Resumen
A new isovaleronitrile diglycoside, named microcarposide (1), together with 6 known compounds: lupeol (2), betulinic acid (3), β-sitosterol glucoside (4), methyl gallate (5), luteolin (6), and epicatechin (7), was isolated from the methanolic extract of the fruits of Detarium microcarpum Guill. Perr. The structures of the compounds were determined by extensive analysis of 1D- and 2D-1H and 13C NMR spectroscopic data in conjunction with mass spectrometry and by comparison with data reported in the literature. Compound 1 was characterized as (2 R)-2-[(6″- O-β-l-rhamnopyranosyl-β-d-glucopyranosyl)oxy]-3-methylbutanenitrile. Some of the isolated compounds were evaluated for their antibacterial activities against several microorganisms; only compound 1 was active against Salmonella typhi, Salmonella enteritidis, and Salmonella typhimurium with minimum inhibition concentration values of 153.4, 76.7, and 76.7 μM, respectively.

Cardiac muscle adapts well to changes in loading conditions. For example, left ventricular (LV) hypertrophy may be induced physiologically (via exercise training) or pathologically (via hypertension or valvular heart disease). If hypertension is treated, LV hypertrophy regresses, suggesting a sensitivity to LV work. However, whether physical inactivity in nonathletic populations causes adaptive changes in LV mass or even frank atrophy is not clear. We exposed previously sedentary men to 6 ( n = 5) and 12 ( n = 3) wk of horizontal bed rest. LV and right ventricular (RV) mass and end-diastolic volume were measured using cine magnetic resonance imaging (MRI) at 2, 6, and 12 wk of bed rest; five healthy men were also studied before and after at least 6 wk of routine daily activities as controls. In addition, four astronauts were exposed to the complete elimination of hydrostatic gradients during a spaceflight of 10 days. During bed rest, LV mass decreased by 8.0 ± 2.2% ( P = 0.005) after 6 wk with an additional atrophy of 7.6 ± 2.3% in the subjects who remained in bed for 12 wk; there was no change in LV mass for the control subjects (153.0 ± 12.2 vs. 153.4 ± 12.1 g, P = 0.81). Mean wall thickness decreased (4 ± 2.5%, P = 0.01) after 6 wk of bed rest associated with the decrease in LV mass, suggesting a physiological remodeling with respect to altered load. LV end-diastolic volume decreased by 14 ± 1.7% ( P = 0.002) after 2 wk of bed rest and changed minimally thereafter. After 6 wk of bed rest, RV free wall mass decreased by 10 ± 2.7% ( P = 0.06) and RV end-diastolic volume by 16 ± 7.9% ( P = 0.06). After spaceflight, LV mass decreased by 12 ± 6.9% ( P = 0.07). In conclusion, cardiac atrophy occurs during prolonged (6 wk) horizontal bed rest and may also occur after short-term spaceflight. We suggest that cardiac atrophy is due to a physiological adaptation to reduced myocardial load and work in real or simulated microgravity and demonstrates the plasticity of cardiac muscle under different loading conditions.

Objective: to analyze the association between medications intake and the development of type 2 diabetes mellitus (T2DM) in patients with gout. Material and methods. The study included 444 patients with gout without T2DM. The median follow-up time was 5.9 [2.9; 8.7] years. The primary end point was the diagnosis of T2DM. At baseline, therapy was initiated or adjusted according to current guidelines. Medication use was recorded: allopurinol, febuxostat, diuretics, glucocorticoids (GC), canakinumab, for which the odds ratio (OR) of developing T2DM was calculated. Results and discussion. T2DM occurred in 108 (24.3 %) patients enrolled in the study. 405 patients completed the study. 311 (76.7 %) patients were taking urate-lowering drugs: 263 (90.7 %) allopurinol, 48 (9.3 %) febuxostat. The mean dose of allopurinol was 153.4 ± 28.4 mg/day, and that of febuxostat was 91.6 ± 12.1 mg/day. During treatment with febuxostat, the probability of developing T2DM was lower: OR 0.433 (95 % confidence interval, CI 0.188–0.996; p = 0.044). When diuretics were used OR was 2.212 (95 % CI 1.303–3.753; p = 0.003), GC – 1.566 (95 % CI 1.003–2.445; p = 0.048). Conclusion. Febuxostat use is associated with a lower likelihood of developing T2DM.

The objectives of this work were the study of the volatile chemical composition of essential oils (EO’s) from Swinglea glutinosa, as well as to evaluate their antioxidant, repellent and fumigant properties. The EO was obtained by hydrodistillation from the peel of the fruit, gathered in the city of Cartagena, Bolívar (Colombia). The volatile composition was analyzed by gas chromatography coupled to mass spectrometry (GC-MS). The major compounds found in S. glutinosa were germacrene D (4.8%), limonene (5.2%), a-terpineol (6.5%), b-pinene (8.5%), nerolidyl acetate (9.8%), and trans-nerolidol (34.6%). S. glutinosa showed antioxidant potential (85.8%) (IC50=142.49 µg mL-1). The EO deployed repellent activity against the Tribolium castaneum weevil at a concentration of 15.73 nL cm-1 at 2 hours of exposure (72%), while the result for the commercial repellent was 50% at the same concentration. EO from S. glutinosa displayed the best fumigant activity with LC50 of 153.4 μg mL-1 air. The essential oil from S. glutinosa can be considerated as a natural source of biocides and antioxidants.

Objectives: To evaluate the association of hypertension and diabetic retinopathy in type 2 DM patients. Study Design: Case Control study. Setting: Diabetic Clinic Department of Medicine, Independent University Hospital, Faisalabad. Period: From January 2019 to June 2019. Material & Methods: One Hundred Type 2 diabetic patients of either sex were reviewed and these patients were screened for diabetic retinopathy using welchallyn ophthalmoscope. The patients with diabetic retinopathy fulfilling the inclusion criteria were placed in group A (cases) and patients without diabetic retinopathy were placed in group B (control). After resting the patient in supine position for 5 minutes, blood pressure (BP) measurements were taken in all these patients using mercury sphygmomanometer in two successive out-patient department visits, and mean value of B.P. was taken. All patients were asked for duration of diabetes, hypertension and visual problems specifically. These patients were advised following investigations: - FBS. - RBS. - HbAlc. Relevant statistics, mean & standard deviation were computed for variables. T-test was applied on hypertension (independent) and DM retinopathy (dependent) variables. Results: Systolic and diastolic blood pressure were significantly higher in the patients with retinopathy (mean systolic B.P 153.4± 17.13 and mean diastolic B.P. 84.1±9.26) than in those without retinopathy (mean systolic B.P. 130.65±11.94 and mean diastolic B.P 77.3 ±6.64). There was significant correlation of diabetic retinopathy with systolic hypertension (P<0.05) and diastolic hypertension (P<0.001). Conclusion: There is strong association between diabetic retinopathy and hypertension. So early detection and treatment of hypertension can retard the development and progression of diabetic retinopathy.

O-alkyl monothiocarbonate (monoxanthate) derivatives of tin were obtained by the reaction of a sodium salt of the monothiocarbonic acid with an organotin chloride to give Ph3Sn[SCO2R], Ph2Sn[SCO2R]2, and Me3Sn[SCO2R], where R = Me and i-Pr. The compounds have been characterized by infrared, Raman, 1H NMR, and 13C NMR spectroscopy, as well as mass spectrometry, and in two cases by X-ray crystallography. Ph3Sn[SCO2Me] (1) and Ph3Sn[SCO2(i-Pr)] (2), crystallize in the triclinic space group P[Formula: see text] (no. 2) with cell parameters a = 10.218(4), b = 10.568(6), c = 9.366(7) Å, α = 106.73(5), β = 96.99(5), γ = 85.55(4)°, V = 960(1) Å3, and Z = 2 for 1; and a = 14.793(2), b = 17.856(3), c = 9.813(3) Å, α = 103.86(5), β = 98.36(5), γ = 106.85(4)°, V = 2343(1) Å3, and Z = 2 for 2. The latter has two molecules in the asymmetric unit. The immediate environment about tin in both 1 and 2 is that of the expected distorted tetrahedron. However, the orientation of the monothiocarbonate group is such that there is an Sn-O intramolecular interaction of 3.040(8) for 1 and 3.05(2) Å on average for 2. Thus, the considerable distortion is consistent with a tendency to form a five-coordinate, trigonal bipyramidal species with one of the O-Sn-C angles approaching 180o (153.4(4) for 1 and an average of 157.1(6) for 2). Estimations of the Pauling partial bond orders suggest this weak Sn-O interaction is slightly stronger than the corresponding Ge-O interaction in the analogous germanium derivative, Ph3Ge[SCO2Me].Key words: structure, tin, methyl, phenyl, isopropyl, monothiocarbonates.

Lungs from fetal guinea pigs (62 ± 2 days of gestation) were supported in vitro for 3 h, and lung liquid production was measured by dye dilution. Eighteen untreated preparations produced fluid at 1.76 ± 0.30 mL∙kg−1 body weight∙h−1 during the first hour, with no significant changes in later hours. When inhibitors of respiratory processes were placed in the outer saline during the middle hour, production changed significantly, as follows: (a) sodium iodoacetate at 10−3 M stopped production (87.2 ± 10.3 and 100% reductions, successive hours; n = 6), at 10−4 M it reduced production (60.0 ± 10.3 and 63.4 ± 9.3% reduction, successive hours; n = 12); (b) sodium fluoride, 10−3 M, almost stopped production (93.2 ± 12.1 and 89.5 ± 9.3% reductions, successive hours; n = 6); (c) sodium cyanide at high concentration (10−3 M) reduced production slowly (35.5 ± 12.3 and 73.1 ± 22.4%, successive hours; n = 6); (d) sodium azide, 10−3 M, also reduced production (67.6 ± 14.2 and 59.7 ± 14.0%, successive hours; n = 6); total lactate lost rose 1.8 ± 0.5 fold; (e) dinitrophenol produced strong reabsorptions; at 10−3 M, production fell 115.4 ± 15.9 and 113.1 ± 47.3%, successive hours (n = 4), and at 2 × 10−4 M it fell 143.8 ± 33.8 and 153.4 ± 26.7%, successive hours (n = 6); total lactate lost rose 2- to 3-fold. Control preparations showed no significant changes. The results suggest that lung liquid production requires glycolysis and aerobic metabolism. However, reabsorption appears to continue on glycolysis alone, a particularly useful situation for neonates suffering respiratory distress.Key words: fetus, lung liquid, metabolic inhibitors.

Objective: This study was aimed to isolate and identify the chemical compounds from the methanol extract of the stems of Etlingera calophrys as well as to evaluate their potential as free radical scavengers. Methods: The isolated compounds were obtained using vacuum liquid chromatography (VLC) and radial chromatography techniques. The structures of these compounds were determined on the basis of their measured spectroscopic evidence (infrared, one dimentional (1D), and 2D nuclear magnetic resonance) and by comparison with literature data. The free radical scavenging activity of the methanol extract of the stems and the isolated compounds were measured using qualitative and quantitative 2,2-diphenyl-1-picrylhydrazyl (DPPH) assays. Results: Yakuchinone A (1), p-hydroxybenzoic acid (2), and stigmasterol (3) were successfully isolated and identified from the stems of E. calophyrs. The methanol extract and compounds 1–3 showed free radical scavenging activity with half maximal inhibitory concentration (IC50) values of 38.36, 39.07, 46.60, and 153.4 μg/mL, respectively, as compared to ascorbic acid with an IC50 value of 10.53 μg/mL. Conclusions: Yakuchinone A (1) and p-hydroxybenzoic acid (2) are first reported from the genus Etlingera. The presence of these compounds may contribute to the free radical scavenging activity of the methanol extract of the stems of E. calophrys. This finding suggested that E. calophrys may serve as a source of natural antioxidant compounds and could be used in the development of antioxidant herbal preparation.

Solid-state batteries (SSBs) are an ideal next-generation energy storage system due to their safety and high energy density. However, many interfacial problems, such as high interfacial resistance and poor compatibility between electrodes and electrolytes, limit the development of SSBs. In this study, an ultrathin interfacial layer composed of poly(ethylene oxide) (PEO) and ethylene carbonate (EC) are in situ fabricated on the surface of electrodes, integrating ceramic/polymer composite electrolyte and electrodes and reducing the interfacial impedance. Meanwhile, the excellent chemical compatibility between PEO and lithium metal effectively improves the interfacial stability. Also, the modification of the interfacial layer leads to more F- participation in the formation of solid electrolyte interphase (SEI), and the relative content of LiF, Li3N, and other by-products increases, which facilitates the formation of dense and stable SEI, thus inhibiting the growth of dendritic lithium and improving the ionic conductivity. With the interfacial layer, a highly stable interface against Li is maintained for more than 1400 h at a current density of 0.1 mA cm−2. A solid-state battery with LiNi0.5Co0.2Mn0.3O2 (NCM523) as the cathode delivers a capacity of 153.4 mAh g−1 and a capacity retention of 92.1% after 100 cycles. This study shows that the interfacial layer PEO-EC is an effective way to improve the performance of SSBs.

Laboratory studies were conducted to examine the leaf surface, epicuticular wax content, and spray droplet behavior on common lambsquarters, common purslane, and velvetleaf. Adaxial and abaxial leaf surfaces were examined using scanning electron microscopy, and leaf wax was extracted and quantified for all three weed species. The spread of 1-μl droplets of distilled water, primisulfuron solution (without surfactant), primisulfuron solution with a nonionic low foam wetter/spreader adjuvant (0.25% v/v), and with an organosilicone wetting agent (0.1% v/v) was determined on the adaxial leaf surfaces of each of the weed species. Glands and trichomes were present on both the adaxial and abaxial leaf surfaces of velvetleaf. Common purslane had neither glands nor trichomes on either side of the leaf. Common lambsquarters did not have any glands or trichomes, but it had globular bladder hairs on both adaxial and abaxial leaf surfaces. Stomata were present on both adaxial and abaxial leaf surfaces in all three weed species. Common purslane had a much lower number of stomata per unit area of leaf as compared with velvetleaf or common lambsquarters. Common lambsquarters had the highest epicuticular wax content on the leaf surface (274.5 μg cm−2), followed by common purslane (153.4 μg cm−2) and velvetleaf (7.4 μg cm−2). There were no significant variations in the spread of the 1-μl droplet of distilled water and primisulfuron (without adjuvant) among the species. Spread of primisulfuron droplets with surfactant was highest on the leaf surface of velvetleaf that had the lowest wax content. Droplet spread was greatest with organosilicone surfactant followed by the nonionic surfactant.

(1) Background: There is evidence in the recent literature that the incidence patterns of colorectal cancer (CRC) have changed considerably over the years, tending to rise rapidly in individuals under 50 years old compared with those over 50 years. The current study aimed to assess the incidence of CRC in Crete from 1992–2021 and compare them among younger and older adults. (2) Methods: Data on malignant neoplasms of colon, rectosigmoid junction, and rectum have been extracted from the database of the Regional Cancer Registry of Crete. (3) Results: The number of these cases for the period 1992–2021 was 3857 (n = 2895 colon and n = 962 rectum). The mean age-specific incidence rate (ASpIR/100,000/year) of colon cancer patients <50 years was 7.2 (95% CI 5.1–9.7), while for patients ≥50 years the ASpIR was 149 (95% CI 146.2–153.4). ASpIR presented a 29.6% increase from 2001 to 2011 in the age group of 20–34 years and further increase is expected from 2022–2030 (projected change, 42.8%). The main risk factors were the pack years (p = 0.01), alcohol consumption (0.02), and farmer occupation (0.04), especially during 2012–2021. (4) Conclusions: We confirmed an increased incidence of CRC in young adults <50 in a European population with low cancer incidence in the past and a worrisome prediction for the near future. The observed trends clearly indicate that starting CRC screening at an earlier age may be essential.

Abstract Rogowski is a special current transformer and has been categorised as a low-power stand-alone current sensor. This sensor is designed by using a Saw blade pattern with 50 turns and 100 turns. This number of turns will impact the increment of the sensitivity of the sensor. Rogowski coil is used for measuring the alternating current (AC) and high-speed current pulses. The PCB Rogowski Coil will detect the arcing fault that design with AutoCAD software after that import to CST software. An arc is created when the electrical current jumps the gap between two conductive materials. The most common causes of arcs include worn contacts in electrical equipment, damage to insulation, break in a cable and loose connections. In order to reduce the arc fault risk, the arc fault interrupter (AFI) technology and arc fault detectors (AFD) have been developed. However, these devices are most suitable for domestic application or low voltage (LV). This paper proposed a simulation using AutoCAD and CST software to design a Rogowski coil and the Rogowski Coil sensor will be tested by using CST software in terms of radiation pattern. The biggest radiation pattern in red colour is the saw blade pattern in 100 turns at 2 KHz frequency, the radiation pattern covering entire sensor area compared with other radiation patterns with other frequency. The main lobe magnitude for 2 KHz frequency is 46.8 dB, main lobe direction is 180 deg, angular width is 153.4 deg and the side lobe level is zero. This is the best number of turn and frequency compared to 50 turn saw blade sensor.

The synthesis of the triphenylmethyl-phosphonic and -phosphonothioic dihalides Ph3CP(:X)Y2 (X = O, S; Y = F, Cl) is described. The NMR parameters of these compounds are discussed and compared to these of other phosphonic and thiophosphonic dihalides. Single crystal X-ray analysis was used to confirm the structure of Ph3CP(:O)F2 2 [C2/c, a = 1599.6(4), b = 1096.3(3), c = 1863.1(7) pm, ß = 109.11(2)°, T = 178 K, R = 0.034]. The geometry at the phosphorus atom is approximately tetrahedral. The bonds between phosphorus and fluorine are extremely short [P— F 153.1(1) and 153.3(1) pm]. Bond distances and angles for this compound are compared to those of methylphosphonic difluoride which were obtained from microwave spectra.

Object. This study was conducted to determine whether perfluorocarbons (PFCs) improve brain oxygenation and reduce ischemic brain damage in an acute subdural hematoma (SDH) model in rats. Methods. Forty adult male Sprague—Dawley rats were allocated to four groups: 1) controls, acute SDH treated with saline and 30% O2; 2) 30-PFC group, acute SDH treated with PFC infusion in 30% O2; 3) 100-O2 group, acute SDH treated with 100% O2; and 4) 100-PFC group, acute SDH treated with PFC plus 100% O2. Ten minutes after the induction of acute SDH, a single dose of PFC was infused and 30% or 100% O2 was administered simultaneously. Four hours later, half of the rats were killed by perfusion for histological study to assess the extent of ischemic brain damage. The other half were used to measure brain tissue oxygen tension (PO2). The volume of ischemic brain damage was 162.4 ± 7.6 mm3 in controls, 165.3 ± 11.3 mm3 in the 30-PFC group, 153.4 ± 17.3 mm3 in the 100-O2 group, and 95.9 ± 12.8 mm3 in the 100-PFC group (41% reduction compared with controls, p = 0.002). Baseline brain tissue PO2 values were approximately 20 mm Hg, and after induction of acute SDH, PO2 rapidly decreased and remained at 1 to 2 mm Hg. Treatment with either PFC or 100% O2 improved brain tissue PO2, with final values of 5.14 and 7.02 mm Hg, respectively. Infusion of PFC with 100% O2 improved brain tissue PO2 the most, with a final value of 15.16 mm Hg. Conclusions. Data from the current study demonstrated that PFC infusion along with 100% O2 can significantly improve brain oxygenation and reduce ischemic brain damage in acute SDH.

SUMMARY INTRODUCTION: Obesity is a growing public health problem associated with many comorbid diseases. The aim of this study was to evaluate the relationship between body mass index and complications of thyroidectomy. METHODS: Patients who underwent total thyroidectomy between January 2015 and December 2018 were enrolled. Patients were divided into two groups, i.e., BMI <25 (group A) and BMI≥ 25 (group B). Demographics, operative time, and complications were retrospectively reviewed. RESULTS: The study included 145 patients (66 in Group A and 79 in Group B). There was no significant difference between the two groups in terms of age (p=0.330) and gender (p=0.055). No surgical site infection and bleeding complications were observed in any patients. The mean operative time was 148.4 minutes (90-235) in Group A and 153.4 minutes (85-285) in Group B (p=0.399). Transient hypocalcemia was observed in 25 (37.9%) patients in group A, and 23 (29.1%) patients in Group B (p = 0.291). Permanent hypocalcemia was not observed in any patient in group A, and in 2 patients in Group B (2.5%) (p = 0.501). Transient recurrent nerve palsy was observed in 1 (1.5%) patient in Group A and in 3 (3.8%) patients in Group B (p=0.626). None of the patients had permanent recurrent nerve palsy. Parathyroid autotransplantation was performed on 1 patient (1.5%) in group A and on 7 (8.9%) patients in Group B (p=0.055). CONCLUSION: We think there is no relationship between a high BMI and thyroidectomy complications, and surgery can also be performed safely in this patient group.

This study investigated whether the putative physiological benefits induced by growth hormone (GH) and insulin-like growth factor-1 (IGF-1) are countered at supra-physiological concentrations because of an augmentation in the production of mitochondrial-derived free radicals with a subsequent increase in oxidative damage, compromising mitochondrial function. To test this hypothesis, peripheral blood mononuclear cells were incubated for 4 h with either recombinant human GH (rhGH) (range = 0.25–100 μg/L) or recombinant IGF-1 (rIGF-1) (range = 100–600 μg/L) and along with control samples were subsequently analyzed by flow cytometry for the determination of cellular viability, mitochondrial membrane potential (Δψm), mitochondrial superoxide (O2–) generation, and mitochondrial permeability transition pore (mtPTP) activity. Results showed levels of mitochondrial O2– generation to be significantly reduced compared with control samples (lymphocytes: 21.5 ± 1.6 AU; monocytes: 230.2 ± 9.8 AU) following rhGH treatment at both concentrations of 5 μg/L (13.5 ± 1.3 AU, P ≤ 0.05) and 10 μg/L (12.3 ± 1.5 AU, P ≤ 0.05) in lymphocytes and at 10 μg/L (153.4 ± 11.4 AU, P ≤ 0.05) in monocytes. However, no significant effect was found at either higher rhGH concentrations or following treatment with any concentration of rIGF-1. In addition, neither of the 2 hormones had any significant effect on Δψm, mtPTP activity, or on cellular viability. In conclusion, physiological concentrations of rhGH elicited a protective cellular effect through the reduction of oxidative free radicals within mitochondria. This antioxidant effect was diminished at supra-physiological concentrations but not to a level that would elicit disruption of mitochondrial function.

AbstractAs one economically important fish in the southeastern Himalayas, the giant devil catfish (Bagarius yarrelli) has been known for its extraordinarily large body size. It can grow up to 2 m, whereas the non-Bagarius sisorids only reach 10–30 cm. Another outstanding characteristic of Bagarius species is the salmonids-like reddish flesh color. Both body size and flesh color are interesting questions in science and also valuable features in aquaculture that worth of deep investigations. Bagarius species therefore are ideal materials for studying body size evolution and color depositions in fish muscles, and also potential organisms for extensive utilization in Asian freshwater aquaculture. In a combination of Illumina and PacBio sequencing technologies, we de novo assembled a 571-Mb genome for the giant devil catfish from a total of 153.4-Gb clean reads. The scaffold and contig N50 values are 3.1 and 1.6 Mb, respectively. This genome assembly was evaluated with 93.4% of Benchmarking Universal Single-Copy Orthologs completeness, 98% of transcripts coverage, and highly homologous with a chromosome-level-based genome of channel catfish (Ictalurus punctatus). We detected that 35.26% of the genome assembly is composed of repetitive elements. Employing homology, de novo, and transcriptome-based annotations, we annotated a total of 19,027 protein-coding genes for further use. In summary, we generated the first high-quality genome assembly of the giant devil catfish, which provides an important genomic resource for its future studies such as the body size and flesh color issues, and also for facilitating the conservation and utilization of this valuable catfish.

ABSTRACT OBJECTIVE: To evaluate the effects of aliskiren on blood pressure and myocardial functionassessed by global longitudinal strain in patients with uncontrolled arterial hypertension. PATIENTS AND METHODS: Forty-fi ve patients were included in the study (29 males, 16 females, mean age 58.7 ± 12.4 years) with BP > 140/90 mmHg despite treatment with combined antihypertensive therapy and echocardiographic data for diastolic dysfunction: Е/Е’ratio ≤ 8, Е/А ratio < 0.8, deceleration time (DT) > 200 msec. Aliskiren (2 х 150 mg per day) was added to the previous therapy. The follow-up period was 1 year, including monthly clinical visits. Echocardiographic assessment of the left ventricular function by longitudinal strain and Doppler analysis of the trans-mitral blood flow was performed at months 1, 6, 12. RESULTS: The baseline systolic and diastolic blood pressures scores were 153.4 ± 14.4/99.2 ± 6.7 mmHg and 157.6 ± 12.5/97.3 ± 8.2 mmHg for males and females, respectively. The systolic and diastolic values at 1 month were 131.7 ± 7.4/83.6 ± 5.2 mmHg for males and 132.4 ± 5.3/81.8 ± 6.9 mmHg for females (р < 0.05 vs. baseline). The baseline Е/Е’ was 6.5 ± 0.9, Е/А - 0.6 ± 001, DT - 258 ± 32.7 msec. These indicators at month 12 were as follows: Е/Е’ - 7.0 ± 0.64, Е/А - 0.7 ± 0.05, DT - 239 ± 16.5 msec, р = NS. Baseline global longitudinal strain in males was -10.4 ± 0.7% and -11.0 ± 0.9% in females and at month 12 - 16.3 ± 0.9% and -17.5 ± 0.7% for males and females, respectively, р < 0.05. For the period of follow-up no adverse effects due to aliskiren treatment were registered. CONCLUSIONS: Adding aliskiren to combined antihypertensive therapy leads to significant improvement of hypertension control and myocardial function assessed by global longitudinal strain.

Objective To investigate the performance of different b values and regions of interest (ROI) for diagnosing liver fibrosis in patients with chronic viral hepatitis by using diffusion-weighted (DW) magnetic resonance imaging (MRI). Methods Eleven healthy participants and 33 patients with viral hepatitis B or C were enrolled. The stage of liver fibrosis and the grade of necroinflammation were determined by using a histologic activity index. Single-shot spin-echo echo-planar DW-MRI was performed in all participants at b values of 0-500, 0-700, and 0-1000 s/mm2 by using 2 circular small and large ROIs of 100 and 200 mm2. To evaluate the performance of different b values for determining cirrhosis, the receiver-operating characteristic curves were depicted, and the areas under the curves were compared. Results The average values of apparent diffusion coefficients significantly decreased with increasing stage or grade categories at all the 3 b values and for both small and large ROIs. The performance at b = 500 s/mm2 was significantly better than b = 1000 s/mm2 for determining cirrhosis or bridging fibrosis. The cut point of 153.4 for apparent diffusion coefficient (×10−5 mm2/s) at b = 500 s/mm2 could determine cirrhosis or bridging fibrosis with a sensitivity of 96% and specificity of 82%. No difference was found between the average apparent diffusion coefficient values of large or small ROIs. Also, there was no difference in performance of large or small ROIs in the diagnosis of liver fibrosis. Conclusions This study provided beneficial data for clinical utilisation of DW-MRI in diagnosing liver fibrosis: b = 500 s/mm2 is better in performance than b = 1000 s/mm2, and a small ROI of 100 mm2 is sufficient for determining cirrhosis or bridging fibrosis.

Insufficient data exist to evaluate the comparative effects of inhaled corticosteroids (ICS) versus leukotriene receptor antagonist (LTRA) on airway inflammation and quality of life (QoL). The aim of the study was to compare the effectiveness of montelukast compared to budesonide at different doses on QoL and bronchial reactivity in mild-asthmatic adult patients. 45 subjects with bronchial asthma were randomly assigned to a different treatment and divided in 3 treatment groups: A: 400 μg of budesonide twice a day; B: 10 mg of montelukast daily; C: 10 mg of montelukast daily plus 400 μg of budesonide twice a day. At the beginning of the study and at the end of the treatment period (16 weeks) all patients underwent complete clinical evaluation, pulmonary function testing and methacholine challenge test (MCHt). In group A the increase from baseline was 153.4%, in group C was 133.2%, and in group B 247.7%, the latter increase being statistically significant compared to that in the other 2 groups (p< 0.005 Wilcoxon test). In all domains the improvement in quality of life in the group treated with montelukast (group B) was significantly greater than that in the group treated with both medications (group C): in particular, the improvement was consistent in the symptoms (p< 0.01) and emotions (p< 0.01) domains, and weaker in the physical activity (p< 0.05). A similar difference was observed between group B and A, but only in the symptoms (p<0.01), emotions (p<0.01), and environmental stimuli domains (p<0.05). The personal perception of their own disease is important for a correct therapeutic management of asthma. In order to optimize the treatment, a complete adherence of the patient to the treatment itself is required, to be achieved through simplification of therapeutic schedule and easy administration of medications. Montelukast may be considered a valid alternative in the treatment of mild-persistent asthma, both for the clinical and functional benefits and for the great advantage of the once-daily dosage, which consistently improves the compliance with the chronic treatment of the disease.

Abstract Background: Anemia is a common condition among elderly and its prevalence increases with age. Fatigue is well recognized as a prominent feature of chronic anemia and may be especially symptomatic in this population. In order to evaluate the effect of epoetin alfa (EPO) treatment on hemoglobin (Hb), fatigue, and health related quality of life (QOL) in elderly patients (pts) with chronic anemia a 32 week(w) randomized, double-blind, cross-over treatment trial among community-dwelling participants ≥65y was conducted. Pts were ambulatory and had anemia for ≥3 months with inclusion Hb ≤11.5g/dL. Pts with GI bleeding, active cancer, CKD (GFR&lt;30), thrombocytopenia, iron deficiency, hypothyroidism, untreated depression, dementia, or expected survival ≤6months were excluded. Pts were randomized to receive either subcutaneous (sc) placebo or EPO weekly for 16w (Phase 1) and then crossed over to the other treatment (Phase 2). QOL was measured using the Functional Assessment of Cancer Therapy-Anemia (FACT-An) instrument, with specific focus on the Anemia and Fatigue subscales of the instrument. Hb was measured every 4w. Analyses of all efficacy endpoints were based on pts who had who had at least one Phase 1 (58/62) and one Phase 2 (55/58) visit (using analysis of covariance controlling for baseline values). Results: 62 pts, 85% women, and 95% African American with a mean (SD) age of 76.1y (±7.2). Mean baseline Hb was 10.5±0.9 g/dL (7.3–11.5); 75% had anemia of chronic disease and 25% had unexplained anemia. Common baseline comorbidities included hypertension, CHF, osteoarthritis, and Type 2 diabetes. Seven pts did not complete the study. There was 1 death, 1 stroke and 1 DVT on placebo and 1 death and 1 pulmonary embolus on EPO. None of the serious adverse events or deaths were considered treatment-related. Pts receiving EPO had significantly increased Hb compared to placebo at the end of phases 1 and 2 (p&lt;0.0001; Table 1) and, significantly improved scores on the composite FACT-An (p&lt;0.0107, phase 1; p=&lt;0.0001, phase 2), FACT-An anemia subscale (p=0.0017, phase 1; p=0.0003, phase 2) and FACT-An fatigue subscale (p=0.0012, phase 1; p=0.0011, phase 2). Conclusion: A direct relationship exists between increasing Hb during EPO therapy, fatigue scores and corresponding QOL improvements among elderly persons with anemia. EPO was well tolerated in this trial among anemic elderly pts. Improving fatigue may enhance ability to perform daily activities in the elderly. The study was aimed to investigate the effect of corn stalk pre-treatment duration on biogas production when cow dung and corn stalk was co-digested in an anaerobic digestion. Corn stalks were pre-treated in different durations: 2-days, 5-days, and 8-days before being added to cow dung into anaerobic co-digesters. The experiments were set up randomly by using triplicate batch anaerobic apparatus in 21 L containers that run in 60-days. The mixing ratio between a corn stalk and cow dung was 50%: 50% (based on the volatile solid value of each material), but corn stalk was cut into small pieces with around 10 cm length, while the cow dung was air dried. The results of the study indicated that all operation parameters such as temperature, pH, and alkalinity in the anaerobic batch were suitable for biogas production. The results showed that there was a significant improvement in total gas produced in the pre-treated 5-days treatment (206.4±8.4 L) compared to 2-days (153.4±9.6 L), and 8-days ones (174±11.1 L). The biogas yield of the pre-treated 2-days, 5-days, and 8-days treatments were 392.7±9.8 L/kg VSfermented, 469.8±10.1 L/kg VSfermented and 497.1±13.3 L/kg VSfermented, respectively, that was not significantly different (5%). In all treatments, low concentration of methane in the beginning phase had been observed but increased and reached the optimum value for energy use after 10 days. The result of the study showed that it is preferable to have 5-days pre-treatment of corn stalk before the corn stalk is loaded to an anaerobic digester in combination with cow dung. Các thí nghiệm được bố trí ngẫu nhiên trong các bình ủ yếm khí theo mẻ 21 L, vận hành trong 60 ngày liên tiếp và có 3 lần lặp lại. Nguyên liệu ủ được phối trộn theo tỷ lệ 50% phân bò và 50% thân bắp, trong đó thân bắp được cắt nhỏ cỡ 10 cm. Kết quả thí nghiệm cho thấy tất cả các thông số pH, nhiệt độ, độ kiềm của mẻ ủ đều phù hợp để vận hành. Lượng khí sinh ra của các nghiệm thức xử lý ở 2 ngày, 5 ngày, 8 ngày được ghi nhận là 153,4±9,6 L, 206,4±8,4 L và 174±11,1 L; năng suất sinh khí của các nghiệm thức không khác biệt và đạt giá trị 392,7±9,8 L/kg VSphânhủy, 469,8±10,1 L/kg VSphânhủy và 497,1±13,3 L/kg VSphânhủy. Tất cả các nghiệm thức đều sản sinh lượng CH4 thấp ở giai đoạn đầu nhưng tăng dần theo thời gian ủ và đạt hiệu quả sử dụng sau 10 ngày ủ. Kết quả cho thấy có thể chọn mốc thời gian 5 ngày để xử lý thân cây bắp trước khi đưa vào hầm ủ biogas.

Background:Systemic lupus erythematosus (SLE) is an autoimmune disease mainly affecting women of childbearing age. The interplay between genetic and environmental factors may contribute to disease pathogenesis1. At today, no robust data are available about the possible contribute of diet in SLE. Caffeine, one of the most widely consumed products in the world, seems to interact with multiple components of the immune system by acting as a non-specific phosphodiesterase inhibitor2.In vitrodose-dependent treatment with caffeine seems to down-regulate mRNA levels of key inflammation-related genes and similarly reduce levels of different pro-inflammatory cytokines3.Objectives:We evaluated the impact of caffeine consumption on SLE-related disease phenotype and activity, in terms of clinimetric assessment and cytokines levels.Methods:We performed a cross-sectional study, enrolling consecutive patients and reporting their clinical and laboratory data. Disease activity was assessed by SLE Disease Activity Index 2000 (SLEDAI-2k)4. Caffeine intake was evaluated by a 7-day food frequency questionnaire, including all the main sources of caffeine. As previously reported, patients were divided in four groups according to the daily caffeine intake: <29.1 mg/day (group 1), 29.2-153.7 mg/day (group 2), 153.8-376.5 mg/day (group 3) and >376.6 mg/day (group 4)5. At the end of questionnaire filling, blood samples were collected from each patient to assess cytokines levels. These were assessed by using a panel by Bio-Plex assays to measure the levels of IL-6, IL-10, IL-17, IL-27, IFN-γ, IFN-α and Blys.Results:We enrolled 89 SLE patients (F/M 87/2, median age 46 years, IQR 14; median disease duration 144 months, IQR 150). The median intake of caffeine was 195 mg/day (IQR 160.5). At the time of the enrollment, 8 patients (8.9%) referred a caffeine intake < 29.1 mg/day (group 1), 27 patients (30.3%) between 29.2 and 153.7 mg/day (group 2), 45 patients (51%) between 153.8 and 376.5 mg/day (group 3) and 9 patients (10.1%) >376.6 mg/day (group 4). A negative correlation between the levels of caffeine and disease activity, evaluated with SLEDAI-2K, was observed (p=0.01, r=-0.26). By comparing the four groups, a significant higher prevalence of lupus nephritis, neuropsychiatric involvement, haematological manifestations, hypocomplementemia and anti-dsDNA positivity was observed in patients with less intake of caffeine (figure 1 A-E). Furthermore, patients with less intake of caffeine showed a significant more frequent use of glucocorticoids [group 4: 22.2%,versusgroup 1 (50.0%, p=0.0001), group 2 (55.5%, p=0.0001), group 3 (40.0%, p=0.009)]. Moving on cytokines analysis, a negative correlation between daily caffeine consumption and serum level of IFNγ was found (p=0.03, r=-0.2) (figure 2A); furthermore, patients with more caffeine intake showed significant lower levels of IFNα (p=0.02, figure 2B), IL-17 (p=0.01, figure 2C) and IL-6 (p=0.003, figure 2D).Conclusion:This is the first report demonstrating the impact of caffeine on SLE disease activity status, as demonstrated by the inverse correlation between its intake and both SLEDAI-2k values and cytokines levels. Moreover, in our cohort, patients with less caffeine consumption seems to have a more severe disease phenotype, especially in terms of renal and neuropsychiatric involvement. The aim of this work was to evaluate the stress generated on the prosthetic intermediary in prosthetic crowns made with different materials using finite element analysis. Three-dimensional models of a portion of the maxilla with an implant and abutment screwed onto it were created. Three crowns for the first molar were constructed, and load simulations were carried out with axial (100 Newtons) and oblique (50 Newtons) forces, as follows: Model 1: prosthetic crown with a cobalt chromium infrastructure, 0.3 mm thick, and feldspathic porcelain roof; Model 2: prosthetic crown with a zirconia infrastructure on the abutment, 0.5 mm thick, and covering feldspathic porcelain; Model 3: zirconia prosthetic crown on the abutment, 1.5 mm thick. The results obtained in the external and inner portions of the intermediary were as follows: in the axial load, model M1 presented peaks in megapascals of 153.5, M2 of 153.6, and M3 of 156.1. In the oblique load, the peaks were in model M1 220.6, M2 220.7, and M3 220.5. In the inner portion of the intermediary, the peaks in megapascals in the axial load were 482.4 in the M1 model, 482.3 in the M2, and 482.3 in the M3. In the oblique load, they presented peaks of 672.8 in M1, 672.9 in M2, and 682.7 in M3. Quantitatively and qualitatively, there were no significant variations between the stress peaks in the outer and inner portions of the abutment, both in axial and oblique loads. Considering the models and methodology used, it is concluded that the mechanical performance of the different treatments analyzed is similar.

Twenty five buffalo cows of Mothbaria Upazila of Pirojpur district and another twenty five indigenous buffalo cows of Pathorghata Upazila of Borguna district of Bangladesh were randomly selected and interviewed to collect data on reproductivity (gestation length, birth weight, age of first calving, post partum heat and calving interval), productivity (lactation length and daily milk yield) and management system (general management, housing, feeding, calf rearing and treatment facilities) of indigenous buffalo cows using pre-tested questionnaire during the period from January to June 2010. The reproductive parameters including gestation length, birth weight, age of first calving, post partum heat and calving interval of Mothbaria Upazila of Pirojpur district were 319.56 ± 5.93 days, 24.28 ± 4 kilograms, 50.88 ± 1.71 months, 153.6 ± 6.13 days, 547.92 ± 10.88 days respectively whereas in case of Pathorghata Upazila of Borguna district these parameters were 319.12 ± 4.69 days, 24.12 ± 3.6 kilograms, 51 ± 1.8 months, 153.44 ± 6.78 days, 547.24 ± 14.32 days respectively. The productive parameters including lactation length and daily milk yield of Mothbaria Upazila of Pirojpur district were 286.12 ± 11.27 days and 3.33 ± 0.68 liters/day respectively while in case of Pathorghata Upazila of Borguna district these parameters 290.44 ± 10.92 days and 3.43 ± 0.744 liters/day respectively. So, reproduction and production are almost regular comparing with other published reports with the exception of age of first calving which is almost 12 months later than the buffaloes of other countries. It could be concluded that the buffalo owners of these areas do not follow any scientific method of buffalo husbandry; they just follow traditional rearing system. BackgroundPulmonary manifestations of idiopathic inflammatory myopathies (IIM) include interstitial lung disease (ILD) and respiratory muscle weakness, but function of small airways has not been studied in these patients (1). Spirometry, impulse oscillometry and measurement of respiratory resistance by the interrupter technique (Rocc) are tools to evaluate function of small airways. In addition, body plethysmography is the gold standard for measuring lung volumes, whereas nitrogen washout seems impaired, “underestimating” lung volumes in patients with small airways obstruction due to gas trapping (2). Thus, discrepancies between lung volumes measured by those two techniques may be an indication of early small airways dysfunction.ObjectivesTo compare pulmonary and small airways function between IIM patients with and without ILD.MethodsThis prospective observational study included 13 IIM patients with ILD (8 patients with dermatomyositis and 5 with polymyositis) and 13 IIM patients without ILD (9 patients with dermatomyositis and 4 with polymyositis) who were diagnosed and followed up in the outpatient rheumatology clinic of the Department of Pathophysiology between June and December 2021. All IIM patients fulfilled the 2017 EULAR/ACR classification criteria (3). The presence of ILD was determined by high resolution computed tomography of the lungs that was performed as baseline standard of care at the time of diagnosis and was evaluated by a special radiologist blindly, according to Fleischner Society definitions for ILD (4). Clinical, laboratory and immunological data were recorded at the time of diagnosis and pulmonary function was assessed by spirometry, body plethysmography, single and multiple breath nitrogen washout, impulse oscillometry and measurement of Rocc, diffusing capacity for carbon monoxide (DLco) and maximal inspiratory and expiratory pressures. Statistical analysis for categorical data was performed by Fisher exact test or χ2 square test accordingly and numerical data with Man-Whitney test or t test.ResultsIIM-ILD patients presented more frequently with dyspnea (53.8% vs 0%, p<0.01), fever (61.5% vs 7.7%, p<0.05) and arthralgias (76.9% vs 23%, p<0.05), compared to IIM patients without ILD. Inflammatory markers, serum muscle enzymes, myositis specific autoantibodies and classic spirometric parameters did not differ between the two groups. IIM-ILD patients had markedly lower predicted DLco compared to those without (mean: 56.3% vs 78.2%, p<0.01). Predicted total lung capacity and residual volume (TLCN2WO%, RVN2WO%) measured by nitrogen washout and the TLC nitrogen washout: TLC body plethysmography ratio (TLCN2WO/TLCpleth) were significantly lower in IIM-ILD patients compared to those without ILD (mean: 111.1% vs 153.4%, p<0.05, median: 171% vs 210%, p<0.05 and median: 1.28 vs 1.45, p<0.05, respectively). Similarly, ΔΝ2(%) was found significantly higher among non-ILD than ILD-IIM patients [5.4 vs 2.8%, p=0.013]. Predicted normal value of Rocc tended to be higher in IIM-ILD patients, although not statistically significantly.ConclusionAbnormal single breath nitrogen washout and discrepancies between lung volumes measured by body plethysmography and nitrogen washout in IIM-ILD patients, indicate an early small airways dysfunction in these patients.References[1]Selva-O’Callaghan et al. Abstract The transcription factor PLZF is required to drive lineage diversification of would-be conventional T cells into the natural killer T cell fate. However, the molecular machinery linking extracellular signaling to this fate choice is unknown. Recently, the early growth response 2 (Egr2) transcription factor, a calcineurin-dependent lineage resolution factor, was found to be important for NKT development. Here, we utilize an Egr2/IRES-GFP transgenic mouse model to show that Egr2 is uniquely induced immediately after TCR engagement in NKT cells. Competitive bone marrow chimeras with T cell conditional knockout mice indicate that Egr2 is required for both stage zero to stage 1, and stage 2 to stage 3 transitions of NKT development. Importantly, Egr1/Egr2 double deficient, stage 1 arrested NKT cells fail to express PLZF, in striking contrast to wild type stage 1 NKTs. Gel shift analysis shows in vitro translated Egr2 protein binds to a conserved site within the zbtb16 (PLZF) promoter. ChIP-seq of Egr2-bound chromatin from thymic NKTs confirmed in vivo binding of the zbtb16 promoter. Finally, we show that strong TCR signals are important for Egr2 induction in NKT cells. We propose that Egr2 links the NKT TCR signal with resolution of a mixed lineage state, driving PLZF expression in addition to other critical NKT lineage properties. We also suggest other receptor-dependent cell fates may be controlled by similar induction of resolution factors.

Abstract Expression of the IL-7 receptor alpha (IL-7R) is a distinguishing feature of common lymphoid progenitors (CLP) in mice. Human B cell development has been thought to differ from that in mouse with respect to the requirement for IL-7. Our previous studies show that IL-7 plays a critical role in human B cell production (J Immunol. 2009, 182:4255). Here we use IL-7R expression to identify a human CLP population that can be generated from hematopoietic stem cells (HSCs) in umbilical cord blood (CB). Following two weeks of co-culture with primary human bone marrow (BM) stroma, CB CD34+ cells give rise to a CD34+CD19-IL-7R+ progenitor population that is absent from fresh, uncultured CB. IL-7R+ progenitors make up ~17% of the CD19-CD34+ cells present in culture at 2 weeks. A similar IL-7R+ human progenitor population is generated in the BM of immunodeficient mice following transplant with human CD34+ CB cells. FACS-sorted CD34+CD19-IL-7R+ progenitors produced in co-culture or in the human-mouse xenograft model give rise to CD19+ B cells and CD56+ NK cells in bulk and single cell culture, but lack myeloid potential in CFU assays and when transplanted into immunodeficient mice. Ongoing experiments will evaluate the T lineage potential of IL-7R+ progenitors in the human-mouse xenograft model. These data will be important for developing therapies to accelerate and enhance lymphoid reconstitution from HSCs in CB, a hematopoietic source used increasingly for stem cell transplant.

The homotetrameric holo-D-glyceraldehyde-3-phosphate dehydrogenase from the hyperthermophilic archaeon Methanothermus fervidus has been crystallized in the presence of NADP+ using the hanging-drop vapour-diffusion method. Crystals grew from a solution containing 2-methyl-2,4-pentanediol and magnesium acetate. A native data set has been collected to 2.1 Å using synchrotron radiation and cryocooling. Diffraction data have been processed in the orthorhombic system (space group P21212) with unit-cell dimensions a = 136.7, b = 153.3, c = 74.9 Å and one tetramer per asymmetric unit.

Abstract Pale yellow crystals of the title compound were obtained by reacting an intimate mixture of K2S, K2S2O4, Ti and S at 650 °C. K6Ti2OS6 is monoclinic, mP60, s. g. P21/c. It is characterized by the formation of bitetrahedral complex anions, [S3TiOTiS3]6−, which adopt a staggered conformation. The mean Ti-S and Ti-O bond lengths are 2.242(1) and 1.836(2) Å , respectively, the Ti-O-Ti bond angle is 153.8(2)°. Two K+ ions complete the coordination of the bridging oxygen atom by the cations to a heavily distorted tetrahedral configuration. The anions are arranged in corrugated slabs running parallel to (100). The packing and crystallographic relationship to K6Co2O7 are discussed.

(1) Background: The purpose of this study was to investigate zinc contents in leaves and soils of the Valdepeñas Protected Designation of Origin (PDO), situated in central Spain. Zn distribution maps of leaves and soils were obtained. (2) Methods: Ninety soil profiles were described, sampled and analyzed. Furthermore, vineyard leaves were collected randomly in each of the analyzed soil vineyard profiles. Soil and leaf samples were analyzed by X-ray fluorescence. (3) Results: The mean total Zn concentrations in vineyard soils were in the range of 16.2–153.7 mg·kg−1, with a mean of 47.5 mg·kg−1. The obtained values above the 95th percentile (between 81.3 and 153.7 mg·kg−1) could be affected by different parent materials or Zn agricultural treatments in vineyards. Contents in different soils follow the order Entisol > Alfisol > Inceptisol. The average Zn content value in leaves was 23.8 mg·kg−1 and oscillated between 11.5 and 93.3 mg·kg−1; minor differences were detected between soil types, with the highest value in plants grown on soils without carbonates. (4) Conclusions: The obtained values are optimal for vine cultivation. The bioaccumulation factors in leaves were lower than unity (0.24–0.53 range). This means that the Zn bioaccumulation process is relatively low in the soil–grapevine system. This study serves as a reference to identify areas that present Zn deficiencies or risk of contamination.

Based on kinetic data of thermal gas-phase elimination reactions, the following Arrhenius log A (s–1) and Ea (kJ mol–1) values, respectively, are obtained: 10.76 and 153.5 for 3-thiopheneacetic acid (1), 10.08 and 149.4 for 2-thiopheneacetic acid (2), 12.04 and 207.1 for 2-(3-thienyl)ethanol (3), 11.55 and 203.3 for 2-(2-thienyl)ethanol (4), 10.91 and 123.4 for 2-thiopheneglyoxylic acid (5), 11.05 and 223.8 for 1-(2-thienyl)propan-1-one (6), and 10.33 and 149.8 for 3-thiophenemalonic acid (7). The products of these pyrolytic reactions were either carbon dioxide or formaldehyde in addition to methylthiophene or thiophenecarboxaldehyde. Both positional and molecular reactivities of the substrates and related compounds are compared, and the results are rationalized on the basis of a reaction pathway involving a concerted six-membered transition state.Key words: thiophenes, gas-phase, pyrolysis, kinetics, mechanism.

Abstract Introduction: For fit multiple myeloma (MM) patients, autologous hematopoietic stem cell transplant (HSCT) is standard of care as part of first line therapy, demonstrating longer progression-free survival when compared to upfront bortezomib, lenalidomide, and dexamethasone (IFM/DFCI 2009, ASH 2015). The use of granulocyte colony stimulating factor (G-CSF) after HSCT accelerates time to neutrophil recovery by 1 - 6 days when compared with control (Klumpp TR et al, JCO, 1995 & Schmitz N et al, BMT 2004). The American Society of Clinical Oncology guidelines recommend that G-CSF should be initiated 1-5 days after administration of high-dose chemotherapy and should be continued until the absolute neutrophil count (ANC) is 2000-3000/mm3 (Smith TJ, JCO, 2015). We have evaluated the role of G-CSF starting day +1, day +5, and day +7 post-transplant in three sequential cohorts of MM patients focusing on the duration of severe neutropenia (rather than the time to neutrophil engraftment), infections, and mucositis. Methods: As part of changes in the standard of care institutional protocols for autologous HSCT of myeloma patients at Ohio State University, three sequential cohorts of myeloma patients were identified that received G-CSF daily post-transplant until ANC>1500/mm3 or WBC>5/mm3. Two hundred twenty-six (226) patients received G-CSF on day +1 (n=43), day +5 (n=78), and day +7 (n=105) from May 2012 to August 2015. The majority of the patients received levofloxacin, acyclovir and fluconazole as prophylaxis. We evaluated the duration of severe neutropenia (ANC<500), the onset of bacteremia, WHO grade 2-4 mucositis, and the time to biochemical progression as part of an IRB approved protocol (NCT01653106). Results:The cohort of patients receiving G-CSF on day +5 had a shorter median follow up (455 days versus 979 days in the day +1 and 1355 days in the day +7 cohorts), since the patients were enrolled at a later timepoint. No statistically significant difference was noted in terms of age, gender, cytogenetic or ISS stage distribution, number of infused cells per kg, and melphalan dose among the three cohorts of patients. The duration of severe neutropenia was significantly increased for patients receiving G-CSF on day +7 (mean 153.4 hrs, SD 36.9) compared to those receiving G-CSF on day +1 (mean: 104.8 hrs, SD 19.4; p-value <0.0001) or day +5 (mean: 120.5 hrs, SD 27.4; p-value <0.0001). There was also a statistically significant difference between the duration of severe neutropenia for G-CSF day +1 or day +5 (p-value 0.013). The duration of neutropenia was not influenced by gender, International Staging System or R-ISS at diagnosis. Moreover, no statistically significant difference was noted in terms of duration of severe neutropenia when patients were subdivided based on the number of stem cell infused (< or > of 5 x 106 cells). Patients who received G-CSF on day +1 or day +5 had a significantly decreased incidence of grade 2 or higher mucositis (4.65% vs 10.3%) for day +1 and day +5 versus 21.9% for day +7 (fisher-test, p = 0.014 and 0.0255). Similarly, the incidence of bacteremia was decreased in those patients treated with G-CSF on days +1 and +5 versus day +7 (21.8%, 23.2%, and 33.3% for those treated on days +1, +5, and +7, respectively; p = 0.09). The group of patients who received G-CSF on day +5 had a longer length of stay, with two patients with prolonged hospitalization (mean 17.7 days, SD 5.6 days, range 12-49) when compared to the other groups (day +1 group mean 13.88 days, SD 2.8 days, range 10-21 versus day +7 group mean 15.2 days, SD 2.7 days, range 11-21 days; Two-tailed t-test p = 0.012). Lastly, our data indicates a trend, though not statistically significant, toward a longer time to biochemical progression in patients who received G-CSF on day +1 when compared to the other two groups. Conclusion: We conclude that starting G-CSF injections the day after stem cell infusion (day +1) decreases the duration of severe neutropenia, infections, and mucositis in comparison to day +5 and day +7 in patients receiving autologous transplant for MM. A prospective trial would definitively test this, but a multi-institution meta-analysis via CIBMTR would be more cost-effective, acknowledging that differing anti-infective prophylaxis regimens would prevent effective between institution comparisons of infection. Zizyphus spina-christi (Rhamnaceae family) is an edible plant used in folk medicine. Therefore, it is of interest to report the cytotoxic effects of Z. spina-christi bark crude extract on human cell lines. Crude ethanol extract of Z. spina-christi bark was fractionated with increasing polarity (diethyl ether, chloroform, ethyl acetate and butanol fractions). The fractions were examined for their cytotoxicity against human colon cancer (HCT-116 and CACO-2), cervical cancer (HeLa and HEp-2), lung carcinoma (A-549), hepatocellular carcinoma (HepG-2), breast cancer (MCF-7) and prostate cancer (PC-3) cell lines using viability assay. Diethyl ether fraction of Z. spina-christi showed the highest cytotoxic effects among the four extracts of Z. spina-christi. The IC50 of diethyl ether fraction was 7.14, 11.2, 11.6, 15.4, 39.8, 42.2, 84.2 and 153.8 Ĵg/ml on HepG-2, A-549, CACO-2, HCT-116, MCF-7, PC-3, HeLa, and HEp-2 cell lines, respectively. Data shows that the diethyl ether fraction of Z. spina-christi showed effective cytotoxic effects in colon, lung and hepatocellular cancer cell lines.

A chemical investigation of the fibrous roots of Anemarrhena asphodeloides Bge. led to the isolation of four benzophenones, including one new compound (1) and three known ones (2–4). Comprehensive 1D, 2D NMR and HRESIMS data established the structures of the isolated compounds. The absolute configurations were determined by comparison of the calculated optical rotation (OR) with experimental data. All the isolates were evaluated for their cytotoxicities on hepatocellular carcinoma cell lines (HepG2 and Hep3B). Compound 1 showed strong cytotoxicity against HepG2 and Hep3B cells, with IC50 values at 153.1 and 180.6 nM. Through MTT assay, flow cytometry and Western blot analysis, compound 1 demonstrated the ability to stimulate apoptosis via the NF-κB signaling pathway in HepG2 cells. These benzophenones are potential lead compounds for the development of better treatments for hepatocellular carcinoma.

Abstract. Yearling red deer (Cervus elaphus) hinds of identical initial body weight were reared on a monocotyledonous grass (group 1) or on a papillonaceous plant pasture (group 2) for 212 days. At the end of the experiment (when deer were shot) blood was taken from ten animals of each group for serum biochemical analysis. Hinds of group 2 provided higher final body weight (90±3.5 vs. 101±6.6 kg) and higher daily body weight gain (105.7±10.7 vs. 153.8±26.8 g/day). Within serum nitrogenous compounds group 2 provided higher total protein concentrations, while from the lipids only serum triglyceride levels were higher in this group. Serum potassium was in both groups higher than the reference range with a superposed slight hyperkalaemia in group 2. Higher lactate dehydrogenase and alkaline phosphatase activities were found in group 2 and lower aspartate aminotransferase activity values. Inorganic phosphate concentration showed a significant difference (group 1 provides higher values). Results refer to an expressed venison growth as a result of the rich dietary protein supply of group 2. Findings were evaluated as well with discriminant factor analysis, outlining the relative importance of the single blood biochemical parameters in shaping the inter-group differences.

An experimental study was conducted to find the impact of interactive teaching method on science learning. 70 students from seventh grade were selected to participate in this study. A control group with 35 randomly assigned students was taught by the traditional lecture method, and an experimental group with another 35 students was taught by an interactive teaching method. A pre-test was conducted prior to implementing the intervention to measure the baseline score, while a post-test was administered to measure the impact of the intervention. An ANCOVA (Analysis of Covariance) was used to find the significant difference in science achievement score of students between the traditional teaching method and interactive teaching method after controlling the effect of pretest scores. There was a significant difference in students' science achievement score between students in the traditional teaching group and students in the interactive teaching group, F(2, 67) = 153.47, p <.001n 2 = .82. It was concluded that the interactive teaching method significantly improves student science learning.

In Jalisco, the "mezcal azul" Agave tequilana Weber grciws in 43 000 hectares of non-irrigated farmland representing 153.6 mi Ilion plants belonging to 2 500 owners. The industry of tequila employs 23 202 persons in Jalisco and produces 70 million liters per year, exporting 90% in bulk to the United States and the remainder to more than 40 countries. The traditional cultivation system has barely been studied and has been changing in the last decade, especially to a monoculture with an excessive use of fertilizers and pesticides, but the consequences of their use have not been evaluated. Some producers consider as an advantage the decrease by one to two years in the crop period, but the majority acknowledge productivity and environmental costs. Small growers are losing the agave market due to surplus agave production and their incipient social organization.

This paper aims to understand the bioaccumulation of pyraoxystrobin in fish. Using a flow-through bioconcentration method, the bioconcentration factor (BCF) and clearance rate of pyraoxystrobin in zebrafish were measured. The measured BCF values were then compared to those estimated from three commonly used predication models. At the exposure concentrations of 0.1 μg/L and 1.0 μg/L, the maximum BCF values for pyraoxystrobin in fish were 820.8 and 265.9, and the absorption rate constants (K1) were 391.0 d−1 and 153.2 d−1, respectively. The maximum enrichment occurred at 12 d of exposure. At the two test concentrations, the clearance rate constant (K2) in zebrafish was 0.5795 and 0.4721, and the half-life (t1/2) was 3.84 d and 3.33 d, respectively. The measured BCF values were close to those estimated from bioconcentration predication models.

ObjectiveTo evaluate the balance and functional capacity of fibromyalgia patients’ with and without migraine.MethodsObservational study with 60 women, between 20 and 60 years old (47±9 years), who were allocated into the following groups: 1. fibromyalgia and migraine (n=49); 2. only fibromyalgia (n=11). The following were used: Migraine Disability Assessment Test (MIDAS) to analyze the impact of migraine on quality of life; Revised Fibromyalgia Impact Questionnaire (FIQ-R) to assess the impact of fibromyalgia; six-minute walk test (6MWT)to assess functional capacity and Berg Balance Scale (BBE) for balance analysis.ResultsThere was no difference between groups 1 and 2 regarding functional capacity (TC6M: 308.7 m ±107.33 vs. 298.8 m ±153.54; p=0.55) and balance (BBE: 48± 14 vs. 47±16; p=0.68) as well as the impact on quality of life (FIQ: 78.8±14.77 vs. 74.4±13.99; p=0.28 and MIDAS: 31 ±24).ConclusionsOur study showed that there was no difference in functional capacity, balance and quality of life in fibromyalgia patients compared to patients with fibromyalgia associated with migraine.

Sixty bacterial strains isolated from the soils sample in the presence of organic solvent were screened for xylanase production. Among them, strain RSPP-15 showed the highest xylanase activity which was identified asBacillus vallismortis. The isolate showed maximum xylanase production (3768 U/mL) in the presence of birch wood xylan and beef extract at 55°C pH 7.0 within 48 h of incubation. The enzyme activity and stability were increased 181.5, 153.7, 147.2, 133.6, and 127.9% and 138.2, 119.3, 113.9, 109, and 104.5% in the presence of Co2+, Ca2+, Mg+2, Zn+2, and Fe+3ions (10 mM). Xylanase activity and stability were strongly inhibited in the presence of Hg and Cu ions. The enzyme was also stable in the presence of 30% ofn-dodecane, isooctane,n-decane, xylene, toluene,n-hexane,n-butanol, and cyclohexane, respectively. The presence of benzene, methanol, and ethanol marginally reduced the xylanase stability, respectively. This isolate may be useful in several industrial applications owing to its thermotolerant and organic solvent resistance characteristics.

Abstract Semarang has experienced floods every year, with a peak in the rainy season. At the end of 2022, there was heavy rain which caused the East Flood Canal River to overflow. As a result, the floods inundated several areas in Semarang City, including Kota Lama of Semarang. This study aims to determine the value of the designed discharge, the area of the flood inundation that occurred and the flood inundation map for return periods of 2, 5, 10, 25, 50 and 100 years. The methodology of this research consist of hydrological analysis, hydrological modelling with HEC-HMS and hydraulics modelling with HEC-RAS and integration with ArcGIS. In this study, it was found that for a return period of 2 years the planned flood discharge value was 153.7 m3/second with an inundation area of 406 Ha and a flood height of 17 cm to 68 cm, up to a return period of 100 years with a planned flood discharge value of 455.7 m3/second with an inundation area of 796 Ha and a flood height of 83 cm to 180 cm.

In this study, effects of bloodmeal on onion (Allium cepa L. CV. BARI peaj-1) cultivation to increase yield and related attributes have been tested. The effect of blood meal as fertilizers has been compared with growth hormone treatment and other conventional fertilizer. In the field, the different amount of growth elements treated were at the rate of 2550, 241.90 and 0.17 kg/ha for blood meal, urea and gibberellic acid (GA3), respectively. The yield performance was 172.84, 189.59 and 153.34 MT/ha for GA3, urea and blood meal, respectively. Using GA3 is expensive, and use of urea, a chemical fertilizer, can have adverse effect on soil. Blood meal, a biological ingredient, appears to be a bio-rationale amendment for production and sustained yield of onion. The results will need to ground truth tested in the field. Bangladesh J. Sci.
