tests evidence hysteresis. The FB structural stiffness increases as the bumps-foil radial deflection increases (hardening effect). The assembly preload results in notable stiffness changes, in particular for small radial loads. A simple analytical model assembles individual bump stiffnesses and renders predictions for the FB structural stiffness as a function of the bump geometry and material, dry-friction coefficient, load orientation, clearance and preload. The model predicts well the test data, including the hardening effect. The uncertainty in the actual clearance (gap) upon assembly of a shaft into a FB affects most the predictions.

The remarkable energetic versatility and adaptability of skeletal muscle provides great inspiration to develop advanced adaptive structures and materials. These notable properties may arise from the assembly of skeletal muscle’s nanoscale cross-bridges into microscale structures known as sarcomeres. Essential understanding of muscle energetics has been developed from models of the micro/nanoscale constituents which incorporate an intriguing, asymmetric, bistable potential energy landscape to capture trends in the experimental data on cross-bridge power stroke motions. Inspired by the capability of cross-bridges to strategically absorb and store elastic energy for achieving high energetic efficiency of sudden motions, this research studies the assembly of modular mechanical structures from asymmetrically bistable constituents to absorb and trap energy in higher-energy stable configurations. Specifically, energy features of a module with an asymmetrically bistable element are first studied; these modules are then assembled in series and parallel to generate structures with multiple stable configurations having different quantities of stored energy. Dynamic analyses are performed to illustrate the ability of these structures to trap a portion of the kinetic energy due to an impulsive excitation as recoverable and reusable strain energy, and insight is gained into how asymmetries and damping influence the energy trapping performance.

Abstract Based on input from key operators in the Middle East region, a new rotary steerable system (RSS) was launched after a compressed development schedule. This paper describes the development and introduction of the larger tool sizes needed for both onshore and offshore hole sections, including hole sizes from 12 in. and up, in the Middle East. It also outlines the deliberate design of the tool for local assembly and repair. Large diameter (9-1/2 and 11 in.) RSS designs used an existing, smaller design for Middle East applications in both offshore and onshore wells as a basis. When designing these new sizes, engineers took note of lessons learned with smaller sizes of the tool and incorporated design elements for local manufacturing, assembly, and repair. The resulting simple, modular construction enables increased levels of local content and provides for significant reductions in transportation, and therefore associated emissions. Of course, although local content and sustainability are highly desirable, performance is essential, and this paper describes case histories demonstrating how well the new tool worked in real-world Middle East applications. In one notable example, the newly introduced 9 1/2-in. diameter RSS was used to drill an offshore section in the Gulf of Arabia. The tool was mobilized after two older generation RSS had become stuck for days. Consisting primarily of argillaceous limestone, the formation had a history of stuck-pipe events. The new RSS was recommended for this application because of a slicker construction, with a fully rotational bias unit, minimal bottom hole assembly (BHA) stabilization, and an optimized junk slot area, which together help to reduce stuck-pipe risks. The tool drilled to the target depth in a single run, thereby achieving all directional requirements. Notably, after reaching the target depth, the assembly was tripped out of the hole without any requirement for backreaming. This seamless exit, in turn, indicated achieving a smooth wellbore. Other case histories demonstrate results with both new sizes of this tool. The paper also discusses in detail the ability to repair locally and engage the local supply chain. Specifically with Middle East applications in mind, a new, simple RSS design in large diameter versions has demonstrated success in offshore and onshore applications across the region. The design has also proven capabilities for manufacturing and repair local to operations, which enables maximizing in-country value, optimizing use of the tools, and energizing local supply chains.

ASME PCC-1 “Guidelines for Pressure Boundary Bolted Joint Assembly” was first published in 2000 [1]. Since that time, there have been advances in bolted joint technology and industry practices that necessitated an update of the original document. The new revision of PCC-1 [2] is over twice the length of the original document and contains many new and updated guidelines. Significant additional detail has been added to the document in an effort to provide the broadest possible array of bolted joint assembly considerations to cover situations commonly encountered in industry, and thereby improve the overall safety and performance of bolted joints. This paper identifies some of the notable changes/additions that are included in the updated document.

Recently, low-dimensional quantum structures such as quantum dots (QDs) and quantum wires (QWIs), has been attracting much interest due to their novel physical properties and consequent improvements in device performances.1) When the ideal QD or QWI structures are achieved, higher gain and lower threshold current in laser diodes are expected.2) Among the many fabrication methods reported for such structures, self-assembled quantum-dot (SAQD) growth techniques3-5) are particularly notable. They positively utilize the islanding growth in highly strained heteroepitaxial systems, such as InGaAs on GaAs. The SAQDs can be simply fabricated by molecular beam epitaxy (MBE)3) or metal-organic vapor phase epitaxy (MOVPE)4),5) and they have high crystal quality and uniform size distributions of within 10% as well as high surface densities of more than about 1011cm-2. Using these SAQDs, low-threshold QD edge-emitting lasers have been fabricated.6-8) We expect to make even more advanced lasers, such as QD vertical-cavity surface-emitting lasers (VCSELs) using QDs in the active region.9) The QD-VCSEL is especially attractive for controlling both the electron and photon modes in a microcavity structure.10) When the cavity mode coincides with the narrow bandwidth light emission that originates from the delta-function-like density of states in uniform QDs, a high-performance light source with very low threshold current can be realized. On the other hand, the gain width, which critically determines the temperature characteristics of the VCSEL,11) can be designated in QD-VCSELs by controlling the dot size distribution. Therefore, for improving and modifying device performances, we believe that the QD-VCSEL is the optimum optical device utilizing the QD structure. In this article, we report the fabrication of a QD-VCSEL and the observation of lasing oscillation at room temperature.

Due to the safety issues arising from the Fukushima accident, a novel completely passive spent fuel pool cooling system is proposed using the high-efficiency heat pipe cooling technology that is available in an emergency condition such as a station blackout. This cooling system’s ability to remove the decay heat released by the spent fuel assemblies is evaluated by a computational fluid dynamics (CFD) simulation. The spent fuel pool of CAP1400 (a passive PWR developed in China) is selected as the reference pool, and the passive cooling system is designed for this spent fuel pool. The pool with the passive cooling system is simulated using Fluent 13.0 with 4 million meshes. Four different cases have been studied, and some notable results have been obtained through this work. The simulation results reveal that the passive cooling system effectively removes the decay heat from the SFP with the storage of 15-year-old spent fuel assemblies with emergency reactor core unloading and prevents the burnout of the fuel rods. The results indicate that the water in the SFP will never boil, even in a severe accident with a lack of emergency power and outside aid.

As environmental issues are raising more interest and are becoming crucial factors in all parts of the world, more and more environmental-friendly electronics products are emerging. Usually this means the introduction of products with lead-free solders. However, the reliability of lead-free solders is still a serious concern despite the vast research done in this field. This paper will describe the interconnect reliability of three kinds of solder joints respectively prepared with lead-free solder paste and lead-free PBGA components, lead-free solder paste and tin-lead-silver PBGA components, and tin-lead solder paste and tin-lead-silver PBGA components. Lead-free and tin-lead solders were composed of eutectic tin-silver-copper and tin-lead, respectively. In addition, the study also presents the effect of multiple reflow times. The study focuses on the microstructures of different assemblies. The particular interest is on the assemblies soldered with lead-free solder paste and tin-lead-silver PBGA components, since the SnPbAg solder on the bumps of the PBGA components were exposed to the reflow profile meant for the lead-free SnAgCu solder. Thus, these SnPbAg solder bumps were in the molten state almost twice as long as the rest of the solders. This had a notable effect on the reliability of these solder joints as we will be showing later in this paper. The test boards were temperature-cycled for 2500 cycles between −40 and +125°C (a 30-minute cycle). PBGA solder joint failures were monitored with a real time monitoring system. Optical and scanning electron microscopy was used to inspect the broken solder joints and their microstructure. The results of tests indicate that the number of reflow times can significantly affect the lifetime of PBGA solder joints. The most notable changes can be seen in the solder joints made with tin-lead-silver PBGA components and tin-silver-copper solder paste soldered with a lead-free reflow profile. The general trend was that the reliability of the solder joints increased in proportion to the number of reflow times. Mainly two factors are believed to have the major effect on the reliability of PBGA solder joints, voids, and microstructural changes in solder.

Abstract Over the past two years, the accuracy and repeatability of equipment used to assemble bolted flanged joints have been studied, and progress has been reported in past papers. One of the findings has been that pneumatic torque wrenches demonstrated a broader plus/minus range on bolt stress from the targeted value (e.g., lack of accuracy) than manual or hydraulic torque wrenches. What was notable, however, was that the repeatability of the bolt stress that was achieved did remain consistent with the other types of torque wrenches. Since the repeatability has been shown to be consistent, attention has turned to investigating and determining the reason for the perceived lack of accuracy. One aspect that might account for this is the calibration of the pneumatic torque wrenches. This paper outlines and documents the research that has recently taken place to identify the parameters that are key elements in improving the accuracy of pneumatic torque wrenches that might form the basis of development of a standard for calibration of powered torqueing equipment.

This study investigates the behavior of a turbopump assembly during critical cavitation of the propellant pumps in the upper rocket engine of the Korea Space Launch Vehicle-II. Turbopumps operate under conditions involving low pressure at the pump inlet and high rotational speeds to allow for a lightweight design. This severe environment can easily cause cavitation to occur in the pump. This cavitation can then cause the pump operation to fail. As the cavitation number in the pump decreases below the critical point, the pump fails to operate. There is concern regarding the behavior of the turbopump assembly arising from pump failure due to cavitation. It is necessary to verify the problems that may occur if the turbopump assembly operates under extreme conditions, such like the critical cavitation. This study performed tests to investigate the breakdown of pumps in the turbopump assembly. Tests were conducted with liquid nitrogen, water, and high-pressure air instead of the mediums used during actual operation of liquid oxygen, kerosene, and hot gas. The turbopump was tested at the design point of 27,000 rpm, while the inlet pressure of each pump was controlled to approach the critical cavitation number. The turbine power output was maintained during the tests. The results show that the breakdown point of the oxidizer pump using liquid nitrogen, which is a cryogenic medium, occurred at a lower cavitation number than during an individual component suction performance test using water. The fuel pump using water, meanwhile, experiences breakdown at similar cavitation numbers in both tests. As the breakdown of the pump occurs, the power required by that pump decreases, and the rotational speed of the turbopump increases. Compared with individual pump suction performance tests, this breakdown test can be used to determine the limit of the propellant inlet pressure of the turbopump and to characterize the behavior of the turbopump assembly when a breakdown occurs. Vibrations were also analyzed for tests at a high cavitation number and at the critical cavitation number. The vibration increased with breakdown and notable frequencies were analyzed.

Canada 1 Water is a 3-year governmental multi-department-private-sector-academic collaboration to model the groundwater-surface-water of Canada coupled with historic climate and climate scenario input. To address this challenge continental Canada has been allocated to one of 6 large watershed basins of approximately two million km2. The model domains are based on natural watershed boundaries and include approximately 1 million km2 of the United States. In year one (2020-2021) data assembly and validation of some 20 datasets (layers) is the focus of work along with conceptual model development. To support analysis of the entire water balance the modelling framework consists of three distinct components and modelling software. Land Surface modelling with the Community Land Model will support information needed for both the regional climate modelling using the Weather Research & Forecasting model (WRF), and input to HydroGeoSphere for groundwater-surface-water modelling. The inclusion of the transboundary watersheds will provide a first time assessment of water resources in this critical international domain. Modelling is also being integrated with Remote Sensing datasets, notably the Gravity Recovery and Climate Experiment (GRACE). GRACE supports regional scale watershed analysis of total water flux. GRACE along with terrestrial time-series data will serve provide validation datasets for model results to ensure that the final project outputs are representative and reliable. The project has an active engagement and collaborative effort underway to try and maximize the long-term benefit of the framework. Much of the supporting model datasets will be published under open access licence to support broad usage and integration.

The project sought to understand factors and mechanisms involved in the hardening of potato tubers. This syndrome inhibits heat softening due to intercellular adhesion (ICA) strengthening, compromising the marketing of industrially processed potatoes, particularly fresh peeled-cut or frozen tubers. However, ICA strengthening occurs under conditions which are inconsistent with the current ideas that relate it to Ca-pectate following pectin methyl esterase (PME) activity or to formation of rhamnogalacturonan (RG)-II-borate. First, it was necessary to induce strengthening of the middle lamellar complex (MLX) and the ICA as a stress response in some plant parenchyma. As normally this syndrome does not occur uniformly enough to study it, we devised an efficient model in which ICA-strengthening is induced consistently under simulated stress by short-chain, linear, mono-carboxylic acid molecules (OAM), at 65 oC [appendix 1 (Shomer&Kaaber, 2006)]. This rapid strengthening was insufficient for allowing the involved agents assembly to be identifiable; but it enabled us to develop an efficient in vitro system on potato tuber parenchyma slices at 25 ºC for 7 days, whereas unified stress was reliably simulated by OAMs in all the tissue cells. Such consistent ICA-strengthening in vitro was found to be induced according to the unique physicochemical features of each OAM as related to its lipophilicity (Ko/w), pKa, protonated proportion, and carbon chain length by the following parameters: OAM dissociation constant (Kdiss), adsorption affinity constant (KA), number of adsorbed OAMs required for ICA response (cooperativity factor) and the water-induced ICA (ICAwater). Notably, ICA-strengthening is accompanied by cell sap leakage, reflecting cell membrane rupture. In vitro, stress simulation by OAMs at pH<pKa facilitated the consistent assembly of ICAstrengthening agents, which we were able to characterize for the first time at the molecular level within purified insoluble cell wall of ICA-strengthened tissue. (a) With solid-state NMR, we established the chemical structure and covalent binding to cell walls of suberin-like agents associated exclusively with ICA strengthening [appendix 3 (Yu et al., 2006)]; (b) Using proteomics, 8 isoforms of cell wall-bound patatin (a soluble vacuolar 42-kDa protein) were identified exclusively in ICA-strengthened tissue; (c) With light/electron microscopy, ultrastructural characterization, histochemistry and immunolabeling, we co-localized patatin and pectin in the primary cell wall and prominently in the MLX; (d) determination of cell wall composition (pectin, neutral sugars, Ca-pectate) yielded similar results in both controls and ICA-strengthened tissue, implicating factors other than PME activity, Ca2+ or borate ions; (e) X-ray powder diffraction experiments revealed that the cellulose crystallinity in the cell wall is masked by pectin and neutral sugars (mainly galactan), whereas heat or enzymatic pectin degradation exposed the crystalline cellulose structure. Thus, we found that exclusively in ICA-strengthened tissue, heat-resistant pectin is evident in the presence of patatin and suberinlike agents, where the cellulose crystallinity was more hidden than in fresh control tissue. Conclusions: Stress response ICA-strengthening is simulated consistently by OAMs at pH< pKa, although PME and formation of Ca-pectate and RG-II-borate are inhibited. By contrast, at pH>pKa and particularly at pH 7, ICA-strengthening is mostly inhibited, although PME activity and formation of Ca-pectate or RG-II-borate are known to be facilitated. We found that upon stress, vacuolar patatin is released with cell sap leakage, allowing the patatin to associate with the pectin in both the primary cell wall and the MLX. The stress response also includes formation of covalently bound suberin-like polyesters within the insoluble cell wall. The experiments validated the hypotheses, thus led to a novel picture of the structural and molecular alterations responsible for the textural behavior of potato tuber. These findings represent a breakthrough towards understanding of the hardening syndrome, laying the groundwork for potato-handling strategies that assure textural quality of industrially processed particularly in fresh peeled cut tubers, ready-to-prepare and frozen preserved products.
