Abstract:
One of the main objectives of many biological studies is to explore differential gene expression profiles between samples. Genes are referred to as differentially expressed (DE) if the read counts change across treatments or conditions systematically. Poisson and negative binomial (NB) regressions are widely used methods for non-over-dispersed (NOD) and over-dispersed (OD) count data respectively. However, in the presence of excessive number of zeros, these methods need adjustments. In this paper, we consider a zero-inflated Poisson mixed effects model (ZIPMM) and zero-inflated negative binomial mixed effects model (ZINBMM) to address excessive zero counts in the NOD and OD RNA-seq data respectively in the presence of random effects. We apply these methods to both simulated and real RNA-seq datasets. The ZIPMM and ZINBMM perform better on both simulated and real datasets.

Abstract:
Background and aims We analyze the possible predictive variables for Adverse Events (AEs) during sedation for gastrointestinal (GI) endoscopy. Methods We consider 23,788 GI endoscopies under sedation on adults between 2012 and 2019. A Zero-Inflated Poisson Regression Mixture (ZIPRM) model for count data with concomitant variables is applied, accounting for unobserved heterogeneity and evaluating the risks of multi-drug sedation. A multinomial logit model is also estimated to evaluate cardiovascular, respiratory, hemorrhagic, other AEs and stopping the procedure risk factors. Results In 7.55% of cases, one or more AEs occurred, most frequently cardiovascular (3.26%) or respiratory (2.77%). Our ZIPRM model identifies one population for non-zero counts. The AE-group reveals that age >75 years yields 46% more AEs than age <66 years; Body Mass Index (BMI) ≥27 27% more AEs than BMI <21; emergency 11% more AEs than routine. Any one-point increment in the American Society of Anesthesiologists (ASA) score and the Mallampati score determines respectively a 42% and a 16% increment in AEs; every hour prolonging endoscopy increases AEs by 41%. Regarding sedation with propofol alone (the sedative of choice), adding opioids to propofol increases AEs by 43% and adding benzodiazepines by 51%. Cardiovascular AEs are increased by age, ASA score, smoke, in-hospital, procedure duration, midazolam/fentanyl associated with propofol. Respiratory AEs are increased by BMI, ASA and Mallampati scores, emergency, in-hospital, procedure duration, midazolam/fentanyl associated with propofol. Hemorrhagic AEs are increased by age, in-hospital, procedure duration, midazolam/fentanyl associated with propofol. The risk of suspension of the endoscopic procedure before accomplishment is increased by female gender, ASA and Mallampati scores, and in-hospital, and it is reduced by emergency and procedure duration. Conclusions Age, BMI, ASA score, Mallampati score, in-hospital, procedure duration, other sedatives with propofol increase the risk for AEs during sedation for GI endoscopy.

Abstract:
Public transport and private cars, when combined with urban planning, tend to contribute to the development of local sustainability. The objective of the research is to analyze the population perception regarding the use of public transportation and private cars in the city of Passo Fundo, RS, Brazil, seeking to understand the population's preference for private transportation and public transport. Methodologically, a 90% confidence interval and a 10% sample error were used to select the 68 questionnaires applied, where multiple choice questions were used. The results revealed that 66% of the interviewed people use public transport, but they evaluate it as of poor quality. As a proposal for the improvement of motorized mobility, the Car Sharing system of three organizations: ZipCar, ZasCar and Car2Go was carried out to suggest the implementation of the system in the city. The results of the study demonstrated the feasibility of the Car Sharing System as an alternative for improvements in sustainable urban mobility.

AbstractThe sharing economy has raised hopes that online platforms will usher in a new era of sharing, even though economic theory suggests that income growth may reduce sharing in the long run. This paper presents evidence that that high-income people are less likely than low-income people to use traditional institutions for sharing goods, including carpools, multi-person households, and garage sales. While it first appears that high-income people are equally likely to use new institutions, such as Craigslist, Airbnb, and Zipcar, this partly reflects the fact that many low-income households in the US still lack an internet connection. Conditional on having internet access, this paper finds that online sharing platforms are also disproportionately used by the poor. The future of sharing likely depends on countervailing forces. Economic growth may continue to dampen incentives to share goods, but this effect could be offset by the proliferation of institutions, norms, and preferences that facilitate sharing.

Abstract The recently developed Hi-C technique has been widely applied to map genome-wide chromatin interactions. However, current methods for analyzing diploid Hi-C data cannot fully distinguish between homologous chromosomes. Consequently, the existing diploid Hi-C analyses are based on sparse and inaccurate allele-specific contact matrices, which might lead to incorrect modeling of diploid genome architecture. Here we present ASHIC, a hierarchical Bayesian framework to model allele-specific chromatin organizations in diploid genomes. We developed two models under the Bayesian framework: the Poisson-multinomial (ASHIC-PM) model and the zero-inflated Poisson-multinomial (ASHIC-ZIPM) model. The proposed ASHIC methods impute allele-specific contact maps from diploid Hi-C data and simultaneously infer allelic 3D structures. Through simulation studies, we demonstrated that ASHIC methods outperformed existing approaches, especially under low coverage and low SNP density conditions. Additionally, in the analyses of diploid Hi-C datasets in mouse and human, our ASHIC-ZIPM method produced fine-resolution diploid chromatin maps and 3D structures and provided insights into the allelic chromatin organizations and functions. To summarize, our work provides a statistically rigorous framework for investigating fine-scale allele-specific chromatin conformations. A economia compartilhada é um tema recente da sociedade contemporânea, em que tem se observado um crescimento de organizações com este perfil e a atenção de investidores às empresas que surgem nesse segmento. A economia compartilhada pode ser considerada uma atividade de peer-to-peer com base na obtenção, concessão ou compartilhamento de acesso a bens e serviços, coordenada através de serviços comunitários online. Plataformas de diversos segmentos como Uber (transporte), Airbnb e CouchSurfing (Turismo e Hospitalidade), Zazcar e Zipcar (compartilhamento de carros), Coursera e Cinese (educação) são alguns dos exemplos que têm surgido nesse sentido. O objetivo do presente artigo é investigar quais são os fatores que contribuem para propensão de consumidores para participar de plataformas de economia compartilhada. Para isso, realizou-se um estudo qualitativo, de caráter exploratório, com grupos de foco, empregando-se a análise de conteúdo como método de análise. Como resultado, concluiu-se que a economia de custos, a influência social e a confiança, e a idade e o estágio no ciclo de vida são os fatores que mais contribuem para o interesse em utilizar as plataformas de economia compartilhada, na visão dos participantes dos grupos de foco.

The study assessed the level of adoption and profitability of summer tomato varieties at farm level. Data were collected from 90 randomly selected tomato farmers of Bagherpara, Jashore Sadar and Jhikorgacha upazila of Jashore district. The results indicated that BARI Hybrid Tomato-4 was highly adopted summer tomato variety (75%) followed by BARI Hybrid Tomato-8 (16%) and ACI summer king tomato variety (9%). The adoption level of ploughing, manure and fertilizer use were low, whereas planting time and irrigation were high. Total cost of production of summer tomato was Tk 584822 per hectare whereas Tk 507355 per hectare was variable cost and fixed cost was Tk 77467 per hectare. Among the cost items mancha preparation cost was the highest (26.89 %) and 26.10 % cost was for labor. The average yield of summer tomato was 50.41 t/ha and gross return was 1542300 tk/ha. On the average, benefit cost ratio was found to be 2.64 on full cost basis and 3.04 on cash cost basis. MoP, zipsum and manure were significant effect on summer tomato cultivation. Attack by pest and disease, lack of seed at proper time, lack of agricultural credit and high cost of production were the major constraints for the adoption of summer tomato.Bangladesh J. Agril. The variety of problem conditions and the specific dynamics of college student demographics demands a holistic measurement. Various measurements to reveal the attributes of students' problems have been developed in Indonesia, but they are not optimal because they are still partial. Even though a holistic measurement tool has been developed, its effectiveness is still a matter of debate, especially in measuring instrument duration and the number of items that are too large. The development of the Zadrian-Ifdil Problem Checklist (ZIPC) measuring instrument aims to obtain an effective and efficient measuring tool in identifying problems experienced by students daily. This study consists of three phases: item development, testing, validation, and item revision. The sample involved in this study were 639 people spread all over Indonesia. Analysis and validation of measuring tools using the Rasch model approach. The development and validation results revealed that the student version of ZIP-C was reliable and valid to reveal daily personal problems in 8 variables of problem conditions and 50 items. This measuring tool can be used for research purposes and practice services and mental health interventions by professionals.

As transport systems are pushed to the limits in many cities, governments have tried to resolve problems of traffic and congestion by increasing capacity. Miller (2013) contends the need to identify new capabilities (instead of capacity) of the transport infrastructure in order to increase efficiency without extending the physical infrastructure. Kenyon and Lyons (2003) identified integrated traveller information as a facilitator for better transport decisions. Today, with further developments in the use of geographic information systems (GIS) and a greater disposition by the public to provide volunteered geographic information (VGI), the potential of information is not only integrated across modes but also user-generated, real-time and available on smartphones anywhere. This geographic information plays today an important role in sectors such as politics, businesses and entertainment, and presumably this would extend to transport in revealing people’s preferences for mobility and therefore be useful for decision-making. The widespread availability of networks and smartphones offer new opportunities supported by apps and crowdsourcing through social media such as the successful traffic and navigation app Waze, car sharing programmes such as Zipcar, and ride sharing systems such as Uber. This study aims to develop insights into the potential of governments to use voluntary (crowdsourced) geographic information effectively to achieve sustainable mobility. A review of the literature and existing technology informs this article. Further research into this area is identified and presented at the end of the paper.

Respondiendo a las tendencias de la globalización, diferentes actores están invirtiendo en alta tecnología para asumir la cuarta era tecnológica. En consecuencia, el concepto de economía compartida se ha convertido en un nuevo método de producción económica, que nace de una nueva forma de producción mediante compartir lo que se tiene y ganar una remuneración a través de las plataformas virtuales como Uber, Cabify, Airbnb, inDriver, Zipcar y otras. Estas nuevas tecnologías representan retos en varias dimensiones, entre esas la gestión de la ciudad. En este estudio, se analizó el caso de Airbnb en la capital del Ecuador, Quito urbano y la conurbación de Los Valles. El objetivo es analizar los patrones territoriales del servicio, las variables que permiten que exista un Airbnb y su relación con una posible gentrificación, aunque no existan estudios sustanciales de la misma. Se estudia también el rol de la proximidad a hitos, la dotación de servicios y la heterogeneidad territorial en la dinámica socioespacial de los alojamientos Airbnb. Este estudio permitirá a los tomadores de decisiones diagnosticar cómo el servicio de Airbnb se está posicionando en la ciudad y poder anticiparse ante una posible gentrificación urbana no planificada producto de los procesos de “nuevo colonialismo urbano”.

It can be used entirely in business classes in marketing, entrepreneurship, operations management, and transportation/logistics, and parts of it can be used for discussions in classes related to emerging economies/markets, environmental management, sustainability, and technology management. Case overview The case builds on the expansion plan considered by a young software company, called Hangzhou Omnipay located in the city of Hangzhou, China. Mr Chao, Vice President (VP) of Omnipay, is the main character of the case. He was aware of the current car-sharing industry leader – Zipcar headquartered in Boston and also identified multiple stakeholders in the city for decision making. By collaborating with a global student project team, Mr Chao collected a great deal of information and data. This teaching case provides students and educators ample opportunities to examine, from a multitude of aspects, the viability of a car-sharing service in Hangzhou. Expected learning outcomes The central goal is to help students gain a comprehensive understanding of the role of car-sharing service in a country's development in sustainability, socio-economy, environmental commitment, and new urban life style, as well as in a technological company's active pursuit of business expansion opportunity. Abstract Background Wearable activity trackers may facilitate walking for chronic pain management. Objective We assessed the acceptability of a commercially available tracker and three alternative modes of reporting daily steps among older adults in a low-income, urban community. We examined whether using the tracker (Fitbit ZipTM) was associated with improvements in functioning and activity. Design Randomized controlled pilot and feasibility trial. Subjects Fifty-one African American adults in Detroit, Michigan, aged 60 to 85 years, with chronic musculoskeletal pain (28 in the intervention group, 23 controls). Methods Participants completed telephone surveys at baseline and eight weeks. Intervention participants wore trackers for six weeks, alternately reporting daily step counts via text messages, automated telephone calls, and syncing (two weeks each). We used multimethods to assess satisfaction with trackers and reporting modalities. Adherence was indicated by the proportion of expected days on which valid step counts were reported. We assessed changes in pain interference, physical function, social participation, walking frequency, and walking duration. Results More than 90% of participants rated trackers as easy to use, but some had technical or dexterity-related difficulties. Text reporting yielded 79% reporting adherence vs 69% each for automated calls and syncing. Intervention participants did not show greater improvement in functioning or walking than controls. Conclusions With appropriate support, wearable activity trackers and mHealth reporting for chronic pain self-care are feasible for use by vulnerable older adults. Future research should test whether the effects of trackers on pain-related outcomes can be enhanced by incorporating behavior change strategies and training in evidence-based cognitive-behavioral techniques.

Adolescents face various challenges and changes in order to realize their developmental tasks. The challenges are coupled with the technological disruption that requires adolescents to face new problems that have never existed before as millennials. However, the problems experienced by adolescents are often not expressed because of various obstacles, such as reluctance to tell spontaneously to the adults around them, high social conformity, so it is difficult to find help. For this reason, an alternative assessment is needed, which serves as an effort to map the problems experienced by adolescents and their mental health conditions accurately and actually. This study aims to find the value of the construct validity that forms the ZIPC to be used more widely and measure the actual problem conditions of the assessment subject. Participants involved in this study were 978 high school adolescents from all over Indonesia. The analysis used in finding the construct validity of this instrument is Exploratory Factor Analysis (EFA). The impact of changes and instrument conditions after analysis will be discussed.

The Sharing Economy (SE) has been growing at an impressive rate across the globe (Cohen - Kietzmann, 2014) and emerging as an innovative and rapidly growing sector of the economy (Hira - Reilly, 2017), which attracted the attention of the scientific community. An increasing number of studies have been brought to light helping to document and analyze how SE manifests and evolves across economic systems, thus, contributing to refine and recast existing management theory (Mair - Reischauer, 2017). Nevertheless, there still is a lack of a common understanding of SE and its underlying mechanisms (Knote - Blohm, 2016). As an emergent category, SE has been contoured by being a mutant process, as it has been crafted by multiple and distinct temporal identity and legitimacy events, mechanisms and claims. Showing signs of being an on-going process of evolution, there is a constant need for further research to identify developments in the evolution of SE considering both identify claims (self-referential) and legitimacy (granted by stakeholders), which would offer additional comprehension about the SE phenomenon. The research addresses it by studying what is the role of SE in establishing the identity of organizations belonging to the field, considering both identify claims (self-referential) and legitimacy (granted by stakeholders)? To answer the research question, the research was designed involving two components inspired on category creation studies (Durand - Paolella, 2013; Glynn - Navis, 2013; Navis - Glynn, 2010; Kennedy - Fiss, 2013; Kennedy et al., 2010; Lounsbury - Rao, 2004; and Wry et al., 2014): 1) four prototypical SE organizations were selected, each of them belonging to Schor’s (2014) four SE archetypes of activities: (i) Airbnb – peer-to-peer, for-profit activity –, (ii) Zipcar – business-to-peer, for-profit activity –, (iii) TimeBanks – peer-to-peer, non-profit – and (iv) Make: makerspaces – business-to-peer, non-profit. The content of the evolution of their identity claims was analyzed, using data from their public available reports, as well as, other secondary data available on-line; and 2) considering the same set of SE organizations, legitimacy evolution was analyzed considering how scientific community, investors, customers, media, other analysts and other interested audiences have been constructing category meaning to them, conferring the formation of SE categorical and organizational identities, and perceptions about the viability of their business models (Tripsas, 2009, as cited in Navis - Glynn, 2010). In short, it was analyzed how stakeholders assess the viability of SE categories and organizations and can grant or withhold legitimacy to SE organizations (Zuckerman, 1999, as cited in Navis - Glynn, 2010). Hasil penelitian menunjukkan bahwa perceived ease of use dan trust berpengaruh signifikan terhadap behavioral intention, demikian juga perceived ease of use terhadap trust. Penelitian ini juga berimplikasi pada ilmu pemasaran Islam, yaitu mengenai niat perilaku konsumen yang akan berefek pada penggunaan aplikasi m-banking bank umum syariah saat ini dan di masa mendatang, seperti perekomendasian aplikasi tersebut ke orang lain. Maka dari itu, bank umum syariah diharapkan dapat meningkatkan kualitas fitur yang terdapat di aplikasi m-banking, mempromosikan secara gencar kemudahan dan manfaat yang akan didapat dari penggunaan aplikasi m-banking agar nasabah tertarik untuk menggunakannya, serta menyederhanakan baik tampilan dan cara penggunaan, sehingga aplikasi tersebut bisa digunakan dengan mudah oleh berbagai kalangan.Kata Kunci: Perceived ease of use, trust, behavioral intention, aplikasi m-banking. ABSTRACTThe intention of this study is to discover perceived ease of use’s impact on behavioral intention with trust as the intervening variable in the usage of Islamic commercial banks’ m-banking applications. This research uses a quantitative approach with path analysis method and involves 50 participants who are customers of one of Islamic commercial banks as well as users of m-banking application of the bank concerned with online purposive sampling method. It also uses path analysis method by using AMOS program to analyze the data. The result of this study indicates both perceived ease of use and trust significantly affect behavioral intention, and so is perceived ease of use to trust. This study also implies to Islamic marketing, which refers to consumers’ behavior that will affect the current and future usage of Islamic comercial banks’ m-banking applications, such as recommending it to other people. Therefore, Islamic commercial banks are expected to improve their applications’ features, promote its convenience and benefits vigorously to attract customers to use it, and simplify both its appearance and usage method, so it can be used by anyone easily.Keyword: Perceived ease of use, trust, behavioral intention, m-banking application. Problematika sdílené ekonomiky je v současné době velmi aktuální téma mající svůj význam v mnoha odvětvích národního hospodářství. V oblasti dopravy je možné pozorovat využití některých principů sdílené ekonomiky, zejména v rámci služeb, které spočívají ve sdílení kapacit osobních aut při přepravě osob (BlaBlaCar, Zipcar, DriveNow atd.). S přihlédnutím k situaci, kdy meziročně narůstá počet přepravovaných zásilek, naskýtá se prostor pro využití principů sdílené ekonomiky nejen pro přepravu osob, jak tomu již je, ale také právě při zajištění přepravy zásilek. Vzhledem k tomu, že v České republice nebyla tato problematika komplexně řešena, je nutné před případným stanovením strategií implementace principů sdílené ekonomiky při přepravě zásilek ze strany příslušných veřejných institucí či ze strany podniků ověřit, zda jsou v České republice vytvořeny vhodné podmínky pro takovouto implementaci. Cílem článku je zhodnotit aktuální situaci v České republice v rámci implementace principů sdílené ekonomiky při přepravě zásilek z vnějšího prostředí, konkrétně s využitím PEST analýzy.

AbstractSchizophrenia is a complex and heterogenous disease that presents with abnormalities in glutamate signaling and altered immune and inflammatory signals. Genome-wide association studies have indicated specific genes and pathways that may contribute to schizophrenia. We assessed the impact of the functional missense variant SLC39A8 (ZIP8)-A391T (ZIP8A391T) on zinc transport, glutamate signaling, and the neuroinflammatory response. The ZIP8A391T mutation resulted in reduced zinc transport into the cell, suggesting a loss in the tight control of zinc in the synaptic cleft. Electrophysiological recordings from perturbed neurons revealed a significant reduction in NMDA- and AMPA-mediated spontaneous EPSCs (sEPSCs) and a reduction in GluN2A and GluA1/2/3 receptor surface expression. All phenotypes were rescued by re-expression of wild-type ZIP8 (ZIP8WT) or application of the membrane-impermeable zinc chelator ZX1. ZIP8 reduction also resulted in decreased BBB integrity, increased IL-6/IL-1β protein expression, and increased NFκB following TNFα stimulation, indicating that ZIP8 loss-of-function may exacerbate immune and inflammatory signals. Together, our findings demonstrate that the A391T missense mutation results in alterations in glutamate and immune function and provide novel therapeutic targets relevant to schizophrenia.

The vital data about the electrical activities of the brain are carried by the electroencephalography (EEG) signals. The recordings of the electrical activity of brain neurons in a rhythmic and spontaneous manner from the scalp surface are measured by EEG. One of the most important aspects in the field of neuroscience and neural engineering is EEG signal analysis, as it aids significantly in dealing with the commercial applications as well. To uncover the highly useful information for neural classification activities, EEG studies incorporated with machine learning provide good results. In this study, a Fusion Hybrid Model (FHM) with Singular Value Decomposition (SVD) Based Estimation of Robust Parameters is proposed for efficient feature extraction of the biosignals and to understand the essential information it has for analyzing the brain functionality. The essential features in terms of parameter components are extracted using the developed hybrid model, and a specialized hybrid swarm technique called Hybrid Differential Particle Artificial Bee (HDPAB) algorithm is proposed for feature selection. To make the EEG more practical and to be used in a plethora of applications, the robust classification of these signals is necessary thereby relying less on the trained professionals. Therefore, the classification is done initially using the proposed Zero Inflated Poisson Mixture Regression Model (ZIPMRM) and then it is also classified with a deep learning methodology, and the results are compared with other standard machine learning techniques. This proposed flow of methodology is validated on a few standard Biosignal datasets, and finally, a good classification accuracy of 98.79% is obtained for epileptic dataset and 98.35% is obtained for schizophrenia dataset.

Introduction: Behavioral economics-based interventions for increasing steps/day may improve cardiorespiratory fitness (CRF; Peak O2 uptake - VO2peak), central/aortic blood pressure, and vascular stiffness. The purpose of this study was to examine the effects of adaptive (Ad) physical activity (PA) goals (daily step goals based on prior 10-day window and a percentile algorithm) vs. static (St) PA goals (10,000 steps/day) combined with either immediate (Im) financial micro-incentives or delayed (De) incentives (Groups: Ad+Im; Ad+De; St+Im; St+De) on VO2peak, blood pressure and vascular stiffness. We hypothesized that participants randomized to adaptive goals with immediate reinforcement (Ad+Im) would have greater improvements in cardiovascular health measures than participants assigned to static goals and/or delayed reinforcement conditions (active controls). Methods: Inactive adults (Age = 41.8 ± 6.7yrs; BMI = 34.5 ± 6 kg/m2) were randomized to an adaptive/static goal intervention with either immediate micro-incentives or delayed incentives in a 4-month factorial RCT to improve steps/day. Additionally, all four groups received daily text-message prompts-to-action and Fitbit ZipTM activity monitors. Pre and post VO2peak (n=81) were estimated using a modified, sub-maximal Balke treadmill protocol while central blood pressures, augmentation index (Aix@75) (n=70) and carotid-femoral pulse wave velocity (cf-PWV) (n=64) were assessed with the Sphygmocor XCELTM device. Between- and within-group differences were examined using linear mixed models, adjusted for sex, age, race/ethnicity, and smoking history. Alpha was set at 0.05, and data are presented as estimated marginal means ± standard errors (SE). Results: All groups improved from pre to post, but we found no significant between-group differences in the degree of improvement. Across all groups, we found the following: VO2peak and all vascular markers improved significantly pre to post intervention (p<0.05). VO2peak improved by ~5% (pre = 24.9 ± 0.65 vs. post = 26.3 ± 0.66 ml/kg/min). Central systolic blood pressures (cSBP) decreased (cSBP pre = 117.5 ± 2.1 vs. post = 113.1 ± 2.2 mmHg). Cf-PWV (pre = 7.5 ± 0.18 vs. post = 7.3 ± 0.18 m/sec) and AIX@75 (pre =14.1 ± 2.2 vs. post = 11.4 ± 2.2) decreased. Conclusion: Cardiorespiratory fitness and markers of vascular health improved when participants were simply provided a Fitbit and prompted daily to be more physically active, regardless of incentive or goal-setting approach. Implications for designing behavioral interventions for cardiovascular risk reduction will be discussed.
