Resumen
ABSTRACT Cell-bound C-signal guides the building of a fruiting body and triggers the differentiation of myxospores. Earlier work has shown that transcription of the csgA gene, which encodes the C-signal, is directed by four genes of the act operon. To see how expression of the genes encoding components of the aggregation and sporulation processes depends on C-signaling, mutants with loss-of-function mutations in each of the act genes were investigated. These mutations were found to have no effect on genes that are normally expressed up to 3 h into development and are C-signal independent. Neither the time of first expression nor the rate of expression increase was changed in actA, actB, actC, or actD mutant strains. Also, there was no effect on A-signal production, which normally starts before 3 h. By contrast, the null act mutants have striking defects in C-signal production. These mutations changed the expression of four gene reporters that are related to aggregation and sporulation and are expressed at 6 h or later in development. The actA and actB null mutations substantially decreased the expression of all these reporters. The other act null mutations caused either premature expression to wild-type levels (actC) or delayed expression (actD), which ultimately rose to wild-type levels. The pattern of effects on these reporters shows how the C-signal differentially regulates the steps that together build a fruiting body and differentiate spores within it.

ABSTRACT Mutations within the −12 and −24 elements provide evidence that the act promoter is recognized by sigma-54 RNA polymerase. Deletion of the −20 base pair, which lies between the two conserved elements of sigma-54 promoters, decreased expression by 90%. In addition, mutation of a potential enhancer sequence, around −120, led to an 80% reduction in act gene expression. actB, the second gene in the act operon, encodes a sigma-54 activator protein that is proposed to be an enhancer-binding protein for the act operon. All act genes, actA to actE, are expressed together and constitute an operon, because an in-frame deletion of actB decreased expression of actA and actE to the same extent. After an initially slow phase of act operon expression, which depends on FruA, there is a rapid phase. The rapid phase is shown to be due to the activation of the operon expression by ActB, which completes a positive feedback loop. That loop appears to be nested within a larger positive loop in which ActB is activated by the C signal via ActA, and the act operon activates transcription of the csgA gene. We propose that, as cells engage in more C signaling, positive feedback raises the number of C-signal molecules per cell and drives the process of fruiting body development forward.

Resumen
Abstract This article studies the discourse in the late eighteenth- and early nineteenth-century French periodical press on the topic of translations of travel writing. It reveals that travel reviews were arenas for discussing the political and ideological value of translating travelogues into French, notably from English. In the context of the Franco-British conflicts at the turn of the century, the French press perceived translations of British travel writing as potential patriotic tools that allowed different ways of countering or subverting British global influence. Paratextual elements of translations, the translator’s prefaces and notes, appeared to be particularly important in this respect. By analysing the periodical discourse on travel book translations, the article shows how travel writing was constructed as a politically invested genre.

Abstract Alternative Complex III (ACIII) is an example of the robustness and flexibility of prokaryotic respiratory chains. It performs quinol:cytochrome c oxidoreductase activity, being functionally equivalent to the bc1 complex but structurally unrelated. In this work we further explored ACIII investigating the role of its monoheme cytochrome c subunit (ActE). We expressed and characterized the individually isolated ActE, which allowed us to suggest that ActE is a lipoprotein and to show its function as a direct electron donor to the caa3 oxygen reductase.

Resumen
Abstract Nephroblastomas or Wilms tumors are the most common pediatric renal cancer. Across all cases, the 5-year overall survival is over 90%. This is largely in part due to brute-force, chemotherapy regimens such as dactinomycin (actD) that have been routinely used to treat Wilms tumors. However, those that do respond to treatment still face a 15% chance of relapse. Patients with anaplastic tumor histology require more toxic therapies, and yet they still face a worse prognosis of only 66% 4-year survival and relapse rate of 43%. Furthermore, recurrence cuts survival rate down to 50%. With the widespread use of actD in Wilms tumors and other childhood malignancies, understanding the mechanism to recalcitrance against this chemotherapy can potentially reveal important vulnerabilities that can be exploited in the clinic. While transcriptomic and genomic analyses have been done in Wilms tumors, no targetable pathways have been identified in actD chemotherapy-resistant and anaplastic tumor cases. To that end, we performed ribosome sequencing of actD-treated anaplastic Wilms tumor cell line WiT49 and uncovered that the proteasome pathway is preferentially translated. Moreover, loss of proteasome subunits in a CRISPR library dropout screen sensitized the cells to actD. Consistent with these findings we found upregulation of proteasome subunit levels and proteasome enzymatic activity in Wilms tumor lines following actD treatment. To interrogate the effect of cotreating bortezomib with dactinomycin in vivo, we subcutaneously implanted anaplastic Wilms tumor patient-derived xenograft (PDX) lines in NOD/SCID Gamma (NSG) mice. Combinatorial treatment was found to be retard tumor growth compared to vehicle and single drug conditions. With further investigation, our findings can provide a foundation for exploring proteasome inhibitors with dactinomycin in clinical trials. Our study has important implications to improving the outcomes for not just for recurrent and anaplastic Wilms tumors, but to other pediatric cancers that use actD as well.

According to Thomas of Aquinas, form is that which instantiates substance in a determinate nature; without it, substance would not be what it is. It should be emphasized that defining what something is (its quid est) is not not the same as to assert that it is, for in this case what is asserted is its existence, not its nature. That indicates that existence is not given by the form of the substance, but by something else, which in his Exposition to the Boethius\'s Ebdomadibus, Thomas calls act of being (actus essendi). Mingling in the substance its subject , the act of being gives it its existence, turning it into an entity, which is vinculated to being itself (ipsum esse), or God. The Aquinate calls this nexus participation, whence the entity participates in being as the effect of its cause. The problem is set when one traverses the ontological level to the gnoseological, that is, when the intellect tries to grasp the participation of the entity in being. In this intellection, the limits of the human intellect become clear: the participation of entity in being is understood not as such, but as the concrete in relation to the abstract. The entity is signified in concrete, for in it the act of being is found concretized; the difficulty, however, reveals itself when the intellect tries to abstract the act of being from the entity: in this abstraction, the act of being is not grasped as such, but as abstract. Thus it becomes inevitable to formulate the following questions: which is the mode of abstraction which tries to obtain the act of being of the entity? Why is the intellect not able to grasp being as being, but only as abstract? Is the act of being, inconceivable to the human intellect, in fact superior to the substantial form? And does participation of the concrete in the abstract correspond to that of the entity in being? These are the main questions that this study aims to answer.

Resumen
This thesis examines the transmission of the idea of crusade to Poland and the subsequent Polish response to the idea of crusade in the twelfth century. It determines the key mechanisms by which the idea of crusade was transmitted to Poland and through an analysis of primary contemporary sources the thesis provides compelling evidence that the Polish elites were highly familiar with, and receptive to, the idea of crusade. Moreover, this thesis argues that the Poles were conscious participants in the crusades and undertook various crusading activities during the twelfth century. These findings are contrary to the established position in Polish historiography, and bring many sources to an English-speaking audience for the first time.
Through an examination of narrative sources this thesis demonstrates that the Polish elites were enthusiastic supporters of the idea of crusade within a decade of the First Crusade. This thesis shows that under the leadership of the Piast dynasty the Poles engaged in Christian holy war (proto-crusading): the conquest of Pomerania (1102–1128), contributed armies to three theatres of the Second Crusade (1147–1148), and launched major expeditions against the Prussian apostates (1166 and 1192).
This thesis identifies the influence of the idea of crusade on the actions of the Piasts and in particular, Boleslaw III’s sons (the Piast Juniors), and the importance of family traditions. The thesis presents new propositions in regards to the crusades in Prussia and key Polish crusader participants: the involvement of the Polish crusader army under the command of Mieszko III in the Wendish Crusade (July–August 1147); previously unreported in English historiography, the first known example of crusading in Prussia led by Boleslaw IV (November–December 1147); and Henry of Sandomierz’s leadership of the Polish crusader contingent to the Holy Land (1147–1148).

General Measure Abstract This diploma thesis is focused on general measure as one of the activities of public administration in Czech Republic. The thesis is systematically divided into four consecutive parts. First part is devoted to theroetical treatsie on the various forms of admnistrative acts. This part emphasizes the distinction between abstract administrative acts, individual administrative acts and mixed administrative acts, which are divided into two groups - specifically abstract and abstractly specific. Second part is focused on generals measures in general and its enshrining in czech legal order. This part includes definiton of general measure, its regulation in foreign law, wich is one of the inspirations for czech regulation. Second part of the thesis also includes chapters devoted to materiál and formal aspects of general measures as well as chapter about some particular general measuers. Following part is about creation of a general measure. This is a procedure that is generally regulated by the sixth part of the Administrative procedure code and applies whenever a special law does not says otherwise. This section is divided into five chapters, where each of the last four chapters deals with a different phase of the process. Last part is devoted possibilities of reviewing general measures....

The topic of this master's thesis is the legislation of local self-governing units from the moment of its creation to its possible repeal by the constitutional Court. The thesis is divided into eight chapters which are divided into subchapters and further. The first chapter describes basic concepts which are mentioned in this thesis and are related to the topic. This is e.g. explanation of the concept of an abstract act and examples of what abstract acts exist in our country, the definition and division of public administration, explanation of what self-govern is and how it is divided, what are generally binding ordinances and regulations and what is the difference between the two. More basic concepts which are important for the comprehension of this master's thesis are explained. The following two chapters talk about the process of creation of generally binding ordinances of Municipalities and Regions and their regulations too. There are described requirements and structures generally binding ordinances and regulations of Municipalities and Regions, their receiving authority, who can the legislation suppose, its approval, force and effect. The fourth chapter focuses on the effectiveness of generally binding ordinances. It shows what areas municipalities and regions can issue generally binding...

Resumen
Intellectual integrity and a challenge to rhetoric are the two strategic objectives of those who take up the hazardous path of sociological knowledge. This book does not presume to respond fully, but at least attempts to target these aims. The fruit of many years' teaching and research experience, it adopts a line of interpretation that highlights the point of view of the social agent considered in his close, symbiotic and procedural relation with the society in which he acts; this society is not abstract and generic but explored and construed in the tangible dimension of daily life and social relations. The book is organised with a practically identical layout in all the chapters: in dialogue format it proceeds from the identification of the categories central to the issue addressed through to its empirical application/s, hinging the two together with contributions from the sociological school or writer most relevant to the subject in question.

Resumen
Abstract The chapter discusses the five-act structure to longer forms of scientific presentations. The flow of the five-act structure moves from the current state of knowledge, through conflicting evidence, the formation of a hypothesis, to data collection and conclusions. The outline of the structure looks like this: Act 1: current state of knowledge; Act 2: conflicting evidence; new hypothesis; Act 3: first presentation of results; Act 4: problems and their solution; and Act 5: conclusions. To make this form more interesting, it can introduce at least one BUT in each of Acts 3 and 4. Act 2 already has a BUT in the form of conflicting evidence. To hold the audience's interest it is best to introduce this same tension into the other acts. Without it the acts can easily become a list of facts (ANDs). Each BUT serves to wake up the audience. The BUTs tell them that something important is about to happen. The revised structure looks like this: Act 1: current state of knowledge; Act 2: conflicting evidence (BUT), new hypothesis; Act 3: results, BUT, results; Act 4: BUT, results, BUT results; and Act 5: conclusions.

Resumen
Abstract This essay is concerned with a particular form of undertaking which appears to break every principle of law governing the formation of contracts. I refer to the abstract payment undertaking, a money promise which is independent of the transaction that gives it birth and which is considered binding when received by the beneficiary (or sometimes even when issued by the promisor) without acceptance, consideration, reliance, or execution in solemn form. The type of contract (if it be such) generated by an abstract payment undertaking fits neither the bilateral nor the unilateral model. It is not bilateral, for no promise is given in exchange; nor is it unilateral in the ordinary sense, for its irrevocability does not depend on the performance, or even the commencement of performance, of a designated act or forbearance

Resumen
Abstract Some right acts have what philosophers call moral worth. A morally worthy act manifests the agent’s virtuous motives such that they deserve credit for having acted rightly. And, for an agent to deserve credit for having acted rightly, not only must their act be right but their motives must be such that their acting rightly was no mere accident. More precisely, then, a right act has moral worth if and only if it was motivated in a way that makes it non-accidentally right. This chapter argues that a right act was motivated in a way that makes it non-accidentally right if and only if it issued from an appropriate set of concerns, where the appropriateness of these concerns is a function of what her ultimate moral concerns should be. Two important upshots of the resulting account of moral worth are that (1) an act can have moral worth even if it doesn’t manifest a concern for doing what’s right and that (2) an act can lack moral worth even if it is performed for the right reasons.

Resumen
Review question / Objective: P: Patients with AML or MDS after allo-SCT; I: Hypomethylating agents after allo-SCT; C: Observation after allo-SCT; O: Overall survival rates. Condition being studied: Hypomethylating agents (HMAs) seem to have a range of properties favorable to post-allogeneic hematopoietic stem cell transplantation (allo-SCT) maintenance in acute myeloid leukemia (AML) patients. This meta-analysis was performed to review all relevant studies to compare the outcomes of patients undergoing allo-SCT for AML or MDS receiving HMA maintenance therapy with observation only. Information sources: The systematic search of the Embase and MEDLINE databases identified 4,416 articles, from which 512 duplicates were removed. This resulted in 3,904 articles available for title and abstract review. Subsequently, 3,875 articles were excluded as the article type and study design did not fulfill the inclusion criteria, or there was no report on a primary outcome of interest. The remaining 29 articles underwent full-length review and 18 of those were excluded for the aforementioned reasons. Ultimately, the eligibility criteria for our meta-analysis were met by 11 studies: 2 RCTs, 1 prospective cohort study, and 8 retrospective cohort studies.
