Texto completoThis work aimed to evaluate control strategies of hairy fleabane (Conyza bonariensis) and also study the arthropods associated with this weed. For this, the work was divided into three chapters. The Chapter I aimed to evaluate the selectivity of herbicides with potential use to control hairy fleabane on forming species winter pastures, which are: oats, black oats, ryegrass and tetraploid ryegrass. Experiments were conducted under greenhouse conditions and evaluated the phytotoxicity of plants and production of shoot dry matter (SDM). The treatments were: 2,4-D amine (806 and 1612 g a.i. ha-1), metsulfuron-methyl (2.4 and 4.8 g a.i. ha-1); clorimuronethyl (20 and 40 g a.i. ha-1); imazetaphyr (350 and 700 g a.i. ha-1); diclosulam (16.8 and 33.6 g a.i. ha-1); saflufenacil (35 and 70 g a.i. ha-1) and flumioxazin (50 and 100 g a.i. ha-1), and the control. According to the results obtained, the selectivity of herbicides is species cultivated dependent and it is important to know the botanical composition of pasture before applying any herbicide. In Chapter II aimed to evaluate the combination effect of the herbicides glyphosate and saflufenacil at different doses on the control of hairy fleabane plants. The treatments consisted of doses that followed the logarithmic scale, and used the herbicides glyphosate and saflufenacil both applied alone and in mixture. Were evaluated the visual control and the SDM production of hairy fleabane plants. The results indicate that there is a synergistic relationship between the herbicide glyphosate and saflufenacil. Also, the association of saflufenacil and glyphosate is effective in controlling glyphosate-resistant hairy fleabane. The addition of the herbicide glyphosate with saflufenacil, besides improve control, prevents the occurrence of regrowth of hairy fleabane plants. In Chapter III, hairy fleabane plants were collected fortnightly in Boa Vista and Sao Vicente Incra South in order to evaluate the arthropods associated with this weed. The results show that hairy fleabane takes an important role as an alternative host of important pests of the soybean, such bugs and caterpillars, and too natural enemies.
Os tratamentos avaliados foram: 2,4-D amina (806 e 1612 g i.a. ha-1); metsulfuron-methyl (2,4 e 4,8 g i.a. ha-1); clorimuron-ethyl (20 e 40 g i.a. ha-1); imazetaphyr (350 e 700 g i.a. ha-1); diclosulam (16,8 e 33,6 g i.a. ha-1); saflufenacil (35 e 70 g i.a. ha-1) e flumioxazin (50 e 100 g i.a. ha-1), além da testemunha.

Texto completoMost human coding genes generate alternative transcripts (isoforms) through alternative splicing (AS) and alternative polyadenylation (APA), most often within the coding region and the 3’ untranslated region (3’UTR), respectively. Both AS and 3’UTR-APA regulations have been increasingly involved in oncogenesis. In particular, AS networks connecting oncogenic splicing factors and oncogenic splicing variants have been recently identified. AS is also widely regulated by genotoxic anticancer drugs, like doxorubicin and cisplatin that induce different types of DNA lesions and are widely used in breast cancer and non-small-cell lung cancer (NSCLC) therapy, respectively. Given the frequent occurrence of resistance to chemotherapy, understanding the underlying mechanisms is crucial to overcome this major issue. There are examples of AS events associated with anticancer drug resistance, but very little is known about the splicing factors and therefore the AS networks involved. In addition, a previous study showed that doxorubicin represses a large set of alternative last exons (ALE) corresponding to the use of intronic polyadenylation (IPA) sites. ALEs have an emerging role in cancer, but little is known about its regulation by other anticancer drugs, like cisplatin. In order to better understand the role of AS and APA regulation in cell response and resistance to chemotherapy, my PhD project had two main aims: 1) determine the extent, regulatory networks and function of AS regulation in breast cancer cell resistance to doxorubicin, and 2) determine the extent, mechanism and impact of ALE regulation in response to cisplatin in NSCLC cells. In the first part, I identified by RNA-seq thousands of AS events and dozens of splicing factors regulated in a cell model of acquired resistance to doxorubicin in ER+ breast cancer. Through an siRNA miniscreen, I found two splicing factors, ZRANB2 and SYF2, involved in doxorubicin resistance. Further RNA-seq analyses revealed the AS events regulated by depletion of these poorly characterized splicing factors, and their convergence on the alternative exon 5 of the oncogene ECT2. Depletion of ZRANB2, SYF2 and the ECT2-Ex5 variant reduces doxorubicin-induced S phase arrest and doxorubicin resistance. In addition, high inclusion levels of ECT2-Ex5 correlate with poor survival specifically in ER+ breast cancer treated with chemotherapy. In the second part, I found by 3’-seq that in NSCLC cell treatment with cisplatin (but not oxaliplatin) induces ALE/IPA in thousands of genes enriched in cell cycle and cell death. This effect is linked to an inhibition of transcription elongation processivity in long genes. 3’-seq analysis on polysomes showed that this ALE regulation impacts the translatome, and revealed a set of particularly short isoforms that were inefficiently translated, including a transcript with a non-coding function. In conclusion, during my thesis, I could identify a novel AS network involved in doxorubicin resistance in ER+ breast cancer, and widespread ALE regulation impacting the translatome in lung cancer cisplatin response. This work increases our understanding of AS and IPA role in cell response and resistance to anti-cancer chemotherapy. In the longer term, the identified alternative transcripts and regulators constitute candidate biomarkers of chemoresistance

Texto completoThis essay is written within my education for Technical Officer in the Swedish armed forces. The essay describes the alternative fuels that the Swedish armed forces can make use of if the supply of fossil fuels is insecure. The alternative fuels that are described and compared are; Synthetic diesel, Organic diesel, Methanol and Ethanol. The essay describes the process of producing the alternatives, the need for modification and the economics for each alternative. The alternatives are compared and assessed for their ability to secure the supply of fuel to the Swedish Armed Forces. The result of the study is that, while synthetic diesel has the best chemical constitution but is not produced in Sweden, the best alternative fuel for the Swedish armed forces at this moment is organic diesel.

Texto completoAlternative economies are commonly depicted as a product of the will of individuals or groups, or as a spontaneous and cumulative reaction to an impact, be it crisis or neoliberalism more generally. Their fate is to transform the world, either gradually or through the clash of models. On the other hand, critics usually see them as a product of neoliberalism, or even capitalism. They are condemned thus to co-optation and marginality, or they just embody neoliberal forms, practices, and subjectivities. In this thesis, I chart an alternative explanation for why and how alternative economies emerge and develop, as well as provide a different lens through which to understand their transformative potential. I investigate these questions by looking at alternative food economies in the post-2008 economic crisis. In order to gain a deep comprehension of real-life events embedded in context, I base my research on two case-studies: the case of new agroecological ‘peasants’ in the Basque Country (Spain), and that of ‘no-middlemen’ solidarity food distributions in Greece. Drawing on fieldwork research, on analytical tools derived from political ecology and food sovereignty literatures, and on Bensaïd’s and Gramsci’s insights on radical politics, this thesis deals with important conceptual and practical questions regarding resistance to neoliberalism, emancipatory strategies, and political agency. My main argument is that alternative food economies can be an integral part of activist strategies engaged in struggles over hegemony, which seek to produce critical and active subjects and, ultimately, move the subaltern to a position of leadership. In the Basque Country, denaturalizing hegemonic ideas and practices regarding agribusiness, and normalizing peasant alternatives, is a key focus of small farmers’ strategy of building alliances and a large social movement fighting for food sovereignty. In Greece, tackling famers’ difficulties and food insecurity through ‘solidarity’ is a strategic step towards advancing counter-austerity ideas and practices to engage people in ‘practical-critical’ activity. Whereas alternative food economies may provide opportunities to politicize politics, create spaces of politicization and self-organization of the subaltern, and generate learning processes on how society-nature relations can be organized differently, they also face challenges, as they are not outside (because there is no outside to) capitalism. The difficulties faced by agroecological producers call us to pay more attention to the relation between working-time and free-time for politics in alternative models. Efforts to develop alternatives must focus on providing the subaltern with the material and subjective conditions that enable them to become ‘agents of their own history’. A politics that tackles social reproduction needs and builds a ‘politics of hope’ is therefore relevant. Indeed, environmental struggles may involve broader social and political goals, beyond concerns over access to resources and the environment or securing livelihoods; this shows the productive relationship between diverse struggles.

This master s dissertation aims to investigate how alternative sanctions are enforce in Brazil in order to identify if this practice constitutes a model for preventing criminal recurrence and protecting the fundamental rights of persons who have been sanctioned due to low and medium gravity crimes. The research starts by analyzing the State punitive power through the point of view of the Democratic State under the Rule of Law and the penalty principles in the Brazilian Constitution, situating the alternative sanctions and measures within the minimalist approach of Criminal Law. Subsequently it analyzes the public policies which approach the public security problem in a broader way, focusing mainly in the criminality prevention, and also points out the pendular character of Brazilian criminal law, which moves between greater and lesser strictness. Further on, it highlights the failure of imprisonment, and the alternative sanctions are presented as criminal sanctions with an educational and reintegrating character. From the treatment provided to alternative sanctions and measures in the United Nations Standard Minimum Rules for Non-custodial Measures (The Tokyo Rules), the research emphasizes the alternative sanctions and measures in the Brazilian legislation and analyzes such alternatives as an interaction process between the State, the community and individuals. Furthermore, it stresses the course taken by the Brazilian experience in the implementation of public policies aimed at the enforcement of alternative sanctions, highlighting the Alternative Sanctions and Measures Incentive Program from the Brazilian Ministry of Justice (Programa de Fomento às Penas e Medidas Alternativas do Ministério da Justiça). The sustainability of the support policy to alternative sanctions and measures in the states, taking into account experiences in several ones, is also analyzed, as well as the proposal for a public security policy with citizenship in the enforcement of alternative sanctions and measures; last, but not least, this research also evidences the possibility of structuring an autonomous alternative criminal justice system in the country.
Srivastava, Diwas. "Modulation of hippo pathway by alternative splicing". Thesis, Montpellier, 2019. http://www.theses.fr/2019MONTT015/document.
Texto completoThe Hippo pathway is a conserved pathway involved in tissue growth and tumor suppression. Studies have demonstrated its significance in the development of human cancers. This cascade controls the activity of the transcription co-activator Yorkie (Yki) in flies and Yes-associated protein (YAP) in mammals. Due to Alternative Splicing (AS), both Yki and YAP proteins exist as two isoforms containing one (Yki1/YAP1) or two (Yki2/YAP2) WW domains. Since WW domains are essential for interaction with specific partners, the alternative inclusion of this domain in Yki/YAP protein may remodel their interaction network and therefore their activity. The regulation and functional consequences of AS of yki/YAP in vivo are unknown. In this Ph.D. project, we identified that depletion of splicing factor B52 in Drosophila lowers inclusion of the alternative exon in yki mRNAs and favors the expression of Yki1 isoform at the expense of the Yki2 isoform. B52 depletion in the wing reduces growth and Yki activity. We demonstrate that Yki1 isoform is an attenuated version of Yki protein that can compete with Yki2 isoform in the nucleus. To ascertain the role of yki AS in vivo and the importance of short isoform Yki1, we abrogated this splicing by using CRISPR/Cas9 technology and created flies that can express Yki2 isoform only. yki2only flies are viable but display a random phenotype of asymmetric wing size. This rise in “fluctuating asymmetry” that is the consequence of subtle deviation from normal development, suggests that AS of yki is crucial for the development robustness. Taking together, these results highlight a new layer of modulation of Hippo pathway via AS of yki.Alternative inclusion of the second WW domain is a conserved feature between Yki and YAP. This further supports the idea that Yki1 and YAP1 isoforms have an important function in vivo and that AS of yki/YAP is a conserved mechanism of control of the Hippo pathway. This study opens up new perspectives for modulation of the Hippo pathway in cancer cells by altering YAP AS

Was used 32 castrated piglets of commercial lineage, weaned at 21 days old and approximately 6,0 kg live weight, with the objective of evaluating the effect of the inclusion of different levels of dried cassava root meal (0, 12, 24 and 36% of DCRM)and feed forms on the performance of piglets in the initial phase (21 to 70 days) and its residual effects on the phases of growth, termination, total period and the 90 kg slaughtering age. The experimental design used were randomized blocks in a factorial scheme 4 x 2 (four levels of inclusion of DCRM and two feed forms dried and wet diets) with four replications by treatment, being the experimental unit one animal. The criteria used for the block formation was the initial piglets weight. The daily average weight gain (DMWG), daily average fees intake (DMFI) and the feeds conversion (FC) weren’t affect significantly (P> 0,05) by inclusion of different levels of DCRM of dry or wet diets. It was concluded that independent of the feed forms of the integral cassava scrap can be used in rations for piglets in the day-care center phase until the level of 36% without compromising the zootecnic performance between the different phases and the 90 kg slaughtering age of live weight.

Texto completoMEMS test and calibration are challenging issues due to the multi-domain nature of MEMS devices. They therefore require the application of physical stimuli, using expensive test equipments, to test and to calibrate their specifications. The main objective of this thesis is to develop alternative electrical-only test and calibration procedures for MEMS convective accelerometers.First, a behavioral model that includes the influence of sensor geometrical parameters on sensitivity is developed and validated with respect to FEM simulations. Second, the model is used to perform fault simulations and to identify correlation that may exist between device sensitivity to acceleration and some electrical parameters. Third, this correlation is exploited to develop alternative test and calibration methods where the sensitivity is estimated using only electrical measurements and without applying any physical stimulus (acceleration). The efficiency of these methods is demonstrated. Finally, two architectures that allow on-chip test and calibration are proposed

Abstract: This research consists of an integrative review of national and international literature about metaphysical self-healing, published between the years 2001 to 2011. The methodology is grounded on the phases proposed by Ganong (1987): selection of hypotheses, establishing criteria for sample selection, presentation of the characteristics of the primary research, data analysis, interpretation and presentation of the results of the review. The online search on scientific work on these databases: Lilacs, Medline, Web of Science, Pubmed, Scopus, using the keywords: "Self-healing", "Self-Healing and Complementary Therapies" and "Self- Healing and Integrative Therapies". Full articles were included, in Portuguese, English or Spanish, and incomplete articles, which were repeated between databases and were not relevant to the study object, resulting in a sample of 89 articles. Content analysis of the thematic side proposed by Bardin (2011) was used as a qualitative methodological strategy to organize the data. The characterization of the corpus analysis, presented in nine tables and subjected to statistical analysis, it points to a production majority of international publications (93%), with a language predominance of English (94%), of which 39% are from the United States. The quantitative method was predominantly (66%) compared to qualitative (34%) with 69.5% and 30.5% prospective cross. The number of publications grew by 2008 (18%), but since 2009, there is a decrease of more than half on numbers of publications (8%), mainly qualitative methodology presented no studies in the past two years (2010 and 2011). Qualitative analysis unveiled three categories: "characteristics of users of practices governed by the principles of self-healing", "the relationship between health professionals... Texto completoAlternative splicing is a process by which the exons of the primary gene transcript are linked in different ways during RNA processing resulting in distinctive proteins. It is an important mechanism in eukaryotic gene expression that enhances proteome diversity and, therefore, the coding capacity of the genome. Different mechanisms seem affect alternative splicing regulation, including metabolic stresses. In the present study, a detailed informatics analysis of ORESTES sequences from head and neck tissues was performed. This in silico analysis revealed that gain of exon sequences is more frequent than exon skipping and GT/AG rule is predominant in splice sites. We observed that alternative splicing usually does not alter the reading frame but may disrupt a protein domain and remove or add new phosphorylation and glycosylation sites. Repetitive and potential regulator elements were frequent in the alternative sequences or in their neighbors. The expression of putative splicing isoforms was investigated in different tissues, including upon stress conditions. We validated approximately 50 new splicing events in normal and tumor cells. Several variants, such as those from HNRNPK, ACTN1, BAT3, CEP192, MPV17, PDK1, PRKAR1A, RAG1AP1 and TRIP6 genes showed distinctive expression pattern in different cell types, in normal and cancer samples from head and neck carcinoma patients and, in some cases, in different tumor stages. We also validated a new transcript of RIPK2 gene, which codes a serine/threonine kinase that activates the NF-kB pathway, and observed a shift in the expression of this variant as a response to temperature stress in vitro. It is currently not clear whether alternative splicing is the cause or the consequence of the neoplastic process. Our data add new information to this topic and provide some examples on the importance of RNA processing in gene expression regulation, both in normal and disease conditions.

Texto completoAccording to Rooth'' alternative semantics (Rooth, 1985, 1992), focus evokes a set of alternatives and that focus particles make reference to this set. The present thesis examines the impact of focus particles and intonational focus on the mental representation of alternatives. In particular, it explores the linguistic and cognitive mechanisms that underlie the establishment of alternative sets in a series of psycholinguistic experiments. Two delayed recall experiments (Exp. 1a and 1b) revealed that participants were better at recalling mentioned alternatives to a focused element when the discourse contained the particles only or even. Experiments 2 and 3 found interference effects of focus particles in the recognition of mentioned alternatives and the rejection of unmentioned alternatives. An additional analysis and Experiment 4 indicate that the set of alternatives consists of various possible replacements of the focused element. Experiment 5 showed that contrastive accents facilitated the retrieval of alternatives while focus particles caused interference effects relative to the condition with an L+H* accent. Overall, this thesis indicates that intonational focus activates alternatives in the listeners mind and helps identifying relevant alternatives. Focus particles, on the other hand, cause stronger competition between the focused element and its alternatives.

This study sought to understand present manifestations of cultural publications which pose an alternative to cultural journalism made by local traditional vehicles, so as to understand whether these publications produce a discourse on culture and everyday life different from the one produced by large media companies. To accomplish this, we chose as corpus four issues of Fome de Quê? (fmq?) magazine and seven issues of Cenário Cultural magazine, the main publications of this genre that circulate in the state of Paraíba showing the local culture and which are not connected to large communication companies. We analyze the characteristics of these two publications in order to understand which alternatives to the journalism model practiced by conventional vehicles are presented in these magazines. We focus our analysis on the way these magazines are organized as a communication vehicle and on the selection and approach of the guidelines, i. e., we observe how their organization modes, subjects, languages, agendas and goals, among other elements, combine in the creation of their discourse on the culture and the daily life they represent. The research approach was Everyday Life Sociology and the method of procedure was the formism. By highlighting the social forms, everyday life studies give us resources to analyze the parts, the alternative forms, in a context that does not favor the definition of the whole, in our case, the framing of the magazines as completely alternatives.

Successful attainment of a high school diploma is essential in today's society. For some students, however, internal and external pressures present significant barriers to school completion. Disengagement from school by these at-risk students is not only detrimental to the individual students, but to the school community and society as a whole. For some students, Alternative Education placements may be a way to reengage them and aid them in school completion. This case study examined the Park Run School District, a pseudonym for the school district, to discover the perceptions of Alternative Education for Disruptive Youth (AEDY) placements by school officials responsible for making the decision to place students as well as other key educators who work directly with at-risk youth. Interviews were used to seek to determine the needs of at-risk students, why administrators choose to refer (or not to refer) students to placement, and if current programming was sufficient. The findings were categorized into three themes, people, placement, and practice. The first theme, people, looked at the concept of risk, the reasons students drop out of school, and they role key individuals play in the lives of at-risk youth. The second theme, placement, looked at the non-traditional nature of alternative education settings, the referral process itself, and the factors administrators consider prior to student outplacement. Finally, the third theme, practice, focused on the nexus of the first two, looking at the most common reasons for referral, the perceived inhibitors to student placement, and the need for additional settings to meet student need.

This thesis concerns the concepts, structure, and applications of the Alternative Energy Design Toolkit. The toolkit is aimed to provide a widely accessible, easy to use, flexible, yet powerful modeling environment for assisting design associated with renewable energy technologies. Exchanges of ideas and knowledge among the users are also highly encouraged and facilitated. The toolkit is composed of three important components: a collection of models that are fundamentals of renewable energy design, a modeling environment called DOME, which is used as the toolkit's enabler, and a supporting Web site. At the beginning, a comprehensive survey of existing tools for renewable energy design is presented. Then, the detailed descriptions and key capabilities of the toolkit's components are provided. In addition, a collection of solar energy models, which is the initial set of models in the toolkit, is also presented. The toolkit is utilized in two design scenarios: a design of a stand-alone PV system, and a trade-off analysis of a hybrid PV-diesel electricity system. In both design scenarios, the models in the toolkit are proven to be useful and convenient resources. The processes of making the representations of the systems are straight-forward, and the analysis mechanisms that the toolkit provides make the design process simple yet effective.
