Texto completoThe exposure of school communities to new information and communication technologies has positive impact on technological appropriation. Our objective was to analyze the use of computer laboratories in three public schools in Diamantina. We have used the evolutive stages of the ACOT Project (Apple Classroom of Tomorrow) as parameters. The theoretical reference works from five evolutive stages: (1) exposure; (2) adoption; (3) adaptation; (4) appropriation and (5) innovation. These categories have derived from observations recorded during the 10-year period of the Apple Project. Each stage shows behaviors and attitudes which have gradually incorporated the new technologies. The source of data consists of (a) interviews with teachers, students, principals and mothers, and (b) field observations recorded between 2004 and 2005. The analytical procedure has searcherd for meaningful units of discourse. The stages of the ACOT Project may be used as parameters to show the technological appropriation in schools. In the three schools analyzed, there has been a significant appropriation of the use of new technologies in the classroom, thus allowing for innovation in the school environment and change of position in the teacher-student relashionship. This work has been carried out in Programa de Pós-Graduação em Educação (FACED/UFRGS) as part of the research area related to the use of computers in education and is associated with the Núcleo de Educação Digital.

Bamboo mosaic virus (BaMV), belongs to genus Potexvirus of the family Flexiviridae, is a positive-sense RNA virus. Since the virus can not complete its infection cycle by itself, it needs host factors in different stages, such as RNA replication, folding, and spreading. Therefore, studying the interaction between virus and its host is important and useful in developing antivirus strategy. The aim of this study is identifying the host proteins that involve in the infection of BaMV. According to the previous study, many of differentially expressed genes from BaMV infected Nicotiana benthamiana plants were identified using cDNA-amplified fragment length polymorphism (cDNA-AFLP) technique. Then I focus on one of the upregulated cDNA fragments, ACCT8-1. Rapid amplification of cDNA ends (RACE) technique is used to extend the 5’ and 3’-end of the cDNA fragments to 3199 nucleotides in length. The sequence the hypothetic protein derived from the coding region is blasted with the database in National Center for Biotechnology Information. ACCT8-1 is an ortholog of N. tabacum EILP (elicitor inducible LRR protein) gene and Cladosporium fulvum resistance gene (cf-2 and cf-5). The expression level of ACCT8-1 is knocked down by Tobacco rattle virus (TRV)-based silencing system to inspect the coat protein accumulation after virus inoculation. Results indicate that the coat protein accumulation is reduced about 40% on the knockdown plants to that of control plants. The accumulation levels of viral RNA analyzed in Northern blot also show similar reduction in knockdown plants. However, the coat protein accumulation levels of Potato Virus X (PVX) and Cucumber mosaic virus (CMV) show no significant difference in ACCT8-1 knockdown and control plants. Further, I also find that the coat protein accumulation has no significant difference and a 40% reduction in the protoplasts and systemic levels derived from the knockdown plants, respectively, compared to those of control plants. Finally, I also find that the lesion sizes in the knockdown plants are smaller than those observed in control plants. Overall of these results suggest that ACCT8-1 may involve in viral movements.

Bamboo mosaic virus (BaMV) is a single-stranded positive sense RNA virus. To study the infection cycle of BaMV in its assay host, Nicotiana benthamiana, we have used cDNA-AFLP technique to identify the differentially expressed genes after BaMV infection. The first project of my thesis is analyzing one downregulated gene, ACAC3, and two upregulated genes, ACAC9-1 and ACCT6-1, to examine whether these genes are involved in the infection of BaMV in N. benthamiana plants. I have used virus induced gene silencing technique to knock down the expression levels of these three genes, and then inoculated BaMV on the knockdown N. benthamiana leaves. The accumulation levels of virus coat protein are measured by Western blotting analysis. The results showed that the accumulation of BaMV was increased to 113% in ACAC3 knockdown plants at 5 dpi compared to that of the control plants. However, the accumulation levels of BaMV coat protein in ACAC9-1 (135%) and ACCT6-1 (60%) knockdown plants were increased and decreased, respectively, significantly to that of control plants at 5 dpi. The results imply that the two upregulated gene ACAC9-1 and ACCT6-1 may play different roles as restricting and assisting, respectively, in BaMV infection cycle. The second project of my thesis is characterizing an up-regulated gene ACCT8-1 which was studied previously. This gene was involved in the movement of BaMV in N. benthamiana plants. The identity of ACCT8-1 is annotated to an elicitor inducible leucine-rich repeats receptor-like protein of N. tabacum and the tomato homologous of cladosporium fulvum resistance gene Cf-2 (Hcr2). To study the interaction of ACCT8-1 with BaMV in the infection cycle, I generated a deletion construct of ACCT8-1 which removed the leucine-rich repeats domain (ΔLRR). I transiently expressed ACCT8-1/ΔLRR-GFP in BaMV inoculated N. benthamiana. The accumulation of BaMV coat protein was enhanced in ACCT8-1/ΔLRR-GFP expressed N. benthamiana at 5 dpi. The results indicate that the leucine-rich repeats domain is not essential for BaMV movement.
