In this work, Oxford Nanopore sequencing is tested as an accessible method for quantifying heterogeneity of amplified DNA. This method enables rapid quantification of deletions, insertions, and substitutions, the probability of each mutation error, and their locations in the replicated sequences. Amplification techniques tested were conventional polymerase chain reaction (PCR) with varying levels of polymerase fidelity (OneTaq, Phusion, and Q5) as well as rolling circle amplification (RCA) with Phi29 polymerase. Plasmid amplification using bacteria was also assessed. By analyzing the distribut

A novel decoupled XY compliant micro-positioning stage, based on a bridge-type amplification mechanism and parallelogram mechanisms, is designed in this paper. Analytical models of the bridge-type amplification mechanism and parallelogram mechanisms are developed by Castigliano’s second theorem and a Beam constrained model. The amplification ratio, input stiffness, and output stiffness of the stage are further derived, based on the proposed model. In order to verify the theoretical analysis, the finite element method (FEM) is used for simulation and modal analysis, and the simulation results i

Abstract Xi’an 200 MeV proton application facility (XiPAF) upgrading project is now in the design phase. A six super periods synchrotron is the main component of this project. Using MADX program, a statical analysis of the relationship between closed-orbit distortion (COD) and different types of magnet errors was carried out. Because we do not know the exact error in the design phase, we use rms value to represent errors and CODs. The results show that COD is in direct proportion to magnet error in some cases while the others are square proportional. We regard all the cases as direct proportio

SUMMARYThis research presents a comprehensive and useful survey of the kinematic models of wheeled mobile robots and their optimal configurations. The kinematic modeling of wheeled mobile robots with no-slip is presented, by considering four common types of wheels: fixed, orientable, castor, and Swedish. Next, the accuracy of the kinematic models is discussed considering their sensitivity or relative error amplification, giving rise to the isotropy concept. As practical application of the previous theory, all types of three-wheeled mobile robots are modeled and their optimal (isotropic) config

Abstract Microbial cells under growth-limiting stress can generate mutations by mechanisms distinct from those in rapidly growing cells. These mechanisms might be specific stress responses that increase mutation rates, potentially altering rates of evolution, or might reflect non-stress-specific processes in rare growing cells. In an Escherichia coli model system, both frameshift reversion mutations and gene amplifications occur as apparent starvation-induced mutations. Whereas frameshift reversion (“point mutation”) requires recombination proteins, the SOS response, and error-prone DNA polyme

Hence, the main aim of this study was to compare two types of robotic rehabilitation on the immediate improvement of timing accuracy: haptic guidance (HG), which consists

Step-by-step instructions are given to implement these methods, as well as research supporting their efficacy. The motivation for this project developed from testing of a full scale building isolated with triple friction pendulum bearings on the E-defense shake table in Japan. The test demonstrated experimentally that the vertical component of ground motion can amplify both the base shear and the story acceleration in the isolated building. Vertical shaking introduced high-frequency variation in the axial force of the bearings, and, consequently, a high-frequency component in the bearing lateral force, which excited higher structural modes in the building.
