Texto completoThe gray mold caused by Botrytis cinerea has a devastating impact on various economically important crops, including grapevine (Vitis vinifera L.), with annual economic losses exceeding10 to 100 billion dollars worldwide. Currently, pesticides remain the main method used to reduce the incidence of this phytopathogenic fungus. However, in addition to emergence of multidrug resistance, chemicals must be increasingly restricted in order to limit their impact on the environment and human health. Thus, in recent years, biological protection is gaining renewed interest. Therefore, the aim of our project is the development of new biotechnologies allowing the grapevine to better resist pathogenic pressures, through the use of beneficial microorganisms. The rhizosphere is a rich source of microorganisms with strong abilities in the biocontrol of plant diseases. In the present study, isolation of plant beneficial microorganisms was carried out on healthy plants. A total of 42 micro-organisms were isolated from different rhizospheric semi-arid soils collected in vineyards of Meknes in Morocco (Latitude 33.75989, Longitude -5.43909). The in vitro antagonism test of the various isolates towards B. cinerea evealed that among all the isolates tested the strains S3, S4, S5 and S6 showed a positive result. These isolates inhibit the growth of B. cinerea. The four strains were identified by the study of biochemical characters and phylogenetic analysis of 16S rRNA gene sequences. The results of the analyzes showed that the bacterial strains retained were related to the following species, of the Bacillus genus: S3: B. velezensis; S4: B. velezensis; and S5: B. halotollerans. Isolate S6 was classified in the genus Enterobacter and identified as E. cloacae. The antagonism test carried out in planta on vine vitroplants indicates that the four rhizobacteria reduce significantly (59%, 39%, 55%, and 17%, respectively), the symptoms of the disease and reduce damage to photosynthetic activity (PSII) due to attack by B. cinerea. This study revealed that strains of the genus Bacillus and Enterobacter isolated from the rhizosphere of the vine could be used as biological control agents in the protection of the vine

Texto completoThe research investigates the application of biomimetic calcium phosphate as innovative delivery system for grapevine (Vitis vinifera L.) protection purposes. This smart material was successfully studied in the biomedical field, from the functionalization of biomimetic calcium phosphate with anti-cancer molecules for localized releases, to the development of an innovative toothpaste for oral hygiene. Preliminary assays to implement the control of the grapevine fungal diseases, have revealed promising results. In this framework, the biomimetic inorganic hydroxyapatite was investigated as potential delivery system of bioactive substances allowed in organic agriculture for plant protection.Through a multidisciplinary approach, the study was aimed to evaluate the efficiency of hydroxyapatite in enhancing the biological activity of copper(II) compounds, on the control of relevant common diseases, like downy mildew, and complex fungal diseases, such as the grapevine trunk diseases. This aim is related to further ambitious goals: the significant reduction of the fungicides amounts applied in plant protection and the optimization of the distribution and persistence of the bioactive substances in the plant tissues, including the vascular ones, where harmful pathogens can develop. Overall, the experimental activities allowed: (i) to understand the interaction between delivery system, functional substance and grapevine tissues; (ii) to demonstrate the mechanism on which the higher efficacy of the functional substance is based; (iii) to collect new information on the mechanisms involved in the symptoms expression by studying the plant defense reactions induced by the treatments

Texto completoRecombinant antibodies isolated from non-human primates represent very promising medical countermeasures against bioweapons. The advantages and methodological aspects of this approach have been described. An scFv, neutralizing the lethal toxin of Bacillus anthracis (2LF: KD= 1. 02 nM) and directed against the “lethal factor” subunit was isolated from a phage-displayed immune library, and characterised. Another scFv, 43RCA, neutralizing ricin, was obtained with the same methodology and tested (in particular: KD= 40 pM). This approach may also be used beyond bioweapons, as an scFv directed toward Aspergillus fumigatus was also isolated (MS130i-IIIC3: KD= 0,96 nM). On-line sequence analysis with IMGT tools allowed to show the high degree of similarity between these scFvs and their human counterparts. Antibody fragments were engineered, including an in vitro affinity maturation (KD initial = 3. 4 nM; KD final = 0. 18 nM). Utilizing IMGT standardisation and on-line tools, a “germline humanization” - utilizing FR derived from IgM, encountered by every Human, as opposed to IgG FR - was realized in order to ensure an optimal tolerance for one of our scFvs. A primatized IgG was tested in vivo and showed therapeutic and prophylactic capacities

Texto completoCold storage is used to obtain large quantities of natural enemies in biological control programs for inundative releases. The objective of this thesis is to compare different storage treatments, considered as thermal stresses, to evaluate the fitness of the survivors, using as a biological model the parasitoid Aphidius ervi. We evaluated the effects of each tretament on different life history traits such as longevity, fecundity at emergence, lifetime fecundity, mating capacity, sex ratio of offspring, but also on physiological parameters such as the amount of lipids. The main results showed that fluctuating temperature regimes significantly improve survival rates (mainly after four weeks of storage) compared to constant regimes, but parasitoids consume larger amounts of lipids during storage. During long storage (4 weeks), we highlighted for the first time that a combination of fluctuating regime with low temperature at 0°C produced the best emergence rates associated with higher fitness. When mortality rate is negligible after storage, longevity was the first parameter affected. Fecundity did not decreased significantly, whereas the sex ratio of the progeny was male biased. When mortality rate is high, the individuals succeeding to emerge, showed performances as high as the control. These individuals, however, were small in size compared to control. After cold storage, larger individuals were more affected by stress than smaller ones. Furthermore, smaller individuals had a higher egg load at emergence, which was observed for the first time. In parasitoids, it is well known that larger individuals have better fitness, and this may thereby affect the success of biological control

Texto completoTo combat pests of crops (herbivores and pathogens) new pathways have been explored, in particular biological control. Used the nature and its organisms to regulate the populations of pathogens, that is the goal of the biological fight also called biocontrol. Among the organisms commonly found in biocontrol are micro-organisms, be they fungi or bacteria. These microorganisms have very interesting capabilities for farmers and agronomists. Indeed, studies have shown that they are able to interact with plants to allow a better growth and health of it.Then, in this thesis, we were interested in bacteria to find alternative methods of struggle with Fusarium graminearum, a pathogen of cereals responsible for high yield losses, especially thanks to the production of mycotoxins that will make the seeds unfit for human and animal consumption.Through an original approach of keeping noninhibitory strains in vitro for in planta tests, we have been able to show the limitations of in vitro selection. Thanks to the metabolic exploration of a particularly efficient strain, we have also been able to highlight potential new antifungal molecules. Through a model strain, we explored the mechanisms of the establishment of systemic resistance in wheat induced by bacteria. And finally, we have explored the potential of bacterial combinations in wheat protection that seems to represent a real future in the world of biocontrol.The work done is in line with the needs of new resources to limit the use of pesticides, but also in a need to better understand the tripartite interactions between wheat, pathogen and beneficial bacteria

Texto completoInbreeding and hybridization are two genetic processes that may have antagonistic effects in biological control agents (BCAs) rearing. Hybridization can minimize the risk of inbreeding depression (decrease in fitness of inbred individuals), and favor the emergence of advantageous phenotypes. Although the mechanisms of inbreeding depression and the consequences of hybridization are well known, very few studies have been carried out in the context of biological control. The actual effects of inbreeding and hybridization on the fitness of BCAs should be assessed to guide the practices of the biocontrol industry. In a public-private collaboration, data were generated on the effects of inbreeding in the four BCAs Allotropa burrelli, Chrysoperla near comanche, Cryptolaemus montrouzieri and Macrolophus pygmaeus. Inbreeding resulted in a 30% decrease in reproductive success in M. pygmaeus. A thorough study of the consequences of intra-specific hybridization was then carried out using four populations of M. pygmaeus. Three genetic groups separated by strong reproductive isolation were identified. Nevertheless, a fitness advantage for individuals from crosses between genetically distant parents has been highlighted within each genetic group. This work brings data usable in the short term by the partner companies. It also helps to create a corpus of data to better evaluate the real importance of genetic effects in BCAs rearing
