It is known that for $M=M_{*}^{(N)}$, $\sOm=B_R(0)$, the above problem has a continuum of solutions. The case when $M>M_{*}^{(N)}$ is referred to as supercritical in the literature. We show that for $M$ near $KM_{*}^{(N)}$, $K>1$, there exist solutions with multiple condensations in $\sOm$. This study aims to determine the influence of E-product quality, e-promotion, and e-word of mouth on e-buying decisions. This study used an explanatory research type with a non-probability sampling technique through a purposive sampling approach. The data collection was carried out by distributing research questionnaires to 100 students of the Faculty of Social and Political Sciences, Diponegoro University. E-product quality, e-promotion, and e-word of mouth have a positive effect on e-buying decisions on Shopee consumers, FISIP Diponegoro University students. E-product qualitys accounted for 20.92% of e-buying decisin, e-ormotion accounted for 41.10%, and e-word of mouth accounted for 35.40% of e-buying decisions. The three variables simultaneously affecting the e-buying decision accounted for 46.20%. So, it is recommended that Shopee Mall can be more flexible about the use of promos offered, increase the speed of product delivery so that it does not exceed the estimated delivery and provide qualifications for sellers to provide more detailed and clear information and be easily accessible to consumers.Penelitian ini bertujuan untuk mengetahui pengaruh E-product quality, e-promotion, dan e-word of mouth terhadap e-buying decision. Penelitian ini menggunakan tipe explanatory research dengan teknik pengambilan sampel non probability sampling melalui pendekatan purposive sampling. Pengumulan data dilakukan dengan menyebarkan kuesioner penelitian pada 100 Mahasiswa Fakultas Ilmu Sosial dan Politik Universitas Diponegoro. E-product quality, e-promotion, dan e-word of mouth berpengaruh positif terhadap e-buying decision pada konsumen Shopee Mahasiswa FISIP Universitas Diponegoro. . E-product quality menyumbang 20,92% terhadap e-buying decisin, e-ormotion menyumbang sebesar 41,10%, dan e-word of mouth menyumbang sebesar 35,40% terhadap e-buying decision. ketiga variabel secara simultan mempengaruhi e-buying decision menyumbang sebesar 46,20%. Maka, disarankan agar Shopee Mall dapat lebih flexible mengenai penggunaan promo yang ditawarkan, meningkatkan kecepatan pengiriman produk agar tidak melebihi estimasi pengiriman dan memberikan kualifikasi bagi seller untuk memberikan informasi lebih detail dan jelas serta mudah diakses konsumen.

Resumen

Background Elastic scattering spectroscopy (ESS) is a noninvasive optical biopsy technique that can distinguish between normal and abnormal tissue in vivo. The handheld device measures ESS spectra of skin lesions and classifies lesions with an output of “Investigate Further” or “Monitor.” The algorithm was trained and validated with over 11,000 spectral scans from over 3500 skin lesions. The device performance was also evaluated in an associated clinical study. Objective The aim of this paper was to establish whether the use of a handheld ESS tool can improve the detection of skin malignancies by evaluating clinical performance while emulating a real-world telemedicine clinical care setting. Methods The associated clinical study examined an independent test set of 332 lesions in a prospective multicenter study that compared algorithm performance to biopsy results for diagnosing malignant lesions. A total of 50 cases were randomly selected from the study data base (25 malignant and 25 benign lesions). Device performance on these lesions had a 96% sensitivity. High-resolution digital images and the patient’s clinical information including prior skin cancer history, risk factors, and physical examination results were available for evaluation. A total of 57 primary care physicians participated in this study in 2 phases, the first phase with their standard-of-care diagnostic and the second phase regarding their evaluation with the device output. The physicians were educated on the ESS device before evaluating the cases in a random order. Case evaluation included the physician reporting their diagnosis, management decision, and confidence level without the device output in the first phase and with the device output in the second phase. The results were evaluated for sensitivity and specificity with confidence intervals. Results The diagnostic sensitivity of the readers without and with the use of the handheld ESS device increased significantly from 67% to 88% (P<.001). There was no significant difference in specificity at 40% and 53% (P=.05). The management sensitivity of the readers increased significantly with and without the use of the device, which, respectively, was 94% (91%-96%) and 81% (77%-85%) (P<.001), suggesting that the use of the device may reduce false negatives by 68%. Specificity was comparable for management decisions (P=.36) at 31% compared to 36% without the device. Conclusions The use of the handheld ESS device significantly improved diagnostic and management sensitivity over standard-of-care, with comparable specificity. While telemedicine has shown promise in many fields, studies have shown that in-person skin evaluation is superior to telemedicine evaluations; however, integration with this type of tool has the potential to improve early detection.

Background Digital technology and media use is integral to adolescents’ lives and has been associated with both positive and negative health consequences. Previous studies have largely focused on understanding technology behaviors and outcomes within adolescent populations, which can promote assumptions about adolescent technology use as homogeneous. Furthermore, many studies on adolescent technology use have focused on risks and negative outcomes. To better understand adolescent digital technology use, we need new approaches that can assess distinct profiles within study populations and take a balanced approach to understanding the risks and benefits of digital technology use. Objective The purpose of this study was to identify profiles of adolescent technology use within a large study population focusing on four evidence-based constructs: technology ownership and use, parental involvement, health outcomes, and well-being indicators. Methods Adolescent-parent dyads were recruited for a cross-sectional web-based survey using the Qualtrics (Qualtrics International, Inc) platform and panels. Technology use measures included ownership of devices, social media use frequency, and the Adolescents’ Digital Technology Interactions and Importance scale. Parent involvement measures included household media rules, technology-related parenting practices, parent social media use frequency, and the parent-child relationship. Health outcome measures included physical activity, sleep, problematic internet use, and mental health assessments. Well-being indicators included mental wellness, communication, and empathy. We used latent class analysis (LCA) to identify distinct profile groups across the aforementioned 4 critical constructs. Results Among the 3981 adolescent-parent dyads recruited, adolescent participants had a mean age of 15.0 (SD 1.43) years; a total of 46.3% (1842/3981) were female, 67.8% (2701/3981) were White, and 75% (2986/3981) lived in a household with an income above the poverty line. The LCA identified 2 discrete classes. Class 1 was made up of 62.8% (2501/3981) of the participants. Class 1 participants were more likely than Class 2 participants to report family-owned devices, have lower technology importance scores, have household technology rules often centered on content, have positive parent relationships and lower parent social media use, and report better health outcomes and well-being indicators. Conclusions Findings from this national cross-sectional survey using LCA led to 2 distinct profile groups of adolescent media use and their association with technology use and parent involvement as well as health and well-being outcomes. The two classes included a larger Class 1 (Family-Engaged Adolescents) and a smaller Class 2 (At-Risk Adolescents). The findings of this study can inform interventions to reinforce positive technology use and family support.

Background The COVID-19 outbreak has not only changed the lifestyles of people globally but has also resulted in other challenges, such as the requirement of self-isolation and distance learning. Moreover, people are unable to venture out to exercise, leading to reduced movement, and therefore, the demand for exercise at home has increased. Objective We intended to investigate the relationships between a Nintendo Ring Fit Adventure (RFA) intervention and improvements in running time, cardiac force index (CFI), sleep quality (Chinese version of the Pittsburgh Sleep Quality Index score), and mood disorders (5-item Brief Symptom Rating Scale score). Methods This was a randomized prospective study and included 80 students who were required to complete a 1600-meter outdoor run before and after the intervention, the completion times of which were recorded in seconds. They were also required to fill out a lifestyle questionnaire. During the study, 40 participants (16 males and 24 females, with an average age of 23.75 years) were assigned to the RFA group and were required to exercise for 30 minutes 3 times per week (in the adventure mode) over 4 weeks. The exercise intensity was set according to the instructions given by the virtual coach during the first game. The remaining 40 participants (30 males and 10 females, with an average age of 22.65 years) were assigned to the control group and maintained their regular habits during the study period. Results The study was completed by 80 participants aged 20 to 36 years (mean 23.20, SD 2.96 years). The results showed that the running time in the RFA group was significantly reduced. After 4 weeks of physical training, it took females in the RFA group 19.79 seconds (P=.03) and males 22.56 seconds (P=.03) less than the baseline to complete the 1600-meter run. In contrast, there were no significant differences in the performance of the control group in the run before and after the fourth week of intervention. In terms of mood disorders, the average score of the RFA group increased from 1.81 to 3.31 for males (difference=1.50, P=.04) and from 3.17 to 4.54 for females (difference=1.38, P=.06). In addition, no significant differences between the RFA and control groups were observed for the CFI peak acceleration (CFIPA)_walk, CFIPA_run, or sleep quality. Conclusions RFA could either maintain or improve an individual’s physical fitness, thereby providing a good solution for people involved in distance learning or those who have not exercised for an extended period. Our analysis covers the case of non-Lipschitz nonlinearities both in the differential equations and in the boundary conditions. Abstract Killer immunoglobulin-like receptors (KIR), usually restricted to NK cells, are expressed on T-cells with increasing age and contribute to age-related diseases. We examined the epigenetic changes associated with KIR expression. In T cells with absent KIR2DL4 transcripts such as the Jurkat cell line or CD4+CD28+ T cells, CpG sites of the proximal KIR2DL4 promoter were hypermethylated. DNMT inhibition in such cells induced low but appreciable KIR2DL4 transcription. DNMT inhibition was strikingly more effective in T cells that had some constitutive KIR2DL4 transcription, such as CD4 and CD8 T cells from elderly adults or the CD4+CD28−T cell line HUT78. Such cells had evidence of partial promoter demethylation; however, although the proximal nucleosome was partially relaxed, the only histone modification was an increase in dimethylated H3-Lys 4. In contrast, NK cells had a fully demethylated KIR2DL4 promoter and the full spectrum of histone modifications indicative of active transcription with H3 and H4 acetylation, di- and trimethylated H3-Lys 4, and reduced dimethylated H3-Lys 9. These results suggest that early steps of promoter activation in T cells include selective increase in H3-Lys 4 dimethylation and limited DNA demethylation. This partially accessible promoter is sensitive to DNMT inhibition, which is sufficient to induce full transcription without further histone acetylation and methylation. OBJECTIVES/SPECIFIC AIMS: In the context of skeletal muscle, IL-6 plays a major role in muscle quality. The goal of this project was to study the influence of systemic IL-6 on skeletal muscle mitochondrial physiology, most notably mitochondrial function (respiration and ROS production) and mitochondrial content. METHODS/STUDY POPULATION: To determine the influence of interleukin-6 (IL-6) on skeletal muscle mitochondria, high-resolution respirometry was performed to simultaneously measure oxygen consumption (JO2) and ROS production in differentiated myotubes incubated with increasing IL-6 (0, 10, 50, 100 ng/mL) for 18 hours in serum free conditions. To evaluate the impact of IL-6 on mitochondrial content we performed western blots on cell lysates from treated cells, measuring proteins of the mitochondrial electron transport chain (ETC) using a cocktail antibody and PGC-1α/PGC-1ß for mitochondrial biogenesis. To determine the role of mitochondrial ROS production on JO2 and mitochondrial content, we co-treated differentiated myotubes for 18 hours with 50 and 100ng/mL IL-6 and the mitochondrial specific antioxidant, MitoQ and performed respirometry for mitochondrial functional measurements and western blots for mitochondrial content.Statistical significance was evaluated by using a 2-tailed Student’s t-test and two-way ANOVA. Post hoc all-group analyses were conducted to determine which groups were different when the model was significant. RESULTS/ANTICIPATED RESULTS: Mitochondrial functional measurements show increased JO2 and increased ROS production in an IL-6 dose-dependent manner. Targeting mitochondrial ROS production with 0.5µm MitoQ attenuated IL-6 induced increases in JO2 and ROS production. Complexes I and II (CI, CII) of the ETC increased significantly in an IL-6 dose-wise fashion, and co-treatment with MitoQ normalized increases at 100ng/mL Il-6. 100ng/mL IL-6 significantly increased protein expression of PGC-1α and PGC-1ß. Co-treatment with MitoQ normalized IL-6 induced increase in PGC-1α. DISCUSSION/SIGNIFICANCE OF IMPACT: Our data suggest that when treated chronically at a high dose, IL-6 increases mitochondrial respiration, ROS production, and content. Targeting mitochondrial ROS production normalizes these mitochondrial adaptations. The present study provides new insights into mitochondrial physiology in the context of inflammation. Therapeutically targeting mitochondrial ROS production may impact skeletal muscle quality in certain populations.

MOOCs are open, online courses that use information technologies to enhance the learning experience and attract various people from the entire world. The current study uses the Technology Acceptance Model (TAM), as well as personal characteristics such as learning strategies, cognitive appraisal, and Kuhlthau’s (1991) model of information seeking as theoretical bases for defining factors that may influence students adopting MOOCs in their learning process, as well as describe their feelings during the learning process. The study was conducted in Israel during the 2014 academic year, and used both quantitative and qualitative techniques and involved 102 students who participated in a MOOC as part of the requirements in an offline course. They were requested to keep study diaries. The quantitative analysis revealed that perceived usefulness (PU) and perceived ease of use (PEOU) have a major influence on the intention to enroll in a MOOC. PEOU can be increased by improving the current MOOC platforms. PU can also be improved by providing content that suits the students’ needs. The qualitative analysis showed mood changes over time; the feelings of uncertainty were replaced by expressions of confidence. We found that students have different needs and expectations. Therefore, the MOOC’s platforms should provide multiple options to accommodate these needs.

Abstract Background: Hepatoblastoma (HB) is the most frequent liver tumor in children. While most tumors respond well to treatment with good outcomes, there are a subgroup of refractory tumors for which novel therapies are required. Emerging literature has suggested a role of Hsp90 in HB progression. One possible mechanism hypothesizes that Hsp90 helps stabilize misfolded proteins that contribute to HB progression including β-catenin. Our study aimed to investigate if inhibition of Hsp90 would affect the cell viability of hepatoblastoma cell lines in vitro. Methods: Commercially available HEPG2, and patient xenograft-derived hepatoblastoma cell lines (HB214, HB279, and HB303) were analyzed for differences in Hsp90 gene expression by qRTPCR as well as cell viability (n = 3 replicates for each cell line) using the MTS assay. Differences in cell viability for each cell line were further examined after treatment with a 0.1µM or 0.25µM dose of 17-AAG (an HSP90 inhibitor) for 48H. We examined gene expression and the MTS assay cell viability data using linear mixed effects modelling followed by t-tests to examine specific comparisons (significance at p &lt; 0.05). Results: Hsp90 gene expression was significantly increased in HEPG2 cells (p &lt; 0.001) and in HB279 cells (p &lt; 0 .05), but not in HB214, or HB303 cells, compared to HUVECs control cells. Examining clinically relevant characteristics, significant improvements to model fit were found with the inclusion of sex (p &lt; 0.01), β-catenin deletion status (p &lt; 0.01), and a higher PRETEXT stage but not from examining if cell lines were derived from metastatic parent tumors. While we observed no significant cell line viability differences between individual HB cell lines at baseline (0µM 17-AAG treatment) or overall significant differences for treatment with either 0.1µM or 0.25µM dosages of 17-AAG, there were differences in cell viability for individual HB cell lines. We observed significant decreases in cell viability in HEPG2 cells with 0.1µM (p &lt; 0.05) and 0.25µM (p &lt; 0.05), as well as in HB279 cells with 0.1µM (p &lt; 0.05) and 0.25µM (p &lt; 0.05) 17-AAG treatments but observed no effect of 17-AAG treatment on cell viability in HB214, or HB303 cells. Similar improvements in model fit were observed with the inclusion of sex (p &lt; 0.01), β-catenin deletion status (p &lt; 0.01), and a higher PRETEXT stage (p &lt; 0.001), but not from examining if cell lines were derived from metastatic parent tumors. Conclusion: HB cell lines express Hsp90 at levels correlated with age, β-catenin deletion status, and risk stage. Moreover, the expression levels of Hsp90 corresponded to the sensitivity of each cell line to the Hsp90 inhibitor 17-AAG, with cells more highly expressing Hsp90 having a greater reduction in cell viability. Continuing to understand the mechanisms of Hsp90 action in HB, particularly through the previously demonstrated connection with β-catenin, may identify potential targetable therapies in the care of HB. Camponotus sericeiventris Guérin são formigas descritas como onívoras, incluindo em sua dieta néctar floral e extrafloral, exsudato de hemípteros e de lepidópteros, artrópodes mortos e presas vivas, além de sementes e frutos forrageados no ambiente em que residem. O objetivo deste trabalho foi analisar o comportamento de forrageio de C. sericeiventris, correlacionando a atividade forrageadora com os fatores climáticos, bem como quantificar e identificar os recursos explorados pela espécie, o tempo de forrageio e o raio de ação. A espécie estudada revelou ser influenciada positivamente pela variação de temperatura do ambiente, aumentando sua atividade com a elevação da temperatura (p<0,05). Apenas 5,19% (n= 387) dos retornos foram possíveis de serem identificados, distribuídos em fezes (35,40%, n = 137); proteína animal (27,65%, n = 107) e fibra vegetal (36,95%, n = 143). Na maior parte dos retornos (94,81%, n= 7.072) as formigas não carregavam uma carga visível. Duas trilhas de forrageio, que partiam da colônia até as árvores onde as formigas buscavam recursos, foram medidas (73 e 86 m), representando um raio médio de distância por procura de recursos de 79,5 ±9,19 m o que resulta em 19.596 m² de área de ação de uma colônia. O tempo de forrageio de C. sericeiventris apresentou uma média de duração de 67 ±16’97’’ (37’03’’ – 101’) min. Os resultados encontrados no presente estudo fornecem importantes subsídios para a compreensão da dinâmica da atividade forrageadora da formiga C. sericeiventris em ambiente antrópico. The aim of this study was to examine the foraging behavior of C. sericeiventris, correlating the foraging activity and climatic factors as well as quantify and identify the resources exploited by the species and time of the foraging and action range. The specie studied was influenced positively by variations in the temperature. In most of the returns (94.81%, n = 7,072) the ants did not carry a load visible. Only 5.19% (n = 387) of the returns were identified and distributed as feces (35.40%, n = 137), animal protein (27.65%, n = 107) and vegetable fiber (36, 95%, n = 143). Two foraging trails, from colony to trees where ants were seeking resources, were measured (73 and 86 m) representing an average of the distance of 79.5 ± 9.19 m, resulting in 19,596 m² of colony action. The duration of foraging of the C. sericeiventris had an average of 67 ± 16’97’’ (37’03’’- 101’) minutes. The results of this study provide important insights into understanding the dynamics of foraging activity of the C. sericeiventris in the human environment. BACKGROUND: High body mass index (obesity) is a great health threat to aging individuals which results in increased blood pressure. MATERIALS AND METHODS: The Study was carried out in four different communities namely: Ikom, Akamkpa, Calabar South and Calabar Municipal of Cross River State, Nigeria. It was carried out amongst 130 women who voluntarily gave their consent during a cervical screening done in their various locations during the period of December 2016 to January 2017 in Cross River State, Nigeria. Blood pressure was measured using a standard mercury sphygmomanometer to obtain both the diastolic and systolic blood pressure of the various individuals. The height and weight were obtained using a weighing balance and a stadiometer respectively to get the weight and height. It was summarized age group wise and cut off points gotten from the individuals and reviewed using the WHO classification of body mass index classification. Data obtained were presented using frequencies and percentages. RESULTS: Underweight individuals (8)had a total percentage of 6.15%.A total of 46(35.40%) of the individuals were of normal weight..46 and 30 of the individuals had a percentage of 35.40% and 23.08%respectively(overweight and obese).A maximum number of the individuals were normal(were neither prehypertensive nor hypertensive). 49 and 19 individuals were prehypertensive in systolic and diastolic blood pressure respectively and 7 and 16 were hypertensive in systolic and diastolic blood pressure respectively. CONCLUSION: A high body mass index and hypertension correlate with advancing age. and can be prevented by living an acceptable lifestyle devoid of it Triggers like alcohol, smoking ,reduced salt intake etc and having a regular routine check-up done periodically.

The object of the study was oilseeds of winter camelina grown in regions with contrasting climatic conditions: the forest-steppe of the Middle Volga region (Penza Research Institute of Agriculture) and the steppe zone of Crimea (Research Institute of Agriculture of Crimea). The aim of the research was to determine the biochemical parameters of winter camelina oilseeds and to study the qualitative and quantitative fatty acid composition depending on the region of cultivation. The climate of the Middle Volga region is temperate continental. The amount of annual precipitation varies from 350 to 750 mm, the average annual temperature is 5.3 ?С. The climate of the steppe Crimea is continental, the average annual temperature reaches 10.2 ?С, the amount of precipitation per year is 350-450 mm. The fat content in camelina seeds varied slightly depending on hydrothermal parameters and averaged 39.04% and 37.81% depending on the region of cultivation. The smallest accumulation of fat (35.40 and 36.17%) was noted in drier years with HTC 0.23 (Crimea) and 0.40 (Penza). The main part in the fatty acid composition of camelina falls on polyunsaturated fatty acids, the average total content of which is 54.40-54.86%. The total content of monounsaturated fatty acids is 34.15-35.40% and saturated - 9.43-9.80%. The largest share in camelina seed oil is linolenic acid, the percentage of which was 33.79 (Crimea) and 34.87 (Penza). The concentration of linoleic and oleic acids is 16.12-17.98% and 14.82-15.70%, depending on the growing region. The content of palmitic acid is 4.93 and 5.17%, eicosene - 15.46 and 15.84 %, erucic 3.15 and 3.16%. It was found that camelina oilseeds contain tocopherols 0.085-0.098%, carotenoids 0.002%, steroids 0.144-0.145% and squalene 0.09-0.10%, depending on the region.

Informal and formal support to parents of children with disabilities affects family functioning and life satisfaction of family members in many ways. Objective. The aim of this research was to determine the extent to which parents of children with disabilities expected support of their family and friends. Methods. The sample included 65 parents of children with disabilities, most of whom were mothers (N = 62, P = 95.4%). The Social Support Behaviors - SS-B scale (Vaux et al., 1987) was used to examine the expected support of family and friends. Results. Descriptive statistics showed that parents counted on family members' support much more than on their friends' support. Parents of children with multiple disabilities, unemployed parents, and those who lived in rural areas had lower expectations regarding family and friends' support compared to other groups. Their expectations were the lowest in the segments of financial and physical support. Conclusion. Extended family members were expected to provide greater help in all five support dimensions (emotional, social, physical, financial, and counselling). The analysis of individual items within these dimensions indicates the existence of various expectation levels within the same dimension, which is the result of specific requirements.

The recent pandemic COVID-19 has become inevitably a comprehensive crisis that challenges, like a war, the very state and society functioning. The situation of global instability and fear puts a logical question: who is responsible for vital things - security and safety of citizens? The traditional answer is the national state, as it constitutes its natural function. However, in last decades this postulate has only caused controversy and extended debates among academics and practitioners who support an idea of reducing the state role and loosing some of its functions in globalizing world in favour of civil society as well as transand supranational institutions. The principle purpose of this article is to reveal objective grounds for reetatization of public life and renationalization of international relations during the period of countering pandemic and at the post-pandemic stage. The authors are also endeavoring to join discussions concerning setting the limits of state powers expanding in a crisis, as well as the validity of the absolutism in etatistic, authoritarian and paternalistic practices as universal tools of crisis management. Data were collected through observation, documentation, and literature study with quantitative content analysis method. The research was limited to the manifest function of the research object. The results showed that: (i). The most dominant of communication intimacy in the film was the intimacy at the level of stabilization (56%), followed by affective (19.5%), orientation and exploration (9.8%), and depenetration (4.9%), (ii). The most dominant of communication intimacy featured in the film was an intensive communication (22%), and followed by personal topic (stabilization) (17%), confirmation status (15%), and personal topic (exploration) (10%).Keywords: Communication Intimacy, Computer Technology, Quantitative Content Analysis, “Her” Film
