Resumen
260 Background: Recent placebo-controlled phase III trials of the mTOR inhibitor everolimus and the VEGF/ PDGF receptor inhibitor sunitinib in PNET noted improved progression-free survival (PFS). However, objective response rates (RR) with these agents are <10%. Preclinical studies suggest enhanced anti-tumor effects with combined mTOR and VEGF targeted therapy. Methods: We conducted a phase II trial of the mTOR inhibitor TEM (25 mg IV q week) and the VEGF-A monoclonal antibody BEV (10 mg/kg IV q 2 weeks) in patients (pts) with well or moderately differentiated PNET and progressive disease by RECIST within 7 months of study entry. Co-primary endpoints were RR and 6-month PFS. Planned enrollment is 50 patients, with interim analysis after the first 25 evaluable pts. Pts had no prior mTOR or VEGF targeted agents, ECOG PS 0-1, and adequate hematologic and organ function. Continued octreotide was allowed, but not required. Prior interferon, embolization, and ≤ 2 chemotherapy regimens were allowed. Results: Confirmed PR was documented in 11 of the first 25 (44%) evaluable patients. 20 of 25 (80%) patients were progression-free at 6 months. Both endpoints exceeded pre-defined criteria to continue enrollment. For 35 evaluable patients, the most common grade 3-4 adverse events attributed to therapy were leukopenia (12%), hypertension (12%), hyperglycemia (12%), mucositis (9%), and fatigue (9%). Conclusions: The combination of TEM/BEV has substantial activity in a multi-center phase II trial with RR of 44%, well in excess of single targeted agents in PNET. 6-month PFS was a notable 80% in a population of patients with RECIST criteria progression within 7 months of study entry. Accrual is ongoing. Supported by NCI N01 Contracts: 662205, 62203, 62208, 62209, 62206, 62204, 62207, 62201.

Resumen
67 Background: The HH pathway is overexpressed in gastroesophageal (GE) tumors. Pre-clinically, HH inhibitors have demonstrated a reduction in GE tumor growth, cell motility and invasiveness. V, an oral small-molecule antagonist of the Hh pathway, has previously been safely combined with FOLFOX chemotherapy. Methods: Pts with untreated, metastatic or locally advanced gastric or GEJ adenocarcinoma were randomized 1:1, stratified by institution and disease status (with or w/o distant mets) to FOLFOX (ox 85 mg/m2, LV 200 mg/m2, 5-FU bolus 400 mg/m2, 5-FU infusion 2400 mg/ m2 over 48 hrs) q14d plus V or placebo (P) (150mg PO daily). Cycle defined as 2 weeks and no crossover allowed at progression. FFPET and blood were collected for biomarker analyses. Response assessed every 8 weeks (RECIST 1.1). Primary endpoint was progression-free survival (PFS), secondary objectives were overall survival (OS), response rate (RR), and toxicity. Results: 124 pts (V/P 60/64) enrolled at 20 sites between 10/09-2/12. Pt characteristics (V/P): median age 58/62; ECOG PS 0: 23 (40%) / 30 (47%); male 39 (65%) / 53 (83%); GEJ 37 (62%) / 39 (61%); diffuse histology 20 (43%) / 14 (30%). Median number of FOLFOX cycles 9.5/11. Most common Gr ≥3 toxicities: (% pts V/P) neutropenia 50.0/31.7 (p=0.07), neuropathy 23.1/14.3 (p=0.33), fatigue 15.4/9.5 (p=0.50), thrombosis 13.5/11.1 (p=0.92), anemia 9.6/9.5 (p=0.99), hypokalemia 9.6/4.8 (p=0.52), nausea 7.7/9.5 (p=0.99). Death on or within 30 days of treatment 6.7%/15.6% (p=0.20). Median PFS in ITT population 11.5/9.3 mo (95% CI 8.5-14.4/6.7-11.9; p=0.34) and median OS 12.2/13.9 mo (95% CI 10.2-14.3/11.5-16.3; p=0.48). Non-statistically significant trends toward improved PFS with V were noted in female pts, diffuse histology, and PS 1 (p≤ 0.08). RR (%) 33/27 (p=0.64). Conclusions: Addition of V to FOLFOX did not improve PFS in an unselected advanced GE carcinoma population. Blood and tissue biomarker analyses are ongoing to determine if there is a subset of patients who may derive benefit from V. Resumen
5517 Background: Anti-angiogenic therapy is active in OC; the combination of VEGF and mTOR inhibitors is hypothesized to further improve activity. This report is the OC cohort of a multi-histology phase II study assessing the activity and toxicity of B/T. Methods: Patients (Pts) with recurrent epithelial OC who had received ≤ 2 chemotherapy regimens and no prior treatment with a VEGF or mTOR inhibitor were eligible. A two-stage design was used with second stage accrual if >6 pts had objective responses (OR) or >10 pts of the first 25 remained progression-free (PF) at six months (mo). Pre-defined end-points for a recommendation for further clinical trial evaluation included at least 15/50 with OR or 26/50 PF at six mo. Treatment included T 25 mg IV wkly and B 10 mg/kg IV q14 days on 28 day cycles. Results: 58 pts were enrolled (the first 50 pts are used to determine a final recommendation). Median age=62 (range 35-82). A median of 4 (range 1-23) cycles were administered. 24 were platinum-sensitive, 34 resistant. Off-study reasons included 13 adverse events and disease progression in 38. 3 refused further therapy due to toxicity. 14 of the first 50 pts had partial response (PR) (9 platinum-resistant); 25/50 remained PF (8 PR, 15 SD, 2 non-progressing) at 6 mo. Grade (gr) 3/4 toxicities occurring >2 events include: fatigue (4), stomatitis (7), hypertension (5), neutropenia (4), thrombocytopenia (4), hypokalemia (3). One rectal and one vaginal fistula, and two colonic perforations (one gr 2 and one gr 3 during cycles 3 and 1 respectively) were observed. Episodes of gr 1/2 oral, nasal, pulmonary, vaginal and gastrointestinal hemorrhage were also observed. Conclusions: Although the OR and PFS did not reach pre-defined standards, the numbers of OR and 6 mo PFS suggest potential enhanced activity with a combination of mTOR inhibitor with anti-angiogenic therapy. Other combinations of these targeted agents may result in more satisfactory activity with less toxicity. Resumen
8004 Background: Vorinostat, a HDAC inhibitor, enhances paclitaxel and platinum-mediated anti-cancer activity in preclinical studies by enhanced tubulin acetylation and DNA fragmentation respectively. Promising activity with carboplatin (C), paclitaxel (P), and vorinostat in patients with advanced NSCLC in the phase I study (Ramalingam et al, Clin Cancer Res, 2007) prompted this placebo-controlled, randomized phase II study. Methods: Pts. with stage IIIB (wet) or IV NSCLC, performance status (PS) 0/1, no prior therapy and adequate bone marrow, renal and hepatic function were randomized (2:1) for therapy with PC with either vorinostat or placebo. Treatment consisted of C: AUC=6 mg/ml.min; and P 200 mg/m2 both given on day 3 along with either vorinostat (400 mg PO QD) or placebo on days 1–14 of each 3 wk cycle to a maximum of 6 cycles. The estimated sample size to demonstrate a 50% improvement in response rate for vorinostat over placebo was 93 pts. (one-sided P, type I error 10%). Results: Ninety-four pts. were enrolled (vorinostat-64; placebo-32). Pts. baseline characteristics were similar between the two arms (median age 64, male 60%, PS 0 40%, brain mets 16%). Median # cycles: vorinostat-3.5; placebo - 4. The confirmed response rate was superior with vorinostat over placebo (34% vs. 12.5%, P = 0.02). At the time of analysis, the preliminary median PFS for vorinostat and placebo were 5.75 and 4.1 m respectively (ITT). Follow up for survival is ongoing. Common grade 3/4 toxicities (vorinostat vs. placebo): neutropenia (44% vs. 47%); thrombocytopenia (33% vs. 16%); fatigue (13% vs. 3%); hyponatremia (21% vs. 6%); diarrhea (5% vs. 0). Discontinuation from study after cycle 1 was higher with vorinostat (27% vs. 16%). Biomarker studies on baseline tumor tissue and peripheral blood cells are ongoing. Conclusions: Administration of vorinostat with carboplatin and paclitaxel resulted in a significantly superior response rate for pts.with advanced NSCLC. HDAC inhibition is a novel therapeutic strategy for treatment of NSCLC. Supported by ASCO Career Development Award to S.S.Ramalingam, and NCI NO1-CM-62209, NO1-CM-62201, NO1-CM-62208. Resumen
A novel, strictly aerobic, heterotrophic, gliding, Gram-negative, oxidase-, catalase-, β-galactosidase- and alkaline phosphatase-positive marine bacterium, designated strain KMM 6220T, was isolated from seawater and studied by using a polyphasic taxonomic approach. The DNA G+C content of strain KMM 6220T was 59.9 mol%. The predominant fatty acids were iso-C15 : 1, iso-C15 : 0, iso-C15 : 0 3-OH, iso-C17 : 0 3-OH and C16 : 1 ω7/iso-C15 : 0 2-OH. Phylogenetic analysis based on 16S rRNA gene sequencing revealed that strain KMM 6220T formed a cluster with the misclassified strains [Flexibacter] aggregans NBRC 15974 and [Flexibacter] tractuosus NBRC 16035 and with the type strains of Reichenbachiella agariperforans and Roseivirga ehrenbergii with levels of similarity of 95.9, 94.4, 92.0 and 91.8 %, respectively. On the basis of its phenotypic, chemotaxonomic, genotypic and phylogenetic characteristics, strain KMM 6220T is considered to represent a novel species of a new genus in the phylum Bacteroidetes, for which the name Fulvivirga kasyanovii gen. nov., sp. nov. is proposed. The type strain of the type species is KMM 6220T (=CCTCC AB 206119T=KCTC 12832T).

Pickard, Stephen. "The purpose of stating the faith : an historical and systematic inquiry into the tradition of fundamental articles with special reference to Anglicanism". Thesis, Durham University, 1990. http://etheses.dur.ac.uk/6220/.

Texto completo
Resumen
Stating the faith in the form of fundamental articles has, historically, provided an important strategy by which the identity and continuity of the Church has been expressed. The issue underlying this ecclesiological context of fundamental articles concerns the truth of the one-in-Christ bond in Christianity. However discussion of fundamental articles of the faith has, from the Post-Reformation period, tended to occur as somewhat disconnected from wider concerns to do with the belief, discipleship and mission of the Church. One result is that important issues and motives implicit in the attempt to articulate the fundamental articles of Christianity remain undisclosed and undeveloped. By means of a multi-level approach - contemporary relevance (Part One), historical development (Part Two), case studies (Part Three) and systematic inquiry (Part Four) - this thesis develops an understanding of fundamental articles which shows how the theme is enmeshed within and contributes to the dynamic of Christian faith in the Church. The resources for this inquiry are drawn from an extensive, but hitherto largely unexamined treatment, of the theme of fundamental articles in Anglicanism. The Protestant tradition of speaking about fundamental articles of faith is found to offer an important medium through which the reality of being one-in-Christ can be identified, communicated and strengthened. In this way the tradition proves a valuable means for uncovering and examining the purpose(s) of stating the faith. The problematic role of fundamental articles in Anglican self- understanding reveals itself as an instance of a more general, controversial and unfinished task in theology to state the truth of God’s creating and redeeming love. The thesis thus draws attention to the significance of fundamental articles for expressing the nature and form of ecclesial faith and discipleship. A positive rationale emerges for a more intensive and discerning engagement with the fundamental articles tradition as a strategy by which theology can serve the mission of the Church.

Lawrence, Peter James. "Mesh generation by domain bisection". Thesis, University of Greenwich, 1994. http://gala.gre.ac.uk/6220/.

Texto completo
Resumen
The research reported in this dissertation was undertaken to investigate efficient computational methods of automatically generating three dimensional unstructured tetrahedral meshes. The work on two dimensional triangular unstructured grid generation by Lewis and Robinson [LeR76] is first examined, in which a recursive bisection technique of computational order nlog(n) was implemented. This technique is then extended to incorporate new methods of geometry input and the automatic handling of multiconnected regions. The method of two dimensional recursive mesh bisection is then further modified to incorporate an improved strategy for the selection of bisections. This enables an automatic nodal placement technique to be implemented in conjunction with the grid generator. The dissertation then investigates methods of generating triangular grids over parametric surfaces. This includes a new definition of surface Delaunay triangulation with the extension of grid improvement techniques to surfaces. Based on the assumption that all surface grids of objects form polyhedral domains, a three dimensional mesh generation technique is derived. This technique is a hybrid of recursive domain bisection coupled with a min-max heuristic triangulation algorithm. This is done to achieve a computationlly efficient and reliable algorithm coupled with a fast nodal placement technique. The algorithm generates three dimensional unstructured tetrahedral grids over polyhedral domains with multi-connected regions in an average computational order of less than nlog(n).

Texto completo
Resumen
The thesis work concerns X-ray spectrometry for both medical and space applications and is divided into two sections. The first section addresses an X-ray spectrometric system designed to study radiological beams and is devoted to the optimization of diagnostic procedures in medicine. A parametric semi-empirical model capable of efficiently reconstructing diagnostic X-ray spectra in 'middle power' computers was developed and tested. In addition, different silicon diode detectors were tested as real-time detectors in order to provide a real-time evaluation of the spectrum during diagnostic procedures. This project contributes to the field by presenting an improved simulation of a realistic X-ray beam emerging from a common X-ray tube with a complete and detailed spectrum that lends itself to further studies of added filtration, thus providing an optimized beam for different diagnostic applications in medicine. The second section describes the preliminary tests that have been carried out on the first version of an Application Specific Integrated Circuit (ASIC), integrated with large area position-sensitive Silicon Drift Detector (SDD) to be used on board future space missions. This technology has been developed for the ESA project: LOFT (Large Observatory for X-ray Timing), a new medium-class space mission that the European Space Agency has been assessing since February of 2011. The LOFT project was proposed as part of the Cosmic Vision Program (2015-2025).

Recent international reports, for example TIMSS (2011 & 2015), point to serious challenges in South African learner performance in Mathematics and Science. Of greatest concern is that research findings (e.g. Graven, Venkat, Westaway and Tshesane 2013) suggest that many South African learners show signs of mathematical knowledge gaps in the lower grades. Hence, there is a need to address challenges of this nature very early in Foundation Phase. This study was undertaken with a view to contribute towards addressing mathematical challenges encountered by learners in Foundation Phase This empirical enquiry was undertaken under the auspices of the South African Numeracy Chair Project (SANCP) at Rhodes University whose mission is to develop sustainable ways of improving quality teaching and learning of Mathematics in South Africa. A relatively new SANCP programme called Pushing for Progression (PfP) run as part of the after-school Maths Clubs to develop the number sense and four Operations in learners was used to achieve the research aims of this study. Research participants were drawn from the Maths Clubs established by the researcher in a small rural town of Ottosdal in the North West Province of South Africa. This Study is grounded on the Vygotskian perspective and uses the interpretivist qualitative research method for data collection and analysis. Sampling was done opportunistically by enlisting participants (12 teachers and 117 learners) on the basis of their availability and willingness to participate. Pre- and post-assessment of learners’ proficiency on the four Basic Operations was conducted at the beginning and at the end of the research project, respectively. This was done to determine the impact of the project on learner performance. Data analysis was done thematically and through the comparison of learner results of the pre- and post-assessment. The findings point to the effectiveness of the PfP Programme in learner performance. This can be deduced from improved scores between pre- and post-assessment and the observations made by participant-teachers on their respective club learners’ mathematical proficiencies. Accordingly, based on the findings, this study recommends, inter alia, that since the PfP programme is still in its early stages, similar research be conducted elsewhere. Additionally, the Department of Basic Education could consider exploring the PfP programme as one of several other strategies to help improve learner proficiency in Mathematics.

En ASME 2004 International Mechanical Engineering Congress and Exposition. ASMEDC, 2004. http://dx.doi.org/10.1115/imece2004-62202.

Texto completo
Resumen
In the case of internal finishing of the bent section of a complex shaped tube, such as found in high purity fittings, by a magnetic abrasive finishing process, the magnetic field at the finishing area and, therefore, the finishing force are hardly uniform over the entire finishing area due to the geometry. This affects the abrasive behavior against the inner surface of the bent section, changing the finishing characteristics of SUS304 stainless steel fittings. In practice, non-uniformities in the surface finish remain at the bent section between the inside, outside, and lateral regions. This unevenness combines to cause difficulties in achieving uniform finishing. Magnetic abrasive is generally supplied with ferrous particles, and the ferrous particles experience greater magnetic force and play a role in pressing the magnetic abrasive against the target surface. This paper studies the finishing mechanism in view of the relationship between the magnetic field, the ferrous particles mixed with magnetic abrasive, and the finishing characteristics. The experiments identify the finishing conditions required for successfully diminishing the non-uniformity in the finished surface, and methods are recommended to satisfy the required conditions. The experiments using the proposed methods show the feasibility of producing a uniformly finished mirror surface.

Resumen
In this paper, a novel method to realize three-dimensional microstructures is presented. The presented technology is based on a direct write technique using a scanning laser system to pattern thick-film SU-8. This technique combines the best features of photolithography techniques in multi-layer processing with the versatility of existing 3D prototyping technologies. It enables laser pulsing spot-by-spot for both in-plane and in-depth processing. The results presented here represent the first step towards the realization of categories of MEMS devices with mechanical, optical, and/or biological functionalities.

Texto completo
Resumen
In this paper, a computational approach is adopted to solve the optimal control and optimal parameter selection problems of a rotating flexible beam fully covered with active constrained layer damping (ACLD) treatment. The beam rotates in a vertical plane under the gravitational effect with variable angular velocity and carries an end mass. Tangent coordinate system and the moving coordinate system are used in the system modeling. Due to the highly nonlinear and coupled characteristics of the system, a relative description method is used to represent the motion of the beam and the motion equations are set up by using relative motion variables. Finite element shape functions of a cantilever beam [1] are used as the displacement shape functions in this study. Lagrangian formulation and Raleigh-Ritz approach [2] are employed to derive the governing equations of motion of the nonlinear time-varying system. The problem is posed as a continuous-time optimal control problem. The control function parameters are the control gains. The two system parameters are the thickness of the constraining layer and the viscoelastic material layer. The software package MISER3.2, which is based on the Control Parametrization and the Control Parametrization Enhancing Transform (CPET) techniques is used to solve the combined problems. The optimal solution takes the end deflection, control voltage and the total weight into account. Results show that substantial improvements are obtained with ACLD as compared to the passive constrained layer damping (PCLD) treatment.

"Singular Superposition/BEM Inverse Technique for Reconstruction of Multi-Dimensional Heat Flux Distributions With Application to Film-Cooling Slots". En ASME 2004 International Mechanical Engineering Congress and Exposition. ASMEDC, 2004. http://dx.doi.org/10.1115/imece2004-62208.

Texto completo
Resumen
A hybrid singularity superposition/boundary element-based inverse problem method for the reconstruction of multi-dimensional heat flux distributions is developed. Cauchy conditions are imposed at exposed surfaces that are readily reached for measurements while convective boundary conditions are unknown at surfaces that are not amenable to measurements such as the edges of cooling slots. The purpose of the inverse analysis is to determine the heat flux distribution along cooling slot surfaces. This is accomplished in an iterative process by distributing a set of singularities at the vicinity of the cooling slot surface inside a fictitious extension of the physical domain with a given initial strength distribution. A forward steady-state heat conduction problem is solved using the boundary element method (BEM), and an objective function is defined to measure the difference between the heat flux measured at the exposed surfaces and the heat flux predicted by the BEM under the current strength distribution of the singularities. A genetic algorithm iteratively alters the strength distribution of the singularities until the measuring surfaces heat fluxes are matched, thus, satisfying Cauchy conditions. Subsequent to the solution of the inverse problem, the heat flux at the inaccessible surface is computed using the BEM. The hybrid singularity superposition/BEM approach thus eliminates the need to mesh the surface of the film cooling slot and the need to parametrize the heat flux over that surface. Rather, the heat flux is determined in a post-processing stage after the inverse problem is solved. This constitutes a tremendous advantage in solving the inverse problem, particularly in three-dimensional applications.

Resumen
Unidirectional stratified flow of two fluids between two parallel plates is modeled using the Level-Set method. A localized mass correction term is used to ensure mass conservation at every axial cross section. The mass correction term is based on the mass flowrates. Results for various combinations of density, viscosity and mass flowrate ratios are presented. Available fully-developed exact solutions for unidirectional stratified flow are used to validate the numerical simulations. The evolutions of the interface in the developing region are also captured and compared well with “exact” solutions.

Texto completo
Resumen
The paper presents analysis and optimization of the sonic drill head rotor housing assembly. The project was initiated by Layne Christensen DTC and performed at Purdue University Calumet in cooperation with the company engineers. Full finite element (FE) models of the rotor housing assembly were developed. Real world working conditions were applied onto the FE models in Ansys Workbench. With different Material setups, multiple trials were simulated so that the performances of material changes can be observed. Results of the total deformations were collected and analyzed both graphically and numerically. Based on the analysis results, recommendations on optimization of the rotor housing assembly were provided. A proposed new design was also developed and analyzed, and a numerical comparison of the new and original designs were made to give the company further recommendations. Support of Layne Christensen DTC is greatly appreciated.

Texto completo
Resumen
Abstract ASME PVP conference papers from 2007 and 2008 (References [1] and [2]) discuss fatigue analyses related to 304 stainless steel instrumentation spacer rings (rings) that had cracked while installed in a thermal fatigue test loop. The rings contain circular radial penetrations extending through the ring that allow access for instrumentation probes to monitor the fluid temperature and pressure. Routine inspection of the test loop revealed actively growing cracks in each port that appeared to start near the inner diameter (ID) of the ring. In this work, three-dimensional finite element analysis (FEA) thermal and fully-elastic structural Abaqus models were made from the available test measurements. The commercial crack modeling software, Franc3D, was used to complete an M-integral based fatigue crack growth (FCG) analysis. An assumed initial crack was inserted in a single port of interest near the ID of the FEA model. The crack was explicitly grown in an automated fashion over iterative growth steps. The explicit FCG analysis was able to match the directional path and final flaw size seen in the destructive evaluation metallographic images of the rings with the assumed input parameters exceptionally well, including shifting directions and splitting the crack without need of user input. The results of this work indicate that an explicit FCG analysis is feasible for a non-trivial thermal-structural problem.

En ASME 2021 Pressure Vessels & Piping Conference. American Society of Mechanical Engineers, 2021. http://dx.doi.org/10.1115/pvp2021-62201.

Resumen
Abstract During a Spring 2019 refueling outage, Plant A, a pressurized water reactor (PWR), experienced difficulty removing the reactor vessel upper internals because of degraded thermal shield (TS) support block bolts (TSSBBs), TS flexures, and baffle-former bolts (BFBs). None of the degradation was expected based on prior industry operating experience. Dynamic finite element analyses (FEA) and neutron noise monitoring were performed to diagnose the degraded condition. The FEA simulated the indications from the 2019 outage to assess the effect that degradation would have on the dynamic response of the lower reactor vessel internals. In parallel, neutron noise data was collected and evaluated to characterize the vibratory signature of the Plant A lower internals. The results of the FEA and neutron noise monitoring were coupled to compare degradation scenarios with the measured data, and it was thus predicted that multiple non-functional (fully failed) flexures existed at Plant A. Those predictions were subsequently validated during the Fall 2020 outage. This use of neutron noise to detect and diagnose degradation represents the practice of proactive structural health monitoring (SHM) for reactor vessel internals. This remote condition monitoring and diagnostic computational framework is currently being enhanced by quantifying the sensitivity of the structural dynamics to different degradation scenarios. This methodology leverages benchmarked FEA models and machine learning methods to enhance interpretability of neutron noise results and proactively diagnose different degradation scenarios, thus enabling prognostic asset management for reactor structures.
