Abstract:
Conservation of genetic resources is of great concern globally to maintain genetic diversity for sustainable food security. Comprehensive identification of the breed composition, estimation of inbreeding and effective population size are essential for the effective management of farm animal genetic resources and to prevent the animals from genetic erosion. The Zhongwei male (ZWM), Arbas Cashmere male (ACM) and Jining Grey male (JGM) goats are conserved in three different state goat farms in China but their family information, level of inbreeding and effective population size are unknown. We investigated the genomic relationship, inbreeding coefficient and effective population size in these three breeds from three state goat farms using the Illumina goat SNP50 BeadChip. Genomic relationships and phylogenetic analysis revealed that the breeds are clearly separated and formed separate clusters based on their genetic relationship. We obtained a high proportion of informative SNPs, ranging from 91.8% in the Arbas Cashmere male to 96.2% in the Jining Grey male goat breeds with an average mean of 96.8%. Inbreeding, as measured by FROH, ranged from 1.79% in ZWM to 8.62% in ACM goat populations. High FROH values, elevated genomic coverage of very long ROH (>30 Mb) and severe decline in effective population size were recorded in ACM goat farm. The existence of a high correlation between FHOM and FROH indicates that FROH can be used as an alternative to inbreeding estimates in the absence of pedigree records. The Ne estimates 13 generations ago were 166, 69 and 79 for ZWM, ACM and JGM goat farm, respectively indicating that these goat breeds were strongly affected by selection pressure or genetic drift. This study provides insight into the genomic relationship, levels of inbreeding and effective population size in the studied goat populations conserved in the state goat farms which will be valuable in prioritizing populations for conservation and for developing suitable management practices for further genetic improvement of these Chinese male goats.

Abstract:
AbstractOne of the basic methods of mechanical rock mining is cutting, which faces increasingly difficult working conditions. Despite the rapid development of machines used in underground and opencast mining as well as in tunnel building, construction industry and road engineering, the problem of insufficient durability of mining tools remains unsolved. In addition to drilling and, to a lesser extent, planing, cutting provides a huge market for tools. Currently, the process of cutting is mainly based on conical picks. The cutterheads of cutting machines are equipped with several dozen, and frequently – more than one hundred conical picks, which, due to their workability and abrasiveness, sometimes work only a few hours. There is a market demand for over two hundred models of conical picks. This is due to the huge variety of shapes and sizes of picks as well as the methods of their mounting in the holder. The article briefly presents various solutions of conical picks, their construction, methods of protection, dimensions and materials used. Next, based on materials produced by ZWM Carbonex, the classic method of their manufacture using the turning technology has been described. The authors have also presented briefly the use of die forging for the large-scale production of picks, applied by Górnicza Fabryka Narzędzi Sp. z o.o.

Abstract:
The Y or W sex chromosome of a heteromorphic pair is usually heterochromatinised and degenerated. However, whether chromosome degeneration constantly proceeds toward an extreme end is not fully understood. Here, we present a case of intermittent evolution of W chromosomes caused by interpopulation hybridisation in the Japanese soil-frog, Glandirana rugosa. This species includes two heteromorphic sex chromosome systems, which are separated into geographic populations, namely the XY and ZW groups. In this study, to uncover the evolutionary mechanisms of the heterogeneous W chromosomes, we genetically investigated the geographic differentiation of the ZW populations along with the closely located XY populations. Analysis of mitochondrial cytochrome b sequences detected three distinct clades, named ZW1, ZW2, and ZW3. High throughput analyses of nuclear genomic DNA showed that autosomal alleles of XY populations were deeply introgressed into the ZW3 sub-group. Based on the genotypes of sex-linked single nucleotide polymorphisms, W-borne androgen receptor gene expression, and WW developmental mortality, we concluded that the X chromosomes were recycled to W chromosomes. Upon inclusion of two cases from another group, Neo-ZW, we observed that the X chromosomes were recycled independently at least four times to the new W chromosomes: a repetition of degeneration and resurrection.

Full text
Abstract:
Abstract Southern Hemisphere mid- to upper-tropospheric planetary wave activity is characterized by the superposition of two zonally oriented, quasi-stationary waveforms: zonal wavenumber 1 (ZW1) and zonal wavenumber 3 (ZW3). Previous studies have tended to consider these waveforms in isolation and with the exception of those studies relating to sea ice, little is known about their impact on regional climate variability. A novel approach is taken to quantifying the combined influence of ZW1 and ZW3, using the strength of the hemispheric meridional flow as a proxy for zonal wave activity. The methodology adapts the wave envelope construct routinely used in the identification of synoptic-scale Rossby wave packets and improves on existing approaches by allowing for variations in both wave phase and amplitude. While ZW1 and ZW3 are both prominent features of the climatological circulation, the defining feature of highly meridional hemispheric states is an enhancement of the ZW3 component. Composites of the mean surface conditions during these highly meridional, ZW3-like anomalous states (i.e., months of strong planetary wave activity) reveal large sea ice anomalies over the Amundsen and Bellingshausen Seas during autumn and along much of the East Antarctic coastline throughout the year. Large precipitation anomalies in regions of significant topography (e.g., New Zealand, Patagonia, and coastal Antarctica) and anomalously warm temperatures over much of the Antarctic continent were also associated with strong planetary wave activity. The latter has potentially important implications for the interpretation of recent warming over West Antarctica and the Antarctic Peninsula.

Abstract:
Respiratory impedances (Zrs) measured with the cheeks manually supported (Zsc) demonstrate an antiresonance (AR) at approximately 170 Hz, whereas those measured with the head generator (Zhg) have an AR at significantly lower frequencies (150 Hz). The differences in the AR between methods were previously attributed to upper airway wall (UAW) shunting in Zsc, and Zhg was believed to represent shunt-free impedance. To test this hypothesis, we measured four independent estimates of Zrs (4–256 Hz) in five healthy adults. Applying the oscillations at the mouth, we measured Zrs with the cheeks unsupported (Zuc) and when the subject's head and neck were enclosed in a rigid chamber completely filled with water (Zwa). We also measured Zhg with the oscillations applied around the head. Because water is incompressible, Zwa should minimize UAW shunt if not completely eliminate it. There were no significant differences in any of the AR features (e.g., location and bandwidth) of Zuc, Zsc, and Zwa. Conversely, all AR features of Zwa and Zhg were significantly different. To assess wall motion during both forms of oscillations (i.e., applied at the mouth and around the head), we measured two UAW local impedances by use of accelerometers placed on the cheeks (Zch) and submental (Zsm) region. Above 40 Hz, Zch and Zsm estimates were significantly dependent on the forcing method. Furthermore, Zch and Zsm made with the standard oscillations were consistent with the lumped UAW impedances measured with a head plethysmograph, whereas those made with the oscillations around the head were not. Therefore we conclude that, at > 32 Hz, Zsc more closely represents shunt-free impedance than does Zhg and hence should be preferred when Zrs are measured.

Abstract:
Abstract Endothelial cells synthesize a heterodimeric adhesion molecule, the vitronectin receptor (VnR), which is similar to the platelet glycoprotein (GP)IIb/IIIa complex. The subunits of the endothelial VnR (VnR alpha and GPIIIa) have been studied for their ability to express alloantigens associated with platelet GPIIb and IIIa. We previously showed that endothelial GPIIIa can express the platelet alloantigen Zwa or PIA1, which is associated with GPIIIa. We studied the relationship between the expression of Zwa on platelets and endothelial cells in neonates (n = 13). Using immunoprecipitation and immunofluorescence techniques, we showed that the Zwa antigen is either expressed or absent from both platelets and endothelial cells of the same individual. This finding indicates that in both cell types the same gene is expressed. We also showed that Zwa-negative endothelial cells express Zwb (PIA2), in analogy to Zwa-negative platelets. Moreover, our results strongly suggest expression on endothelial cells of Yukb, a recently described platelet alloantigen, also located on GPIIIa. However, we could not demonstrate expression on the endothelial VnR alpha subunit of Baka, an alloantigen located on platelet GPIIb. These findings are in agreement with the concept that the endothelial GPIIIa subunit is more closely related to its platelet counterpart than to the endothelial VnR alpha subunit.

Abstract:
Long time series of reanalyses, from NCEP–NCAR and from ECMWF, are used to investigate the occurrence of persistent positive anomalies (PPAs) in the 500-hPa geopotential height field over the Southern Hemisphere extratropics during 1958–2001. Defining persistent anomalies as those of at least 100 m in magnitude lasting for at least 5 days, it is found that the region of most frequent occurrence is over the South Pacific. A cluster analysis of monthly PPA counts shows two distinct patterns, one a zonal wavenumber-1 (ZW1) pattern centered over the southeast Pacific near 60°S and the other a zonal wavenumber-3 (ZW3) pattern with centers near New Zealand and over the southern Atlantic and Indian Oceans. Results were insensitive to the choice of dataset, and to the removal of a linear trend from the daily height fields. The southeast Pacific PPA region is strongly modulated by ENSO, while the ZW3 pattern appears only weakly related to ENSO variability. A strong upward trend is apparent in occurrence of the ZW3 cluster, related to a matching trend in the variance of the height fields, particularly those from ECMWF. Such trends are at least in part a consequence of changes in the observing system, particularly the introduction of satellite soundings in the late 1970s.

Abstract:
Levitation Mass Method (LMM) is the method as a material tester to evaluate the mechanical response of general objects against impact forces. In this method, a mass is made to collide with material to be tested and the impulse, i.e. the time integration of the impact force, is measured highly accurately as a change in momentum of the mass. To realize linear motion with sufficiently small friction acting on the mass, a pneumatic linear bearing is used. The inertial force acting on the mass is calculated from the velocity of the mass. The velocity is determined, highly accurately by means of measuring the Doppler shift frequency of a laser light beam reflected on the mass using an optical interferometer. To determine the Doppler frequency shift for LMM data processing, the method for estimating the frequency is necessary. Several methods have been developed to estimate the frequency for the LMM data processing with high accuracy, i.e. Zero-Crossing Average Method (ZAM), Zero-Crossing Fitting Method (ZFM), Sine Wave Fitting, and Zero-crossing Sine Wave Fitting. All methods realized using the zero-crossing point of the waveform obtained from the digitizer. A better method to estimate frequency on the digitized waveform will enable higher precision for a more accurate result. In this research, a new method that can improve the accuracy has been developed. The program was developed using data segmentation to obtain the frequency of the digitized waveforms. The developed program has the smallest error ( 1,98 X 10^-10 for N= 200) compare to other methods (2,31 X 10-3 for ZAM; 1,10X10-3 for ZFM; and 8,69 X10-4 for Zero-crossing Sine Wave Fitting).

Abstract:
In the brief overview of the procedure for enacting an administrative act it is stated that in the German understanding the administrative procedure with its inherent control mode, goal orientation and role of the various institutions and bodies as well as persons involved in the intermediary role between constitutional requirements and administrative reality is not purely technical one. The authority decides ex officio about the opening of the procedure except a citizen applied for, it decides at its discretion except when it must act ex officio how to proceed. Its important principles named as informality, simplicity, expediency, speed, obligation t o cooperate and freedom of form. Special importance is attributed to the principle of officiality of the investigation of a given case. The relevance of circumstances arises from the relevant legal norms and – i case of – discretionary decisions. The extent of the proof and the burden of proof are also important for the scope of the official investigation principle. Authorities are obliged to grant the citizen a right to be heard in the administrative procedure and to influence the course and outcome of the procedure. It carries direct and indirect external impact, revealing the basic understanding of relations between the state and the individual derived from the constitution. Equal treatment of all citizens and material substantivity proves the rule of law and prohibits the abuse of power at the expense of the citizen.
