Abstract Interleukin-13 receptor alpha 2 (IL-13Rα2) is overexpressed in various types of cancers including glioma and melanoma and is known to be associated with increased tumor invasiveness and metastases. Here, we present the preclinical efficacy and toxicity results for a novel, anti-IL-13Rα2 ADC-LM-306. In vitro binding activity and internalization of LM-306 and LM-106 (unconjugated antibody of LM-306) were assessed in human IL-13Rα2 overexpressing and endogenous cell lines using flow cytometry. The antibody-dependent cell-mediated cytotoxicity (ADCC) and direct cytotoxicity were assessed using viability assays. Cross-species reactivity of drug was assessed in cells expressing cynomolgus, rat and mouse IL-13Rα2. In vivo A375 CDX model was used to assess the effect of LM-306 anti-tumor activity. The toxicity of LM-306 was evaluated in repeated-dose intravenous dose range-finding (DRF) toxicity and single intravenous maximum tolerated dose (MTD) study in cynomolgus monkeys. Both LM-106 and LM-306 showed concentration dependent binding affinity to IL-13Rα2 in various cell lines (Table 1). In addition, LM-106 also showed a dose dependent internalization and ADCC and LM-306 showed significant cytotoxicity in all cell lines (Table 1). Furthermore, LM-306 showed comparable binding affinity to human IL-13Rα2 to its unconjugated counterpart LM-106 (KD:7.43E-10 vs 1.77E-9). LM-306 was also found to be cross-reactive toward cynomolgus IL-13Rα2. In vivo, LM-306 showed tumor growth inhibition at a dose of 1, 3, 10 mg/kg given intravenously once weekly, indicating its strong anti-tumor activity. The highest non-severely toxic dose of LM-306 was determined to be 9 mg/kg in the DRF study and MTD was determined to be 18mg/kg in MTD study for male monkey. In summary, LM-306 showed high binding affinity to human IL13Rα2 expressing cells with potent cytotoxicity in vitro and significant in vivo inhibition of tumor growth, suggesting its therapeutic benefit in immuno-oncology. Background: Children in Afghanistan are at greater risk of malnutrition. Over 50% of children were suffering from malnutrition in Afghanistan in 2012, which has a long-lasting physical, mental, social, and impact on children. Study purpose: The purpose of this research study was to explore the association between parents knowledge, attitudes, and practices about complementary feeding and stunting in children in Afghanistan. The predictor variables were mothers knowledge, attitudes, and practices about complementary feeding and the outcome variable was stunting status of children aged 6 to 24 months. Research methods:A quantitative cross-sectional survey was conducted with randomly selected participants who were living in Kabul, Afghanistan. Logistic regressionmodels were used to analysethe data. Research setting: This research was conducted in 6 hospitals in Kabul, Afghanistan. Study participants: The study participants included 306 mothers who had children aged between 6-24 months. Results: Mothers knowledge, attitudes, and practices were significant predictors of stunting in children, χ2 (9, N = 306) = 45.33, p < .001 χ2 (9, N = 306) = 26.71, p < .01 and χ2 (9, N = 306) = 56.97, p <.001.Mothers who did not practice responsive feeding were 7.1 times more likely to have stunted children than mothers who practiced responsive feeding. Social and policy implications: The social and policy implications of this research study include reviewing nutrition policies, investing in nutrition programs, and increasing public education and awareness in promoting appropriate complementary infant feeding practices in Afghanistan.

Objective To define the role and early results of aortic valve surgery through a right minithoracotomy. Methods A retrospective analysis was performed on 306 consecutive patients undergoing aortic valve replacement through an 8-cm right minithoracotomy in the second intercostal space. The initial experience was included. The right second and third ribs were detached from the sternum in most cases and repaired at the end of each case. Most operations were performed using anterograde and retrograde cardioplegic arrest with percutaneous femoral venous cannulation and direct aortic cannulation through the incision. Standard instruments were used with direct digital knot tying. Results Mean age was 65 ± 14 (range, 20–90) years. Aortic valve disease cause was calcific disease in 160 of 306 (52%) patients, bicuspid disease in 95 of 306 (31%) patients, and endocarditis in 9 of 306 (3%) patients. Previous cardiac surgery was present in 13 of 306 (4%) patients. Biologic prostheses were used in 240 of 306 (78%) patients. Median valve size was 23 mm. Mean clamp times and pump times were 103 ± 26 and 158 ± 35 minutes, respectively. Median postoperative length of stay was 5 days. Thirty-day mortality was found in 4 of 306 (1%) cases. There were no deep wound infections or mediastinitis. Stroke occurred in 5 of 306 (1.6%) patients, and new pacemaker required in 11 of 306 (4%) patients. Reoperation for bleeding occurred in 2 of 306 (1%) patients. Conversion to median sternotomy occurred in 15 of 306 (5%) patients caused by chest wall anatomy (n = 7), bleeding (n = 3), coronary disease (n = 2), or aortic disease (n = 3). Patients were allowed to return to driving or preoperative activity in 2 weeks. With a mean follow-up of 2.8 ± 2.2 years, one patient required reoperation for aortic root disease. Conclusions Right minithoracotomy is a safe but limited alternative to sternotomy in isolated aortic valve replacement. This approach may be particularly valuable in some higher risk, elderly patients and opens options for a hybrid approach combined with percutaneous coronary angioplasty.

General and specific combining ability variances and effects for seven fodder characters were studied in, bajra-napier interspecific hybrids obtained in line x tester programme involving 9 CMS lines of bajra as female and 6 genotypes of napier grass, as male. The mean squares due to general and specific combining ability were significant indicating the importance of both additive and dominance components. The magnitude of sca variances was greater than that of gca for all characters suggesting the predominance of non-additive gene action. The parents, 405 A, 306 A, and 732 A among the CMS lines of bajra and FD 466 and FD 435 among napier grass were identified as best general combiners. The crosses 306 A x FD 466, 306 Ax FD 435, 437 were the best specific combinations for forage yield. High heterosis observed for different forage yield characters was mostly due to the desirable sca effects.

Resumen
Enzymatic additives, particularly alkaline proteases, play a crucial role in enhancing detergent effectiveness against protein-based stains. Despite advancements in enzyme stabilization techniques, there is a need for innovative strategies to further improve protease stability in laundry detergents. However, research exploring the utilization of substrate imprinting technology to achieve this objective remains limited. Therefore, this study aims to enhance the stability of alkaline proteases in laundry detergents by employing casein as an imprinting substrate and utilizing transglutaminase-mediated (TGase) crosslinking to modify proteases 102 and 306. The optimal temperature, pH, and thermal stability of the modified alkaline proteases 102 and 306 showed no significant changes. However, these two modified alkaline proteases exhibited varying degrees of improvement in stability among the 14 detergent additives tested. Under 40 °C incubation for 24 h, the relative enzyme activity of modified alkaline protease 102 increased approximately 1.4–15-fold in AEO-9, BS-12, CMI, APG, FMEE, and SOE, while the relative enzyme activity of modified alkaline protease 306 increased approximately 1.2–3.7-fold across different additives (FMEE, AEO-9, BS-12, SOE, FAA, and AEC-9Na). These modified proteases demonstrated improved stability and wider applicability across commercial detergent formulations available. Integrated into standard laundry detergent at a 1:7 ratio before and after modification, they effectively removed protein stains from the cotton fabric after 24 h of 40 °C incubation. These findings provide insights into more effective stain-removal techniques.

Context.— The Bethesda System for Reporting Thyroid Cytopathology recommends against the use of intraoperative frozen section (FS) during lobectomy of a thyroid nodule with a fine-needle aspiration (FNA) diagnosis of malignant. Bethesda recommendations for FS in the FNA category of suspicious for malignancy (SFM) is less well-defined. In some institutions in China, FS examination is performed during lobectomy even for FNA-proven malignant cases. Objective.— —To compare the efficacy of FNA versus FS in the evaluation of malignant thyroid lesions. Design.— A 3-year retrospective analysis from a single institution was performed on cases with an FNA diagnosis of SFM or malignant with subsequent FS examination during thyroidectomy. The results of FNA and FS findings were compared to the final thyroidectomy pathology. Results.— —A total of 5832 thyroidectomy procedures were performed: 1265 cases had FNA and FS results available. Fine-needle aspiration gave a diagnosis of SFM to 306 cases and a diagnosis of malignant to 821 cases. Of the SFM cases, 10.5% (32 of 306) had benign/indeterminate, 4.6% (14 of 306) suspicious, and 84.9% (260 of 306) malignant FS results. Final pathology showed 56.3% (18 of 32), 64.3% (9 of 14), and 100% (260 of 260) malignancy rates, respectively. For the malignant FNA group, 10.0% (82 of 821) had benign/indeterminate, 4.4% (36 of 821) suspicious, and 85.6% (703 of 821) malignant FS results. The final pathology showed 96.4% (79 of 82), 97.2% (35 of 36), and 99.9% (702 of 703) malignancy rates, respectively. Conclusions.— Frozen section should not be performed for the malignant FNA category because FS evaluation may result in 10% falsely negative findings. Performing FS for SFM may be better justified; however, more than half of FS cases read as benign in this category had malignant final pathology. Therefore, caution should be taken for FS results even in the SFM group.

This study explores borderlands as a function of the imposition of the post-colonial state upon primary structures of identity, polity and social organisation which may be sub-state, national or trans-state in nature. This imposition, particularly in the postcolonial experience of Asia, manifests itself in incongruence between identities of nation and state, between authority and legitimacy, and between beliefs and systems, each of which is most acutely demonstrated in the dynamic borderlands where the competition for influence between non-state and state centres of political gravity is played out. The instability in borderlands is a product of the re-territorialisation of pre-state primary structures, and the state's efforts in accommodating, assimilating or suppressing these structures through a combination of militarisation, providing opportunities for greater political enfranchisement, and the structure of trans-borderland economic flows. The Pashtun tribes of the Afghan borderland between Pakistan and Afghanistan are exhibiting a resurgence of autonomy from the state, as part of the re-territorialisation of the primary substructure of Pakhtunkhwa that underlies southern Afghanistan and north-western Pakistan. This phenomenon is localised, tribally driven, and replicated across the entirety of Pakhtunkhwa. It is a product of the pashtunwali mandated autonomy of zai from which every kor, killi and khel derives its security, and through the protection of which each is able to raise its nang, and is able to realise its position within the larger clan or tribe. Other examples of competition between postcolonial states and primary structures are the Kurdish experience in south-eastern Turkey and the experience of the Arab state. While manifesting significant peculiarities, all three cases - the Kurds, the Arabs and the Pashtuns - demonstrate that the current configuration of the postcolonial state system in Asia is a fragile construction, imposed upon enduring, pre-state primary structures which are resurgent through competition with the state.

Through the lens of identity process theory, the present thesis explores: (i) the qualitative nature of British national and ethnic attachments and their respective outcomes for identity processes among British South Asians (BSA); and (ii) the impact of media representations for identification and identity processes. In study I, 20 first generation South Asians (FGSA) were interviewed regarding identity, national and ethnic group memberships and inter-ethnic relations. The results revealed that (i) social representations of the ethnic 'homeland' could accentuate national attachment, but that both national and ethnic identities could have positive outcomes for identity processes in distinct social contexts; (ii) the phenomenological importance of 'special moments' and family identity can shape and accentuate national identification; (iii) ethnic and national identities are strategically 'managed' in order to achieve psychological coherence. In study II, 20 second generation South Asians (SGSA) were interviewed regarding similar issues. The results revealed that (i) SGSAs' awareness of the hardship faced by FGSA in the early stages of migration could induce disidentification with Britishness and accentuate identification with the ethnic group; (ii) the Press may be regarded as excluding BSA from Britishness; (iii) SGSA may manifest hybridised identities to enhance psychological coherence. In study III, a sample of 50 tabloid articles regarding BSA was analysed qualitatively. The results revealed that (i) BSA are constructed as 'deviating' from self-aspects of Britishness; (ii) BSA may be represented in terms of a hybridised threat to the ethno-national ingroup. Study IV investigated some of the findings of the previous studies quantitatively. The questionnaire was administered to 215 BSA. A series of statistical analyses confirmed (i) the impact of negative media representations of one's ethnic group for identity processes; (ii) the accentuation of ethnic identity and attenuation of British national identity as a result of exposure to negative media representations; (iii) a weaker national attachment among British Pakistanis than British Indians. It is argued that levels of British national and ethnic identities will likely fluctuate in accordance with social and temporal context and that BSA will make strategic use of both identities in order to optimise identity processes.

Resumen
It has been suggested that a selective lipoxygenase deficiency may be a mechanism for hyperfunction of platelets in myeloproliferative disorders. 24 out of 60 patients with one of the myeloproliferative disorders polycythemia vera, myeloid metaplasia and myelofibrosis, essential thrombo-cythemia or chronic myeloid leukemia have been reported to be deficient for platelet lipoxygenase activity (NEJM 1982; 306:381). These patients paradoxically tended to have a greater hemorrhagic tendency rather than thrombotic episodes. We investigated platelet lipoxygenase activity in clearly defined patients with erythromelalgic, thrombotic thrombocythemia (til) (Ann IntMed 1985; 102:466) while on treatment with low dose aspirin in order to prevent platelet activation in vivo or in vitro. The production of hydroxyheptodecatrienoic acid (HHT) by cyclo-oxygenase and 12-hydroxy-eicosatetraenoic acid (12-HETE) by lipoxygenase in 14C labeled platelets after stimulation with thrombin or arachidonic acid were estimated in 12 ETT patients and in 3 patients with reactive thrombocytosis (RT). HHT and 12-HETE were measured with the technique of High Performance Liquid Chromatography. Platelet lipoxygenase activity was completely normal in the 3 patients with RT and in 9 of 12 patients with ETT. Three ETT patients were severely deficient for platelet lipoxygenase activity. Two of them had progressive myeloproliferative disease with very high platelet counts, many large and some giant platelets, a few megakaryoblasts and chromosomal abnormalities of bone marrow cells (trisomy of chromosome 9 and 18p+, 1P-, 11a- respectively). Normal bone marrow karyograms were found in the 9 ETT patients with normal platelet lipoxygenase activity and stable disease. In conclusion platelet lipoxygenase deficiency is not a feature of ETT, but may reflect abnormal megakaryopoesis in progressive myeolo-proliferative disorders.
