Abstract:

Abstract:
In the world of business nowadays there is a focus on shareholders, and a lack of separation between ownership and management. Innovations in investment technology have led to a growth in the assets of institutional investors and an increased influence of investors. Indeed, many investors have assumed the role of 'speaking shareholder' for the companies in question. This is referred to as hedge fund activism, with once silent investors becoming shareholders and, as a result, individuals and hedge funds are able to dominate the activities of companies. From the late 1990s to the early 2000s this situation was centred around pension funds but now revolves around hedge funds. Professor Yumiko Miwa, Graduate School of Commerce, Meiji University, Japan, is conducting an international comparative study of the influence of investment funds on enterprise management. This research investigates anti-takeover measures, preferential treatments and addresses potential conflicts of interest. In other research, Miwa is working to address the stance taken by some companies that women are socially disadvantaged. The topic of this research is Women Working in the Capital Markets.

Abstract:
As social beings, humans have a desire to create interpersonal relationships and communication. Interpersonal communication is also often created in the use of social media. However, this can lead to the merging of private space with public space so that private activities can be disseminated, including sexual orientation. One person who dares to reveal her sexual orientation is Yumi. In the @just.yumi account, she shows a lesbian relationship with her girlfriend. For this reason, this study wants to know the views of Generation Z on self-disclosure of lesbian couples on Yumi’s Instagram account. The theory used in this research are interpersonal communication and self-disclosure. The author uses a qualitative research approach with a case study method. The research data were obtained through interviews with six sources and one expert resource, simple surveys, documentation and literature studies. The results showed that Generation Z didn’t mind with Yumi’s content. According to them it is a right and freedom of expression. However, the majority of generation Z also said that Yumi's content could have a negative impact on Instagram users.

Abstract:
Food 3D printing is expected to be a technology to create foods with different tastes and hardness depending on individual preferences and to solve the problem of food loss problems. In this study, we focus on the 3D printing of chocolate, whose product value is greatly affected by its appearance, entertainment and texture. Although 3D printers of chocolate are gradually appearing in the market, most of them only focus on the modeling of shape, and there are quite few printers that can form chocolate with designed texture. We develop a chocolate 3D printer using two chocolate materials. In addition to the syringe type extruder, we introduce the gear pump type extruder, which has a fine and stable extrusion. We 3d print chocolates with various internal structures using two chocolate materials and evaluate the differences in their textures by equipment measurement and by sensory tests. In addition, the melt-in-mouth feeling and the snappiness, which are important for the texture of chocolate, are investigated. In this presentation, we report the results of texture tests of black chocolate formed by a single syringe type 3D printer. Abstract:
Animation is not only about the presentation of 3D caricatures but also about something beyond the scope of aesthetics, such as Emotional Intelligence which is portrayed in the animated film Inside Out and South Korean drama series Yumi’s Cells. The animated emotional intelligence characters who ‘lives inside the human brain’ are visualized as communicating and interacting with each other, representing the concept of Symbolic Interactionism Theory. This qualitative research, with a conversation analysis approach, offered a new point about the relationship and sequence process in the formation of three concepts in symbolic interactionism theory from George Herbert Mead and Herbert Blummer through the interactions of the animated characters in the film Inside Out and drama Yumi’s Cells. The results of the study showed that the characters of animated cells with emotional intelligence in the animated films Inside Out and Yumi's Cells could dominate the representation of the symbolic interactionist concepts by visualizing emotions and feelings.

Abstract:
Background:In clinical rheumatology, interferon-γ release assays (IGRAs) have been reported as a useful diagnostic test for latent tuberculosis infection (LTBI) before beginning the administration of biologics such as anti-TNF therapies (1). CD4-positive T cells are the main target in Human T-cell leukaemia virus type 1 (HTLV-1) infection. Several reports suggest that the reaction of tuberculin skin test (TST) is attenuated in HTLV-1-positive individuals compared with that in HTLV-1-negative individuals (2). However, it remains unclear whether IGRAs are reliable for detecting TB infection among HTLV-1-positive RA patients. Objectives:The present study aimed to investigate the usefulness of the T-SPOT.TBassay in HTLV-1-positive RA patients. In addition, the association between the existence of IFN-γ producing T cells and HTLV-1 proviral loads (PVLs) in HTLV-1-positive RA patients was analysed on the basis of the T-SPOT.TBassay results. Methods:We reviewed the medical records of 75 HTLV-1-negative and 29 HTLV-1-positive RA patients were suspected cases of LTBI and evaluated using the T-SPOT.TBassay as a clinical practice from April 2012 to July 2019. The results of T-SPOT.TBwere collected from medical records, retrospectively. Peripheral blood samples were obtained from HTLV-1-positive RA patients for the analysis of HTLV-1 PVLs values. The study protocol was approved by the research ethics committees of our hospitals. Results:Approximately 55% of the HTLV-1-positive RA patients showed invalid results for the T-SPOT.TBassay (p < 0.0001); the cause of invalid results was a spot-forming count of >10 spots in the negative controls of the T-SPOT.TBassay among HTLV-1-positive RA patients. Among HTLV-1-positive RA patients, HTLV-1 PVL values were significantly higher in 16 patients who showed invalid results than in 13 patients who did not (p = 0.003). There were no between-group differences in female patient ratio, age, RA disease activity and therapeutic regimens. IFN-γ producing cells were detected in the peripheral blood of HTLV-1-positive RA patients without stimulation with TB-specific antigens. Conclusion:The incidence of invalid results for the T-SPOT.TBassay has been reported to be as low as 0.6% (3). The results of this assay for screening of LTBI in HTLV-1-positive RA patients should be interpreted with caution. Furthermore, our results show that an increase in IFN-γ producing T cell numbers due to HTLV-1 infection in RA patients may affect the pathogenesis of RA.References:[1]Iannone, F., et al.J. Abstract:

Abstract:
The purpose of the study was to determine changes in central hemodynamics under the influence of the Yumeiho therapy procedure and to develop criteria for their evaluation Material & Methods: 55 people who received Yumeiho therapy procedures at the "Yumeiho" center in Lviv were under supervision. Of them, 29 women aged 38.7 (32.0; 52.4) years and 26 men aged 35.8 (29.0; 46.6) years. The average age of the examinees was 37.5 (31.5; 47.2) years. All individuals had non-specific signs of manifestations of osteochondrosis of the spine. The methodology for the study of the indicators of the cardiovascular system involved the recording of blood pressure (BP) and heart rate (HR) in the supine position after 5 minutes of rest before the procedure, as well as after 5 minutes of rest in the supine position after the completion of the Yumeiho therapy Results: stroke volume (SV, cm3), cardiac output (CO, dm3), general peripheral vascular resistance (GPVR, dyn/s/cm−5), cardiac index (CI, dm3/m2) and stroke index (SI, cm3/m2) were calculated according to known formulas. The analysis of changes in hemodynamic parameters during 1014 procedures was carried out. It is shown that in the Yumeiho-therapy procedure, there is a significant decrease in SV, CO, CI, SI and an increase in GPRV, both in men and in women. There is a decrease in the kinetics of blood circulation to the hypokinetic level. Conclusions: the conducted study made it possible to establish that under the influence of Yumeiho therapy in patients there is a significant decrease in SV (cm3), CO (dm3), CI (dm3/m2), SI (cm3/m2), and a significant increase in GPRV (dyn/s/cm–5). According to the apriori data of the percentile analysis of changes in hemodynamic parameters, the limits of pronounced deviations, which may indicate an inadequate reaction of the body to the procedure, are established. That is, the obtained results can be useful during operational control of the effect of the Yumeiho therapy procedure on the patient's body. In the future, it is possible to consider the use of Yumeiho therapy in cardiac patients.

Methods: Pooled analysis included study data from AMEERA-1 (NCT03284957), in which pts received single-agent amcenestrant in dose escalation (20-600 mg QD or 300 mg BID) and dose expansion (400 mg QD), and from AMEERA-2 (NCT03816839), in which Japanese pts received single-agent amcenestrant at 400 mg QD or 300 mg BID. Mutational profiling of plasma circulating cell-free DNA was conducted by droplet digital polymerase chain reaction (ddPCR) for 12 pathogenic ESR1 mutations at baseline (BL) and on treatment (OT; Cycle 2, Day 28), and by next-generation sequencing (NGS; 77-gene panel) at BL and end of treatment (EOT). Clinical benefit (CB; complete response + partial response [PR] + stable disease ≥ 24 weeks) was assessed in pooled pts who received amcenestrant≥ 150 mg QD or BID. In tumor tissue, ER, progesterone receptor (PgR), Ki67 and Bcl-2 expression levels over time were assessed by immunohistochemistry; functional ER pathway activity was assessed by gene set variation analysis with RNA-seq data (ER activity score). Results: Among pts with data for ESR1 mutation status at BL (N = 80; ddPCR), 41 pts were WT and 39 pts had at least one ESR1 mutation, with D538G and Y537S the most prevalent. CB was observed in pts with both WT (13 pts) and mutated ESR1 (12 pts) at BL. The most frequent pathogenic mutations detected in pts (N = 79; NGS) at BL were ESR1 (49%), followed by PI3KCA (46%)and TP53 (33%). In response-evaluable pts with ESR1 mutational profiling at BL and OT (N = 40; ddPCR), 17 of 19 pts with ESR1 mutation at BL showed a decrease in ESR1 mutation allele frequency, independently of clinical benefit. Eight pts showed an increase (7/25 no CB and 1/15 CB). Among 17 pts with biopsies, pts with ER or PgR -/low (H-score: 0-3) at BL did not derive benefit from treatment. In the remaining patients with ER high (H-score ≥ 140), no difference between pts with and without CB was observed in ER, Bcl-2 or ER activity score, while pts with CB had a tendency for lower Ki67. ER and PgR decreased upon treatment independently of CB, while ER activity score and Ki67 mostly decreased in pts with CB and mostly increased in pts without CB. Conclusions: In postmenopausal women with ER+/HER2- aBC treated with single-agent amcenestrant, low Ki67 at BL and the pharmacodynamic decrease of Ki67 and ER activation score by amcenestrant trended toward an association with CB, whereas increase in ESR1 mutation allele frequency on treatment trended toward an association with lack of CB. Clinical benefit was observed in both pts with WT and mutated ESR1 at BL. Citation Format: Hiroji Iwata, Aditya Bardia, Simon Lord, Hannah M. Abstract:
Abstract Background: Biomarkers predicting therapeutic response to immunotherapy have been widely explored via monitoring the liquid and tissue-derived components. Increasing treatment options for recurrent/metastatic head and neck squamous cell carcinoma (R/M HNSCC) mandates prediction of the therapeutic response of anti-PD-1 antibody alone as well as optimization of the treatment sequence. In view of improving biomarkers predicting the efficacy of immunotherapy for R/M HNSCC, we hypothesized that biomarkers can be personalized depending on clinicopathological backgrounds and treatment sequence. Methods: In this study, we retrospectively included formalin-fixed paraffin-embedded (FFPE) samples, peripheral blood cell counts at treatment, clinicopathological information, and outcome data for patients with R/M HNSCC receiving nivolumab across 22 institutions in Japan (N = 100). FFPE samples were subjected to 14-marker multiplex immunohistochemistry (IHC) and image cytometry analysis (Tsujikawa T et al. Cell Reports, 2017) to quantitatively evaluate CD8+ T cells, helper T cells, regulatory T cells, B cells, natural killer (NK) cells, macrophages, dendritic cells, CD66b+ granulocytes, mast cells, programmed death ligand 1 (PD-L1) and PD-1 expression in a single slide. Intratumoral and circulating immune cell frequencies were comparatively analyzed between responders (CR, n = 14; PR, n = 39) and non-responders (SD, n = 2; PD, n = 45). Results: Of 100 patients included, responders had significantly lower smoking and alcohol index, higher incidence of immune related adverse events, and higher PD-L1 expression in immune cells as well as PD-L1 combined positive score (CPS) than non-responders. Next, focusing on the history of prior therapy, stratified analysis revealed that the frequency of NK cells was associated with nivolumab response in patients with prior cetuximab use, but not in cetuximab-naïve status. Furthermore, stratified analysis by patient age revealed that nivolumab response was significantly associated with high CPS and lymphoid-inflamed profiles based on cell densities of nine immune cell lineages in the group aged 65 years or older, but not in the group under 65 years of age. On the contrary, the neutrophil/lymphocyte ratios (NLR) in peripheral blood counts at nivolumab treatment were significantly lower in responders (mean 4.96) than those in non-responders (mean 10.46) in the group under 65 years of age, but not in those over 65 years of age (7.41 versus 8.47). Conclusions: Using peripheral blood data and tumor tissue profiling stratified by patient age and prior treatment might provide better predictive biomarkers in nivolumab response to HNSCC. Further preclinical and clinical studies elucidating immune mechanisms in different patient backgrounds will be warranted. Abstract:
Abstract Introduction: Immunotherapy with immune checkpoint inhibitors (ICI) is the standard of care for advanced non-small-cell lung cancer (NSCLC) without driver gene alterations. However, survival benefits with ICI are limited to a small subset of NSCLC patients, and particularly ICI combinations with cytotoxic agents produce serious physical and large financial toxicities. Tumor PD-L1 expression levels/proportion score (TPS) are universally used as predictive biomarkers in ICI monotherapy response and outcomes. However, PD-L1/TPS assays are imperfect because of low analytical validity and reproducibility due to the visual-scoring system by pathologists. Therefore, additional biomarkers are urgently needed to predict ICI therapy response and outcomes. Recently, we reported that serum antibodies (Abs) against NY-ESO-1 and XAGE1 cancer-testis antigens were probably useful biomarkers in the prediction of clinical benefits with anti-PD-1 monotherapy for NSCLC (J Thorac Oncol, 2019). Additionally, we developed a fully automated immunoassay system (HISCLTM) to measure the Abs easily and rapidly (Sakai Y, et al. Clin Chim Acta 2021). In this study, we retrospectively examined whether the Abs measured by HISCLTM predict the clinical benefits with ICI monotherapy for NSCLC.Patients and Methods: Sera were obtained from controls of non-malignant lung diseases (n=75) and healthy subjects (n=100), and advanced NSCLC patients (n=263) to to determine a cutoff value in HISCLTM. In NSCLC patients analyzed here (n=78), sera were obtained before anti-PD-1 monotherapy with nivolumab in second-line or later settings. The serum NY-ESO-1/XAGE1 Abs were measured by HISCLTM, and we examined the relationships between the Abs levels, objective response rate (ORR), progression free survival (PFS), and overall survival (OS) after nivolumab monotherapy. Results: NY-ESO-1/XAGE1 Abs levels in NSCLC patients (n=263) were significantly higher than those in the controls (n=175). A cutoff value was determined as the Abs level of 10 SU/mL, calculated from the Abs values in the controls. The Abs (≥ 10 SU/mL) were detected in 21/78 (27%) of the NSCLC patients treated with nivolumab, and one patient had both NY-ESO-1 and XAGE1 Ab. An ORR was 62%, 16%, and 29% in the Abs-positives, the Abs-negatives, and overall, respectively. The Abs levels in responders were significantly higher than those in non-responders. The NSCLC patients with high-Abs values (≥ 10 SU/mL) had significantly better survivals with nivolumab monotherapy than those with low-Abs (PFS, HR 0.51, 95%CI 0.31-0.83, p < 0.01; OS, HR 0.51, 95%CI 0.31-0.84, p < 0.01). Interestingly, a few NSCLC patients with both high-Abs and driver genes responded well to nivolumab. Conclusions: Serum NY-ESO-1/XAGE1 Abs measured by HISCLTM are potential biomarkers that predict clinical benefits with anti-PD-1 monotherapy for NSCLC, even including driver genes. These findings warrant further biomarker studies using NY-ESO-1/XAGE1 Abs in clinical trial and practice of NSCLC immunotherapy. Abstract:
Palaeohydrochemical and palaeohydrological changes of lakes have seldom been reconstructed from the fossil shells of the gasropod Radix auricularia, which is a new, and potentially high-resolution environnmental archive. We conducted a geochemical and stable isotope study of the shells of Radix from the exposed fluvio-lacustrine sediments near Lake Yamdrok Yumtso in the southern Tibetan Plateau. Our aims were to determine the sedimentary environment, palaeo-lake hydrochemistry and hydrological status. AMS 14C and OSL dating indicates that a lake-level stage of Yamdrok Yumtso higher than that of today occurred during ~4.7–1.2 cal. kyr BP. Results of Sr/Ca, δ13C and δ18O analysis of the fossil shells of Radix auricularia indicate that the lake-level fluctuations were mainly controlled by changes of the Indian Summer Monsoon; decreasing evaporation during the mid- to late-Holocene was also responsible. In addition, based on the geochemical relationship between Radix sp. shells and the ambient water in lakes, the values of δ18OPalaeo-water and Sr/CaPalaeo-water reconstructed using the fossil shells of Radix auricularia are −8.2‰ to −5.1‰ and 0.0012 to 0.0057, respectively. Further, based on the values of δ18Oshell, together with geomorphological evidence, we infer that Yamdrok Yumtso was a closed lake system, and we estimate its possible extent during the interval of high lake-level. In addition, we speculate that the water level of Yamdrok Yumtso at this time exceeded 4448.9 m a.s.l., but was less than 4451 m a.s.l., and that the major separation of various components of the Yamdrok Yumtso system occurred after 1.2 kyr BP.

Abstract:
Background:Residual synovitis can be detected by sensitive modalities such as ultrasonography in patients with rheumatoid arthritis in clinical remission. On the other hand, a previous study has shown that ultrasound-guided treatment provides modest benefit compared to a conventional strategy aiming clinical remission in early patients. It is still unclear how discordant clinical remission is from imaging remission by ultrasonography in patients treated with biologic agentsObjectives:To clarify the discordance between clinical remission and imaging remission in patients with rheumatoid arthritis treated with biologic agents. Methods:Patients with rheumatoid arthritis who were treated with biologic agents and in clinical remission defined as disease activity score for 28 joints (DAS28)<2.6 were enrolled. All patients were performed comprehensive ultrasound examination of 44 joints as well as physical examinations. Ultrasound images of gray scale (GS) and power doppler (PD) were evaluated with a semi-quantitative score of 0-3. Imaging remission with ultrasound was defined as no PD signal detected in any joints. Clinical information was collected from their medical charts. Results:A total of 41 patients were enrolled with 22 patients treated with tumor necrosis factor (TNF)-α inhibitors and 19 with interleukin (IL)-6 inhibitors. The mean age, female ratio, the mean disease duration, and the mean duration of clinical remission were 60 years old, 87%, 5.1 years and 11.5 years. The imaging remission by ultrasonography was observed only in 51.2 %. When patients were divided according to biologic agents, baseline characteristics including median age, disease duration and clinically remission duration were comparable between both groups, while the rates of seropositivity and the stage of radiological progression was higher in IL-6 group (seropositivity, p=0.04; radiological progression, p=0.02). The mean DAS28 was 1.93 in the TNFα group and 1.02 in the IL-6 group. The discordance of clinical remission and imaging remission was observed in 28.6% of the TNFα group and 71.4% of the IL-6 group (p=0.03). The residual synovitis scores of GS and PD in 44 joints were significantly lower in the TNFα than the IL-6 group (GS, 1.1±1.8 vs 4.7 ± 4.6, p<0.01; PD, 0.6 ± 1.3 vs 3.3 ± 3.5, p<0.01, respectively). A receiver operating characteristic curve demonstrated an optimal score of DAS28 that discriminated imaging remission as 1.89 in the TNFα group and 1.25 in the IL-6 group. Conclusion:Our results showed that there was substantial discordance between clinical remission and imaging remission, especially in the patients treated with IL-6 inhibitors. In patients treated with biologic agents, clinical remission should be assessed more stringently than the usual 2.6, and ulltrasound-guided management may be useful.References:[1]Smolen JS, et al. Abstract:

Abstract:
Abstract. The contrasting patterns of lake-level fluctuations across the Tibetan Plateau (TP) are indicators of differences in the water balance over the TP. However, little is known about the key hydrological factors controlling this variability. The purpose of this study is to contribute to a more quantitative understanding of these factors for four selected lakes in the southern-central part of the TP: Nam Co and Tangra Yumco (increasing water levels), and Mapam Yumco and Paiku Co (stable or slightly decreasing water levels). We present the results of an integrated approach combining hydrological modeling, atmospheric-model output and remote-sensing data. The J2000g hydrological model was adapted and extended according to the specific characteristics of closed-lake basins on the TP and driven with High Asia Refined analysis (HAR) data at 10 km resolution for the period 2001–2010. Differences in the mean annual water balances among the four basins are primarily related to higher precipitation totals and attributed runoff generation in the Nam Co and Tangra Yumco basins. Precipitation and associated runoff are the main driving forces for inter-annual lake variations. The glacier-meltwater contribution to the total basin runoff volume (between 14 and 30 % averaged over the 10-year period) plays a less important role compared to runoff generation from rainfall and snowmelt in non-glacierized land areas. Nevertheless, using a hypothetical ice-free scenario in the hydrological model, we indicate that ice-melt water constitutes an important water-supply component for Mapam Yumco and Paiku Co, in order to maintain a state close to equilibrium, whereas the water balance in the Nam Co and Tangra Yumco basins remains positive under ice-free conditions. These results highlight the benefits of linking hydrological modeling with atmospheric-model output and satellite-derived data, and the presented approach can be readily transferred to other data-scarce closed lake basins, opening new directions of research. Future work should go towards a better assessment of the model-chain uncertainties, especially in this region where observation data are scarce.

Abstract:
Abstract. Lake-level fluctuations in closed basins on the Tibetan Plateau (TP) indicate climate-induced changes in the regional water balance. However, little is known about the region's key hydrological parameters, hampering the interpretation of these changes. The purpose of this study is to contribute to a more quantitative understanding of these controls. Four lakes in the south-central part of the TP were selected to analyze the spatiotemporal variations of water-balance components: Nam Co and Tangra Yumco (indicating increasing water levels), and Mapam Yumco and Paiku Co (indicating stable or slightly decreasing water levels). We present the results of an integrated approach combining hydrological modeling, atmospheric-model output and remote-sensing data. The hydrological model J2000g was adapted and extended according to the specific characteristics of closed lake basins on the TP and driven with "High Asia Refined analysis (HAR)" data at 10 km resolution for the period 2001–2010. Our results reveal that because of the small portion of glacier areas (1 to 7% of the total basin area) the contribution of glacier melt water accounts for only 14–30% of total runoff during the study period. Precipitation is found to be the principal factor controlling the water-balance in the four studied basins. The positive water balance in the Nam Co and Tangra Yumco basins was primarily related to larger precipitation amounts and thus higher runoff rates in comparison with the Paiku Co and Mapam Yumco basins. This study highlights the benefits of combining atmospheric and hydrological modeling. The presented approach can be readily transferred to other ungauged lake basins on the TP, opening new directions of research. Future work should go towards increasing the atmospheric model's spatial resolution and a better assessment of the model-chain uncertainties, especially in this region where observational data is missing.
