Resumen
We sintered α (6H)- and β(3C)-SiC powders using an Al-B-C additive. SiC powders were densified to > 98% of the theoretical density from 1950 to 2150oC with 0.67-2.7 mass % AlB2 and 2.0 mass % C. Sintering temperatures are 150-200 oC lower than the conventional. During sintering, 6H polytype in α-SiC powder was partly transformed to 4H. α-SiC powder grew moderately into plate-shaped grains. β-/SiC powder was completely transformed to 6H and subsequently 4H with large grain growth. Low-temperature sintering enabled us to use hot isostatic pressing resulting in pore-free SiC materials.

AlN-SiC ceramics with 0 to 75 mol% of AlN were fabricated through pressureless sintering of very fine AlN and SiC. Powder compacts with different amounts of AlN were fired at 2000°C for 1 h in Argon gas flow using an induction-heating furnace. The microstructure and phases present in the products were evaluated using SEM and XRD. The AlN-SiC ceramics had a porous structure with 30% porosity, and the grain size was increased with the addition of AlN. XRD analysis showed that 2H was a main phase in all samples, though 3C and 6H phases were found in 25 mol%AlN-75 mol%SiC ceramic. The electrical properties of the AlN-SiC ceramics were evaluated at various temperatures ranging from room temperature to 300°C. The electrical conductivity of the AlN-SiC ceramics depended on the amount of AlN and on the temperature. The 75 mol%AlN-SiC ceramic had higher electrical resistance, though the other samples were electrical conductors. The highest electrical conductivity was obtained with the 25 mol% AlN composition, which was 7 S/m at room temperature and 30 S/m at 300°C. The Seebeck coefficient for the AlN-SiC ceramics increased with rising temperatures. The AlN-SiC ceramics with 50 mol%AlN had the highest Seebeck coefficient of 220 2V/K at 300°C.

ABSTRACT Deformed wing virus (DWV) of honeybees (Apis mellifera) is closely associated with characteristic wing deformities, abdominal bloating, paralysis, and rapid mortality of emerging adult bees. The virus was purified from diseased insects, and its genome was cloned and sequenced. The genomic RNA of DWV is 10,140 nucleotides in length and contains a single large open reading frame encoding a 328-kDa polyprotein. The coding sequence is flanked by a 1,144-nucleotide 5′ nontranslated leader sequence and a 317-nucleotide 3′ nontranslated region, followed by a poly(A) tail. The three major structural proteins, VP1 (44 kDa), VP2 (32 kDa), and VP3 (28 kDa), were identified, and their genes were mapped to the N-terminal section of the polyprotein. The C-terminal part of the polyprotein contains sequence motifs typical of well-characterized picornavirus nonstructural proteins: an RNA helicase, a chymotrypsin-like 3C protease, and an RNA-dependent RNA polymerase. The genome organization, capsid morphology, and sequence comparison data indicate that DWV is a member of the recently established genus Iflavirus.

Includes of suffix, prefix and infix to word in this research the writer want to know the third semester students at FKIP UIR capability in changing the derivational word. This research is descriptive quantitative there are one variable in this research. The location of this research was in English department of FKIP UIR and the writer took the data on November 20011 there are seven clases in third semester that consist of 317 student as population and the sample of this research was 47 students in 3c class. There is one instrument in this research, it is test and this test was given consist of three test. It was used to know students ability in changing the derivational word. From the data analysis, it can be seen for the first test there are 89,36% students feel difficult in changing infix 87,23% students feel difficult in changing suffix and 0% in changing prefix, after that from the second test there are 100% students feel difficult in changing infix, 80,85% students feel difficult in changing suffix and 21,27% students feel difficult in changing prefix and the last test there are 95,74% students feel difficult in changing infix, 87,23% students feel difficult in changing suffix and 68,08% students feel difficult in changing prefix, so it can be concluded the students more difficult in changing derivational word into infix. It can be seen from the explanation above from three times taken the data.

Resumen
We report observations of the radio galaxy 3C 317 (at z = 0.0345) located at the center of the Abell cluster A2052, obtained with the VLT/MUSE integral field spectrograph. The Chandra images of this cluster show cavities in the X-ray emitting gas, which were produced by the expansion of the radio lobes inflated by the active galactic nucleus (AGN). Our exquisite MUSE data show with unprecedented detail the complex network of line emitting filaments enshrouding the northern X-ray cavity. We do not detect any emission lines from the southern cavity, with a luminosity asymmetry between the two regions higher than ~75. The emission lines produced by the warm phase of the interstellar medium (WIM) enable us to obtain unique information on the properties of the emitting gas. We find dense gas (up to 270 cm−3) that makes up part of a global quasi spherical outflow that is driven by the radio source, and obtain a direct estimate of the expansion velocity of the cavities (265 km s−1). The emission lines diagnostic rules out ionization from the AGN or from star-forming regions, suggesting instead ionization from slow shocks or from cosmic rays. The striking asymmetric line emission observed between the two cavities contrasts with the less pronounced differences between the north and south sides in the hot gas; this represents a significant new ingredient for our understanding of the process of the exchange of energy between the relativistic plasma and the external medium. We conclude that the expanding radio lobes displace the hot tenuous phase of the interstellar medium (ISM), but also impact the colder and denser ISM phases. These results show the effects of the AGN on its host and the importance of radio mode feedback.

Introduction N6-methyladenosine (m6A) is the most prevalent post-transcriptional modification of eukaryotic mRNA. Accumulating evidence suggests that RNA m6A methylation exerts crucial roles in oncogenesis. However, the mRNA m6A methylation pattern in chronic lymphocytic leukemia (CLL) has not been investigated. Hence, the aim of this study is to perform a comprehensive profiling to identify distinct m6A methylation signatures in CLL patients. Methods Peripheral blood samples from de novo CLL patients were collected with informed consents at the Department of Hematology in Shandong Provincial Hospital Affiliated to Shandong First Medical University. CD19+ B cells were isolated with informed consents from healthy donors. Methylated RNA immunoprecipitation sequencing (MeRIP-Seq) was conduct to profile mRNA m6A methylation of CLL-B cells and normal CD19+ B cells at Novogene (Beijing, China). The library preparations were sequenced on an Illumina Novaseq platform with a paired-end read length of 150 bp according to the standard protocols. The sequencing was carried out with 3 independent biological replicates. After mapping reads to the reference genome, exomePeak R package was used for the m6A peak identification in each anti-m6A immunoprecipitation group with the corresponding input samples serving as a control, and q-value threshold of enrichment of 0.05 was used for all data sets. The m6A-enriched motifs of each group were identified by HOMER. Differential peak calling was performed using exomePeak R package with parameters of p-value < 0.05 and fold_change > 1. Functional enrichment analyses of gene ontology (GO) and kyoto encyclopedia of genes and genomes (KEGG) of differentiated peaks associated genes were performed. All investigators comply with the guiding principles for experimental procedures found in the Declaration of Helsinki of the World Medical Association. Results By MeRIP sequencing, significant distribution of methylation peaks were detected in 5'UTR, 3'UTR and CDS regions of CLL primary cells (Figure 1A) and normal B cells (Figure 1B). Figure 1C-D illustrated the percentage of methylation peaks in the five regions, suggesting distinct m6A patterns in CLL cells. Moreover, Figure 1E-F revealed the positions of m6A methylation peaks in chromosomes of CLL and normal B cells. Furthermore, the compared distributions of m6A methylation peaks in CLL and normal B cells were presented in Figure 2A. Besides, the bean plot visibly displayed the obvious differentiation of methylation peaks in CLL group and normal B cells in read density (Figure 2B). Importantly, a total of 1836 significantly changed peaks, of which 1519 were significantly up-regulated and 317 peaks were significantly down-regulated (p<0.05, |log2Foldchange|>1; Figure 2C). These m6A peaks were located across 1850 genes. Functional enrichment analyses identified that differentiated peaks associated genes were potentially regulate RNA metabolic process via oncogenic pathways in CLL pathogenesis (Figure 3A-B). In addition, HOMER analysis identified 38 significant de novo m6A peak motifs, top 10 most significant peak motifs of which were presented (Figure 3C), illuminating potential detailed mechanism of m6A RNA methylation in the tumorigenesis and progression of CLL. Conclusion Taken together, our investigations explored for the first time the m6A methylation pattern of mRNA in CLL. m6A modifications play crucial roles in the progression and survival of CLL patients,highlighting m6A modifications-targeted intervention formulating a novel treatment paradigm in progressed CLL that warrants clinical investigation. The pH and conductometric study of the electrochemical properties of aqueous solutions (1∙10-4 ÷ 9∙10-3 М) YNHCH2SO3Na (Y = H, CH3, HOCH2CH2, (CH3)3C and C6H5CH2) was carried out. The values of the limiting electrical conductivity were calculated by extrapolation according to Shidlovsky. The activation parameters of the electrical conductivity of the systems “sodium aminomethanesulfonate – H2O” at 293-313 K were obtained.

This work investigates the 3C-SiC heteroepitaxial growth on silicon substrates having a wide variety of orientations, i.e. (100) on axis and 2°off, (111), (110), (211), (311), (331), (510), (553) and (995). All the 3C-SiC layers were grown using the same two-step CVD process with a growth rate of 2 μm/h. According to X-ray diffraction characterizations, direct heteroepitaxy (layer having exactly the same orientation as the substrate) was successful on most of the Si substrates except for (110) one which was the only orientation leading to obvious polycrystalline deposit. Each layer led to a specific surface morphology, the smoothest being the ones grown on Si (100)2°off, and (995) substrates. None of these layers cracked upon cooling though those grown on Si (111), (211) and (553) substrates were highly bowed.

3C-SiC films were grown on Si by VPE using CBr4 as the carbon source, at temperatures ranging between 1100 to 1250°C. XRD, TEM, AFM, and SEM results indicate that the epitaxy proceeds as a 3D growth of uncoalesced islands at low temperature, whereas a continuous layer with hillocks on top is obtained above 1200°C. The shape and faceting of the islands are analyzed by AFM, showing (311) preferred facets.

ABSTRACT Unlike 3C protease, the severe acute respiratory syndrome coronavirus (SARS-CoV) 3C-like protease (3CLpro) is only enzymatically active as a homodimer and its catalysis is under extensive regulation by the unique extra domain. Despite intense studies, two puzzles still remain: (i) how the dimer-monomer switch is controlled and (ii) why dimerization is absolutely required for catalysis. Here we report the monomeric crystal structure of the SARS-CoV 3CLpro mutant R298A at a resolution of 1.75 Å. Detailed analysis reveals that Arg298 serves as a key component for maintaining dimerization, and consequently, its mutation will trigger a cooperative switch from a dimer to a monomer. The monomeric enzyme is irreversibly inactivated because its catalytic machinery is frozen in the collapsed state, characteristic of the formation of a short 310-helix from an active-site loop. Remarkably, dimerization appears to be coupled to catalysis in 3CLpro through the use of overlapped residues for two networks, one for dimerization and another for the catalysis.

An intercontinental VLBI experiment at 327 MHz involving the Ooty Radio Telescope and telescopes at Jodrell Bank, Westerbork, Torun and Crimea was carried out during December 8–12, 1983. The purpose of the experiment was to (i) establish the feasibility of performing MKII VLBI observations from India and (ii) to study the low frequency structure of nearby galaxies, quasars and some other well known radio sources at metre wavelengths. The sources observed were NGC 262, 315, 1052, 1068, 1265, 1275, 4151, 4486, 7674 and 3C 120; PKS 1055+018, 1148−001 and CTA 102; 3C 237 and SS 433. The bright NGC galaxies chosen were selected from an Interplanetary Scintillation survey of 150 nearby galaxies (Bagri and Ananthakrishnan 1983); they were found to have a flux density greater than about 0.2 Jy in subarcsec components at 327 MHz and have high frequency VLBI observations. The quasars chosen are amongst the most compact sources known at metrewavelengths. The other objects were the well known sources which have been studied extensively at higher frequencies.

Abstract Background Type 3c Diabetes Mellitus (DM3c) or Pancreatogenic Diabetes is defined as diabetes secondary to pancreatic disease. The most common cause of Type 3c Diabetes is chronic pancreatitis, accounting for 80% of cases.1 Hereditary pancreatitis, with mutations such as SPINK1, CFTR, CTRC, is an underdiagnosed cause of chronic pancreatitis leading to Type 3c Diabetes Mellitus.1 The typical disease course for hereditary pancreatitis is recurrent episodes of acute pancreatitis that leads to chronic pancreatitis.1 The average time between hereditary pancreatitis diagnosis and development of DM3c has been shown to be 80 +/- 24 months.1 We present a case of previously undiagnosed hereditary pancreatitis presenting as new onset Type 3c Diabetes Mellitus. Clinical Case A 33-year-old male presents with a two month history of diarrhea, epigastric pain, and weight loss. Vital signs on presentation were blood pressure 105/74, heart rate 59, BMI 13. Physical exam revealed a cachectic appearing male with epigastric tenderness. Lab-work showed blood glucose 819 mg/dL, SOsm 313 mOsm/kg, lipase 21 U/L, bicarbonate 33 mmol/L. A computed tomography (CT) of the abdomen revealed punctate pancreatic parenchymal calcifications, suggestive of chronic pancreatitis. He was admitted to general medicine for management of HHS, where he was treated with IV hydration and insulin therapy. Workup for new onset diabetes revealed a negative anti-GAD Ab <5.0 U/mL, and C peptide level 0.5 ng/mL which suggested β cell dysfunction and decreased endogenous insulin secretion. With these lab values, along with radiographic findings consistent with chronic pancreatitis, the likely driving factor for his diabetes was pancreatic disease and he was diagnosed with DM3c. Severe pancreatic disease was further shown by a reduced fecal elastase level of <50, suggesting exocrine pancreatic insufficiency. Investigating the etiology of his chronic pancreatitis, history revealed no prior episodes of acute pancreatitis or abdominal pain, and no family history of pancreatitis. Workup for common causes of pancreatitis, including alcohol use, hypertriglyceridemia, and gallstones was negative, and genetic workup was pursued. The patient was found to be heterozygous for a SPINK1 mutation, which is a known mutation in hereditary pancreatitis.1 Therefore, hereditary pancreatitis was determined to be the likely cause of his chronic pancreatitis which led to Type 3c Diabetes Mellitus. A small number of radio galaxies have two sets of classical double radio lobes with the radio axes aligned in different directions. Furthermore differences in the properties of the radio lobes such as the surface brightness and spectral index indicate that the two sets of double radio structure have different ages. The radio ejection axis has therefore changed direction with time. In the first two known radio galaxies of this type, 3C 315 and B2 0055+26, the host galaxy is a member of a close pair of ellipticals in a common optical envelope suggesting that the complex radio structure is caused by gravitational interaction.

Background: Obstetric anal sphincter injuries (OASIS) might be associated with long-term urinary and anorectal morbidities. The aim of the study was to investigate the risk factors and clinical implications of OASIS associated with vacuum-assisted deliveries versus normal vaginal deliveries. Methods: A series of 413 consecutive OASIS cases were retrospectively analyzed. A comparison was made between OASIS cases diagnosed following vacuum-assisted deliveries versus OASIS cases diagnosed following normal vaginal deliveries. Multivariable analysis was used to study the association between vacuum-assisted deliveries and superficial (3A and 3B) versus deep (3C and 4) perineal tears. Results: The study population comprised 88,123 singleton vaginal deliveries. Diagnosis of OASIS was made in 413 women (0.47% of the total cohort), 379 (91.8%) of whom had third-degree tears and 34 (8.2%) of whom had fourth-degree tears. Among the 7410 vacuum-assisted deliveries, 102 (1.37%) had OASIS, whereas, among the 80,713 normal vaginal deliveries, only 311 (0.39%) had OASIS. In a multivariate analysis, only vacuum-assisted delivery was found to be associated with a significant risk of deeper (3C or 4) perineal tears (OR = 1.72; 95% CI 1.02–2.91; p = 0.043). Conclusions: Vacuum-assisted instrumental intervention is a significant risk factor for OASIS and especially for deeper tears, independent of other maternal and obstetric risk factors.

Background: Numerous scales exist for the classification of major bleeding events. Limited data compare the most commonly used bleeding scales within a single at-risk cohort of patients with atrial fibrillation. Here, we analyze bleeding outcomes according to the ISTH (International Society on Thrombosis and Hemostasis), TIMI (Thrombolysis in Myocardial Infarction), GUSTO (Global Usage of Strategies to Open Occluded Arteries), and BARC (Bleeding Academic Research Consortium) bleeding scales in the ENGAGE AF (Effective Anticoagulation With Factor Xa Next Generation in Atrial Fibrillation)–TIMI 48 trial (NCT00781391) of edoxaban versus warfarin. Methods: A total of 21 105 patients with atrial fibrillation at risk for stroke (CHADS 2 score ≥2) were enrolled in the ENGAGE AF-TIMI 48 trial comparing warfarin with a higher- (60/30 mg daily) or lower- (30/15 mg daily) dose edoxaban regimen. Median follow-up was 2.8 years. Bleeding events occurring among on-treatment patients were examined. Annualized event rates were calculated by the ISTH, TIMI, GUSTO, and BARC scales and compared across treatment arms. Cox proportional hazards for a first bleeding event of each type were calculated for higher-dose edoxaban regimen vs warfarin and lower-dose edoxaban regimen versus warfarin. Results: A total of 10 311 bleeding events were reported. In a comparison of the most severe events in each scale, ISTH major bleeding was the most common (n=1289), followed by TIMI major (n=548), GUSTO severe/life-threatening (n=347), and BARC 3c+5 (n=276) bleeding. Lower bleeding risk with edoxaban compared with warfarin was seen regardless of bleeding scale (higher-dose edoxaban regimen range: hazard ratio [HR], 0.47 [95% CI, 0.35–0.62] for BARC 3c+5 versus HR, 0.80 [95% CI, 0.71–0.91] for ISTH major; lower-dose edoxaban regimen range: HR, 0.32 [95% CI, 0.23–0.45] for BARC 3c+5 versus HR, 0.47 [95% CI, 0.41–0.55] for ISTH major). Furthermore, a gradient of more pronounced risk reduction with edoxaban was observed with greater severity of first bleeding event (higher-dose edoxaban regimen: HR, 0.47 [95% CI, 0.35–0.62] for BARC 3c+5 bleeds versus HR, 0.86 [95% CI, 0.81–0.91] for any BARC bleed; lower-dose edoxaban regimen: HR, 0.32 [95% CI, 0.23–0.45] for BARC 3c+5 bleeds versus HR, 0.68 [95% CI, 0.63–0.72] for any BARC bleed). The direction of this trend was consistent for both gastrointestinal bleeding and nongastrointestinal bleeding. Conclusions: Among patients with atrial fibrillation at risk for stroke, there was a >4-fold difference in the frequency of the most severe bleeding events across commonly used bleeding scales. Furthermore, the relative safety of edoxaban compared with warfarin tended to increase with greater severity of bleeding. Clinical Trial Registration: URL: https://www.clinicaltrials.gov . Nine herbaceous perennial species were evaluated for use as flowering pot plants for late winter and early spring sales. Plugs of Achillea `King Edward', Arabis sturii, Armeria `Alba', Bergenia `New Hybrid', Chrysogonum virginianum, Dianthus `War Bonnet', Phlox `Chattahoochee', Platycodon `Sentimental Blue', and Veronica `Sunny Border Blue' were established in 14-cm (0.8-liter) round plastic containers, grown for one season, and covered with a thermoblanket for winter. Five plants of each species were transferred to a 21 ± 3C glasshouse for forcing under natural daylength at six 10-day intervals beginning 1 Dec. 1993. By this date plants had experienced approximately four weeks of temperatures below 5C. Ambis, Chrysogonum, and Phlox, species that naturally flower in spring, were the most floriferous. Days to first flower for Arabis decreased from 30 to 26 while flower number increased 44% by the 20 Dec. forcing date. For Phlox, days to first flower decreased from 36 to 31 by 20 Dec., but flower numbers were similar regardless of forcing date. Chrysogonum averaged eight flowers throughout the study, but days to first flower increased from 25 (1 Dec.) to 31 in all following forcing dates.

Model pengembangan perangkat yang digunakan model pengembangan 4D dari pendefinisian, perancangan, pengembangan, dan penyebaran oleh Thiagarajan, dkk (yang dimodifikasi). Ujicoba perangkat pembelajaran dilakukan di kelas 3C dan uji keefektifan dilakukan di kelas 3B di SDN Ketintang 1 Surabaya. Ujicoba perangkat dan uji keefektifan dilaksanakan dengan melakukan melakukan pengamatan aktivitas siswa selama mengikuti proses pembelajaran dan kemampuan guru mengelola pembelajaran. Tes hasil belajar siswa yang digunakan kemudian dianalisis validitas, sensitifitas, dan reliabilitas. Kemudian tes hasil belajar dianalisis dengan menggunakan persentase ketuntasan belajar. Berdasarkan hasil penelitian yang telah dilakukan, bahwa perangkat pembelajaran matematika realistik yang dikembangkan telah memenuhi kriteria valid,praktis dan efektif. Pada kelas ujicoba data yang diperoleh adalah aktivitas siswa mencapai 83.29%. Kemampuan guru mengelola pembelajaran termasuk dalam kategori baik. Hasil belajar siswa mengalami peningkatan yang baik yaitu 87%. Respon siswa memberikan respon yang positif yaitu 95.2%. Dengan demikian perangkat pembelajaran yang telah dikembangkan dapat digunakan sebagai alternative dalam pembelajaran di kelas. ABSTRACT The family 9 cellulase gene celI of Clostridium thermocellum, was previously cloned, expressed, and characterized (G. P. Hazlewood, K. Davidson, J. I. Laurie, N. S. Huskisson, and H. J. Gilbert, J. Gen. Microbiol. 139:307-316, 1993). We have recloned and sequenced the entire celI gene and found that the published sequence contained a 53-bp deletion that generated a frameshift mutation, resulting in a truncated and modified C-terminal segment of the protein. The enzymatic properties of the wild-type protein were characterized and found to conform to those of other family 9 glycoside hydrolases with a so-called theme B architecture, where the catalytic module is fused to a family 3c carbohydrate-binding module (CBM3c); CelI also contains a C-terminal CBM3b. The intact recombinant CelI exhibited high levels of activity on all cellulosic substrates tested, with pH and temperature optima of 5.5 and 70°C, respectively, using carboxymethylcellulose as a substrate. Native CelI was capable of solubilizing filter paper, and the distribution of reducing sugar between the soluble and insoluble fractions suggests that the enzyme acts as a processive cellulase. A truncated form of the enzyme, lacking the C terminal CBM3b, failed to bind to crystalline cellulose and displayed reduced activity toward insoluble substrates. A truncated form of the enzyme, in which both the cellulose-binding CBM3b and the fused CBM3c were removed, failed to exhibit significant levels of activity on any of the substrates examined. This study underscores the general nature of this type of enzymatic theme, whereby the fused CBM3c plays a critical accessory role for the family 9 catalytic domain and changes its character to facilitate processive cleavage of recalcitrant cellulose substrates.

Champion & Seth classified Indian forests into different ‘forest types and sub-types’, based on similarity of dominant vegetation and structural arrangement of species in each. However, it is not known if the species composition and community structure of butterflies is also different in each forest sub-type. If this is the case then each forest sub-type harbouring unique species can be taken as units of conservation on a sub-regional scale. The present study assesses for the first time the species composition and community structure of butterflies across 20 different and prominent ‘forest sub-types’ found across the state of Uttarakhand, western Himalaya. Data collected over eight years (2006–2009; June 2012; 2017–2020) using random seasonal sampling covering 307 transects revealed 370 butterfly taxa. Hierarchical clustering of butterfly abundances revealed seven different butterfly communities spread over 19 forest subtypes. Of these four forest sub-types (3C/C2a moist Shiwalik sal forest; 12/C2c moist temperate deciduous forest; 12/C1a ban oak forest; & 3C/C2c moist Terai sal forest) were identified as most important as they hold most of the butterfly diversity of the state including 58 rare taxa identified according to ‘rarity’ out of the total. GIS based mapping of these 58 priority species over laid on the protected area network and forest cover distribution in the state revealed many forested sites outside the PA network supporting these rare taxa. These sites along a physio-geographical gradient with important forest sub-types and rare taxa can be recommended and listed as new sites for conservation in the state.

AimsA conventional arthroscopic capsuloligamentous repair is a reliable surgical solution in most patients with scapholunate instability. However, this repair does not seem to be sufficient for more advanced injuries. The aim of this study was to evaluate the functional results of a wide arthroscopic dorsal capsuloligamentous repair (WADCLR) in the management of severe scapholunate instability.MethodsThis was a prospective single-centre study undertaken between March 2019 and May 2021. The primary outcome was the evaluation of the reduction of the radiological deformity and the functional outcomes after WADCLR. A secondary outcome was the evaluation of the effectiveness of this technique in patients with the most severe instability (European Wrist Arthroscopy Society (EWAS) stage 5). The patients were reviewed postoperatively at three, six, and 12 months.ResultsThe study included 112 patients (70 male and 42 female). Their mean age was 31.6 years (16 to 55). A total of three patients had EWAS stage 3A injuries, 12 had stage 3B injuries, 29 had stage 3C injuries, 56 had stage 4 injuries, and 12 had stage 5 injuries. There was a significant improvement of the radiological signs in all patients with a return to normal values. There was also a significant improvement in all aspects of function except for flexion, in which the mean increase was negligible (0.18° on average). There was also a significant improvement in all criteria for patients with a stage 5 injury, except for some limitation of extension, flexion, and radial and ulnar deviation, although these showed a trend towards improvement (except for flexion).ConclusionWADCLR is a minimally invasive, easy, and reproducible technique with few complications, offering a clear improvement in function and a reduction in the radiological deformity at one year postoperatively.Cite this article: Bone Joint J 2023;105-B(3):307–314.

ABSTRACT B lymphocytes converted into lymphoblastoid cell lines (LCLs) by an Epstein-Barr virus that expresses a conditional EBNA3C require complementation with EBNA3C for growth under nonpermissive conditions. Complementation with relatively large EBNA3C deletion mutants identified amino acids (aa) 1 to 506 (which includes the RBP-Jκ/CSL [RBP-Jκ] binding domain) and 733 to 909 to be essential for LCL growth, aa 728 to 732 and 910 to 992 to be important for full wild-type (wt) growth, and only aa 507 to 727 to be unimportant (S. Maruo, Y. Wu, T. Ito, T. Kanda, E. D. Kieff, and K. Takada, Proc. Natl. Acad. Sci. USA 106:4419-4424, 2009). When mutants with smaller deletions were used, only aa 51 to 400 and 851 to 900 were essential for LCL growth; aa 447 to 544, 701 to 750, 801 to 850, and 901 to 992 were important for full wt growth; and aa 4 to 50, 401 to 450, 550 to 707, and 751 to 800 were unimportant. These data reduce the EBNA3C essential residues from 68% to 40% of the open reading frame. Point mutations confirmed RBP-Jκ binding to be essential for wt growth and indicated that SUMO and CtBP binding interactions were important only for full wt growth. EBNA3C aa 51 to 150, 249 to 311, and 851 to 900 were necessary for maintaining LCL growth, but not RBP-Jκ interaction, and likely mediate interactions with other key cell proteins. Moreover, all mutants null for LCL growth had fewer S+G2/M-phase cells at 14 days, consistent with EBNA3C interaction with RBP-Jκ as well as aa 51 to 150, 249 to 311, and 851 to 900 being required to suppress p16INK4A (S. Maruo, Y. Wu, S. Ishikawa, T. Kanda, D. Iwakiri, and K. Takada, Proc. Natl. Acad. Sci. USA 103:19500-19505, 2006). We have confirmed that EBNA3C upregulates TCL1 and discovered that EBNA3C upregulates TCL1 through RBP-Jκ, indicating a central role for EBNA3C interaction with RBP-Jκ in mediating cell gene transcription.

ABSTRACTWe report on recent doping experiments of cubic GaN epilayers by Ge and investigate in detail the optical properties by photoluminescence spectroscopy. Plasma-assisted molecular beam epitaxy was used to deposit Ge-doped cubic GaN layers with nominal thicknesses of 600 nm on 3C-SiC(001)/Si(001) substrates. The Ge doping level could be varied by around six orders of magnitude by changing the Ge effusion cell temperature. A maximum free carrier concentration of 3.7×1020 cm-3 was measured in the GaN layers via Hall-effect at room temperature. Low temperature photoluminescence (PL) showed a clear shift of the donor-acceptor emission to higher energies with increasing Ge-doping. Above a Ge concentration of ∼ 2x1018cm-3 the near band edge lines merge to one broad band. From temperature dependent measurements of the observed excitonic and donor-acceptor transitions a donor-energy of ∼ 36 meV could be estimated for Ge.

Crystalline dicaesium mercury tetrachloride (Cs2HgCl4) is isomorphous with \beta-K_2SO_4 (space group Pnma, Z = 4) in its normal phase at room temperature. On cooling a sequence of incommensurate and commensurate superstructures occurs, below T = 221 K with modulations parallel to a*, and below 184 K with modulations along c*. The commensurately modulated structures at T = 185 K with {\bf q}= {{1}\over{5}}\bf{a}^* and at T = 176 K with {\bf q} = {{1}\over{3}}\bf{c}^* were determined using X-ray scattering with synchrotron radiation. The structure at T = 185 K has superspace group Pnma(\alpha,0,0)0ss with \alpha = 0.2. Lattice parameters were determined as a = 5\times9.7729\kern2pt(1), b = 7.5276\kern2pt(4) and c = 13.3727\kern2pt(7) Å. Structure refinements converged to R = 0.050 (R = 0.042 for 939 main reflections and R = 0.220 for 307 satellites) for the section t = 0.05 of superspace. The fivefold supercell has space group Pn2_1a. The structure at T = 176 K has superspace group Pnma(0,0,\gamma)0s0 with \gamma = {{1}\over{3}}. Lattice parameters were determined as a = 9.789\kern2pt(3), b = 7.541 \kern2pt(3) and c = 3 \times 13.418\kern2pt(4) Å. Structure refinements converged to R = 0.067 (R = 0.048 for 2130 main reflections, and R = 0.135 for 2382 satellite reflections) for the section t = 0. The threefold supercell has space group P112_1/a. It is shown that the structures of both low-temperature phases can be characterized as different superstructures of the periodic room-temperature structure. The superstructure of the 5a-modulated phase is analysed in terms of displacements of the Cs atoms, and rotations and distortions of HgCl4 tetrahedral groups. In the 3c-modulated phase the distortions of the tetrahedra are relaxed, but they are replaced by translations of the tetrahedral groups in addition to rotations.

Abstract. The eastern limestone cliff of Mount Néron (French Alps) was the theater for two medium-size rockfalls between summer and winter 2011. On 14 August 2011, a ~2000 m3 rock compartment detached from the cliff, fell 100 m below and propagated down the slope. Although most of the fallen rocks deposited on the upper part of the slope, some blocks of about 15 m in size were stopped by a ditch and an earthen barrier after a run-out of 800 m. An unstable overhanging ~2600 m3 compartment remained attached to the cliff and was blasted on 13 December 2011. During this artificially triggered event, 7 blocks reached the same ditch, with volumes ranging from 0.8 to 12 m3. A semi-permanent seismic array located about 2.5 km from the site recorded the two events, providing a unique opportunity to understand and to compare the seismic phases generated during natural and artificially triggered rockfalls. Both events have signal duration of ~100 s with comparable maximum amplitudes recorded at large distances (computed local magnitude of 1.14 and 1.05, respectively), most of the energy lying below 20 Hz. Remote sensing techniques (photogrammetry and lidar) were employed before and after the provoked rockfall, allowing the volume and fracturing to be characterized. This event was filmed by two video cameras, and the generated ground motions were recorded using two temporary 3C seismic sensors and three seismic arrays deployed at the slope toe. Videos and seismogram processing provided estimates of the propagation velocity during the successive rockfall phases, which ranges from 12 to 30 m s−1. The main seismic phases were obtained from combined video and seismic signal analyses. The two most energetic phases are related to the ground impact of fallen material after free fall, and to individual rock block impacts into the ditch and the earthen barrier. These two phases are characterized by similar low-frequency content but show very different particle motions. The discrete element technique allowed reproducing the key features of the rockfall dynamics, yielding propagation velocities compatible with experimental observations.

A revisited study of perovskite-based manganite La0.67Sr0.33MnO3 (LSMO) deposited onto corning-glass (CG) substrates is reported here. The LSMO films were fabricated with RF sputtering with different deposition times, varying from 60 to 180 min, resulting in films with thicknesses of 35.0–109.7 nm. Rietveld refinements of the XRD spectrum showed that the samples exhibited hexagonal structures with a space group of R-3c (167) and remained unmodified structures. Clusters of monolayer growth were observed from the morphologies of the films. Interestingly, the average particle size obtained with an AFM perceived an insignificant variation of 31.7–35.6 nm despite the film thickness increment. Optical, electrical and magnetic properties were studied, as particle-size variation usually has a notable contribution to changes in these properties. It was found that the optical band gap (Eg) was in direct variation with the deposition time due to enhanced thickness and surface roughness. The temperature dependence of the resistivity graph was fitted with the percolation model to obtain further enlightenment on the electrical transport in the films. Low-field magnetoresistance (LFMR) was observed for all samples. Ferromagnetic to paramagnetic transition was observed in the vicinity of all samples’ TC values.

AbstractThe 127I-NQR of(HOCH2)3CNH3+ I- was determined in the range 77 ≤ T/K ≤ 310. At T = 310 K the NQR signal fades out (Tm = 463 K). The 127I spectrum ( T =77 K.): v1 =29.195 MHz, v2 = 14.597 MHz, η(121l)=0, e QΦzz h-1 (127I) = 97.315 MHz, is in agreement with the crystal structure. The 127I NQR spectrum of 1,3-diammonium-2,2-dimethylpropane diiodide, (H3NCH2C(CH3)2CH2NH3)2+ ·2I- , is a quartet within the whole temperature range investigated, and the lines correspond to two crystallographically independent iodines: Space group P21/c, Z = 4, a = 731.2(3) pm, b = 689.0(3) pm, c = 2255.1(8) pm, β = 104.90(1)°. At 7 = 7 7 K the 127I NQR quartet is (MHz): v1 = 34.145, v2 = 32.805, v3 = 22.113, v4 = 16.787; at 295 K (same order, MHz): 30.559, 29.729, 19.810, 15.651. There are two combinations of the NQR frequencies. Considering the coordination of I-, the hydrogen bonds N -H ··· I, eQΦzzQ h-1 and η, we choose for I(1) v1 and v3, for I(2) v2 and v4. At 77 K eQΦzzQ h-1 (I(1))= 118.86 MHz,η (127I(1)) = 0.498, eQΦzzQ h-1 (I(2)) = 109.75 MHz, η(127I(2)) = 0.135 follow for the two iodine atoms. Both, eQΦzzQ h-1 (I(1)) and e eΦzzQ h-1(I(2)) decrease smoothly with increasing T: η I(2)) increases with increasing T whereas η(127I(1)) is almost constant within 77 ≤ T /K ≤ 4 0 6 . The 79,81Br NQR spectrum of l,3-diamino-2,2-dimethylpropane dihydrobromide is also a quartet, showing two crystallographic inequivalent Br atoms in the unit cell. The frequencies are (T =273 K, MHz): v1 (79Br)= 14.303, v2 (79Br)= 12.884, (81Br)= 11.951, v2(81Br) = 10.781; space group C2/c, Z = 8 , a = 2136.4(6) pm, b = 854.6(3) pm, c = 1125.8(3) pm, β = 93.23(1)°. Crystal structures and NQR results are discussed.

The method of high temperature physical vapor transport (HTPVT) is an available approach to prepare silicon carbide (SiC) ceramics with high density and high purity. In the present work, α-SiC (6H-SiC) and β-SiC (3C-SiC) powders were used as starting materials respectively to fabricate SiC ceramics with HTPVT process, and the effects of starting materials on nucleation, density, microstructure and mechanical properties of SiC ceramics were investigated. It showed that at high temperature, the decomposition rate of β-SiC was higher than that of α-SiC, and at the initial nucleation stage, the average grain size of SiC crystal obtained with β-SiC starting materials was smaller than that with α-SiC starting materials, because higher vapour pressure of gas phase which decomposed by β-SiC starting materials facilitated nucleation and growth of SiC grains. Density of the resulted SiC ceramics using α-SiC and β-SiC as starting materials was 3.16 g·cm-3 and 3.17 g·cm-3, indicating close values, while, using β-SiC as the starting materials, the grain size was smaller, consequently, the flexure strength was higher. Increasing growth temperature from 2200°C to 2300°C, the densities and the flexure strength of the SiC ceramics using either α-SiC or β-SiC were decreased.

BackgroundInterleukin(IL)-12 activates T cells pivoting the switch that turns lingering inflammation into acute inflammation and cancer rejection. However, its clinical utilization is limited by severe systemic toxicity. IL-12 is a potent inducer of PD-1 expression in T cells. Here, we present a conditional, antigen-dependent, non-editing CRISPR-activation (CRISPRa) circuit (RB-312) that delivers nanoscale doses of IL-12 for autocrine activation of CAR-T cells. RB-312 was also tested in combination with PD-L1 blocking antibody (atezolizumab).MethodsRB-312 is a CAR T cell engineered to express the IL-12 heterodimer via conditional transcription of its two endogenous subunits p35 and p40. The circuit includes two lentiviral constructs with one encoding HER2-specific chimeric antigen receptor and two sgRNAs targeting IL-12A or IL-12B and the other encoding linker for activation of T cells, complexed to dead Cas9 (dCas9)-VP64-p65-Rta transcriptional activator (VPR) (LdCV). Activation of CAR allows the release of dCas9 for nuclear localization and hence conditionally and reversibly induces the secretion of IL-12 p70 heterodimer.ResultsRB-312 induced low concentrations of IL-12 upon exposure to HER2+ FaDu cancer cells engineered to overexpress PD-L1 and this resulted in significantly enhanced production of IFN-γ, cytotoxicity and CAR-T proliferation (figure 1A). These effects were comparable to co-culturing conventional HER2 CAR with FaDu cells modified to express high doses of IL-12 (figure 1B). In vivo administration of RB-312 significantly enhanced survival of mice carrying FaDu xenografts compared to mice treated with the respective conventional HER2 CAR or cRB-312 (control lacking the IL-12 sgRNAs, figure 2A). RB-312 induced a strong upregulation of PD-1 in CAR-T cells in vivo (figure 2B). The critical role of the PD-1/PD-L1 interaction was demonstrated in vitro by comparing RB-312 proliferation when exposed to FaDu overexpressing PD-L1 or PD-L1 knock out cells (figure 3A). The autocrine effects of nanoscale IL-12 production limit the risk of off-tumor leakage and systemic toxicity. Here, we tested the combination of PD-1/PD-L1 blockade with IL-12-induced CAR-T cell activation demonstrated dramatic synergistic effects. We are currently evaluating the intrinsic combination of IL-12 delivery and PD-L1 resistance for the next generation of RB-312 product eliminating the need for systemic checkpoint blockade.

Abstract A metabolic syndrome (MetS) phenotype is characterized by insulin-resistance, atherogenic dyslipidemia, oxidative stress, and elevated cardiovascular risk and frequently involves subnormal levels of high-density lipoprotein (HDL) cholesterol. We evaluated the capacity of physicochemically distinct HDL subfractions from MetS subjects to protect low-density lipoprotein against oxidative stress. MetS subjects presented an insulin-resistant phenotype, with central obesity and elevation in systolic blood pressure and plasma triglyceride, LDL-cholesterol, apolipoprotein B, glucose, and insulin levels. Systemic oxidative stress, assessed as plasma 8-isoprostanes, was significantly higher (3.7-fold) in MetS subjects (n = 10) compared with nonobese normolipidemic controls (n = 11). In MetS, small, dense HDL3a, 3b, and 3c subfractions possessed significantly lower specific antioxidative activity (up to −23%, on a unit particle mass basis) than their counterparts in controls. In addition, HDL2a and 3a subfractions from MetS patients possessed lower total antioxidative activity (up to −41%, at equivalent plasma concentrations). The attenuated antioxidative activity of small, dense HDL subfractions correlated with systemic oxidative stress and insulin resistance and was associated with HDL particles exhibiting altered physicochemical properties (core triglyceride enrichment and cholesteryl ester depletion). We conclude that antioxidative activity of small, dense HDL subfractions of altered chemical composition is impaired in MetS and associated with elevated oxidative stress and insulin resistance. Induction of selective increase in the circulating concentrations of dense HDL subfractions may represent an innovative therapeutic approach for the attenuation of high cardiovascular risk in MetS.

A new series of di-spirooxindole analogs, engrafted with oxindole and cyclohexanone moieties, were synthesized. Initially, azomethine ylides were generated via reaction of the substituted isatins 3a–f (isatin, 3a, 6-chloroisatin, 3b, 5-fluoroisatin, 3c, 5-nitroisatin, 3d, 5-methoxyisatin, 3e, and 5-methylisatin, 3f, and (2S)-octahydro-1H-indole-2-carboxylic acid 2, in situ azomethine ylides reacted with the cyclohexanone based-chalcone 1a–f to afford the target di-spirooxindole compounds 4a–n. This one-pot method provided diverse structurally complex molecules, with biologically relevant spirocycles in a good yields. All synthesized di-spirooxindole analogs, engrafted with oxindole and cyclohexanone moieties, were evaluated for their anticancer activity against four cancer cell lines, including prostate PC3, cervical HeLa, and breast (MCF-7, and MDA-MB231) cancer cell lines. The cytotoxicity of these di-spirooxindole analogs was also examined against human fibroblast BJ cell lines, and they appeared to be non-cytotoxic. Compound 4b was identified as the most active member of this series against prostate cancer cell line PC3 (IC50 = 3.7 ± 1.0 µM). The cyclohexanone engrafted di-spirooxindole analogs 4a and 4l (IC50 = 7.1 ± 0.2, and 7.2 ± 0.5 µM, respectively) were active against HeLa cancer cells, whereas NO2 substituted isatin ring and meta-fluoro-substituted (2E,6E)-2,6-dibenzylidenecyclohexanone containing 4i (IC50 = 7.63 ± 0.08 µM) appeared to be a promising agent against the triple negative breast cancer MDA-MB231 cell line. To explore the plausible mechanism of anticancer activity of di-spirooxindole analogs, molecular docking studies were investigated which suggested that spirooxindole analogs potentially inhibit the activity of MDM2.
